Abstract:Interactions between metals and anaerobic ammonium oxidizing consortia substantially affect the quality of wastewater treatment plant effluent. In this study, we conducted acid-base titrations to ascertain the surface reactivity and proton adsorptive capacity of anammox consortia. A combination of titration data modeling and infrared spectroscopy suggested the presence of carboxyl, amine, and hydroxyl groups. Cd adsorption experiments demonstrate that 1 g of dry biomass could bind an equivalent of 7.12 x 10(-6) mol/L of Cd. Density functional theory calculations further reveal that carboxyl and hydroxyl groups are able to form stable Cd complexes. Furthermore, considerable carboxyl and hydroxyl groups promote bacterial aggregation, and thus solid-liquid separation. The results of this study highlight the potential role of anammox consortia in adsorbing metal cations, and thus help to improve the understanding of the universally significant contribution of anammox consortia at the detoxification of metal cations in wastewater treatment systems. Abstract:Heavy metals (HMs) have been widely reported to pose an adverse effect on anaerobic ammonia oxidation (anammox) bacteria, yet the underlying mechanisms remain unclear. This study provides new insights into the potential mechanisms of interaction between HMs and functional enzymes through big date analysis, molecular docking and molecular dynamics simulation. The statistical analysis indicated that 10 mg/L Cu(II) and Cd(II) reduced nitrogen removal rate (NRR) by 85% and 43%, while 5 mg/L Fe(II) enhanced NRR by 29%. Additionally, the results of molecular simulations provided a microscopic interpretation for these macroscopic data. Molecular docking revealed that Hg(II) formed a distinctive binding site on ferritin, while other HMs resided at iron oxidation sites. Furthermore, HMs exhibited distinct binding sites on hydrazine dehydrogenase. Concurrently, the molecular dynamics simulation results further substantiated their capacity to form complexes. Cu(II) displayed the strongest binding affinity with ferritin for −1576 ± 79 kJ/mol in binding free energy calculation. Moreover, Cd(II) bound to ferritin and HDH for −1052.67 ± 58.49 kJ/mol, −290.02 ± 49.68 kJ/mol, respectively. This research addressed a crucial knowledge gap, shedding light on potential applications for remediating heavy metal-laden industrial wastewater.

Electrokinetic deposition and removal of heavy metals from sewage sludge enhanced with ammonia complexation

Abstract:The experiment used an in-house designed electrokinetic equipment in order to remove heavy metals from municipal wastewater sludge. Ammonia was added and mixed with four groups of sludge samples at ratios of 0, 0.08, 0.10, 0.12(V:V, NH3·H2O/sludge sample). The samples were electrolyzed for 120 hours at a constant DC voltage. The effects of anodic acid generation coupled with ammonia complexation on the enhanced removal of heavy metals from sewage sludge was measured. The results indicated that anodic acidification had a moderate removal rate on heavy metals. Although adding ammonia had no impact on p H and electric current density in the electrokinetic process, it enhanced the trapping and removal rate of heavy metals in the sludge. Results further indicated that different amount of ammonia exerted different levels of removal rates under electrokinetic treatment. When ammonia water was added at a ratio of 0.12, removal rates of Zn and Cu were 91.62% and 71.49%, respectively. Optimal remediation was achieved ata ratio of 0.08 for Pb with a removal rate of 47.85%. The enhanced removal effect by adding ammonia was presumably the result of decreased residual fractions and organic bound fractions, as well as the increased exchangeable fractions of these metals.

Abstract:Persistence of ammonium nitrogen and heavy metals in wastewater still remains a challenge, and many wastewater treatment plants face the challenge of removing nitrogen under heavy metal stresses. There is no preferred method for the biological treatment of wastewater containing nitrogen and heavy metals with the possible exception of the anaerobic ammonium oxidation (anammox), since it has shown promise for removing nitrogen under heavy metal stresses. This article reviews the recent research results of the nitrogen-removal performance and mechanism by the anammox process under heavy metal stresses, mainly discussing the enhancing and inhibition effects of heavy metals on the performance of the Anammox reactor. The influencing mechanism of heavy metals on the microbial community and extracellular polymeric substances is also presented, and examples are given for explanation. The main problems of the present research are pointed out, and it is proposed that unifying the metal ion concentrations of inhibiting or promoting anammox activity is necessary for the development and industrial application of the anammox process. The information of this review can offer a great possibility for achieving desired nitrogen removal in wastewater treatment under heavy metal stresses and with significant energy savings.

Abstract:Anaerobic ammonium oxidation (anammox) is a promising biological technology for treating ammonium-rich wastewaters. However, due to the high sensitivity of anammox bacteria, many external factors have inhibitory effects on this process. As one of the commonly found toxic substances in wastewater, heavy metals (HMs) are possible to cause inhibition on anammox sludge, which then results in a declined treatment performance. Getting insights into the response mechanism of anammox sludge to HMs is meaningful for its application in treating this kind of wastewater. This review summarized the effect of different HMs on treatment performance of anammox bioreactor. In addition, the mechanism of toxication raised by HMs was discussed. Also, the potential mitigation strategies were summarized and the future prospects were outlooked. This review might provide useful information for both scientific research on and engineering application of anammox process for treating HMs containing wastewater.

Abstract:The anaerobic ammonium oxidation (anammox) process has been acknowledged as an effective nitrogen removal technology with extensive application potential. However, the growth rates of anammox bacteria (AnAOB) are extremely low, and susceptible to a handful of operational and environmental conditions, which limits the process' extensive application. In recent years, numerous researchers have conducted studies of exogenous substance addition on the performance and microbial community of anammox, and related findings have been analyzed with a focus on the effects of their promotion on performance and microbial community. This study summed up the current reports based on various metals (Zero-valent iron, Fe 2+ , Fe 3+ , iron-based material, Mn, Ni, K, Ca), organic matter (fulvic acid, humic acid), inorganic matter (biochar, activated carbon, graphene, zeolite, inorganic carbon), reaction intermediates (hydroxylamine, hydrazine) and N-acyl-homoserine lactones influencing anammox by enhancing the functional bacteria and electronic delivery, obtaining higher biological retention and specific anammox activity (SAA). This review is intended to provide the groundwork for prospective advancement and large-scale utilization of the anammox process.

Abstract:Extracellular polymeric substance (EPS) of anaerobic ammonium oxidation (anammox) sludge plays an important role in resisting the toxicity when treating heavy metal containing wastewater. Understanding the interaction mechanism between EPS and heavy metal is essential for stable and optimal operation of such technology. In this work, the identification and preferential response of the binding sites, as well as the binding thermodynamic and the conformational changes of EPS to Cu2+ were explored. Unlike the activated sludge system, functional groups in EPS would coordinate with Cu2+ to form inner sphere complexes, which is an endothermic and entropy-increasing process. Low concentration of Cu2+ was found to result in the reduced electrostatic repulsion and bridging effect, which promoted the compaction and agglomeration of EPS colloid. Two-dimensional correlation spectroscopy analysis confirmed that carboxyl groups of proteins were preferentially responded to Cu2+ compared to polysaccharides and hydrocarbons. More importantly, the secondary structure of the protein in EPS was disrupted and changed into loose state after combining with Cu2+. This work is of great significance for understanding the binding properties of anammox sludge EPS with Cu2+ and may provide theoretical support for elucidating the resist mechanism of anammox bacteria to heavy metals.

Abstract:Here we unveiled a novel magnetically separable amphoteric biosorbent (PD-Fe3O4@CCS) and investigated its adsorption behavior toward two classes of heavy metals, hexavalent chromium (Cr(VI)) and copper (Cu(II)) ions from water. Results indicated that the adsorption behavior of PD-Fe3O4@CCS for Cr(VI) was well described by Langmuir model; while for Cu(II) adsorption, the Freundlich model was the better one. Based on the kinetic results, both Cr(VI) and Cu(II) adsorption on PD-Fe3O4@CCS fitted well with the pseudo-second-order kinetic model. To evaluate the reusability and stability of PD-Fe3O4@CCS, regeneration tests were carried out for five cycles. Furthermore, the applicable feasibility of PD-Fe3O4@CCS in the real water matrix (including the single and binary pollutant systems) was studied, and results suggested the promising potential of PD-Fe3O4@CCS for large-scale application. Apart from these, the surface interactions between PD-Fe3O4@CCS and heavy metal ions in single and binary systems were systematically investigated based on FTIR and XPS analyses, which provided an essential implication for comprehending the interactions between biosorbents and contaminants in wastewater.

Abstract:Complex heavy metal (HM)-containing wastewater discharges pose substantial risks to global water ecosystems and human health. Aerobic granular sludge (AGS) has attracted increased attention as an efficient and low-cost adsorbent in HM-containing wastewater treatment. Therefore, this study systematically evaluates the effect of Cu(II), Ni(II), and Cr(III) addition on the characteristics, performance and mechanism of AGS in complex HM-containing wastewater treatment process by means of fourier transform infrared spectroscopy, inductively coupled plasma spectrocopcy, confocal laser scanning microscopy, extracellular polymeric substances (EPS) fractions detection and scanning electron microscope-energy dispersive X-ray. The results showed that AGS efficiently eliminated Cu(II), Ni(II), and Cr(III) by the orchestrated mechanisms of ion exchange, three-layer EPS adsorption [soluble microbial products EPS (SMP-EPS), loosely bound EPS (LB-EPS), tightly bound EPS (TB-EPS)], and inner-sphere adsorption; notably, almost 100% of Ni(II) was removed. Three-layer EPS adsorption was the dominant mechanism through which the HM were removed, followed by ion exchange and inner-sphere adsorption. SMP-EPS and TB-EPS were identified as the key EPS fractions for adsorbing Cr(III) and Cu(II), respectively, while Ni(II) was adsorbed evenly on SMP-EPS, TB-EPS, and LB-EPS. Moreover, the rates at which the complex HM penetrated into the granule interior and their affinity for EPS followed the order Cu(II) > Ni(II) > Cr(III). Ultimately, addition of complex HM stimulated microorganisms to excrete massive phosphodiesterases (PDEs), leading to a pronounced decrease in cyclic diguanylate (c-di-GMP) levels, which subsequently suppressed EPS secretion due to the direct linkage between c-di-GMP and EPS. This study unveils the adaptability and removal mechanism of AGS in the treatment of complex HM-containing wastewater, which is expected to provide novel insights for addressing the challenges posed by intricate real wastewater scenarios.

Abstract:The feasibility of using anaerobic ammonium oxidation (anammox) process to treat wastewaters containing antibiotics and heavy metals was evaluated in this study. The nitrogen removal performance and characteristic parameters were monitored during the whole experimental period of 258 d. The single and joint effects of zinc and tetracycline on the microbial community were studied in upflow anaerobic sludge blanket (UASB) reactors. The anammox performance remained at levels comparable with the initial state at the lower inhibitor concentrations (zinc, 0-2.26 mg L-1; tetracycline, 0-0.5 mg L-1). When the concentrations of zinc and tetracycline increased to 3.39 mg L-1 in R1 and 1.0 mg L-1 in R2, an obvious deterioration in performance was observed. Dual inhibitors with a total concentration of ≥3 mg L-1 caused dramatic decreases in the nitrogen removal efficiency of R3. The quantification results showed that the abundances of eight antibiotic resistance genes (ARGs), czcA and intI1 in the experimental reactors generally increased under stress from metals or/and antibiotics, with final values higher than in the control, while the functional gene abundances were lower. Moreover, most genes exhibited significant correlations. Microbial community analysis indicated that Planctomycetes (represented by Candidatus Kuenenia) was inhibited by both zinc and tetracycline, but still held the dominant position. Furthermore, Caldilinea (belonging to Chloroflexi) maintained a higher abundance during the inhibitory period, implying its potential resistance to both inhibitors. These findings suggested that anammox could be inhibited by metals and antibiotics, but it has the potential to remove nitrogen from wastewaters containing both of them within the concentration threshold.

Abstract:Cadmium (Cd) can cause the deterioration of biological systems through inhibiting the enzymes activity and disturbing the microbial metabolism. Although the influence of Cd on conventional wastewater treatment process has been studied, the response of anammox to Cd exposure still remains unclear. This study firstly investigated the adaption and restoration of anammox biomass to Cd(II) stress. Results showed that long-term exposure of anammox bacteria to 2 mg L-1 Cd(II) was beneficial for the reactor performance, while 5 mg L-1 Cd(II) would cause the decline of SAA, extracellular polymeric substance content and relative abundance of Candidatus kuenenia by 40%, 25% and 31%, respectively. Furthermore, these indexes could approximately recover to the initial status after withdrawing Cd(II) from the influent. Overall, the anammox biomass exhibited a certain adaption and restoration ability to the suppression of Cd(II). This study may provide key valuable information for the biological treatment of wastewater containing Cd(II).
Selective targeted adsorption and inactivation of antibiotic-resistant bacteria by Cr-loaded mixed metal oxides

Abstract:Herein we provide a novel high-efficiency nanocomposite for bacterial capture based on mixed metal oxides (MMOs) with deleterious chromium properties. With both the layer structure of layered double hydroxides (LDHs) and the magnetic properties of Fe, MMOs enrich the location of ionic forms on the surface, providing a good carrier for adsorption of the heavy metal Cr(VI). The capacity for adsorption of Cr(VI) by MMOs can be as high as 98.80 mg/g. The prepared Cr(VI)-MMOs achieved extremely expeditious location of gram-negative antibiotic-resistant E. coliNDM-1 by identifying lipid bilayers. Cr-MMOs with a Cr loading of 19.70 mg/g had the best bactericidal effect, and the concentration of E. coliNDM-1 was decreased from ∼10 to ∼10 CFU/mL after 30 min of reaction. The binding of nitrogen and phosphorus hydrophilic groups to chromate generated realistic models for density functional theory (DFT) calculations. The specific selectivity of MMOs toward bacterial cells was improved by taking Cr(VI) as a transferable medium, thereby enhancing the antibacterial activity of Cr-MMOs. Under the combined action of chemical and physical reactions, Cr(VI)-MMOs achieved high capacity for inactivation of bacteria. Moreover, the metallic elements ratio in Cr-MMOs remained stable in their initial valence states after inactivation. This guaranteed high removal efficiency for both heavy metals and bacteria, allowing recycling of the adsorbent in practical applications.

Abstract:Anaerobic ammonium-oxidizing (anammox) bacteria can play an important role in nitrogen elimination in the environment. However, the effect of heavy metals on anammox bacteria in aquatic ecosystem remains largely unknown. The present study investigated the variability of anammox bacterial community in a freshwater reservoir after a severe heavy metal spill. The richness (Chao1 richness estimator = 2–18), diversity (Shannon index = 0.26–2.04) and community structure of anammox bacteria changed considerably with sampling date, while anammox bacterial abundance (from 1.38 × 105 to 3.09 × 105 anammox bacterial 16S rRNA gene copies per gram dry sediment) was less responsive to metal spill. Anammox bacterial communities were mainly composed of Brocadia- and Anammoxoglobus-like bacteria as well as novel phylotype, however, there relative abundance varied among sampling dates. This work could add the knowledge of the response of anammox bacteria to heavy metal contamination.
Mechanism of Deep-Sea Microbial-Mediated Synergistic Synthesis of Ferromanganese Oxidation Products Process for Heavy Metal Ion Adsorption

Abstract:Here we report the mechanism of adsorption and immobilization of heavy metal ions during the formation of deep-sea microbial-mediated Mn-Fe oxidation synergistic synthesis products under Mn concentration of 1 g/L, pH = 6, temperature of 25°C, adsorption time of 12 days, salinity of 3% and OMF inoculation of 20%. The adsorption amounts of heavy metal ion (Cu 2+ , Ni 2+ , and Co 2+ ) by the deep-sea microbial-mediated Mn-Fe oxidation co-synthesis products were 11.81 mg/g, 9.52 mg/g, and 7.14 mg/g, respectively. The process was consistent with the Langmuir isothermal adsorption model and quasi-secondary kinetic model, indicating that the adsorption of heavy metal ion during the formation of the deep-sea microbial-mediated Mn-Fe oxidation co-synthesis products was controlled by chemisorption. FTIR and XPS results showed that the adsorption and fixation of heavy metal ions in the system mainly depended on the physical adsorption, ion exchange, functional group complexation, precipitation and electrostatic attraction effects of the functional groups on the surface of the manganese and iron oxidation products and oxidized low-valent manganese colonies. The introduction of iron ions improved the negative charge, specific surface area, active sites and surface activity of the adsorbent surface, which enhanced the adsorption of heavy metal ion by the oxidized low-valent manganese functional microbial-mediated manganese oxidation synergistic synthesis products. Meanwhile, this study provides a new theoretical basis for the pollution treatment of Cu 2+ , Ni 2+ , Co 2+ , Fe 2+ , and Mn 2+ in heavy metal polluted water bodies and deep-sea manganese nodule mining processes with local exceedances.

Abstract:Activated sludge adsorbs heavy metals in the water during the water treatment process, which brings trouble to the subsequent treatment of the waste sludge. However, the role of bacterial cell and extracellular polymeric substance (EPS) on adsorption of heavy metal remains elusive. Here, we found the anammox sludge possessed the highest Cu(II) adsorption capacity (14.68 mg/g SS), followed by the activated sludge and denitrifying sludge. The Cu(II) adsorption capacities of sludges increased when sludge dosages and initial Cu(II) concentrations increased. These sorption processes were all well explained by Langmuir isotherms. Analysis of the Cu(II) distribution found that Cu(II) adsorbed by bacterial cells (35 %) were much higher than the Cu(II) adsorbed by EPS (4–11 %). This implied that bacterial cells were the most important carrier for the sludge to adsorb Cu(II). Additionally, the difference in Cu(II) adsorption of sludge was due to the inconsistent Cu(II) content adsorbed by EPS. Correlation analysis showed that the EPS content highly positively correlated with the Cu(II) content adsorbed by EPS, which indicates that the EPS content was crucial factor causing the difference in Cu(II) adsorption. FITR analysis shows that another key factor for disparate Cu(II) adsorption was functional groups, including amide I and amide II of protein, C–H from aliphatic, as well as OH and CO from carboxylic acid. This study provides more information for understanding the role of the bacterial cell and EPS in the adsorption of heavy metals, which might provide a theoretical basis for the treatment of heavy metals-containing sludge.

Abstract:Heavy metal oxoanions and cations often co-exist in the environment and it is a critical challenge to achieve their simultaneous and efficient removal due to their presence of opposite charges. Here, a polyacrylate-coated ferrous sulfide and layered double hydroxide (PAA/FeS@LDH) nanocomposite was developed. Results illustrated that PAA/FeS@LDH exhibited excellent reductivity and stability due to the dispersion of PAA and the support by LDH. The removal efficiency of single Cr(VI) (9.73 mg/L) using PAA/FeS@LDH reached 100 % within 120 min, which was significantly higher than that of LDH (35.39 %), FeS (73.13 %), and FeS/LDH (96.47 %). Attractively, an enhanced removal of Cr(VI) (19.46 mg/L) by PAA/FeS@LDH in the presence of Cu(II), Ni(II), or Cd(II) was observed, with Cr(VI) removal efficiency increasing from 49.63 % (single Cr(VI)) to 99.51 %, 82.46 %, and 72.18 % within 30 min, respectively. The adsorption capacities of Cr(VI) significantly enlarged from 140 mg Cr/g (single Cr(VI)) to 221, 175, and 171 mg Cr/g when co-existing with Cu(II), Ni(II), or Cd(II), respectively. Another interesting outcome was the significant enhancement of reusability of PAA/FeS@LDH by the co-existence of Cr(VI) and Cu(II). After 5 cycles, PAA/FeS@LDH could still remove 96.97 % of Cr(VI) together with >70 % of Cu(II) in the mixed solution, by contrast, the removal efficiency of Cr(VI) was only 38.04 % in a single solution. The characterization analysis and density functional theory calculations demonstrated that besides surface reduction, complexation and precipitation, the cation-anion bridge interaction was an essential mechanism of achieving the synergistic removal of heavy metal oxoanions and cations.

Abstract:In this study, the short- and long-term effects of Cd(II) on the bioactivity, nitrogen removal and microbial community of the anaerobic ammonia oxidation (Anammox) process were investigated by step-wise increasing Cd(II). The self-recovery ability of Anammox was also studied after long-term exposure in Cd(II). The bioactivity of anaerobic ammonia-oxidizing bacteria (AAOB) within short-term exposure of Cd(II) were detected by batch experiments. Results showed that Cd(II) had both acute and persistent toxicity to Anammox, the IC50 in short-term exposure was calculated as 5.43 mg L-1. Long-term exposure led to the microbial diversity increase, as well as the relative abundance decrease of AAOB in Anammox system. AAOB was continuously suppressed by Cd(II) in 1-20 mg L-1, while it had self-adaption to Cd(II) in 1-10 mg L-1. Anammox biofilm showed prominent adsorbing ability for cadmium, and the cadmium in biofilm was the key factor affecting Anammox.
Interaction Structure and Affinity of Zwitterionic Amino Acids with Important Metal Cations (Cd2+, Cu2+, Fe3+, Hg2+, Mn2+, Ni2+ and Zn2+) in Aqueous Solution: A Theoretical Study

Abstract:Heavy metals are non-biodegradable and carcinogenic pollutants with great bio-accumulation potential. Their ubiquitous occurrence in water and soils has caused serious environmental concerns. Effective strategies that can eliminate the heavy metal pollution are urgently needed. Here the adsorption potential of seven heavy metal cations (Cd2+, Cu2+, Fe3+, Hg2+, Mn2+, Ni2+ and Zn2+) with 20 amino acids was systematically investigated with Density Functional Theory method. The binding energies calculated at B3LYP-D3/def2TZVP level showed that the contribution order of amino acid side chains to the binding affinity was carboxyl > benzene ring > hydroxyl > sulfhydryl > amino group. The affinity order was inversely proportional to the radius and charge transfer of heavy metal cations, approximately following the order of: Ni2+ > Fe3+ > Cu2+ > Hg2+ > Zn2+ > Cd2+ > Mn2+. Compared to the gas-phase in other researches, the water environment has a significant influence on structures and binding energies of the heavy metal and amino acid binary complexes. Collectively, the present results will provide a basis for the design of a chelating agent (e.g., adding carboxyl or a benzene ring) to effectively remove heavy metals from the environment.
