Abstract:Herein, we report a mild and operationally simple photoredox/NHC dual catalysis strategy for the α-carboxylation of tertiary amine C(sp3)-H bonds using diethyl pyrocarbonate. This method offers a novel approach for synthesizing α-amino acid derivatives. The protocol features a broad substrate scope, accommodating both N-aryl tetrahydroisoquinolines (THIQ) and N-methyl aniline and is scalable to gram quantities. Additionally, it is suitable for the late-stage derivatization of certain pharmaceutical compounds.

Abstract:The asymmetric cross-coupling reaction is developed as a straightforward strategy toward 1,1-diaryl alkanes, which are a key skeleton in a series of natural products and bioactive molecules in recent years. Here we report an enantioselective benzylic C(sp(3))-H bond arylation via photoredox/nickel dual catalysis. Sterically hindered chiral biimidazoline ligands are designed for this asymmetric cross-coupling reaction. Readily available alkyl benzenes and aryl bromides with various functional groups tolerance can be easily and directly transferred to useful chiral 1,1-diaryl alkanes including pharmaceutical intermediates and bioactive molecules. This reaction proceeds smoothly under mild conditions without the use of external redox reagents.

Abstract:The functionalization of α-C(sp 3 )–H bonds in amines has become a focal point of contemporary research. Here, we report a new approach utilizing photocatalysis α-C(sp 3 )–H bond functionalization in alicyclic and aliphatic secondary amines facilitated by intramolecular 1,5-hydrogen atom transfer (HAT). This finding unlocks a sustainable method for rapidly constructing complex heterocyclics via cross-dehydrogenative C–C coupling of protected amines and nitrogen-containing heterocycles. This protocol boasts broad applicability to various substrates, exhibits tolerance to numerous functional groups, and supports the late-stage modification of drug molecules.

Abstract:Herein, we have reported a fully atom- & highly step-economic cascade dearomatization with non-activated phenyl and other hetero-aryl rings for accesses to tethered-aniline spiro-lactam with broad substrate scopes. This protocol involves a photoredox-induced hydrogen transfer/Giese addition/ dearomative cyclization/ protonation cascade and enables the introduction two fragments of the C(sp3)-H bond of N-methylaniline into N-benzyl-acrylamide. Interestingly, dearomatice products of pyridine ring can be further transformed into pentacyclic alkaloid skeletons in a a fully atom-economic fashion again.

Abstract:In this work, we have established a photoredox/Ti dual‐catalyzed dehydroxylation of cyclobutanone oximes. The resulting γ‐cyanoalkyl radicals undergo a cascade radical addition/cyclization with N‐acrylamides to afford cyanoalkyl substituted oxoindolines under mild conditions. This reaction also exhibits broad substrate scope and good functional group tolerance. Furthermore, it is applicable to late‐stage functionalization of complex molecules and gram‐scale synthesis, which illustrates its potential value in organic synthesis.

Abstract:Herein, we disclose a visible-light-driven photoredox-catalyzed protocol for site-selective alkylation of glycine derivatives via 1,2-hydrogen atom transfer, which is distinguished by metal free and mild conditions, high chemoselectivity, and good functional group compatibility. This protocol provides a unique approach for synthesizing valuable α,β-diamino acid derivatives. Abstract:α-Chiral alkyl phosphines are privileged structural motifs with a wide application in organic and medical synthesis. It is highly desirable to develop stereoselective methods to prepare these enantioenriched molecules. The incorporation of C( sp 3 )─H functionalization and chiral phosphine chemistry is much less explored, probably because of the weak reactivity of C( sp 3 )─H bonds and/or the challenging site- and stereoselectivity issues. Herein, we disclose a synergistic catalysis system to enable an enantioselective radical addition process of α-substituted vinylphosphine oxides. An array of diverse α-chiral alkyl phosphors compounds is smoothly accessed by using the readily available chemicals as the inert C( sp 3 )─H bond reagent, such as sulfides, amines, alkenes, and toluene derivatives, exerting remarkable chemo-, site-, and enantioselectivity. On the basis of the mechanistic studies, both the C( sp 3 )─H bond activation and the stereochemistry-determining step are proposed to involve a single-electron transfer/proton transfer process.

Abstract:Conjugation of amino acids to bioactive molecules has emerged as a promising strategy for optimizing pharmacological profiles of lead candidates in drug discovery. This study describes a photocatalytic Minisci‐type transfer reaction of N‐acyl amino acids into various N‐heterocycles. Notably, this protocol enables direct conjugation of amino acids into heterocyclic C–H bonds, eliminating the need for prefunctionalized substrates. A diverse array of N‐heterocycles, amino acids, oligopeptides, and drugs were used to demonstrate the potential of the proposed approach. In addition, the importance of this approach is demonstrated through its application in the DNA‐encoded library chemistry. Various synthetic transformations and preliminary mechanistic investigations were also explored.

Abstract:Catalytic ring-opening/functionalization of unactive cyclopropanes has proven to be a significant but challenging task in organic synthesis. Herein, we disclose the photoredox and cobalt cocatalyzed ring-opening/acceptorless dehydrogenative functionalization of monodonor cyclopropanes, which provides a promising platform to achieve a sustainable and atom-economic approach to assemble allylic N -acyl-acetal derivatives. Abstract:Tetrahydropyridazines constitute an important structural motif found in numerous natural products and pharmaceutical compounds. Herein, we report an aminoacylation reaction of alkenes that enables the synthesis of 1,4,5,6-tetrahydropyridazines through cooperative N-heterocyclic carbene (NHC) and photoredox catalysis. This approach involves the 6-endo-trig cyclization of N-centered hydrazonyl radicals, generated via single-electron oxidation of hydrazones, followed by a radical-radical coupling step. The mild process tolerates a wide range of common functional groups and affords a variety of tetrahydropyridazines in moderate to high yields. Preliminary investigations using chiral NHC catalysts demonstrate the potential of this protocol for asymmetric radical reactions.

Abstract:Lactone motifs are prevalent in both naturally and synthetically bioactive molecules. We report herein a photoredox-catalyzed formal [5 + 2] annulation of O-acylhydroxamide derivatives with 1,3-dienes/1,3-enynes for rapid synthesis of structurally diverse alkene/alkyne-containing 7-membered lactones. The reaction features selective transformation of distal unactivated C(sp3)–H bonds and a broad substrate scope. A radical mechanism involving nitrogen-directed 1,5-hydrogen atom transfer (HAT), facile coupling of alkyl radicals with 1,3-dienes/1,3-enynes, and the construction of 7-membered lactone motifs via C–O bond formation is proposed.

Abstract:An asymmetric catalytic strategy in photocatalysis utilizing a chirality-induced approach through an organocatalytic/photoredox relay catalysis strategy is successfully achieved for the rapid construction of polycyclic compounds containing vicinal amino alcohols in a one-pot protocol. This methodology facilitates the efficient synthesis of diverse substituted polycyclic tetra-hydroquinoline and benzofuran-derived vicinal amino alcohols, each containing five consecutive chiral centers, with high yields, excellent diastereoselectivities and enantioselectivities (up to 95% yield, >20:1 dr and 98% ee), under mild reaction conditions driven by sequential bifunctional squaramide organocatalyst-catalyzed [4+2] annulation and photocatalyst-catalyzed ketyl radical addition cyclization reaction process. Furthermore, investigations into the stereoselectivity mechanism and high-resolution mass spectrometry (HRMS) experiments on free radical trapping have provided evidence for elucidating the detailed mechanism of chirality-induced processes and chiral intermediate conversions in this procedure.
