Analysis of Regioselectivity of Lipases Using Acidolysis of Lauric Acid and Camellia Oil

Abstract:Regioselectivity of lipases played a crucial role in structured lipids synthesis and oil modification.Because of differences between aqueous and non-aqueous systems,regioselectivity analyzed by the conventional hydrolysis method might not always match that in synthesis reactions.In this paper,acidolysis of lauric acid and camellia oil was developed to directly evaluate regioselectivity of lipases in solvent-free system.Fatty acid composition and positional distribution of reaction products were detected by gas chromatography.Through this protocol,Lipozyme RM IM,L02,L03 and L04 were identified as sn-1 (3) regioselectivity,L01 was partially selective,and Novozym 435 was nearly non-selective.Predictability of the model reaction had been verified by replacing substrates of lipases.According to the comparative analysis of results of the acidolysis method and hydrolysis one,regioselectivity of lipases under two situations was generally the same,except selectivity of Novozym 435 which was susceptible to solvent systems.Therefore the present method avoided limits of hydrolysis determination results in synthesis reactions.And moreover it lowered the requirement on purity of reactants,which means it can be used to screen numerous lipases at low cost.

Abstract:Lipases are ubiquitous in animals, plants and microorganisms, but industrial lipases are mainly produced by microorganisms. The industrial application of lipases is constrained by its high cost and enzymatic characteristics such as temperature and pH tolerance, specific activity, substrate specificity and solvent resistance. Despite the abundance of information regarding microbial lipase genes and proteins, there are currently very few commercial lipase preparations that are suitable for application in the food, pharmaceutical and energy industries. Accordingly, tremendous research efforts have been made to mine novel alternative lipase resources. This article mainly presents a review of the approaches used to mine microbial lipase resources including environmental screening and metagenomics screening for mining new lipases with good properties, genetic improvement of lipase-producing strains, recombinant expression of lipases in genetically engineered strains, lipase modification by protein engineering, lipase immobilization and optimization of reaction conditions of immobilized lipase.

Abstract:Lipase is the key enzyme for catalyzing triglyceride hydrolysis in vivo, and lipase inhibitors have been used in the management of obesity. We present the first report on the use of lipase-adsorbed halloysite nanotubes as an efficient medium for the selective enrichment of lipase inhibitors from natural products. A simple and rapid approach was proposed to fabricate lipase-adsorbed nanotubes through electrostatic interaction. Results showed that more than 85% lipase was adsorbed into nanotubes in 90 min, and approximately 80% of the catalytic activity was maintained compared with free lipase. The specificity and reproducibility of the proposed approach were validated by screening a known lipase inhibitor (i.e., orlistat) from a mixture that contains active and inactive compounds. Moreover, we applied this approach with high performance liquid chromatography-mass spectrometry technique to screen lipase inhibitors from the Magnoliae cortex extract, a medicinal plant used for treating obesity. Two novel biphenyl-type natural lipase inhibitors magnotriol A and magnaldehyde B were identified, and their IC50 values were determined as 213.03 and 96.96 mu M, respectively. The ligand-enzyme interactions of magnaldehyde B were further investigated by molecular docking. Our findings proved that enzyme-adsorbed nanotube could be used as a feasible and selective affinity medium for the rapid screening of enzyme inhibitors from complex mixtures. (C) 2015 Elsevier B.V. Abstract:Lipase is a highly utilized enzyme that holds significant value in various biotechnological and industrial applications, such as the food, paper, and oleochemical sectors, as well as in pharmaceutical contexts. Nevertheless, the application of the substance is relatively challenging and costly due to its aqueous solubility and instability. The immobilization technique is frequently employed to enhance the enzymatic activity and stability of lipase, and this approach has demonstrated considerable potential. The performance of immobilized lipase on nanomaterials (NMs) is subject to various factors, including the immobilization mechanisms and the specific type of matrix employed. This review examines recent advancements, mechanisms, and effects of nanomaterials (NMs) on lipase immobilization and activity. We also discuss different methods for lipase activity determination. The potential for multiple applications of immobilized lipases has been taken into consideration.

Abstract:As some of the most widely used biocatalysts, lipases have exhibited extreme advantages in many processes, such as esterification, amidation, and transesterification reactions, which causes them to be widely used in food industrial production. However, natural lipases have drawbacks in terms of organic solvent resistance, thermostability, selectivity, etc., which limits some of their applications in the field of foods. In this systematic review, the application of lipases in various food processes was summarized. Moreover, the general structure of lipases is discussed in-depth, and the engineering strategies that can be used in lipase engineering are also summarized. The protocols of some classical methods are compared and discussed, which can provide some information about how to choose methods of lipase engineering. Thermostability engineering and solvent tolerance engineering are highlighted in this review, and the basic principles for improving thermostability and solvent tolerance are summarized. In the future, comput er-aided technology should be more emphasized in the investigation of the mechanisms of reactions catalyzed by lipases and guide the engineering of lipases. The engineering of lipase tunnels to improve the diffusion of substrates is also a promising prospect for further enhanced lipase activity and selectivity.

Abstract:Enzymes are natural catalysts highly specific to the substrate type and operate under mild conditions of temperature, pressure, and pH with high conversion rates, which makes them more efficient than conventional chemical catalysts. The enzymes can be obtained from various sources, animal, vegetable, and microbiological. Lipases are very versatile enzymes, and this has aroused the interest of the industries. However, the great problem of the use of soluble lipases is the high cost of acquisition, low operational stability, and difficulties of recovery, and reuse. Enzymatic immobilization has been suggested as an alternative to reduce the limitations of soluble enzymes, increasing their stability and facilitating recovery, and reuse, significantly reducing the cost of processes involving the use of enzymes. This review presents a discussion on the different immobilization methods for lipase, as well as the challenges of use lipases immobilized on the industrial scale.

Abstract:The demand for adequate and ecologically acceptable procedures to produce the most differentiated products has been growing in recent decades, with enzymes being excellent examples of the advances achieved so far. Lipases are astonishing catalysts with a vast range of applications including the synthesis of esters, flavours, biodiesel, and polymers. The broad specificity of the substrates, as well as the regio‐, stereo‐, and enantioselectivity, are the differentiating factors of these enzymes. Structural modification is a current approach to enhance the activity of lipases. Chemical modification of lipases to improve catalytic performance is of great interest considering the increasingly broad fields of application. Together with the physical immobilization onto solid supports, different strategies have been developed to produce catalysts with higher activity and stability. In this review, practical insights into the different strategies developed in recent years regarding the modification of lipases are described. For the first time, the impact of the modifications on the activity and stability of lipases, as well as on the biotechnological applications, is fully compiled. Abstract:To the Editor: Epidermolytic palmoplantar keratoderma (EPPK, OMIM 144200) is an autosomal dominant genodermatosis characterized by typically diffuse keratoderma on the palms and soles with a sharp demarcated erythematous margin and histologically by epidermolytic hyperkeratosis (EHK). The involvement of the dorsal aspects of the hands and feet is rare in this disease, but several cases with these features are reported (1–5). The molecular basis of EPPK was recently shown to result from mutations in the KRT9 gene encoding keratin 9 (K9), a cytoskeletal protein expressed exclusively in suprabasal keratinocytes of palmoplantar epidermis. We herein report a case of EPPK with knuckle pad-like keratoses over the dorsal aspects of the hands and its mutation analysis in KRT9. The patient was a thirteen-year-old Japanese boy who was the offspring of healthy non-consanguineous parents. He also has an older sister who is clinically normal. A single and continuous spectrophotometric assay for various lipolytic enzymes, using natural, non-labelled lipid substrates

Abstract:A rapid, continuous spectrophotometric assay for measuring the amount and activity of several lipolytic enzymes is described. It is based on the metachromatic properties of the cationic dye safranine, and makes use of the fact that an adequate combination of a lipolytic enzyme with one of its substrates leads to a change in the net negative charge at the lipid/water interface, which is monitored by the absorbance change of safranine. Utilizing this method, most lipolytic enzymes can be detected in very low amounts (milliunit or less) in about 1 min without employing radiolabelled lipids or synthetic lipid analogues. Over a wide range of enzyme concentrations, there is a good linearity between the initial hydrolysis rate (determination by the safranine method) and the amount of enzyme. The versatility of the assay is illustrated by examples showing how phospholipase A2, triacylglycerol hydrolase, phospholipase D or phospholipase C (either general or phosphatidylinositol-specific) activities can be detected, either separately or sequentially. Due to its high sensitivity, simplicity, and rapidity, this assay should find its main application in monitoring column effluents during the purification steps of lipolytic enzymes.
Lipase applications in oil hydrolysis with a case study on castor oil: a review

Abstract:Lipase (triacylglycerol acylhydrolase) is a unique enzyme which can catalyze various types of reactions such as hydrolysis, esterification, alcoholysis etc. In particular, hydrolysis of vegetable oil with lipase as a catalyst is widely studied. Free lipase, lipase immobilized on suitable support, lipase encapsulated in a reverse micelle and lipase immobilized on a suitable membrane to be used in membrane reactor are the most common ways of employing lipase in oil hydrolysis. Castor oil is a unique vegetable oil as it contains high amounts (90%) of a hydroxy monounsaturated fatty acid named ricinoleic acid. This industrially important acid can be obtained by hydrolysis of castor oil. Different conventional hydrolysis processes have certain disadvantages which can be avoided by a lipase-catalyzed process. The degree of hydrolysis varies widely for different lipases depending on the operating range of process variables such as temperature, pH and enzyme loading. Immobilization of lipase on a suitable support can enhance hydrolysis by suppressing thermal inactivation and estolide formation. The presence of metal ions also affects lipase-catalyzed hydrolysis of castor oil. Even a particular ion has different effects on the activity of different lipases. Hydrophobic organic solvents perform better than hydrophilic solvents during the reaction. Sonication considerably increases hydrolysis in case of lipolase. The effects of additives on the same lipase vary with their types. Nonionic surfactants enhance hydrolysis whereas cationic and anionic surfactants decrease it. A single variable optimization method is used to obtain optimum conditions. In order to eliminate its disadvantages, a statistical optimization method is used in recent studies. Statistical optimization shows that interactions between any two of the following pH, enzyme concentration and buffer concentration become significant in presence of a nonionic surfactant named Span 80.

Abstract:Lipase can catalyze varieties of reactions at the interface of aqueous and organic phase. Among various alternatives to modify catalytic performance of lipase, the addition of surfactants, particularly nonionic surfactants, has been widely studied. Low concentrations of nonionic surfactants augment lipase catalysis; on increasing surfactant concentration, often the catalytic performance decreases. Mole ratio of water to (nonionic) surfactant also has a profound effect on lipase activity. Catalytic abilities of some lipases are either enhanced or reduced in the presence of all nonionic surfactants of the same type, whereas for some other lipases, nonionic surfactants of the same type have mixed effect. Nonionic surfactant even changes substrate specificity of lipase. Water-in-ionic liquid microemulsion involving nonionic surfactant often performs better than other systems in improving catalytic ability of lipase. Tween and Triton surfactants often enhance enantiomeric separation catalyzed by lipase. Nonionic surfactants significantly affect activities of immobilized lipase, being present either as a component during immobilization or as a component in reaction medium. Lipases coated with nonionic surfactants act better than reverse micelles and microemulsions containing lipase. Thus, nonionic surfactants help lipase catalyzed processes in various media to enhance production of useful compounds like flavor ester, structured lipids, optically pure compounds, and noncrystalline polymers.

Abstract:Lipid hydrolysis monitoring, including especially glycerides, is necessary for comprehending the anaerobic digestion process in lipid-rich substrates processing. This reaction has not been investigated in such detail so far, despite its potential to be crucial in assuring a stable process. This study suggested and thoroughly validated an uncomplicated method of monitoring lipid hydrolysis during anaerobic digestion, achieving recovery values >95 % with an average relative standard deviation <5 %. Subsequently, the method was applied on the very first detailed observation of glyceride hydrolysis in the anaerobic sludge, tracking even changes in fatty acid profiles during anaerobic digestion. Results showed that lipid hydrolysis can take several days, thus likely affecting the whole anaerobic digestion of lipids. The method aims to provide answers to improve understanding of lipids' fate and their inhibition phenomena in anaerobic digestion.

Abstract:<p>Immobilized enzymes have received incredible interests in industry, pharmaceuticals, chemistry and biochemistry sectors due to their various advantages such as ease of separation, multiple reusability, non-toxicity, biocompatibility, high activity and resistant to environmental changes. This review in between various immobilized enzymes focuses on lipase as one of the most practical enzyme and chitosan as a preferred biosupport for lipase immobilization and provides a broad range of studies of recent decade. We highlight several aspects of lipase immobilization on the surface of chitosan support containing various types of lipase and immobilization techniques from physical adsorption to covalent bonding and cross-linking with their benefits and drawbacks. The recent advances and future perspectives that can improve the present problems with lipase and chitosan such as high-price of lipase and low mechanical resistance of chitosan are also discussed. According to the literature, optimization of immobilization methods, combination of these methods with other techniques, physical and chemical modifications of chitosan, co-immobilization and protein engineering can be useful as a solution to overcome the mentioned limitations.</p>

Abstract:Lipases are industrial biocatalysts, which are involved in several novel reactions, occurring in aqueous medium as well as non-aqueous medium. Furthermore, they are well-known for their remarkable ability to carry out a wide variety of chemo-, regio- and enantio-selective transformations. Lipases have been gained attention worldwide by organic chemists due to their general ease of handling, broad substrate tolerance, high stability towards temperatures and solvents and convenient commercial availability. Most of the synthetic reactions on industrial scale are carried out in organic solvents because of the easy solubility of non-polar compounds. The effect of organic system on their stability and activity may determine the biocatalysis pace. Because of worldwide use of lipases, there is a need to understand the mechanisms behind the lipase-catalyzed reactions in organic solvents. The unique interfacial activation of lipases has always fascinated enzymologists and recently, biophysicists and crystallographers have made progress in understanding the structure-function relationships of these enzymes. The present review describes the advantages of lipase-catalyzed reactions in organic solvents and various effects of organic solvents on their activity.

Abstract:AbstractLipase is one of the most widely used enzymes and plays an important role in biotechnological and industrial processes including food, paper, and oleochemical industries, as well as in pharmaceutical applications. However, its aqueous solubility and instability make its application relatively difficult and expensive. The immobilization technique is often used to improve lipase performance, and the strategy has turned out to be a promising method. Immobilized lipase on nanomaterials (NMs) has shown superiority to the free lipase, such as improved thermal and pH stability, longer stable time, and the capacity of being reused. However, immobilization of lipase on NMs also sometimes causes activity loss and protein loading is relatively lowered under some conditions. The overall performance of immobilized lipase on NMs is influenced by mechanisms of immobilization, type of NMs being used, and physicochemical features of the used NMs (such as particle size, aggregation behavior, NM dimension, and type of coupling/modifying agents being used). Based on the specific features of lipase and NMs, this review discusses the recent developments, some mechanisms, and influence of NMs on lipase immobilization and their activity. Multiple application potential of the immobilized lipases has also been considered.

Abstract:With the industrialization and development of modern science, the application of enzymes as green and environmentally friendly biocatalysts in industry has been increased widely. Among them, lipase (EC. 3.1.1.3) is a very prominent biocatalyst, which has the ability to catalyze the hydrolysis and synthesis of ester compounds. Many lipases have been isolated from various sources, such as animals, plants and microorganisms, among which microbial lipase is the enzyme with the most diverse enzymatic properties and great industrial application potential. It therefore has promising applications in many industries, such as food and beverages, waste treatment, biofuels, leather, textiles, detergent formulations, ester synthesis, pharmaceuticals and medicine. Although many microbial lipases have been isolated and characterized, only some of them have been commercially exploited. In order to cope with the growing industrial demands and overcome these shortcomings to replace traditional chemical catalysts, the preparation of new lipases with thermal/acid-base stability, regioselectivity, organic solvent tolerance, high activity and yield, and reusability through excavation and modification has become a hot research topic.

Abstract:In the absence of colipase and bile salts, using tributyrin emulsions or monomolecular films of dicaprin at low surface pressure, we observed that no significant lipase activity can be measured with Human Pancreatic Lipase (HuPL), Horse Pancreatic Lipase (HoPL) or Dog Pancreatic Lipase (DPL). Only Porcine Pancreatic Lipase (PPL) and recombinant Guinea Pig Pancreatic Lipase Related Protein of type 2 (r-GPL) hydrolyse pure tributyrin in the absence of any additive, as well as dicaprin films at low surface pressures. The former lipases may lack enzyme activity because of irreversible interfacial denaturation due to the high energy existing at the tributyrin/water interface and at the dicaprin film surface at low surface pressures. The enzyme denaturation cannot be reflected in the number of disulfide bridges, since all the pancreatic lipases tested here contain six disulfide bridges, but behaved very differently at interfaces. We propose to use the surface pressure threshold, as determined using the monomolecular technique, as a criterion for classifying lipases in terms of their sensitivity to interfacial denaturation.
