Abstract:<span>The organophosphorus-based OPs) nerve agents and pesticides have been applied in the agriculture industry for a long time. However, they were found to have a persistent effect on the environment and threaten human health. Traditional methods, including incineration and landfilling, could not thoroughly remove these organophosphorus compounds (OPs). Meanwhile, chemical hydrolysis for decontamination was also inhibited due to the presence of corrosive materials and high costs. Biological remediation for OPs employing microorganisms and organophosphorus-degrading enzymes is promising due to a mild and controllable procedure, environmental-friendly reactions, and high efficacy. A wide variety of enzymes have shown latent ability in degrading OPs hazards like organophosphorus hydrolase (OPH), organophosphorus acid anhydrolase (OPAA), the diisopropylfluorophosphatase (DFPase), and mammalian paraoxonase 1 (PON 1). To this end, increasing efforts have been made on these intriguing enzymes to increase their expression level, enhance the catalytic activity, modify the optimal substrate, and expand the practical application. In this review, the enzyme resource, crystal structure, molecular modification, and industry application were compared and discussed in detail. Moreover, the proposed ideas and positive results could be useful for the other relevant OPs-degrading enzymes.</span>

Development of a multimetal-based phosphotriesterase hybrid nanoflowers for decontamination of environmental organophosphorus compounds pollution

Abstract:The biodegradation of organophosphorus compounds (OPs) by phosphotriesterase (PTE) is an efficient and ecofriendly way. However, the low catalytic activity and instability of PTE becomes the key limiting step for the practical application. In this study, a new multimetal-based organic-inorganic self-assembly hybrid system was proposed to ameliorate this situation. A series of characterization methods demonstrated that the synthesized multimetallic PTE hybrid complex (Co/MnHF@PTE) exhibited a hierarchical flower-like structure. The activity recovery and the effective encapsulation yield of the Co/MnHF@PTE was 408% and 84.5%, respectively. The kcat/Km of Co/MnHF@PTE was separately 2.9-, 1.3-, and 2.7-fold higher than MnHF@PTE, CoHF@PTE, and free PTE. The hybrid nanoflowers showed preferable durability, stability, and reusability. When Co/MnHF@PTE was applied in a pump-flow reactor, the degradation percent of methyl parathion could be maintained at more than 93% with the injection of 24 ml of reaction mixture. More importantly, the Co/MnHF@PTE can effectively break P-F or P-S bond of soman (GD) and nerve agent VX to produce non-toxic products through hydroxyl nucleophilic attack. This study provides a feasible method to improve the activity and stability of PTE, promoting the biodegradation of OPs in the environment.

Abstract:Organophosphorus compounds (OPs) are widely used as pesticides and chemical warfare agents (CWAs). These synthetic compounds reside in the environment and cause serious pollution problems. Organophosphorus compounds can be detoxified rapidly by hydrolysis on exposure to environment, which are liable to be influenced by abiotic and biotic factors. Microorganisms isolated from different niches are the predominant entities in the biosphere with an exceptional ability to metabolize various OPs for their growth. The focus of this work is organophosphorus hydrolase (OPH, E.C. 8.1.3.1), which catalyzes the hydrolysis of many organophosphorus compounds and greatly reduces the toxicity and even can completely mineralize them. Several OPH enzymes, including o-phenylenediamine dihydrochloride (OPD), methyl parathion hydrolase (MPH) mevalonate pyrophosphate decarboxylase (MPD) etc, have been identified which accomplish hydrolysis for specific classes of organophosphorus compounds. The functional gene encoding OPH protein have been cloned, expressed and purified in both prokaryotic and eukaryocyte expression vector. To increase the enzyme activity or enhance their broad-spectrum property, the wild activated OPH was used to modify activated sites by the chemical modification of specific amino acid residues with the use of appropriately designed coenzyme analogs. The applications of the functional strains and OPH enzymes in bioremediation of OPs pollutants included: (i) Bioremediation: the technologies can be generally classified as in situ or ex situ; (ii) immobilization: an immobilized enzyme is the OPH that is attached to an inert, insoluble material such as calcium alginate and agar; immobilized whole cell, the target cell is the functional strains that is capable of degrading special OPs; (iii) construction of genetically engineering bacteria. A versatile genetically engineering bacterium that gained more functional genes from different sources was constructed by gene engineering and enzyme engineering, showing a creative and promising application in OPs detoxification. Abstract:Mining of organophosphorous (OPs)-degrading bacterial enzymes in collections of known bacterial strains and in natural biotopes are important research fields that lead to the isolation of novel OP-degrading enzymes. Then, implementation of strategies and methods of protein engineering and nanobiotechnology allow large-scale production of enzymes, displaying improved catalytic properties for medical uses and protection of the environment. For medical applications, the enzyme formulations must be stable in the bloodstream and upon storage and not susceptible to induce iatrogenic effects. This, in particular, includes the nanoencapsulation of bioscavengers of bacterial origin. In the application field of bioremediation, these enzymes play a crucial role in environmental cleanup by initiating the degradation of OPs, such as pesticides, in contaminated environments. In microbial cell configuration, these enzymes can break down chemical bonds of OPs and usually convert them into less toxic metabolites through a biotransformation process or contribute to their complete mineralization. In their purified state, they exhibit higher pollutant degradation efficiencies and the ability to operate under different environmental conditions. Thus, this review provides a clear overview of the current knowledge about applications of OP-reacting enzymes. It presents research works focusing on the use of these enzymes in various bioremediation strategies to mitigate environmental pollution and in medicine as alternative therapeutic means against OP poisoning.

Abstract:In the past decades, the organophosphorus compounds had been widely used in the environment and food industries as pesticides. Owing to the life-threatening and long-lasting problems of organophosphorus insecticide (OPs), an effective detection and removal of OPs have garnered growing attention both in the scientific and practical fields in recent years. Bacterial organophosphorus hydrolases (OPHs) have been extensively studied due to their high specific activity against OPs. OPH could efficiently hydrolyze a broad range of substrates both including the OP pesticides and some nerve agents, suggesting a great potential for the remediation of OPs. Abstract:Organophosphorus compounds (OP) are highly toxic pesticides and nerve agents widely used in agriculture and chemical warfare. The extensive use of these chemicals has severe environmental implications, such as contamination of soil, water bodies, and food chains, thus endangering ecosystems and biodiversity. Plants absorb pesticide residues, which then enter the food chain and accumulate in the body fat of both humans and animals. Numerous human cases of OP poisoning have been linked to both acute and long-term exposure to these toxic OP compounds. These compounds inhibit the action of the acetylcholinesterase enzyme (AChE) by phosphorylation, which prevents the breakdown of acetylcholine (ACh) neurotransmitter into choline and acetate. Thus, it becomes vital to cleanse the environment from these chemicals utilizing various physical, chemical, and biological methods. Biological methods encompassing bioremediation using immobilized microbes and enzymes have emerged as environment-friendly and cost-effective approaches for pesticide removal. Cell/enzyme immobilized systems offer higher stability, reusability, and ease of product recovery, making them ideal tools for OP bioremediation. Interestingly, enzymatic bioscavengers (stoichiometric, pseudo-catalytic, and catalytic) play a vital role in detoxifying pesticides from the human body. Catalytic bioscavenging enzymes such as Organophosphate Hydrolase, Organophosphorus acid anhydrolase, and Paraoxonase 1 show high degradation efficiency within the animal body as well as in the environment. Moreover, these enzymes can also be employed to decontaminate pesticides from food, ensuring food safety and thus minimizing human exposure. This review aims to provide insights to potential collaborators in research organizations, government bodies, and industries to bring advancements in the field of bioremediation and bioscavenging technologies for the mitigation of OP-induced health hazards.

Abstract:Organophosphates (OPs) compounds are universally used as pesticides and maintained as chemical warfare agents by many nations across the globe. These OPs compounds due to their molecular structure are highly persistent in nature, contaminating soil and water equally, thereby adversely affecting terrestrial and aquatic life, and contributing to millions of poisoning cases every year worldwide. Therefore, there are urgent requirements for safe and rapid method for environmental restoration and therapeutic detoxications. Organophosphate hydrolyzing enzymes are emerging as an attractive candidate for the degradation of OPs compounds. The biologically driven approach is safe, rapid, and environment-friendly. As genetically modified microbes are not in practice worldwide, scientists are exploring different bioremediation approaches that mainly focus on cell-free biocatalytic systems. In this review, we have discussed the prevalence of OPs hydrolyzing enzymatic systems and the recent advancement of enzyme engineering in enhancing the catalytic activity, substrate specificity, and half-life. It highlights the application in OPs detection, decontamination (environmental bioremediation), and therapeutic detoxification using approaches like immobilization. We have also described the remaining challenges and future prospects.

Abstract:Organophosphate esters (OPEs) are widely used commercial additives, but their environmental persistence and toxicity raise serious concerns necessitating associated remediation strategies. Although there are various existing technologies for OPE removal, comprehensive screening for them is urgently needed to guide further research. This review provides a comprehensive overview of the techniques used to remove OPEs from soil and water, including their related influencing factors, removal mechanisms/degradation pathways, and practical applications. Based on an analysis of the latest literature, we concluded that (1) methods used to decontaminate OPEs include adsorption, hydrolysis, photolysis, advanced oxidation processes (AOPs), activated sludge processes, and microbial degradation; (2) factors such as the quantity/characteristics of the catalysts/additives, pH value, inorganic ion concentration, and natural organic matter (NOM) affect OPE removal; (3) primary degradation mechanisms involve oxidation induced by reactive oxygen species (ROS) (including •OH and SO4•-) and degradation pathways include hydrolysis, hydroxylation, oxidation, dechlorination, and dealkylation; (5) interference from the pH value, inorganic ion and the presence of NOM may limit complete mineralization during the treatment, impacting practical application of OPE removal techniques. This review provides guidance on existing and potential OPE removal methods, providing a theoretical basis and innovative ideas for developing more efficient and environmentally friendly techniques to treat OPEs in soil and water.

Abstract:Background: Organophosphorus compounds, widely used in agriculture and industry, pose a serious threat to human health due to their acute neurotoxicity. Although traditional interventions for organophosphate poisoning are effective, they often come with significant side effects. Objective: This paper aims to evaluate the potential of enzymes within biological organisms as organophosphorus bioclearing agents. It analyses the technical challenges in current enzyme research, such as substrate specificity, stereoselectivity, and immunogenicity, while exploring recent advancements in the field. Methods: A comprehensive review of literature related to detoxifying enzymes or proteins was conducted. Existing studies on organophosphorus bioclearing agents were summarised, elucidating the biological detoxification mechanisms, with a particular focus on advancements in protein engineering and novel delivery methods. Results: Current bioclearing agents can be categorised into stoichiometric and catalytic bioclearing agents, both of which have shown some success in preventing organophosphate poisoning. Technological advancements have significantly improved various properties of bioclearing agents, yet challenges remain, particularly in substrate specificity, stereoselectivity, and immunogenicity. Future research will focus on expanding the substrate spectrum, enhancing catalytic efficiency, prolonging in vivo half-life, and developing convenient administration methods. Conclusion: With the progression of clinical trials, bioclearing agents are expected to become widely used as a new generation of therapeutic organophosphate detoxifiers.
Microbial degradation of organophosphorus pesticides: novel degraders, kinetics, functional genes, and genotoxicity assessment

Abstract:Farmland soil sprayed with organophosphorus pesticides (OPs) annually was investigated for the identification and characterization of OP-degrading microorganisms. Six bacterial strains were identified, including Brevundimonas faecalis MA-B12 and Alcaligenes faecalis subsp. parafaecalis MA-B13 for methamidophos degradation, Citrobacter freundii TF-B21 and Ochrobactrum intermedium TF-B23 for trichlorfon degradation, Ochrobactrum intermedium DV-B31 for dichlorvos degradation, and Bacillus cereus for dimethoate degradation. The optimal biodegradation conditions for OPs were obtained at pH 7.0 and incubation temperature ranging from 28 to 37 °C. In an 8-day batch test, biodegradation of the four OPs all followed first-order kinetics, with biodegradation rates ranging from 58.08 to 96.42%. Functional genes responsible for OPs degradation were obtained, including ophB, ampA, opdE, opd, opdA, and mpd. As these strains were indigenous strains isolated from farmland soils, they can be potentially used as bacterial consortium for the bioremediation of mixed OP-contaminated soils. A time-course genotoxicity assessment of the degradation products was done by a bacterial whole-cell bioreporter, revealing that biodegradation of trichlorfon, dichlorvos, and dimethoate resulted a decreased genotoxicity within 5 days, which, however, significantly increased on day 8. The result demonstrated that more toxic products may be produced during the biodegradation processes of OPs, and more attention should be put not only on the pesticides themselves, but also on the toxic effects of their degradation products. To the best of our knowledge, this is for the first time that the genotoxicity of OP degradation products was evaluated by the bioreporter assay, broadening our understanding on the genotoxic risks of OPs during biodegradation process.Graphical Abstract

A Novel Organophosphorus Acid Anhydrolase from Deep Sea Sediment with High Degradation Efficiency for Organophosphorus Pesticides and Nerve Agent

Abstract:Organophosphorus compounds (OPCs), including highly toxic nerve agents and pesticides, have been used widely in agricultural and military applications. However, they have aroused widespread concern because they persistently pollute the environment and threaten human life. Organophosphorus acid anhydrolase (OPAA) is a promising enzyme that can detoxify OPCs. Here, a novel OPAA (OPAA114644) was isolated and characterized from deep-sea sediment (−3104 m). It exhibited excellent alkaline stability, and the loss of activity was less than 20% in the pH range 5.0–9.0, even after being incubated for 30 d at 4 °C. It also exhibited high salt tolerance, and its enzymatic activity increased by approximately fourfold in the presence of 20% NaCl (w/v). Additionally, OPAA114644 exhibited high degradation efficiency for soman, dichlorvos, paraoxon, coumaphos, and chlorpyrifos with a concentration of up to 250 mg/L, with the degradation rate being 100%, 100%, 100%, 80% and 51%, respectively, in 20 min under optimal conditions. Notably, OPAA114644 dissolved in different solutions, such as 20% NaCl, 1 mM SDS, 0.05% soap, 10% methanol, and tap water, could efficiently decontaminate the residual paraoxon on the surfaces of glasses, cotton tissues, and apples. These results indicate that OPAA114644 has excellent potential for the biodegradation and bioremediation of OPCs pollution and represents a real application of OPAA in the decontamination and detoxification of foods and clothes, and in the remediation of sites such as floors. Deep-sea sediment might also be an abundant resource for various functional microorganisms and enzymes.

Abstract:Organophosphorus pesticides (OPPs) are one of the most widely used types of pesticide that play an important role in the production process due to their effects on preventing pathogen infection and increasing yield. However, in the early development and application of OPPs, their toxicological effects and the issue of environmental pollution were not considered. With the long-term overuse of OPPs, their hazards to the ecological environment (including soil and water) and animal health have attracted increasing attention. Therefore, this review first clarified the classification, characteristics, applications of various OPPs, and the government's restriction requirements on various OPPs. Second, the toxicological effects and metabolic mechanisms of OPPs and their metabolites were introduced in organisms. Finally, the existing methods of degrading OPPs were summarized, and the challenges and further addressing strategy of OPPs in the sustainable development of agriculture, the environment, and ecology were prospected. However, methods to solve the environmental and ecological problems caused by OPPs from the three aspects of use source, use process, and degradation methods were proposed, which provided a theoretical basis for addressing the stability of the ecological environment and improving the structure of the pesticide industry in the future.

Abstract:Organophosphates (OPs) are an integral part of modern agriculture; however, due to overexploitation, OPs pesticides residues are leaching and accumulating in the soil, and groundwater contaminated terrestrial and aquatic food webs. Acute exposure to OPs could produce toxicity in insects, plants, animals, and humans. OPs are known for covalent inhibition of acetylcholinesterase enzyme in pests and terrestrial/aquatic organisms, leading to nervous, respiratory, reproductive, and hepatic abnormalities. OPs pesticides also disrupt the growth-promoting machinery in plants by inhibiting key enzymes, permeability, and trans-cuticular diffusion, which is crucial for plant growth. Excessive use of OPs, directly/indirectly affecting human/environmental health, raise a thoughtful global concern. Developing a safe, reliable, economical, and eco-friendly methods for removing OPs pesticides from the environment is thus necessary. Bioremediation techniques coupled with microbes or microbial-biocatalysts are emerging as promising antidotes for OPs pesticides. Here, we comprehensively review the current scenario of OPs pollution, their toxicity (at a molecular level), and the recent advancements in biotechnology (modified biocatalytic systems) for detection, decontamination, and bioremediation of OP-pesticides in polluted environments. Furthermore, the review focuses on onsite applications of OPs degrading enzymes (immobilizations/biosensors/others), and it also highlights remaining challenges with future approaches.

Abstract:Organophosphorus adulteration in the environment creates terrestrial and aquatic pollution. It causes acute and subacute toxicity in plants, humans, insects, and animals. Due to the excessive use of organophosphorus pesticides, there is a need to develop environmentally friendly, economical, and bio-based strategies. The microbiomes, that exist in the soil, can reduce the devastating effects of organophosphates. The use of cell-free enzymes and yeast is also an advanced method for the degradation of organophosphates. Plant-friendly bacterial strains, that exist in the soil, can help to degrade these contaminants by oxidation-reduction reactions, enzymatic breakdown, and adsorption. The bacterial strains mostly from the genus Bacillus, Pseudomonas, Acinetobacter, Agrobacterium, and Rhizobium have the ability to hydrolyze the bonds of organophosphate compounds like profenofos, quinalphos, malathion, methyl-parathion, and chlorpyrifos. The native bacterial strains also promote the growth abilities of plants and help in detoxification of organophosphate residues. This bioremediation technique is easy to use, relatively cost-effective, very efficient, and ensures the safety of the environment. This review covers the literature gap by describing the major effects of organophosphates on the ecosystem and their bioremediation by using native bacterial strains.

Azinophos-methyl, chlorpyrifos, diazinon, fonofos and disulfoton are well-known organophosphate pesticides (OPs) having extensive applications in agriculture, horticulture, pest control, plastic making, flame retardants and for several household applications. OPs are the ester forms of phosphoric acid, usually considered as safe for agriculture use due to their relatively fast degradation rates. Acute or chronic exposure to OPs can produce varying levels of toxicity in humans, animals, plants, and insects. These are known to inhibit acetylcholinesterase activity, not only in insects but also in aquatic and terrestrial organisms leading to respiratory, reproductive, nervous, hepatic and renal abnormalities. OPs disrupt the growth promoting mechanism by inhibiting various enzymes, transcuticular diffusion and permeability which is essential for the growth of plants. Regular use of OPs subside the microbial community and reduces soil fertility. Due to environmental concerns associated with the accumulation of OPs in food products and water supplies, there is an urgent need to develop rapid, reliable and economical method amenable to onsite applications. Here, we review different classes of organophosphate pesticides, their environmental issues, analytical techniques for estimation, and eco-friendly biodegradation approaches for its efficient bioremediation.

Abstract:BACKGROUND Raised serum uric acid (SUA) levels are associated with conditions at high risk for coronary heart disease. SUA levels can be partly modified by the diet, but little is known about the association of SUA with several dietary factors, particularly the quality of ingested lipids. METHODS The associations between SUA levels and biological and dietary factors were investigated in a sample of 90 healthy subjects (45 males and 45 females; ages 55.6 +/- 18 years) living in Nove, a village near Vicenza (Veneto Region, northern Italy). A careful selection of subjects to exclude those with chronic or acute disease (influencing SUA directly or indirectly through their pharmacological treatments) was obtained with the collaboration of three general practitioners operating in Nove. RESULTS SUA was higher among men than among women and higher among postmenopausal women than among women of child-bearing age (P < 0.01). Stepwise multiple linear regression analysis of the population as a whole showed that serum triglycerides and total RBC omega-6 PUFA were significant and independent predictors of a consistent proportion of serum uric acid variability (R = 0.59; R2 = 0.332; P < 0.001). Taking men and postmenopausal women together, the result was the same. CONCLUSIONS Triglycerides and SUA are related even in the normal population and not only in "Syndrome X." Dietary enrichment with omega-6 PUFA might be useful for the negative modulation of SUA.

Abstract:Complex rhizoremediation is the main mechanism of phytoremediation in organic-contaminated soil. Low molecular weight organic acids (LMWOAs) in root exudates have been shown to increase the bioavailability of contaminants and are essential for promoting the dissipation of contaminants. The effects of root exudates on the dissipation of organophosphate esters (OPEs) in soil are unclear. Consequently, we studied the combined effects of root exudates, soil enzymes and microorganisms on OPEs (tri (1-chloro-2-propyl) phosphate (TCPP) and triphenyl phosphate (TPP)) dissipation through pot experiments. Oxalic acid (OA) was confirmed to be the main component of LMWOAs in root exudates of ryegrass. The existence of OA increased the dissipation rate of OPEs by 6.04%-25.50%. Catalase and dehydrogenase activities were firstly activated and then inhibited in soil. While, urease activity was activated and alkaline phosphatase activity was inhibited during the exposure period. More bacteria enrichment (e.g., Sphingomonas, Pseudomonas, Flavisolibacter, Pontibacter, Methylophilus and Massilia) improved the biodegradation of OPEs. In addition, the transformation paths of OPEs hydrolysis and methylation under the action of root exudates were observed. This study provided theoretical insights into reducing the pollution risk of OPEs in the soil.

Abstract:Phosphotriesterase (PTE) biodegradation of organophosphorus pesticides (OPs) is an efficient and environmentally friendly method. However, the instability and nonreusability of free PTE become the key factors restricting its practical application. In this study, a novel cross-linked magnetic hybrid nanoflower (CLMNF) was prepared. Molecular dynamics (MD) simulations were performed to further investigate the enhanced catalytic efficiency of the enzymes. The recovery rate of enzyme activity was 298% due to the large specific surface area and metal ion activation effect. More importantly, the immobilization scheme greatly improved the stability and reuse performance of the catalyst and simplified the recovery operation. CLMNFs retained 90.32% relative activity after 5 consecutive cycles and maintained 84.8% relative activity after 30 days at 25 °C. It has a good practical application prospect in the degradation and detection of OPs. Consequently, the immobilized enzyme as a biocatalyst has the characteristics of high efficiency, stability, safety, and easy separation, establishing the key step in a biodetoxification system to control organophosphorus contamination in food and the environment.

Abstract:The excessive or illegal use of organophosphorus pesticides (OPs) in agriculture has led to potential residue problems, making it essential to develop an effective analytical method. As reliable and rapid analytical methods, biosensors open an avenue for OP detection. However, traditional biosensors mainly depend on bio-recognition elements, consequently limiting their practical application. To address these issues, nanozymes have been widely applied because of their low cost, excellent stability, and favorable catalytic activity. This review summarizes the related research progress of nanozyme-based biosensors for OP detection in the last six years (2018–2023). The structural classification, catalytic activity, and biosensing strategy are discussed. Emphatically, this paper introduces the practical applications of nanozyme-enabled biosensors in the direction of simple operation, low cost, and on-site detection, such as optical and electrochemical biosensors. Finally, the main challenges and future trends are prospected, which could provide theoretical references for nanozyme-based biosensors in OP detection.
