Abstract:Four albino rabbits were treated with 4 different doses of chloroquine phosphate (30–50 mg kg−1) given intragastrically and 10 others were given the hydrochloride (2.5–12.5 mg kg−1 i.v.). The i.v. doses were given as bolus into the median ear vein over 5–8 min. Samples (3–5 mL) of whole blood were subsequently collected at intervals for up to 6 weeks and analysed by HPLC. From the results, log concentration‐time curves were drawn and the i.v. data fitted by computer to 3‐compartment (8 rabbits) or 2‐compartment curves (2 rabbits). The area under the curve (AUC) and half‐lives were calculated for the different doses. A plot of AUC versus dose yielded a straight line but the half‐lives showed wide inter‐individual variation from the same doses.
Fabrication of Enzyme-Immobilized Halloysite Nanotubes for Affinity Enrichment of Lipase Inhibitors from Complex Mixtures.

Abstract:Lipase is the key enzyme for catalyzing triglyceride hydrolysis in vivo, and lipase inhibitors have been used in the management of obesity. We present the first report on the use of lipase-adsorbed halloysite nanotubes as an efficient medium for the selective enrichment of lipase inhibitors from natural products. A simple and rapid approach was proposed to fabricate lipase-adsorbed nanotubes through electrostatic interaction. Results showed that more than 85% lipase was adsorbed into nanotubes in 90 min, and approximately 80% of the catalytic activity was maintained compared with free lipase. The specificity and reproducibility of the proposed approach were validated by screening a known lipase inhibitor (i.e., orlistat) from a mixture that contains active and inactive compounds. Moreover, we applied this approach with high performance liquid chromatography-mass spectrometry technique to screen lipase inhibitors from the Magnoliae cortex extract, a medicinal plant used for treating obesity. Two novel biphenyl-type natural lipase inhibitors magnotriol A and magnaldehyde B were identified, and their IC50 values were determined as 213.03 and 96.96 mu M, respectively. The ligand-enzyme interactions of magnaldehyde B were further investigated by molecular docking. Our findings proved that enzyme-adsorbed nanotube could be used as a feasible and selective affinity medium for the rapid screening of enzyme inhibitors from complex mixtures. (C) 2015 Elsevier B.V. Abstract:Decorating liquid crystals (LCs) with stimuli-responsive materials provides a bridge between the LCs and the biochemical interactions. The alignments of the LC molecules are particularly sensitive. Even small perturbations to the LC interface can substantially direct the orientation of bulk LC phases, which results in transduction and amplification of the chemical and biological events into readily detectable optical outputs. These characteristics make the LCs extremely alluring for the development of biosensors that allow highly sensitive and label-free detection of biomarkers at the aqueous/LC interface. In this review, we organize the LC-based sensing systems into six groups, including monolayer formation, monolayer removal, monolayer disruption, floating LC droplets, sessile LC droplets, and LC elastomer microspheres. The principle, utilization, advancement, and limitations of each platform for the detection of various biomarkers are discussed in detail, which will help expand our understanding of the development of LC-based sensors for advanced diagnostic and therapeutic approaches.

Abstract:An approach to optical transduction and amplification of amphiphile-triggered orientational responses of liquid crystals (LCs) based on the interference effect was developed. The sensitive substrate was obtained by lading 4'-pentyl-4-cyanobiphenyl (5CB) into three-dimensionally ordered silica colloidal crystal (SCC) films. Changes in the optical thickness (Delta OT) of the substrates, which are inverted by their Fabry-Perot fringes, depend on the changes of the refractive index caused by the differences in the orientations of LCs. The orientation changes of LCs loading into SCC films have the effect of amplifying signals. These are based on the interactions between surfactants (alkyl trimethylammonium halides (CnTABs, n = 8, 10, 12, 14, and 16) and sodium lauryl sulfonate (SLS)) and LCs, which induce a particular orientation of the LCs molecules. In this flowing system, the reversibility of the signal response for the adsorption of amphiphile was related to the length of the surfactant chain and its critical micelle concentration (CMC). A new method capable of real-time sensing adsorbate-triggered anchoring transitions based on LC-infiltrated SCC films was accomplished. These results provide basics and principles for online, label-free, and real-time analysis of molecules and their interactions in a flowing environment based on the interference effect.
Fluorometric probe for the lipase level: Design, mechanism and biological imaging application

Abstract:To realize accurate regulation for fluorescent substrate of lipase, two series compounds (I, II) with similar structure were designed and synthesized. The flexible diphenylmethane was permitted I to go deep into the catalytic site of lipase, while rigid structure of 9H-fluoren makes itself difficult to approach the center. Series I could be effectively hydrolyzed by porcine pancreatic lipase (PPL), and the product emitted fluorescence based on aggregation-induced emission (AIE) mechanism, meanwhile different substituent groups (–Br, –Cl, –Ph, –H) on I induced the tunable hydrolysis rate. The limit of detection (LOD) was 0.05 U/mL, and linear calibration range was 0.1-4.0 U/mL, this would be effectively applied to biological detection. This result was well supported by theories calculation and molecular docking. Moreover, substrate 1 was selected to describe the distribution of lipase in many living biology such as zebrafish, Locusta migratoria and Helianthus annuus seeds coat. It was found that lipase and lipid droplets (LDs) coexist in biological systems, but there is no lipase in LDs. The designed probe gives us an opportunity to describe lipase distribution in many important tissues, including biology and medical science.

Abstract:A rapid, continuous spectrophotometric assay for measuring the amount and activity of several lipolytic enzymes is described. It is based on the metachromatic properties of the cationic dye safranine, and makes use of the fact that an adequate combination of a lipolytic enzyme with one of its substrates leads to a change in the net negative charge at the lipid/water interface, which is monitored by the absorbance change of safranine. Utilizing this method, most lipolytic enzymes can be detected in very low amounts (milliunit or less) in about 1 min without employing radiolabelled lipids or synthetic lipid analogues. Over a wide range of enzyme concentrations, there is a good linearity between the initial hydrolysis rate (determination by the safranine method) and the amount of enzyme. The versatility of the assay is illustrated by examples showing how phospholipase A2, triacylglycerol hydrolase, phospholipase D or phospholipase C (either general or phosphatidylinositol-specific) activities can be detected, either separately or sequentially. Due to its high sensitivity, simplicity, and rapidity, this assay should find its main application in monitoring column effluents during the purification steps of lipolytic enzymes.
High-throughput screening of lipase inhibitors as anti-obesity drugs on liquid crystal-infused porous surfaces

Abstract:High-throughput screening of lipase inhibitors is critical for developing anti-obesity drugs. Herein, a robust and versatile sensing platform based on the difference in the sliding time of droplets on liquid crystal-infused porous surfaces (LCIPS) is developed to detect lipase inhibitors. Lipase catalyzes glycerol trioleate (GT) to produce oleate that can adsorb at the solution/liquid crystal (LC) interface due to its high amphiphilicity, which impedes the sliding of droplets on LCIPS, resulting in an extended sliding time. However, in the presence of lipase inhibitors, the enzymatic hydrolysis of GT by lipase is inhibited, which shortens the sliding time. The screening of three lipase inhibitors, orlistat, cetilistat, and epigallocatechin gallate (EGCG), popular anti-obesity drugs, is demonstrated to have IC 50 values of 17.45±3.15 nM, 72.15±6.32 nM, and 2.19±0.24 μM, respectively. This study provides a simple, inexpensive, portable, user-friendly, and instrument-free method for rapid, accurate, and high-throughput screening of lipase inhibitors. The LCIPS-based sensing platform is up-and-coming in developing advanced bioanalytical sensors.

Highly sensitive and label-free detection of catalase by a H2O2-responsive liquid crystal sensing platform

Abstract:Abnormal levels of catalase are implicated in multiple clinical diseases, so it is of great importance to monitor the levels of catalase in real samples. In this report, a H2O2-responsive liquid crystal (LC) sensing platform was developed and applied to the detection of catalase in human serum for the first time. The platform was constructed by doping LCs with a synthesized surfactant, namely N,N-dimethyl-N-(4-(4,4,5,5-tetramethyl-1,3,2-dioxaborolan-2-yl)benzyl)tetradecan-1-aminium bromide (PBEC14A), which can be decomposed by H2O2. Self-assembled monolayers formed at the aqueous/LC interface when immersing the surfactant-doped LCs into an aqueous solution. Accordingly, the LCs displayed a dark image that indicates a perpendicular state of the LCs at the interface. However, when H2O2 was present in the aqueous solution, the optical image of the LCs became bright that represents a planar state of the LCs at the interface. It is caused by disruption of the surfactant monolayers due to decomposition of PBEC14A by H2O2. Interestingly, it was found that the LCs still displayed the dark image when the pre-incubated mixture of catalase and H2O2 was transferred onto the LC sensing platform due to enzymatic hydrolysis of H2O2 by catalase. The detection limit of catalase reached as low as 5.5 mU/mL. In addition, it also shows excellent performance for the detection of catalase in human serum. As a simple and convenient method, it shows high promise in the highly sensitive and label-free detection of catalase in practical applications.

Immobilization of Lipase Onto Functional Cyclomatrix Polyphosphazene Microspheres

Abstract:Functional polyphosphazene cyclomatrix microspheres (PMS) containing glucose and dodecyl moieties were utilized for physical adsorption of lipase. In this regard, Candia Rugosa lipase was immobilized onto the above microspheres through physical adsorption, which exhibited catalytic hydrolysis of glycerol triacetate as the model reaction. As observed under transmission electron microscope (TEM) and scanning electron microscope (SEM), the size of all the microspheres ranged from 400 nm to 1 mu m with a mean diameter of 680 nm. Zeta potential analysis demonstrated that the microspheres were negatively charged. The influences of buffer pH, substrate concentration and temperature were also studied along with reusability. The obtained results demonstrated that the immobilized lipase on deprotected glycosylated PMS (D-GPMS) showed the best performance and possessed the highest enzymatic loading activity around 1.0884 mmol L-1 min(-1) g(-1), and retained nearly 50% of its initial activity even after 10 cycles of application. Moreover, the lipase-microsphere complex displayed high pH adaptability from 7 to 9 and temperature stability above 40 degrees C. From the viewpoint of biocompatible PMS support, the GPMS-lipase conjugate have potential applications in field such as food, medicine and environment. (C) 2016 Elsevier B.V. All rights reserved.

Abstract:A cost-effective and label-free optical fiber sensor was proposed to detect phospholipase A2 (PLA2) in nM concentration. The sensor is made of an alkoxysilane-modified side-polished fiber (SPF) coated with 4′-pentyl-4-cyanobiphenyl (5CB) and self-assembled phospholipid (L-DLPC). It is found that the relative transmission optical power (RTOP) of the fiber sensor decreases due to the 5CB realignment and redistribution induced by the PLA2 hydrolysis of L-DLPC. The response-time at 5 dB RTOP variation exhibits an exponential dependence on PLA2 concentration, allowing us to detect the PLA2 by the 5 dB-response time. This detection method can reduce the detection time. Compare with the traditional copper-grid sensor, the proposed novel fiber sensor has a lower detection limit (<1 nM). Furthermore, the sensor has good repeat-ability and specificity.The sensor's RTOP variation for PLA2 detection at 1 nM is ∼21 times higher than that for five other enzymes (trypsin, amylase, thrombin, glucose oxidase, pepsin) at 1000 nM and lipase at 50 nM. This confirms the sensor's excellent PLA2 specificity. Furthermore, the fiber sensor provides a potential way to be incorporated into micro-flow chips to quantitatively detect biological molecules in a real-time and online manner.

Abstract:A briefness, convenient, label-free and sensitive liquid crystal (LC) sensing platform is demonstrated to detect catalase in human serum with the assistance of CeO2 nanomaterials and the single-stranded DNA (ssDNA). In the presence of H2O2 and ssDNA/CeO2 nanomaterials complex, a colourful optical appearance of LCs doped with octadecyl-trimethylammonium bromide (OTAB) was observed under a polarized optical microscopy (POM), which is corresponding to the oblique alignment of LCs at the aqueous-LC interface. This can be ascribed to the disruption of the OTAB monolayers by ssDNA that is set free from the surface of CeO2 nanomaterials in the presence of H2O2. The stronger coordination interaction between H2O2 and CeO2 nanomaterials causes a replacement for ssDNA. Interestingly, with the hydrolysis of H2O2 by catalase, the LCs exhibit a dim image, which suggests the upright alignment of LCs at the aqueous-LC interface. It is because there is no free ssDNA discharged into the aqueous solution after the hydrolysis of H2O2. Hence, by observing the changes of colourful and dim optical images of LCs, catalase can be analyzed with remarkable sensitivity and excellent specificity. The detection limit of catalase reaches as low as ∼ 1 mU/mL. Besides, the detection of catalase in human serum is also successfully achieved, which makes this assay meets the requirement of practical applications. Therefore, this work offers an effective and appealing strategy for the real-time and label-free detection of catalase in human serum.

Abstract:The interface between a nematic liquid crystal phase, 4-cyano-4′-pentylbiphenyl (5CB) and water was examined for protein detection by monitoring the formation of a complex between sodium polystyrene sulfonate (PSSNa) and a positively charged biological species on the 5CB in a transmission electron microscopy (TEM) grid cell coated with a strong anionic polyelectrolyte-containing block copolymer, LCP-b-PSSNa (LCP:poly(4-cyanobiphenyl-4′-oxyundecylacrylate)). This block copolymer was successfully synthesized by reversible addition-fragmentation chain transfer polymerization. A monolayer of LCP-b-PSSNa in a Langmuir Blodgett trough (in which PSSNa and LCP were located in and above water, respectively, in the TEM grid cell) was transferred to the 5CB/water interface in the 5CB-filled TEM grid that was already placed on octadecyltrichlorosilane-coated glass. Model proteins such as bovine serum albumin (BSA), hemoglobin (Hb), α chymotrypsinogen-A (ChTg), and lysozyme (LYZ) having different isoelectric points (pIs) were tested for non-specific protein detection. When the protein solutions were injected into the TEM grid cell, the initial homeotropic orientation of 5CB in the TEM grid cell changed to a planar one below the pIs of the proteins due to electrostatic interactions between PSSNa (- charge) and the proteins (+ charge); this did not occur above the pIs of the tested proteins. The minimum concentrations at which the homeotropic to planar configurational changes (H-P changes) occurred were 0.02, 0.04, 0.04, and 0.08 wt% for BSA, Hb, ChTg, and LYZ, respectively. Therefore, the positively charged biomaterials were visually detected at the PSSNa-coated LC/water interface during an H-P change by using polarized optical microscopy under crossed polarizers. This simple set-up for non-specific biomaterial detection paves a way for the development of efficient and excellent quality biosensors.

Abstract:Reliable and sensitive detection of lipase activity is essential for the early diagnosis and monitoring of acute pancreatitis or progression of digestive diseases. However, the available fluorescent probes for detection of lipase activity are only implemented in a hexane-water two-phase system due to the nature of heterogeneous catalysis of lipase, thus limiting their applications in direct imaging of lipase activity in living cells and tissues. Here we designed and synthesized a "turn on" fluorescent probe CPP based on self-assembled micelles for hydrolysis of lipase. The CPP probe exhibits high selectivity and excellent sensitivity for the detection of lipase in such a homogeneous system and is successfully applied for monitoring lipase activity in pancreatic AR42J cells, tissues, and serums. Taken together, the fluorescent CPP probe not only provides a tool for diagnostic potential in pancreatic disease but also demonstrates an application potential for micelle self-assembly-based development of biological probes.
An integrated liquid crystal sensing device assisted by the surfactant-embedded smart hydrogel

Abstract:The abnormal level of trypsin in biological fluids can cause some acute illnesses, such as acute pancreatitis, cystic fibrosis and malnutrition. In this paper, we report the development of an integrated liquid crystal (LC) sensing device for simple, rapid and sensitive detection of trypsin assisted by the surfactant-embedded smart hydrogel. The gelatin hydrogel mixed with CTAB is added into the side channel of the LC sensing device. In the presence of trypsin, the gelatin hydrogel is decomposed, which triggers instant release of CTAB into the aqueous solution. The CTAB molecules are then captured by the LCs and form CTAB monolayers at the aqueous/LC interface, which leads to change of the LC images from the bright to the dark appearance under the crossed polarizers. The integrated LC sensing device has a remarkable detection limit of 3.4×10-5 mg/mL. It is successfully employed to single-step detection of trypsin in human serum within 30 min. The integrated LC sensing device with use of the surfactant-embedded hydrogel takes advantages of single-step detection, high portability, remarkable sensitivity and fast response time, which provides a new perspective to facilitate development of user-friendly LC-based sensors.

Rapid disposable lipase activity sensor for automatic industrial application

Abstract:A low-cost disposable lipase activity sensor was developed and characterized overcoming the main drawbacks of the already existing disposable ones: insufficient reproducibility to be applied for automatic lipase activity quantification and complicated and long manual analytical procedure performed by qualified personnel. The sensor is based on a capacitor formation on an electrode surface by a substrate layer deposition possessing high electrical resistivity and dielectric constant (composite of olive oil mixed with BaTiO3 nanoparticles) serving as both: sensor sensitive layer and capacitor dielectric. Its decomposition by lipase catalyzed hydrolysis causes its thickness decrease resulting in capacitance increase proportional to the lipase activity. The sensor was characterized in terms of limit of quantification, linear quantification range, sensitivity, response time, precision, repeatability, storage time and the results are presented and discussed. The validation experiment showed excellent agreement with the standardized titrimetric method.

Abstract:The development of liquid crystal (LC)-based sensors with superior performances such as high portability, excellent stability, great convenience, and remarkable sensitivity is highly demanded. This work proposes a new strategy for constructing the LC-based sensor using enzyme-linked dual-functional nucleic acid (d-FNA) on magnetic beads (MBs). The detection of kanamycin (KA) is demonstrated as a model. Acetylcholinesterase (AChE) is assembled onto the KA aptamer-modified MBs with a d-FNA strand that consists of an AChE aptamer and the complementary sequence of a KA aptamer. As the specific recognition of KA by its aptamer triggers the release of AChE from the MBs, the myristoylcholine (Myr) solution after incubation with the MBs causes the black image of the LCs due to the formation of the Myr monolayer at the aqueous/LC interface. Otherwise, in the absence of KA, AChE is still decorated on the MBs and causes the hydrolysis of Myr. Therefore, a bright image of LCs is obtained. The detection of KA is successfully achieved with a lower detection limit of 48.1 pg/mL. In addition, a thin polydimethylsiloxane (PDMS) layer-coated glass and a portable optical device are used to improve the stability and portability of the LC-based sensor to advance potential commercial applications. Furthermore, the detection of KA in milk with a portable device is demonstrated, showing the potential of the proposed enzyme-linked LC-based sensor.
Long-term live-cell lipid droplet-targeted biosensor development for nanoscopic tracking of lipid droplet-mitochondria contact sites

Abstract:Background: Lipid droplets (LDs) establish a considerable number of contact sites with mitochondria to enable energy transfer and communication. In this study, we developed a fluorescent biosensor to image LD-mitochondria interactions at the nanoscale and further explored the function of LD-mediated matrix transmission in processes involving multi-organelle interactions. Methods: A fluorescent probe called C-Py (C21H19N3O2, 7-(diethylamino) coumarin-3-vinyl-4-pyridine acetonitrile) was designed and synthesized. Colocalization of C-Py and the commercial LD stain Nile Red was analyzed in HeLa cells. The fluorescence stability and signal to background ratio of C-Py under structured illumination microscopy (SIM) were compared to those of the commercial probe BODIPY493/503. The cytotoxicity of C-Py was assessed using CCK-8 assays. The uptake pattern of C-Py in HeLa cells was then observed under various temperatures, metabolic levels, and endocytosis levels. Contact sites between LDs and various organelles, such as mitochondria, nuclei, and cell membrane, were imaged and quantitated using SIM. Physical changes to the contact sites between LDs and mitochondria were monitored after lipopolysaccharide induction. Results: A LD-targeted fluorescent biosensor, C-Py, with good specificity, low background signal, excellent photostability, low cytotoxicity, and high cellular permeability was developed for tracking LD contact sites with multiple organelles using SIM. Using C-Py, the subcellular distribution and dynamic processes of LDs in living cells were observed under SIM. The formation of contact sites between LDs and multiple organelles was visualized at a resolution below ~200 nm. The number of LD-mitochondria contact sites formed was decreased by lipopolysaccharide treatment inducing an inflammatory environment. Conclusions: C-Py provides strategies for the design of ultra-highly selective biosensors and a new tool for investigating the role and regulation of LDs in living cells at the nanoscale.

Harnessing Liquid Crystal Sensors for High-Throughput Real-Time Detection of Structural Changes in Lysozyme during Refolding Processes

Abstract:Despite the rapid advances in process analytical technology, the assessment of protein refolding efficiency has largely relied on off-line protein-specific assays and/or chromatographic procedures such as reversed-phase high-performance liquid chromatography and size exclusion chromatography. Due to the inherent time gap pertaining to traditional methods, exploring optimum refolding conditions for many recombinant proteins, often expressed as insoluble inclusion bodies, has proven challenging. The present study describes a novel protein refolding sensor that utilizes liquid crystals (LCs) to discriminate varying protein structures during unfolding and refolding. An LC layer containing 4-cyano-4'-pentylbiphenyl (5CB) intercalated with 1,2-dioleoyl-sn-glycero-3-phosphoethanolamine (DOPE) is used as a sensing platform, and its proof-of-concept performance is demonstrated using lysozyme as a model protein. As proteins unfold or refold, a local charge fluctuation at their surfaces modulates their interaction with zwitterionic phospholipid DOPE. This alters the alignment of DOPE molecules at the aqueous/LC interface, affecting the orientational ordering of bulk LC (i.e., homeotropic to planar for refolding and planar to homeotropic for unfolding). Differential polarized optical microscope images of the LC layer are subsequently generated, whose brightness directly linked to conformational changes of lysozyme molecules is quantified by gray scale analysis. Importantly, our LC-based refolding sensor is compatible with diverse refolding milieus for real-time analysis of lysozyme refolding and thus likely to facilitate the refolding studies of many proteins, especially those lacking a method to determine structure-dependent biological activity.
Quantitative Biosensing Based on a Liquid Crystal Marginally Aligned by the PVA/DMOAP Composite for Optical Signal Amplification

Abstract:The working principle for a liquid crystal (LC)-based biosensor relies on the disturbance in the orderly aligned LC molecules induced by analytes at the LC-aqueous or LC-solid interface to produce optical signals that can be typically observed under a polarizing optical microscope (POM). Our previous studies demonstrate that such optical response can be enhanced by imposing a weak electric field on LCs so that they are readily tilted from the homeotropic alignment in response to lower concentrations of analytes at the LC-glass interface. In this study, an alternative approach toward signal amplification is proposed by taking advantage of the marginally tilted alignment configuration without applying an electric field. The surface of glass substrates was modified with a binary aligning agent of poly(vinyl alcohol) (PVA) and dimethyloctadecyl[3-(trimethoxysilyl)propyl] ammonium chloride (DMOAP), in which the amount of PVA was fine-tuned so that the interfacing LC molecules were slightly tilted but remained virtually homeotropically aligned to yield no light leakage under the POM in the absence of an analyte. Two nematic LCs, E7 and 5CB, were each sandwiched between two parallel glass substrates coated with the PVA/DMOAP composite for the detection of bovine serum albumin (BSA), a model protein, and cortisol, a small-molecule steroid hormone. Through image analysis of the optical appearance of E7 observed under the POM, a limit of detection (LOD) of 2.5 × 10−8 μg/mL for BSA and that of 3 × 10−6 μg/mL for cortisol were deduced. Both values are significantly lower than that obtained with only DMOAP as the alignment layers, which correspond to signal amplification of more than six orders of magnitude. The new approach for signal amplification reported in this work enables analytes of a wide range of molecular weights to be detected with high sensitivity.
