This is likely due to the smaller size/higher specific surface area of nanoplastics which works more efficiently as a vector for other contaminants' transportation and fate, to release the formulation additives and to be accumulated in the human body. The research on nanoplastics is currently hampered by the absence of an effective characterisation method, although X-ray photoelectron spectroscopy/Scanning Electron Microscopy (XPS/SEM) is reportedly dominating the nanoplastics characterisation approaches. Recently, imaging technology, which provides direct visualisation of the targets, is attracting increased attention. In this study, we demonstrate that Raman imaging can be employed to visualise and identify microplastics and nanoplastics down to 100 nm, by distinguishing the laser spot, the pixel size/image resolution, the nanoplastics size/position (within a laser spot), the Raman signal intensity, and via the sample preparation etc. We also validate this technique for the analysis of paint-polishing dust samples that have been collected from a driveway when a vehicle's clear coating of poly-acrylic was polished by hand. We estimate that billions-trillions of microplastics/nanoplastics ranging from similar to 7 mu m down to similar to 200 nm have been generated by hand-polishing an engine hood. (C) 2020 Elsevier Ltd. Airborne Polystyrene Microplastics and Nanoplastics Induce Nasal and Lung Microbial Dysbiosis in Mice.

Abstract:Microplastics (MP) and nanoplastics (NP) have been found in multiple environments and creatures. However, their effects on the airway microbiota still remain poorly understood. In this study, a series of bioinformatic and statistical analyses were carried out to explore the influence of airborne MP and NP on the nasal and lung microbiota in mice. Both MP and NP were capable of inducing nasal microbial dysbiosis, and MP had a stronger influence on the lung microbiota than NP. Multiple nasal and lung bacteria were associated with MP and NP groups, among which nasal Staphylococcus and lung Roseburia were most associated with MP group, while nasal Prevotella and lung unclassified_Muribaculaceae were most associated with NP group. The nasal Staphylococcus, lung Roseburia, lung Eggerthella and lung Corynebacterium were associated with both MP and NP groups, which were potential biomarkers of micro/nanoplastics-induced airway dysbiosis. SAR11_Clade_Ia and SAR11_Clade_II were associated with both nasal and lung microbiota in MP group, while no such bacterium was determined in NP group. The relevant results suggest that both airborne MP and NP could induce nasal and lung microbial dysbiosis, and the relevant preventative and curable strategies deserve further investigations.
Measurement of microplastic release after use of polypropylene nasal irrigation bottles

Abstract:Objectives: Microplastics from plastic materials can affect human health. This study aimed to assess their presence in nasal irrigation fluids obtained from reused bottles based on usage duration. Methods: Readily available nasal irrigation bottles made of polypropylene were purchased. Unused irrigation bottles served as controls. Bottles mimicking 1-, 3-, and 6-month reuse were prepared as test samples. Nasal irrigation fluid samples (n=12) were collected from each set of irrigation bottles: three from new control bottles and nine from bottles mimicking 1-, 3-, and 6-month reuse. Raman spectroscopy was employed to detect microplastics in the nasal irrigation samples; the results were compared according to the bottle use duration. Results: An average of 33.00±20.42 (ea/300 mL) microplastic particles was detected in the nasal irrigation fluid from the control bottles relative to an average of 68.66±30.07, 261.66±20.59, and 204.33±52.16 (ea/300 mL) from bottles used for 1, 3, and 6 months, respectively. The majority of the detected microplastics were 10-100 μm in size and fragmentshaped in form. The predominant microplastic was polypropylene, indicating direct release from irrigation bottles. Conclusion: We found microplastics in nasal irrigation fluids, likely originating from the repeated use of nasal irrigation bottles. The quantity of microplastics was notably elevated in the samples obtained from bottles simulating 3 months of use compared to those in the control samples. Thus, we propose developing guidelines regulating the duration of nasal irrigation bottle usage to mitigate microplastic infiltration into the body through the sinonasal cavity.

Abstract:Plastic is produced, used, and accumulated exponentially. Humans can take it through the food chain, and microplastics can be potentially toxic to humans. In this study, we analyzed the surface patterns of microplastics and their effects on humans using polyvinyl chloride (PVC) and acrylonitrile butadiene styrene (ABS). The surface analysis of these microplastics will be used as a standardized measurement method for microplastic detection and will be used as an indicator of the impact of environmental pollution caused by microplastics on humans. We have used a statistical method to analyze the shape of the milled microplastics and estimated the local curvature for each plastic type and found that PVC microplastics have a more rugged shape than the ABS microplastics. We also analyzed the toxicity of PVC and ABS microplastics. Larger PVC tended to induce the release of interleukin 6 (IL-6) and tumor necrosis factor-α (TNF-α). Smaller ABS induced the release of IL-6 at high concentrations. However, larger ABS tended to induce the release of TNF-α at all concentrations. Thus, we concluded that microplastics can induce immune responses.This article has not yet been cited by other publications.

Abstract:Cloning of the opioid receptor family has added a new dimension to opioid pharmacology. In common with the endogenous opioid subtypes, recombinant opioid receptors, when expressed in a variety of cell types, couple to K channels, Ca channels and adenylyl cyclase. The net effect in the CNS is reduction in neuronal firing and excitability. If this occurs in neurones involved in the processing of noxious stimuli then analgesia results. Despite these monumental advances the clinical “holy grail” of analgesia without respiratory depression remains to be realized. In this editorial we summarize briefly a new development in opioid pharmacology—isolation of a novel receptor involved in pain processing and discovery of an endogenous ligand for this receptor. Shortly after the publication of the and opioid receptor sequences several groups reported the isolation of an atypical “opioid-like” receptor subtype with approximately 50% homology to and opioid receptors. There is general consensus that these different clones represent the same receptor. From sequence and structural analysis the receptor is predicted to be a new member of the G-protein coupled receptor superfamily and is known euphemistically as the orphan opioid receptor (ORL1). A common characteristic of the orphan receptor was that opioid receptor ligands bound with very low affinity compared with opioid receptors. In the rat, orphan receptors are found “scattered” throughout the CNS with high levels found in the thalamus, hypothalamus, amygdala, cortex (diffuse but layered), hippocampus, periaqueductal gray, dorsal raphe, locus coeruleus and spinal cord. At the time of cloning no endogenous ligand had been isolated. In November 1995, Reinscheid and colleagues and Meunier and colleagues reported the isolation of a heptadecapeptide (fig. 1) that was named orphanin FQ or nociceptin (throughout the remainder of this editorial orphanin FQ (OFQ) is used and refers to the full 17 amino acid peptide). The heptadecapeptide displayed nanomolar binding affinity for the recombinant ORL1 receptor in radioligand binding studies. The first amino acid differed in OFQ compared with opioid peptides, but of these peptides its closest match was with dynorphin (fig. 1). When injected intracerebroventricularly (i.c.v.) (i.e. supraspinally) in the mouse, the peptide appeared to be hyperalgesic. 13 In addition, it has been reported recently that OFQ is a functional opioid antagonist. OFQ was shown to completely block supraspinal antinociception in mice mediated by selective and agonists. 15 Stanfa and colleagues reported that OFQ inhibited C-fibreevoked wind-up and post-discharge of dorsal horn neurones, concluding that OFQ was antinociceptive at the spinal level. These differences at spinal and supraspinal sites clearly need further exploration. Unlike receptor activation orphan receptor activation with OFQ (and hence a clinical orphan receptor agonist when available) does not produce dependence and as such has little or no abuse potential. The cellular effects of OFQ have received some attention and have been shown to inhibit cAMP formation. 13 In addition, OFQ increases inwardly rectifying K currents and inhibits voltage-sensitive Ca channels. 22 Activation of K channels, closing of voltage-sensitive Ca channels and inhibition of adenylyl cyclase are features common to opioid receptors. We have demonstrated recently that OFQ inhibits the release of glutamate from rat cerebrocortical slices in a naloxone-insensitive manner. These data are in agreement with the inhibitory effects of OFQ on glutamatergic transmission in the rat spinal cord. If this depression in glutamatergic transmission is mirrored in the hippocampus then this might provide an explanation for the effects of OFQ on spatial learning. Similarly, if observed in the cerebellum, this may go some way to explain the effects of OFQ on locomotion. 25 In addition to glutamate, OFQ also inhibits the release of tachykinin, acetylcholine 28 and dopamine. These data suggest the presence of presynaptic ORL1 receptors involved in neurotransmitter release. Is OFQ released in significant quantities to provide inhibition of release under physiological conditions? This is another important issue that needs to be resolved in the near future. The recent development of a sensitive radioimmunoassay should prove useful in this regard.

Abstract:BACKGROUND: Microplastics are able to enter the human body through food chains, personal care products, contact (skin) from the soil, water or inhalation of microplastics in the air. AIM: This article aims to identify the presence of microplastics that are exposed to humans. METHODS: Systematic review using four bibliographic databases (Google Scholar, Wiley Online Library, PubMed, and Science Direct). Literature search from 2016 to 2021, in the search found as many as 10983 papers extracted using PRISMA guidelines, a total of five papers were selected for review. RESULTS: In this review article, the presence of oral, dermal, and inhalation exposed microplastics is found in feces, colon, placenta, scalp hair, hand skin, facial skin, and saliva. Several methods are used to identify microplastics according to polymer types, shape, and sizes microplastics in humans. The polymer type often found in the article is polypropylene. In addition, fibers, fragments and films are detectable shape of microplastic, while the size of microplastics >5 mm. CONCLUSION: This literature study provides evidence that microplastics of various types of polymers, shapes and sizes are ubiquitous in humans. Hence, the possibility of health risks resulting from microplastic exposure in the human body should not be ignored.

Abstract:Micro(nano)plastics are considered an emerging threat to human health because they can interact with biological systems. In fact, these materials have already been found in the human body, such as in the lungs. However, limited data are available on the behavior of these materials under biological conditions and their impact on human cells, specifically on alveolar epithelial cells. In this study, micro(nano)plastics were exposed to various simulated biological fluids (artificial lysosomal fluids and Gamble's solution) for 2–80 h. Pristine and treated plastic particles were characterized based on their surface chemistry, zeta potentials, and elemental composition. Various toxicological endpoints (mitochondrial membrane potential, lactate dehydrogenase, protein, and antioxidant levels) were examined using A549 lung carcinoma cells. The surface characteristics of the treated micro(nano)plastics and the toxicological endpoints of A549 cells were found to be influenced by the simulated biological media, specifically with high concentrations of the treated micro(nano)plastics and increasing exposure under biological conditions. Moreover, the toxicological endpoints were strongly linked to the chemistry of plastics and included multiple processes in response to the plastics; different biological pathways were obtained in artificial lysosomal fluid and Gamble's solution.

Abstract:Microplastics (MPs) (including nanoplastics (NPs)) are pieces of plastic smaller than 5 mm in size. They are produced by the crushing and decomposition of large waste plastics and widely distributed in all kinds of ecological environments and even in organisms, so they have been paid much attention by the public and scientific community. Previously, several studies have reviewed the sources, occurrence, distribution, and toxicity of MPs in water and soil. By comparison, the review of atmospheric MPs is inadequate. In particular, there are still significant gaps in the quantitative analysis of MPs and the mechanisms associated with the toxic effects of inhaled MPs. Thus, this review summarizes and analyzes the distribution, source, and fate of atmospheric MPs and related influencing factors. The potential toxic effects of atmospheric MPs on animals and humans are also reviewed in depth. In addition, the common sampling and analysis methods used in existing studies are introduced. The aim of this paper is to put forward some feasible suggestions on the research direction of atmospheric MPs in the future.

Abstract:Plastics have enormous impacts to every aspect of daily life including technology, medicine and treatments, and domestic appliances. Most of the used plastics are thrown away by consumers after a single use, which has become a huge environmental problem as they will end up in landfill, oceans and other waterways. These plastics are discarded in vast numbers each day, and the breaking down of the plastics from micro- to nano-sizes has led to worries about how toxic these plastics are to the environment and humans. While, there are several earlier studies reported the effects of micro- and nano-plastics have on the environment, there is scant research into their impact on the human body at subcellular or molecular levels. In particular, the potential of how nano-plastics move through the gut, lungs and skin epithelia in causing systemic exposure has not been examined thoroughly. This review explores thoroughly on how nanoplastics are created, how they behave/breakdown within the environment, levels of toxicity and pollution of these nanoplastics, and the possible health impacts on humans, as well as suggestions for additional research. This paper aims to inspire future studies into core elements of micro- and nano-plastics, the biological reactions caused by their specific and unusual qualities.

Abstract:We are directly exposed to microplastic contamination via indoor air that we breathe daily, for which the characterisation of microplastics is still a challenge. Herein, two typical air filter samples were collected, one from an air-conditioner and another from a personal computer, both of which have been working for around half a year to collect and accumulate microplastics in the indoor air, like microplastic banks. After the sample preparation to remove the mineral dusts, Raman imaging was employed to directly and simultaneously identify and visualise microplastics of polyethylene terephthalate (PET) fibres, distinguish them from other fibres such as cellulose and cross-check them with a scanning electron microscope (SEM). To count the microplastics and to avoid the quantification bias, several areas were randomly scanned and imaged to statistically estimate the percentage of microplastic fibres in the analysed samples. The microplastics amount, which has been estimated at 73-88,000 fibers per filter per half a year, varies and depends on the indoor environment so that the air filter can work as a good indicator to monitor the quality of the indoor air from the microplastic perspective. Overall, human are directly exposed to this emerging contamination every day, raising environmental concerns. Raman imaging characterisation and its corresponding statistical information can help pursue further research on microplastics.

Abstract:Micro (nano) plastics, MNPLs, have become emerging particulate human-made pollutants, and quickly become the fields of increasing concern of the public. Due to the lack of analytical technology with sufficient sensitivity and selectivity, it is difficult to fully detect and determine the characteristics of MNPLs in the environment and biological matrix. Atomic spectrometry has many advantages, such as low detection limit and high sensitivity, and shows great potential in the analysis of MNPLs. However, there is a lack of systematic summary in this field at present in order to give full play to its advantages in the analysis of plastics. In this review, the whole process of analyzing MNPLs in complex matrix based on various atomic spectroscopy techniques have been discussed, including sample labeled, treatment, purification, and detection. Furthermore, the application of atomic spectrometry in tracing the source of MNPLs in the environment, in situ identification and classification, analysis surface morphology and functional groups changes during aging process, assessments of environmental ecological risk assessment and toxicological effects were addressed aiming to improve our understanding on the environmental fate and transport of the fragments. Finally, future outlooks and research directions have been recommended based on the existing research gaps in the area.
Hormone concentrations and ovulatory response in rats treated with antiprogestagens.

Abstract:Administration of antiprogestagens (2 mg/day) to female rats for 21 days induces high serum prolactin levels. These levels stimulate luteal progesterone production and an increase in ovarian weight. Compared with RU486 (mifepristone) the increase in prolactin is less after treatment with ZK299 (onapristone), an antiprogestagen with lower antiglucocorticoid activity. To study whether cyclic ovulations occur in rats treated with antiprogestagens, 5-day cyclic rats were given daily injections of RU486 or ZK299 (2 mg) from metoestrus (day 1) to pro-oestrus. This treatment advanced the forthcoming ovulation by 1 day; however, the ovulation rate was low. Injection of 10 IU human chorionic gonadotrophin on the afternoon of pro-oestrus (day 3) increased the ovulation rate, but not to the level found in oil-treated rats. Serum LH concentrations measured from metoestrus to oestrus at 10.00 and 17.00 h were higher in antiprogestagen- than in oil-treated rats from day 2 (17.00 h) onwards. A low preovulatory LH surge was found in antiprogestagen-treated rats on the afternoon of pro-oestrus (day 3). Ovarian histology at the day of oestrus (day 4) confirmed the presence of a low LH surge as, besides ruptured follicles, unruptured follicles with dispersion of cumulus cells were present. The pro-oestrus surge of prolactin was also advanced by 24 h. The magnitude, however, was not different from that in oil-treated rats at day 4.(ABSTRACT TRUNCATED AT 250 WORDS)

Abstract:Plastics are widely utilized in industry, agriculture and our daily life due to their low cost and high corrosion resistance, leading to substantial plastic waste in the environment. Under environmental stressors, plastics degrade into micro- and nano-plastics (MNPs) with various size, shape, morphology and composition. However, detailed studies on the methods for collecting, treating, and detecting MNPs remain limited. Herein, the review firstly summarizes the sampling procedures of MNPs from environmental and biological compartments. Additionally, methods for the extraction and separation of MNPs are summarized and discussed, followed by a comprehensive overview of the detection methods used for MNPs identification and quantification. Beyond microplastics - investigation on health impacts of submicron and nanoplastic particles after oral uptake in vitro

Abstract:Abstract The continuously increasing use of plastics is supposed to result in a rising exposure of MNPs to humans. Available data on human health risks of microplastics after oral uptake increased immensely in the past years and indicates very likely only low risks after oral consumption. Concerning nanoplastics, uptake, transport and potential adverse effects after oral uptake are less well understood. This study aims to investigate differences between microplastic particles and particles in the submicron- and nanoscaled size derived from food-relevant polymers with a particle size range consistent with higher potential for cellular uptake, fate, and effects when applied to human intestinal and liver cells. This work includes the development of cellular and subcellular detection methods for synthetic polymeric particles in the micro- and nanometer-range, using Scanning Electron Microscopy, Small-Angle X-ray and Dynamic Light Scattering methods, Asymmetric Flow Field Flow Fractionation, octanol-water fractionation, fluorescence microscopy and flow cytometry. Polylactic acid (250 nm and 2 μm (polydisperse)), melamine formaldehyde (366 nm) and polymethylmethacrylate (25 nm) were thoroughly characterized. The submicro- and nanoplastic test particles showed an increased uptake and transport quantity through intestinal cells. Both types of particles resulted in observed differences of uptake behavior, most likely influenced by different lipophilicity, which varied between the polymeric test materials. Toxic effects were detected after 24 h only in overload situations for the particles in the submicrometer range. This study provides further evidence for gastrointestinal uptake of submicro- and nanoplastics and points towards differences regarding bioavailability between microplastics and smaller plastic particles that may result following the ingestion of contaminated food and beverages. Furthermore, the results reinforce the importance for studying nanoplastics of different materials of varying size, surface properties, polymer composition and hydrophobicity.

Abstract:Microplastics (MPs) are plastic residues with a size <5 mm, which are often further broken into micro/nano size particles in the environment. Owing to their minute scale, widespread distribution, and substantial toxicity potential, microplastics has emerged as a critical environmental concern. Therefore, accurate characterization of these particles becomes a formidable yet imperative challenge in environmental science. In this review, a comprehensive overview on current microplastic characterization techniques, including microscopy/electron microscopy, spectroscopy, and mass spectrometry, have been summarized and discussed. We assess their applicability, strengths, limitations, as well as recent advancements. We also highlight the features offered by different techniques and the particles size range for which each technique is suitable. Furthermore, the combination of different characterization techniques and some novel techniques that can be used in microplastic characterization are also mentioned. This work offers a reference for microplastic characterization, which will facilitate the detection of microplastic in the environment.

Abstract:The ubiquitous occurrence of micro/nano plastics (MNPs) poses potential threats to ecosystem and human health that have attracted broad concerns in recent decades. Detection of MNPs in several remote regions has implicated atmospheric transport as an important pathway for global dissemination of MNPs and hence as a global health risk. In this review, the latest research progress on (1) sampling and detection; (2) origin and characteristics; and (3) transport and fate of atmospheric MNPs was summarized. Further, the current status of exposure risks and toxicological effects from inhaled atmospheric MNPs on human health is examined. Due to limitations in sampling and identification methodologies, the study of atmospheric nanoplastics is very limited today. The large spatial variation of atmospheric MNP concentrations reported worldwide makes it difficult to compare the overall indoor and outdoor exposure risks. Several in vitro, in vivo, and epidemiological studies demonstrate adverse effects of immune response, apoptosis and oxidative stress caused by MNP inhalation that may induce cardiovascular diseases and reproductive and developmental abnormalities. Given the emerging importance of atmospheric MNPs, the establishment of standardized sampling-pretreatment-detection protocols and comprehensive toxicological studies are critical to advance environmental and health risk assessments of atmospheric MNPs.
