Abstract:N-cadherin serves as an important oncobiomarker of epithelial-to-mesenchymal transition (EMT) progression, which identifies invasion and metastasis of malignant tumor cells. Although many efforts have been devoted to quantitative detection of N-cadherin, efforts to analyzing the protein of interest at intact cellular levels are scarce. Herein, a metal cluster-based electrochemical biosensing system is developed to determine the expressing levels of N-cadherin during the EMT process of tumor cells. To be specific, a peptide with a unique sequence and function is designed as a reductant and an anchor to synthesize metal clusters in a precise manner. Consequently, peptide-modified metal clusters possess N-cadherin-targeting, photoluminescence, and electrocatalytic properties. Especially, the redox-active metal clusters function as both an electron-transfer mediator and an electronic conductor for enhanced electrochemical sensing. These favorable features enable them as a rapid, sensitive, and reliable whole-cell biosensor, which integrates the fluorescence and electrochemical signals. This cytosensor can accurately quantify the expression levels of N-cadherin on at least 5000 tumor cells. Further, the current signals of model cancer cells gradually increase with EMT progression, indicating tumor cell-type evolution. Abstract:Abstract Epithelial-mesenchymal transition (EMT) is critically involved in a variety of biological processes. Electrochemical sensing offers potential to develop more effective technology for EMT detection. In this study, by using the unique performance of quantum dot (QD)-nanocomposite materials, we establish an electrochemical biosensor that can specifically detect the change of E-cadherin and analyze different stages of EMT. The signal for EMT is largely magnified due to the transmission of molecular information to the electronic device. In addition, differential pulse voltammetry reveals that the response of the electrochemical signals is rapid and sensitive, due to the synergistic effect of QDs and carbon nanotube-gold nanoparticles. Our study thus suggests that electrochemical sensing is an effective technology for detecting EMT and may have broad applications in analyzing various cell type transitions.

Abstract:In this study, cell biosensor chip with micro-electrode arrays was used to monitor the culture behaviour of mammalian cancer cells and evaluate the chemosensitivity of anti-cancer drugs using electrochemical impedance spectroscopy. The electrode arrays were fabricated by semiconductor technology with diameter from 50 pm to 10 pm of each electrode. The human oesophageal cancer cell lines (KYSE 30) were cultured on the surface of the electrode with the help of fibronectin. Morphology changes of cells adhesion, spreading, and proliferation can be detected by impedimetric analysis in a real time and non-invasive way. Anti-caner drug of cisplatin was added to cells for potential drug screening applications. The experimental results show that drug's chemosensitivity effects can be detected by this cell based biosensor.

Abstract:We construct a peptide-conjugated metal cluster as an enzyme-like catalytic bioprobe to enhance quantitative analysis of a membrane protein biomarker and detect epithelial-to-mesenchymal transition of tumor cells. This bioprobe with atomically precise formula, termed clusterzyme, possesses selective recognition and intrinsic enzyme-like activity. These favorable features facilitate sensitive quantitative analysis of the membrane protein in situ through on-cell catalytic signal amplification. This clusterzyme-based analytical method exhibits excellent compatibility with a traditional enzyme-linked immunosorbent assay and improved detection sensitivity with accuracy and robustness. Further, the expression level of the membrane protein reflects the ability of migration and invasion of model tumor cells, revealing epithelial-to-mesenchymal transition process. This work offers a facile and sensitive approach to monitor tumor cell type evolution at the molecular level, demonstrating a potential application of early cancer diagnosis and therapy assessment.

Abstract:Cluster of differentiation 147 (CD147), also known as extracellular matrix metalloproteinase inducer (EMMPRIN), plays an essential role in tumor progression and metastasis, the expression of which on cell surface is a critical clinical testing index for cancer therapy. In this work, an electrochemical method to assay CD147/EMMPRIN expression on tumor cell surface is proposed. While the oxidation of 3,3′,5,5′-tetramethylbenzidine (TMB) catalyzed by horseradish peroxidase (HRP) can be employed for electrochemical measurement, the signal enhancement amplified by gold nanoparticles (GNPs) can be also utilized in this study. Therefore, under optimized conditions, the fabricated biosensor responds linearly to the CD147/EMMPRIN concentration from 125 to 1000pg/mL with a detection limit as low as 52pg/mL. High sensitivity can also be achieved for the quantification of breast cancer cells in a linear range from 6.2×104 to 6.25×105 cells/mL. Moreover, the CD147/EMMPRIN expressed on a single breast cancer cell can be calculated as 2.57×104 molecules/cell. The proposed strategy in this study is considerably potential for monitoring the dynamic protein expression on cancer cells and for the effective cancer diagnosis and treatment in the future.

Abstract:The cell surface protein epithelial cell adhesion molecule (EpCAM) has oncogenic effects, and its overexpression is strongly correlated with tumor development. Here, an electrochemical method is proposed to detect and identify EpCAM at the level of purified proteins and living cells. The core concept of this method is the use of DNA nanotetrahedron (NTH)-based capture probes and aptamer-modified quasi-ZIF-67@Au@methylene blue (QZIF-67@Au@MB@APT) as signal probes. In our design, NTH-assisted targeted immobilization can greatly improve the accessibility of nucleotide-containing artificial aptamers to suspended targets and the specificity of the target EpCAM. Moreover, following partial deligandization, ZIF-67 was converted to QZIF-67 with an extensively improved porous structure that significantly enhanced the loading efficiency of signaling molecules and thus led to a remarkable increase in detection sensitivity. QZIF-67@Au@MB can be integrated with an aptamer to act as a signal reporter with an electrochemical sensing platform for EpCAM detection. By tracing the electrochemical signals from the QZIF-67@Au@MB@APT, the method demonstrated the detection of targets as low as 3 pg/mL within a wide linear range from 0.005 to 100 ng/mL. We have successfully applied this aptasensor for in situ characterization of EpCAM on the cellular surface and monitoring alterations in EpCAM expression during drug treatment, which provides a low-cost but robust alternative to highly expensive and experience-dependent flow cytometry. Overall, electrochemical characterization may provide promising technical support for the precise management of tumors.

Abstract:The epithelial cell adhesion molecule (EpCAM) is a known biomarker of circulating tumor cells that plays an important role in tumor metastasis. The detection of EpCAM is vital for personalized diagnosis and therapy but is challenging because of its extremely low concentration in peripheral blood. In our work, an electrochemical aptasensor has been developed to quantitatively detect EpCAM at low concentrations. A gold electrode modified with an aptamer can capture EpCAM, which can then be further recognized by a second aptamer acting as a signal reporter. To amplify the signal, a silica nanoparticle/CdSe complex has been designed to enhance the detection system. First, silica nanoparticles were synthesized to act as carriers for loading many CdSe quantum dots which are capable of binding with the second aptamer by biotin-streptavidin interactions. A molecule of EpCAM can bind with numerous CdSe quantum dots (QDs) to realize significant signal amplification. By stripping square wave voltammetry (SSWV), concentrations down to 10 aM EpCAM can be successfully detected and quantified based on the newly developed aptasensor. The detection linear range is from 10 aM to 100 pM. This principle can also be extended to other biomarkers when a suitable aptamer or antibody is available. Abstract:Cancer development and progression associated with epithelial-mesenchymal transition (EMT) not only results in biological and functional abnormalities but also leads to changes in mechanical and structural characteristics of cells. Mechanical characterization has been considered as a promising, label-free, alternative way to cancer diagnosis. In this paper, we fabricate a microchip to quantify mechanical deformability of cancer cells during EMT in an electrodeformation-based way. For three typical cancer cells, i.e., MCF-7, MDA-MB-231 and A549 cells, our experimental outcome manifests that there exists significant difference in the deformability before and after EMT induced by TGF-beta 1. Both MCF-7 and MDA-MB-231 cells after EMT show more than 40% reduction in elasticity and viscosity, whereas A549 cells appear to have a similar to 25% decrease in elasticity and similar to 35% in viscosity, respectively. These findings imply it is feasible to monitor the EMT process by means of mechanical clue of cells involved. It is expected that the present electrodeformation-based technique can be developed into a label-free, high-throughput method to evaluate metastatic potential of cancer cells, which is crucial to early detection and diagnosis of cancer. Multifunctional Gold Nano-Cytosensor With Quick Capture, Electrochemical Detection, and Non-Invasive Release of Circulating Tumor Cells for Early Cancer Treatment

Abstract:Circulating tumor cells (CTCs) are metastatic tumor cells that shed into the blood from solid primary tumors, and their existence significantly increases the risk of metastasis and recurrence. The timely discovery and detection of CTCs are of considerable importance for the early diagnosis and treatment of metastasis. However, the low number of CTCs hinders their detection. In the present study, an ultrasensitive electrochemical cytosensor for specific capture, quantitative detection, and noninvasive release of EpCAM-positive tumor cells was developed. The biosensor was manufactured using gold nanoparticles (AuNPs) to modify the electrode. Three types of AuNPs with controllable sizes and conjugated with a targeting molecule of monoclonal anti-EpCAM antibody were used in this study. Electrochemical impedance spectroscopy (EIS) and differential pulse voltammetry (DPV) of the cytosensors were performed to evaluate the cell capture efficiency and performance. The captured 4T1 cells by the AuNPs hindered electron transport efficiency, resulting in increased EIS responses. The cell capture response recorded using EIS or DPV indicated that the optimal AuNPs size should be 17 nm. The cell capture response changed linearly with the concentration range from 8.0 × 10 to 1 × 10 7 cells/mL, and the limit of detection was 50 cells/mL. After these measurements, glycine-HCl (Gly-HCl) was used as an antibody eluent to destroy the binding between antigen and antibody to release the captured tumor cells without compromising their viability for further clinical research. This protocol realizes rapid detection of CTCs with good stability, acceptable assay precision, significant fabrication reproducibility with a relative standard deviation of 2.09%, and good recovery of cells. Our results indicate that the proposed biosensor is promising for the early monitoring of CTCs and may help customize personalized treatment options.

Abstract:Epithelial cell adhesion molecule (EpCAM), which is overexpressed in breast cancer cells and participates in cell signaling, migration, proliferation, and differentiation, has been utilized as a biomarker for cancer diagnosis and therapeutic prognosis. Here, a dual-signal readout nonenzymatic aptasensor is fabricated for the evaluation of EpCAM at the level of three breast cancer cell lines. The central principle of this enzyme-free aptasensor is the use of double hook-type aptamers (SYL3C and SJ3C2)-functionalized magnetic iron oxide (Fe3O4) as capture probes and quasi-CoFe prussian blue analogs (QCoFe PBAs) as nonenzymatic signal probes for colorimetric and electrochemical analysis. Following ligand detachment, the CoFe PBA was transformed to QCoFe PBA (calcined at 350 °C for 1 h), with its metal active sites exposed by controllable pyrolysis. We found that the enhanced sensitivity was attributed to the resonance effect of QCoFe PBA with the remarkable enzymatic properties. The dual-signal readout nonenzymatic aptasensor exhibited limits of detection for EpCAM as low as 0.89 pg mL-1 and 0.24 pg mL-1, within a wide linear range from 0.001 to 100 ng mL-1, respectively. We successfully employed this nonenzymatic aptasensor for monitoring EpCAM expression in three breast cancer cell lines, which provides an economical and robust alternative to costly and empirical flow cytometry. The dual-signal readout nonenzymatic aptasensor provides rapid, robust, and promising technological support for the accurate management of tumors.
Electrical Impedance Spectroscopy as a Tool to Detect the Epithelial to Mesenchymal Transition in Prostate Cancer Cells

Abstract:Prostate cancer (PCa) remains a significant health threat, with chemoresistance and recurrence posing major challenges despite advances in treatment. The epithelial to mesenchymal transition (EMT), a biochemical process where cells lose epithelial features and gain mesenchymal traits, is linked to chemoresistance and metastasis. Electrical impedance spectroscopy (EIS), a novel label-free electrokinetic technique, offers promise in detecting cell phenotype changes. In this study, we employed EIS to detect EMT in prostate cancer cells (PCCs). PC3, DU145, and LNCaP cells were treated with an EMT induction media for five days. EIS characterization revealed unique impedance spectra correlating with metastatic potential, distinguishing DU145 EMT+ and EMT- cells, and LNCaP EMT+ and EMT- cells (in combination with dielectrophoresis), with comparisons made to epithelial and mesenchymal controls. These changes were supported by shifts in electrical signatures, morphological, and protein expression, including downregulation of E-cadherin and upregulation of vimentin. No phenotype change was observed in PC3 cells, which maintained a mesenchymal phenotype. EMT+ cells were also distinguishable from mixtures of EMT+ and EMT- cells. This study demonstrates key advancements: application of EIS and dielectrophoresis for la-bel-free EMT detection in PCCs, characterization of cell electrical signature after EMT, and EIS sensitivity to EMT transitions. Detecting EMT in PCa is important to the development of more effective treatments and overcoming the challenges of chemoresistance.

Abstract:Exosomes, as a non-invasive biomarker, perform an important role in breast cancer screening and prognosis monitoring. However, establishing a simple, sensitive, and reliable exosome analysis technique remains challenging. Herein, a one-step multiplex analysis electrochemical aptasensor based on a multi-probe recognition strategy was constructed to analyze breast cancer exosomes. HER2-positive breast cancer cell (SK-BR-3) exosomes were selected as the model targets and three aptamers including CD63, HER2 and EpCAM aptamers were used as the capture units. Methylene blue (MB) functionalized HER2 aptamer and ferrocene (Fc) functionalized EpCAM aptamer, which were modified on gold nanoparticles (Au NPs), i.e. MB-HER2-Au NPs and Fc-EpCAM-Au NPs, were used as signal units. When the mixture of target exosomes, MB-HER2-Au NPs and Fc-EpCAM-Au NPs were added on the CD63 aptamer modified gold electrode, two Au NPs modified by MB and Fc could be specifically captured on the electrode by the recognition of three aptamers with target exosomes. Then one-step multiplex analysis of exosomes was achieved by detecting two independent electrochemical signals. This strategy can not only distinguish breast cancer exosomes from other exosomes (including normal exosomes and other tumor exosomes) but also HER2-positive breast cancer exosomes and HER2-negative breast cancer exosomes. Besides, it had high sensitivity and can detect SK-BR-3 exosomes with a concentration as low as 3.4 × 103 particles mL-1. Crucially, this method can be applicable to the examination of exosomes in complicated samples, which is anticipated to afford assistance for the screening and prognosis of breast cancer.

Abstract:Breast cancer is one of the leading causes of cancer-related death. An effective diagnostic system that enables early cancer detection is required for timely diagnosis and better treatment outcomes. Here, we developed an ultrasensitive electrochemical aptasensor for the multiplex detection of exosome biomarkers based on the electrochemical signals of metal ions. Specifically, a screen-printed carbon electrode (SPCE) was first modified with a multi-walled carbon nanotube (MWCNT), ionic liquid (IL), and chitosan (CHT) composite, and then gold nanoparticles (GNPs) were deposited via electrodeposition (GNPs/MWCNT-IL-CHT). To capture target exosomes, an aptamer specific for CD63, the universal exosome surface protein, was immobilized on the GNPs/MWCNT-IL-CHT/SPCE. When EpCAM or HER-2 positive exosomes were present in the sample, they could bind to EpCAM or HER-2 aptamers with primer sequences that acted as a rolling circle amplification reaction initiator, thereby generating numerous poly-guanine and poly-thymine repeats of a metal ion binding sequence, which produced strong electrochemical signals upon complexation with copper and lead ions. Using the proposed, multiplex exosome analysis system, EpCAM- and HER-2-positive exosomes were simultaneously detected with high specificity and a detection limit of 1 particle mL-1. In addition, its clinical applicability was validated via spike-and-recovery experiments using human serum samples.
Reusable and Dual-Potential Responses Electrogenerated Chemiluminescence Biosensor for Synchronously Cytosensing and Dynamic Cell Surface N-glycan Evaluation.

Abstract:A novel reusable and dual-potential responsive electrogenerated chemiluminescence (ECL) biosensor was fabricated for synchronous detection of cancer cells and their surface N-glycan. In this strategy, a cancer cell recognized aptamer hybridized with a capture DNA was immobilized on electrochemically reduced MoS2 nanosheets, and Ru(phen)3(2+) as ECL probes was intercalated into the grooves of the double-strand DNA. In the presence of target cells, the capture DNA and Ru(phen)3(2+) were released from the electrode interface owing to the specific interaction between cancer cells and the aptamer. Meanwhile, concanavalin A (Con A), a mannose binding protein, and a conjugated gold nanoparticle modified graphite-C3N4 (Con A@Au-C3N4) was used as a negative ECL nanoprobe and applied for the cell surface N-glycan evaluation owing to the excellent ECL properties of g-C3N4 at negative potential. The cytosensing and cell surface N-glycan evaluation could be simultaneously realized with high sensitivity and excellent selectivity based on the ratio of ECL intensity between the negative potential and positive potential (ΔECLn/ΔECLp), avoiding the traditional routing cell counting procedures. Moreover, the aptamer modified electrode can be regenerated in the presence of capture DNA solutions for cyclic utilization. As a proof-of-concept, the ECL cytosensor showed excellent performances for the analysis of the MCF-7 cancer cell and its surface N-glycan evaluation in human serum samples. The reusable and dual potential response ECL biosensor endows a feasibility tool for clinical diagnosis and drug screening especially in complex biological systems.

Abstract:Epithelial-mesenchymal transition (EMT) is closely related to tumor metastasis and invasion. Thereinto, mesenchymal tumor mitochondria are the critical target for tumor inhibition. Therefore, real-time in vivo monitoring of EMT as well as inhibiting mesenchymal tumor mitochondria is of great diagnosis and therapy significance. Herein, we construct a multi-stage recognition and morphological transformable self-assembly-peptide nano biosensor NDRP which can response the EMT marker and specifically damage the mesenchymal tumor cell in vivo. This nano-molar-affinity sensor is designed and screened with sensitive peptides containing a molecular switching which could be specifically triggered by the receptor to achieve the vesicle-to-fibril transformation in living system with enhanced fluorescent signal. Isolating and targeting the real-time plasticity and malignant properties of epithelial-mesenchymal transition in cancer

Abstract:Epithelial-mesenchymal transition (EMT) is a driving force in promoting malignant cancer, including initiation, growth, and metastasis. EMT is a dynamic process that can undergo a mesenchymal-epithelial transition (MET) and partial transitions between both phenotypes, termed epithelial-mesenchymal plasticity (EMP). In cancer, the acquisition of EMP results in a spectrum of phenotypes, promoting tumor cell heterogeneity and resistance to standard of care therapy. Here we describe a real-time fluorescent dual-reporter for vimentin and E-cadherin, biomarkers of the mesenchymal and epithelial cell phenotypes, respectively. Stable dual-reporter cell lines generated from colorectal (SW620), lung (A549), and breast (MDA-MB-231) cancer demonstrate a spectrum of EMT cell phenotypes. We used the dual-reporter to isolate the quasi epithelial, epithelial/mesenchymal, and mesenchymal phenotypes. Although EMT is a dynamic process, these isolated quasi-EMT-phenotypes remain stable to spontaneous EMP in the absence of stimuli and during prolonged cell culture. However, the quasi-EMT phenotypes can readily be induced to undergo EMT or MET with growth factors or small molecules. Moreover, isolated EMT phenotypes display different tumorigenic properties and are morphologically and metabolically distinct. 3D high-content screening of ~23,000 compounds using dual-reporter mesenchymal SW620 tumor organoids identified small molecule probes that modulate EMT, and a subset of probes that effectively induced MET. The tools, probes, and models described herein provide a coherent mechanistic understanding of mesenchymal cell plasticity. Future applications utilizing this technology and probes are expected to advance our understanding of EMT and studies aimed at therapeutic strategies targeting EMT.

Abstract:The epithelial-mesenchymal transition (EMT) process plays a pivotal role in acquiring invasive and metastatic properties and has been recognized as a crucial driver of epithelial-derived tumor malignancies. It is necessary to determine the role of EMT in promoting or suppressing carcinoma progression through investigating the relationship between EMT and apoptosis. We designed a multicolor fluorescent nanoprobe for simultaneously imaging the epithelial biomarker E-cadherin mRNA, the mesenchymal marker vimentin mRNA, and the apoptotic marker caspase-3. EMT and apoptosis progresses could be visually detected, which were used to study the effect of EMT on apoptosis and further assess the influence of EMT on drug efficacy in different cancer cells. We believe the designed nanoprobe can offer a new strategy for visualizing EMT and apoptosis in tumor cells and will be a promising tool to investigate the efficiency of drugs targeting EMT-related therapies in living cells.

Abstract:The development of sensitive and efficient cell sensing strategies to detect circulating tumor cells (CTCs) in peripheral blood is crucial for the early diagnosis and prognostic assessment of cancer clinical treatment. Herein, an array of hierarchical flower-like gold microstructures (HFGMs) with anisotropic nanotips was synthesized by a simple electrodeposition method and used as a capture substrate to construct an ECL cytosensor based on the specific recognition of target cells by aptamers. The complex topography of the HFGMs array not only catalyzed the enhancement of ECL signals, but also induced the cells to generate more filopodia, improving the capture efficiency and shortening the capture time. The effect of topographic roughness on cell growth and adhesion propensity was also investigated, while the cell capture efficiency was proposed to be an important indicator affecting the accuracy of the ECL cytosensor. In addition, the capture of cells on the electrode surface increased the steric hindrance, which caused ECL signal changes in the Ru(bpy)32+ and TPrA system, realizing the quantitative detection of MCF-7 cells. The detection range of the sensor was from 102 to 106 cells mL-1 and the detection limit was 18 cells mL-1. The proposed detection method avoids the process of separation, labeling and counting, which has great potential for sensitive detection in clinical applications.

Abstract:Here we developed a novel hybrid bipolar electrode (BPE)-electrochemiluminescence (ECL) biosensor based on hybrid bipolar electrode (BPE) for the measurement of cancer cell surface protein using ferrocence (Fc) labeled aptamer as signal recognition and amplification probe. According to the electric neutrality of BPE, the cathode of U-shaped ITO BPE was electrochemically deposited by Au nanoparticles (NPs) to enhance its conductivity and surface area, decrease the overpotential of O-2 reduction, which would correspondingly increase the oxidation current of Ru(bpy)(3)(2+)/tripropylamine (TPA) on the anode of BPE and resulting a similar to 4-fold enhancement of ECL intensity. Then a signal amplification strategy was designed by introducing Fc modified aptamer on the anode surface of BPE through hybridization for detecting the amount of mucin-1 on MCF-7 cells. The presence of Fc could not only inhibit the oxidation of Ru(bpy)(3)(2+) because of its lower oxidation potential, its oxidation product Fc(+) could also quench the ECL of Ru(bpy)(3)(2+)/TPA by efficient energy-transfer from the excited-state Ru(bpy)(3)(2+)* to Fc(+), making the ECL intensity greatly quenched. On the basis of the cathodic Au NPs induced ECL enhancing coupled with anodic Fc induced signal quenching amplification, the approach allowed detection of mucin-1 aptamer at a concentration down to 0.5 fM and was capable of detecting a minimum of 20 MCF-7 cells. Besides, the amount of mucin-1 on MCF-7 cells was calculated to be 9041 +/- 388 molecules/cell. This approach therefore shows great promise in bioanalysis.
