Abstract:Amines are a quintessential moiety in bioactive molecules, pharmaceuticals and organic materials. Transition-metal-catalysed C–N coupling of aryl electrophiles has been established as a powerful and reliable method for amine synthesis. However, the analogous C–N coupling of alkyl electrophiles is largely under-developed due to the decomposition of metal alkyl intermediates by β-hydrogen elimination and difficulty in C(sp3)–N reductive elimination. Here, we provide a general strategy for amination of alkyl electrophiles by merging photoredox and copper catalysis. Photoredox catalysis allows the use of alkyl redox-active esters, recently established as a superior class of alkyl electrophiles, whereas copper catalysis enables C(sp3)–N cross-coupling. Decarboxylative amination can be used for the synthesis of a diverse set of alkyl anilines with high chemoselectivity and functional-group compatibility. Rapid functionalization of amino acids, natural products and drugs is demonstrated.

What problem does this paper attempt to address?
The problem that this paper attempts to solve is how to achieve the C - N bond - forming reaction between alkyl electrophiles and amine nucleophiles through a new method in organic synthesis. Specifically, the paper focuses on developing a general strategy to realize the decarboxylative amination of redox - active esters by combining photoredox and copper catalysis to synthesize various types of alkyl anilines. This method aims to overcome several major challenges in traditional C - N coupling reactions: 1. **β - Hydrogen Elimination**: Under metal catalysis, the metal - alkyl intermediates generated from alkyl electrophiles are prone to β - hydrogen elimination, resulting in a decrease in the yield of the target product. 2. **Difficulty in C(sp³) - N Reductive Elimination**: In the process of C(sp³) - N bond formation, the reductive elimination step is difficult to achieve, which is a crucial step in C - N coupling reactions. 3. **Functional Group Compatibility**: Traditional methods often have problems with selectivity and compatibility when dealing with substrates containing multiple functional groups. The method proposed in the paper can not only efficiently synthesize various alkyl anilines, but also has high chemoselectivity and compatibility with multiple functional groups, thus providing new approaches for drug synthesis and the preparation of organic materials. In addition, this method also avoids the over - alkylation problem common in the direct alkylation process and is applicable to bulky primary and secondary alkyl groups.
