Abstract:In this study, the short- and long-term effects of 1 mg L-1 of CuO nanoparticles (NPs), Cu NPs, and Cu(II) on the Anammox process were comparatively investigated. Results showed that short-term exposure to both CuO NPs and Cu NPs decreased the Anammox rate, while it slightly increased with Cu(II) (p < 0.05). Long-term exposure to CuO, Cu NPs, and Cu(II) all significantly suppressed nitrogen removal of Anammox, and the inhibition level decreased in the following order: CuO NPs > Cu NPs > Cu(II). The relative abundance of Candidatus Kuenenia decreased to 1.72%, 4.07%, and 3.93%, respectively, from their abundance of 8.29% in the control and that of Candidatus Brocadia decreased to 0.80%, 1.18%, and 1.87%, respectively, from 4.08%. The presence of Cu NPs elevated the expression of the copA metal resistance gene (p < 0.05) to 12.4 +/- 3.2 x 10(-4) from 3.56 +/- 0.39 x 10(-4) in the control, while CuO NPs and Cu(II) induced no significant variation. The copper content of sludge showed high negative correlation with nitrogen removal, and the nanoeffect was also attributed to inhibition by NPs. Cu NPs induced the highest expression to help the Anammox resist NPs when compared with CuO NPs. As a result, Cu NPs inhibited Anammox to a slightly greater extent than CuO NPs. The high accumulation of copper in the Cu(II) exposed sludge also led to Anammox suppression after long-term exposure.
Strengthened attachment of anammox bacteria on iron-based modified carrier and its effects on anammox performance in integrated floating-film activated sludge (IFFAS) process

Abstract:Moving-bed biofilm reactor (MBBR) or integrated floating-film activated sludge (IFFAS) process has been proved to be one of the ideal candidates for anammox application. However, the slow development of anammox bacteria (AnAOB) biofilm and unstable bioactivity always limit their wide application. This study developed a type of novel zero-valent iron (ZVI)-based modified carrier for strengthening AnAOB attachment and enhancing anammox performance. Surface properties analysis indicated the iron-based modified carrier revealed electropositive, less hydrophobic, and higher surface free energy compared with conventional high density polyethylene (HDPE) carrier. These surface parameters were positively correlated with total biomass attachment, anammox biofilm development, EPS secretion and heme-c production. IFFAS process filled with iron-based modified carriers could keep relatively stable and high anammox activity at different influent TN loadings (varied from 0.6 to 1.4 kg/(m3∙d)) and showed potential to keep and recover AnAOB bioactivity after six-months-freeze. Microbial analysis confirmed that anammox genus, Candidatus Kuenenia, had a significant niche preference on iron-based modified carrier than conventional HDPE carrier. As a result, the population of Candidatus Kuenenia in IFFAS process filled with modified carriers that contained 2 wt% or 3 wt% ZVI was 1.34 × 106–1.55 × 106 copies/ mg DNA, increased by 20.7–39.6% comparing with that in the control reactor (1.11 × 106 copies/ mg DNA). This study demonstrated AnAOB could be enriched and maintained in situ with high abundance and bioactivity on the iron-based modified carriers, which would be significant for anammox process wide application in full-scale.

Abstract:The individual and combined impacts of copper ion (Cu2+) and oxytetracycline (OTC) on anaerobic ammonium oxidation (anammox) performance and its self-recovery process were examined. Experimental results showed that the anammox performance and activity of anammox bacteria were inhibited by 1.0 mg L-1 OTC, Cu2+ and OTC + Cu2+, and both single and combined inhibitions were reversible. The abundance of functional genes and parts of antibiotic resistance genes (ARGs) were positively related to the dominant bacterium Ca. Kuenenia, implying that the recovery of the performance was associated with the progressive induction of potentially resistant species after inhibition. The above outcomes illustrated that anammox bacteria were stressed by metals and antibiotics, but they still could remove nitrogen at a rate higher than 20.6 ± 0.8 kg N m-3 d-1, providing guidance for engineering applications of anammox processes.

Abstract:The influence of copper ion (Cu2+) on anaerobic ammonium oxidation (anammox) performance and microbial community structures after oxytetracycline (OTC) stress recovery were assessed. Experimental results demonstrated that anammox performance were stressed by 1.0 mg L−1 Cu2+ and inhibitions were reversible with total nitrogen removal rate higher than 3.08 ± 0.2 kg N m−3 d−1. The residual OTC in the anammox sludge could combine with Cu2+ introduced and thereby retarded inhibition on performance in the presence of 2.0 mg L−1 Cu2+. Moreover, the positive relation of dominant bacterium Ca. Anammoxoglobus with the abundance of functional genes and parts of antibiotic resistance genes were observed, suggesting that regain of performance was the results of the gradual domestication of latent resistant species after inhibition. This investigation reveals new insights into resistance of anammox performance for Cu2+ and OTC.

Abstract:Metal and metal oxide nanoparticles (NPs) show differences in antimicrobial activity due to different chemical and physical properties. Using copper as a representative example, this study compared the NP effects on anaerobic ammonium oxidation (anammox) bacteria in wastewater treatment. Long-term exposure to 5 mgCu L-1 CuNPs reduced the physiological activity and abundance of anammox bacteria, thereby causing deterioration of reactor performance. However, anammox granules exhibited stronger resistance and resilience to perturbation by 1-160 mgCu L-1 CuONPs, and no adverse effects on performance were observed. Moreover, the level of Cu(II) released from NPs in the influent exhibited good correlations with variations of the community structure and sludge properties. Therefore, the effects of Cu-based NPs on anammox sludge properties are dependent on their forms and levels, and their discrepant effects are partially attributed to their ability to release ionic copper.
Effect of Ferrous Ion on the Performance of an Anammox Reactor

Abstract:The effect of ferrous ion on the performance of an anaerobic ammonium oxidation (anammox) reactor was investigated. The substrate removal rate of the reactor was boosted by addition of ferrous ion within 205 d. The maximum NH+4-N and NO-2-N removal rate with 0.075 mmol·L-1 Fe2+ was 1.8-fold and 1.6-fold of the control (Fe2+ concentration = 0.03 mmol·L-1). Microbial growth,denoted by production of nitrate and volatile solids (VS),was also enhanced. The biomass increment at 0.075 mmol·L-1 Fe2+ was 1.36-fold over the control,and the VS of the test reactor was 2.15-fold of the control. The anammox bacteria varied in cellular structure under iron-replete conditions (0.075 mmol·L-1). An unknown grey area appeared between the light area and the dark area,which was unprecedented in anammox bacteria. Based on this research,anammox bacteria have a large demand for iron and the former medium for the anammox bacteria was iron limited.

Abstract:Nitrogen removal by anaerobic ammonium oxidation (Anammox) has attracted increasing attention in nowadays. An Anammox biofilter was subjected to a continuous loading of elevated Zn (II). The influence of Zn (II) on the nitrogen removal, microbial community and biofilm property was investigated in the condition of 23-26 degrees C and 3.5 h HRT. The nitrogen removal greatly decreased to 0.054 from the initial 0.502 kg m(-3) d(-1), with the Zn (II) addition. Anaerobic ammonia-oxidizing bacteria (AAOB) had self-adaption to Zn (II) in 1-10 mg L-1 and was significantly enhanced after long-term acclimatization, while the suppression threshold was 20 mg L-1. Soluble microbial products (SMP) increased correspondingly with Zn (II), while extracellular polymeric substance (EPS) climbed up initially and then decreased. Anammox biofilm performed the highest zinc adsorption as 158.27 mg g(-1) SS in biofilm. High Zn (II) improved the microbial diversity and lowered the Candidatus Kuenenia abuandance to 1.38% from 20.89%.

Abstract:The increasing use of engineered nanoparticles (NPs) poses an emerging challenge to biological wastewater treatment. The long-term impact of CuNPs on anaerobic ammonium oxidation (anammox) process was firstly investigated in this study. The nitrogen removal capacity of anammox reactor was nearly deprived within 30days under the stress of 5.0mgL-1 CuNPs and the relative abundance of anammox bacteria (Ca. Kuenenia) was decreased from 29.59% to 17.53%. Meanwhile, copper resistance genes associated with the Cus, Cop and Pco systems were enriched to eliminate excess intracellular copper. After the withdrawal of CuNPs from the influent, the nitrogen removal capacity of anammox biomass recovered completely within 70days. Overall, anammox biomass showed susceptibility, resistance and resilience to the stress of CuNPs. Therefore, the potential impacts of ENPs on anammox-based processes should be of great concern.

Abstract:Anaerobic ammonium oxidation (anammox) is a promising biological technology for treating ammonium-rich wastewaters. However, due to the high sensitivity of anammox bacteria, many external factors have inhibitory effects on this process. As one of the commonly found toxic substances in wastewater, heavy metals (HMs) are possible to cause inhibition on anammox sludge, which then results in a declined treatment performance. Getting insights into the response mechanism of anammox sludge to HMs is meaningful for its application in treating this kind of wastewater. This review summarized the effect of different HMs on treatment performance of anammox bioreactor. In addition, the mechanism of toxication raised by HMs was discussed. Also, the potential mitigation strategies were summarized and the future prospects were outlooked. This review might provide useful information for both scientific research on and engineering application of anammox process for treating HMs containing wastewater.

Abstract:Heavy metals (HMs) have been widely reported to pose an adverse effect on anaerobic ammonia oxidation (anammox) bacteria, yet the underlying mechanisms remain unclear. This study provides new insights into the potential mechanisms of interaction between HMs and functional enzymes through big date analysis, molecular docking and molecular dynamics simulation. The statistical analysis indicated that 10 mg/L Cu(II) and Cd(II) reduced nitrogen removal rate (NRR) by 85% and 43%, while 5 mg/L Fe(II) enhanced NRR by 29%. Additionally, the results of molecular simulations provided a microscopic interpretation for these macroscopic data. Molecular docking revealed that Hg(II) formed a distinctive binding site on ferritin, while other HMs resided at iron oxidation sites. Furthermore, HMs exhibited distinct binding sites on hydrazine dehydrogenase. Concurrently, the molecular dynamics simulation results further substantiated their capacity to form complexes. Cu(II) displayed the strongest binding affinity with ferritin for −1576 ± 79 kJ/mol in binding free energy calculation. Moreover, Cd(II) bound to ferritin and HDH for −1052.67 ± 58.49 kJ/mol, −290.02 ± 49.68 kJ/mol, respectively. This research addressed a crucial knowledge gap, shedding light on potential applications for remediating heavy metal-laden industrial wastewater.

[Effect of simulated heavy metal leaching solution of electroplating sludge on the bioactivity of Acidithiobacillus ferrooxidans]

Abstract:An Acidithiobacillus ferrooxidans strain WZ-1 was isolated from the tannery sludge in Wenzhou, Zhejiang Province in China. The cell of WZ-1 strain is Gram negative and rod-shaped, its 16S rDNA sequence is closely related to that of Acidithiobacillus ferrooxidans ATCC23270 with 99% similarity. These results reveal that WZ-1 is a strain of Acidithiobacillus ferrooxidans. The effects of Ni2+, Cr3+, Cu2+, Zn2+ and 5 kinds of simulated leaching solutions of electroplating sludge on the bioactivity of Fe2+ oxidation and apparent respiratory rate of WZ-1 were investigated. The results showed that Ni2+ and Cr3+ did not have any influence on the bioactivity of WZ-1 at concentrations of 5.0 g x L(-1) and 0.1 g x L(-1), respectively. WZ-1 showed tolerance to high levels of Ni2+, Zn2+ (about 30.0 g x L(-1)), but it had lower tolerance to Cr3+ and Cu2+ (0.1 g x L(-1) Cr3+ and 2.5 g x L(-1) Cu2+). Different kinds of simulated leaching solution of electroplating sludge had significant differences in terms of their effects on the bioactivity of WZ-1 with a sequence of Cu/Ni/Cr/Zn > Cu/Ni/Zn > Cu/Cr/Zn > Cu/Ni/Cr > Ni/Cr/Zn.
Aerobic granular sludge shows enhanced resistances to the long-term toxicity of Cu(II)

Abstract:Cu(II) is one of the most widely-existed heavy metal ions in industrial effluents. A high concentration of Cu(II) leads to strong toxic effects on microorganisms and sludge for treating industrial wastewater which often contains aromatic pollutants. Granular sludge has different characteristics compared with floc sludge, and it may exhibit unique responses to the high concentration of Cu(II). Therefore, in this study, the variations of sludge properties and pollutant removal were investigated in the aerobic granular sludge (AGS) system with 0, 5, and 10 mg L−1 of Cu(II). The results suggested that both levels of Cu(II) promoted protein secretion and bounded with extracellular polymeric substances; thus, led to more compact granules with better settleability. Cu(II) had limited impacts on the overall organic degradation and denitrification efficiency, while it exerted significant negative effect on nitrification. The average NH4+-N concentration reached 1.4 ± 0.5, 6.7 ± 3.1, and 8.4 ± 1.5 mg L−1 in the effluent when the influent contained 0, 5, and 10 mg L−1 of Cu(II), respectively. The microbial community succession showed that no reduction was observed for the total relative abundance of main groups involved in organic removal such as Pseudoxanthomonas, Acidovorax, Acinetobacter, and Thauera. However, the growth of some functional groups such as Saccharibacteria for nitrification was inhibited by the toxic effect of Cu(II). These findings suggested that AGS could resist to the long-term toxic effects of Cu(II) by multiple rationales.

Abstract:The feasibility of using anaerobic ammonium oxidation (anammox) process to treat wastewaters containing antibiotics and heavy metals was evaluated in this study. The nitrogen removal performance and characteristic parameters were monitored during the whole experimental period of 258 d. The single and joint effects of zinc and tetracycline on the microbial community were studied in upflow anaerobic sludge blanket (UASB) reactors. The anammox performance remained at levels comparable with the initial state at the lower inhibitor concentrations (zinc, 0-2.26 mg L-1; tetracycline, 0-0.5 mg L-1). When the concentrations of zinc and tetracycline increased to 3.39 mg L-1 in R1 and 1.0 mg L-1 in R2, an obvious deterioration in performance was observed. Dual inhibitors with a total concentration of ≥3 mg L-1 caused dramatic decreases in the nitrogen removal efficiency of R3. The quantification results showed that the abundances of eight antibiotic resistance genes (ARGs), czcA and intI1 in the experimental reactors generally increased under stress from metals or/and antibiotics, with final values higher than in the control, while the functional gene abundances were lower. Moreover, most genes exhibited significant correlations. Microbial community analysis indicated that Planctomycetes (represented by Candidatus Kuenenia) was inhibited by both zinc and tetracycline, but still held the dominant position. Furthermore, Caldilinea (belonging to Chloroflexi) maintained a higher abundance during the inhibitory period, implying its potential resistance to both inhibitors. These findings suggested that anammox could be inhibited by metals and antibiotics, but it has the potential to remove nitrogen from wastewaters containing both of them within the concentration threshold.
Influence of Cu application on ammonia oxidizers in fluvo-aquic soil

Abstract:Nitrification driving by archaeal and bacterial ammonia oxidizers is an essential step of the global nitrogen cycle. This laboratory study screened toxicity of copper (Cu) stress in the fluvo-aquic system, investigated the ecological niche of ammonia-oxidizing archaea (AOA) and ammonia-oxidizing bacteria (AOB) communities and analyzed the response of AOA and AOB communities to Cu stress in the system. We added Cu as CuCl2 solution at 0 mg, 100 mg, 300 mg, 600 mg, and 1200 mg Cu per kg soil in the fresh fluvo-aquic soil. Soil samples were incubated for 6 h, 12 h, 1 d, 3 d, 7 d, 14 d, 28 d, and 56 d in lab condition. The soil potential nitrification rates (PNR) were measured at each stage. Denaturing gradient gel electrophoresis (DGGE) analysis of amoA of AOA and AOB were performed at 56 d of Cu incubation. Soil RNA was extracted and the abundance of amoA transcripts was analyzed by real time polymerase chain reaction (real-time PCR) at 1, 3, and 7 d. The amoA gene abundance analysis and DGGE profile analysis showed that the abundance and diversity of AOA were higher than that of AOB in fluvo-aquic soil. However, the correlations between the soil PNR and the number of amoA transcript indicated that the AOB community, rather than AOA community, dominates ammonia oxidation in Cu-polluted fluvo-aquic soil ecosystems. Combining the results of the soil PNR and the abundance and diversity of ammonia oxidizers community, Cu hormetic concentration was identified as a range from 100 to 600 mg kg−1, while the toxic concentration was higher than 600 mg kg−1 in fluvo-aquic soil system. Furthermore, the results of amoA transcript numbers showed that AOA may be resistant to Cu stress and AOB may be more sensitive to Cu stress under our experiment conditions.

Abstract:The presence of organic-complexed copper and zinc in anaerobic digestate effluent (ADE) poses persistent ecological toxicity. This study investigated the detoxification performance and biotic responses of indigenous bacteria against ethylene diamine tetraacetic acid (EDTA)-complexed Cu(II) and Zn(II). Heavy metals (HMs) stress induced reactive oxygen species (ROS) generation and enhanced extracellular polymeric substances (EPS) secretion. At a Cu(II) influent concentration of 20.0 mg·L-1, indigenous bacteria removed 88.2% of Cu(II) within nine days. The majority of copper and zinc sequestered by bacteria were stored in the cell envelope, with over 50% of copper and 60% of zinc being immobilized. Transmission electron microscopy mapping (TEM-mapping) revealed significant mineralization of copper and zinc on the cell wall. Proteins abundant in EPS, alongside humic acid-like substances, effectively adsorbed HMs. Indigenous bacteria exhibited the capacity to reduce cupric to the cuprous state and cupric is preferentially reduced to cuprous before reaching reducing capacity saturation. Sulfur precipitation emerges as a crucial pathway for Zn(II) removal. Metagenomic analysis indicated that indigenous bacteria upregulated genes related to HMs homeostasis, efflux, and DNA repair, enhancing its resistance to high concentrations HMs. This study provided theoretical guidance for employing bacterial consortia to eliminate HMs in complex aquatic environments.

Comprehensive analysis of the impacts of iron-based nanoparticles and ions on Anammox process

Abstract:The short- and long-term effects of 5 mg L−1 iron-based nanoparticles (NPs) and ions on Anammox sludge were comparatively investigated in the present study, including Fe3O4 NPs, Fe2O3 NPs, Fe NPs, Fe (II) and Fe (III). Results showed that short-term exposure to Fe (II) obviously increased the Anammox activity, while Fe2O3 NPs and Fe (III) both decreased it (p < 0.05), and Fe3O4 and Fe NPs presented slight impacts. After long-term exposure, the TN removal efficiencies varied to 89.4%, 42.5%, 74.8%, 91.3% and 56.2%, respectively, while that in the control was 82.6%. The OTUs showed the highest difference from others in the sludge exposed to Fe2O3 NPs, while the microbial biodiversity in the iron exposed reactors all increased. The relative abundance of Candidatus Kuenenia varied to 11.67%, 3.99%, 10.07%, 13.89% and 8.07%, respectively, from the abundance as 11.26% in the control. Denitrifying bacteria, including Arenimonas and Denitritisoma were enhanced by the iron. The impacts (positive (+) or negative (-)) of the irons on Anammox process were in the order Fe (II)(+)>Fe3O4 NPs(+)>Fe NPs( ± )> Fe (III)(-)>Fe2O3 NPs(-). Fe (II) and Fe3O4 NPs could be adopted for stimulating Anammox activity, while using Fe NPs need to pre-cultivate the sludge or optimize the dosing strategy.

Abstract:To understand the temporal and spatial toxic effect of heavy metals on the microbial activities of biofilms, microelectrodes were used to measure the inhibitory oxygen (O(2)) concentration profiles resulted from the effects of zinc (Zn(2+)) and copper (Cu(2+)). Using the O(2) microprofiles as bases, the spatial distributions of net specific O(2) respiration were determined in biofilms with and without treatment of 5 mg/L Zn(2+) or 1 mg/L Cu(2+). Results show that microbial activities were inhibited only in the outer layer (∼400 μm) of the biofilms and bacteria present in the deeper sections of the biofilms became even more active. The inhibition caused by the heavy metals was evaluated by two methods. One was derived from the oxygen influx at the interface and the other was based on the integral of the oxygen consumption calculated from the entire O(2) profile. The two methods yielded significantly different results. We argue that the integral method results in more accurate assessment of toxicity than the surface flux determination.
