Abstract:Activation of invasion and metastasis is recognized as one of the hallmarks of cancer. There are 90% of cancer-related deaths due to metastasis and given that it is worthy of note to study cancer progression and metastasis. Owing to restricted tools used to underpin the study of tumor invasion process, an on-site platform was developed to monitor this event in vitro. We used interdigital gold electrodes to monitor the dynamic process of cancer cells invading into extracellular matrix in situ continuously. Influences of collagen concentration and number of cancer cells on the measured impedance was exhibited. In addition, the parameters used to demonstrate the experiment results were optimized. The change of impedance magnitude indicated the cell-matrix interaction during invasion process. The potential further use of this platform would be complementary in cell studies when concerning metastasis.

Abstract:Cancer development and progression associated with epithelial-mesenchymal transition (EMT) not only results in biological and functional abnormalities but also leads to changes in mechanical and structural characteristics of cells. Mechanical characterization has been considered as a promising, label-free, alternative way to cancer diagnosis. In this paper, we fabricate a microchip to quantify mechanical deformability of cancer cells during EMT in an electrodeformation-based way. For three typical cancer cells, i.e., MCF-7, MDA-MB-231 and A549 cells, our experimental outcome manifests that there exists significant difference in the deformability before and after EMT induced by TGF-beta 1. Both MCF-7 and MDA-MB-231 cells after EMT show more than 40% reduction in elasticity and viscosity, whereas A549 cells appear to have a similar to 25% decrease in elasticity and similar to 35% in viscosity, respectively. These findings imply it is feasible to monitor the EMT process by means of mechanical clue of cells involved. It is expected that the present electrodeformation-based technique can be developed into a label-free, high-throughput method to evaluate metastatic potential of cancer cells, which is crucial to early detection and diagnosis of cancer. Mechanomics Biomarker for Cancer Cells Unidentifiable through Morphology and Elastic Modulus

Abstract:Cellular mechanical properties are potential cancer biomarkers used for objective cytology to replace the current subjective method relying on cytomorphology. However, heterogeneity among intra/intercellular mechanics and the interplay between cytoskeletal prestress and elastic modulus obscured the difference detectable between malignant and benign cells. In this work, we collected high density nanoscale prestress and elastic modulus data from a single cell by AFM indentation to generate a cellular mechanome. Such high dimensional mechanome data was used to train a malignancy classifier through machine learning. The classifier was tested on 340 single cells of various origins, malignancy, and degrees of similarity in morphology and elastic modulus. The classifier showed instrument-independent robustness and classification accuracy of 89% with an AUC-ROC value of 93%. Abstract:There is spatiotemporal heterogeneity in cell phenotypes and mechanical properties in tumor tissues, which is associated with cancer invasion and metastasis. It is well-known that exogenous growth factors like transforming growth factor (TGF)-β, can induce epithelial-mesenchymal transition (EMT)-based phenotypic transformation and the formation of EMT patterning on geometrically confined monolayers with mechanics heterogeneity. In the absence of exogenous TGF-β stimulation, however, whether geometric confinement-caused mechanics heterogeneity of cancer cell monolayers alone can trigger the EMT-based phenotypic heterogeneity still remains mysterious. Here, we develop a micropattern-based cell monolayer model to investigate the regulation of mechanics heterogeneity on the cell phenotypic switch. We reveal that mechanics heterogeneity itself is enough to spontaneously induce the emergence of mesenchymal-like phenotype and asymmetrical activation of TGF-β-SMAD signaling. Spatiotemporal dynamics of patterned cell monolayers with mesenchymal-like phenotypes is essentially regulated by tissue-scale cell behaviors like proliferation, migration as well as heterogeneous cytoskeletal contraction. The inhibition of cell contraction abrogates the asymmetrical TGF-β-SMAD signaling activation level and the emergence of mesenchymal-like phenotype. Our work not only sheds light on the key regulation of mechanics heterogeneity caused by spatially geometric confinement on regional mesenchymal-like phenotype of cancer cell monolayers, but highlights the key role of biophysical/mechanical cues in triggering phenotypic switch.

Abstract:Epithelial-mesenchymal transition (EMT) is a developmental cell state transition co-opted by cancer to drive metastasis and resistance. Stable EMT intermediate states play a particularly important role in cell state plasticity and confer metastatic potential. To explore the dynamics of EMT and identify marker genes of highly metastatic intermediate cells, we analyzed EMT across multiple tumor types and stimuli via mathematical modeling with single-cell RNA-sequencing (scRNA-seq) data. We identified pan-cancer genes consistently expressed or upregulated in EMT intermediate states, most of which were not previously annotated as markers of EMT. Using Bayesian parameter inference, we fit a simple mathematical model to scRNA-seq data, revealing tumor-specific transition rates. This mathematical model offers a framework to quantify EMT progression. A consensus analysis of differential intermediate expression, regulation, and model-derived dynamics identified marker genes associated with persistence of the intermediate EMT state. SFN and NRG1 emerged as genes with the strongest evidence for their role influencing intermediate EMT dynamics. Through analysis of an independent cell line, we verified the role of SFN as a marker intermediate EMT transition. Modeling and inference of genes associated with EMT dynamics offer means to find biomarkers and to identify therapeutic approaches to harness or reverse tumor-promoting cell state transitions driven by EMT.

Abstract:Normal-to-cancer (NTC) transition is known to be closely associated to cell´s biomechanical properties which are dependent on the dynamics of the intracellular medium. This study probes different human cancer cells (breast, prostate and lung), concomitantly to their healthy counterparts, aiming at characterising the dynamical profile of water in distinct cellular locations, for each type of cell, and how it changes between normal and cancer states. An increased plasticity of the cytomatrix is observed upon normal-to-malignant transformation, the lung carcinoma cells displaying the highest flexibility followed by prostate and breast cancers. Also, lung cells show a distinct behaviour relative to breast and prostate, with a higher influence from hydration water motions and localised fast rotations upon NTC transformation. Quasielastic neutron scattering techniques allowed to accurately distinguish the different dynamical processes taking place within these highly heterogeneous cellular systems. The results thus obtained suggest that intracellular water dynamics may be regarded as a specific reporter of the cellular conditions-either healthy or malignant.

Abstract:Epithelial to mesenchymal transition (EMT) is crucial during embryonic development, tissue fibrosis, and cancer progression. Epithelial cells that display a cobblestone-like morphology can undergo a switch to mesenchymal-like phenotype, displaying an elongated spindle shape or a fibroblast-like morphology. EMT is characterized by timely and reversible alterations of molecular and cellular processes. The changes include loss of epithelial and gain of mesenchymal marker expression, loss of polarity, increased cell migratory and invasive properties. Epithelial cells can progress unevenly during this transition and attain hybrid E/M states or metastable EMT states, referred to as epithelial cell plasticity. To gain a deeper insight into the mechanism of EMT, understanding the dynamic aspects of this process is essential. One of the most prominent factors to induce EMT is the cytokine transforming growth factor-β (TGF-β). This chapter discusses molecular and cellular techniques to monitor TGF-β-induced signaling and EMT changes in normal and cancer cell lines. These methods include measuring the TGF-β-induced activation of its intracellular SMAD effectors proteins and changes in epithelial/mesenchymal marker expression and localization. Moreover, we describe assays of cell migration and dynamic reorganization of the actin cytoskeleton and stress filaments that are frequently part of the TGF-β-induced EMT cellular response.

Abstract:Cancer metastasis is a multistep complex process accompanied by changes in the mechanical properties of cancer cells, such as adhesion, viscoelasticity, deformability, and motility. Connexin 43 (Cx43), a gap junction protein with a putative tumor suppressor function, plays a pivotal role in Epithelial to Mesenchymal transition (EMT). It affects cell motility indicators and promotes cytoskeleton reorganization and, ultimately, metastasis when downregulated, corroborating resistance to chemotherapy. The lack of universal markers that can predict the metastatic ability of cancer cells necessitates the evaluation of parameters, such as cell adhesion, viscoelasticity, and stiffness, to serve as unique biophysical metrics that predict cancer cell metastatic potential and elucidate how metastasis and cell mechanics are coupled. Therefore, we utilized fluidic-based single-cell force spectroscopy (SCFS) and Quartz Crystal Microbalance with Dissipation Monitoring (QCM-D) to measure the mechanical properties of single cancer cells with different metastatic potential. To explore the relationship between cell mechanics and metastatic cell behavior, we quantitatively demonstrate the progression of cancer cell biomechanics across different metastatic states using MDA-MB-231 breast cancer cells with varying connexin 43 (Cx43) expression as a model of cells with varying metastatic potential. We measured, at a single-cell level and in real-time, the adhesion and viscoelastic properties of MDA-MB-231 cells and their response to chemotherapy. Our data show that cells with higher metastatic potential (down-regulated Cx43 expression) exhibited a softer, less adherent, and more viscoelastic phenotype, consistent with their metastatic state when in clusters compared to cells at the lower spectrum of metastatic potential (up-regulated Cx43 expression). These mechanical biomarkers are significantly absent in single cells outside of a cluster. Of particular interest, we also observed significant cellular stiffening upon treatment with docetaxel (DTX), a chemotherapeutic agent that targets microtubules, with a rapid and profound response of highly aggressive cells when compared to cells with attenuated metastatic potential. Such treatment schemes could render cancer cells less deformable, potentially impeding their ability to invade and metastasize to other tissues. Our study demonstrates a unique quantitative metric that unfolds biomechanical markers for discerning the metastatic state of cancer cells. Our data represents a stepping stone to understanding how mechanical changes in cancer cells may determine how cancer progresses and responds to chemotherapeutic agents and may provide novel modalities to inhibit metastasis. Abstract:Mechanical phenotypes of cells are found to hold vital clues to reveal cellular functions and behaviors, which not only has great physiological significance but also is crucial for disease diagnosis. To this end, we developed a set of electrodeformation-based biomechanical microchip assays to quantify mechanical phenotypes on the single-cell level. By investigating the spatiotemporal dynamics of cancer cells driven by dielectrophoresis forces, we captured the key global viscoelastic indexes including cellular elasticity, viscosity, and transition time that was defined as the ratio of the transient viscosity and elasticity, simultaneously, and thus explored their intrinsic correlation with cell cycle progression. Our results showed that both global elasticity and viscosity have a significant periodic variation with cell cycle progression, but the transition time remained unchanged in the process, indicating that it might be an intrinsic property of cancer cells that is independent of the cell cycle and the type of cell in the experiments. Further, we investigated the molecular mechanism regulating cellular viscoelastic phenotypes on the biomechanical chips through intracellular cytoskeletal perturbation assays. These findings, together with the electrodeformation-based microchip technique, not only reveal the relation between mechanical phenotypes of cancer cells and cell cycle progression but also provide a platform for implementing multi-index mechanical phenotype assays associated with cancer cell cycles in the clinic.
Characterizing cellular mechanical phenotypes with mechano-node-pore sensing

Abstract:The mechanical properties of cells change with their differentiation, chronological age, and malignant progression. Consequently, these properties may be useful label-free biomarkers of various functional or clinically relevant cell states. Here, we demonstrate mechano-node-pore sensing (mechano-NPS), a multi-parametric single-cell-analysis method that utilizes a four-terminal measurement of the current across a microfluidic channel to quantify simultaneously cell diameter, resistance to compressive deformation, transverse deformation under constant strain, and recovery time after deformation. We define a new parameter, the whole-cell deformability index (wCDI), which provides a quantitative mechanical metric of the resistance to compressive deformation that can be used to discriminate among different cell types. The wCDI and the transverse deformation under constant strain show malignant MCF-7 and A549 cell lines are mechanically distinct from non-malignant, MCF-10A and BEAS-2B cell lines, and distinguishes between cells treated or untreated with cytoskeleton-perturbing small molecules. We categorize cell recovery time, ΔTr, as instantaneous (ΔTr~0 ms), transient (ΔTr⩽40 ms), or prolonged (ΔTr>40 ms), and show that the composition of recovery types, which is a consequence of changes in cytoskeletal organization, correlates with cellular transformation. Through the wCDI and cell-recovery time, mechano-NPS discriminates between sub-lineages of normal primary human mammary epithelial cells with accuracy comparable to flow cytometry, but without antibody labeling. Mechano-NPS identifies mechanical phenotypes that distinguishes lineage, chronological age, and stage of malignant progression in human epithelial cells.

Abstract:The transition between epithelial and mesenchymal (EMT) is a fundamental cellular process that plays critical roles in development, cancer metastasis, and tissue wound healing. EMT is not a binary process but involves multiple partial EMT states that give rise to a high degree of cell state plasticity. Here, we first reviewed several studies on theoretical predictions and experimental verification of these intermediate states, the role of partial EMT on kidney fibrosis development, and how quantitative signaling information controls cell commitment to partial or full EMT upon transient signals. Next, we summarized existing knowledge and open questions on the coupling between EMT and other biological processes, such as the cell cycle, epigenetic regulation, stemness, and apoptosis. Taken together, EMT is a model system that has attracted increasing interests for quantitative experimental and theoretical studies.

Abstract:High‐speed scanning ion conductance microscopy reveals sub‐cellular morphologies and elastic modulus in dynamics and their correlations of basal surface derived from metastatic intestinal 3D organoids in physiological condition, where the elastic modulus as a key factor allows to differ malignant tumor cells from benign tumor cells and normal cells. Studying mechanobiology is increasing of scientific interests in life science and nanotechnology since its impact on cell activities (e.g., adhesion, migration), physiology, and pathology. The role of apical surface (AS) and basal surface (BS) of cells played in mechanobiology is significant. The mechanical mapping and analysis of cells mainly focus on AS while little is known about BS. Here, high‐speed scanning ion conductance microscope as a powerful tool is utilized to simultaneously reveal morphologies and local elastic modulus (E) of BS of genotype‐defined metastatic intestinal organoids. A simple method is developed to prepare organoid samples allowing for long‐term BS imaging. The multiple nano/microstructures, i.e., ridge‐like, stress‐fiber, and E distributions on BS are dynamically revealed. The statistic E analysis shows softness of BS derived from eight types of organoids following a ranking: malignant tumor cells > benign tumor cells > normal cells. Moreover, the correlation factor between morphology and E is demonstrated depending on cell types. This work as first example reveals the subcellular morphologies and E distributions of BS of cells. The results would provide a clue for correlating genotype of 3D cells to malignant phenotype reflected by E and offering a promising strategy for early‐stage diagnosis of cancer.

Abstract:The dynamical change of cellular mechanical properties plays an important role in cell metastasis process, while how the cancer cells modulate their mechanical properties during metastasis are still not fully understood. In this report, the cellular detaching and seeding processes, two vital steps of cell metastasis, were simulated in vitro using a self-developed protocol and characterized by the dynamical mechanical properties using AFM. The measured results show that cells decrease their stiffness and increase their surface adhesion force as they are detaching from substrate, while cells present an opposite change in mechanical properties as they seeding. Additionally, the effect of anti-cancer drug (docetaxel) on the detaching and attaching process of cancer cells (PC-3) was also investigated from the aspect of mechanical properties. The results shows that the docetaxel can increase stiffness, decrease surface adhesion force of PC-3 cell, and slow down the change speed of these mechanical properties during PC-3 cell detaching and seeding process. These discoveries demonstrated that a dynamical change of cell mechanical properties is required for cancer cell metastasis, which provide a new drug development strategy for cancer treatment.

Abstract:Metastasis is a complicated, multistep process that is responsible for over 90% of cancer-related death. Metastatic disease or the movement of cancer cells from one site to another requires dramatic remodeling of the cytoskeleton. The regulation of cancer cell migration is determined not only by biochemical factors in the microenvironment but also by the biomechanical contextual information provided by the extracellular matrix (ECM). The responses of the cytoskeleton to chemical signals are well characterized and understood. However, the mechanisms of response to mechanical signals in the form of externally applied force and forces generated by the ECM are still poorly understood. Furthermore, understanding the way cellular mechanosensors interact with the physical properties of the microenvironment and transmit the signals to activate the cytoskeletal movements may help identify an effective strategy for the treatment of cancer. Here, we will discuss the role of tumor microenvironment during cancer metastasis and how physical forces remodel the cytoskeleton through mechanosensing and transduction.

Abstract:BackgroundThe Epithelial-Mesenchymal Transition (EMT) endows epithelial-looking cells with enhanced migratory ability during embryonic development and tissue repair. EMT can also be co-opted by cancer cells to acquire metastatic potential and drug-resistance. Recent research has argued that epithelial (E) cells can undergo either a partial EMT to attain a hybrid epithelial/mesenchymal (E/M) phenotype that typically displays collective migration, or a complete EMT to adopt a mesenchymal (M) phenotype that shows individual migration. The core EMT regulatory network - miR-34/SNAIL/miR-200/ZEB1 - has been identified by various studies, but how this network regulates the transitions among the E, E/M, and M phenotypes remains controversial. Two major mathematical models – ternary chimera switch (TCS) and cascading bistable switches (CBS) - that both focus on the miR-34/SNAIL/miR-200/ZEB1 network, have been proposed to elucidate the EMT dynamics, but a detailed analysis of how well either or both of these two models can capture recent experimental observations about EMT dynamics remains to be done.ResultsHere, via an integrated experimental and theoretical approach, we first show that both these two models can be used to understand the two-step transition of EMT - E→E/M→M, the different responses of SNAIL and ZEB1 to exogenous TGF-β and the irreversibility of complete EMT. Next, we present new experimental results that tend to discriminate between these two models. We show that ZEB1 is present at intermediate levels in the hybrid E/M H1975 cells, and that in HMLE cells, overexpression of SNAIL is not sufficient to initiate EMT in the absence of ZEB1 and FOXC2.ConclusionsThese experimental results argue in favor of the TCS model proposing that miR-200/ZEB1 behaves as a three-way decision-making switch enabling transitions among the E, hybrid E/M and M phenotypes.

Isolating and targeting the real-time plasticity and malignant properties of epithelial-mesenchymal transition in cancer

Abstract:Epithelial-mesenchymal transition (EMT) is a driving force in promoting malignant cancer, including initiation, growth, and metastasis. EMT is a dynamic process that can undergo a mesenchymal-epithelial transition (MET) and partial transitions between both phenotypes, termed epithelial-mesenchymal plasticity (EMP). In cancer, the acquisition of EMP results in a spectrum of phenotypes, promoting tumor cell heterogeneity and resistance to standard of care therapy. Here we describe a real-time fluorescent dual-reporter for vimentin and E-cadherin, biomarkers of the mesenchymal and epithelial cell phenotypes, respectively. Stable dual-reporter cell lines generated from colorectal (SW620), lung (A549), and breast (MDA-MB-231) cancer demonstrate a spectrum of EMT cell phenotypes. We used the dual-reporter to isolate the quasi epithelial, epithelial/mesenchymal, and mesenchymal phenotypes. Although EMT is a dynamic process, these isolated quasi-EMT-phenotypes remain stable to spontaneous EMP in the absence of stimuli and during prolonged cell culture. However, the quasi-EMT phenotypes can readily be induced to undergo EMT or MET with growth factors or small molecules. Moreover, isolated EMT phenotypes display different tumorigenic properties and are morphologically and metabolically distinct. 3D high-content screening of ~23,000 compounds using dual-reporter mesenchymal SW620 tumor organoids identified small molecule probes that modulate EMT, and a subset of probes that effectively induced MET. The tools, probes, and models described herein provide a coherent mechanistic understanding of mesenchymal cell plasticity. Future applications utilizing this technology and probes are expected to advance our understanding of EMT and studies aimed at therapeutic strategies targeting EMT.

Abstract:Cancer cells are in vivo situated in a complex and heterogeneous tumor microenvironment (TME) that includes various biochemical and biophysical cues, such as the elasticity of extracellular matrix (ECMwo) and the viscosity of extracellular fluid (ECF). ECF viscosity in TME is much higher than in normal tissue, but it is unclear how this increased viscosity works simultaneously with other biophysical cues (e.g., ECM stiffness) to influence cancer cell behavior. We experimentally observed that ECF viscosity can significantly enhance cellular mechanosensing behaviors (e.g., cell spreading, cell adhesion, and YAP/TAZ nuclear translocation) only on a stiff substrate, representing a novel enhancement of cell behaviors by distinct mechanical signals. To explore the mechanical mechanisms behind such enhancement phenomenon, we developed a viscosity-based motor-clutch model, with which we found that cells sense and respond to ECF viscosity and ECM stiffness by regulating integrin-ECM bonds in cell adhesion dynamics. These findings help us understand how different mechanical signals in the complex tumor microenvironment collaborate to influence cancer cell behaviors during the development of cancers.

Abstract:Somatic cells that change from one mature phenotype to another exhibit the property of plasticity. It is increasingly clear that epithelial and endothelial cells enjoy some of this plasticity, which is easily demonstrated by studying the process of epithelial-mesenchymal transition (EMT). Published reports from the literature typically rely on ad hoc criteria for determining EMT events; consequently, there is some uncertainty as to whether the same process occurs under different experimental conditions. As we discuss in this Personal Perspective, we believe that context and various changes in plasticity biomarkers can help identify at least three types of EMT and that using a collection of criteria for EMT increases the likelihood that everyone is studying the same phenomenon - namely, the transition of epithelial and endothelial cells to a motile phenotype.

Abstract:Epithelial-to-mesenchymal transition (EMT) plays an important role in many biological processes during development and cancer. The advent of single-cell transcriptome sequencing techniques allows the dissection of dynamical details underlying EMT with unprecedented resolution. Despite several single-cell data analysis on EMT, how cell communicates and regulates dynamics along the EMT trajectory remains elusive. Using single-cell transcriptomic datasets, here we infer the cell–cell communications and the multilayer gene–gene regulation networks to analyze and visualize the complex cellular crosstalk and the underlying gene regulatory dynamics along EMT. Combining with trajectory analysis, our approach reveals the existence of multiple intermediate cell states (ICSs) with hybrid epithelial and mesenchymal features. Analyses on the time-series datasets from cancer cell lines with different inducing factors show that the induced EMTs are context-specific: the EMT induced by transforming growth factor B1 (TGFB1) is synchronous, whereas the EMTs induced by epidermal growth factor and tumor necrosis factor are asynchronous, and the responses of TGF-β pathway in terms of gene expression regulations are heterogeneous under different treatments or among various cell states. Meanwhile, network topology analysis suggests that the ICSs during EMT serve as the signaling in cellular communication under different conditions. Interestingly, our analysis of a mouse skin squamous cell carcinoma dataset also suggests regardless of the significant discrepancy in concrete genes between in vitro and in vivo EMT systems, the ICSs play dominant role in the TGF-β signaling crosstalk. Overall, our approach reveals the multiscale mechanisms coupling cell–cell communications and gene–gene regulations responsible for complex cell-state transitions.
