Abstract:Physical methods present promising avenues for inducing covalent modifications of proteins by polyphenols, circumventing the safety and sustainability issues associated with traditional approaches. This study sought to enhance the physicochemical properties of coconut globulin (CG) by facilitating covalent cross-linking with tannic acid (TA) through atmospheric cold plasma (ACP). The ACP treatment effectively transitioned the interaction between CG and TA from non-covalent to covalent in a voltage-dependent manner at pH 6.0, resulting in structural modifications of CG. The treatment with TA enhanced the spherical structure of CG, with a reduction in particle size from 474 to 384 nm. This size reduction was further amplified by the exposure of charged groups induced by ACP treatment. Consequently, the solubility, surface hydrophobicity, and viscosity of ACP-treated CG-TA increased, leading to an elevated denaturation temperature and enhanced physical stability. These results suggest a viable approach to improving the suboptimal physicochemical properties of plant proteins.

Abstract:Atmospheric cold plasma (ACP) represents a promising approach for enhancing covalent interactions between proteins and polyphenols, circumventing the drawbacks associated with traditional methods. This study aims to investigate the enhancement of covalent interactions between coconut globulin (CG) and tannic acid (TA) facilitated by ACP at varying pH levels. At acidic pH, ACP treatment was found to promote free radical-induced covalent cross-linking between CG and TA, whereas at pH 7.0 and 9.0, ACP treatment enhanced quinone-induced covalent cross-linking. In contrast, the covalent crosslinking induced by quinone significantly disrupted the protein structure, leading to greater exposure of hydrophobic groups. At pH 9.0, the CG-TA complex treated with ACP exhibited the highest interfacial activity, with an interfacial adsorption mass of 5292 ng/cm2. This was accompanied by improvements in droplet size, viscosity, and stability of the CG-TA-stabilized emulsion. These findings offer novel insights into the covalent modification of proteins and polyphenols, thereby broadening the potential applications of food protein.

Abstract:This work demonstrated the decomposition products of several amino acids when exposed to nonthermal atmospheric pressure plasma. A floating electrode dielectric barrier discharge plasma was used to treat 20 mM solutions of cysteine, glutamine, alanine, and lysine for varying treatment times to investigate possible mechanisms in cell wall degradation and treatment of tissue. The treated solutions were analyzed primarily using gas chromatography-mass spectrometry (GC-MS) and nuclear magnetic resonance (NMR). The primary targets of the plasma treatment were found to be the sulfur or nitrogen species. Acetic acid and lactic acid were observed throughout all solutions as oxidative products from the plasma treatment. This work suggests that the primary targets for plasma treatment of cell wall components is the amino acids.
Influence of atmospheric-pressure cold plasma-induced oxidation on the structure and functional properties of egg white protein

Abstract:This study examined the effects of atmospheric-pressure cold plasma (ACP) on the structure and functional properties of egg white protein (EWP). Egg white solution was treated with ACP for 0, 10, 20, 30, and 40 min. The structural unfolding of EWP was confirmed as the exposed free SH group content and surface hydrophobicity increased during the 30 min treatment; however, 40 min of ACP treatment showed the opposite tendency, suggesting the formation of protein aggregates. The emulsion stability of egg white emulsions was the highest at 30 min but decreased again at 40 min. In addition, the proportion of large droplets decreased as the treatment time increased up to 30 min. As a result, moderate oxidation was induced during 30 min of ACP treatment, which could contribute to the enhancement of functional properties of EWP. However, excessive oxidation caused by further ACP treatment negatively affected the enhanced functional properties. Industrial relevance Egg whites are used as functional ingredients for a variety of food products, so the quality and functionality of egg whites can directly affect the productivity of the final products. The present study suggests the moderate oxidation of egg white protein induced by atmospheric-pressure cold plasma, which could contribute to the enhancement of functional properties. These applied studies can serve as a cornerstone for the establishment of advanced oxidation techniques that can enhance the functionality of egg whites without chemical additives, and consequently contribute to the improvement of food quality and productivity.

Abstract:The present study explored the interaction and kinetics of cobalt ferrite nanoparticles (NPs) with acid phosphatase (ACP) by utilizing diverse range of spectroscopic techniques. The results corroborate, the CoFe2O4 NPs cause fluorescence quenching in ACP by static quenching mechanism. The negative values of van't Hoff thermodynamic expressions (Delta H= -0.3293 J mol(-1)K(-1) and Delta G= -3.960 kJ mol(-1)K(-1)) corroborate the spontaneity and exothermic nature of static quenching. The positive value of Delta S (13.2893 J mol(-1)K(-1)) corroborate that major contributors of higher and stronger binding affinity among CoFe2O4 NPs with ACP were electrostatic. In addition, FTIR, UV-CD, UV-vis spectroscopy and three dimensional fluorescence (3D) techniques confirmed that CoFe2O4 NPs binding induces microenvironment perturbations leading to secondary and tertiary conformation changes in ACP to a great extent. Furthermore, synchronous fluorescence spectroscopy (SFS) affirmed the comparatively significant changes in microenvironment around tryptophan (Trp) residue by CoFe2O4 NPs. The effect of CoFe2O4 NPs on the activation kinetics of ACP was further examined in Chlorella vulgaris. Apparent Michaelis constant (Km) values of 0.57 and 26.5 mM with activation energy values of 0.538 and 3.428 kj mol(-1) were determined without and with 200 mu M CoFe2O4 NPs. Apparent V-max value of -7 Umml(-1) corroborate that enzyme active sites were completely captured by the NPs leaving no space for the substrate. The results confirmed that CoFe2O4 NPs ceased the activity by unfolding of ACP enzyme. This suggests CoFe2O4 NPs perturbed the enzyme activity by transitions in conformation and hence the metabolic activity of ACP. Abstract:Atmospheric cold plasma (ACP) is an emerging technology which has increased attraction due to the consumers' tendency toward fresh and minimally processed food products. This non-thermal technology has been considered as a promising tool for decontamination of foods, modification of food components as well as food packaging. The potential interactions of cold plasma species with food components and consequently its effect on food quality is of high importance. Proteins are the main food constituent in food formulations regarding both nutritional and technological points of view. The susceptibility of native proteins to reactive species created through ACP treatment should be considered regarding the power supply, type of feeding gas and its pressure, exposure time, input voltage and current flow. However, the protein characteristics and the manner in which they are exposed are also important to be considered. This review article is aimed to investigate the technological and nutritional characteristics of proteins during atmospheric cold plasma treatment.

Abstract:The present study aims to investigate the structural and functional properties of oxidated walnut protein isolates (WPI) by 2,2′-azobis (2-amidinopropane) dihydrochloride (AAPH). The oxidation degree, changes in structural characteristics, processing properties, and protein modifications of WPI were measured. The results showed that oxidation significantly induced structural changes, mainly reflected by the increasing carbonyl content, and decreasing sulfhydryl and free amino groups. Moreover, the secondary structure of WPI was altered in response to oxidation, and large aggregates formed through disulfide cross-linking and hydrophobic interactions. Almost all the property indicators were significantly decreased by oxidation except the foaming property and water/oil holding capacity. Mass spectrometry analysis showed that 16 different modifications occurred in amino acid side chains, and most of the protein groups with higher numbers of modifications were found to be associated with allergies, which was further confirmed by the reduction in antigenicity of the major allergen (Jug r 1) in WPI. Meanwhile, we used oxidation-related proteins for gene ontology (GO) enrichment analyses, and the results indicated that 115, 204 and 59 GO terms were enriched in terms of biological process, molecular function, and cellular component, respectively. In conclusion, oxidation altered the groups and conformation of WPI, which in turn caused modification in the functional properties correspondingly. These findings might provide a reference for processing and storage of walnut protein foods.

Abstract:In addition to microbial spoilage and lipid peroxidation, protein oxidation is increasingly recognized as a major cause for quality deterioration of muscle-based foods. Although protein oxidation in muscle-based foods has attracted tremendous interest in the past decade, specific oxidative pathways and underlying mechanisms of protein oxidation in aquatic products remain largely unexplored. The present review covers the aspects of the origin and site-specific nature of protein oxidation, progress on the characterization of protein oxidation, oxidized proteins in aquatic products, and impact of protein oxidation on protein functionalities. Compared to meat protein oxidation, aquatic proteins demonstrate a less extent of oxidation on aromatic amino acids and are more susceptible to be indirectly oxidized by lipid peroxidation products. Different from traditional measurement of protein carbonyls and thiols, proteomics-based strategy better characterizes the targeted oxidation sites within proteins. The future trends using more robust and accurate targeted proteomics, such as parallel reaction monitoring strategy, to characterize protein oxidation in aquatic products are also given.

Abstract:Walnut protein is a key plant protein resource due to its high nutritional value, but walnuts are prone to oxidation during storage and processing. This article explored the oxidative modification and digestion mechanism of walnut protein isolates by peroxyl radical and obtained new findings. SDS-PAGE and spectral analysis were used to identify structural changes in the protein after oxidative modification, and LC-MS/MS was used to identify the digestion products. The findings demonstrated that as the AAPH concentration increased, protein carbonyl content increased from 2.36 to 5.12 nmol/mg, while free sulfhydryl content, free amino content, and surface hydrophobicity decreased from 4.30 nmol/mg, 1.47 μmol/mg, and 167.92 to 1.72 nmol/mg, 1.13 μmol/mg, and 40.93 nmol/mg, respectively. Furthermore, the result of Tricine-SDS-PAGE in vitro digestion revealed that protein oxidation could cause gastric digestion resistance and a tendency for intestinal digestion promotion. Carbonyl content increased dramatically during the early stages of gastric digestion and again after 90 min of intestine digestion, and LC-MS/MS identified the last digestive products of the stomach and intestine as essential seed storage proteins. Oxidation causes walnut proteins to form aggregates, which are then re-oxidized during digestion, and proper oxidative modification may benefit intestinal digestion.

Abstract:The aim of this study was to investigate the effect of 2,2'-azobis (2-amidinopropane) dihydrochloride (AAPH)-induced oxidation on the functional, structural properties and proteomic information of arachin. The results showed that moderate oxidation improved the water/oil holding capacity of proteins and increased the emulsifying stability, while excessive oxidation increased the carbonyl content, reduced the thiol content, altered the structure and thermal stability, and reduced most of the physicochemical properties. Through LC-QE-MS analysis, it was observed that oxidation leads to various modifications in arachin, including carbamylation, oxidation, and reduction, among others. In addition, 15 differentially expressed proteins were identified. Through gene ontology (GO) analysis, these proteins primarily affected the cellular and metabolic processes in the biological process category. Further Kyoto encyclopedia of genes and genomes (KEGG) analysis revealed that the "proteasome; protein processing in the endoplasmic reticulum (PPER)" pathway was the most significantly enriched signaling pathway during the oxidation process of arachin. In conclusion, this study demonstrated that AAPH-induced oxidation can alter the conformation and proteome of arachin, thereby affecting its corresponding functional properties. The findings of this study can potentially serve as a theoretical basis and foundational reference for the management of peanut processing and storage.

Abstract:The oxidation and degradation of fats lead to a decrease in the nutritional value of food and pose safety concerns. Saturated fatty acids also hold a significant position in the field of lipid oxidation. In this study, the oxidation products of methyl palmitate were investigated by using gas chromatography mass spectrometry (GC-MS). Seven monohydroperoxides and 72 secondary oxidation products were detected. Combined with density functional theory (DFT) calculations, the formation mechanisms of oxidation products can be summarized into four stages. The initial stage involved the formation of monohydroperoxides and alkanes, followed by the subsequent stage involving methyl x-oxo(hydroxy)hexadecanoates. The third stage involved the formation of methyl ketones, carboxylic acids, and aldehydes, while the final stage involved lactones. Meanwhile, methyl ketones were the most abundant oxidation product, approximately 25 times more abundant than aldehydes; the calculated results agreed well with the experimental results. The establishment of a comprehensive thermal oxidation mechanism for palmitic acid provided a new foundation for future lipid oxidation analyses.

Abstract:Ozone is used to in surimi production and affects the conformation of myosin and gelling properties. Amino acid analysis, SDS-PAGE, in-gel trypsin hydrolysis and LC-MS/MS were used to investigate the effect of the ozone treatment time on the oxidation principle of amino acids to identify the oxidation site and oxidation mechanism of myosin with different oxidation degrees. The results showed that the order of ozonation of amino acids from easy to difficult was tyrosine > cysteine > histidine > proline. The protein structure near the SH1-SH2 region initially changed when the ozone treatment time was 50 s. Prolonging the oxidation to 80 s leads to an irregular distribution of oxidation sites. Ten min of ozone treatment resulted in the aggregation from the SH1 helical region and myosin rod. This study helped to clarify the mechanism of ozone oxidation, thus providing a theoretical basis for producing surimi products of improved quality.
Dual modification of the aggregation behavior and in vitro digestion of oxidized pine kernel protein via a combination of homogenization and succinylation

Abstract:Protein oxidation affects the high-value utilization of nuts as oxidative attack causes protein aggregation, thereby challenging their technological functionality. Herein, a strategy using homogenization-assisted octenyl succinic anhydride (OSA) modification was proposed to tailor the structure, aggregation behavior, and digestive characteristics of oxidized pine kernel proteins. Results indicated that the ratio of α-helices to β-turns ranged from as low as 0.43 up to 0.67 after homogenization, suggesting greater molecular flexibility. With increasing protein oxidation, the acylation degree exhibited an inverted V-shaped trend, peaking at 67.22 %. OSA treatment reduced the aggregation rate and prolonged the lag time of proteins by stabilizing the α-helices and β-turns, increasing hydrogen bonding, and decreasing hydrophobicity. This increased the solubility of oxidized pine kernel proteins by ~30 % and improved their fluidity and thermal stability. A lower degree of succinylation was associated with higher free sulfhydryl content and surface hydrophobicity, which facilitated the thermal aggregation and the formation of elastomeric gels. Furthermore, the in vitro dynamic digestion and morphological observations of hydrolysis products indicated that cotreated proteins exhibited higher digestibility and formed small spherical particles ranging from 405.50 to 676.50 nm. These findings provide a promising approach to mitigate the adverse effects of oxidation on nut proteins.

Abstract:Studies on the atmospheric pressure cold plasma (ACP) exposure of meat and meat products mainly determine microbial inactivation, lipid oxidation, and meat color. Some studies include sensory evaluation, but only a few determine the changes in volatile composition due to ACP treatment. The results of sensory evaluation are inconclusive and range from "improvement" to "off-odor". This could be due to differences in the food matrix, especially in processed foods, or different experimental settings, including inadvertent effects such as sample heating. The few studies analyzing volatile composition report changes in alcohols, esters, aldehydes, and other compounds, but not necessarily changes that are novel for meat and meat products. Most studies do not actually measure the formation of reactive species, although this is needed to determine the exact reactions taking place in the meat during ACP treatment. This is a prerequisite for an adjustment of the plasma conditions to achieve antimicrobial effects without compromising sensory quality. Likewise, such knowledge is necessary to clarify if ACP-exposed meat and products thereof require regulatory approval.

Abstract:To elucidate the formation of protein oxidation biomarkers alpha-aminoadipic semialdehyde (AAS) and gamma-glutamic semialdehyde (GGS) in food proteins was the main purpose of the present study. Food proteins, namely, myofibrillar proteins, alpha-lactalbumin, and soy proteins, as well as bovine serum albumin (BSA), were suspended in a piperazine-1,4-bis(2-ethanesulfonic acid) (PIPES) buffer and oxidized by Fe(3+) and H(2)O(2) while kept in an oven for 14 days at 37 degrees C. For the analysis of semialdehydes, a derivatization procedure with p-aminobenzoic acid (ABA) and NaCNBH(3) followed by liquid chromatography (LC)-electrospray ionization (ESI)-multistage tandem mass spectrometry (MS) was performed. For comparative purposes, the dinitrophenylhydrazine (DNPH) method was also employed as a routine method to assess carbonyl gain. Both semialdehydes were specifically and accurately detected by LC-MS in all oxidized proteins proving that GGS and AAS are formed as a consequence of the oxidation of lysine, proline, and arginine amino acid residues from BSA and other food proteins. Proteins from an animal source and, particularly, BSA were more susceptible to undergo oxidative reactions than soy proteins. The results from the present paper highlight the significance of using both semialdehydes as protein oxidation indicators in meat and dairy products. The analysis of GGS and AAS in real food systems would contribute to the understanding of the precise mechanisms involved in food protein oxidation and shed light on the fate of oxidizing amino acids during food processing and storage.

Optimization of the Extraction of Coconut Protein and the Evaluation of Antioxidant and ACE Inhibitory Activities of its Enzymatic Hydrolysates

Abstract:The extraction of proteins from coconut meat was performed utilizing alkaline extraction. On the basis of single factor experiment, an orthogonal experiment was designed to optimize the extraction conditions (including extraction time, temperature, NaOH concentration and liquid to solid ratio). The highest extraction rate (7.3%) was obtained at 120 min, 45 °C, 0.1 M and liquid-solid ratio 25:1. Coconut protein was consequently hydrolyzed with four commercial enzymes (alcalase, trypsin, pepsin, papain). Furthermore, activities of hydrolysates on DPPH radical scavenging and ACE inhibition were assayed respectively. The results indicated that pepsin hydrolysate exhibited the highest DPPH radical scavenging activity (100%, 1 mg protein/mL), and trypsin hydrolysate possessed the most potent ACE inhibitory activity (40.2%, 1 mg protein/mL).
