Abstract:The biodegradation of organophosphorus compounds (OPs) by phosphotriesterase (PTE) is an efficient and ecofriendly way. However, the low catalytic activity and instability of PTE becomes the key limiting step for the practical application. In this study, a new multimetal-based organic-inorganic self-assembly hybrid system was proposed to ameliorate this situation. A series of characterization methods demonstrated that the synthesized multimetallic PTE hybrid complex (Co/MnHF@PTE) exhibited a hierarchical flower-like structure. The activity recovery and the effective encapsulation yield of the Co/MnHF@PTE was 408% and 84.5%, respectively. The kcat/Km of Co/MnHF@PTE was separately 2.9-, 1.3-, and 2.7-fold higher than MnHF@PTE, CoHF@PTE, and free PTE. The hybrid nanoflowers showed preferable durability, stability, and reusability. When Co/MnHF@PTE was applied in a pump-flow reactor, the degradation percent of methyl parathion could be maintained at more than 93% with the injection of 24 ml of reaction mixture. More importantly, the Co/MnHF@PTE can effectively break P-F or P-S bond of soman (GD) and nerve agent VX to produce non-toxic products through hydroxyl nucleophilic attack. This study provides a feasible method to improve the activity and stability of PTE, promoting the biodegradation of OPs in the environment.
Aerobic Biodegradation Kinetics and Pathway of the Novel Herbicide ZJ0273 in Soils

Abstract:Degradation of ZJ0273, a recently developed pyrimidynyloxybenzoic‐based herbicide, was investigated in five different soils under aerobic conditions. ZJ0273 degradation rate was strongly affected by soil physico‐chemical characteristics and the inoculation of ZJ0273‐degrading bacteria. Greater organic matter (OM) content, neutral pH and inoculation of ZJ0273‐degrading bacteria can increase degradation rate and decrease the half‐life value (DT50). At 30°C the biodegradation rate of ZJ0273 reached 41–85% in natural (unsterilized) soils. It ranged from 69 to 96% at 90 days after treatment (DAT) in five different types of soils after re‐inoculation of Amycolatopsis sp. M3‐1 and DT50 decreased by 34 , 81, 16, 20 and 32 days, respectively, in soils S1, S2, S3, S4 and S5. Furthermore, using the six metabolites (M1–M6) identified six metabolites (M1–M6) by liquid chromatography‐mass spectrometry (LC‐MS) and their behaviour, a biodegradation pathway of ZJ0273 in soils was proposed. New metabolites, M5 and M6, were found in soils. Biodegradation of ZJ0273 involved continuous biocatalytic reactions, such as de‐estering, hydrolysis, acylation, C‐N cleavage, de‐methyl and ether cleavage reactions. Finally, ZJ0273 was bio‐transformed into M4 and M6, which could be degraded and oxidized into CO 2 and H 2 O through the tricarboxylic acid (TCA) cycle.
Recent insights into the microbial catabolism of aryloxyphenoxy-propionate herbicides: microbial resources, metabolic pathways and catabolic enzymes

Abstract:Aryloxyphenoxy-propionate herbicides (AOPPs) are widely used to control annual and perennial grasses in broadleaf crop fields and are frequently detected as contaminants in the environment. Due to the serious environmental toxicity of AOPPs, there is considerable concern regarding their biodegradation and environmental behaviors. Microbial catabolism is considered as the most effective method for the degradation of AOPPs in the environment. This review presents an overview of the recent findings on the microbial catabolism of various AOPPs, including fluazifop- P -butyl, cyhalofop-butyl, diclofop-methyl, fenoxaprop- P -ethyl, metamifop, haloxyfop- P -methyl and quizalofop- P -ethyl. It highlights the microbial resources that are able to catabolize these AOPPs and the metabolic pathways and catabolic enzymes involved in their degradation and mineralization. Furthermore, the application of AOPPs-degrading strains to eliminate AOPPs-contaminated environments and future research hotspots in biodegradation of AOPPs by microorganisms are also discussed.

Abstract:Fenoxaprop-P-ethyl (FE) is widely used as a post-emergence aryloxyphenoxy propionate (AOPP) herbicide in agriculture. An efficient FE-degrading strain DL-2 was isolated from the enrichment culture and identified as Acinetobacter sp. and the metabolite fenoxaprop acid (FA) was identified by HPLC/MS analysis. The strain DL-2 could also degrade a wide range of other AOPP herbicides. A novel FE hydrolase esterase gene afeH was cloned from strain DL-2 and functionally expressed in Escherichia coli BL21(DE3). The specific activities of recombinant AfeH was 216.39 U mg(-1) for FE with Km and Vmax values of 0.82 μM and 7.94 μmol min(-1) mg(-1). AfeH could also hydrolyze various AOPP herbicides, p-nitrophenyl esters and triglycerides. The optimal pH and temperature for recombinant AfeH were 9.0 and 50°C, respectively; the enzyme was activated by Co(2+) and inhibited by Ca(2+), Zn(2+), Ba(2+). AfeH was inhibited strongly by phenylmethylsulfonyl and SDS and weakly by dimethyl sulfoxide.

Abstract:Diclofop-methyl, an aryloxyphenoxypropionate (AOPP) herbicide, is a chiral compound with two enantiomers. Microbial detoxification and degradation of various enantiomers is garnering immense research attention. However, enantioselective catabolism of diclofop-methyl has been rarely explored, especially at the molecular level. This study cloned two novel hydrolase genes (dcmA and dcmH) in Sphingopyxis sp. DBS4, and characterized them for diclofop-methyl degradation. DcmA, a member of the amidase superfamily, exhibits 26.1-45.9% identity with functional amidases. Conversely, DcmH corresponded to the DUF3089 domain-containing protein family (a family with unknown function), sharing no significant similarity with other biochemically characterized proteins. DcmA exhibited a broad spectrum of substrates, with preferential hydrolyzation of (R)-(+)-diclofop-methyl, (R)-(+)-quizalofop-ethyl, and (R)-(+)-haloxyfop-methyl. DcmH also preferred (R)-(+)-quizalofop-ethyl and (R)-(+)-haloxyfop-methyl degradation while displaying no apparent enantioselective activity towards diclofop-methyl. Using site-directed mutagenesis and molecular docking, it was determined that Ser175 was the fundamental residue influencing DcmA's activity against the two enantiomers of diclofop-methyl. For the degradation of AOPP herbicides, DcmA is an enantioselective amidase that has never been reported in research. This study provided novel hydrolyzing enzyme resources for the remediation of diclofop-methyl in the environment and deepened the understanding of enantioselective degradation of chiral AOPP herbicides mediated by microbes.

Abstract:The enantioselective catabolism of the two enantiomers of the aryloxyphenoxypropionate (AOPP) herbicide, quizalofop-ethyl (QE), in environment by microorganisms is of great interest. In this study, Sphingobium sp. strain QE-1, capable of transforming both enantiomers of QE, was successfully isolated. A novel gene qeH, encoding an esterase capable of initially transforming (R)/(S)-QE to (R)/(S)-quizalofop acid [(R)/(S)-QA], was cloned. QeH showed the highest identity (36.9%) to the β-lactamase MT1414, and the purified QeH displayed optimal activity for (R)-QE at 40 °C and pH 7.0. QeH preferentially hydrolyzed (R)-QE than (S)-QE. Based on molecular docking analyses, the distance between the ligand molecule and the coordination atom was shorter when (R)-QE occupied the active sites as compared to that of (S)-QE, which resulted in a stronger ligand binding affinity for (R)-QE. Besides the two enantiomers of QE, QeH could also convert many other AOPP herbicides with different catalytic efficiency (clodinafop-propargyl > fenoxaprop-P-ethyl > fluazifop-P-butyl > cyhalofop-butyl). This study will enhance our understandings of the enantioselective catabolism of AOPP herbicides by microorganisms.

Abstract:Clodinafop-propargyl (CP) is a widely used post-emergence aryloxyphenoxy propionate (AOPP) herbicide and causes serious environmental problems. The elimination of CP from the environment is urgently required, but there are few reports on microbial degradation of CP. In this study, a bacterial consortium WP was enriched from the activated sludge of CP containment, which could completely degrade 50 mg L −1 of CP within 7 days. UPLC-MS analysis showed that the main intermediate metabolites of CP degradation were clodinafop acid (CA), 4-(5-chloro-3-fluoro-2-pyridyloxy) phenol, and 5-chloro-3-fluoropyridin-2-ol. And the metabolic pathway of CP degradation by consortium WP has been proposed. High-throughput sequencing revealed that the consortium WP had rich bacterial diversity, including 4 phyla, 14 classes, and 148 genera. Ten bacteria capable of converting CP to CA were isolated from the consortium WP using LB medium containing CP, and the strain Pseudoxanthomonas sp. WP66 had the strongest conversion ability, which could utilize CP as the sole source for growth and degrade 91.20% of 100 mg L −1 CP within 15 h. This study presented the main degradation pathways of CP by microorganisms, and provided resource of bacterial strain for the removal of CP from the environment.

Abstract:The enantioselective mechanism of the esterase QeH against the two enantiomers of quizalofop–ethyl (QE) has been primitively studied using computational and experimental approaches. However, it is still unclear how the esterase QeH adjusts its conformation to adapt to substrate binding and promote enzyme–substrate interactions in the catalytic kinetics. The equilibrium processes of enzyme–substrate interactions and catalytic dynamics were reproduced by performing independent molecular dynamics (MD) runs on the QeH-(R)/(S)-QE complexes with a newly developed residue-specific force field (RSFF2C). Our results indicated that the benzene ring of the (R)-QE structure can simultaneously form anion–π and cation–π interactions with the side-chain group of Glu328 and Arg384 in the binding cavity of the QeH-(R)-QE complex, resulting in (R)-QE being closer to its catalytic triplet system (Ser78-Lys81-Tyr189) with the distances measured for the hydroxyl oxygen atom of the catalytic Ser78 of QeH and the carbonyl carbon atom of (R)-QE of 7.39 Å, compared to the 8.87 Å for (S)-QE, whereas the (S)-QE structure can only form an anion–π interaction with the side chain of Glu328 in the QeH-(S)-QE complex, being less close to its catalytic site. The computational alanine scanning mutation (CAS) calculations further demonstrated that the π–π stacking interaction between the indole ring of Trp351 and the benzene ring of (R)/(S)-QE contributed a lot to the binding stability of the enzyme–substrate (QeH-(R)/(S)-QE). These results facilitate the understanding of their catalytic processes and provide new theoretical guidance for the directional design of other key enzymes for the initial degradation of aryloxyphenoxypropionate (AOPP) herbicides with higher catalytic efficiencies.

Abstract:Esterase, as a type of powerful catabolic enzyme for the degradation of pyrethroid pesticides (PYRs), appears promising in improving the quality of crops and the environment contaminated by pesticide residues. The purpose of this research is to provide a detailed introduction to the enzymatic properties, optimal production and immobilization conditions, and the degradation ability of Est804 for PYRs. The study on enzymatic properties indicated that Est804 was an alkaline esterase with an optimal pH of 8.0 and a broad optimal temperature in the range of 35−50°C. The optimal activity of free Est804 was calculated to be 112.812 U, and the specific enzyme activity was 48.97 U/mg. The kinetic parameters of Est804 were K m = 0.613 mM, k cat = 12,371 s –1 , and V m = 0.095 mM/min. The results of the fermentative optimization demonstrated that the optimal conditions included 1.5% of inoculation amount, 30 mL of liquid volume, 28°C of the fermentation temperature, and 18 h of the fermentation time. The optimal medium consists of 15.87 g of yeast powder, 8.00 g of glycerol, and 9.57 g of tryptone in 1 L of liquid. The optimized enzyme activity was 1.68-fold higher than that before optimization. Immobilized Est804 exhibited the highest activity under the optimum preparation conditions, including 0.35 g of chitosan dosage, 0.4 mL of an enzyme, and 4 h at 40°C for adsorption. The degradation rates of Cypermethrin (CYP), fenpropathrin (FE), and lambda-cyhalothrin (LCT) by Est804 within 30 min were 77.35%, 84.73%, and 74.16%, respectively. The present study indicated that Est804 possesses great potential for the treatment of pesticide residues on crops and environmental remediation, conducive to the development of SGNH family esterase against pyrethroid accumulation.

Abstract:Complex rhizoremediation is the main mechanism of phytoremediation in organic-contaminated soil. Low molecular weight organic acids (LMWOAs) in root exudates have been shown to increase the bioavailability of contaminants and are essential for promoting the dissipation of contaminants. The effects of root exudates on the dissipation of organophosphate esters (OPEs) in soil are unclear. Consequently, we studied the combined effects of root exudates, soil enzymes and microorganisms on OPEs (tri (1-chloro-2-propyl) phosphate (TCPP) and triphenyl phosphate (TPP)) dissipation through pot experiments. Oxalic acid (OA) was confirmed to be the main component of LMWOAs in root exudates of ryegrass. The existence of OA increased the dissipation rate of OPEs by 6.04%-25.50%. Catalase and dehydrogenase activities were firstly activated and then inhibited in soil. While, urease activity was activated and alkaline phosphatase activity was inhibited during the exposure period. More bacteria enrichment (e.g., Sphingomonas, Pseudomonas, Flavisolibacter, Pontibacter, Methylophilus and Massilia) improved the biodegradation of OPEs. In addition, the transformation paths of OPEs hydrolysis and methylation under the action of root exudates were observed. This study provided theoretical insights into reducing the pollution risk of OPEs in the soil.

Abstract:Phthalate ester (PAE) pollution is an increasing problem globally. Paracoccus kondratievae BJQ0001 was isolated from the fermentation starter of Baijiu and showed an efficient degradation capability toward PAEs. To our poor knowledge, this is the first report of a P. kondratievae strain capable of degrading PAEs. The first complete genome sequence of P. kondratievae was presented without gaps, and composed of two circular chromosomes and one plasmid. The species simultaneously degraded di-methyl phthalate (DMP), di-ethyl phthalate (DEP), di-butyl phthalate (DBP), di-isobutyl phthalate (DIBP) and di-(2-ethylhexyl) phthalate (DEHP), with DMP and DEP as the preferred substrates. The half-life (t1/2) of DMP was only 6.34 h with an initial concentration of 200 mg/L. Combined with gene annotation and metabolic intermediate analysis, a metabolic pathway was proposed for the species. Benzoic acid, the intermediate of anaerobic PAE metabolism, was identified in the aerobic degradation process. Two key enzymes for alkyl ester bond hydrolysis were obtained, and belonged to families IV and VI of hydrolases, respectively. These results will promote the investigation of PAE degradation by P. kondratievae, and provide useful information for improving the quality control of food and environmental PAE treatment.

Abstract:Farmland soil sprayed with organophosphorus pesticides (OPs) annually was investigated for the identification and characterization of OP-degrading microorganisms. Six bacterial strains were identified, including Brevundimonas faecalis MA-B12 and Alcaligenes faecalis subsp. parafaecalis MA-B13 for methamidophos degradation, Citrobacter freundii TF-B21 and Ochrobactrum intermedium TF-B23 for trichlorfon degradation, Ochrobactrum intermedium DV-B31 for dichlorvos degradation, and Bacillus cereus for dimethoate degradation. The optimal biodegradation conditions for OPs were obtained at pH 7.0 and incubation temperature ranging from 28 to 37 °C. In an 8-day batch test, biodegradation of the four OPs all followed first-order kinetics, with biodegradation rates ranging from 58.08 to 96.42%. Functional genes responsible for OPs degradation were obtained, including ophB, ampA, opdE, opd, opdA, and mpd. As these strains were indigenous strains isolated from farmland soils, they can be potentially used as bacterial consortium for the bioremediation of mixed OP-contaminated soils. A time-course genotoxicity assessment of the degradation products was done by a bacterial whole-cell bioreporter, revealing that biodegradation of trichlorfon, dichlorvos, and dimethoate resulted a decreased genotoxicity within 5 days, which, however, significantly increased on day 8. The result demonstrated that more toxic products may be produced during the biodegradation processes of OPs, and more attention should be put not only on the pesticides themselves, but also on the toxic effects of their degradation products. To the best of our knowledge, this is for the first time that the genotoxicity of OP degradation products was evaluated by the bioreporter assay, broadening our understanding on the genotoxic risks of OPs during biodegradation process.Graphical Abstract

Abstract:however, because the principal treatment method used with alcoholics is group therapy. A further major difficulty (perhaps to a degree unique among alcoholics and drug addicts) is a low degree of motivation and, more important, an unwillingness to co-operate, certainly over a long period of treatment. This is well shown in Dr Gareth Lloyd's article (18 September, p 785). Many patients frankly tell us at a later date that while they were attending treatment and going through the motions they had no intentions of stopping drinking, nor did they accept that they were alcoholic. More important, they soon made it clear by their non-attendance that they would not co-operate with treatment. It is as if hypertensive patients did not accept that they were hypertensive and refused to take their drugs or attend follow-up clinics. In these circumstances it is not likely that the antihypertensive drugs or hypertension clinic would be regarded as of no value. In order to show that treatment is ineffective in the severely dependent alcoholic, which is the type of patient who attends hospital, it will be necessary to show at least that there is no change occurring in drinking behaviour in patients who have shown quantifiably that they have undergone regular persistent treatment over a period of at least several years. Abstract:This work performed a large scale assessment for organophosphorus pesticides (OPPs) degradation activity of 121 Lactobacillus (L.) plantarum strains. Six L. plantarum strains (P9, IMAU80110, IMAU40100, IMAU10585, IMAU10209, and IMAU80070) were found to possess high capacity of degrading three commonly used OPPs, namely dimethoate, phorate, and omethoate; and they were selected for more detailed characterization. Moreover, the three OPPs were mainly detected in the culture supernatants but not in the cell extracts, further confirming that the OPPs were degraded rather than absorbed by the cells. Among the six selected strains, P9 was most tolerant to gastrointestinal juices and bile. We thus used ultra-high performance liquid chromatography electron spray ionization coupled with time-of-flight mass spectrometry (UPLC/ESI-Q-TOF/MS) to generate the metabolomic profiles of the strain P9 growing in MRS medium with and without containing phorate. By using orthogonal partial least squares discriminant analysis, we identified some potential phorate-derived degradative products. This work has identified novel lactic acid bacteria resources for application in pesticide degradation. Our results also shed light on the phorate degradation mechanism by L. plantarum P9.

Biodegradation of phthalate acid esters by a versatile PAE-degrading strain Rhodococcus sp. LW-XY12 and associated genomic analysis

Abstract:Di-(2-ethylhexyl) phthalate (DEHP), the most extensively used phthalate acid esters (PAEs), poses a potential risk to human and environment. A novel bacterial strain, Rhodococcus sp. LW-XY12, with efficient PAE-degrading capability, was isolated from activated sludge. Strain LW-XY12 could degrade 96.91 ± 0.68% of DEHP (500 mg L−1) within 32 h. The degradation curves of DEHP (50–1500 mg L−1) and PAE mixture (500 mg L−1 each) fitted well with the modified Gomperz kinetics model. DEHP metabolic pathway was reconstructed by using genome annotation and metabolic intermediate analyses. The DEHP metabolic pathway might comprise de-esterification and β-oxidation. Homologous modeling and molecular docking analysis revealed that DEHP and MEHP (mono-(2-ethylhexyl) phthalate) could be bound to putative carboxylesterase (KXC42_04905) via hydrogen bonding and hydrophobic interaction. A conserved catalytic triad (Ser195-Glu319-His412) might act as an active protein pocket and catalyze the hydrolysis of DEHP. Carboxylesterase (KXC42_04905) could hydrolyze ester bond and a possible mechanism underlying ester bond hydrolysis catalyzed by the carboxylesterase was proposed. Protocatechuate 3,4-dioxygenase, 3-oxoadipate CoA-transferase, benzoate 1,2-dioxygenase, catechol 1,2-dioxygenase, and catechol 2,3-dioxygenase transcripts were detected and significantly up-regulated under DEHP induction. This study sheds light on the functional genes associated with DEHP degradation and metabolic mechanism in this versatile bacterium for degradation of PAEs.

Abstract:Ochratoxin A (OTA) is a toxic secondary metabolite that widely contaminates agro-products and poses a significant dietary risk to human health. Previously, a carboxypeptidase CP4 was characterized for OTA degradation in Lysobacter sp. CW239, but the degradation activity was much lower than its host strain CW239. In this study, an amidohydrolase ADH2 was screened for OTA hydrolysis in this strain. The result showed that 50 μg/L OTA was completely degraded by 1.0 μg/mL rADH2 within 5 min, indicating ultra-efficient activity. Meanwhile, the two hydrolases (i.e., CP4 and ADH2) in the strain CW239 showed the same degradation manner, which transformed the OTA to ochratoxin α (OTα) and l-β-phenylalanine. Gene mutants (Δcp4, Δadh2 and Δcp4-adh2) testing result showed that OTA was co-degraded by carboxypeptidase CP4 and amidohydrolase ADH2, and the two hydrolases are sole agents in strain CW239 for OTA degradation. Hereinto, the ADH2 was the overwhelming efficient hydrolase, and the two types of hydrolases co-degraded OTA in CW239 by synergistic effect. The results of this study are highly significant to ochratoxin A contamination control during agro-products production and postharvest.
[Isolation and analysis of human leukocyte interferons synthesized by bacterial producer strains].

Abstract:Rapid and sensitive methods for immunochemical estimation of bacterially-made human leukocytic interferons were developed. A large number of strains producing such interferons was analyzed with these methods and optimal ones capable of synthesizing up to 10(10), 10(8), 10(9) and 10(9) IU/l of interferons alpha A, alpha N, alpha F and alpha I1 respectively were selected. The proteins were isolated in homogenous state and their main physico-chemical parameters were characterized. Interferon oligomers were detected. Their formation was partially due to intermolecular disulphide exchange. The effect on interferon alpha A of retinal (vitamin A) and its derivatives efficiently interacting with this protein was studied.

Abstract:Organophosphorus pesticides are highly toxic phosphate compounds with the general structure of O=P(OR)3 and threaten human health seriously. Methyl parathion hydrolase from microbial is an important enzyme to degrade organophosphorus pesticides (OPs) into less toxic or nontoxic compounds likep-nitrophenol and diethyl phosphate. Here, a gene encoding methyl parathion hydrolase from Azohydromonas australica was firstly cloned and expressed in Escherichia coli. The recombinant hydrolase showed its optimal pH and temperature at pH 9.5 and 50 °C. Leveraging 1 mM Mn2+, the enzyme activity was significantly enhanced by 29.3-fold, and the thermostability at 40 and 50 °C was also improved. The recombinant MPH showed the specific activity of 4.94 and 16.0 U/mg towards methyl parathion and paraoxon, respectively. Moreover, A. australica MPH could effectively degrade various of OPs pesticides including methyl parathion, paraoxon, dichlorvos and chlorpyrifos in a few minutes, suggesting a great potential in the bioremediation of OPs pesticides.

Abstract:Di-n-butyl phthalate (DBP) is one of the most extensively used phthalic acid esters (PAEs) and is considered to be an emerging, globally concerning pollutant. The genus Streptomyces holds promise as a degrader of various organic pollutants, but PAE biodegradation mechanisms by Streptomyces species remain unsolved. In this study, a novel PAE-degrading Streptomyces sp. FZ201 isolated from natural habitats efficiently degraded various PAEs. FZ201 had strong resilience against DBP and exhibited immediate degradation, with kinetics adhering to a first-order model. The comprehensive biodegradation of DBP involves de-esterification, β -oxidation, trans-esterification, and aromatic ring cleavage. FZ201 contains numerous catabolic genes that potentially facilitate PAE biodegradation. The DBP metabolic pathway was reconstructed by genome annotation and intermediate identification. Streptomyces species have an open pangenome with substantial genome expansion events during the evolutionary process, enabling extensive genetic diversity and highly plastic genomes within the Streptomyces genus. FZ201 had a diverse array of highly expressed genes associated with the degradation of PAEs, potentially contributing significantly to its adaptive advantage and efficiency of PAE degradation. Thus, FZ201 is a promising candidate for remediating highly PAE-contaminated environments. These findings enhance our preliminary understanding of the molecular mechanisms employed by Streptomyces for the removal of PAEs.
