Abstract:Introduction: The oxygen uptake ( V ̇ O 2 ) vs power output relationship from ramp incremental exercise is used to prescribe aerobic exercise. As power output increases, there is a delay in V ̇ O 2 that contributes to a misalignment of V ̇ O 2 from power output; the mean response time (MRT). If the MRT is not considered in exercise prescription, ramp incremental-identified power outputs will elicit V ̇ O 2 values that are higher than intended. We compared three methods of determining MRT (exponential modeling (MRT EXP ), linear modeling (MRT LIN ), and the steady-state method (MRT SS )) and evaluated their accuracy at predicting the V ̇ O 2 associated with power outputs approximating 75% and 85% of gas exchange threshold and 15% of the difference between gas exchange threshold and maximal V ̇ O 2 (Δ15). Methods: Ten males performed a 30-W∙min −1 ramp incremental and three 30-min constant power output cycle ergometer trials with intensities at 75% gas exchange threshold, 85% gas exchange threshold, and ∆15. At each intensity, the measured steady-state V ̇ O 2 during each 30-min test was compared to the V ̇ O 2 predicted after adjustment by each of the three MRTs. Results: For all three MRT methods, predicted V ̇ O 2 was not different ( p = 1.000) from the measured V ̇ O 2 at 75%GET (MRT EXP , 31 mL, MRT LIN , −35 mL, MRT SS 11 mL), 85%gas exchange threshold (MRT EXP −14 mL, MRT LIN −80 mL, MRT SS −32 mL). At Δ15, predicted V ̇ O 2 based on MRT EXP was not different ( p = .767) from the measured V ̇ O 2 , but was different for MRT LIN ( p < .001) and MRT SS ( p = .03). Conclusion: Given that the intensity is below gas exchange threshold, all model predictions implemented from the current study matched the exercise prescription.

Mechanisms of slowed V̇O2 on-kinetics in second step of two-step-incremental exercise in skeletal muscle

Abstract:Simulations using a computer model of the skeletal muscle bioenergetic system demonstrate that the slowed V̇O2 on-kinetics of the second step in two-step incremental exercise (exercise initiated from elevated baseline metabolic rate) can be accounted for by a decrease in the stimulation of oxidative phosphorylation (OXPHOS) and/or increase in the stimulation of glycolysis through each-step activation (ESA) in working skeletal muscle. This effect can be caused by either a recruitment of more glycolytic type IIa, IIx and IIb fibers or metabolic regulation in already recruited fibers, or both. The elevated-glycolysis-stimulation mechanism predicts that the end-second-step pH in two-step-incremental exercise is lower than the end-exercise pH in constant-power exercise with the same work intensity (power output). The lowered-OXPHOS-stimulation mechanism predicts higher end-exercise ADP and Pi, and lower PCr in the second step of two-step-incremental than in constant-power exercise. Comparison of laser-induced surface damage density measurements with small and large beams: toward representativeness.

Abstract:Pulsed laser damage density measurements obtained with diverse facilities are difficult to compare, due to the interplay of numerous parameters, such as beam area and pulse geometry, which, in operational large beam conditions, are very different from laboratory measurements. This discrepancy could have a significant impact; if so, one could not even pretend that laser damage density control is a real measurement process. In this paper, this concern is addressed. Tests with large beams of centimeter size on a high-power laser facility have beam performed according to a parametric study and are compared to small beam laboratory tests. It is shown that laser damage densities obtained with large and small beams are equal, within calculated error bars.
The Oxygen Uptake Response to Incremental Ramp Exercise

Abstract:An incremental ramp exercise is a protocol that is frequently used in the domain of exercise testing to get an insight into the exercise tolerance of both healthy active populations (including athletes) and patients, due to the specific characteristics of the protocol. The continuous and linear increase in work rate is not only less strenuous for populations with a very low exercise capacity but it requires the aerobic metabolism to adapt to the continuously changing conditions. Therefore, this protocol can provide important information on the adaptive capacity of individuals to exercise in non-steady-state conditions. The ramp exercise has also been used in the past two decades to get an insight into the underlying mechanisms of the oxygen uptake ($$\dot V$$O2) response (and kinetics) to exercise. Against the expectations, it has been shown that the parameters that quantify the $$\dot V$$O2 response to ramp exercise do not completely correspond to those obtained from constant work-rate transitions and incremental step exercise. For that reason, it could be concluded that the $$\dot V$$O2 response is specific to ramp exercise, and thus is determined by other mechanisms than those which determine other protocols. Although the $$\dot V$$O2 response to ramp exercise has a high level of reproducibility and a uniform pattern can be observed, especially for the $$\dot V$$O2 response below the gas exchange threshold (GET) [above the GET, the $$\dot V$$O2 response is less clear, some prudence is necessary when interpreting potential differences in the $$\dot V$$O2 response between populations. Several methodological issues (e.g. baseline work rate, ramp slope) exert an important impact on the $$\dot V$$O2 response to ramp exercise. The main purpose of this review is to provide an overview of the methodological and physiological factors that have an impact on the $$\dot V$$O2 response to ramp exercise. It is of importance that exercise physiologists take these factors into consideration, not only prior to the conductance of the ramp exercise in a variety of subjects, but also when interpreting the obtained results.

Abstract:ABSTRACT Introduction Traditional neuromuscular fatigue assessments are not task-specific and are unable to characterize neuromuscular performance decline during dynamic whole-body exercise. This study used interleaved maximal isokinetic cycling efforts to characterize the dynamics of the decline in neuromuscular performance during ramp-incremental (RI) cycle ergometry exercise to intolerance. Methods Eleven young healthy participants (10 male/1 female) performed two RI cycle ergometry exercise tests to intolerance: [1] RI-exercise with peak isokinetic power (Piso) at 80 rev·min -1 measured at baseline and immediately at intolerance from a maximal ~6 s effort; [2] RI-exercise where additional Piso measurements were interleaved every 90 s to characterize the decline in neuromuscular performance during the RI-test. Muscle excitation was measured using EMG during all Piso assessments, and pulmonary gas exchange was measured throughout. Results Baseline Piso was 832 ± 140 W and RI-exercise reduced Piso to 349 ± 96 W at intolerance (p = 0.001), which was not different from flywheel power at intolerance (303 ± 96 W; p = 0.292). There was no reduction in Piso between baseline cycling and gas exchange threshold (GET; baseline Piso vs. mean Piso below GET: 828 ± 146 vs. 815 ± 149 W; p = 1.00). Piso fell progressively above GET until intolerance (Piso every 90 s above GET: 759 ± 139; 684 ± 141; 535 ± 144; 374 ± 117 W; each p < 0.05 vs. baseline and mean Piso below GET). Peak muscle excitation (EMG) was also reduced only above GET (73 ± 14 % of baseline, at intolerance; p < 0.05). However, the reduction in peak Piso preceded the reduction in peak muscle excitation. Conclusions The dynamics of the decline in neuromuscular performance (reduction in Piso and EMG) during RI-exercise are consistent with known intensity-dependent metabolic and traditional pre-post neuromuscular fatigue responses to discrete bouts of constant-power exercise.

Abstract:Abstract. The purposes of this study were to compare the acute cardiorespiratory responses and muscle oxygenation trends during incremental cycle exercise to exhaustion with those observed during 30 s and 45 s Wingate tests in healthy men and women, and to examine the relationships between selected variables among these tests. Seventeen healthy junior badminton players, nine men [mean age, height, body mass and maximal oxygen uptake ($$\dot V{\rm O}_{{\rm 2max}} $$) were 15.8 (SD 0.8) years, 1.73 (SD 0.08) m, 65.6 (SD 6.3) kg and 50.6 (SD 6.9) ml·kg–1·min–1 respectively] and eight women [mean age, height, body mass and $$\dot V{\rm O}_{{\rm 2max}} $$ were 16.6 (SD 1.0) years, 1.65 (SD 0.03) m, 62.7 (SD 4.5) kg and 42.0 (SD 5.0) ml·kg–1·min–1 respectively] completed a stepwise incremental exercise test to voluntary exhaustion and two Wingate tests lasting 30 s and 45 s in three separate sessions in random order. Cardiorespiratory responses were monitored breath-by-breath using a metabolic cart interfaced with an electrocardiogram. Tissue absorbancy trends were continuously recorded from the right vastus lateralis muscle using dual wavelength near infrared spectroscopy. Oxygen uptake and heart rate were significantly higher during the incremental test when compared to the two Wingate tests in the men and women. However, the oxygen pulse (oxygen utilization per heart beat, i.e., the product of stroke volume and arterio-venous oxygen difference) was not significantly different among the three tests in both sexes. The minimal tissue absorbancy, an index of muscle deoxygenation, was also not significantly different among the three tests in both sexes. Significant relationships were observed for the oxygen uptake (r2=0.72) and oxygen pulse (r2=0.60) between the incremental and 45 s Wingate tests in the sample for both sexes combined. The minimal tissue absorbancy, however, was not significantly related between the two tests. It was concluded that the significantly higher oxygen uptake during the incremental test was due to the higher heart rate because: firstly, oxygen pulse was not significantly different among the three tests, and secondly, peripheral factors, as indicated by the changes in muscle oxygenation, were not significantly different among the three test conditions. Although the peak values of the oxygen pulse during the incremental and 45 s Wingate tests were significantly correlated, the common variance of the minimal tissue absorbancy measurements between these two tests was quite low, suggesting considerable variation in the peripheral contribution during these two tests.

Abstract:Purpose: This study aimed to cross-validate a recently proposed equation for the prediction of maximal oxygen uptake ( V ̇O2max) in cycling exercise by using the average power output normalized by the body mass from a 5-minute time trial (RPO5-min) as the independent variable. Further, the study aimed to update the predictive equation using Bayesian informative prior distributions and meta-analysis. Methods: On different days, 49 male cyclists performed an incremental graded exercise test until exhaustion and a 5-minute time trial on a stationary cycle ergometer. We compared the actual V ̇O2max with the predicted value obtained from the RPO5-min, using a modified Bayesian Bland–Altman agreement analysis. In addition, this study updated the data on the linear regression between V ̇O2max and RPO5-min, by incorporating information from a previous study as a Bayesian informative prior distribution or via meta-analysis. Results: On average, the predicted V ̇O2max using RPO5-min underestimated the actual V ̇O2max by −6.6 mL·kg–1·min–1 (95% credible interval, −8.6 to −4.7 mL·kg–1·min–1). The lower and upper 95% limits of agreement were −17.2 (−22.7 to −12.3) and 3.8 (−1.0 to 9.5) mL·kg–1·min–1, respectively. When the current study's data were analyzed using the previously published data as a Bayesian informative prior distribution, the accuracy of predicting sample means was found to be better when compared with the data combined via meta-analyses. Conclusions: The proposed equation presented systematic bias in our sample, in which the prediction underestimated the actual V ̇O2max. We provide an updated equation using the previous one as the prior distribution, which could be generalized to a greater audience of cyclists.

Abstract:Peak oxygen uptake (VO 2 ), evaluated as exercise tolerance, is a strong predictor of life prognosis regardless of health condition. Several previous studies have reported that peak VO 2 is higher in those with a greater decrease in muscle oxygen saturation (SmO 2 ) in the active muscles during incremental exercise. However, the skeletal muscle characteristics of individuals exhibiting a greater decrease in SmO 2 during active muscle engagement in incremental exercise remain unclear. This study aimed to clarify the relationship among muscle strength, muscle endurance, and skeletal muscle oxygenation dynamics in active leg muscles during incremental exercise. Twenty-four healthy young men were included and categorized into the non-moderate-to-high muscular strength and endurance group (those with low leg muscle strength, endurance, or both; n = 11) and the moderate-to-high muscular strength and endurance group (those with both moderate-to-high leg muscle strength and endurance; n = 13). All participants underwent cardiopulmonary exercise testing combined with near-infrared spectroscopy to assess whole-body peak VO 2 and the change in SmO 2 at the lateral vastus lateralis from rest to each exercise stage as skeletal muscle oxygenation dynamics. A linear mixed-effects model, with the change in SmO 2 from rest to each stage as the dependent variable, individual participants as random effects, and group and exercise load as fixed effects, revealed significant main effects for both group ( P = 0.001) and exercise load ( P < 0.001) as well as a significant interaction between the two factors ( P < 0.001). Furthermore, multiple-comparison test results showed that the change in SmO 2 from rest to 40%–100% peak VO 2 was significantly higher in the moderate-to-high muscular strength and endurance group than in the non-moderate-to-high muscular strength and endurance group. Maintaining both muscle strength and endurance at moderate or higher levels contributes to high skeletal muscle oxygenation dynamics (i.e., greater decrease in SmO 2 ) during moderate- or high-intensity exercise.

Abstract:ABSTRACT Introduction This study assessed the effect of individualized, domain-based exercise intensity prescription on changes in maximal oxygen uptake (V̇O 2max ) and submaximal thresholds. Methods Eighty-four young healthy participants (42 Females, 42 Males) were randomly assigned to six age, sex, and V̇O 2max -matched groups (14 participants each). Groups performed continuous cycling in the 1) moderate (MOD)-, 2) lower heavy (HVY1)-, and 3) upper heavy-intensity (HVY2)- domain; interval cycling, in the form of 4) high-intensity interval training (HIIT) in the severe-intensity domain, or 5) sprint-interval training (SIT) in the extreme-intensity domain; or no exercise for, 6) control (CON). All training groups except SIT, were work-matched. Training participants completed three sessions per week for six weeks with physiological evaluations performed at PRE, MID and POST intervention. Results Compared to the change in V̇O 2max (∆V̇O 2max ) in CON (0.1 ± 1.2 mL·kg -1 ·min -1 ), all training groups except MOD (1.8 ± 2.7 mL·kg -1 ·min -1 ), demonstrated a significant increase (p < 0.05). HIIT produced the highest increase (6.2 ± 2.8 mL·kg -1 ·min -1 ) followed by HVY2 (5.4 ± 2.3 mL·kg -1 ·min -1 ), SIT (4.7 ± 2.3 mL·kg -1 ·min -1 ), and HVY1 (3.3 ± 2.4 mL·kg -1 ·min -1 ), respectively. The Δ PO at the estimated lactate threshold (θ LT ) was similar across HVY1, HVY2, HIIT and SIT which were all greater than CON (p < 0.05). The Δ V̇O 2 and Δ PO at θ LT for MOD was not different from CON (p > 0.05). HIIT produced the highest Δ PO at maximal metabolic steady state, which was greater than CON, MOD, and SIT (p < 0.05). Conclusions This study demonstrated that i) exercise intensity is a key component determining changes in V̇O 2max and submaximal thresholds and ii) exercise intensity domain-based prescription allows for a homogenous metabolic stimulus across individuals.

Metabolic and Perceptual Responses to Constant Heart Rate Exercise at Vigorous Intensities in Women

Abstract:ABSTRACT Purpose This study quantified the metabolic demands (oxygen uptake [V̇O 2 ]), power output adjustments, changes in the V̇O 2 /power output ratio, and perceptual responses (rating of perceived exertion [RPE]) during constant heart rate (HR) exercise performed within the vigorous intensity range (77-95% HR peak ). Methods Twelve women (mean ± SD age: 22 ± 4 years) performed a graded exercise test to exhaustion to determine peak parameters and 3, randomly ordered, constant HR trials to exhaustion or for 60 min at the lower (HR L = 77% HR peak ), middle (HR M = 86% HR peak ), and higher (HR H = 95% HR peak ) end of the vigorous intensity range. Time course of changes and patterns of responses were examined for V̇O 2 , power output, V̇O 2 /power output, and RPE for the composite and for each subject. Results Across the HR L (time to exhaustion [T lim ] = 56.3 ± 9.9 min), HR M (51.8 ± 13.5 min), and HR H (27.2 ± 17.7 min) trials, V̇O 2 and power output decreased quadratically ( p < 0.05) relative to the initial value from 10-100% of T lim , while the V̇O 2 /power output increased quadratically from 20-100% T lim , and RPE increased linearly from 50-100% T lim . The V̇O 2 and RPE, collapsed across time, for HR L (54.3 ± 3.3% V̇O 2peak , 11 ± 1.5 RPE) were lower than HR M (64.9 ± 4.5% V̇O 2peak , 14 ± 1.7 RPE) and both were lower than HR H (80.1 ± 4.1% V̇O 2peak , 17 ± 1.4 RPE). None of the 12 subjects at HR L , 6 at HR M , and 7 at HR H were within the vigorous V̇O 2 range. Conclusions The HR L was not sufficient to meet the desired metabolic intensity for vigorous exercise, while the middle to higher end of the range elicited a V̇O 2 within the prescribed range of only ~50-60% of the subjects. This study indicated that exercise held constant at a percentage of HR peak cannot consistently be used to prescribe a desired metabolic stimulus.

Abstract:Abstract Purpose Power output at the moderate-to-heavy-intensity transition decreases during prolonged exercise, and resilience to this has been termed ‘durability’. The purpose of this study was to assess the relationship between durability and the effect of prolonged exercise on severe-intensity performance, and explore intramuscular correlates of durability. Methods On separate days, 13 well-trained cyclists and triathletes (V̇O 2 peak, 57.3 ± 4.8 mL kg −1 min −1 ; training volume, 12 ± 2.1 h week −1 ) undertook an incremental test and 5-min time trial (TT) to determine power output at the first ventilatory threshold (VT 1 ) and severe-intensity performance, with and without 150-min of prior moderate-intensity cycling. A single resting vastus lateralis microbiopsy was obtained. Results Prolonged exercise reduced power output at VT 1 (211 ± 40 vs. 198 ± 39 W, ∆ -13 ± 16 W, ∆ -6 ± 7%, P = 0.013) and 5-min TT performance (333 ± 75 vs. 302 ± 63 W, ∆ -31 ± 41 W, ∆ -9 ± 10%, P = 0.017). The reduction in 5-min TT performance was significantly associated with durability of VT 1 (r s = 0.719, P = 0.007). Durability of VT 1 was not related to vastus lateralis carnosine content, citrate synthase activity, or complex I activity ( P > 0.05). Conclusion These data provide the first direct support that durability of the moderate-to-heavy-intensity transition is an important performance parameter, as more durable athletes exhibited smaller reductions in 5-min TT performance following prolonged exercise. We did not find relationships between durability and vastus lateralis carnosine content, citrate synthase activity, or complex I activity.

Comparative Bilateral Measurements of Vastus Lateralis Muscle Oxygen Desaturation Kinetics during 30 S Sprint Cycling Exercise: Effects of Age and Performance

Abstract:The study assessed vastus lateralis oxygen desaturation kinetics (SmO2) in 32 male cyclists (16 Seniors, 16 Juniors) during a 30 s sprint, examining effects of age and performance. An incremental test was used to determine ventilatory thresholds (VT1, VT2) and maximal oxygen uptake (VO2kg), followed by a sprint test to evaluate anaerobic performance. Cyclists' performance phenotype was determined as the ratio of power at VT2 to 5 s peak sprint power. Juniors exhibited sprinter-like traits, excelling in all functional tests except for lactate levels post-sprint. SmO2 data showed no age-related or bilateral differences across participants. The combined mean response time (MRT) revealed stronger bilateral goodness of fit (R2 = 0.64) than individual time delay (TD) and time constant (τ). Higher VO2kg at VT2, peak power, and maximal uptake were linked to longer TD, while shorter TD correlated with higher lactate production and increased fatigue. Bilaterally averaged SmO2 kinetics distinguished between sprint and endurance athletes, indicating the potential to reflect the alactic anaerobic system's capacity and depletion. Age did not affect desaturation rates, but younger cyclists showed greater response amplitude, attributed to a higher initial baseline rather than maximal desaturation at the end of the exercise.
A comparison of critical power and the respiratory compensation point at slower and faster pedaling cadences

Abstract:We investigated whether pedal cadence (60 vs. 100 rpm) affects oxygen uptake (V̇O2) and power output (PO) at two indexes of the heavy-to-severe-intensity domain boundary (i.e., critical power (CP) and respiratory compensation point (RCP)) and their correspondence. Fourteen adults (7 females, 23 ± 2 years) cycled at 60 and 100 rpm during: (i) a "step-ramp-step" protocol to identify V̇O2 and PO at RCP; (ii) 4-5 exhaustive constant-PO bouts for CP identification; and (iii) a constant-power bout at CP to identify V̇O2 at CP. Separate two-way repeated measures Analysis of variance assessed whether V̇O2 and PO were affected by index (CP vs. RCP) and cadence (60 vs. 100 rpm). The V̇O2 was not affected by index (mean difference (MD) = 73 ± 197 mL·min-1; p = 0.136) but there was an index × cadence interaction (p = 0.014), such that V̇O2 was higher at 100 versus 60 rpm for CP (MD = 142 ± 169 mL·min-1; p = 0.008), but not RCP (p = 0526). The PO was affected by cadence (MD = 13 ± 9 W; p < 0.001) and index (MD = 8 ± 11 W; p = 0.016), with no cadence × index interaction (p = 0.168). The systematic bias in PO confirms cadence-specificity of CP and RCP. The relationship between these indexes and their change in unison in PO suggests a mechanistic link between these two heavy-to-severe domain boundary candidates.
Maximal Lactate Steady State Versus the 20-Minute Functional Threshold Power Test in Well-Trained Individuals: "Watts" the Big Deal?

Abstract:Purpose: To (1) compare the power output (PO) for both the 20-minute functional threshold power (FTP20) field test and the calculated 95% (FTP95%) with PO at maximal lactate steady state (MLSS) and (2) evaluate the sensitivity of FTP95% and MLSS to training-induced changes. Methods: Eighteen participants (12 males: 37 [6] y and 6 females: 28 [6] y) performed a ramp-incremental cycling test to exhaustion, 2 to 3 constant-load MLSS trials, and an FTP20 test. A total of 10 participants returned to repeat the test series after 7 months of training. Results: The PO at FTP20 and FTP95% was greater than that at MLSS (P = .00), with the PO at MLSS representing 88.5% (4.8%) and 93.1% (5.1%) of FTP and FTP95%, respectively. MLSS was greater at POST compared with PRE training (12 [8] W) (P = .002). No increase was observed in mean PO at FTP20 and FTP95% (P = .75). Conclusions: The results indicate that the PO at FTP95% is different to MLSS, and that changes in the PO at MLSS after training were not reflected by FTP95%. Even when using an adjusted percentage (ie, 88% rather than 95% of FTP20), the large variability in the data is such that it would not be advisable to use this as a representation of MLSS.
Heart Rate Acquisition and Threshold-Based Training Increases Oxygen Uptake at Metabolic Threshold in Triathletes: A Pilot Study

Abstract:Exercise intensity is a critical component of the exercise prescription model. However, current research employing various non-specific exercise intensity protocols have reported wide variability in maximum oxygen uptake (VO2max) improvement after training, suggesting a present lack of consensus regarding optimal heart rate (fC) training zones for maximal athletic performance. This study examined the relationship between percentage of time (%time) spent training between the metabolic (VO2θ) and ventilatory thresholds (VEθ), and the resultant change in markers of aerobic performance. Thirteen (6 males) collegiate club-level triathletes were recruited for eight weeks of remote fC monitoring during all running and cycling sessions. Participants donned a forearm-worn optical fC sensor paired to a smartphone that collected and stored fCs. Subjects were categorized into Low and High groups based on %time spent training between the VO2θ and VEθ. Significant increases were observed in relative VO2max (P = 0.007, g = 0.48), VO2θ (P = 0.018, g = 0.35), and VEθ (P = 0.030, g = 0.29) from baseline after eight weeks for both groups. A 95% bootstrapped confidence interval that did not include zero (−0.38, −0.03; g = 1.26) revealed a large and significantly greater change in VO2θ in the High group (0.37 ± 0.15 L/min) versus the Low group (0.17 ± 0.14 L/min). No significant differences were observed in other variables between groups. Increasing triathletes’ %time spent exercising between VO2θ and VEθ may optimize increases in VO2θ after eight weeks of training.

Limitations to systemic and locomotor limb muscle oxygen delivery and uptake during maximal exercise in humans

Abstract:Reductions in systemic and locomotor limb muscle blood flow and O2 delivery limit aerobic capacity in humans. To examine whether O2 delivery limits both aerobic power and capacity, we first measured systemic haemodynamics, O2 transport and O2 uptake during incremental and constant (372 +/- 11 W; 85% of peak power; mean +/- S.E.M.) cycling exercise to exhaustion (n = 8) and then measured systemic and leg haemodynamics and during incremental cycling and knee-extensor exercise in male subjects (n = 10). During incremental cycling, cardiac output and systemic O2 delivery increased linearly to 80% of peak power (r2 = 0.998, P < 0.001) and then plateaued in parallel to a decline in stroke volume (SV) and an increase in central venous and mean arterial pressures (P < 0.05). In contrast, heart rate and increased linearly until exhaustion (r2 = 0.993; P < 0.001) accompanying a rise in systemic O2 extraction to 84 +/- 2%. In the exercising legs, blood flow and O2 delivery levelled off at 73-88% of peak power, blunting leg per unit of work despite increasing O2 extraction. When blood flow increased linearly during one-legged knee-extensor exercise, per unit of work was unaltered on fatigue. During constant cycling, , SV, systemic O2 delivery and reached maximal values within approximately 5 min, but dropped before exhaustion (P < 0.05) despite increasing or stable central venous and mean arterial pressures. In both types of maximal cycling, the impaired systemic O2 delivery was due to the decline or plateau in because arterial O2 content continued to increase. These results indicate that an inability of the circulatory system to sustain a linear increase in O2 delivery to the locomotor muscles restrains aerobic power. The similar impairment in SV and O2 delivery during incremental and constant load cycling provides evidence for a central limitation to aerobic power and capacity in humans.
Profiles of muscle-specific oxygenation responses and thresholds during graded cycling incremental test

Abstract:Compared to the determination of exercise thresholds based on systemic changes in blood lactate concentrations or gas exchange data, the determination of breakpoints based on muscle oxygen saturation offers a valid alternative to provide specific information on muscle-derived thresholds. Our study explored the profiles and timing of the second muscle oxygenation threshold (MOT2) in different muscles. Twenty-six cyclists and triathletes (15 male: age = 23 ± 7 years, height = 178 ± 5 cm, body mass = 70.2 ± 5.3 kg; 11 female: age = 22 ± 4 years, height = 164 ± 4 cm, body mass = 58.3 ± 8.1 kg) performed a graded exercise test (GXT), on a cycle ergometer. Power output, blood lactate concentration, heart rate, rating of perceived exertion, skinfolds and muscle oxygen saturation were registered in five muscles (vastus lateralis, biceps femoris, gastrocnemius medialis, tibialis anterior and triceps brachii) and percentage at which MOT2 occurred for each muscle was determinated using the Exponential Dmax. The results of Statistical Parametric Mapping and ANOVA showed that, although muscle oxygenation displayed different profiles in each muscle during a GXT, MOT2 occurred at a similar percentage of the GXT in each muscle (77% biceps femoris, 75% tibalis anterior, 76% gastrocnemius medialis and 72% vastus lateralis) and it was similar that systemic threshold (73% of the GXT). In conclusion, this study showed different profiles of muscle oxygen saturation in different muscles, but without notable differences in the timing for MOT2 and concordance with systemic threshold. Finally, we suggest the analysis of the whole signal and not to simplify it to a breakpoint.
NFBD1/MDC1 regulates ionizing radiation‐induced focus formation by DNA checkpoint signaling and repair factors

Abstract:NFBD1/MDC1 (mediator of DNA damage checkpoint 1) is a nuclear factor with an aminoterminal FHA (forkhead‐associated) domain and a tandem repeat of BRCT (breast cancer susceptibility gene‐1 carboxyl terminus) domains. We have previously shown that NFBD1 is an early participant in DNA damage signaling pathways and that ionizing radiation‐induced nuclear foci (IRIF) of NFBD1 colocalize with several DNA checkpoint signaling and repair factors. We report here that NFBD1 physically associates with ATM, p53, components of the MRE11‐RAD50‐NBS1 (MRN) complex, and γ‐H2AX. An overexpressed FHA domain‐containing fragment of NFBD1 binds to endogenous NFBD1 and components of the MRN complex, but not to γ‐H2AX. This fragment interferes with IRIF formation by endogenous NFBD1, MRE11, or NBS1. A BRCT domain‐containing fragment of NFBD1 binds to γ‐H2AX and 53BP1, but not to components of the MRN complex, and abolishes IRIF formation by NFBD1, MRE11, NBS1, 53BP1, CHK2 phospho‐T68, γ‐H2AX, and possible ATM/ATR substrates recognized by anti‐phospho‐SQ/TQ antibody. These results suggest that NFBD1 is an ATM/ATR‐dependent organizer that recruits DNA checkpoint signaling and repair proteins to the sites of DNA damage.—Xu, X., Stern, D. F. Abstract:It has been suggested that time at a high fraction (%) of maximal oxygen uptake (VO2max) plays a decisive role for adaptations to interval training. Yet, no study has, to date, measured the % of VO2max during all interval sessions throughout a prolonged training intervention and subsequently related it to the magnitude of training adaptations. Thus, the present study aimed to investigate the relationship between % of VO2max achieved during an interval training intervention and changes in endurance performance and its physiological determinants in well‐trained cyclists. Twenty‐two cyclists (VO2max 67.1 (6.4) mL·min−1 ·kg−1; males, n = 19; females, n = 3) underwent a 9‐week interval training intervention, consisting 21 sessions of 5 × 8‐min intervals conducted at their 40‐min highest sustainable mean power output (PO). Oxygen uptake was measured during all interval sessions, and the relationship between % of VO2max during work intervals and training adaptations were investigated using linear regression. A performance index was calculated from several performance measures. With higher % of VO2max during work intervals, greater improvements were observed for maximal PO during the VO2max test (R2adjusted = 0.44, p = 0.009), PO at 4 mmol·L−1 [blood lactate] (R2adjusted = 0.25, p = 0.035), the performance index (R2adjusted = 0.36, p = 0.013), and VO2max (R2adjusted = 0.54, p = 0.029). Other measures, such as % of maximal heart rate, were related to fewer outcome variables and exhibited poorer session‐to‐session repeatability compared to % of VO2max. In conclusion, improvements in endurance measures were positively related to the % of VO2max achieved during interval training. Percentage of VO2max was the measure that best reflected the magnitude of training adaptations.

Abstract:ABSTRACT Purpose This study aimed to determine physiological and metabolic responses to two different sprint interval exercises (SIE) matched for total sprint duration and sprint-to-rest ratio. Methods After having measured peak oxygen uptake (V̇O 2peak ), fourteen healthy males (27.1 ± 4.8 years, 169.6 ± 6.0 cm, 64.5 ± 8.4 kg, V̇O 2peak : 47.2 ± 7.7 ml·kg -1 ·min -1 ) performed four 10-s sprints with 80-s recovery (SIE10) and two 20-s sprints with 160-s recovery (SIE20) on different occasions in a counterbalanced crossover manner. Pulmonary V̇O 2 and changes in tissue oxygenation index (∆TOI) at vastus lateralis (VL) and rectus femoris (RF) were measured during the SIEs. Furthermore, T2-weighted MRI was taken immediately before and after the SIEs to determine the activation levels of VL, RF, vastus medialis, vastus intermedius, adductor magnus, biceps femoris long head, semitendinosus and semimembranosus at 50% of right thigh length. Results In SIE10, increases in V̇O 2 and ∆TOI at VL and RF plateaued after the second sprint, whereas session-averaged ∆TOI was greater in SIE20 than SIE10 in both muscles (VL: 20.9 ± 7.4 vs. 14.2 ± 5.9%, RF: 22.8 ± 9.3 vs. 12.9 ± 6.6%, p = 0.00). While both SIEs significantly increased T2 values in all eight muscles, those magnitudes were similar between the conditions (SIE10 vs. SIE20: 5-16 vs. 8-16%). Conclusions This study showed blunted responses of whole-body (V̇O 2 ) and peripheral (∆TOI) oxidative responses with successive sprints (sprint 1 < sprint 2-4) in SIE10, suggesting that increasing sprint repetitions does not necessarily induce greater oxidative metabolism or stimulus. Moreover, greater peripheral oxygen extraction (∆TOI) was achieved with SIE20, whereas %changes of T2 indicates that the thigh muscles were similarly activated between the SIE conditions.
