Abstract:OBJECTIVE: To develop and validate a liquid chromatography-tandem mass spectrometry (LC-MS/MS) method for the quantification of paclitaxel in rat plasma and to study the pharmacokinetics of paclitaxel thermosensitive liposomes in rats. METHODS: The plasma was alkalized with sodium bicarbonate (pH 8.0) and extracted with tert-butyl methyl ether (TBME). Paclitaxel and IS(docetaxel) were separated on an Agilent Zorbax SB C18 column (2.1 mm × 50 mm, 3.5 μm) with the mobile phase consisting of methanol-0.5 mmol · L-1 ammonium acetate (70: 30, V/V) at a flow rate of 300 μL · min-1. The injection volume was 5 μL An Agilent 6410 A QQQ triple-quadrople mass spectrometer equipped with an electrospray ionization (ESI) source was used as detector and was operated in positive ion mode. Selected ion monitoring (SIM) was performed and the m/z of ions selected for quantitation were 876.3 → 308.1 (paclitaxel) and 830.2 → 549.2 (internal standard, docetaxel) . SD rats were injected paclitaxel thermosensitive liposomes and paclitaxel injection via tail vein at the dose of 7 mg · kg-1, respectively. Pharmacokinetic parameters were calculated. RESULTS: The chromatograms showed no endogenous interfering peaks in blank rat plasma. Each analysis was completed within 3.5 min. The linear range of paclitaxel in plasma was 2-1 000 ng · mL-1. The lower limit of quantification was 2 ng · mL-1. The intra-run and inter-run relative standard deviations (RSDs) were less than 15%. The differences of f1/2β, MRT, AUC and CL were significant between the two preparations. CONCLUSION: The method is sensitive, accurate, and convenient for the pharmacokinetic study of paclitaxel thermosensitive liposomes. Compared with paclitaxel injection, paclitaxel thermosensitive liposomes has a controlled release rate, which can alleviate the acute toxicity and improve the tolerance to a certain extent.
The Development of a Quantitative and Qualitative Method Based on UHPLC-QTOF MS/MS for Evaluation Paclitaxel–tetrandrine Interaction and Its Application to a Pharmacokinetic Study

Abstract:Paclitaxel is a broad-spectrum anti-cancer drug by targeting microtubulin. However, multidrug resistant (MDR) makes its clinical application more difficult and results in failure of chemotherapy. Tetrandrine as a potential multidrug resistant modulator could be combined with other anti-cancer drugs. In this study, ultra-performance liquid chromatography (UHPLC) combined with quadrupole time-of-flight mass spectrometry (QTOF) was applied to simultaneously qualitative and quantitative analysis of paclitaxel for the pharmacokinetic studies while combined with tetrandrine. This method was developed based on non-target screening mode IDA (Information Dependent Acquisition). As a result, the validated range was 0.25-64ng/ml (30µl plasma) for paclitaxel. Totally 33 metabolites of paclitaxel and tetrandine were identified in vivo and in vitro. The main metabolites of PTX were dose-dependent decreased with different amounts of tetrandine co-administration no matter in vivo and in vitro, the exposure of PTX increased in pharmacokinetic study. The verified method is sensitive accurate and effective for the simultaneous determination of paclitaxel and its metabolites in blood, urine and live microsome incubation samples and it was successfully applied to evaluate the pharmacokinetics and drug-drug interaction between paclitaxel and tetrandine. Furthermore, a biosensor technology, surface plasmon resonance (SPR) analysis was applied to preliminary evaluate the competitive protein binding of multiple components. The SPR analysis indicated that the affinity between 6-hydroxy-paclitaxel and micotubulin is similar to that between paclitaxel and micotubulin, and tetrandrine also does not form a competitive combination with paclitaxel. For human, 6-hydroxy-paclitaxel is the one of main metabolites of paclitaxel, so the results suggested that tetrandine has an influence on the metabolite of paclitaxel, but tetrandine and the main metabolites of PTX probably do not affect PTX's biological targeting, the effect of its pharmacological action needs to be further studied.

Abstract:Matrine and oxymatrine are the major bioactive compounds extracted from the root of Sophora flavescens Ait, which have been widely used in traditional Chinese medicines. The objective of the study was to investigate the effects of matrine or oxymatrine on hepatic cytochrome P450 (CYP450) and the underlying molecular mechanisms. Matrine (15, 75 and 150 mg/kg) or oxymatrine (36, 180 and 360 mg/kg) was administered to rats for 14 days and the activities of CYP450 were measured by the quantification of the metabolites from multiple CYP450 probe substrates, using validated liquid chromatography coupled with liquid chromatography-tandem mass spectrometry detection (LC-MS/MS) and high-performance liquid chromatography methods. The mRNA and protein expression levels of CYPs were determined by quantitative real-time reverse-transcription polymerase chain reaction and Western blotting analysis respectively. Interactions between matrine or oxymatrine and human constitutive androstane (CAR), pregnane X receptor were evaluated by means of the reporter gene assay in CV-1 cells. Our study showed that matrine and oxymatrine significantly induced the activity and gene expression of CYP2B1 in a dose-dependent manner; matrine (150 mg/kg) slightly induced the mRNA and protein expression of CYP2E1 and mildly inhibited the mRNA and protein expression of CYP3A1 in rats. Matrine or oxymatrine could activate human CAR and induce the CYP2B reporter construct in CV-1 cells. These results reveal that matrine and oxymatrine can induce the activity and expression of CYP2B1/2 in rats, and the underlying mechanism may be related to the activation of CAR.

Abstract:We investigated cytochrome P450 (P450)-catalyzed metabolism of the important cancer drugs paclitaxel and docetaxel in rat, pig, minipig, and human liver microsomes and cDNA-expressed P450 enzymes. In rat microsomes, paclitaxel was metabolized mainly to C3'-hydroxypaclitaxel (C3'-OHP) and to a lesser extent to C2-hydroxypaclitaxel (C2-OHP), di-hydroxypaclitaxel (di-OHP), and another unknown monohydroxylated paclitaxel. In pig and minipig microsomes, this unknown hydroxypaclitaxel was the main metabolite, whereas C3'-OHP was a minor product. In minipigs, C2-OHP was the next minor product. In human liver microsomes, 6 alpha-hydroxypaclitaxel (6 alpha-OHP) was the main metabolite, followed by C3'-OHP and C2-OHP. Among different cDNA-expressed human P450 enzymes (CYP1A2, 1B1, 2A6, 2C9, 2E1, and 3A4), only CYP3A4 enzyme formed C3'-OHP and C2-OHP. Docetaxel was metabolized in pig, minipig, rat, and human liver microsomes mainly to hydroxydocetaxel (OHDTX), whereas CYP3A-induced rat microsomes produced primarily diastereomeric hydroxyoxazolidinones. Human liver microsomes from 10 different individuals formed OHDTX at different rates correlated with CYP3A4 content. Troleandomycin as a selective inhibitor of CYP3A inhibited the formation of C3'-OHP, C2-OHP, and di-OHP, as well as the unknown OHP produced in rat, minipig, and pig microsomes. In human liver microsomes, troleandomycin inhibited C3'-OHP and C2-OHP formation, and a suitable inhibitor of human CYP2C8, fisetin, strongly inhibited the formation of 6 alpha-OHP, known to be catalyzed by human CYP2C8. In conclusion, the metabolism of docetaxel is the same in all four species, but metabolism of paclitaxel is different, and 6 alpha-OHP remains a uniquely human metabolite. Pigs and minipigs compared with each other formed the same metabolites of paclitaxel.

Abstract:Paclitaxel (PTX) is heralded as one of the most successful natural-product drugs for the treatment of refractory cancers. In humans, the hepatic metabolic transformation of PTX is primarily mediated by two cytochrome P450 enzymes (P450s): CYP3A4 and CYP2C8. The impact of P450 metabolism on the anticancer effectiveness of PTX is significant. However, the precise mechanism underlying selective P450-catalyzed reactions in PTX metabolism remains elusive. To address this knowledge gap, we conducted molecular docking and molecular dynamics simulations using multiple crystal structures of CYP3A4, which originally contained other ligands. These methods enabled us to determine the most plausible binding structure of PTX within the enzyme. By further employing hybrid quantum mechanics and molecular mechanics calculations, we successfully identified two primary pathways for the reaction between compound I (Cpd I) of CYP3A4 and PTX. One of these pathways involves the formation of an epoxide, while the other proceeds through a ketone intermediate.

Abstract:Many pharmacokinetic studies of paclitaxel formulations with or without Cremophor (CrEL) have been performed on experimental animals. However, limited studies describe the different pharmacokinetic behaviors of paclitaxel in animals. The different distribution of drug in blood fractions may have great effect on its pharmacokinetic behaviors. Our present study was designed to study the characteristics of paclitaxel distribution in human, rabbit and rat blood, by measuring plasma-to-blood ratio (PBR) of paclitaxel in vitro and in vivo, and analyzing the results of equilibrium dialysis of paclitaxel with erythrocyte, plasma and hemoglobin. It was demonstrated that the paclitaxel PBR values in rat, unlike those in rabbit, are most significantly different from those in human, which may be due to distinct affinity of paclitaxel to blood fractions among different species. The effect of CrEL on increasing paclitaxel plasma concentration and in vitro & in vivo correlation in animal PBR values were observed. The findings in this study are of significance in the evaluation of the newly developed formulations of paclitaxel.
Absolute Quantification of Cytochrome P450 and Uridinediphosphate Glucuronosyl Transferase Isoforms by Proteomics-based Approach

Abstract:A strategy for the absolute quantitation of metabolic enzymes in rat liver microsomes was developed based on shotgun-based proteomics approach. Rat liver microsomes were digested with trypsin, and drug metabolizing enzymes were determined by liquid chromatography tandem mass spectrometry (LC-MS/MS) using standard curves based on custom synthesized specific peptides for each enzyme. The assays for cytochrome P450 (CYP450) and uridine diphosphoglucuronosyl transferase (UGT) showed good linearity (r > 0.995) with lower limits of quantitation of 10 nM. A synthetic isotope labeled specific peptide was then used as internal standard to determine UGT1A1 by stable isotope dilution method. The labeled peptide behaved similarly to the unlabeled peptide in LC-MS/MS and the assay was linear in matrix solution. The UGT1A1 concentration was detected by the labeled peptide method to be 18.2 pmol mg−1 protein compared with 17.3 pmol mg−1 protein obtained by the standard curve method. Although the consistent results were achieved by the two methods, the stable isotope dilution method was more convenient and more suitable for high throughput determinations in complex systems.

Abstract:To study the pharmacokinetic characteristic of antitumorcaffeic acid-3,4-dihydroxybenzylethyl (-3,4-dihydroxyphenethyl) ester (CADPE) in human liver microsomes.Human liver microsomes were incubated in vitrowith CADPE for different times.LC-MS method was applied to determine the concentrations of CADPE and its metabolites of hydroxytyrosol and caffeic acid at the designed time points.AnLC-MS analytic method was established and validated to determine the concentrations of CADPE and its metabolites hydroxytyrosol and caffeic acid inhuman liver microsomes.CADPE metabolism showed single compartment model and it's half-life (T1/2) was 0.72 ± 0.09 h.The time to peak (Tmax) and the half-life(T1/2) for hydroxytyrosol was 1.3 ± 0.3 h,3.97 ± 0.58 h,respectively.Hydroxytyrosol metabolism showed single compartment characteristic.The relationship of caffeic acid concentration and time was logarithmic.The concentration of caffeic acid gradually increased and finally reached steady state with the gradual metabolism of CADPE.This study clarified the pharmacokinetic characteristics of CADPE and its metabolites hydroxytyrosol and caffeic acid in human liver microsomes.The data obtained also provided scientific basis for the further investigation of the pharmacokinetic characteristics of CADPE in animals and human.

Abstract:The pharmacokinetics, safety, and anticancer efficacy profiles of nanoparticle albumin-bound (nab)-paclitaxel formulations are superior to those of solvent-based paclitaxel formulations. The aims of the present study were to study the effects of nab-paclitaxel and solvent-based paclitaxel formulations on the metabolic profiles of the model cell line (A549) and attempt to elucidate the associated metabolic pathways. A mass spectrometry-based cell metabolomics approach and viability evaluation were used to explore the potential difference. Western blotting was utilized to measure the levels of relevant proteins, and carnitine palmitoyltransferase 1 (CPT1) activities were quantified. Fold changes normalized to controls in levels of carnitine and several acylcarnitines were significantly different (p < 0.05) between A549 cells treated with nab-paclitaxel and those treated with solvent-based paclitaxel. Relative to the controls, there were also significant fold change differences in palmitic and linoleic acid levels in the cell lysates, mitochondrial CPT1 activities, and mitochondrial medium-chain acyl-CoA dehydrogenase (MCAD) protein levels in the A549 cells subjected to the nab-paclitaxel and solvent-based paclitaxel formulations. Results suggested that the two formulations differentially modulated fatty acid oxidation in the A549 cells. While cell viability results did not reveal significant differences, the findings implied that a mass spectrometry-based cell metabolomics approach could be a sensitive tool to explore the differences caused by formulation changes without using animals. Since uncertainties of products containing nanomaterials warrant holistic screening to address safety concerns, the aforementioned approach may be of regulatory importance and is worth further investigation.This article has not yet been cited by other publications.

Abstract:The prominent stromal compartment surrounds pancreatic ductal adenocarcinoma (PDAC) protects the tumor cells from chemo- or radio-therapy. We hypothesized that our nano formulation carried cyclopamine (CPA, stroma modulator) and paclitaxel (PTX, anti-tumor agent) could increase the permeation of PTX through the stromal compartment and improve the intratumoral delivery of PTX. In the present study a sensitive, reliable UPLC-MS/MS method was developed and validated to quantify PTX and CPA simultaneously in mouse whole blood, pancreas, liver and spleen samples. Docetaxel was used as the internal standard. The method demonstrated a linear range of 0.5-2000 ng/mL for whole blood and tissue homogenates for both PTX and CPA. Accuracy and precision of the assay were all within ±15 %. Matrix effects for both analytes were within 15%. Recoveries from whole blood; liver, spleen and pancreas homogenates were 92.7-105.2% for PTX and 72.8-99.7% for CPA. The stability was within ±15% in all test biomatrices. The validated method met the acceptance criteria according to FDA regulatory guidelines. The method was successfully applied to support pharmacokinetic and biodistribution study for PTX and CPA in mice biomatrices.
Preparation and Pharmacokinetics of Paclitaxel Nanometric Liposomes in Rats

Abstract:Objective:To evaluate the pharmacokinetic profile of a novel paclitaxel nanometric liposomes in rats. Methods: Paclitaxel nanometic liposomes (PNL) were prepared by thin-film method with sonication and lyophilization. Rats were iv injected with PNL or Anzatax via the tail. The plasma samples of the rats were collected to extract paclitaxel residuals with ether and measured paclitaxel concentration using RP-HPLC method. The RP-HPLC condition was as followed; a Gemini ODS (150 mm x 4. 6 mm,5μm) , an eluate consisted of acetonitrile-ammonium acetate buffer (0. 035 mol·L-1 , pH 5. 0) (50:45) with a flow rate of 1. 0 mL·min-1 , and a detection wavelength at 230 nm. Diazepam was used as an internal control. Results:The PNL with a particle size of (54. 1±26. 0) nm and entrapment efficiency of 80% or more was compatible well with glucose solutions for injection within 24 hours. The plasma concentration-time curves of the PNL obeyed a two-compartment model. The half-life of PNL (t1/2β) was longer than Anzatax [ (3. 38±0. 39)h vs. (2. 49±0. 63)h,P 0. 05]. No statistical difference of PNL from Anzatax was found in the other pharmacokinetic parameters. The AUC ratio of PNL over Anzatax was 88. 13% . Conclusion:The paclitaxel nanometric liposomes had similar pharmacokinetic profiles to Anzatax.
Structural and Functional Insights into Polymorphic Enzymes of Cytochrome P450 2C8

Abstract:The cytochrome P450 (CYP) superfamily plays a key role in the oxidative metabolism of a wide range of drugs and exogenous chemicals. CYP2C8 is the principal enzyme responsible for the metabolism of the anti-cancer drug paclitaxel in the human liver. Nearly all previous works about polymorphic variants of CYP2C8 were focused on unpurified proteins, either cells or human liver microsomes; therefore their structure–function relationships were unclear. In this study, two polymorphic enzymes of CYP2C8 (CYP2C8.4 (I264M) and CYP2C8 P404A) were expressed in E. coli and purified. Metabolic activities of paclitaxel by the two purified polymorphic enzymes were observed. The activity of CYP2C8.4 was 25% and CYP2C8 P404A was 30% of that of WT CYP2C8, respectively. Their structure–function relationships were systematically investigated for the first time. Paclitaxel binding ability of CYP2C8.4 increased about two times while CYP2C8 P404A decreased about two times than that of WT CYP2C8. The two polymorphic mutant sites of I264 and P404, located far from active site and substrate binding sites, significantly affect heme and/or substrate binding. This study indicated that two important nonsubstrate recognition site (SRS) residues of CYP2C8 are closely related to heme binding and/or substrate binding. This discovery could be valuable for explaining clinically individual differences in the metabolism of drugs and provides instructed information for individualized medication.
Metabolic Profiling of the Novel Hypoxia-Inducible Factor 2α Inhibitor PT2385 In Vivo and In Vitro.

Abstract:PT2385 is a first-in-class, selective small-molecule inhibitor of hypoxia-inducible factor-2α (HIF-2α) developed for the treatment of advanced clear cell renal cell carcinoma. Preclinical results demonstrated that PT2385 has potent antitumor efficacy in mouse xenograft models of kidney cancer. It also has activity toward metabolic disease in a mouse model. However, no metabolism data are currently publically available. It is of great importance to characterize the metabolism of PT2385 and identify its effect on systemic homeostasis in mice. High-resolution mass spectrometry-based metabolomics was performed to profile the biotransformation of PT2385 and PT2385-induced changes in endogenous metabolites. Liver microsomes and recombinant drug-metabolizing enzymes were used to determine the mechanism of PT2385 metabolism. Real-time polymerase chain reaction analysis was employed to investigate the reason for the PT2385-induced bile acid dysregulation. A total of 12 metabolites of PT2385 was characterized, generated from hydroxylation (M1, M2), dihydroxylation and desaturation (M3, M4), oxidative-defluorination (M7), glucuronidation (M8), N-acetylcysteine conjugation (M9), and secondary methylation (M5, M6) and glucuronidation (M10, M11, and M12). CYP2C19 was the major contributor to the formation of M1, M2, and M7, UGT2B17 to M8, and UGT1A1/3 to M10-M12. The bile acid metabolites taurocholic acid and tauro-β-muricholic acid were elevated in serum and liver of mice after PT2385 treatment. Gene expression analysis further revealed that intestinal HIF-2α inhibition by PT2385 treatment upregulated the hepatic expression of CYP7A1, the rate-limiting enzyme in bile acid synthesis. This study provides metabolic data and an important reference basis for the safety evaluation and rational clinical application of PT2385.

Abstract:The in vivo fate of nanoformulated drugs is governed by the physicochemical properties of the drug and the functionality of nanocarriers. Nanoformulations such as polymeric micelles, which physically encapsulate poorly soluble drugs, release their payload into the bloodstream during systemic circulation. This results in three distinct fractions of the drug-nanomedicine: encapsulated, protein-bound, and free drug. Having a thorough understanding of the pharmacokinetic (PK) profiles of each fraction is essential to elucidate mechanisms of nanomedicine-driven changes in drug exposure and PK/PD relationships pharmacodynamic activity. Here, we present a comprehensive preclinical assessment of the poly (2-oxazoline)-based polymeric micelle of paclitaxel (PTX) (POXOL hl-PM), including bioequivalence comparison to the clinically approved paclitaxel nanomedicine, Abraxane®. Physicochemical characterization and toxicity analysis of POXOL hl-PM was conducted using standardized protocols by the Nanotechnology Characterization Laboratory (NCL). The bioequivalence of POXOL hl-PM to Abraxane® was evaluated in rats and rhesus macaques using the NCL's established stable isotope tracer ultrafiltration assay (SITUA) to delineate the plasma PK of each PTX fraction. The SITUA study revealed that POXOL hl-PM and Abraxane® had comparable PK profiles not only for total PTX but also for the distinct drug fractions, suggesting bioequivalence in given animal models. The comprehensive preclinical evaluation of POXOL hl-PM in this study showcases a series of widely applicable standardized studies by NCL for assessing nanoformulations prior to clinical investigation.
Sensitive and Rapid UHPLC-MS/MS Assay for Simultaneous Quantifications of Calcipotriol and Paclitaxel in Rat Whole Blood and Plasma Samples

Abstract:Vitamin-D analogues have emerged as potential stroma-modulating agents for the treatment of pancreatic ductal adenocarcinoma (PDAC). One such agent, calcipotriol (Cal) has shown significant activity in in vitro and in vivo models of pancreatic ductal adenocarcinoma. Attempts in our lab have been focused on establishing the therapeutic merits of co-formulating this agent with the chemotherapeutic drug paclitaxel (PTX) in animal models. Accurate and reliable quantifications of these agents is critical to successful pharmacokinetic/pharmacodynamic (PK/PD) projections from animals into humans. Herein, we developed a LC–MS/MS assay for measuring Cal and PTX in whole blood and plasma.A liquid-liquid analyte extraction procedure, using a mixture of water-MeOH (50:50, v/v) and hexane-dichloromethane- isopropyl alcohol (150:15:5, v/v/v) was used. Chromatographic separation was carried out on Kinetex C18 column (1.7 μm, 100 × 2.10 mm) under an isocratic elution at a flow rate of 0.4 mL/min with a total runtime of 3.5 min. The mobile phase was composed of ammonium acetate (pH 6.51; 5 mM)-methanol (15:85, v/v). The analytes were ionized by positive electrospray ionization using API 5500-Qtrap triple quadrupole mass spectrometer (Applied Biosystem/AB SCIEX). The linearity of calibration curves for both analytes were established at 0.5 (LLOQ)-500 ng/mL with correlation coefficients exceeding 0.99. Spiked whole blood and plasma samples were used as surrogates for matrix validation. For both analytes, the intra-day and inter-day accuracies were 90.5–105 % and 96.6–106 %, respectively, while the corresponding precisions were 3.09–10.7 % and 5.20–12.9 %. No carryover was observed for the analytes which also remained acceptably stable in the surrogate matrices under relevant conditions.The assay is robust, reliable and sensitive in rat whole blood and plasma. The analytes extraction procedure performs acceptably well in both matrices with high recoveries and minimal matrix effects. Additionally, only 20 μL of rat whole blood or plasma is required and the total run time per sample is 3.5 min.PK studies enabled by the assay revealed that when co-administered, PTX AUC0→∞ and Cmax increased while those of Cal decreased. This finding alerts a potential drug-drug interaction and warrants further investigation in studies using this combination regimen.

Abstract:To identify the metabolite and CYP450 isoforms involved in rat liver microsomal metabolism of TM2O8.The present study investigated the metabolism of TM208 and the effects of selective CYP450 inhibitors on the metabolism of TM208 in rat liver microsomes.Various specific inhibitors of CYP were used to identify the isoforms of CYP involved in the metabolism of TM208.The inhibitor of CYP2D and that of CYP2B had strong inhibitory effects on TM208 metabolism in a concentration-dependant manner,the inhibitor of CYPIA had a modest inhibitory effect,and the inhibitor of CYP3A seemed not to have an obvious inhibitory effect on TM208 metabolism.TM208 might mainly be metabolized by CYP2D and CYP2B in rat liver microsomes.

Abstract:OBJECTIVE To compare and analysis the effect of three different P-glycoprotein(P-gp)inhibitors on the pharmacokinetic behaviors of paclitaxel by intravenous administration to rats.METHODS 24 Rats were divided into 4 groups.One group was given Taxol alone and the other three groups were given Taxol with Cyclosporin A(CsA),verapamil or quercetin,respectively.Plasma concentration of paclitaxel was analyzed by HPLC,and pharmacokinetics parameters were calculated by 3P97.RESULTS CsA,verapamil and quercetin decreased the elimination of paclitaxel and increased the AUC of paclitaxel.CONCLUSION Paclitaxel undergoes the elimination effect of P-gp and CYP3A,the agents that simultaneously inhibit P-gp and CYP3A could significantly increase the bioavailability and decrease the elimination of paclitaxel.

Abstract:3FDT, an analog of docetaxel with a blocked metabolism at its 3'-N-tert-butyloxyl group with three fluorine atoms, exhibits more potent cytotoxicity than docetaxel both with human cancer cell line SKOV-3 in vitro and with human non-small cell lung cancer A549 xenografts in vivo. To further develop pharmacodynamically and pharmacokinetically favorable fluorinated docetaxel analogs as anticancer agents, we chose 3FDT as the model compound to identify the metabolites of 3FDT in RLMs, rats, and HLMs and the cytochrome P450 enzymes responsible for the metabolism of 3FDT. Our findings indicated that the major metabolic site switched from the C3' appendage for docetaxel to the taxane ring for 3FDT, and the main metabolizing P450 enzymes switched from CYP3A to CYP3A4 and CYP2E1.
