Abstract:Prosthesis implantation and fat transplantation are common breast reconstruction methods. In general, prosthesis implantation alone does not achieve a realistic enough appearance, and fat transplantation alone is difficult to achieve in the correct capacity. To date, no reports have focused on methods of combining fat with implanted prostheses for breast reconstruction. Using a newly designed bionic ink (i.e., polyether F127 diacrylate (F127DA) poly(ethylene glycol) diacrylate (PEGDA)) and projection-based three-dimensional bioprinting (PBP), we report the development of a new method for printing porous prostheses. PEGDA was used to improve the printing precision of the prosthesis by increasing the gel point of F127DA and reducing the impact of external temperature. The compression modulus of the printed prosthesis was very close to that of prostheses currently used in clinical practice and to that of natural breasts. Finally, stromal vascular fraction gel (SVF-gel), a human fat extract, was injected into the pores of the synthesized prostheses to prepare a prosthesis mixed with adipose tissue. These were implanted subcutaneously in nude mice to observe their biological performance. After 14 and 28 days of observation, the prosthesis showed good biocompatibility, and adipose tissues grew well in and around the prosthesis. This result shows that a porous prosthesis containing pre-placed adipose tissues is a promising breast reconstruction material.

Abstract:Projection-based 3D bioprinting(PBP):a powerful method to fabricate 3D cellular structures Three-dimensional(3D)bioprinting has played an impor-tant role in tissue engineering and regenerative medicine areas over the past decade[1].Different from traditional cell cultures in Petri dishes,3D bioprinting can build bionic structures with a better potential to become artificial organ substitutes[2-4].With the development of photocurable bio-materials,the projection-based 3D printing method has been successfully applied in biological research[5,6].It pos-sesses significant advantages in precision and speed due to its unique manufacturing principle.On the one hand,light-dependent solidification allows it to build models with resolutions around 50 μm(theoretically a minimum of 1 μm)[7].On the other hand,its surface-to-entity printing process provides duplicates in batches without extra time require-ment[8].Therefore,it has attracted the attention of many researches devoted to the field of 3D cell culture.
Microfabrication of 3D-Polymeric Structures Using Soft Lithography for Tissue Engineering

Abstract:Simple methods for fabricating 3D biodegradable, biocompatible micro polymeric structures with well-defined geometry for tissue engineering have been developed using the soft lithography, which utilized an elastomeric poly-dimethylsiloxane (PDMS) mold. A high-resolution transparency mask was drawn with CorelDraw software and printed on a plastic slide using a high-resolution line printer. The copolymers of poly-lactic-co-glycolic acid (PLGA 85/15) were pretreated with moderate stiffness and flexibility. The PLGA scaffolds with feature sizes of approximately 10~100 μm were fabricated using two molding methods: polymer casting and spin coating. The factors that influenced the integrity and the lateral resolution of scaffolds are discussed and optimized. The two different methods have been compared in terms of lateral and vertical resolution. As a proof-of-concept for scaffold utility in tissue engineering applications, multilayer structures for cell culture were formed by thermal lamination. In conclusion, the soft-lithographic technique provides a useful tool to realize microscale 3D structures with little islands of polymers for tissue engineering.
Digital Light Processing-Based 3D Printing of Cell-Seeding Hydrogel Scaffolds with Regionally Varied Stiffness

Abstract:Cell-seeding heterogeneous scaffolds with regionally varied stiffness play an important role in tissue engineering, e.g., bone and cartilage regeneration, that require the recapitulation of geometric complexity through biocompatible material to mimic the natural cell microenvironment in vivo. Here, we report the digital light processing (DLP)-based 3D printing of cell-seeding hydrogel scaffold with regionally varied stiffness by tuning the exposure time without changing the geometric architecture. Mechanical tests on printed poly(ethylene glycol) diacrylate (PEGDA) hydrogels homogeneous scaffold revealed that a 60% increase in the elastic modulus can be achieved by setting the optimal exposure time. Furthermore, regulating the stiffness by varying the exposure time was demonstrated in the printed three-sectional heterogeneous scaffolds. Uniaxial compression tests showed that no fracture was observed even when the compression strain reached up to 25%, indicating that by adjusting the exposure time, the undesired influence of the scaffold on mechanical integrity could be avoided. 3T3 fibroblasts were then seeded onto the scaffold, and the biocompatibility together with the physical support of the scaffolds were confirmed by observation and cell population assessment.

Abstract:Synthetic polymers possess more reproducible physical and chemical properties than their naturally occurring counterparts. They have also emerged as an important alternative for fabricating tissue substitutes because they can be molecularly tailored to have vast array of molecular weights, block structures, active functional groups, and mechanical properties. To this date however, there has been very few successful and fully functional synthetic tissue and organ substitutes and with the rapidly spreading 3D printing technology beginning to reshape the tissue engineering and regenerative field, the need for an effective, safe, and bio printable biomaterial is becoming more and more urgent. Here, we have developed a synthetic polymer from controlled living radical polymerisation that can be printed into well-defined structures. The polymer showed low cytotoxicity before and after printing. Additionally, the incorporation of gelatine-methacrylate coated PLGA microparticles within the hydrogel provided cell adhesion surfaces for cell proliferation. The results point to possible application of the microparticle seeded, synthetic hydrogel as a direct printable tissue or organ substitute.

Abstract:A facile technique is herein reported to fabricate three-dimensional (3D) polymeric porous scaffolds with interior surfaces of a topographic microstructure favorable for cell adhesion. As demonstration, a well-known biodegradable polymer poly(lactide-co-glycolide) (PLGA) was employed as matrix. Under the porogen-leaching strategy, the large and soft porogens of paraffin were modified by colliding with small and hard salt particles, which generated micropits on the surfaces of paraffin spheres. The eventual PLGA scaffolds after leaching the modified porogens had thus interior surfaces of microscale roughness imprinted by those micropits. The microrough scaffolds were confirmed to benefit adhesion of bone marrow stromal cells (BMSCs) of rats and meanwhile not to hamper the proliferation and osteogenic differentiation of the cells. The insight and technique might be helpful for biomaterial designing in tissue engineering and regenerative medicine.
Direct 3D Printed Biomimetic Scaffolds Based on Hydrogel Microparticles for Cell Spheroid Growth

Abstract:Biocompatible hydrogel inks with shear‐thinning, appropriate yield strength, and fast self‐healing are desired for 3D bioprinting. However, the lack of ideal 3D bioprinting inks with outstanding printability and high structural fidelity, as well as cell‐compatibility, has hindered the progress of extrusion‐based 3D bioprinting for tissue engineering. In this study, novel self‐healable pre‐cross‐linked hydrogel microparticles (pcHμPs) of chitosan methacrylate (CHMA) and polyvinyl alcohol (PVA) hybrid hydrogels are developed and used as bioinks for extrusion‐based 3D printing of scaffolds with high fidelity and biocompatibility. The pcHμPs display excellent shear thinning when injected through a syringe and subsequently self‐heal into gels as shear forces are removed. Numerical simulations indicate that the pcHμPs experience a plug flow in the nozzle with minimal disturbance, which favors a steady and continuous printing. Moreover, the pcHμPs show a self‐supportive yield strength (540 Pa), which is critical for the fidelity of printed constructs. A series of biomimetic constructs with very high aspect ratio and delicate fine structures are directly printed by using the pcHμP ink. The 3D printed scaffolds support the growth of bone‐marrow‐derived mesenchymal stem cells and formation of cell spheroids, which are most important for tissue engineering.

Reducing Branded Prescription Drug Prices: A Review of Policy Options

Abstract:The high prices of specialty pharmaceuticals are causing some public programs to ration care and many private insurers, including Medicare drug plans, to place specialty drugs on high cost‐sharing tiers. As a result, access to these drugs is often restricted, and only a small portion of the population with a disease may receive treatment. This concern has generated a wide range of proposed solutions. We conducted a literature review and identified 52 solutions in the peer‐reviewed literature that we classified into five broad categories: revising the patent system, encouraging research to increase development of new drugs, altering pharmaceutical regulation, decreasing market demand, and developing innovative pricing strategies. We discuss the rationale for these five approaches and summarize the proposed solutions. We also discuss four empirical issues that are particularly important in any discussion of policy options.
Permeability and mechanical properties of gradient porous PDMS scaffolds fabricated by 3D-printed sacrificial templates designed with minimal surfaces

Abstract:In the present study, polydimethylsiloxane (PDMS) porous scaffolds are designed based on minimal surface architectures and fabricated through a low-cost and accessible sacrificial mold printing approach using a fused deposition modeling (FDM) 3D printer. The effects of pore characteristics on compressive properties and fluid permeability are studied. The results suggest that radially gradient pore distribution (as a potential way to enhance mechanically-efficient scaffolds with enhanced cell/scaffold integration) has higher elastic modulus and fluid permeability compared to their uniform porosity counterparts. Also, the scaffolds are fairly strain-reversible under repeated loading of up to 40% strain. Among different triply periodic minimal surface pore architectures, P-surface was observed to be stiffer, less permeable and have lower densification strain compared to the D-surface and G-surface-based pore shapes. The biocompatibility of the created scaffolds is assessed by filling the PDMS scaffolds using mouse embryonic fibroblasts with cell-laden gelatin methacryloyl which was cross-linked in situ by UV light. Cell viability is found to be over 90% after 4 days in 3D culture. This method allows for effectively fabricating biocompatible porous organ-shaped scaffolds with detailed pore features which can potentially tailor tissue regenerative applications. STATEMENT OF SIGNIFICANCE: Printing polymers with chemical curing mechanism required for materials such as PDMS is challenging and impossible to create high-resolution uniformly cured structures due to hard control on the base polymer and curing process. An interconnected porous mold with ordered internal architecture with complex geometries were 3D printed using low-cost and accessible FDM technology. The mold acted as a 3D sacrificial material to form internally architected flexible PDMS scaffolds for tissue engineering applications. The scaffolds are mechanically stable under high strain cyclic loads and provide enough pore and space for viably integrating cells within the gradient architecture in a controllable manner.

Abstract:Three dimension (3D) printing has been increasingly used to make tissue engineering scaffolds. However, polyester-based scaffolds made through traditional 3D printing such as fused deposition modeling (FDM) lack of bioactivity as biomolecules cannot be directly blended with inks to achieve an in situ delivery. In addition, with increasing demand of reducing surgical trauma, implanting scaffolds through a minimally invasive way is of great importance. Here, surface porous tissue engineering scaffolds were made from novel poly(D,L-lactic acid-co-trimethylene carbonate)-based water-in-oil emulsions through a cryogenic 3D plotting. The effects of water content in emulsions on the microstructure of 3D plotted scaffolds were investigated. With heat treatment at 40°C, scaffolds became soft and can be injected into a catheter. Growth factors can be loaded into the scaffolds in situ and showed a sustained release within 30 days.

Abstract:Tissue engineering has emerged as an advanced strategy to regenerate various tissues using different raw materials, and thus it is desired to develop more approaches to fabricate tissue engineering scaffolds to fit specific yet very useful raw materials such as biodegradable aliphatic polyester like poly(lactide-co-glycolide) (PLGA). Herein, a technique of "wet 3D printing" was developed based on a pneumatic extrusion 3D printer after we introduced a solidification bath into a 3D printing system to fabricate porous scaffolds. The room-temperature deposition modeling of polymeric solutions enabled by our wet 3D printing method is particularly meaningful for aliphatic polyester, which otherwise degrades at high temperature in classic fuse deposition modeling. As demonstration, we fabricated a bilayered porous scaffold consisted of PLGA and its mixture with hydroxyapatite (HAp) for regeneration of articular cartilage and subchondral bone. Long-term in vitro and in vivo degradation tests of the scaffolds were carried out up to 36 weeks, which support the three-stage degradation process of the polyester porous scaffold and suggest faster degradation in vivo than in vitro. Animal experiments in a rabbit model of articular cartilage injury were conducted. The efficacy of the scaffolds in cartilage regeneration was verified through histological analysis, micro-CT and biomechanical tests, and the influence of scaffold structures (bilayer versus single layer) on in vivo tissue regeneration was examined. This study has illustrated that the wet 3D printing is an alternative approach to biofabricate tissue engineering porous scaffolds based on biodegradable polymers.

Abstract:Preparation of biological scaffolds with complex shapes and controllable internal structures is a significant development direction for the tissue engineering field. Due to its simplicity, low-cost and cost-effectiveness, fused deposition molding (FDM) is one of the most popular 3D printing technologies. In this paper, polyvinyl alcohol/polylactic acid/hydroxyapatite composites were fabricated successfully. On this basis, composite scaffolds with different pore structures were designed and constructed by using FDM technology. The material structure was characterized by scanning electron microscopy (SEM) and dynamic thermomechanical analyzer. The results showed that the cross-sectional morphology of the system presented a "sea-island" structure. The introduction of PLA and HA significantly increased the modulus of the material, which was beneficial to achieve its FDM processing. The compressibility, mineralization behavior, and biocompatibility of the viscoelastic characteristics to natural cartilage subjected to the condition of filling angle of 45° was systemically invesitgated. Notably, the concentrations of Ca2+ and PO43− in simulated body fluid were reduced by 34.7% and 58.2% on 8th day, indicating a good bone formation and mineralization ability, which was further corroborated by SEM and fourier-transform infrared spectroscopy (FTIR) results. Biotoxicity test showed that the scaffold presented excellent biocompatibility.

Abstract:Tissue engineering is expected to construct complicated hominine organs composed of many different types of cells. One of the key points is the accurate controlling of scaffold material and porous morphology point by point. A new direct rapid prototyping process called low-temperature deposition manufacturing (LDM) was proposed to fabricate scaffolds. The new process integrated extrusion/jetting and phase separation and therefore could fabricate scaffolds with hierarchical porous structures creating a wonderful environment for the growth of new tissue. The interconnected computer-designed macropores allow cells in the new tissue to grow throughout the scaffold. Also, the parameter-controlled micropores let nutrition in and metabolic wastes out. The macrocellular morphology, microcellular morphology, porosity, and mechanical properties of the poly(alpha-hydroxy acid)-TCP composite scaffolds prepared by the proposed method are investigated. These scaffolds with high controllability would potentially play an important role in tissue engineering. LDM could also be combined with multinozzle deposition or cell deposition to exactly control materials or cells point by point. A Novel Poly (Lactic-Co-Glycolic Acid)-Collagen Hybrid Scaffold Fabricated Via Multi-Nozzle Low-Temperature Deposition

Abstract:Scaffolds made from biodegradable materials play an important role in tissue engineering as a temporary support for cellular survival. An ideal scaffold for tissue engineering applications must demonstrate adequate strength and good properties to sustain cell adhesion, proliferation and differentiation. We constructed a PLGA-collagen hybrid scaffold with interconnected macro-channels via a Rapid Prototyping (RP) based Multi-nozzle Low-temperature Deposition and Manufacturing (M-LDM) system. Preliminary results show that the scaffold possesses interconnected micro-pores and macro-pores; the collagen sponge is strongly absorbed to PLGA matrices and a stable mesh is formed; the hybrid scaffold possesses stronger mechanical properties compared to collagen sponge and better hydrophilicity compared to PLGA scaffold; and this hybrid scaffold would be potentially used in bone and cartilage regeneration.

Abstract:A novel method for the fabrication of complexly shaped three-dimensional porous scaffolds has been developed by combining modified compression molding and conventional particulate leaching. The resultant scaffolds of various shapes, including some shaped like auricles, were made of hydrophobic biodegradable and bioresorbable poly(D,L-lactic acid) (PDLLA) and poly(D,L-lactic-co-glycolic acid) (PLGA). A polymer-particulate mixture was first prepared by the conventional solvent casting method and then compressively molded in a specially designed flexible-rigid combined mold which facilitates shaping and mold release during the fabrication process. The molding was carried out at a moderate temperature, above the glass transition temperature and below the flow temperature of these amorphous polymers. A porous scaffold was then obtained after particulate leaching. The pores are highly interconnected and uniformly distributed both in the bulk and on the external surface of the scaffolds, and the porosity can exceed 90%. The mechanical properties of the resultant porous scaffolds are satisfactory as determined by measurements of compressive modulus and compressive stress at 10% strain. Good viability of cells seeded in the porous scaffolds was confirmed. This novel fabrication method is promising in tissue engineering because of its ability to produce precise and complexly (anatomically) shaped porous scaffolds.

Abstract:Some basic requirements of bone tissue engineering include cells derived from bone tissues, three-dimensional (3D) scaffold materials, and osteogenic factors. In this framework, the critical architecture of the scaffolds plays a crucial role to support and assist the adhesion of the cells, and the subsequent tissue repairs. However, numerous traditional methods suffer from certain drawbacks, such as multi-step preparation, poor reproducibility, high complexity, difficulty in controlling the porous architectures, the shape of the scaffolds, and the existence of solvent residue, which limits their applicability. In this work, we fabricated innovative poly(lactic-co-glycolic acid) (PLGA) porous scaffolds, using 3D-printing technology, to overcome the shortcomings of traditional approaches. In addition, the printing parameters were critically optimized for obtaining scaffolds with normal morphology, appropriate porous architectures, and sufficient mechanical properties, for the accommodation of the bone cells. Various evaluation studies, including the exploration of mechanical properties (compressive strength and yield stress) for different thicknesses, and change of structure (printing angle) and porosity, were performed. Particularly, the degradation rate of the 3D scaffolds, printed in the optimized conditions, in the presence of hydrolytic, as well as enzymatic conditions were investigated. Their assessments were evaluated using the thermal gravimetric analyzer (TGA), differential scanning calorimetry (DSC), and gel permeation chromatography (GPC). These porous scaffolds, with their biocompatibility, biodegradation ability, and mechanical properties, have enabled the embryonic osteoblast precursor cells (MC3T3-E1), to adhere and proliferate in the porous architectures, with increasing time. The generation of highly porous 3D scaffolds, based on 3D printing technology, and their critical evaluation, through various investigations, may undoubtedly provide a reference for further investigations and guide critical optimization of scaffold fabrication, for tissue regeneration.

Abstract:A microfluidics system is used to create gelatin methacryloyl (GelMA) nanofibrous microspheres (GNM), which are employed as bioink for 3D printing hierarchically porous GNM scaffolds (HP‐GNMS). Bone marrow stromal cells (BMSCs) are seeded onto these scaffolds and implanted in a rat cranial defect model, promoting cell infiltration, and guiding osteogenic differentiation and extracellular matrix deposition. Current limitations in 3D printing pose significant challenges for the fabrication of hierarchical 3D scaffolds with nanofibrous structures that simulate the natural bone extracellular matrix (ECM) for enhanced bone regeneration. This study presents an innovative approach to 3D printing customized hierarchical porous scaffolds with nanofiber structures using biodegradable nanofibrous microspheres as the bio‐ink. In vitro investigations demonstrate that the hierarchical porous architecture substantially enhances cell infiltration and proliferation rates, while the nanofiber topology provides physical cues to guide osteogenic differentiation and ECM deposition. When serving as a cell carrier, the 3D‐printed nanofibrous scaffold promotes bone tissue regeneration and integration in vivo. Additionally, the facile and versatile chemical modification facilitates the precise tailoring of the scaffold's functionality. Using nanofibrous microspheres with highly biomimetic and versatile modification properties as the foundational constituent in this universal 3D printing methodology enables comprehensive manipulation of scaffolding biological properties, spanning from macroscopic external morphology to molecular‐scale biochemical kinetics, thereby addressing a diverse spectrum of clinical requisites.

Abstract:3D printing is a promising technique for producing bone implants, but there is still a need to adjust efficiency, facilitate production, and improve biocompatibility. Porous materials have a proven positive effect on the regeneration of bone tissue, but their production is associated with numerous limitations. In this work, we described a simple method of producing polymer or polymer-ceramic filaments for 3D-printing scaffolds by adding micrometer-scale porous structures on scaffold surfaces. Scaffolds included polycaprolactone (PCL) as the primary polymer, β-tricalcium phosphate (β-TCP) as the ceramic filler, and poly(ethylene glycol) (PEG) as a porogen. The pressurized filament extrusion gave flexible filaments composed of PCL, β-TCP, and PEG, which are ready to use in fused filament fabrication (FFF) 3D printers. Washing of 3D-printed scaffolds in ethanol solution removed PEG and revealed a microporous structure and ceramic particles on the scaffold's surfaces. Furthermore, 3D-printed materials exhibit good printing precision, no cytotoxic properties, and highly impact MG63 cell alignment. Although combining PCL, PEG, and β-TCP is quite popular, the presented method allows the production of porous scaffolds with a well-organized structure without advanced equipment, and the produced filaments can be used to 3D print scaffolds on a simple commercially available 3D printer.

Abstract:A mouse monoclonal antibody (mAb), designated as mNI-11, has been produced. This mAb strongly induced homotypic cell aggregation (HCA) of monocyte-like cell line U937 cultured with conditioned medium derived from PHA-stimulated peripheral blood mononuclear cells and cytochalasin B. HCA occurred at an optimal temperature of 37 C but not 4 C. mAbs to CD18 (lymphocyte function-associated antigen-1 beta; LAF-1 beta) and CD54 (intercellular adhesion molecule-1; ICAM-1) completely inhibited this HCA. These results suggest that mNI-11 strongly induces HCA of the U937 cells in an activated form after stimulation with conditioned medium and cytochalasin B.

Abstract:This paper reports polymeric scaffolds with spherical internal macropores and relatively large external dimension. Paraffin spheres with the diameter of several hundred microns were prepared by a suspension technique. Particulate leaching technique based on this kind of spherical porogens was combined with room-temperature compression molding technique to fabricate biodegradable poly(D,L-lactic- co -glycolic acid) (PLGA) porous scaffolds potentially for tissue engineering or in situ tissue induction. The scaffolds exhibited ordered macropores with good pore interconnectivity. The porosity ranged from 80 to 97% adjusted simply by varying porogen content. The foams with porosity around 90% have compressive modulus over 3 MPa and compressive strength over 0.2 MPa. As preliminary cell experiments with 3T3 fibroblasts cultured on the porous scaffolds indicate, the processing procedure of the scaffolds has not brought with problem in cytotoxicity.
