Abstract:Previous work has identified characteristic neural signatures of value-based decision-making, including neural dynamics that closely resemble the ramping evidence accumulation process believed to underpin choice. Here, we test whether these signatures of the choice process can be temporally dissociated from additional, choice- value signals. Indeed, EEG activity during value-based choice revealed distinct spatiotemporal clusters, with a stimulus-locked cluster reflecting affective reactions to choice sets and a response-locked cluster reflecting choice difficulty. Surprisingly, of these clusters met the criteria for an evidence accumulation signal. Instead, we found that stimulus-locked activity can an evidence accumulation process when aligned to the response. Re-analyzing four previous studies – including three perceptual decision-making studies – we show that response-locked signatures of evidence accumulation disappear when stimulus-locked and response-locked activity are modelled jointly. Collectively, our findings show that neural signatures of value can reflect choice-independent processes and look deceptively like evidence accumulation.

Abstract:The spontaneous activity of the brain interacts with stimulus-induced activity which is manifested in event-related amplitude and its trial-to-trial variability (TTV). TTV describes the variability in the amplitude of the stimulus-evoked response across trials, and it is generally observed to be reduced, or quenched. While such TTV quenching has been observed on both the cellular and regional levels, its exact behavioral relevance and neuronal basis remains unclear. Applying a novel paradigm for testing neural markers of individuality in internally-guided decision-making, we here investigated whether TTV (i) represents an individually specific response by comparing individualized vs shared stimuli; and (ii) is mediated by the complexity of prestimulus activity as measured by the Lempel-Ziv Complexity index (LZC). We observed that TTV - and other electrophysiological markers such as ERP, ERSP, and ITC - showed first significant differences between individualized and shared stimuli (while controlling for task-related effects) specifically in the alpha and beta frequency bands, and secondly that TTV in the beta band correlated significantly with reaction time and eLORETA activity. Moreover, we demonstrate that the complexity (LZC) of neuronal activity is higher in the prestimulus period while it decreases during the poststimulus period, with the former also correlating specifically with poststimulus individualized TTV in alpha (but not with shared TTV). Together, our results show that the TTV represents a marker of 'neural individualization' which, being related to internal processes on both neural and psychological levels, is mediated by the information complexity of prestimulus activity. More generally, our results inform the pre-post-stimulus dynamics of rest-stimulus interaction, which is a basic and ubiquitous neural phenomenon in the brain and highly relevant for mental features including their individuality.
Value construction through sequential sampling explains serial dependencies in decision making

Abstract:Many decisions are expressed as a preference for one item over another. When these items are familiar, it is often assumed that the decision maker assigns a value to each of the items and chooses the item with the highest value. These values may be imperfectly recalled, but are assumed to be stable over the course of an interview or psychological experiment. Choices that are inconsistent with a stated valuation are thought to occur because of unspecified noise that corrupts the neural representation of value. Assuming that the noise is uncorrelated over time, the pattern of choices and response times in value-based decisions are modeled within the framework of Bounded Evidence Accumulation (BEA), similar to that used in perceptual decision-making. In BEA, noisy evidence samples accumulate over time until the accumulated evidence for one of the options reaches a threshold. Here, we argue that the assumption of temporally uncorrelated noise, while reasonable for perceptual decisions, is not reasonable for value-based decisions. Subjective values depend on the internal state of the decision maker, including their desires, needs, priorities, attentional state, and goals. These internal states may change over time, or undergo revaluation, as will the subjective values. We reasoned that these hypothetical value changes should be detectable in the pattern of choices made over a sequence of decisions. We reanalyzed data from a well-studied task in which participants were presented with pairs of snacks and asked to choose the one they preferred. Using a novel algorithm (Reval), we show that the subjective value of the items changes significantly during a short experimental session (about 1 hour). Values derived with Reval explain choice and response time better than explicitly stated values. They also better explain the BOLD signal in the ventromedial prefrontal cortex, known to represent the value of decision alternatives. Revaluation is also observed in a BEA model in which successive evidence samples are not assumed to be independent. We argue that revaluation is a consequence of the process by which values are constructed during deliberation to resolve preference choices.

Abstract:Accumulating evidence to make decisions is a core cognitive function. Previous studies have tended to estimate accumulation using either neural or behavioral data alone. Here we develop a unified framework for modeling stimulus-driven behavior and multi-neuron activity simultaneously. We applied our method to choices and neural recordings from three rat brain regions — the posterior parietal cortex (PPC), the frontal orienting fields (FOF), and the anterior-dorsal striatum (ADS) — while subjects performed a pulse-based accumulation task. Each region was best described by a distinct accumulation model, which all differed from the model that best described the animal's choices. FOF activity was consistent with an accumulator where early evidence was favored while the ADS reflected near perfect accumulation. Neural responses within an accumulation framework unveiled a distinct association between each brain region and choice. Choices were better predicted from all regions using a comprehensive, accumulation-based framework and different brain regions were found to differentially reflect choice-related accumulation signals: FOF and ADS both reflected choice but ADS showed more instances of decision vacillation. Previous studies relating neural data to behaviorally-inferred accumulation dynamics have implicitly assumed that individual brain regions reflect the whole-animal level accumulator. Our results suggest that different brain regions represent accumulated evidence in dramatically different ways and that accumulation at the whole-animal level may be constructed from a variety of neural-level accumulators.

Abstract:Making adaptive choices in dynamic environments requires flexible decision policies. Previously, we showed how shifts in outcome contingency change the evidence accumulation process that determines decision policies. Using in silico experiments to generate predictions, here we show how the cortico-basal ganglia-thalamic (CBGT) circuits can feasibly implement shifts in decision policies. When action contingencies change, dopaminergic plasticity redirects the balance of power, both within and between action representations, to divert the flow of evidence from one option to another. When competition between action representations is highest, the rate of evidence accumulation is the lowest. This prediction was validated in in vivo experiments on human participants, using fMRI, which showed that (1) evoked hemodynamic responses can reliably predict trial-wise choices and (2) competition between action representations, measured using a classifier model, tracked with changes in the rate of evidence accumulation. These results paint a holistic picture of how CBGT circuits manage and adapt the evidence accumulation process in mammals.

Abstract:Neural representations of perceptual decision formation that are abstracted from specific motor requirements have previously been identified in humans using non-invasive electrophysiology; however, it is currently unclear where these originate in the brain. Here we capitalized on the high spatiotemporal precision of intracranial EEG to localize such abstract decision signals. Participants undergoing invasive electrophysiological monitoring for epilepsy were asked to judge the direction of random-dot stimuli and respond either with a speeded button press ( N = 24), or vocally, after a randomized delay ( N = 12). We found a widely distributed motor-independent network of regions where high-frequency activity exhibited key characteristics consistent with evidence accumulation, including a gradual buildup that was modulated by the strength of the sensory evidence, and an amplitude that predicted participants' choice accuracy and response time. Our findings offer a new view on the brain networks governing human decision-making.

The root of all value: a neural common currency for choice

Abstract:How do humans make choices between different types of rewards? Economists have long argued on theoretical grounds that humans typically make these choices as if the values of the options they consider have been mapped to a single common scale for comparison. Neuroimaging studies in humans have recently begun to suggest the existence of a small group of specific brain sites that appear to encode the subjective values of different types of rewards on a neural common scale, almost exactly as predicted by theory. We have conducted a meta analysis using data from thirteen different functional magnetic resonance imaging studies published in recent years and we show that the principle brain area associated with this common representation is a subregion of the ventromedial prefrontal cortex (vmPFC)/orbitofrontal cortex (OFC). The data available today suggest that this common valuation path is a core system that participates in day-to-day decision making suggesting both a neurobiological foundation for standard economic theory and a tool for measuring preferences neurobiologically. Perhaps even more exciting is the possibility that our emerging understanding of the neural mechanisms for valuation and choice may provide fundamental insights into pathological choice behaviors like addiction, obesity and gambling.

Abstract:Decision making is traditionally thought to be mediated by neurons that accumulate evidence through persistent activity. However, recent decision-making experiments in rodents have observed neurons across the brain that fire sequentially, rather than persistently, with the subset of neurons in the sequence depending on the animal’s choice. We developed two new candidate circuit models in which neurons are active sequentially and transfer evidence faithfully to the next active population. One model encodes evidence in the relative firing of two competing chains of neurons, and the other in the network location of a stereotyped pattern (“bump”) of neural activity. Neural recordings from four brain regions during an evidence accumulation task revealed that different regions displayed evidence tuning consistent with different candidate models. This work provides a mechanistic explanation for how graded information may be precisely accumulated within and transferred between neural populations, a set of computations fundamental to many cognitive operations.

Challenges of Interpreting Frontal Neurons during Value-Based Decision-Making

Abstract:The frontal cortex is crucial to sound decision-making, and the activity of frontal neurons correlates with many aspects of a choice, including the reward value of options and outcomes. However, rewards are of high motivational significance and have widespread effects on neural activity. As such, many neural signals not directly involved in the decision process can correlate with reward value. With correlative techniques such as electrophysiological recording or functional neuroimaging, it can be challenging to distinguish neural signals underlying value-based decision-making from other perceptual, cognitive, and motor processes. In the first part of the paper, we examine how different value-related computations can potentially be confused. In particular, error-related signals in the anterior cingulate cortex, generated when one discovers the consequences of an action, might actually represent violations of outcome expectation, rather than errors per se. Also, signals generated at the time of choice are typically interpreted as reflecting predictions regarding the outcomes associated with the different choice alternatives. However, these signals could instead reflect comparisons between the presented choice options and previously presented choice alternatives. In the second part of the paper, we examine how value signals have been successfully dissociated from saliency-related signals, such as attention, arousal, and motor preparation in studies employing outcomes with both positive and negative valence. We hope that highlighting these issues will prove useful for future studies aimed at disambiguating the contribution of different neuronal populations to choice behavior.

Abstract:Perceptual decision-making is affected by uncertainty arising from the reliability of incoming sensory evidence (perceptual uncertainty) and the categorization of that evidence relative to a choice boundary (categorical uncertainty). Here, we investigated how these factors impact the temporal dynamics of evidence processing during decision-making and subsequent metacognitive judgments. Participants performed a motion discrimination task while electroencephalography was recorded. We manipulated perceptual uncertainty by varying motion coherence, and categorical uncertainty by varying the angular offset of motion signals relative to a criterion. After each trial, participants rated their desire to change their mind. High uncertainty impaired perceptual and metacognitive judgments and reduced the amplitude of the centro-parietal positivity, a neural marker of evidence accumulation. Coherence and offset affected the centro-parietal positivity at different time points, suggesting that perceptual and categorical uncertainty affect decision-making in sequential stages. Moreover, the centro-parietal positivity predicted participants' metacognitive judgments: larger predecisional centro-parietal positivity amplitude was associated with less desire to change one's mind, whereas larger postdecisional centro-parietal positivity amplitude was associated with greater desire to change one's mind, but only following errors. These findings reveal a dissociation between predecisional and postdecisional evidence processing, suggesting that the CPP tracks potentially distinct cognitive processes before and after a decision.

Abstract:A core assumption in decision neuroscience is that individuals decide between options by comparing option-specific subjective reward values. Psychological accounts challenge this view and suggest that decisions are better explained by comparisons between choice attributes than by comparisons between option-specific values, casting doubts on the interpretation of activation in the neural reward system as subjective value signals. Here, we provide neuroimaging and pharmacological evidence that value-related neural activity follows the value comparison strategy employed by an individual on the psychological level. Neural model comparisons reveal that activation in the striatum, rather than generally reflecting attribute-wise or option-wise value comparisons, reflects the value comparison strategy that provides the best explanation for an individual's choice behavior. Strikingly, manipulating activation in the dopaminergic reward system reveals that dopamine antagonism counteracts the engagement in an individual's dominant value comparison strategy. Together, our findings provide evidence for the biological plausibility of psychological accounts of decision making and emphasize the importance of neural model comparisons to prevent misinterpretations of brain activation.

Abstract:Frömer et al (2024, Nature Human Behaviour) apply a deconvolution method to correct for component overlap in the event-related potential. They report that this method eliminates signatures of sensory evidence accumulation from response-aligned measurements of the centro-parietal positivity (CPP), suggesting that these signatures arise artifactually. Here, we argue that the analysis and interpretation of their perceptual choice data are critically flawed. We demonstrate with simulations that the deconvolution analyses used by the authors are not designed to reliably test for the presence or absence of bounded accumulation signals.

Abstract:To investigate the mechanisms through which economic decisions are formed, I examined the activity of neurons in the orbitofrontal cortex while monkeys chose between different juice types. Different classes of cells encoded the value of individual offers (offer value), the value of the chosen option (chosen value), or the identity of the chosen juice (chosen juice). Choice variability was partly explained by the tendency to repeat choices (choice hysteresis). Surprisingly, near-indifference decisions did not reflect fluctuations in the activity of offer value cells. In contrast, near-indifference decisions correlated with fluctuations in the preoffer activity of chosen juice cells. After the offer, the activity of chosen juice cells reflected the decision difficulty but did not resemble a race-to-threshold. Finally, chosen value cells presented an "activity overshooting" closely related to the decision difficulty and possibly due to fluctuations in the relative value of the juices. This overshooting was independent of choice hysteresis.

Abstract:The frontal cortex-striatum circuit plays a pivotal role in adaptive goal-directed behaviours. However, the mediation of decision-related signals through cross-regional transmission between the medial frontal cortex and the striatum by neuronal ensembles remains unclear. We analysed neuronal ensemble activity obtained through simultaneous multiunit recordings in the secondary motor cortex (M2) and dorsal striatum (DS) while the rats performed an outcome-based choice task. Tensor component analysis (TCA), an unsupervised dimensionality reduction approach at the single-trial level, was adopted for concatenated ensembles of M2 and DS neurons. We identified distinct three spatiotemporal neural dynamics (TCA components) at the single-trial level specific to task-relevant variables. Choice-position selective neural dynamics was correlated with the trial-to-trial fluctuation of behavioural variables. This analytical approach unveiled choice-pattern selective neural dynamics distinguishing whether the incoming choice was a repetition or switch from the previous choice. Other neural dynamics was selective to outcome. Choice-pattern selective within-trial activity increased before response choice, whereas outcome selective within-trial activity increased following response. These results suggest that the concatenated ensembles of M2 and DS process distinct features of decision-related signals at various points in time. The M2 and DS may collaboratively monitor action outcomes and determine the subsequent choice, whether to repeat or switch, for coordinated action selection.

Abstract:Neurobiological investigations of perceptual decision-making have furnished the first glimpse of a flexible cognitive process at the level of single neurons. Neurons in the parietal and prefrontal cortex are thought to represent the accumulation of noisy evidence, acquired over time, leading to a decision. Neural recordings averaged over many decisions have provided support for the deterministic rise in activity to a termination bound. Critically, it is the unobserved stochastic component that is thought to confer variability in both choice and decision time. Here, we elucidate this drift-diffusion signal on individual decisions. We recorded simultaneously from hundreds of neurons in the lateral intraparietal cortex of monkeys while they made decisions about the direction of random dot motion. We show that a single scalar quantity, derived from the weighted sum of the population activity, represents a combination of deterministic drift and stochastic diffusion. Moreover, we provide direct support for the hypothesis that this drift-diffusion signal approximates the quantity responsible for the variability in choice and reaction times. The population-derived signals rely on a small subset of neurons with response fields that overlap the choice targets. These neurons represent the integral of noisy evidence. Another subset of direction-selective neurons with response fields that overlap the motion stimulus appear to represent the integrand. This parsimonious architecture would escape detection by state-space analyses, absent a clear hypothesis.

Abstract:Economic choice theories usually assume that humans maximize utility in their choices. However, studies have shown that humans make inconsistent choices, leading to suboptimal behavior, even without context dependent manipulations. Previous studies showed that activation in value and motor networks are associated with inconsistent choices at the moment of choice. Here we investigated if the neural predispositions, measured before a choice task, can predict choice inconsistency in a later risk choice task. Using functional connectivity (FC) measures from resting-state functional magnetic resonance imaging (rsfMRI), derived before any choice was made, we aimed to predict subjects’ inconsistency levels in a later-performed choice task. We hypothesized that rsfMRI FC measures extracted from value and motor brain areas would predict inconsistency. Forty subjects (21 females) completed a rsfMRI scan before performing a risky-choice task. We compared models that were trained on FC that included only hypothesized value and motor regions with models trained on whole-brain FC. We found that both model types significantly predicted inconsistency levels. Moreover, even the whole-brain models relied mostly on FC between value and motor areas. For external validation, we used a neural network pre-trained on FC matrices of 40,000 subjects and fine-tuned it on our data, and again showed significant predictions. Together, this shows that the tendency for choice inconsistency is predicted by predispositions of the nervous system, and that synchrony between the motor and value networks plays a crucial role in this tendency. Significance Statement Humans often make irrational, or inconsistent, choices in value-based decision-making. Studies have suggested potential sources for this suboptimal behavior in momentary brain activity during choice, emphasizing the role of value and motor areas in the process of inconsistent choice behavior. Here, we show that inconsistency is predicted by neural predispositions, evident even before any choice was made. We predict subjects’ inconsistency levels from brain connectivity measures derived before they perform any choice task and find that inconsistency is predicted by the synchrony between the value and motor brain networks. This suggests choice inconsistency is related to a-priori constraints of the nervous system, and that value and motor areas play a central role in this behavior.
