Abstract:Electrochemiluminesence (ECL) of luminol has been enhanced through the catalysis of various artificial nanomaterials to improve its performance in sensing applications. Herein, we report on a biofunctional-catalytic approach based on a simple organic radical, 2,2,6,6-tetramethylpiperidine-1-oxyl (TEMPO), which enhances significantly the ECL emission for both luminol/O 2 and luminol/H 2 O 2 systems. The electrochemical studies showed that the oxidation of TEMPO could produce highly stable oxoammonium cation (TEMPO + ) that has little interaction with dissolved O 2 , but was found to improve the oxidation of luminol. On the other hand, TEMPO as an electrocatalyst could make the oxidation of H 2 O 2 more efficient. This bifunctional-catalysis of TEMPO has facilitated the production of L −• in luminol/dissolved O 2 system and O 2 −• in luminol/H 2 O 2 system for ECL generation. Based on the enhanced ECL of luminol/TEMPO/cholesterol oxidase (ChOx)/cholesterol system, an ECL sensor has been designed for the selective and sensitive detection of cholesterol with a linear range between 0.30 μM and 100 μM and a low detection limit of 0.08 μM. Finally, the present work opens new avenues for studying luminol ECL using the bifunctional-catalysis of nitroxyl radicals.

Abstract:Luminol-based electrochemiluminescence (ECL) can be readily excited by various reactive oxygen species (ROS) electrogenerated with an oxygen reduction reaction (ORR). However, the multiple active intermediates involved in the ORR catalyzed with complex nanomaterials lead to recognizing the role of ROS still elusive. Moreover, suffering from the absence of the direct electrochemical oxidation of luminol at the cathode and poor transformation efficiency of O2 to ROS, the weak cathodic ECL emission of luminol is often neglected. Herein, owing to the tunable coordination environment and structure-dependent catalytic feature, single-atom catalysts (SACs) are employed to uncover the relationship between the intrinsic ORR activity and ECL behavior. Interestingly, the traditionally negligible cathodic ECL of luminol is first boosted (ca. 70-fold) owing to the combination of electrochemical ORR catalyzed via SACs and chemical oxidation of luminol. The boosted cathodic ECL emission exhibits electron-transfer pathway-dependent response by adjusting the surrounding environment of the center metal atoms in a controlled way to selectively produce different active intermediates. This work bridges the relationship between ORR performance and ECL behavior, which will guide the development of an amplified sensing platform through rational tailoring of the ORR activity of SACs and potential-resolved ECL assays based on the high-efficiency cathodic ECL reported.

Abstract:Cathodic electrochemiluminescence (ECL) of luminol (or its analogues)-dissolved oxygen (O2) system is an ideal alternative to the ECL of traditional luminol-hydrogen peroxide (H2O2) system, which can efficiently avoid the self-decomposition of H2O2 at room temperature. However, the mechanism for the generation of cathodic ECL by luminol (or its analogues)-O2 system is still ambiguous. Herein, we report the study of cathodic ECL generation by the L012-O2 system at a glassy carbon electrode (GCE). The types of reactive oxygen species (ROS) involved generated during ECL reactions were verified. A possible reaction mechanism for the system was proposed and the rate constants of related reactions were estimated. Furthermore, several intermediates of L012 involved in the proposed pathways were validated by electrochemistry coupled mass spectrometry. Finally, the cathodic ECL system was successfully used for measuring antioxidant capacity of commercial juice with Trolox as a standard.

Abstract:Electrochemiluminescence (ECL) of luminol is a luminescence process that proceeds in the presence of reactive oxygen species (e.g. hydrogen peroxide (H2O2)) at a suitable electrode potential, the reaction mechanism of which is complicated and remains ambiguous. In this work, we report a visualization approach for measuring the thickness of the ECL layer (TEL) of the luminol/H2O2 system to decipher the reaction process by combined use of the microtube electrode, ECL microscopy, and finite element simulations. With the increase of solution pH, the ECL image captured with the microtube electrode tends to vary from spot to ring, corresponding to the decrease of TEL from >9.1 μm to ca. 4.3 μm. We propose that different intermediates are involved in the course of ECL reaction. At a low pH (e.g. pH < 9), a relatively large TEL is most likely determined by the diffusion of oxidized and deprotonated luminol intermediate that is neutral and has a long lifetime. While at a high pH (e.g. pH in the range of 10 to 12), the ECL reaction is controlled by short-lived radical intermediates of both luminol and superoxide anion. The proposed mechanism is proved theoretically by finite element simulations and experimentally by the apparent effect of concentration ratio of luminol/H2O2.

Abstract:A variety of luminol derivatives with N- and O-substitution have been synthesized with broad functional group tolerance. O-esterification has been demonstrated for the first time as a promising way to prepare enhanced CL reagents for sensing hemin, bloodstain and horseradish peroxidase (HRP). The most effective analogue with a deuterated acetyl group exhibited greater potential than luminol for bloodstain imaging and HRP imaging in western blotting (WB). In addition, O-etherification can greatly suppress CL signal that has been applied to design a CL probe for β-glucosidase (β-Glu). This study offers important and useful information regarding the luminol modification and shows great potential to use O-substituted analogues for enhanced CL analysis.

Abstract:Construction the insitu, real-time monitoring methodology for chemical changes, especially in the mechanism of oxidation, has long been regarded as a significant and challenging concern. Herein, we designed and synthesized a TEMPO-mediated alkynylplatinum(II) terpyridine complex with switchable activity in photoluminescence and EPR (Electron Paramagnetic Resonance) spectra. This complex of alkynylplatinum(II) terpyridine-TEMPO [(2,2,6,6-tetramethylpiperidin-1-yl)oxyl] is paramagnetic, while the insitu formed alkynylplatinum(II) terpyridine-TEMPOH is diamagnetic, so that concentration changes of TEMPO/TEMPOH species were obtained successfully by photoluminescence and EPR spectra in an operando way. As a model example, we investigated the mechanism of alcohol oxidation mediated by TEMPO/ bis(acetoxy)iodo]benzene (BAIB). It was found that, with the consumption of co-oxidant (BAIB), the concentration and conformation of catalyst (TEMPO) were changed gradually, and the vital role of catalyst and co-oxidant in the benzyl alcohol oxidation was clearly showed by photo-luminescence and EPR spectroscopies. On the basis of these observations, we suggested an effective methodology to study the mechanism of chemical reaction, which is promising for further practical applications in analytical or environmental issues.

Abstract:Electrochemiluminescence (ECL) comprises a sophisticated cascade of reactions. Despite advances in mechanistic studies by electrochemistry and spectroscopy, a lack of access to dynamic molecular information renders many plausible ECL pathways unclear or unproven. Here we describe the construction of a real-time ECL mass spectrometry (MS) platform (RT-Triplex) for synchronization of dynamic electrical, luminescent, and mass spectrometric outputs during ECL events. This platform allows immediate and continuous sampling of newly born species at the Pt wire electrode of a capillary electrochemical (EC) microreactor into MS, enabling characterization of short-lived intermediates and the multi-step EC processes. Two ECL pathways of luminol are validated by observing the key intermediates alpha-hydroxy hydroperoxide and diazaquinone and unraveling their correlation with applied voltage and ECL emission. Moreover, a "catalytic ECL route" of boron dipyrromethene (BODIPY) involving homogeneous oxidation of tri-n-propylamine with the BODIPY radical cation is proposed and verified.

Abstract:As a peroxidase mimic, cupric oxide nanoparticles were found to enhance the chemiluminescence (CL) of luminol-H(2)O(2) system up to 400 folds. The CL spectra and radical scavengers were conducted to investigate the possible CL enhancement mechanism. It was suggested that the enhanced CL could be attributed to the peroxidase-like activity of CuO nanoparticles, which effectively catalyzed the decomposition of hydrogen peroxide into hydroxyl radicals. The effects of the reactant concentrations and some organic compounds were also investigated. The proposed method could be used as a sensitive detection tool for hydrogen peroxide and glucose.

Abstract:This work used sulfite as a coreactant to enhance the anodic electrochemiluminescence (ECL) of mercaptopropionic acid modified CdTe quantum dots (QDs). This strategy proposed the first coreactant anodic ECL of QDs and led to a sensitive ECL emission of QDs in aqueous solution at relatively low potential. In the presence of dissolved oxygen, the stable ECL emission resulted from the excited QDs. Thus, an ECL detection method was proposed at +0.90 V (vs Ag/AgCl) based on the quenching of excited QDs by the analyte. Using tyrosine as a model compound, whose electrooxidized product could quench the excited QDs and thus the ECL emission, an analytical method for detection of tyrosine in a wide concentration range was developed. Furthermore, by combining an enzymatic cycle of trace tyrosinase to produce the oxidized product with an energy-transfer process, an extremely sensitive method for ECL detection of tyrosine with a subpicomolar limit of detection was developed. The sulfite-enhanced anodic ECL emission provided an alternative for traditional ECL light emitters and a new methodology for extremely sensitive ECL detection of mono- and dihydroxybenzenes at relatively low anodic potential. This strategy could be easily realized and opened new avenues for the applications of QDs in ECL biosensing.

Abstract:The luminol–dissolved O2 (DO) electrochemiluminescence (ECL) sensing system has recently gained growing interest; however, the drawback of the ultra-low ECL signal response greatly hinders its potential quantitative applications. In this work, for the first time, we explored the use of high entropy oxide (HEO) comprising five metal ingredients (Ni, Co, Cr, Cu, and Fe), to accelerate the reduction reaction of DO into reactive oxygen species (ROS) for boosting the ECL performance of the luminol–DO system. Benefiting from the existing abundant oxygen vacancies induced by the unique crystal structure of the HEO, DO could be efficiently converted into ROS, thus significantly boosting the performance of the corresponding ECL sensor (with an ∼240-fold signal enhancement in this study). Abstract:Luminol electrochemiluminescence (ECL) imaging was developed for the parallel measurement of active membrane cholesterol at single living cells, thus establishing a novel electrochemical detection technique for single cells with high analysis throughput and low detection limit. In our strategy, the luminescence generated from luminol and hydrogen peroxide upon the potential was recorded in one image so that hydrogen peroxide at the surface of multiple cells could be simultaneously analyzed. Compared with the classic microelectrode array for the parallel single-cell analysis, the plat electrode only was needed in our ECL imaging, avoiding the complexity of electrode fabrication. The optimized ECL imaging system showed that hydrogen peroxide as low as 10 μM was visible and the efflux of hydrogen peroxide from cells could be determined. Coupled with the reaction between active membrane cholesterol and cholesterol oxidase to generate hydrogen peroxide, active membrane cholesterol at cells on the electrode was analyzed at single-cell level. The luminescence intensity was correlated with the amount of active membrane cholesterol, validating our system for single-cell cholesterol analysis. The relative high standard deviation on the luminescence suggested high cellular heterogeneities on hydrogen peroxide efflux and active membrane cholesterol, which exhibited the significance of single-cell analysis. This success in ECL imaging for single-cell analysis opens a new field in the parallel measurement of surface molecules at single cells.

Abstract:Electrochemiluminescence (ECL) imaging, as an optical technology, has been developed at full tilt in the field of life science and nanomaterials. However, the relatively low ECL intensity or the high co-reactant concentration needed in the electrochemical reaction blocks its practical application. Here, we developed an ECL imaging system based on the rGO-TiO2-x composite material, where the co-reactant, reactive oxygen species (ROS), is generated in situ under the synergetic effect of of ultrasound (US) and electric irradiation. The rGO-TiO2-x composites facilitate the separation of electron (e-) and hole (h+) pairs and inhibit recombination triggered by external US irradiation due to the high electroconductivity of rGO and oxygen-deficient structures of TiO2, thus significantly boosting ROS generation. Furthermore, the increased defects on rGO accelerate the electron transfer rate, improving the electrocatalytic performance of the composite and forming more ROS. This high ultrasonic-electric synergistic efficacy is demonstrated through the enhancement of photon emission. Compared with the luminescence intensity triggered by US irradiation and electric field, an enhancement of ∼20-fold and 10-fold of the US combined with electric field-triggered emission is observed from this composite. Under the optimized conditions, using dopamine (DA) as a model target, the sensitivity of the US combined ECL strategy for detection of DA is two orders of magnitude higher than that of the ECL method. The successful detection of DA at low concentrations makes us believe that this strategy provides the possibility of applying ECL imaging for cellular single-molecule analysis and cancer therapy.
Self-Supply of H2O2 and O2 by Hydrolyzing CaO2 to Enhance the Electrochemiluminescence of Luminol Based on a Closed Bipolar Electrode.

Abstract:A novel luminol electrochemiluminescence (ECL) sensing platform was developed based on the closed indium tin oxide bipolar electrode (ITO BPE) for detecting the biomarkers of squamous cell carcinomas named cytokeratin 19 fragments (CYFRA 21-1). A strategy of in situ-generated coreactant H2O2 and O2 was proposed to enhance luminol ECL intensity. CaO2, possessing a high capacity of self-supplying H2O2 and O2, was encapsulated by ZIF-67, glucose oxidase, and horseradish peroxidase (GOD/HRP-loaded ZIF-67@CaO2). In the presence of glucose, gluconic acid and H2O2 were generated via the catalytic effect of GOD; then, gluconic acid induced the degradation of ZIF-67, leading to the hydrolysis of the unprotected CaO2 to produce both O2 and H2O2. Therefore, the generated coreactants O2 and H2O2, via the oxidation of glucose and the hydrolysis of CaO2, can effectively enhance the luminol ECL behavior. According to the luminol ECL reaction occurring on the anode surface of the ITO BPE, a sandwich immunosensor was fabricated on the anodic ITO BPE and GOD/HRP-loaded ZIF-67@CaO2 as labels incubated with secondary antibodies. Owing to the cathodic reaction rate influencing the anodic reaction rate, Au nanoparticles were electrodeposited on the cathode surface of the ITO BPE, which not only meliorated oxygen reduction but also further enhanced the ECL intensity of luminol. Under optimal experiments, an ECL immunosensor for detecting CYFRA 21-1 with a wider linear range from 0.0075 to 50 ng mL-1 and a detection limit of 1.89 pg mL-1 (S/N = 3) was obtained.

Abstract:Electrochemiluminescence (ECL) has emerged as a valuable tool for understanding multiphasic and compartmentalized systems, which have crucial wide-ranging applications across diverse fields. However, ECL reactions are limited to the vicinity of the electrode surface due to spatial constraints of electron transfer and the short lifetime of radical species, making ECL emission in bulk multiphasic solution challenging. To address this limitation, we propose a novel bipolar electrochemistry (BPE) approach for wireless dual-color ECL emission at the water/oil (w/o) interface. Firstly, amphiphilic glass carbon (GC) microbeads with distinct hydrophilic and hydrophobic regions are prepared by bipolar electrografting of hydrophobic trifluoromethyl diazonium salt, then the resulting Janus beads are positioned at the w/o interface. Subsequently, two model ECL systems containing luminol and H2O2 in the aqueous phase, and [Ru(bpy)3]2+ and benzoyl peroxide (BPO) in the organic phase, are selected based on their solubility to confine light-emitting reactions to their respective phases. Upon application of an electric field perpendicular to the interface, the Janus microbeads get polarized, triggering simultaneous oxidative blue ECL (425 nm) and reductive red ECL (620 nm) in the aqueous and organic phases, respectively. Taking advantage of ECL imaging, the potential gradient distribution on the GC microbead at the w/o interface is revealed, indicating a "pseudo-closed" bipolar system due to limited ion transfer between phases. We also investigate the effect of changing the electric field direction parallel to the interface, which alters the ECL emission area from a hemisphere to a quarter of the microbead's surface. This bipolar ECL approach at the w/o interface not only offers opportunities for imaging the aqueous phase and organic phase simultaneously, but also enables ECL imaging and light generation in the bulk solution, thus overcoming the usual spatial limitation requiring proximity to the electrode surface.

Abstract:In this work, electrogenerated chemiluminescence resonance energy transfer (ECL-RET) between luminol as a donor and CdSe@ZnS quantum dots (QDs) as an acceptor was reported in neutral conditions. It was observed that a glassy carbon electrode modified with CdSe@ZnS quantum dots (CdSe@ZnS/GCE) can catalyze the luminol oxidation to promote the anodic luminol ECL without coreactants. The intensity of anodic luminol ECL (0.60 V) at the CdSe@ZnS/GCE was enhanced more than 1 order of magnitude compared with that at the bare GCE. Another stronger anodic ECL peak observed at more positive potential (1.10 V) could be assigned to the ECL-RET between the excited state of luminol and the QDs. A label-free ECL aptasensor for the detection of thrombin was fabricated based on the synergic effect of the electrocatalysis and the ECL-RET. The approach showed high sensitivity, good selectivity, and wide linearity for the detection of thrombin in the range of 10 fM-100 pM with a detection limit of 1.4 fM (S/N = 3). The results suggested that the as-proposed luminol-QDs ECL biosensor will be promising in the detection of protein.

Abstract:Tris(bipyridine) ruthenium(II)-based luminophores have been well developed in the area of electrochemiluminescence, while their applications in chemiluminescence (CL) are rarely studied due to the poor luminous efficiency and complicated CL reaction. Herein, a novel tris(bipyridine) ruthenium(II)-based ternary CL system is proposed by introducing cobalt single atoms integrated with graphene-encapsulated cobalt nanoparticles (Co SAs/Co@C) and peroxymonosulfate (PMS) as advanced coreaction accelerator and promising coreactant, respectively. On the basis of the experimental results and density functional theory calculations, it is concluded that Co@C can synergistically modulate the adsorption behavior of PMS on Co SAs and then efficiently activate PMS to produce massive singlet oxygen for remarkable CL emission. Under the optimum conditions, the as-prepared CL biosensor exhibits a good linear range, excellent sensitivity, and selectivity, holding great potential for the practical detection of prostate-specific antigen in human serum.

Ratiometric electrochemiluminescent strategy regulated by electrocatalysis of palladium nanocluster for immunosensing.

Abstract:This work designed a novel ratiometric electrochemiluminescence (ECL) immunosensing approach based on two different ECL emitters: CdS quantum dots (QDs) as cathodic emitter and luminol as anodic emitter. The ECL immunosensor was constructed by a layer-by-layer modification of CdS QDs, Au nanoparticles and capture antibody on a glassy carbon electrode. With hydrogen peroxide as ECL coreactant, the immunosensor showed a cathodic ECL emission of CdS QDs at -1.5 V (vs Ag/AgCl) in air-saturated pH 8.0 buffer. Upon the formation of sandwich immunoassay, the lumiol/palladium nanoclusters (Pd NCs)@graphene oxide probe was introduced to the electrode. Therefore, the cathodic ECL intensity decreased and luminol anodic ECL emission was appeared at +0.3 V (vs Ag/AgCl) owing to the competition of the coreactant of hydrogen peroxide. Using carcino-embryonic antigen as model, this ratiometric ECL strategy could be used for immunoassay with a linear range of 1.0-100 pg mL(-1) and a detection limit of 0.62 pg mL(-1). The enhanced ratiometric ECL signal resulted from the high density and excellent electrocatalysis of the loaded Pd NCs. The immunosensor exhibited good stability and acceptable fabrication reproducibility and accuracy, showing a great promising for clinical application. This electrocatalysis-regulated ratiometric ECL provides a new concept for ECL measurement, and could be conveniently extended for detection of other protein biomarkers. (C) 2015 Elsevier B.V. Abstract:In this work, a novel electrochemical biosensor based on nitronyl nitroxide monoradical 2,2,6,6-tetramethylpiperidine 1-Oxyl (TEMPO) as new electrochemical label for facile nucleic acids detection is developed. This fast and convenient functional microelectrode was designed by fixing the capture probe peptide nucleic acid (PNA) and using the coordination interaction of Zr4+ with both phosphate groups and carboxyl groups. Differential pulse voltammetry (DPV) was used to study the oxidation current of TEMPO which was combined with the electrode surface and labeled. TEMPO electrochemical signal related to target deoxyribonucleic acid (tDNA) concentration was finally detected when tDNA was added on the surface of glassy carbon electrode (GCE). The detection principle, optimization of key factors and performance analysis of the biosensor are also discussed. A great linear relation is acquired within the scope of 10 pM to 100 nM under optimal conditions and the detection limit of this experiment is calculated as low as 2.57 pM (R2 = 0.996). In addition, complex serum samples were used to explore the practical application of this experiment. The results show the developed electrochemical DNA biosensor has wide application prospects in nucleic acids detection and clinical analysis.
