Abstract:In this article, we present a CMOS-based broadband nuclear magnetic resonance (NMR) magnetometer system with magnetic field searching and automatic tuning capabilities. The NMR front-end consists of a custom-designed NMR transceiver application-specific integrated circuit (ASIC) and an external LC resonance circuit formed by an NMR coil and a digitally tunable capacitor (DTC). With a 1-mm NMR coil, the system is designed to measure the magnetic fields between 0.54 and 2.54 T with nanotesla resolution. To this end, the system automatically sweeps its excitation frequency to acquire an NMR signal from a known sample inside the NMR coil. The measured frequency of the NMR signal, together with the known Larmor frequency of the sample, can then be used to determine the unknown magnetic field with very high precision. The presented system provides an optimum sensitivity over its entire frequency range by automatically tuning the LC resonant circuit to the current Larmor frequency using the DTC. With a 0.78-mu L sample of vegetable oil, the presented NMR magnetometer achieves an excellent field resolution of 1.77 nT inside a 1.45-T benchtop MR magnet, corresponding to a relative resolution of 1.2 ppb, which is limited by the field stability of the magnet. With its intrinsic frequency noise of 15 mHz, measured using a synthetic input signal, the presented sensor can support magnetic field resolutions down to 242 pT within a 1.45-T base field.

Abstract:We developed a low field NMR system, which consisting of three parts based on High Tc DC-SQUID. First, the parameters and the relative location of the coils were designed on the basis of thoretical calculation. We made the prepolarization field reached 10mT and the measurement field changed between 200 μT and 1 μT. Second, based on the LabVIEW platform, we designed a programm, by which automatic measurement and data acquisition were achieved perfectly. At last, we fulfilled the test of the SQUID and the system electronics, and all the parts worked well during the process of the comprehensive test. Further experiment and analysis on the system were in process.

Abstract:Accurate measuring magnetic induction intensity is important for some scientific research. In the paper, the conventional nuclear magnetic resonance (NMR) experiment under static magnetic field has been extended to longitudinal time-varying magnetic field (TVMF), and the time domain transient solution of the Bloch equations for spin nuclei exhibits the so-called non-steady state nuclear magnetic resonance (NSS-NMR) phenomenon. Based on the above effect and the precise gyromagnetic ratio of spin nucleus, a new magnetic field measurement method and corresponding experimental apparatus have been developed in longitudinal TVMF environment. The orthogonal dual-coil probe, radio frequency (RF) exciting and receiving hardware, key signal processing algorithms, host computer controlling and analysis software are deeply studied. In order to improve the signal-to-noise ratio of the NSS-NMR signal, the RF signal envelope shaping and the low-frequency pre-emphasis probe matching techniques are designed and verified by the invented circuit-field-nuclear multi-domain joint simulation subassembly (CFN-MDJS) which is integrated in the host computer software. Furthermore, the multi-tone RF excitation signal substituting the bandpass continuous spectrum signal for NSS-NMR experiment is also verified by the CFN-MDJS. The experimental results under longitudinal 50Hz TVMF environment has demonstrated the authentic existence of the NSS-NMR effect and simultaneously verified the effectiveness of the experimental scheme and signal processing algorithms proposed in the paper.

Abstract:Nuclear magnetic resonance (NMR) spectroscopy is a powerful high-resolution tool for characterizing biomacromolecular structure, dynamics, and interactions. However, the lengthy longitudinal relaxation of the nuclear spins significantly extends the total experimental time, especially at high and ultra-high magnetic field strengths. Although longitudinal relaxation-enhanced techniques have sped up data acquisition, their application has been limited by the chemical shift dispersion. Here we combined an evolutionary algorithm and artificial intelligence to design 1H and 15N radio frequency (RF) pulses with variable phase and amplitude that cover significantly broader bandwidths and allow for rapid data acquisition. We re-engineered the basic transverse relaxation optimized spectroscopy experiment and showed that the RF shapes enhance the spectral sensitivity of well-folded proteins up to 180 kDa molecular weight. These RF shapes can be tailored to re-design triple-resonance experiments for accelerating NMR spectroscopy of biomacromolecules at high fields.

Abstract:In magnetic resonance imaging (MRI), the spectrometer is a fundamental component of MRI systems, responsible for the transmission of radiofrequency (RF) pulses that excite hydrogen nuclei and the subsequent acquisition of MR signals for processing. However, detailed knowledge about this component remains largely inaccessible due to the proprietary nature of commercial systems. To address this gap, we present an FPGA-based platform specifically designed for MRI signal transmission and reception in low-field MRI applications. Additionally, with appropriate chip replacements, this platform can be adapted for use in mid- and high-field MRI systems. This platform leverages Direct Digital Synthesis (DDS) technology to generate RF pulses, offering the flexibility to quickly and precisely adjust soft pulse parameters to meet the specific requirements of the MRI system. Additionally, the platform processes MRI signals through digital downconversion techniques and utilizes CIC and FIR filters to obtain baseband signals. Experimental testing of this platform has yielded promising results. We hope that this work will inspire further research and development in the field of MRI spectrometer design. Furthermore, it is worth noting that with the replacement of relevant chips, this system can also be adapted for use in mid- and high-field MRI systems.

Abstract:Object: We studied the feasibility of generating the variable magnetic fields required for ultra-low field nuclear magnetic resonance relaxometry with dynamically adjustable permanent magnets. Our motivation was to substitute traditional electromagnets by distributed permanent magnets, increasing system portability. Materials and methods: The finite element method (COMSOL®) was employed for the numerical study of a small permanent magnet array to calculate achievable magnetic field strength, homogeneity, switching time and magnetic forces. A manually operated prototype was simulated and constructed to validate the numerical approach and to verify the generated magnetic field. Results: A concentric small permanent magnet array can be used to generate strong sample pre-polarisation and variable measurement fields for ultra-low field relaxometry via simple prescribed magnet rotations. Using the array, it is possible to achieve a pre-polarisation field strength above 100 mT and variable measurement fields ranging from 20-50 μT with 200 ppm absolute field homogeneity within a field-of-view of 5 x 5 x 5 cubic centimetres. Conclusions: A dynamic small permanent magnet array can generate multiple highly homogeneous magnetic fields required in ultra-low field nuclear magnetic resonance (NMR) and magnetic resonance imaging (MRI) instruments. This design can significantly reduce the volume and energy requirements of traditional systems based on electromagnets, improving portability considerably.

Abstract:The field of RNA interference depends on the development of safe and efficient carriers for short interfering ribonucleic acid (siRNA) delivery. Conventional cationic monomers for siRNA delivery have utilized the nitrogen heteroatom to produce cationic charges. Here, we polymerized cationic sulfonium (meth)acrylate by activators regenerated by electron transfer (ARGET) atom transfer radical polymerization (ATRP) to form polymers with narrow molecular weight distributions for siRNA delivery. The tertiary sulfonium species was stable toward dealkylation in water but less stable in the polar aprotic solvent dimethyl sulfoxide. Block copolymers poly(ethylene oxide) with poly(meth)acrylate containing sulfonium moieties were prepared as an siRNA delivery platform. Results suggested block copolymers were biocompatible up to 50 μg/mL in vitro and formed polyplexes with siRNA. Additionally, block copolymers protected siRNAs against endonuclease digestion and facilitated knockdown of glyceraldehyde 3-phosphate dehydrogenase (Gapdh) mRNA expression in murine calvarial preosteoblasts. The versatility, biocompatibility, and cationic nature of these tertiary sulfonium groups are expected to find widespread biological applications.

Abstract:High temperature superconducting (HTS) ceramic materials provide magnets with liquid cryogen‐free working environment, no need of oxygen sensors and exhaust ventilation system, reducing the magnet footprint compared to low temperature superconducting (LTS) magnets, reducing long‐term maintenance costs, and enabling installation at smaller facilities. In this mini review, we describe the work successfully done with the prototype 400‐MHz HTS cryogen‐free spectrometer, installed in the fume hood of a chemistry laboratory, for structure elucidation, reaction monitoring, and computer‐assisted 3D structure elucidation (CASE 3D). The discovery of new ceramic materials containing Ba‐La‐Cu oxides in 1986 that exhibited superconducting properties at high temperatures in the range of 35 K or higher, recognized with the Nobel Prize in Physics in 1987, opened a new world of opportunities for nuclear magnetic resonance (NMRs) and magnetic resonance imaging (MRIs) to move away from liquid cryogens. This discovery expands the application of high temperature superconducting (HTS) materials to fields beyond the chemical and medical industries, including electrical power grids, energy, and aerospace. The prototype 400‐MHz cryofree HTS NMR spectrometer installed at Amgen's chemistry laboratory has been vital for a variety of applications such as structure analysis, reaction monitoring, and CASE‐3D studies with RDCs. The spectrometer has been integrated with Amgen's chemistry and analytical workflows, providing pipeline project support in tandem with other Kinetic Analysis Platform technologies. The 400‐MHz cryofree HTS NMR spectrometer, as the name implies, does not require liquid cryogens refills and has smaller footprint that facilitates installation into a chemistry laboratory fume hood, sharing the hood with a process chemistry reactor. Our evaluation of its performance for structural analysis with CASE‐3D protocol and for reaction monitoring of Amgen's pipeline chemistry was successful. We envision that the HTS magnets would become part of the standard NMR and MRI spectrometers in the future. We believe that while the technology is being developed, there is room for all magnet options, including HTS, low temperature superconducting (LTS) magnets, and low field benchtop NMRs with permanent magnets, where utilization will be dependent on application type and costs.

Abstract:We describe here the design and construction of a modern, state-of-the-art nuclear magnetic resonance (NMR) field-cycling instrument. Fourier transform NMR spectra of both liquid and solid samples can be measured, and spin-lattice relaxation times (T1Z) investigated over a broad range of magnetic field strengths ranging from 0 to 2 T. The instrument is based upon an existing personal computer-based NMR spectrometer [C. Job, R. M. Pearson, and M. F. Brown, Rev. Sci. Instrum. 65, 3354 (1994)] which has been expanded into a fully computer-controlled field-cycling instrument. The magnetic field cycling is accomplished electronically by utilizing fast switching thyristors and a storage capacitor based on the Redfield energy storage concept. Unique aspects of the design include the field-cycling magnet, which can reach fields as high as 2 T; the personal computer-based NMR spectrometer and associated waveform electronics; and the use of a commercially available pulse width modulation switching current amplifier, having low internal power dissipation and a fast current settling time. Using this new technology T1Z relaxation times as short as 1 ms can be readily measured.

Abstract:State-of-the-art NMR spectrometers using superconducting magnets have enabled, with their ultrafine spectral resolution, the determination of the structure of large molecules such as proteins, which is one of the most profound applications of modern NMR spectroscopy. Many chemical and biotechnological applications, however, involve only small-to-medium size molecules, for which the ultrafine resolution of the bulky, expensive, and high-maintenance NMR spectrometers is not required. For these applications, there is a critical need for portable, affordable, and low-maintenance NMR spectrometers to enable in-field, on-demand, or online applications (e.g., quality control, chemical reaction monitoring) and co-use of NMR with other analytical methods (e.g., chromatography, electrophoresis). As a critical step toward NMR spectrometer miniaturization, small permanent magnets with high field homogeneity have been developed. In contrast, NMR spectrometer electronics capable of modern multidimensional spectroscopy have thus far remained bulky. Complementing the magnet miniaturization, here we integrate the NMR spectrometer electronics into 4-mm(2) silicon chips. Furthermore, we perform various multidimensional NMR spectroscopies by operating these spectrometer electronics chips together with a compact permanent magnet. This combination of the spectrometer-electronics-on-a-chip with a permanent magnet represents a useful step toward miniaturization of the overall NMR spectrometer into a portable platform.

Abstract:Nuclear magnetic resonance (NMR) spectroscopy is an important tool for studying the structures of molecules and materials. Mainly due to their high costs, the NMR spectrometers are often not available for teaching experiments. Teaching NMR experiments cannot be performed routinely and effectively also because that the students are not able to understand the principles underlying the operational steps without proper and adequate training. In this work, a virtual NMR spectrometer (VMRS1.0) for teaching experiments, which is based on numerical computational simulation technology and can be operated like a real spectrometer, is presented. With the VMRS1.0, the students can perform basic NMR experiments, including tuning, shimming, field lock, power adjustment for radio frequency, selecting samples, acquiring data, fast Fourier transform and spectrum processing (i.e., phase correction, integration, chemical shift correction, auto peak finding and J-coupling constant measurement). In addition to the routine one-dimensional NMR experiments, more complicated experiments such as decoupling, DEPT and HMQC can also be performed on the VMRS1.0. With the virtual NMR spectrometer developed, the students can practice their experimental skills and gain deeper understandings of the underlying principles in a more easy and cost-efficient way.

Near-resonant nuclear spin detection with high-frequency mechanical resonators

Abstract:Mechanical resonators operating in the high-frequency regime have become a versatile platform for fundamental and applied quantum research. Their exceptional properties, such as low mass and high quality factor, make them also very appealing for force sensing experiments. In this Letter, we propose a method for detecting and ultimately controlling nuclear spins by directly coupling them to high-frequency resonators via a magnetic field gradient. Dynamical backaction between the sensor and an ensemble of nuclear spins produces a shift in the sensor's resonance frequency, which can be measured to probe the spin ensemble. Based on analytical as well as numerical results, we predict that the method will allow nanoscale magnetic resonance imaging with a range of realistic devices. At the same time, this interaction paves the way for new manipulation techniques, similar to those employed in cavity optomechanics, enriching both the sensor's and the spin ensemble's features.

Abstract:Ultralow-field nuclear magnetic resonance (NMR) provides a new regime for many applications ranging from materials science to fundamental physics. However, the experimentally observed spectra show asymmetric amplitudes, differing greatly from those predicted by the standard theory. Its physical origin remains unclear, as well as how to suppress it. Here we provide a comprehensive model to explain the asymmetric spectral amplitudes, further observe more unprecedented asymmetric spectroscopy and find a way to eliminate it. Moreover, contrary to the traditional idea that asymmetric phenomena were considered as a nuisance, we show that more information can be gained from the asymmetric spectroscopy, e.g., the light shift of atomic vapors and the sign of Land$\acute{\textrm{e}}$ $g$ factor of NMR systems.

An Inexpensive, Configurable Two-Tone Electron Spin Resonance Spectrometer

Abstract:Electron spin resonance (ESR) is a powerful tool for characterizing and manipulating spin systems, but commercial ESR spectrometers can be expensive and designed to work in narrow frequency bands. This work presents an inexpensive spectrometer that, when coupled with easy-to-design resonators, enables ESR over a broad frequency range, including at frequencies outside the standard bands. It can operate at either a single frequency or at two frequencies simultaneously. The spectrometer is built from off-the-shelf parts and controlled by a field programmable gate array (FPGA), and new capabilities can be easily added by reconfiguring the FPGA and adding or swapping components. We demonstrate the capabilities of the spectrometer using the molecular nanomagnet Cr$_7$Mn.

Abstract:We present the architecture of the versatile NMR spectrometer with software-defined radio (SDR) technology and its application to the dynamically controlled pulsed magnetic fields. The pulse-field technology is the only solution to access magnetic fields greater than 50 T, but the NMR experiment in the pulsed magnetic field was difficult because of the continuously changing field strength. The dynamically controlled field pulse allows us to perform NMR experiment in a quasi-steady field condition by creating a constant magnetic field for a short time around the peak of the field pulse. We confirmed the reproducibility of the field pulses using the NMR spectroscopy as a high precision magnetometer. With the highly reproducible field strength we succeeded in measuring the nuclear spin-lattice relaxation rate $1/T_1$, which had never been measured by the pulse-field NMR experiment without dynamic field control. We also implement the NMR spectrum measurement with both the frequency-sweep and field-sweep modes and discuss the appropriate choice of these modes depending on the magnetic properties of sample to be measured. This development, with further improvement at a long-duration field pulse, will innovate the microscopic measurement in extremely high magnetic fields.

Abstract:Low-field NMR (LF-NMR) relaxation analyzers have been more and more widely used in food science, biomedicine, and petroleum exploration in recent years. An LF-NMR device analyzes various interactions between sample molecules through spin relaxation, diffusion, flow, and imaging experiments. However, temporal field fluctuations in the magnet limit the scope of application. A stable magnetic field is the basic guarantee for detecting weak signals with shorter relaxation time. This paper presents an approach involving a field-frequency lock for a 21.3-MHz high-performance NMR bipolar permanent magnet. The diameter spherical volume (DSV) of the magnet is only 60 mm. The field-frequency lock system uses 19F in a micro coil integrated into the main probe as a lock detector. The required magnetic field compensation can be calculated from the lock free induction decay (FID) signal frequency, which is measured by lock spectroscopy. The compensation coils used to produce the magnetic field were designed based on Helmholtz coils. The system determines the signal frequency by detecting the lock FID signal and calculates the required compensation-coil current to stabilize the main magnetic field of the analyzer. The results of practical locking experiments in a 21.3-MHz high-performance NMR relaxation analyzer showed that this new approach helps to reduce magnetic field fluctuations from 11 ppm/3h (11 × 10–6) to 0.4 ppm/3h (0.4 × 10–6), which meets the application requirements. This approach is especially viable and effective for a permanent magnet with large field fluctuations. This paper also provides observations of the effect of these fluctuations on NMR measurements before and after installation of the field-frequency lock system.

High-Resolution NMR Spectroscopy at Large Fields with Nitrogen Vacancy Centers

Abstract:Ensembles of nitrogen-vacancy (NV) centers are used as sensors to detect NMR signals from micron-sized samples at room temperature. In this scenario, the regime of large magnetic fields is especially interesting as it leads to a large nuclear thermal polarisation -- thus, to a strong sensor response even in low concentration samples -- while chemical shifts and J-couplings become more accessible. Nevertheless, this regime remains largely unexplored owing to the difficulties to couple NV-based sensors with high-frequency nuclear signals. In this work, we circumvent this problem with a method that maps the relevant energy shifts in the amplitude of an induced nuclear spin signal that is subsequently transferred to the sensor. This stage is interspersed with free-precession periods of the sample nuclear spins where the sensor does not participate. Thus, our method leads to high spectral resolutions ultimately limited by the coherence of the nuclear spin signal.
