Abstract:Beta-lactoglobulin (β-LG) is considered to be the major allergenic protein in milk. Lactic acid bacteria (LAB) possess a protein hydrolysis system that holds great promise for hydrolyzing β-LG and reducing its allergenicity. Therefore, this study aimed to screen LAB with β-LG hydrolysis activity from Yunnan traditional fermented foods. The results showed that Pediococcus pentosaceus C1001, Pediococcus acidilactici E1601-1, and Lactobacillus paracasei E1601-2, could effectively hydrolyze β-LG and further reduce its sensitization (more than 40%). All 3 lactic acid bacteria hydrolyzed β-LG allergenic fragments V41-K60 and L149-I162. Moreover, they encode a variety of genes related to proteolysis, such as aminopeptidase pepC and pepN, proline peptidase pepIP and endopeptidase pepO, and L. paracasei E1601-2 contains extracellular protease coding gene prtP. And they encode a variety of genes associated with hydrolyzed proteins. The 3 strains screened in this study can be used to develop hypoallergenic dairy products.
Lactic acid bacteria and yeast co-fermented milk alleviate cow milk allergy

Abstract:Cow milk allergy is one of the common food allergies. Our previous study showed that the allergenicity of fermented milk is lower than that of unfermented skimmed milk in vitro, and the antigenicity of β-lactoglobulin and α-lactalbumin in fermented milk was decreased by 67.54% and 80.49%, respectively. To confirm its effects in vivo, allergic BALB/C mice model was used to further study the allergenicity of fermented milk. It was found that compared with the skim milk (SM) group, the intragastrically sensitization with fermented milk had no obvious allergic symptoms and the fingers were more stable: lower levels of IgE, IgG, and IgA in serum, lower levels of plasma histamine and mast cell protein-1, and immune balance of Th1/Th2 and Treg/Th17. At the same time, intragastrically sensitization with fermented milk increased the α diversity of intestinal microbiota and changed the microbiota abundance: the relative abundance of norank-f-Muribaculaceae and Staphylococcus significantly decreased, and the abundance of Lachnospiraceae NK4A136 group, Bacteroides, and Turicibacter increased. In addition, fermented milk can also increase the level of short-chain fatty acids in the intestines of mice. It turns out that fermented milk is much less allergenicity than SM. PRACTICAL APPLICATION: Fermentation provides a theoretical foundation for reducing the allergenicity of milk and dairy products, thereby facilitating the production of low-allergenic dairy products suitable for individuals with milk allergies.
Detection and analysis of binding activities of IgE antibodies from child patients allergic to cow’s milk protein

Abstract:Aiming to investigate 4kinds of milk protein allergen specific IgE in serum of children with cow milk protein allergy.Specific IgE antibodies for 4kinds of milk protein allergens were detected in milk-allergic children by enzymelinked immunosorbent assay(ELISA).The results demonstrated that:α-LA-andβ-LG-specific IgE in milk allergy children serum were maintained in a high level,and the positive rate reached 44.3% and 39.3%;The positive rates ofα-CNandβ-CN-specific IgE were respectively 31.2% and 361%.These results suggested that milk-allergic children had a certain proportion of specific IgE of 4kinds of milk protein allergen.α-LA andβ-LG were probably the main allergens of milk allergy children with higher positive rate.Most of milk-allergic children were allergic to more than one kind of milk protein allergen.
Immunomodulatory Effects of L. Helveticus WHH2580 Fermented Milk on an Immunosuppressed Murine Model

Abstract:Several bacterial strains from traditional fermented foods were screened on a large scale to identify novel candidate probiotics that can be used as starters. Two in vitro cellular models were used to identify strains with immunomodulatory activity. Three strains with pro-inflammatory activity were selected, and their recovery effect was tested in vivo in an immunosuppressed murine model. Only Lactobacillus helveticus WHH2580 enhanced natural killer cell activity and splenic lymphocyte proliferation. Furthermore, milk fermented using the strain WHH2580 exerted an immunomodulatory effect with enhanced resistance to gastrointestinal conditions, including strong adhesion to HT-29 cells. Whole genome sequencing revealed the presence of a circular chromosome and two circular plasmids; no antibiotic resistance or putative virulence genes were discovered. These findings indicate the safety and positive role of L. helveticus WHH2580 in maintaining host health in mice and suggest that milk fermented with this strain can deliver this strain and thus promote immunity.

Abstract:Reducing the protein antigen fermented by probiotic has become a new research field in recent years,and it is significant to the development of the functional food market.Proteolytic activity of Lactoba- cillus helveticus was studied in this article and the ACE-inhibitory activity of the products was also re- searched.It showed that the products harvested in the period of logarithm growth have better performance. A strain was used to decrease allergen.Proteolytic activity of Lactobacillus was studied in this article and the degradation of antigen of the fermented products was also researched.The decreasing rates of allergen areα- LA31.8%,β-LG20.54%,αs-CN4.08%,β-LCN18.67% and BSA25.47%.The stain is identified as Lactoba- cillus helveticus.It can be used to produce hypoallergenic protein ingredients from milk source.
Evaluation of allergenicity of cow milk treated with enzymatic hydrolysis through a mouse model of allergy

Abstract:Cow milk (CM) allergy is a worldwide concern. Currently, few studies have been performed on the immunoreactivity of CM and fewer still on the antigenicity of CM in vivo and in vitro. In this study, we assessed the potential allergenicity of enzymatically hydrolyzed CM using in vitro ELISA and oral sensitization and challenge of BALB/c mice. Alcalase-, Protamex-, and Flavourzyme-treated CM (all from Novozymes) diminished IgE binding capacity, with greatest reductions of 56.31%, 50.62%, and 56.45%, respectively. Allergic symptoms and levels of total IgG1 were reduced, and allergic inflammation of the lung, jejunum, and spleen was relieved. Moreover, the numbers of CD8+ T and B220+ cells decreased, and the balance of CD4+ T/CD8+ T cells was effectively regulated. These findings suggest that the potential allergenicity of CM was reduced by enzymatic hydrolysis, and our research will lay a solid foundation for developing high-quality hypoallergenic CM products.
Mining Patterns of Disease Progression: A Topic-Model-Based Approach

Abstract:Knowledge of how diseases progress and transform is crucial for clinical decision making. Frequent pattern mining techniques, such as sequential pattern mining (SPM) algorithms, can automatically extract such knowledge from large collections of electronic medical records (EMR). However, EMR data are usually unorganized and highly noisy. Finding meaningful disease patterns often calls for manual manipulation such as cohort and feature selection on EMR data by medical professionals. In this paper, we propose a topic-model-based SPM approach to find disease progression patterns from diagnostic records. We improve the traditional SPM algorithms by filtering and grouping the diagnosis sequences according to different clinical topics. These topics represent certain clinical conditions with closely related diagnoses, and are detected without prior medical knowledge. The experiment on real-world EMR data shows that our approach is able to find meaningful progression patterns with less noises, and can help quickly identify interesting patterns related to a certain clinical condition with less human effort.

Abstract:Background Milk allergy is a global health issue, which affects the quality of life and can even be life threatening. According to epidemiological investigation, 2%–6% of infants are sensitive to beta-lactoglobulin (β-LG) in milk. Avoiding allergens is still the first-rank option for vulnerable group because immunization therapy is hardly available. Therefore, exploring the allergic mechanism of β-LG and applying appropriate antiallergic treatments are the key to solving the problem of milk allergy. Scope and approach This review describes the occurrence of β-LG, including the type, structure, epitopes of β-LG, the performance of gastrointestinal digestion, the mechanism and allergy consequence of β-LG allergic responses. Then, various methods to reduce the sensitization of β-LG were summarized, including covalent and non-covalent interactions, non-thermal processing technologies (fermentation, irradiation, pulsed electric field, ultrasound), thermal processing technology (microwave), and combination therapies. Key findings and conclusions : β-LG allergy is an immediate type I hypersensitivity reaction involving immunoglobulin (IgE) dependent antibody responses that cause a variety of clinical symptoms, such as asthma, eczema, gastrointestinal disorders, and anaphylactic shock in severe cases. To reduce the allergenicity of the β-LG, it is necessary to adopt novel processing strategies with higher efficiency. This review covers the covalent or non-covalent binding of polyphenols, polysaccharides, metal ions and nanoparticles to β-LG, as well as emerging technologies, and finds that structural modifications and conformational epitope changes of β-LG are associated with immunomodulatory properties. The combination of various approaches is a potential new way to reduce β-LG hypersensitivity.

Dietary Polyphenols Reduced the Allergenicity of Β-Lactoglobulin Via Non-Covalent Interactions: a Study on the Structure-Allergenicity Relationship

Abstract:Studies showed that complexation of polyphenols with milk allergens reduced their immunogenic potential. However, the relationship between structures of polyphenols and their hypoallergenic effects on milk allergens in association with physiological and conformational changes of the complexes remain unclear. In this study, polyphenols from eight botanical sources were extracted to prepare non-covalent complexes with β-lactoglobulin(β-LG), a major allergen in milk. The dominant phenolic compounds bound to β-LG with a diminished allergenicity were identified to investigate their respective role on the structural and allergenic properties of β-LG. Extracts from Vaccinium fruits and black soybeans were found to have great inhibitory effects on the IgE-and IgG-binding abilities of β-LG. Among the fourteen structure-related phenolic compounds, flavonoids and tannins with larger MWs and multi-hydroxyl substituents, notably rutin, EGCG, and ellagitannins were more potent to elicit changes on the conformational structures of β-LG to decrease the allergenicity of complexed β-LG. Correlation analysis further demonstrated that a destabilized secondary structure and protein depolymerization caused by polyphenol-binding were closely related to the allergenicity property of formed complexes. This study provides insights into the understanding of structure-allergenicity relationship of β-LG-polyphenol interactions and would benefit the development of polyphenol-fortified matrices with hypoallergenic potential.
Identification of the Critical Amino Acids of IgE-Binding Epitopes in α-Lactalbumin

Abstract:α-Lactalbumin represents one of the major allergens causing cow milk allergy.The identification of the epitopes of α-lactalbumin and the critical amino acids for its allergenicity is of great significance for understanding the mechanism of action of α-lactalbumin and developing hypoallergenic dairy products.In this study,a series of peptides were synthesized by a solid phase method for the characterization of immunological epitopes and critical amino acids.The immunoglobulin (Ig) E-binding epitopes were immunolabeled with individual sera from cow milk-allergic patients as probes by enzyme linked immunosorbent assay (ELISA).Alanine scanning of immunodominant epitopes was used to identify the critical amino acids (aa).The results showed that IgE-binding epitopes were located within the sequences of aa1-15,aa6-20,aa46-60,aa71-85 and aa101-115.Our initial data revealed that Val8,Phe9,Arg10,Tyr103,Leu105 and His107 were the critical amino acids for IgE-binding epitopes.This study will provide the necessary information to alter the cDNA to encode a protein capable of activating milk-specific T cells,but with reduced IgE-binding capacity.
Lactic acid bacteria isolated from Chinese traditional fermented milk as novel probiotic strains and their potential therapeutic applications

Abstract:Lactic acid bacteria (LAB) are believed to have health-promoting properties to the host and can be used in therapeutics interventions; intriguingly, they have the property to produce bio-preservatives substances. Therefore, this study aimed to mine probiotics and evaluate their safety, functional properties, and cholesterol-lowering capability. Seven potential probiotic strains were compared from 56 LAB strains isolated from traditional Chinese fermented milk. The results showed that all tested strains are tolerant to gastric acidity (45.5-83.26) and bile salts (11.92-92.91%) and have antibacterial activity against Staphylococcus aureus ATCC25923 and Escherichia coli ATCC25922. Likewise, it lowered the cholesterol levels in vitro by live cells (26.57-45.76%) and dead cells (29.53-50.97%) with remarkable aggregation ability (13.8-43.71%). Antioxidant properties and produce short chain fatty acids (SCFAs) were strain-dependent features. Upon assessment of the safety, Enterococcus faecium NWAFU-BIO-AS14 exhibited virulence factors genes (VFs) of (mur-2ed, odc, and tet(K)) and + hemolysis activity. While Enterococcus faecium NWAFU-BIO-A-B24 and Limosilactobacillus fermentum NWAFU-BIO-B-S6 have VFs of (odc, vanC2, and ant(6)-Ia). Limosilactobacillus fermentum NWAFU-BIO-D-B2 has only (odc). Thus, they are not considered as safe probiotics. In contrast, Lactiplantibacillus plantarum NWAFU-BIO-BS29, Companilactobacillus crustorum NWAFU-BIO-AS16, and Lactobacillus gallinarum NWAFU-BIO-D-S7 are the safest and best strains, respectively, due to the absence of 16 VFs and their sensitivity to antibiotics such as kanamycin, erythromycin, tetracycline, gentamycin, vancomycin, streptomycin, chloramphenicol, and ampicillin. Accordingly, these strains have a high potentiality to be used as starter cultures or safely applied as perfect probiotics in functionals food and feed. Effect of Lacticaseibacillus rhamnosus and Lactiplantibacillus plantarum isolated from food and human origin on reduction of IgE-dependent hypersensitivity in Balb/c mice

Abstract:Capacity and ability of Lactiplantibacillus plantarum and Lacticaseibacillus rhamnosus probiotic strains isolated from some human and food sources were evaluated for modulating and regulating the immune system against hypersensitivity type 1 (dependent on IgE). In this study, given the probiotic properties of the strains and the use of mouse models, the strains were orally administered (1×109 cfu/ml) during 45 days with gavage needles. Levels of total IgE antibodies, cytokines TGF-β, INF-ɤ, and IL-4 were measured by ELISA and compared with control groups. In addition, the nasopharyngeal lavage (total and differential count) was evaluated. Among the strains, Lactiplantibacillus plantarum a7 and Lacticaseibacillus rhamnosus M1 had the highest suppression of IL-4 cytokine production in splenocytes from ovalbumin-sensitized mice in vitro. Also, a7 and M1 had a significant (P < 0.0001) effect on reducing total IgE levels compared to the control group (OVA). Further, Lactiplantibacillus plantarum M8, a7 and M1 enhanced the production of INF-ɤ and TGF-β cytokines. Examination of nasal lavage results revealed that the total cell count in the a7, M1 and Lacticaseibacillus rhamnosus RHM groups and eosinophil cells in Lactiplantibacillus plantarum LF57, a7, and M1 significantly decreased compared to the control group. It was observed oral feeding of the isolated strains had a beneficial effect on reducing the total count on Plate Count Agar (PCA) and Mac Conkey Agar in mice feces during 7 days. M1 and a7 showed the highest level of Lactic Acid Bacteria at 7.28 and 6.9 Log cfu/g on day14, respectively. In conclusion, the animals that received the strains M1 and a7 (isolated from cow's milk and infant's small intestine) had the highest count of lactic acid bacteria in the mice gastrointestinal tract on day 14 (7.28 and 6.9 Log cfu/g), respectively. In addition, the mentioned strains can modulate immune system of mice by suppressing IL-4 production, increasing INF-ɤ and TGF-β cytokines, and reducing total IgE levels.
Improved Anti-Oxidant and Anti-Bacterial Capacities of Skim Milk Fermented by Lactobacillus plantarum

Abstract:Milk, on account of its abundant protein content, is recognized as a vital source of bioactive substances. In this study, the bioactive ingredients in milk were obtained by a combination of protease hydrolysis and fermentation with Lactobacillus plantarum. The compositions of protease hydrolysate (PM) and fermentation supernatant (FM) were determined, and their anti-oxidant and anti-bacterial activities were evaluated. Using LC-MS/MS, the molecular weights and sequences of the peptides were characterized, among which a total of 25 bioactive peptides were identified. The DPPH radical scavenging results demonstrated that FM exhibited an enhanced anti-oxidant capacity compared to PM. The bacterial survival rate results revealed that FM had a remarkable anti-bacterial ability compared to PM. Additionally, the anti-bacterial component and potential anti-bacterial mechanisms were determined. The results of cytoplasmic membrane depolarization, cell membrane permeability, and morphological observation indicated that FM could interact with bacterial membranes to achieve its anti-bacterial effect. These findings suggested that FM, as a bioactive substance of natural origin, holds potential applications in the functional food, pharmaceutical, and cosmetic industries.
Paracrine growth response as a major determinant in liver-specific colonization by in vivo selected B16 murine melanoma cells.

Abstract:We investigated the respective roles of specific adhesion and paracrine growth stimulation in preferential liver colonization by an in vivo selected B16 murine melanoma cell line (B16-LS9). Comparison of B16-LS9 cells with a lung-specific B16 melanoma cell line (B16-F10) revealed no significant differences between their adhesion properties in vitro, or their immediate organ retention pattern in vivo after tail vein injection. In contrast, B16-LS9 cells grew at slower rates in vitro than B16-F10, unless hepatocytes in coculture, or a liver plasma membrane-extract, were present. In vivo, tumors produced by B16-LS9 cells after subcutaneous or intrafootpad injection also grew at slower rates than those produced by B16-F10, and appeared to progressively lose their liver specificity. However, after intravenous (tail vein) or intrasplenic inoculation, liver colonization was much more pronounced with B16-LS9 than B16-F10 cells. These data indicate that liver colonization by the B16-LS9 cell line depends primarily on the inability of these cells to grow efficiently at sites other than the liver. The liver can indeed provide these cells with a growth-stimulating factor which we found associated with its plasma membrane fraction (i.e. juxtacrine growth stimulation).

Abstract:The fermentation characteristics and aroma-producing properties of Lactobacilli could influence the flavour quality of fermented milk, an important influencing factor of consumers' preference. In this study, fermented milk was prepared using Lactobacillus helveticus and the dynamic changes in the sensory quality of fermented milk throught fermentation were to assess the dynamic changes in sensory quality of fermented milks throughout the fermentation process, including rheological properties and flavour profiles. Styrene, linalool, octanoic acid, and 1-nonanol were considered as the key flavour components during fermentation. The quality of the fermented milk tends to be stabilized after 24-h, showing the minimal off-flavour at 48-h and optimal fermented aroma at 72-h. Three prebiotics (inulin, Galactooligosaccharides and inulin mixed with Galactooligosaccharides) were added to Lactobacillus spiralis fermented milk separately, and the results showed that inulin mixed with Galactooligosaccharides was the most effective group in improving the organoleptic quality of the fermented milk. Overall, the experimental results provide deeper insights into the release and retention of aroma compounds during fermentation and scientific reference for broadening the application of prebiotics and flavour-producing Lactobacilli in fermented milk processing.

Abstract:Lactic acid bacteria are probiotics in the intestines and have been widely used as natural antioxidants in the food industry. In this study, Enterococcus italicus FM5 with strong antioxidant ability was isolated from fresh milk. The safety evaluation showed that E. italicus FM5 was sensitive to ampicillin, chloramphenicol, erythromycin, vancomycin, rifampicin, and tetracycline, and was not hemolytic. Meanwhile, the whole genome information and biofunctional attributes of this strain were determined and analyzed. Subsequently, E. italicus FM5 was co-cultured with traditional yogurt starters (Streptococcus thermophilus and Lactobacillus bulgaricus) to make fermented milk. The results showed that the addition of E. italicus FM5 could improve the oxygen free radical scavenging ability of the fermented milk, and the scavenging rates of DPPH, ABTS, OH-, and O2- radicals reaching up to 95.54 %, 88.35 %, 93.65 %, and 60.29 %, respectively. Furthermore, the addition of E. italicus FM5 reduced the curd time and improved the water holding capacity of the fermented milk. Besides, the growth of Lb. bulgaricus was significantly promoted when co-cultured with E. italicus FM5, thus the survival cells were increased compared with the traditional fermentation processes. Therefore, this study emphasized the potential to manufacture fermented milk by the co-cultivation of E. italicus with traditional yogurt starters.

Abstract:Lactic acid bacteria (LAB) might switch the Th2 biased immune response in allergic patients towards a balanced Th1/Th2 immune profile, leading to amelioration of allergy. To select strains of LAB that could be of potential application for foods in controlling allergy, 35 bacterial strains were screened in vitro using murine splenocytes and peritoneal exudate cells (PECs). Streptococcus thermophilus AHU1838 (FERM AP-21009), and Lactobacillus paracasei subsp. casei AHU1839 (FERM AP-21010) enhanced the secretion of Th1 cytokines such as interferon-gamma (IFN-gamma) and interleukin-12 (IL-12). The two strains of LAB also up-regulated the expression of CD40, and CD86 in dendritic cells (DCs), and activated cytotoxic T lymphocytes (CTL). These two strains could therefore be used in producing fermented food products that can enhance the Th1 immune profile which is important in ameliorating allergy.

Development of innovative fermented products by exploiting the diversity of immunomodulatory properties and fermentative activity of lactic and propionic acid bacteria

Abstract:Many consumers nowadays demand plant-based milk analogs for reasons related to lifestyle, health, diet and sustainability. This has led to the increasing development of new products, fermented or not. The objective of the present study was to develop a plant-based fermented product (based on soy milk analog or on hemp milk analog), as well as mixes, using lactic acid bacteria (LAB) and propionic acid bacteria (PAB) strains, as well as consortia thereof. We screened a collection of 104 strains, from nine LAB species and two PAB species, based on their ability to ferment plant or milk carbohydrates, to acidify goat milk, soy milk analog and hemp milk analog, as well as to hydrolyze proteins isolated from these three products. Strains were also screened for their immunomodulatory ability to induce secretion of two interleukins, i.e., IL-10 and IL-12, in human Peripheral Blood Mononuclear Cells. We selected five strains: Lactobacillus delbrueckii subsp. lactis Bioprox1585, Lactobacillus acidophilus Bioprox6307, Lactococcus lactis Bioprox7116, Streptococcus thermophilus CIRM-BIA251, and Acidipropionibacterium acidipropionici CIRM-BIA2003. We then assembled them in 26 different bacterial consortia. Goat milk and soy milk analog fermented by each of the five strains or by the 26 consortia were tested in vitro, for their ability to modulate inflammation in cultured Human Epithelial Intestinal Cells (HEIC) stimulated by pro-inflammatory Lipopolysaccharides (LPS) from Escherichia coli. Plant-based milk analogs, fermented by one consortium composed of L.delbrueckii subsp. lactis Bioprox1585, Lc.lactis Bioprox7116, and A.acidipropionici CIRM-BIA2003, reduced the secretion of the proinflammatory cytokine IL-8 in HIECs. Such innovative fermented vegetable products thus open perspectives as functional foods targeting gut inflammation.
Enhancing the antioxidant capacity and quality attributes of fermented goat milk through the synergistic action of Limosilactobacillus fermentum WXZ 2-1 with a starter culture

Abstract:This study evaluated 75 strains of lactic acid bacteria (LAB) isolated from traditional dairy products in western China for their probiotic properties. Among them, Limosilactobacillus fermentum WXZ 2-1, Lactiplantibacillus plantarum TXZ 2-35, Companilactobacillus crustorum QHS 9, and Companilactobacillus crustorum QHS 10 demonstrated potential probiotic characteristics. The antioxidant capacity of these 4 strains was assessed, revealing that L. fermentum WXZ 2-1 exhibited the highest antioxidant capacity. Furthermore, when cocultured with Streptococcus salivarius ssp. thermophilus and Lactobacillus delbrueckii ssp. bulgaricus, L. fermentum WXZ 2-1 demonstrated a synergistic effect in growth medium and goat milk. To explore its effect on goat milk fermentation, different amounts of L. fermentum WXZ 2-1 were added to goat milk, and its physicochemical properties, antioxidant activity, flavor substances, and metabolomics were analyzed. The study found that the incorporation of L. fermentum WXZ 2-1 in goat milk fermentation significantly improved the texture characteristics, antioxidant capacity, and flavor of fermented goat milk. These findings highlight the potential of L. fermentum WXZ 2-1 as a valuable probiotic strain for enhancing the functionality and desirability of fermented goat milk, contributing to the development of novel functional foods with improved health benefits and enhanced quality attributes.
