Abstract:A fundamental step in gene-regulatory activities, such as repression, transcription, and recombination, is the binding of regulatory DNA-binding proteins (DBPs) to specific targets in the genome. To rapidly localize their regulatory genomic sites, DBPs reduce the dimensionality of the search space by combining three-dimensional (3D) diffusion in solution with one-dimensional (1D) sliding along DNA. However, the requirement to form a thermodynamically stable protein-DNA complex at the cognate genomic target sequence imposes a challenge on the protein because, as it navigates one-dimensionally along the genome, it may come in close contact with sites that share partial or even complete sequence similarity with the functional DNA sequence. This puzzling issue creates a conflict between two basic requirements: finding the cognate site quickly and stably binding it. Here, we structurally assessed the interface adopted by a variety of DBPs to bind DNA specifically and nonspecifically, and found that many DBPs utilize one interface to specifically recognize a DNA sequence and another to assist in propagating along the DNA through nonspecific associations. While these two interfaces overlap each other in some proteins, they present partial overlap in others and frustrate the protein-DNA interface. Using coarse-grained molecular dynamics simulations, we demonstrate that the existence of frustration in DBPs is a compromise between rapid 1D diffusion along other regions in the genome (high frustration smoothens the landscape for sliding) and rapid formation of a stable and essentially active protein-DNA complex (low frustration reduces the free energy barrier for switching between the two binding modes).
Atmospheric cold plasma pretreatment for effective enhancement of covalent crosslinking between coconut globulin and tannic acid: Improving interfacial activity and emulsifying properties

Abstract:Atmospheric cold plasma (ACP) represents a promising approach for enhancing covalent interactions between proteins and polyphenols, circumventing the drawbacks associated with traditional methods. This study aims to investigate the enhancement of covalent interactions between coconut globulin (CG) and tannic acid (TA) facilitated by ACP at varying pH levels. At acidic pH, ACP treatment was found to promote free radical-induced covalent cross-linking between CG and TA, whereas at pH 7.0 and 9.0, ACP treatment enhanced quinone-induced covalent cross-linking. In contrast, the covalent crosslinking induced by quinone significantly disrupted the protein structure, leading to greater exposure of hydrophobic groups. At pH 9.0, the CG-TA complex treated with ACP exhibited the highest interfacial activity, with an interfacial adsorption mass of 5292 ng/cm2. This was accompanied by improvements in droplet size, viscosity, and stability of the CG-TA-stabilized emulsion. These findings offer novel insights into the covalent modification of proteins and polyphenols, thereby broadening the potential applications of food protein.
Investigating binding mechanism between coconut globulin and tannic acid mediated by atmospheric cold plasma: Protein structure and stability

Abstract:Physical methods present promising avenues for inducing covalent modifications of proteins by polyphenols, circumventing the safety and sustainability issues associated with traditional approaches. This study sought to enhance the physicochemical properties of coconut globulin (CG) by facilitating covalent cross-linking with tannic acid (TA) through atmospheric cold plasma (ACP). The ACP treatment effectively transitioned the interaction between CG and TA from non-covalent to covalent in a voltage-dependent manner at pH 6.0, resulting in structural modifications of CG. The treatment with TA enhanced the spherical structure of CG, with a reduction in particle size from 474 to 384 nm. This size reduction was further amplified by the exposure of charged groups induced by ACP treatment. Consequently, the solubility, surface hydrophobicity, and viscosity of ACP-treated CG-TA increased, leading to an elevated denaturation temperature and enhanced physical stability. These results suggest a viable approach to improving the suboptimal physicochemical properties of plant proteins.

Abstract:Coconut palm (Cocos nucifera L.) is rich in protein and consumed worldwide, coconut protein as a plant protein contains many nutrients and is beneficial for human health, but its allergic components also need to be considered. Therefore, further research on coconut protein is of great significance. In this study, nine proteins were identified through matrix-assisted laser desorption/ionization tandem time-of-flight mass spectrometry (MALDI-TOF/TOF-MS). Three proteins, namely, (1) 63 kDa globulin-like protein [Elaeis guineensis], (2) 63 kDa globulin-like protein [Phoenix dactylifera], and (3) glutelin type-A 3-like [E. guineensis] were initially found in coconut proteins. To further purify and identify the coconut protein, we used dialysate membrane and high-performance liquid chromatography-Ultraviole (HPLC-UV) to concentrate and separate the extracted protein. Two types of 63 kDa globulins were further purified and identified, namely, P. dactylifera and E. guineensis proteins with 564 and 398 amino acids, respectively, were identified. SWISS-MODEL was used to predict the tertiary structure of the 63 kDa globulins and showed that the structures of the coconut proteins were mostly similar to those of vicilin. This work provided a feasible and simple separation approach for the isolation and purification of coconut protein, and predicts the structure of newly identified protein.

Combination of Structural Analysis and Proteomics Strategy Revealed the Mechanism of Ultrasound-Assisted Cold Plasma Regulating Shrimp Allergy

Abstract:Allergic incidents of crustacean aquatic products occur frequently, and tropomyosin (TM) is the main allergen. Therefore, it is worthwhile to develop technologies to efficiently reduce the allergenicity of TM. In this study, ultrasound-assisted cold plasma (UCP) treatment was used to regulate shrimp allergy. The remarkable changes in TM structure were substantiated by alteration in secondary structure, reduction in sulfhydryl content, change in surface hydrophobicity, and disparity in surface morphology. The IgE and IgG binding ability of TM significantly decreased by 52.40% and 46.51% due to UCP treatment. In the Balb/c mouse model, mice in the UCP group showed most prominent mitigation of allergic symptoms, proved by lower allergy score, changes in levels of TM-specific antibodies, and restoration of Th1/Th2 cytokine imbalance. Using a proteomics approach, 439 differentially expressed proteins (DEPs) in the TM group (vs phosphate-buffered saline group) and 170 DEPs in the UCP group (vs TM group) were determined. Subsequent analysis demonstrated that Col6a5, Col6a6, and Epx were potential biomarkers of TM allergy. Moreover, Col6a5, Col6a6, Dcn, and Kng1 might be the target proteins of UCP treatment, while PI3K/Akt/mTOR might be the regulated signaling pathway. These findings proved that UCP treatment has great potential in reducing TM allergenicity and provide new insights into the development of hypoallergenic shrimp products.
Dietary Polyphenols Reduced the Allergenicity of Β-Lactoglobulin Via Non-Covalent Interactions: a Study on the Structure-Allergenicity Relationship

Abstract:Studies showed that complexation of polyphenols with milk allergens reduced their immunogenic potential. However, the relationship between structures of polyphenols and their hypoallergenic effects on milk allergens in association with physiological and conformational changes of the complexes remain unclear. In this study, polyphenols from eight botanical sources were extracted to prepare non-covalent complexes with β-lactoglobulin(β-LG), a major allergen in milk. The dominant phenolic compounds bound to β-LG with a diminished allergenicity were identified to investigate their respective role on the structural and allergenic properties of β-LG. Extracts from Vaccinium fruits and black soybeans were found to have great inhibitory effects on the IgE-and IgG-binding abilities of β-LG. Among the fourteen structure-related phenolic compounds, flavonoids and tannins with larger MWs and multi-hydroxyl substituents, notably rutin, EGCG, and ellagitannins were more potent to elicit changes on the conformational structures of β-LG to decrease the allergenicity of complexed β-LG. Correlation analysis further demonstrated that a destabilized secondary structure and protein depolymerization caused by polyphenol-binding were closely related to the allergenicity property of formed complexes. This study provides insights into the understanding of structure-allergenicity relationship of β-LG-polyphenol interactions and would benefit the development of polyphenol-fortified matrices with hypoallergenic potential.
Evaluation of allergenicity of cow milk treated with enzymatic hydrolysis through a mouse model of allergy

Abstract:Cow milk (CM) allergy is a worldwide concern. Currently, few studies have been performed on the immunoreactivity of CM and fewer still on the antigenicity of CM in vivo and in vitro. In this study, we assessed the potential allergenicity of enzymatically hydrolyzed CM using in vitro ELISA and oral sensitization and challenge of BALB/c mice. Alcalase-, Protamex-, and Flavourzyme-treated CM (all from Novozymes) diminished IgE binding capacity, with greatest reductions of 56.31%, 50.62%, and 56.45%, respectively. Allergic symptoms and levels of total IgG1 were reduced, and allergic inflammation of the lung, jejunum, and spleen was relieved. Moreover, the numbers of CD8+ T and B220+ cells decreased, and the balance of CD4+ T/CD8+ T cells was effectively regulated. These findings suggest that the potential allergenicity of CM was reduced by enzymatic hydrolysis, and our research will lay a solid foundation for developing high-quality hypoallergenic CM products.

Abstract:In this study, monosodium glutamate (MSG) was used to improve the viscosity of coconut milk and the underlying mechanism was explored by investigating the changes in structures of coconut milk protein and physicochemical properties of coconut milk. Firstly, the effect of MSG on the properties of coconut milk was studied. The results showed that MSG increased the pH and zeta potential, reduced the particle size, thus enhancing the droplet interaction and increasing the viscosity of coconut milk. Subsequently, the effects of MSG on the structure and properties of coconut proteins (CP) were investigated. FTIR spectroscopy and circular dichroism spectroscopy showed that MSG was able to change the secondary structure of CP. The results of SDS-PAGE showed that MSG was able to bind to CP to form a larger molecular weight protein, thus improving the viscosity of coconut milk. Moreover, MSG was also able to increase the water-binding capacity of CP. In addition, molecular docking and driving force analysis revealed that hydrogen bonds, electrostatic forces, disulfide bonds, and hydrophobic interactions are the main interactions between MSG and CP. Studying the effect of MSG on the viscosity of coconut milk provides theoretical support to improve the viscosity of other plant protein emulsions.

Abstract:Allergies have become a global problem, and effective control is greatly needed. Here, the inactivation effects of the atmospheric pressure cold plasma (APCP) on aerosolized allergens including Der p 1, Der f 1, Asp f 1, Alt a 1, and Can f 1 as well as those from indoor and outdoor environments were investigated. The effectiveness of the APCP treatment was further studied using blood sera from the allergen sensitized humans. In addition, the allergen samples were also analyzed using sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE). Results revealed that the APCP was highly effective in reducing the allergenicity of both lab-prepared and environmental allergen aerosols. The airborne reductions were shown to range from 30% for Der p 1 to 80% for Can f 1 allergen for 0.12 s exposure. Allergnicity tests showed that the APCP treated Asp f 1 allergens caused 50% less binding with IgEs in the blood sera compared to the control. The observed allergenicity loss was due to hydroxyl radicals produced by the plasma device. The results from SDS-PAGE showed that the plasma treatment resulted in decreased size of the Asp f 1 allergen. The developed technology holds great promise in combating the allergic diseases.
The role of fat content in coconut milk: Stability and digestive properties

Abstract:The fat in coconut milk contributes to unique flavour, while increasing fat content affects stability of the coconut milk. In this study, coconut water and fat were separated, recombined, and homogenized to obtain coconut milk with different fat contents (0-20 %). Emulsifying properties, stability, and digestibility of coconut milk with different fat contents were comprehensively evaluated. The results showed that as the fat content increased from 0 to 20 %, the droplet size increased from 2.18 to 4.70 μm and the viscosity showed an increasing trend. During storage and freeze-thaw, coconut milk with 5 % and 10 % fat content showed excellent stability. In addition, coconut milk with 10 % fat content had superior fat digestibility, which was related to high affinity of pancrelipase. In short, this study revealed that fat content below 10 % can withstand environmental factors such as storage, lipid oxidation, and freeze-thaw, which can be accurately developed as coconut milk products.

Abstract:Background Milk allergy is a global health issue, which affects the quality of life and can even be life threatening. According to epidemiological investigation, 2%–6% of infants are sensitive to beta-lactoglobulin (β-LG) in milk. Avoiding allergens is still the first-rank option for vulnerable group because immunization therapy is hardly available. Therefore, exploring the allergic mechanism of β-LG and applying appropriate antiallergic treatments are the key to solving the problem of milk allergy. Scope and approach This review describes the occurrence of β-LG, including the type, structure, epitopes of β-LG, the performance of gastrointestinal digestion, the mechanism and allergy consequence of β-LG allergic responses. Then, various methods to reduce the sensitization of β-LG were summarized, including covalent and non-covalent interactions, non-thermal processing technologies (fermentation, irradiation, pulsed electric field, ultrasound), thermal processing technology (microwave), and combination therapies. Key findings and conclusions : β-LG allergy is an immediate type I hypersensitivity reaction involving immunoglobulin (IgE) dependent antibody responses that cause a variety of clinical symptoms, such as asthma, eczema, gastrointestinal disorders, and anaphylactic shock in severe cases. To reduce the allergenicity of the β-LG, it is necessary to adopt novel processing strategies with higher efficiency. This review covers the covalent or non-covalent binding of polyphenols, polysaccharides, metal ions and nanoparticles to β-LG, as well as emerging technologies, and finds that structural modifications and conformational epitope changes of β-LG are associated with immunomodulatory properties. The combination of various approaches is a potential new way to reduce β-LG hypersensitivity.

Lipid matrix-specific pretreatment method for enhancing the extractability and allergenicity maintenance of bovine milk allergens in ELISA detection

Abstract:The presence of various components in the food matrix makes allergen detection difficult and inaccurate, and pretreatment is an innovative breakthrough point. Food matrices were categorised based on their composition. Subsequently, a pretreatment method was established using a combination of ultrasound-assisted n-hexane degreasing and weakly alkaline extraction systems to enhance the detection accuracy of bovine milk allergens. Results showed that more allergens were obtained with less structural destruction, as demonstrated using immunological quantification and spectral analysis. Concurrently, allergenicity preservation was confirmed through liquid chromatography-tandem mass spectrometry (LC-MS/MS) analysis, a KU812 cell degranulation model, and western blotting. The method exhibited good accuracy (bias, 8.47%), repeatability (RSDr, 1.52%), and stability (RSDR, 5.65%). In foods with high lipid content, such as chocolate, the allergen content was 2.29-fold higher than that of commercial kits. Laser confocal scanning microscopy (LCSM) and scanning electron microscopy (SEM) analyses revealed a significant decrease in fat content after post-pretreatment using our method. In addition, colloidal stability surpassed that achieved using commercial kits, as indicated through the PSA and zeta potential results. The results demonstrated the superiority of the extractability and allergenicity maintenance of lipid matrix-specific pretreatment methods for improving the accuracy of ELISA based allergen detection in real food.
Analysis of the glycosylation products of peanut protein and lactose by cold plasma treatment: Solubility and structural characteristics

Abstract:Cold plasma (CP) treatment was used to prepare the glycosylation conjugates of high-temperature peanut protein isolate (HPPI) and lactose to improve the solubility of HPPI. We observed that by increasing the CP treatment time to 3 min, the solubility of the conjugates increased to 1.34 mg/mL. An increase in the degree of glycosylation and a decrease in the degree of browning indicated that although CP treatment accelerated the glycosylation of HPPI and lactose, it interfered with the formation of melanoidin. The analysis of protein tertiary structure showed that tryptophan and tyrosine residues in proteins undergoing CP treatment were the primary sites for the Maillard reaction. The relative decrease in surface hydrophobicity and FT-IR analysis indicated that the increase in the -OH stretching vibration intensity on the protein surface represented the formation of the covalent bonds between HPPI and lactose during the CP treatment. An increase in the denaturation temperature of proteins was observed after grafting with lactose. Changes in the secondary structure and surface structure of proteins showed that lactose covalently bonded to the surface of HPPI during CP treatment, forming a more stable ordered structure.

Abstract:This study aims at the production of coconut milk as a substitute for milk from cows (dairy milk) using mature coconut. The purpose of this milk is to address the challenges associated with allergies, lactose intolerance, and the prevalence of hypercholesterolemia. The milk was obtained from coconut by wet extraction. The matured coconut fruit was deshelled, pared, and fragmented. It was pasteurized at 62.8°C for 30 mins. The results obtained for energy in calories, total fat content, pH, water, total sugars, reducing sugars, and sucrose are as follows; 135.94 kcal, 14.12 g, 790.33 g, 0.70 g, 0.15 g, and 0.55 g, respectively. Additionally, the derived results for protein content, total solids, ash, total carbohydrate, milk solids non-fat, calcium, and vitamin are as follows; 2.22 g, 9.67 g, 0.83 g, 0.89 g, 1.99 g, 92.46 mg, and 18.59 mg, respectively. The total phenolic content was found to be between 295.8333 and 312.5 mg GAE/L and antioxidant capacity value ranged from 412.5 to 437.5 mg vitamin C EQ/L.
Advantages of UHT in retaining coconut milk aroma and insights into thermal changes of aroma compounds

Abstract:Coconut milk products are susceptible to bacterial damage, necessitating sterilization methods that often compromise nutrient and aroma integrity. This study investigates the effects of different thermal sterilisation methods on coconut milk aroma using headspace gas chromatography-ion mobility spectrometry (HS-GC-IMS) and headspace solid-phase microextraction gas chromatography-mass spectrometry (HS-SPME-GC-MS). We assessed the impact of pasteurisation (PAS, 70 °C, 25 min), high-temperature sterilisation (HTS, 121.1 °C, 15 min), and ultra-high temperature sterilisation (UHT, 130 °C, 5 s) through clustered heat maps and correlation analyses. Significant differences were observed (p < 0.05), with 37 and 52 substances detected by HS-GC-IMS and HS-SPME-GC-MS, respectively, identifying 12 key aroma compounds. UHT treatment primarily reduced 8 acids, maintaining a compositional structure and sensory profile similar to raw coconut milk. PAS and HTS treatments decreased the sensory intensity of overall coconut milk aroma, creamy, and floral notes, correlating with the presence of 2-heptanol, nonanal, 4-methylvaleric acid, and 2-tridecanone. These methods increased cooked notes, associated with 5-methyl-3-heptanone, 3-butyn-1-ol, hydroxyacetone, and acetoin. Rancidity was linked to acids such as isobutyric acid, isovaleric acid, and heptanoic acid, with high temperatures effectively reducing these compounds. Prolonged temperature changes in PAS and HTS accelerated lipid oxidative degradation and the Maillard reaction, involving free fatty acids in the formation of alcohols, aldehydes, esters, and lactones. These findings provide a theoretical basis for studying coconut milk flavour deterioration.
Allergenicity evaluation of an extensively hydrolyzed infant formula based on cow milk protein

Abstract:Cow milk protein allergy (CMPA) is the most common food allergy in infants. Currently, hypoallergenic infant formulas on the market are mainly divided into extensively hydrolyzed whey protein formulas and extensively hydrolyzed casein formulas. There are few extensively hydrolyzed infant formulas (EHFs) with a similar protein composition to breast milk. Therefore, we developed a hypoallergenic infant formula based on extensively hydrolyzed cow milk protein (whey protein-to-casein ratio of 6 : 4) and evaluated its allergenicity in vitro and in vivo. The results showed that the antigenicity of EHF was significantly decreased. The levels of Treg and Th1 cells were increased, while the levels of Th2 cells, IgE and IgG1, plasma histamine and serum mast cell enzymes were significantly decreased. At the same time, the allergic symptoms of the jejunum and lungs of mice were relieved. This study provides a solution for the development of cow milk protein based hypoallergenic infant formulas.
Cold Plasma Technology: A Sustainable Approach to Milk Preservation by Reducing Pathogens and Enhancing Oxidative Stability

Abstract:Pathogenic microorganisms and lipid oxidation are critical challenges in the dairy industry, influencing both food safety and quality. This study explores the potential of cold plasma (CP) technology as a sustainable alternative for milk preservation compared to conventional pasteurization. CP treatment utilizes ionized gas to generate reactive species, which effectively disrupt microbial cell membranes and inactivate pathogens, thereby sterilizing the milk. We assessed raw, pasteurized, and cold plasma-treated milk samples, focusing on microbial growth, lipid oxidation, and oxidative stability. Our findings indicate that CP treatment significantly reduced microbial contamination, effectively inhibiting the growth of pathogenic bacteria and delaying acidity development in milk. In contrast, pasteurized milk exhibited a notable increase in peroxide values, indicating lipid deterioration. Furthermore, the oxidative stability of cold plasma-treated milk was enhanced, with an induction period extending from approximately five to seven hours, demonstrating its superior resistance to oxidation. In conclusion, CP has emerged as a promising eco-friendly technology for prolonging the shelf life of milk by mitigating microbial growth and lipid oxidation. This method not only aligns with sustainability goals by reducing the need for chemical preservatives but also enhances the overall quality of milk products. Future research should focus on large-scale applications and the impacts of CP on other essential milk components, particularly fat-soluble vitamins, to fully understand its sustainability benefits in the dairy sector.
