Abstract:Lipases are ubiquitous in animals, plants and microorganisms, but industrial lipases are mainly produced by microorganisms. The industrial application of lipases is constrained by its high cost and enzymatic characteristics such as temperature and pH tolerance, specific activity, substrate specificity and solvent resistance. Despite the abundance of information regarding microbial lipase genes and proteins, there are currently very few commercial lipase preparations that are suitable for application in the food, pharmaceutical and energy industries. Accordingly, tremendous research efforts have been made to mine novel alternative lipase resources. This article mainly presents a review of the approaches used to mine microbial lipase resources including environmental screening and metagenomics screening for mining new lipases with good properties, genetic improvement of lipase-producing strains, recombinant expression of lipases in genetically engineered strains, lipase modification by protein engineering, lipase immobilization and optimization of reaction conditions of immobilized lipase.

Abstract:An extracellular lipase from Aureobasidium pullulans was obtained and purified with a specific activity of 17.7 U/mg of protein using ultrafiltration and a DEAE-Sepharose Fast Flow column. Characterization of the lipase indicated that it is a novel finding from the species A. pullulans. The molecular weight of the lipase was 39.5 kDa, determined by sodium dodecyl sulfonate-polyacrylamide gel electrophoresis (SDS-PAGE). The enzyme exhibited its optimum activity at 40 °C and pH of 7. It also showed a remarkable stability in some organic solutions (30%, v/v) including n-propanol, isopropanol, dimethyl sulfoxide (DMSO), and hexane. The catalytic activity of the lipase was enhanced by Ca2+ and was slightly inhibited by Mn2+ and Zn2+ at a concentration of 10 mmol/L. The lipase was activated by the anionic surfactant SDS and the non-ionic surfactants Tween 20, Tween 80, and Triton X-100, but it was drastically inhibited by the cationic surfactant cetyl trimethyl ammonium bromide (CTAB). Furthermore, the lipase was able to hydrolyze a wide variety of edible oils, such as peanut oil, corn oil, sunflower seed oil, sesame oil, and olive oil. Our study indicated that the lipase we obtained is a potential biocatalyst for industrial use.

Microbial lipases are usually more useful than that from plants or animals because of the possible high yields, easiness of genetic manipulation, regular supply due to absence of seasonal fluctuations. In this paper, the screening of lipases producing microbe, the purification, molecular structure, catalytic mechanism and properties of lipases are introduced and the application of microbial lipases in the synthesis of sugar esters, cocoa butter equivalent and human milk fat substitute is reviewed in detail.

Abstract:ANREP and STARLING(1) demonstrated that in the dog a reciprocal action existed between the blood-pressure in the head and neck and in the periphery; a rise in the former causing a fall in the latter and vice versa. These results they attributed to a direct influence of the vascular pressure on the vaso-motor and vagus centres. Hering (2) has described in detail a depressor reflex from the sinus caroticus, and in his recent monograph attributed the results of Anrep and Starling to the presence of this reflex in their preparations, although he did not offer any adequate proof of this contention. Anrep and Segall(3) in further experiments with the innervated heart lung preparation asserted that, even after complete excision of the sinus caroticus itself, a "central" action upon heart rate still existed, and Nash(4), using the same preparation, found that a rise in the cerebral blood-pressure still determined a fall of blood-pressure in the body. During some experiments upon the cerebral blood vessels we obtained results which indicated that in the intact animal there was no reciprocal action between the blood-pressure in the circle of Willis and the peripheral blood-pressure, and in view of the divergence of opinion between the authors stated we extended our observations to elucidate this problem. In the experiments to be described, we have changed the bloodpressure in the circle of Willis by occluding the vertebral arteries. The occlusion of these arteries, while lowering the pressure in the circle of Willis, has no mechanical influence upon the general arterial pressure, and any reciprocal action between the pressure in the circle of Willis and the peripheral blood-pressure can be studied without the interference of the depressor reflex from the sinus3. Abstract:Enzymes play a crucial role in various industrial sectors. These biocatalysts not only ensure sustainability and safety but also enhance process efficiency through their unique specificity. Lipases possess versatility as biocatalysts and find utilization in diverse bioconversion reactions. Presently, microbial lipases are gaining significant focus owing to the rapid progress in enzyme technology and their widespread implementation in multiple industrial procedures. This updated review presents new knowledge about various origins of microbial lipases, such as fungi, bacteria, and yeast. It highlights both the traditional and modern purification methods, including precipitation and chromatographic separation, the immunopurification technique, the reversed micellar system, the aqueous two-phase system (ATPS), and aqueous two-phase flotation (ATPF), moreover, delves into the diverse applications of microbial lipases across several industries, such as food, vitamin esters, textile, detergent, biodiesel, and bioremediation. Furthermore, the present research unveils the obstacles encountered in employing lipase, the patterns observed in lipase engineering, and the application of CRISPR/Cas genome editing technology for altering the genes responsible for lipase production. Additionally, the immobilization of microorganisms' lipases onto various carriers also contributes to enhancing the effectiveness and efficiencies of lipases in terms of their catalytic activities. This is achieved by boosting their resilience to heat and ionic conditions (such as inorganic solvents, high-level pH, and temperature). The process also facilitates the ease of recycling them and enables a more concentrated deposition of the enzyme onto the supporting material. Consequently, these characteristics have demonstrated their suitability for application as biocatalysts in diverse industries.

The Purification and N-Terminal Sequencing of the Lipase from Penicillium Expansum WMC20718

Abstract:The alkaline lipase produced from penicillium expansum WMC20718 was purified 18.5 fould with the recovery of 48.8% by centrifugation, ammonium sulfate precipitation, phenyl Sepharose CL 4B, DEAE Sepharose and Sephadex G 75 column chromatographies. The purified lipase had a specific activity of 4 200 U/mg protein, which were showed to be homogeneous by SDS Page and RP HPLC. The molecular weight of the native lipase was estimated to be about 28.2 kD by gel filtration using Sephades G 150, and that of the denatured lipase about 27.5 kD by SDS PAGE, indication that the lipase was a monomer. The N terminal amino acid sequence was determined as follows: A T A D A A A F P D L H R A A K L S S A. The optimum temperature of the lipase was 30 ℃ and was stable below 35 ℃, but only 30% of its activity remained after 30 min incubation at 45 ℃. The lipase was stable at pH from 6.5 to 10.5, and its optimal pH was 10.0. Anionic surfactant LAS affected the enzymatic stability greatly, but non ionic surfactant AEO 9 had little effect and could lessen the effect of LAS. The alkaline lipase showed resistance to alkaline protease in proper concentration.

Abstract:With the industrialization and development of modern science, the application of enzymes as green and environmentally friendly biocatalysts in industry has been increased widely. Among them, lipase (EC. 3.1.1.3) is a very prominent biocatalyst, which has the ability to catalyze the hydrolysis and synthesis of ester compounds. Many lipases have been isolated from various sources, such as animals, plants and microorganisms, among which microbial lipase is the enzyme with the most diverse enzymatic properties and great industrial application potential. It therefore has promising applications in many industries, such as food and beverages, waste treatment, biofuels, leather, textiles, detergent formulations, ester synthesis, pharmaceuticals and medicine. Although many microbial lipases have been isolated and characterized, only some of them have been commercially exploited. In order to cope with the growing industrial demands and overcome these shortcomings to replace traditional chemical catalysts, the preparation of new lipases with thermal/acid-base stability, regioselectivity, organic solvent tolerance, high activity and yield, and reusability through excavation and modification has become a hot research topic.

Abstract:Enzymes are natural catalysts highly specific to the substrate type and operate under mild conditions of temperature, pressure, and pH with high conversion rates, which makes them more efficient than conventional chemical catalysts. The enzymes can be obtained from various sources, animal, vegetable, and microbiological. Lipases are very versatile enzymes, and this has aroused the interest of the industries. However, the great problem of the use of soluble lipases is the high cost of acquisition, low operational stability, and difficulties of recovery, and reuse. Enzymatic immobilization has been suggested as an alternative to reduce the limitations of soluble enzymes, increasing their stability and facilitating recovery, and reuse, significantly reducing the cost of processes involving the use of enzymes. This review presents a discussion on the different immobilization methods for lipase, as well as the challenges of use lipases immobilized on the industrial scale.

Abstract:As some of the most widely used biocatalysts, lipases have exhibited extreme advantages in many processes, such as esterification, amidation, and transesterification reactions, which causes them to be widely used in food industrial production. However, natural lipases have drawbacks in terms of organic solvent resistance, thermostability, selectivity, etc., which limits some of their applications in the field of foods. In this systematic review, the application of lipases in various food processes was summarized. Moreover, the general structure of lipases is discussed in-depth, and the engineering strategies that can be used in lipase engineering are also summarized. The protocols of some classical methods are compared and discussed, which can provide some information about how to choose methods of lipase engineering. Thermostability engineering and solvent tolerance engineering are highlighted in this review, and the basic principles for improving thermostability and solvent tolerance are summarized. In the future, comput er-aided technology should be more emphasized in the investigation of the mechanisms of reactions catalyzed by lipases and guide the engineering of lipases. The engineering of lipase tunnels to improve the diffusion of substrates is also a promising prospect for further enhanced lipase activity and selectivity.

Abstract:To the Editor: Epidermolytic palmoplantar keratoderma (EPPK, OMIM 144200) is an autosomal dominant genodermatosis characterized by typically diffuse keratoderma on the palms and soles with a sharp demarcated erythematous margin and histologically by epidermolytic hyperkeratosis (EHK). The involvement of the dorsal aspects of the hands and feet is rare in this disease, but several cases with these features are reported (1–5). The molecular basis of EPPK was recently shown to result from mutations in the KRT9 gene encoding keratin 9 (K9), a cytoskeletal protein expressed exclusively in suprabasal keratinocytes of palmoplantar epidermis. We herein report a case of EPPK with knuckle pad-like keratoses over the dorsal aspects of the hands and its mutation analysis in KRT9. The patient was a thirteen-year-old Japanese boy who was the offspring of healthy non-consanguineous parents. He also has an older sister who is clinically normal. Abstract:AbstractLipase is one of the most widely used enzymes and plays an important role in biotechnological and industrial processes including food, paper, and oleochemical industries, as well as in pharmaceutical applications. However, its aqueous solubility and instability make its application relatively difficult and expensive. The immobilization technique is often used to improve lipase performance, and the strategy has turned out to be a promising method. Immobilized lipase on nanomaterials (NMs) has shown superiority to the free lipase, such as improved thermal and pH stability, longer stable time, and the capacity of being reused. However, immobilization of lipase on NMs also sometimes causes activity loss and protein loading is relatively lowered under some conditions. The overall performance of immobilized lipase on NMs is influenced by mechanisms of immobilization, type of NMs being used, and physicochemical features of the used NMs (such as particle size, aggregation behavior, NM dimension, and type of coupling/modifying agents being used). Based on the specific features of lipase and NMs, this review discusses the recent developments, some mechanisms, and influence of NMs on lipase immobilization and their activity. Multiple application potential of the immobilized lipases has also been considered.

Abstract:Many researchers have found fungi as a reliable source of lipase due to the versatility of their properties, ease of mass production, thermal stability, pH stability, broad substrate specificity, retained activity in organic solvents, and their low cost extraction procedure.This review paper presents an overview about the main aspects of fungal lipases screened from several types of strains, as well as their use as biocatalysts. Additionally, some biochemical properties will be reported. As commonly known, lipases can be produced from animals, plants and microorganisms. Compared to other lipases, those obtained from fungi have been found to be more productive, a fact that encouraged the massive production of most fungal lipases due to their considerable commercial importance during the past few years. This paper is concerned about some of the major characteristics that made fungal lipases desirable products in the industrial fields.Due to the enantioselective properties of fungal lipases and their ability to remain active under extreme temperature, pH and organic solvents, enzymes are capable to synthesize esters as well as to catalyze a variety of chemical reactions that include esterification, transesterification, acidolysis and aminolysis in aqueous and non‐aqueous media. Furthermore, lipases are considered to have a commercial importance for biotechnological application fields, which makes them increasingly popular in food, detergent, cosmetic, organic synthesis, and pharmaceutical domains. The biotechnological potential of lipases has made the latter a coveted choice in industries for the present and future as biocatalysts. In addition, a classification of these fungal enzymes is also highlighted in this review. Moreover, the impact of an immobilization strategy of these fungal strains to achieve higher yields and to improve their production is discussed.Finally, fungal enzymes have played a crucial role from ancient times to today in different fields using several types of biological systems, which gives them a great interest for the production of these enzymes in large amounts with low cost and easy viability to enlarge their use in many industries.Likewise, some future perspectives on lipase production will also be discussed by focusing on special cases on lipase engineering.This article is protected by copyright. A novel strategy for partial purification of alkane hydroxylase from P. chrysogenum SNP5 through reconstituting its native membrane into liposome

Abstract:Integral proteins or enzymes are still challenging to purify into their native state because of their need for an amphipathic environment and cofactors. Alkane hydroxylase (AlkB) is a membrane-bound enzyme that catalyzes the hydroxylation of a range of alkanes that have a broad spectrum of applications. In the current study, a novel approach has been explored for partial purification of alkane hydroxylase (AlkB) in its native state through restructuring the lipid bilayer of Penicillium chrysogenum SNP5 into a liposome to extend the native and protective environment to AlkB enzyme. Three different methods i.e., reverse-phase evaporation method (RPEM), detergent-based method (DBM), and ethanol injection method (EIM) have been used for reconstituting its native membrane into liposome. On characterizing liposomes through fluorescence imaging, AFM, and particle size analysis, the reverse-phase evaporation method gave the best results based on the size distribution (i.e., 100–300 nm), the morphology of liposomes, and maximum AlkB specific activity (i.e., 140.68 U/mg). The maximum reconstitution efficiency of 29.48% was observed in RPEM followed by 17.3% in DBM and 12.3% in EIM. On the characterization of the purified AlkB, the molecular weight was measured of 44.6 KDa and the thermostability of liposomes synthesized with the RPEM method was obtained maximum at 55 °C. This approach may open a new strategy for the purification of integral enzymes/proteins in their native state in the field of protein purification and its applications in diversified industries.

Pre-images are later back-loaded to perform high speed reconstructions. One such processor is assigned to each image slice of the Super-PETT. Abstract:Increasing environmental concerns and significant demand for industrial fuels necessitate the development of alternate and sustainable energy sources. Biodiesel is a renewable and environmentally friendly fuel produced by trans-esterifying a variety of feedstocks. Lipases are robust triacylglycerol ester hydrolases that catalyze hydrolysis, esterification, interesterification, transesterification, acylation, acidolysis, alcoholysis, aminolysis, and resolution of racemates. Although the lipase-assisted greener biosynthesis of biodiesel has numerous advantages over the traditional alkali-based process, low catalytic efficiency, marginal stability, and high cost of lipase enzymes limit its widespread industrial processability. Protein engineering methodologies such as directed evolution, semi-rational design, and rational design can effectively tailor or improve the biocatalytic characteristics of lipase enzymes used in biodiesel generation, such as thermostability, solvent tolerance, activity, and substrate specificity. These innovative techniques improved our ability to predict structure-function relationships, engineering qualities, computational tools for designing new biocatalysts, and functional screening to manipulate enzyme traits for application in prevalent industrial bioprocesses. Many recent studies have demonstrated improved lipase performance, such as activity, stability, and specificity via protein engineering. This review spotlights a current overview of lipase engineering at the molecular scale with robust biocatalytic properties for biodiesel synthesis.

Purification and Characterization of a Lipase with High Thermostability and Polar Organic Solvent-Tolerance from Aspergillus niger AN0512

Abstract:An extracellular lipase (EC 3.1.1.3, AN0512Lip) from Aspergillus niger AN0512 was purified and its characteristics were investigated. After the process of ammonium sulfate precipitation followed by ion-exchange chromatography and gel filtration, the purified lipase was achieved with 203.6-fold purification and 22.1 % recovery. AN0512Lip exhibited the highest activity at 50 °C and pH 5.0. It was thermostable and pH-stable, as indicated by that more than 50 % activity retained at 60 °C for 20 h and more than 90 % activity retained at pH 3.0 for 20 h, respectively. AN0512Lip activity was stimulated by some divalent metal ions (especially Cu(2+), Ca(2+)), while greatly suppressed by EDTA, indicating that AN0512Lip was a metal-activated enzyme. Moreover, AN0512Lip exhibited high tolerance for various polar organic solvents with log P < 0.8, and the highest lipase activity (476 % of its original activity) was achieved after addition of 90 % (V/V) isopropanol to the reaction mixture. AN0512Lip also displayed 3-regiospecificity and great affinity for the long-chain fatty ester. The preliminary test showed that AN0512Lip was a candidate for enriching EPA and DHA in fish oil. All the unique properties, such as thermostability, Cu(2+)-dependent, 3-regiospecificity, and polar organic solvent-tolerance, indicated that AN0512Lip could have potential applications in the food industry, even in organic synthesis and the pharmaceutical industry.

Abstract:Constituents of the fruits of Rubus corchorifolius were investigated. A new compound, namely rubusin A (1), along with three known compounds, was isolated and characterized. Among them, the new compound exhibited significant activity against kidney deficiency, and quercetin and kaempferol were isolated from the fruits of R. corchorifolius for the first time.
