Abstract:The synergistic cooperation of microbial cells and their extracellular polymeric substances (EPS) in biofilms is critical for the biofilm's resistance to heavy metals and the migration and transformation of heavy metals. However, the effects of different components of biofilms have not been fully understood. In this study, the spatial distribution and speciation of copper in the colloidal EPS, capsular EPS, cell walls and membranes, and intracellular fraction of unsaturated Pseudomonas putida (P. putida) CZ1 biofilms were fully determined at the subcellular level. It was found that 60-67% of copper was located in the extracellular fraction of biofilms, with 44.7-42.3% in the capsular EPS. In addition, there was 15.5-20.1% and 17.2-21.2% of copper found in the cell walls and membranes or the intracellular fraction, respectively. Moreover, an X-ray absorption fine structure spectra analysis revealed that copper was primarily bound by carboxyl-, phosphate-, and hydrosulfide-like ligands within the extracellular polymeric matrix, cell walls and membranes, and intracellular fraction, respectively. In addition, macromolecule quantification, fourier-transform infrared spectroscopy spectra and sulfur K-edge x-ray absorption near edge structure analysis further showed the carboxyl-rich acidic polysaccharides in EPS, phospholipids in cell walls and cell membranes, and thiol-rich intracellular proteins were involved in binding of copper in the different components of biofilm. The full understanding of the distribution and chemical species of heavy metals in biofilms not only promotes a deep understanding of the interaction mechanisms between biofilms and heavy metals, but also contributes to the development of effective biofilm-based heavy metal pollution remediation technologies. Interaction Between Copper and Extracellular Nucleic Acids in the EPS of Unsaturated Pseudomonas Putida CZ1 Biofilm

Abstract:The role of extracellular DNA (eDNA) in biofilm in heavy metal complexation has been little reported. In this study, the interaction between the extracellular fraction of unsaturated biofilms and Cu2+ was studied using random amplified polymorphic DNA (RAPD) and synchrotron-based X-ray absorption spectroscopy (XAS) analyses. Under Cu2+ stress, the amount of eDNA was about 10-fold higher than the treatment without Cu2+ stress, which was substantially more than the amount of intracellular DNA (iDNA) present in the biofilm. The eDNA content increased significantly under Cu2+ stress and higher eDNA contents were found in colloidal extracellular polymeric substances (EPS) than in capsular EPS in Luria-Bertani medium. It was found that the composition of eDNA was distinctly changed under conditions of Cu2+ stress compared with the treatments without Cu2+ treatments, with specific eDNA bands appearing under Cu2+ treatments as revealed by RAPD analyses. X-ray absorption fine structure (XAFS) analysis assessing the molecular speciation of copper showed that copper in the secreted eDNA mainly existed as species resembling Cu3(PO4)2, followed by Cu-citrate species. This study investigated the interaction between copper and eDNA in unsaturated Pseudomonas putida CZ1 biofilms. Potential function of eDNA in biofilms under Cu2+ stress was found.

Abstract:Heavy metals (HMs) have been widely reported to pose an adverse effect on anaerobic ammonia oxidation (anammox) bacteria, yet the underlying mechanisms remain unclear. This study provides new insights into the potential mechanisms of interaction between HMs and functional enzymes through big date analysis, molecular docking and molecular dynamics simulation. The statistical analysis indicated that 10 mg/L Cu(II) and Cd(II) reduced nitrogen removal rate (NRR) by 85% and 43%, while 5 mg/L Fe(II) enhanced NRR by 29%. Additionally, the results of molecular simulations provided a microscopic interpretation for these macroscopic data. Molecular docking revealed that Hg(II) formed a distinctive binding site on ferritin, while other HMs resided at iron oxidation sites. Furthermore, HMs exhibited distinct binding sites on hydrazine dehydrogenase. Concurrently, the molecular dynamics simulation results further substantiated their capacity to form complexes. Cu(II) displayed the strongest binding affinity with ferritin for −1576 ± 79 kJ/mol in binding free energy calculation. Moreover, Cd(II) bound to ferritin and HDH for −1052.67 ± 58.49 kJ/mol, −290.02 ± 49.68 kJ/mol, respectively. This research addressed a crucial knowledge gap, shedding light on potential applications for remediating heavy metal-laden industrial wastewater.

Abstract:Short-term and long-term effects of Cd(II) on wastewater biological nitrogen and phosphorus removal were investigated with respect to microorganism abundances, enzyme activities, and polyhydroxyalkanoates (PHAs) and glycogen transformations. Though no obvious effects on wastewater biological nutrient removal were observed after short-term exposure, the long-term exposure of 10 mg L(-)(1) Cd(II) inhibited nitrification and phosphorus uptake. Compared with the absence of Cd(II), the presence of 10 mg L(-1) of Cd(II) decreased total nitrogen and phosphorus removal efficiencies from 97% and 98% to 88% and 18%, respectively. Mechanism studies revealed that Cd(II) affected the transformations of intracellular PHAs and glycogen, and the activities of oxidoreductase and polyphosphate kinase, resulted in the decrease of nitrite oxidizing bacteria and polyphosphate accumulating organisms abundance, which might be the major reason for the negative effects of long-term exposure to 10 mg L(-1) Cd(II) on biological nitrogen and phosphorus removal.

Abstract:Extracellular polymeric substance (EPS) of anaerobic ammonium oxidation (anammox) sludge plays an important role in resisting the toxicity when treating heavy metal containing wastewater. Understanding the interaction mechanism between EPS and heavy metal is essential for stable and optimal operation of such technology. In this work, the identification and preferential response of the binding sites, as well as the binding thermodynamic and the conformational changes of EPS to Cu2+ were explored. Unlike the activated sludge system, functional groups in EPS would coordinate with Cu2+ to form inner sphere complexes, which is an endothermic and entropy-increasing process. Low concentration of Cu2+ was found to result in the reduced electrostatic repulsion and bridging effect, which promoted the compaction and agglomeration of EPS colloid. Two-dimensional correlation spectroscopy analysis confirmed that carboxyl groups of proteins were preferentially responded to Cu2+ compared to polysaccharides and hydrocarbons. More importantly, the secondary structure of the protein in EPS was disrupted and changed into loose state after combining with Cu2+. This work is of great significance for understanding the binding properties of anammox sludge EPS with Cu2+ and may provide theoretical support for elucidating the resist mechanism of anammox bacteria to heavy metals.

Abstract:In this study, the short- and long-term effects of Cd(II) on the bioactivity, nitrogen removal and microbial community of the anaerobic ammonia oxidation (Anammox) process were investigated by step-wise increasing Cd(II). The self-recovery ability of Anammox was also studied after long-term exposure in Cd(II). The bioactivity of anaerobic ammonia-oxidizing bacteria (AAOB) within short-term exposure of Cd(II) were detected by batch experiments. Results showed that Cd(II) had both acute and persistent toxicity to Anammox, the IC50 in short-term exposure was calculated as 5.43 mg L-1. Long-term exposure led to the microbial diversity increase, as well as the relative abundance decrease of AAOB in Anammox system. AAOB was continuously suppressed by Cd(II) in 1-20 mg L-1, while it had self-adaption to Cd(II) in 1-10 mg L-1. Anammox biofilm showed prominent adsorbing ability for cadmium, and the cadmium in biofilm was the key factor affecting Anammox.
Competition of Cd(II) and Pb(II) on the bacterial cells: a new insight from bioaccumulation based on NanoSIMS imaging

Abstract:ABSTRACT Polymetallic exposure causes complex toxicity to microorganisms. In this study, we investigated the responses of Escherichia coli under co-existence of cadmium (Cd) and lead (Pb), primarily based on biochemical analysis and RNA sequencing. Cd completely inhibited bacterial growth at a concentration of 2.41 mmol/L, with its removal rate as low as 95% under equimolar sole Pb stress. In addition, the Raman analysis confirmed the loss of proteins for the bacterial cells. Under the co-existence of Cd and Pb, the Cd toxicity to E. coli was alleviated. Meanwhile, the biosorption of Pb cations was more intense during the competitive sorption with Cd. Transmission electron microscopy images showed that a few cells were elongated during incubation, i.e., the average cellular length increased from 1.535 ± 0.407 to 1.845 ± 0.620 μm. Moreover, NanoSIMS imaging showed that the intracellular distribution of Cd and Pb was coupled with sulfur. Genes regulating sulfate transporter were also upregulated to promote sulfate assimilation. Then, the subsequent production of biogenic sulfide and sulfur-containing amino acids was enhanced. Although this strategy based on S enrichment could resist the polymetallic stress, not all related genes were induced to upregulate under sole Cd stress. Therefore, the S metabolism might remodel the microbial resistance to variable occurrence of heavy metals. Furthermore, the competitive sorption (in contrast to sole Cd stress) could prevent microbial cells from strong Cd toxicity. IMPORTANCE Microbial tolerance and resistance to heavy metals have been widely studied under stress of single metals. However, the polymetallic exposure seems to prevail in the environment. Though microbial resistance can alleviate the effects of exogenous stress, the taxonomic or functional response to polymetallic exposure is still not fully understood. We determined the strong cytotoxicity of cadmium (Cd) on growth, and cell elongation would be driven by Cd stress. The addition of appropriate lead (Pb) showed a stimulating effect on microbial bioactivity. Meanwhile, the biosorption of Pb was more intense during co-existence of Pb and Cd. Our work also revealed the spatial coupling of intracellular S and Cd/Pb. In particular, the S assimilation was promoted by Pb stress. This work elucidated the microbial responses to polymetallic exposure and may provide new insights into the antagonistic function during metal stresses.

Abstract:Cadmium (Cd) can cause the deterioration of biological systems through inhibiting the enzymes activity and disturbing the microbial metabolism. Although the influence of Cd on conventional wastewater treatment process has been studied, the response of anammox to Cd exposure still remains unclear. This study firstly investigated the adaption and restoration of anammox biomass to Cd(II) stress. Results showed that long-term exposure of anammox bacteria to 2 mg L-1 Cd(II) was beneficial for the reactor performance, while 5 mg L-1 Cd(II) would cause the decline of SAA, extracellular polymeric substance content and relative abundance of Candidatus kuenenia by 40%, 25% and 31%, respectively. Furthermore, these indexes could approximately recover to the initial status after withdrawing Cd(II) from the influent. Overall, the anammox biomass exhibited a certain adaption and restoration ability to the suppression of Cd(II). This study may provide key valuable information for the biological treatment of wastewater containing Cd(II).

Abstract:The effect of heavy metals on soluble microbial products (SMPs) have been widely studied in the activated sludge system. However, their impact on the biofunctions of SMPs remain unclear, which could reflect state of cellular metabolism. In present work, we investigated toxic impact of Cd 2+ and Cu 2+ on properties of soluble microbial products (SMPs) and cellular metabolism activities via proteomics analysis and genotoxicity. The results indicated all sized fractions in SMPs have been increased by addition of Cd 2+ or Cu 2+ through altering several cellular metabolic pathways; especially for the appearance of Cu 2+ . The toxicity of Cd 2+ impacted cellular substrate consumption, energy generation, and electron transport activities via changing metabolism and/or degradation of amino acid, while the addition of Cu 2+ threated transfer of genetic information and cellular signals, energy generation and storage, and activity of enzymes via affecting metabolic activities of ribonucleic acid, purine, and pyrimidine. Meanwhile, ribosomes played essential roles in toxic resistance under Cd 2+ shock, ATP-binding cassette (ABC) transporters functioned in toxic resistance under Cu 2+ shock. In addition, the expression of functional genes grpE , dnaJ , ibpB , and ytfE were specially regulated by Cd 2+ shock, and the expression of functional genes grxA , grxB , gst , ahpF , marC , dacB , and pbpG are particularly sensitive to the Cu 2+ stress; which confirmed differences in cellular response activities under stressful conditions of toxic Cd 2+ and Cu 2+ . Those findings provided key insights into how toxic metals change cellular metabolism at the molecular level, which would be meaningful to operational control and optimization of biological wastewater treatment.

Treatment of chronic myelogenous leukemia in blastic crisis with chemotherapy incorporating vindesine-prednisolone.

Abstract:Twenty-three patients with chronic myelogenous leukemia (CML) in terminal transformation were treated with a regimen incorporating vindesine (a new derivative of vinblastine), prednisolone, and additional cytarabine. Of 21 patients in blastic crisis, complete remission (CR) was achieved in 15 (71.4%). All of five patients with lymphoid morphology and ten of 12 with myeloid morphology (83.3%) achieved CR, while none of four with monocytoid morphology achieved CR. Two patients in accelerated phase also attained CR. Our results compare favorably with those of other regimens in this refractory disorder and indicate that the regimens incorporating vindesine, prednisolone, and additional cytarabine may be recommended in the treatment of CML-blastic crisis with both lymphoid and myeloid morphology and accelerated phase of CML as well.

Abstract:Highly correlated photons or, accordingly, high-fidelity single-photon states are a prerequisite for closing detection loopholes in experimental tests of local realism and implementing scalable linear optical quantum computation. We demonstrate a parametric down-conversion source exhibiting a conditional detection efficiency of 51% (with corresponding preparation efficiency of 85%) and extraordinarily high detection rates of up to 8.5 x 10(5) coincidences/(s mW). We exploit a novel type-II phase matching configuration in a microstructured waveguide in conjunction with an ultrashort pump.

Abstract:Cadmium (II) can potentially alter the microbial community structure and molecular ecological network in activated sludge systems. In this study, we used Illumina sequencing combined with an RMT-based network approach to show the response of the microbial community and its network structure to Cd (II) in activated sludge systems. The results demonstrated that 1 mg/L Cd (II) did not have chronic negative effects on chemical oxygen demand (COD) reduction and denitrification processes, but negatively affected the nitrification process and phosphorus removal. In contrast, 10 mg/L Cd (II) adversely affected both COD and nutrient removal, and reduced the microbial diversity and changed the overall microbial community structure. The relative abundances of Nitrosomonadaceae, Nitrospira, Accumulibacter and Acinetobacter, which are involved in nitrogen removal, significantly decreased with increases in the Cd (II) concentration. In addition, molecular ecological network analysis showed that the networks sizes in the presence of higher levels of Cd (II) were smaller than in the control, but the nodes were more closely connected with neighbors. These shifts in bacterial abundance and the bacterial network structure may be responsible for the deterioration of COD and nutrient removal. Overall, this study provides new insights into the effects of Cd (II) on the bacterial community and its interactions in activated sludge systems.

Abstract:The presence of organic-complexed copper and zinc in anaerobic digestate effluent (ADE) poses persistent ecological toxicity. This study investigated the detoxification performance and biotic responses of indigenous bacteria against ethylene diamine tetraacetic acid (EDTA)-complexed Cu(II) and Zn(II). Heavy metals (HMs) stress induced reactive oxygen species (ROS) generation and enhanced extracellular polymeric substances (EPS) secretion. At a Cu(II) influent concentration of 20.0 mg·L-1, indigenous bacteria removed 88.2% of Cu(II) within nine days. The majority of copper and zinc sequestered by bacteria were stored in the cell envelope, with over 50% of copper and 60% of zinc being immobilized. Transmission electron microscopy mapping (TEM-mapping) revealed significant mineralization of copper and zinc on the cell wall. Proteins abundant in EPS, alongside humic acid-like substances, effectively adsorbed HMs. Indigenous bacteria exhibited the capacity to reduce cupric to the cuprous state and cupric is preferentially reduced to cuprous before reaching reducing capacity saturation. Sulfur precipitation emerges as a crucial pathway for Zn(II) removal. Metagenomic analysis indicated that indigenous bacteria upregulated genes related to HMs homeostasis, efflux, and DNA repair, enhancing its resistance to high concentrations HMs. This study provided theoretical guidance for employing bacterial consortia to eliminate HMs in complex aquatic environments.

Abstract:The complexation with extracellular polymeric substances (EPS) greatly reduces the toxicity of heavy metals towards organisms in the environment. However, the molecular mechanism of EPS−metal complexation remains unclear owing to the limitation of precise analysis for key fractions and functionalities in EPS that associate with metals. Herein, we explored the EPS−Cd (II) complexation by fluorescence excitation emission matrix coupled with parallel factor (EEM−PARAFAC), two-dimensional Fourier transform infrared correlation spectroscopy (2D-FTIR−COS) and X-ray photoelectron spectroscopy (XPS), attempting to explain the mechanisms of EPS in alleviating Cd (II) toxicity toward a green alga Chlorella vulgaris (C. vulgaris). When the algal EPS were removed, the cell internalizations of Cd (II), growth inhibition rate and chlorophyll autofluorescence increased, but the surface adsorption and esterase activities decreased, indicating that the sorption of Cd (II) by EPS was crucial in alleviating the algal toxicity. Moreover, the complexation with proteins in EPS controlled the sorption of Cd (II) to algal EPS, resulting in the chemical static quenching of the proteins fluorescence by 47.69 ± 2.37%. Additionally, the complexing capability of the main functionalities, COO− and C–OH in proteins with Cd (II) was stronger than that of C–O(H) and C–O–C in polysaccharides or C–OH in the humus-related substances. Oxygen atom in protein carboxyl C–O might be the key site of EPS−Cd (II) complexation, supported by the modified Ryan−Weber complexation model and the obvious shift of oxygen valence-electron signal. These findings provide deep insights into understanding the interaction of EPS with heavy metals in aquatic environment.

Abstract:The adsorption behavior and interaction mechanisms of extracellular polymeric substances (EPS) of Pseudomonas aeruginosa OMCS-1 towards chromium (Cr), lead (Pb), and cadmium (Cd) were investigated. EPS-covered (EPS-C) cells exhibited significantly higher ( p < 0.0001; two-way ANOVA) removal of Cr (85.58 ± 0.39%), Pb (81.98 ± 1.02%), and Cd (73.88 ± 1%) than EPS-removed (EPS-R) cells. Interactions between EPS-heavy metals were spontaneous (ΔG<0). EPS-Cr(VI) and EPS-Pb(II) binding were exothermic (ΔH 0) process. EPS bonded to Pb(II) via inner-sphere complexation by displacement of surrounding water molecules, while EPS-Cr(VI) and EPS-Cd(II) binding occurred through outer-sphere complexation via electrostatic interactions. Increased zeta potential of Cr (29.75%), Pb (41.46%), and Cd (46.83%) treated EPS and unchanged crystallinity (CI XRD =0.13), inferred EPS-metal binding via both electrostatic interactions and complexation mechanism. EPS-metal interaction was predominantly promoted through hydroxyl, amide, carboxyl, and phosphate groups. Metal adsorption deviated EPS protein secondary structures. Strong static quenching mechanism between tryptophan protein-like substances in EPS and heavy metals was evidenced. EPS sequestered heavy metals via complexation with C-O, C-OH, C O/O-C-O, and NH/NH 2 groups and ion exchange with –COOH group. This study unveils the fate of Cr, Pb, and Cd on EPS surface and provides insight into the interactions among EPS and metal ions for metal sequestration.

Abstract:Metal binding to microbial extracellular polymeric substances (EPS) greatly influences the distribution of heavy metals in microbial aggregates, soil and aquatic systems in nature. In this work, the thermodynamic characteristics of the binding between aqueous metals (with copper ion as an example) and EPS of activated sludge were investigated. Isothermal titration calorimetry was employed to estimate the thermodynamic parameters for the binding of Cu2+ onto EPS, while three-dimensional excitation-emission matrix (EEM) fluorescence spectroscopy with parallel factor analysis was used for quantifying the complexation of Cu2+ with the EPS. The binding mechanisms were further explored by X-ray absorption fine structure (XAFS) and Fourier transform infrared (FTIR) spectroscopy analysis. The results show that the proteins and humic substances in EPS were both strong ligands for Cu2+. The binding capacity N, binding constant K, binding enthalpy ΔH were calculated as 5.74 × 10−2 mmol/g, 2.18 × 105 L/mol, and −11.30 kJ/mol, respectively, implying that such a binding process was exothermic and thermodynamically favorable. The binding process was found to be driven mainly by the entropy change of the reaction. A further investigation shows that Cu2+ bound with the oxygen atom in the carboxyl groups in the EPS molecules of activated sludge. This study facilitates a better understanding about the roles of EPS in protecting microbes against heavy metals.
Roles of bacterial cell and extracellular polymeric substance on adsorption of Cu(II) in activated sludges: A comparative study

Abstract:Activated sludge adsorbs heavy metals in the water during the water treatment process, which brings trouble to the subsequent treatment of the waste sludge. However, the role of bacterial cell and extracellular polymeric substance (EPS) on adsorption of heavy metal remains elusive. Here, we found the anammox sludge possessed the highest Cu(II) adsorption capacity (14.68 mg/g SS), followed by the activated sludge and denitrifying sludge. The Cu(II) adsorption capacities of sludges increased when sludge dosages and initial Cu(II) concentrations increased. These sorption processes were all well explained by Langmuir isotherms. Analysis of the Cu(II) distribution found that Cu(II) adsorbed by bacterial cells (35 %) were much higher than the Cu(II) adsorbed by EPS (4–11 %). This implied that bacterial cells were the most important carrier for the sludge to adsorb Cu(II). Additionally, the difference in Cu(II) adsorption of sludge was due to the inconsistent Cu(II) content adsorbed by EPS. Correlation analysis showed that the EPS content highly positively correlated with the Cu(II) content adsorbed by EPS, which indicates that the EPS content was crucial factor causing the difference in Cu(II) adsorption. FITR analysis shows that another key factor for disparate Cu(II) adsorption was functional groups, including amide I and amide II of protein, C–H from aliphatic, as well as OH and CO from carboxylic acid. This study provides more information for understanding the role of the bacterial cell and EPS in the adsorption of heavy metals, which might provide a theoretical basis for the treatment of heavy metals-containing sludge.
