Abstract:Microfluidic magnetophoresis is an effective technique to separate magnetically labeled bioconjugates in lab-on-a-chip applications. However, it is challenging and expensive to fabricate and integrate microscale permanent magnets into microfluidic devices with conventional methods that use thin-film deposition and lithography. Here, we propose and demonstrate a simple and low-cost technique to fabricate microscale permanent magnetic microstructures and integrate them into microfluidic devices. In this method, microstructure channels were fabricated next to a microfluidic channel and were injected with a liquid mixture of neodymium (NdFeB) powders and polydimethylsiloxane (PDMS). After the mixture was cured, the resulted solid NdFeB–PDMS microstructure was permanently magnetized to form microscale magnets. The microscale magnets generate strong magnetic forces capable of separating magnetic particles in microfluidic channels. Systematic experiments and numerical simulations were conducted to study the geometric effects of the microscale magnets. It was found that rectangular microscale magnets generate larger \(({\mathbf {H}}\cdot \nabla ) {\mathbf {H}}\) which is proportional to magnetic force and have a wider range of influence than the semicircle or triangle magnets. For multiple connected rectangular microscale magnet, additional geometric parameters, including separation distance, height and width of the individual elements, further influence the particle separation and were characterized experimentally. With an optimal size combination, complete separation of yeast cells and magnetic microparticles of similar sizes (\(4\;\upmu \hbox {m}\)) was demonstrated with the multi-rectangular magnet microfluidic device.

Abstract:This paper demonstrates simple and cost-effective microfluidic devices for enhanced separation of magnetic particles by using soft magnetic microstructures. By injecting a mixture of iron powder and polydimethylsiloxane (PDMS) into a prefabricated channel, an iron–PDMS microstructure was fabricated next to a microfluidic channel. Placed between two external permanent magnets, the magnetized iron–PDMS microstructure induces localized and strong forces on the magnetic particles in the direction perpendicular to the fluid flow. Due to the small distance between the microstructure and the fluid channel, the localized large magnetic field gradients result a vertical force on the magnetic particles, leading to enhanced separation of the particles. Numerical simulations were developed to compute the particle trajectories and agreed well with experimental data. Systematic experiments and numerical simulation were conducted to study the effect of relevant factors on the transport of superparamagnetic particles, including the shape of iron–PDMS microstructure, mass ratio of iron–PDMS composite, width of the microfluidic channel, and average flow velocity.

Abstract:The accurate and efficient manipulation of particles and droplets is crucial for the fields of disease diagnosis, target drug delivery, and sample purification, specific microstructure-based particle and droplet manipulation have been widely utilized in bioassays. However, current techniques are mainly based on the fixed microstructures, which is extremely difficult to achieve dynamic modulation by adjusting microstructure morphology and excitation conditions. Herein, we present a novel particle capture and droplet manipulation methodology by using morphologically reconfigurable magnetic micropillar arrays incorporate with acoustic streaming effects, which enables reconfigurable particle capturing, cargo transportation, and droplet manipulation. The mechanism is to assemble NdFeB magnetic powders by magnetic gradient fields to form morphologically variable magnetic micropillar arrays with porous surfaces, and the localized acoustic streaming effects generated by surface acoustic waves (SAWs) enable micropillar arrays to capture nearby particles onto the surfaces or to drive perturbations in droplets in the vicinity. To demonstrate the capabilities of this technology, we have achieved the high-volume acoustofluidic particle capture and loading (0.214 MCF), targeted complex maze navigation, efficient droplets manipulation (≤ 3.96mm/s), and acoustofluidic mixing acceleration (≤ 2.5s). Therefore, this technique enables reconfigurable and versatile manipulation of particles and droplets in biomedical applications.

Abstract:We introduce magnetophoresis-based microfluidics for sorting biological targets using positive Magnetophoresis (pM) for magnetically labeled particles and negative Magnetophoresis (nM) for label-free particles. A single, externally magnetized ferromagnetic wire induces repulsive forces and is positioned across the focused sample flow near the main channel's closed end. We analyze magnetic attributes and separation performance under two transverse dual-mode magnetic configurations, examining magnetic fields, hydrodynamics, and forces on microparticles of varying sizes and properties. In pM, the dual-magnet arrangement (DMA) for sorting three distinct particles shows higher magnetic gradient generation and throughput than the single-magnet arrangement (SMA). In nM, the numerical results for SMA sorting of red blood cells (RBCs), white blood cells (WBCs), and prostate cancer cells (PC3-9) demonstrate superior magnetic properties and throughput compared to DMA. Magnetized wire linear movement is a key design parameter, allowing device customization. An automated device for handling more targets can be created by manipulating magnetophoretic repulsion forces. The transverse wire and magnet arrangement accommodate increased channel depth without sacrificing efficiency, yielding higher throughput than other devices. Experimental validation using soft lithography and 3D printing confirms successful sorting and separation, aligning well with numerical results. This demonstrates the successful sorting and separating of injected particles within a hydrodynamically focused sample in all systems. Both numerical and experimental findings indicate a separation accuracy of 100% across various Reynolds numbers. The primary channel dimensions measure 100 μm in height and 200 μm in width. N52 permanent magnets were employed in both numerical simulations and experiments. For numerical simulations, a remanent flux density of 1.48 T was utilized. In the experimental setup, magnets measuring 0.5 × 0.5 × 0.125 inches and 0.5 × 0.5 × 1 inch were employed. The experimental data confirm the device's capability to achieve 100% separation accuracy at a Reynolds number of 3. However, this study did not explore the potential impact of increased flow rates on separation accuracy.

Abstract:Due to the outstanding properties, magnetic particles have been widely used as magnetic carriers and adsorbents in a diverse range of fields, such as biomedical diagnostics, food safety, and environmental monitoring, etc. Sorting different magnetic particles from each other based on size simultaneously can be very useful in multiple targets separation. To the best of our knowledge, this research firstly proposes the arc comb structure microfluidic (ACSM) chip for multi-magnetic particles separation based on size by magnetophoresis. And the parameters affecting particle trajectory and separation efficiencies, such as flow velocity, velocity ratio of the two inlets, the remanent flux density of the magnets, and dynamic viscosity of the buffer solution, were investigated by numerical simulations in this study. Particle magnetophoresis spectrum width Θ and general separation ratio PG are put forward to characterize the particles separation efficiency. Under the recommended simulation condition, six kinds of magnetic beads were separated with a general separation ratio PG of 0.83 using the newly designed chip. And the separation ratio pi of the 0.5, 1.8, 2.5, 3, 3.5 and 4.5 μm magnetic particle is 100.00%, 98.18%, 66.36%, 97.25%, 97.27% and 96.36%, respectively. This numerical simulation study provides a theoretical basis for multiple magnetic particles' separation before experimental trials and also facilitates the utilization and development of microfluidic chips in the future.

Abstract:Magnetic nanoparticle based bioseparation in microfluidics is a multiphysics phenomenon that involves interplay of various parameters. The ability to understand the dynamics of these parameters is a prerequisite for designing and developing more efficient magnetic cell/bio-particle separation systems. Therefore, in this work proof-of-concept experiments are combined with advanced numerical simulation to design and optimize the capturing process of magnetic nanoparticles responsible for efficient microfluidic bioseparation. A low cost generic microfluidic platform was developed using a novel micromolding method that can be done without a clean room techniques and at much lower cost and time. Parametric analysis using both experiments and theoretical predictions were performed. It was found that flow rate and magnetic field strength greatly influence the transport of magnetic nanoparticles in the microchannel and control the capturing efficiency. The results from mathematical model agree very well with experiments. The model further demonstrated that a 12% increase in capturing efficiency can be achieved by introducing of iron-grooved bar in the microfluidic setup that resulted in increase in magnetic field gradient. The numerical simulations were helpful in testing and optimizing key design parameters. Overall, this work demonstrated that a simple low cost experimental proof-of-concept setup can be synchronized with advanced numerical simulation not only to enhance the functional performance of magneto-fluidic capturing systems but also to efficiently design and develop microfluidic bioseparation systems for biomedical applications.

Abstract:This paper reported an efficient method to sizeselective separate magnetic nanospheres using a self-focusing microfluidic channel equipped with a permanent magnet. Under external magnetic field, the magnetophoresis force exerted on particles leads to size-dependent deflections from their laminar flow paths and results in effective particles separation. By adjusting the distance between magnet and main path of channel, we obtained two monodisperse nanosphere samples (Ca. 90 nm, Ca. 160 nm) from polydispersing particles solution whose diameters varied from 40 to 280 nm. Based on the magnetostatic and laminar flow models, numerical simulations were also used to predict and optimize the nanospheres migrations. Two thresholds of particles diameters were obtained by the simulations and diverse at each position of magnet. Therefore, appropriate position of the magnet could be determined at a certain particle sizes' range when the flow rate of the two inlets remains unchanged.
Multiphase Ferrofluid Flows for Micro-Particle Focusing and Separation

Abstract:Ferrofluids have demonstrated great potential for a variety of manipulations of diamagnetic (or non-magnetic) micro-particles/cells in microfluidics, including sorting, focusing, and enriching. By utilizing size dependent magnetophoresis velocity, most of the existing techniques employ single phase ferrofluids to push the particles towards the channel walls. In this work, we demonstrate a novel strategy for focusing and separating diamagnetic micro-particles by using the laminar fluid interface of two co-flowing fluids-a ferrofluid and a non-magnetic fluid. Next to the microfluidic channel, microscale magnets are fabricated to generate strong localized magnetic field gradients and forces. Due to the magnetic force, diamagnetic particles suspended in the ferrofluid phase migrate across the ferrofluid stream at the size-dependent velocities. Because of the low Reynolds number and high Péclet number associated with the flow, the fluid interface is sharp and stable. When the micro-particles migrate to the interface, they are accumulated near the interface, resulting in effective focusing and separation of particles. We investigated several factors that affect the focusing and separation efficiency, including susceptibility of the ferrofluid, distance between the microfluidic channel and microscale magnet, and width of the microfluidic channel. This concept can be extended to multiple fluid interfaces. For example, a complete separation of micro-particles was demonstrated by using a three-stream multiphase flow configuration.

Abstract:A novel continuous switching/separation scheme of magnetic nanoparticles (MNPs) in a sub-microlitre fluid volume surrounded by neodymium permanent magnet is studied in this work using tangential microfluidic channels. Polydimethylsiloxane tangential microchannels are fabricated using a novel micromoulding technique that can be done without a clean room and at much lower cost and time. Negligible switching of MNPs is seen in the absence of magnetic field, whereas 90% of switching is observed in the presence of magnetic field. The flow rate of MNPs solution had dramatic impact on separation performance. An optimum value of the flow rate is found that resulted in providing effective MNP separation at much faster rate. Separation performance is also investigated for a mixture containing non-magnetic polystyrene particles and MNPs. It is found that MNPs preferentially moved from lower microchannel to upper microchannel resulting in efficient separation. The proof-of-concept experiments performed in this work demonstrates that microfluidic bioseparation can be efficiently achieved using functionalised MNPs, together with tangential microchannels, appropriate magnetic field strength and optimum flow rates. This work verifies that a simple low-cost magnetic switching scheme can be potentially of great utility for the separation and detection of biomolecules in microfluidic lab-on-a-chip systems.
Enhancing Magnetic Micro- and Nanoparticle Separation with a Cost-Effective Microfluidic Device Fabricated by Laser Ablation of PMMA

Abstract:Superparamagnetic iron oxide micro- and nanoparticles have significant applications in biomedical and chemical engineering. This study presents the development and evaluation of a novel low-cost microfluidic device for the purification and hyperconcentration of these magnetic particles. The device, fabricated using laser ablation of polymethyl methacrylate (PMMA), leverages precise control over fluid dynamics to efficiently separate magnetic particles from non-magnetic ones. We assessed the device's performance through Multiphysics simulations and empirical tests, focusing on the separation of magnetite nanoparticles from blue carbon dots and magnetite microparticles from polystyrene microparticles at various total flow rates (TFRs). For nanoparticle separation, the device achieved a recall of up to 93.3 ± 4% and a precision of 95.9 ± 1.2% at an optimal TFR of 2 mL/h, significantly outperforming previous models, which only achieved a 50% recall. Microparticle separation demonstrated an accuracy of 98.1 ± 1% at a TFR of 2 mL/h in both simulations and experimental conditions. The Lagrangian model effectively captured the dynamics of magnetite microparticle separation from polystyrene microparticles, with close agreement between simulated and experimental results. Our findings underscore the device's robust capability in distinguishing between magnetic and non-magnetic particles at both micro- and nanoscales. This study highlights the potential of low-cost, non-cleanroom manufacturing techniques to produce high-performance microfluidic devices, thereby expanding their accessibility and applicability in various industrial and research settings. The integration of a continuous magnet, as opposed to segmented magnets in previous designs, was identified as a key factor in enhancing magnetic separation efficiency.
High-Resolution Separation of Nanoparticles Using a Negative Magnetophoretic Microfluidic System

Abstract:The separation and purification of a sample of interest is essential for subsequent detection and analysis procedures, but there is a lack of effective separation methods with which to purify nano-sized particles from the sample media. In this paper, a microfluidic system based on negative magnetophoresis is presented for the high-resolution separation of nanoparticles. The system includes on-chip magnetic pole arrays and permalloys that symmetrically distribute on both sides of the separation channel and four permanent magnets that provide strong magnetic fields. The microfluidic system can separate 200 nm particles with a high purity from the mixture (1000 nm and 200 nm particles) due to a magnetic field gradient as high as 10,000 T/m being generated inside the separation channel, which can provide a negative magnetophoretic force of up to 10 pN to the 1000 nm particle. The overall recovery rate of the particles reaches 99%, the recovery rate of 200 nm particles is 84.2%, and the purity reaches 98.2%. Compared with the existing negative magnetophoretic separation methods, our system not only exhibits high resolution on particle sizes (800 nm), but also improves the sample processing throughput, which reaches 2.5 μL/min. The microfluidic system is expected to provide a new solution for the high-purity separation of nanoparticles, as well as nanobiological samples.

Abstract:The timely detection and diagnosis of diseases and accurate monitoring of specific genetic conditions require rapid and accurate separation, sorting, and direction of target cell types toward a sensor device surface. In that regard, cellular manipulation, separation, and sorting are progressively finding application potential within various bioassay applications such as medical disease diagnosis, pathogen detection, and medical testing. The aim of this paper is to present the design and development of a simple traveling wave ferro-microfluidic device and system rig purposed for the potential manipulation and magnetophoretic separation of cells in water-based ferrofluids. This paper details in full: (1) a method for tailoring cobalt ferrite nanoparticles for specific diameter size ranges (10–20 nm), (2) the development of a ferro-microfluidic device for potentially separating cells and magnetic nanoparticles, (3) the development of a water-based ferrofluid with magnetic nanoparticles and non-magnetic microparticles, and (4) the design and development of a system rig for producing the electric field within the ferro-microfluidic channel device for magnetizing and manipulating nonmagnetic particles in the ferro-microfluidic channel. The results reported in this work demonstrate a proof of concept for magnetophoretic manipulation and separation of magnetic and non-magnetic particles in a simple ferro-microfluidic device. This work is a design and proof-of-concept study. The design reported in this model is an improvement over existing magnetic excitation microfluidic system designs in that heat is efficiently removed from the circuit board to allow a range of input currents and frequencies to manipulate non-magnetic particles. Although this work did not analyze the separation of cells from magnetic particles, the results demonstrate that non-magnetic (surrogates for cellular materials) and magnetic entities can be separated and, in some cases, continuously pushed through the channel based on amperage, size, frequency, and electrode spacing. The results reported in this work establish that the developed ferro-microfluidic device may potentially be used as an effective platform for microparticle and cellular manipulation and sorting.

Abstract:Accurate separation of microparticles by shape has diverse applications in biology and biotechnology, but is a significant challenge in separation science and engineering. We demonstrate a simple and effective mechanism that can achieve shape-based separation of magnetic particles in microscale flows. In this method, a uniform magnetic field is applied perpendicularly to the flow direction, and causes shape-dependent lateral migration of the particles. Using high-speed imaging, we studied the rotational dynamics of the ellipsoidal particles. It is found that the lateral migration is correlated with the asymmetric rotation of the particles. Different from existing techniques that use magnetic forces, our method uses shape-dependent magnetic torque but zero magnetic force.

Abstract:Based on sharp geometry of Ni nanowires, we developed a novel high-gradient magnetic separator that was composed of a nanowires array and a uniform magnetic field. When suspension of magnetic nanoparticles (MNPs) flowed through it, the relatively large nanoparticles or clusters were removed from the suspension so that the size distribution can be improved. The separation resulted from magnetic force so that extra molecules or solvents were unnecessary to add. The performance was proved by scanned electron microscopy characterization and dynamic light scattering measurement. The improvement in magnetic colloidal dispersivity is important for the biomedical application of MNPs. Our results may also play a role in microfluidic application and nanoparticle-based detection.

Abstract:AbstractMicrofluidic-based separation methods have been highlighted for a number of biological applications, such as single cell analysis, disease diagnostics, and therapeutics. Although a number of previous studies have been carried out to minimize the physical damage and chemical deformations of the sample during the separation process, it still remains a challenge. In this paper, we developed a microfluidic device with dual-neodymium magnet-based negative magnetophoresis for the separation of the microparticles and cells. The poly(ethylene oxide) (PEO) was added to the solution to increase the viscoelasticity of the medium which could assist the sorting of the microparticles in the microfluidic device even at low flow rates, while minimizing damage to the cells and microparticles. Following this method, it was possible to separate 10 and 16 μm microparticles with high efficiency of 99 ± 0.1%, and 97 ± 0.8%, respectively. We also demonstrated the separation of glioblastoma cancer cells and neural stem cells (NSCs) in the microfluidic device.

Abstract:The separation of target substances is a significant biological detection procedure, where magnetic microspheres can act as high-performance separation materials. However, challenges are still kept to fulfill all the requirements. A submicron magnetic poly (glycidyl methacrylate) (PGMA) microsphere was synthesized in this investigation by utilizing three distinct techniques: in situ coprecipitation, electrostatic self-assembly, and silica surface coating. The PGMA microspheres were initially produced through a soap-free emulsion polymerization technique, wherein the coagulation process was governed by surface charge density. This factor additionally impacted the size and monodispersity of the microspheres. Then, we discovered that the capping agent sodium citrate (Na 3 Cit) effectively regulated the superparamagnetism properties of magnetic microspheres; the critical size of the superparamagnetic was 10.9 nm. Furthermore, the concentration of Fe 2+ and Fe 3+ effectively regulated the saturation magnetization, a property that correlated with the nucleation rate of the Fe 3 O 4 crystal. Additionally, we demonstrated that the pH regulated the electrostatic self-assembly, and it was suggested that positively charged PGMA–NH 2 microspheres and negatively charged Fe 3 O 4 nanoparticles be tightly coupled. For application, the PGMA@Fe 3 O 4 microspheres were subsequently coated with SiO 2 , which had been surface-modified with carboxyl groups. The PGMA@Fe 3 O 4 and carboxyl-modified microspheres exhibited saturated magnetization values of 23.73 and 17.73 emu/g, respectively. These microspheres have been effectively utilized for the extraction of DNA from various sources such as Salmonella typhi, monkeypox virus, and clinical swab samples, suggesting the potential of these microspheres for nucleic acid separation in the biomedical domain.

Abstract:The migration of magnetic nanoparticles (MNPs) in solution under magnetic field gradients (magnetophoresis) is an essential step in many separation processes in the field of chemical and other engineering disciplines. Previous works show that weak magnetic gradients generated by handheld magnets are enough to induce substantial magnetophoresis of MNPs, due to hydrodynamic effect (magnetophoresis induced convection). However, it is not known how the properties of the MNPs and the fluid influence the significance of this hydrodynamic effect. In this work, we study experimentally and by theoretical simulations how the fluid viscosity, fluid flow confinement and magnetic field distribution affect the intensity of magnetophoresis induced convection. The theoretical simulation conducted in this study is based on the theoretical framework established in our previous works on the hydrodynamic effect of LGMS process involving dilute colloidal MNP system. We show that the intensity of the induced convection increases by lowering the viscosity, decreasing the geometrical confinement and increasing the magnetic field gradient. Also, we show that the effect of all these variables can be characterized by the value of the dimensionless magnetic Grashof number Gr m . These findings are crucial in the design, optimization and intensification of LGMS processes for real time applications.

Abstract:Multiomics studies at single-cell level require small volume manipulation, high throughput analysis and multiplexed detection, characteristics that droplet microfluidics can tackle. However, the initial step of molecules bioseparation remains challenging. Here, we describe a unique magnetic device to trap and extract magnetic particles in sub-nanoliter droplets, for compartmentalisation of detection steps. Relying on electrodeposition of NiFe structures and microfluidic manipulation, this technology has allowed the purification of genetic material at single-cell scale and was able to reach an extraction rate of 72% for a sample of purified oligonucleotides.
