Abstract:Imaging three-dimensional, subcellular structures with high axial resolution has always been the core purpose of fluorescence microscopy. However, trade-offs exist between axial resolution and other important technical indicators, such as temporal resolution, optical power density, and imaging process complexity. We report a new imaging modality, fluorescence interference structured illumination microscopy（FI-SIM）, which is based on three-dimensional structured illumination microscopy for wide-field lateral imaging and fluorescence interference for axial reconstruction. FI-SIM can acquire images quickly within the order of hundreds of milliseconds and exhibit even 30 nm axial resolution in half the wavelength depth range without z-axis scanning. Moreover, the relatively low laser power density relaxes the requirements for dyes and enables a wide range of applications for observing fixed and live subcellular structures.

Abstract:The effects of different lasers on biological tissues are various. With short duration time, great instantaneous power and small focus dimension, the femtosecond laser pulses have expansive application future on the ultra-fast, the ultra-intense and the ultra-fine fields. And the study on tiny cell's dynamics, such as mitosis, metabolism, and apoptosis, is of great importance to understand the biological and developmental properties of cells. Meanwhile the biological big molecules and the water do not absorb the light in the near-infrared region, so it is considered to do experiments on the biological cells with the near-infrared femtosecond laser pulses. This laser technique has been applied to dissect and ablate intracellular structures. Some applications of this technology on cells, such as nanosurgery, gene transfection and chromosomal dissection are introduced. It has higher precision than the traditional technologies, and can be used without compromising the cell's viability.

Abstract:Single cells are increasingly recognized to be capable of wound repair that is important for our mechanistic understanding of cell biology. The lack of flexible, facile, and friendly subcellular treatment methods has hindered single-cell wound repair studies and organelle transport analyses. Here we report a laminar flow based approach, we call it fluid cell knife (Fluid CK), that is capable of precisely cutting off or treating a portion of a single cell from its remaining portion in its original adherent state. Local operations on portions of a living single cell in its adherent culture state were applied to various types of cells. Temporal wound repair was successfully observed. Moreover, we successfully stained portions of a living single cell to measure the organelle transport speed (mitochondria as a model) inside a cell. This technique opens up new avenues for cellular wound repair and subcellular behavior analyses.

Abstract:Minimally invasive surgical techniques, including endoscopic and robotic procedures, continue to revolutionize patient care, for their ability to minimize surgical trauma, thus promoting faster recovery and reduced hospital stays. Yet, the suturing of soft tissues ensuring damage-free tissue bonding during these procedures remains challenging due to missing haptics and the fulcrum effect. Laser tissue soldering has potential in overcoming these issues, offering damage-free seamless tissue fusion. To ensure the precision and safety of laser tissue soldering, we introduce feedback controlled fluorescent nanothermometry-guided laser tissue soldering using nanoparticle-protein solders within endoscopic and robotic contexts. Temperature-sensitive fluorescent nanoparticles embedded in the solder provide surgeons with immediate feedback on tissue temperatures during laser application, all while within the confines of minimally invasive (robotic) surgical setups. By integrating fluorescent nanothermometry-guided laser tissue surgery into endoscopic and robotic surgery, we pave the way for a new approach for safe and atraumatic soft tissue joining, especially in regions where traditional suturing is unfeasible.

High-Precision Implant Cavity Fabrication Using Femtosecond Lasers

Abstract:Objective: This study aims to enhance the precision of implant cavity preparation, addressing a notable challenge in the current state of the field by utilizing femtosecond lasers. Background: The application of femtosecond lasers in implant cavity preparation heralds a noninvasive and efficient technique, characterized by diminished thermal damage and high biocompatibility. Despite these promising attributes, the realization of precise cavity preparation remains a significant challenge in the contemporary domain. Materials and Methods: Our research group devised a specialized femtosecond laser microsurgery robotic system tailored for sophisticated implant cavity preparation. This system facilitated the meticulous analysis of sheep shank bone samples, enabling precise three-dimensional cutting. The analysis included an extensive examination of ablation effects, using a laser scanning microscope and VK Analyzer software. This investigation spanned the phases of laser flux calibration and experimental validation, offering a critical evaluation of the automated preparation process. Results: The study delineated that at the focus position of our custom-made oral clinical femtosecond laser microsurgery robotic system, the laser spot diameter is 75.69 μm, and ascertained the ablation threshold for sheep shank cortical bone to be 1.47 J/cm2. Utilizing low laser flux with minimal ablation craters overlap compromised the sidewall precision of the implant cavity, whereas employing high laser flux with extensive ablation craters overlap resulted in an enlarged ablation angle. At a laser energy setting of 2.2362 J/cm2 and a 50% ablation crater overlap, an implant cavity was successfully crafted featuring a top diameter of 4.41 mm, a bottom diameter of 3.98 mm, and a depth of 3 mm, devoid of any adverse thermal effects such as cracking or carbonization. Conclusions: The oral clinical femtosecond laser microsurgery robotic system can achieve automated and precise implant cavity preparation. This advancement promotes the broader application of femtosecond lasers in the field of orthopedics.

Abstract:New challenges in minimally invasive surgery (MIS), particularly for microsurgery, demand further advances in laser scalpels and medical robots. In this Perspective, we discuss challenges and solutions to develop small-scale and flexible laser scalpels. We review medical continuum robots and propose a fiberbotic laser scalpel that combines advanced laser ablation, actuation, and information acquisition technologies as a novel platform. It would enable surgeons to have safer and quicker access to hard-to-reach sites, thus enhancing the device's effectiveness in complex surgeries.

Abstract:Cellular heterogeneity facilitating specialization of cells is essential to control normal physiological processes as well as drive pathogenesis of many diseases. While it remains common to measure population-averaged properties in biological studies, the assumption that all cells are identical can lead to incorrect or at least imprecise assessments. To better understand heterogeneous characteristics and responses of individual cells, single-cell analytic methods should be applied to profile individual cellular properties. Using micro-fabrication technology, we have developed high-throughput microfluidic platforms, that enable tracking of thousands of single cells on-chip. Building on the cell isolation capability, we innovated a photomechanical actuation method to selectively detach and retrieve target cells for downstream analysis. Irradiation of a nanosecond-pulsed laser is utilized to generate shear force for detaching target cells cultured on carbon nanotube–polydimethylsiloxane (CNT-PDMS) composite. This non-destructive cell retrieval method can preserve cell viability, membrane proteins, and mRNA expression levels, so we have successfully performed single-cell transcriptome analysis as well as functional studies on the retrieved target cells. In addition to cell tracking and retrieval, we applied machine learning techniques to correlate cellular morphological features with cellular functions including migration, cancer drug response, and tumor initiation. Machine made significantly better predictions than experienced researchers, and we found novel morphological features facilitating breakthroughs in mechanistic understanding. The integrated single-cell manipulation and analysis augmented with machine learning will change how we understand cell biology and ultimately improve how we treat diseases.

Abstract:Laser microdissection technology is favored by biomedical researchers for its ability to rapidly and accurately isolate target cells and tissues. However, the precision cutting capabilities of existing laser microdissection systems are hindered by limitations in overall mechanical movement accuracy, resulting in suboptimal cutting quality. Additionally, the use of current laser microdissection systems for target acquisition may lead to tissue burns and reduced acquisition rates due to inherent flaws in the capture methods. To address these challenges and achieve precise and efficient separation and capture of cellular tissues, we integrated a digital micromirror device (DMD) into the existing system optics to modulate spatial light. This allows the system to not only implement the traditional point scanning cutting method but also utilize the projection cutting method.We have successfully cut various patterns on commonly used laser microdissection materials such as PET films and mouse tissues. Under projection cutting mode, we were able to achieve precise cutting of special shapes with a diameter of 7.5 micrometers in a single pass, which improved cutting precision and efficiency. Furthermore, we employed a negative pressure adsorption method to efficiently collect target substances. This approach not only resulted in a single-pass capture rate exceeding 90% for targets of different sizes but also enabled simultaneous capture of multiple targets, overcoming the limitations of traditional single-target capture and enhancing target capture efficiency, and avoiding potential tissue damage from lasers.In summary, the integration of the digital micromirror device into laser microdissection systems significantly enhances cutting precision and efficiency, overcoming limitations of traditional systems. This advancement demonstrates the accuracy and effectiveness of laser microdissection systems in isolating and capturing biological tissues, highlighting their potential in medical applications.

Abstract:Femtosecond lasers enable flexible and thermal-damage-free ablation of solid materials and are expected to play a critical role in high-precision cutting, drilling, and shaping of electronic chips, display panels, and industrial parts. Although the potential applications are theoretically predicted, true 3D nano-sculpturing of solids such as glasses and crystals, has not yet been demonstrated, owing to the technical challenge of negative cumulative effects of surface changes and debris accumulation on the delivery of laser pulses and subsequent material removal during direct-write ablation. Here, a femtosecond laser-induced cavitation-assisted true 3D nano-sculpturing technique based on the ingenious combination of cavitation dynamics and backside ablation is proposed to achieve stable clear-field point-by-point material removal in real time for precise 3D subtractive fabrication on various difficult-to-process materials. As a result, 3D devices including free-form silica lenses, micro-statue with vivid facial features, and rotatable sapphire micro-mechanical turbine, all with surface roughness less than 10 nm are readily produced. The true 3D processing capability can immediately enable novel structural and functional micro-nano optics and non-silicon micro-electro-mechanical systems based on various hard solids.
Baseline Hearing Measurements in Alaskan Belugas

Abstract:Abstract : While hearing is the primary sensory modality for odontocetes, there are few data addressing baseline hearing and subsequent variation within a natural population. To determine the effects of noise on marine mammals we need to understand what they hear. This is imperative in the Arctic where there is both an increase in human activity and a concurrent increase in human-produced noise. This work examines the hearing sensivitiy and variability of wild beluga whales in an effort to understand how belugas may be impacted by noise. A standard audiogram was determined from the wild animals, noting the variation between animals and the audiogram of maximal and minimal sensitivity. These novel data were compared to available hearing results from captive belugas, evaluating differences and variation within the two data sets. The hearing curves were appraised relative to basic demographic meta-data from the animals from which the measurements were made. Through these data analyses we sought to: 1) define the natural and baseline hearing abilities and variability in belugas, 2) place the results in the context of potential ecological influences and that of anthropogenic noise, and 3) evaluate the validity of captive-based hearing data in relation to wild animals. This is part of a larger effort to understand variation in the sensory biology and noise susceptibility of diverse odontocete species.

Abstract:As a noncontact strategy with flexible tools and high efficiency, laser precision engineering is a significant advanced processing way for high-quality micro-/nanostructure fabrication, especially to achieve novel functional photoelectric structures and devices. For the microscale creation, several femtosecond laser fabrication methods, including multiphoton absorption, laser-induced plasma-assisted ablation, and incubation effect have been developed. Meanwhile, the femtosecond laser can be combined with microlens arrays and interference lithography techniques to achieve the structures in submicron scales. Down to nanoscale feature sizes, advanced processing strategies, such as near-field scanning optical microscope, atomic force microscope, and microsphere, are applied in femtosecond laser processing and the minimum nanostructure creation has been pushed down to ~25 nm due to near-field effect. The most fascinating femtosecond laser precision engineering is the possibility of large-area, high-throughput, and far-field nanofabrication. In combination with special strategies, including dual femtosecond laser beam irradiation, ~15 nm nanostructuring can be achieved directly on silicon surfaces in far field and in ambient air. The challenges and perspectives in the femtosecond laser precision engineering are also discussed.

Abstract:Using tight focusing light, optical tweezers (OT) are tools that can manipulate and capture microscopic particles and biological cells as well as characterize a wide range of micro and nanomaterials. In this paper, we focused on fibroblasts, which are widely used in the biomedical area for a variety of purposes, including promoting human wound healing and preventing the early proliferation of tumor cells. We first built an optical tweezer experimental platform, using an 808 nm continuous-wave laser as the capture light source, to confirm that the device can precisely control the movement of single or multiple particles as well as fibroblasts. Then, a 1030 nm femtosecond laser was employed as the capture light source to study the manipulation of microparticles and fibroblasts at different powers. Lastly, a protracted manipulation protocol was used to prevent the fibroblasts from adhering to the wall. This method can be used to isolate and precisely block adherent growth of fibroblasts in cell populations. This experimental result can be further extended to other biological cells.

Abstract:Femtosecond laser microsurgery has become an advanced method for clinical procedures and biological research. The tissue treated by femtosecond laser can become highly fluorescent, indicating the formation of new fluorescent compounds that can naturally label the treated tissue site. We systematically characterized the fluorescence signals produced by femtosecond laser ablation in biological tissues in vivo. Our findings showed that they possess unique fluorescence properties and can be clearly differentiated from endogenous signals and major fluorescent proteins. We further demonstrated that the new fluorescent compounds can be used as in vivo labelling agent for biological imaging and guided laser microsurgery.
Micro-processing of Polymers and Biological Materials Using High Repetition Rate Femtosecond Laser Pulses

Abstract:High repetition rate femtosecond laser micro-processing has been applied to ophthalmological hydrogel polymers and ocular tissues to create novel refractive and diffractive structures. Through the optimization of laser irradiation conditions and material properties, this technology has become feasible for future industrial applications and clinical practices. A femtosecond laser micro-processing workstation has been designed and developed. Different experimental parameters of the workstation such as laser pulse duration, focusing lens, and translational stages have been described and discussed. Diffractive gratings and three-dimensional waveguides have been fabricated and characterized in hydrogel polymers, and refractive index modifications as large as + 0.06 have been observed within the laser-irradiated region. Raman spectroscopic studies have shown that our femtosecond laser micro-processing induces significant thermal accumulation, resulting in a densification of the polymer network and increasing the localized refractive index of polymers within the laser irradiated region. Different kinds of dye chromophores have been doped in hydrogel polymers to enhance the two-photon absorption during femtosecond laser micro-processing. As the result, laser scanning speed can be greatly increased while the large refractive index modifications remain. Femtosecond laser wavelength and pulse energy as well as water and dye concentration of the hydrogels are optimized. Lightly fixed ocular tissues such as corneas and lenses have been micro-processed by focused femtosecond laser pulses, and refractive index modifications without any tissue-breakdown are observed within the stromal layer of the corneas and the cortex of the lenses. Living corneas are doped with Sodium Fluorescein to increase the two-photon absorption during the laser micro-processing, and laser scanning speed can be greatly increased while inducing large refractive index modifications. No evidence of cell death has been observed in or around the laser-induced refractive index modification regions. These results support the notion that femtosecond laser micro-processing method may be an excellent means of altering the refraction or higher order aberration content of corneal tissue without cell death and short-term tissue damage, and has been named as Intra-tissue Refractive Index Shaping (IRIS). The femtosecond laser micro-processing workstation has also been employed for laser transfection of single defined cells. Some preliminary results suggest that this method can be used to trace individual cells and record their biological and morphological evolution, which is quite promising in many biomedical applications especially in immunology science. In conclusion, high repetition rate femtosecond laser micro-processing has been employed to fabricate microstructures in ophthalmological hydrogels and ocular tissues. A high-throughput all-optical laser-scanning imaging flow cytometer with biomolecular specificity and subcellular resolution

Abstract:Image-based cellular assay advances approaches to dissect complex cellular characteristics through direct visualization of cellular functional structures. However, available technologies face a common challenge, especially when it comes to the unmet need for unraveling population heterogeneity at single-cell precision: higher imaging resolution (and thus content) comes at the expense of lower throughput, or vice versa. To overcome this challenge, a new type of imaging flow cytometer based upon an all-optical ultrafast laser-scanning imaging technique, called free-space angular-chirp-enhanced delay (FACED) is reported. It enables an imaging throughput (>20 000 cells s-1 ) 1 to 2 orders of magnitude higher than the camera-based imaging flow cytometers. It also has 2 critical advantages over optical time-stretch imaging flow cytometry, which achieves a similar throughput: (1) it is widely compatible to the repertoire of biochemical contrast agents, favoring biomolecular-specific cellular assay and (2) it enables high-throughput visualization of functional morphology of individual cells with subcellular resolution. These capabilities enable multiparametric single-cell image analysis which reveals cellular heterogeneity, for example, in the cell-death processes demonstrated in this work-the information generally masked in non-imaging flow cytometry. Therefore, this platform empowers not only efficient large-scale single-cell measurements, but also detailed mechanistic analysis of complex cellular processes.

Abstract:Advanced photonic tools may enable researchers and clinicians to visualize, track, control and manipulate biological processes at the single-cell level in space and time. Biological systems are complex and highly organized on both spatial and temporal levels. If we are to study, perturb, engineer or heal biological entities, we must be able to visualize key players in such systems and to track, control and manipulate them precisely and selectively. To achieve this goal, the engineering of non-destructive tools will allow us to interrogate and manipulate the function of proteins, pathways and cells for physicians, enabling the design of 'smart materials' that can direct and respond to biological processes on-demand. Among the potentially exploitable non-destructive tools, light-based actuation is particularly desirable, as it enables high spatial and temporal resolution, dosage control, minimal disturbance to biological systems and deep tissue penetration. Here, we overview existing approaches toward the engineering of light-activated tools for the interrogation and manipulation of single-cell processes, and list the types of studies and types of functions that can be controlled by light. Timely applications, such as studies of inflammation and of crossing brain barrier systems - via super-resolution imaging and optomechanical manipulation - are two representative examples of emerging applications so far never addressed.
High Efficiency Fabrication of Complex Microtube Arrays by Scanning Focused Femtosecond Laser Bessel Beam for Trapping/Releasing Biological Cells

Abstract:In this paper, we present a focused femtosecond laser Bessel beam scanning technique for the rapid fabrication of large-area 3D complex microtube arrays. The femtosecond laser beam is converted into several Bessel beams by two-dimensional phase modulation using a spatial light modulator. By scanning the focused Bessel beam along a designed route, microtubes with variable size and flexible geometry are rapidly fabricated by two-photon polymerization. The fabrication time is reduced by two orders of magnitude in comparison with conventional point-to-point scanning. Moreover, we construct an effective microoperating system for single cell manipulation using microtube arrays, and demonstrate its use in the capture, transfer, and release of embryonic fibroblast mouse cells as well as human breast cancer cells. The new fabrication strategy provides a novel method for the rapid fabrication of functional devices using a flexibly tailored laser beam.

Abstract:Lab-on-a-chip devices have been intensively developed during the last decade when emerging technologies offered possibilities to manufacture reliable devices with increased spatial resolution. These biochips allowed testing chemical reactions in nanoliter volumes with enhanced sensitivity and lower consumption of reagents. We have developed a new technology by combining subtractive ultrafast laser assisted chemical etching of glasses and additive two-photon polymerization to integrate 3D glass microfluidics and polymer microcomponents in a single biochip. The innovative hybrid "ship-in-a-bottle" approach is not only an instrument that can tailor 3D environments but also a tool to fabricate biomimetic in vivo structures inside a glass microfluidic chip. It was possible to create appropriate environment for cell culturing and to offer robustness and transparency for optical interrogation. Cancer cells were cultivated inside biochips and monitored over short and long periods. With the view of understanding cancer cells specific behavior such as migration or invasiveness inside human body, introduction of different geometrical configurations and chemical conditions were proposed. The cells were found responsive to a gradient of nutrient concentration through the microchannels of a 3D polymeric scaffold integrated inside glass biochip.
Optogenetic-controlled immunotherapeutic designer cells for post-surgical cancer immunotherapy

Abstract:Abstract Surgical resection is the main treatment option for most solid tumors, yet cancer recurrence after surgical resection remains a significant challenge in cancer therapy. Recent advances in cancer immunotherapy are enabling radical cures for many tumor patients, but these technologies remain challenging to apply because of side effects related to uncontrollable immune system activation. Here, we develop far-red light-controlled immunomodulatory engineered cells (FLICs) that we load into a hydrogel scaffold, enabling the precise optogenetic control of cytokines release (IFN-β, TNF-α, and IL-12) upon illumination. Experiments with a B16F10 melanoma resection mouse model show that FLICs-loaded hydrogel implants placed at the surgical wound site achieve sustainable release of immunomodulatory cytokines, leading to prevention of tumor recurrence and increased animal survival. Moreover, the FLICs-loaded hydrogel implants elicit long-term immunological memory that prevents against tumor recurrence. Our findings illustrate that this optogenetic perioperative immunotherapy with FLICs-loaded hydrogel implants offers a safe treatment option for solid tumors based on activating host innate and adaptive immune systems to inhibit tumor recurrence after surgery. Beyond extending the optogenetics toolbox for immunotherapy, we envision that our optogenetic-controlled living cell factory platform could be deployed for other biomedical contexts requiring precision induction of bio-therapeutic dosage.

Abstract:The less-invasive non-embedded cell cutting or slicing technique provides opportunities for a bio-study at subcellular scale, but there are few effective solutions available at the current stage. This paper reports a robot-aided vibrating system for less-invasive non-embedded cell cutting and investigates the role of key vibrating parameters in the cell cutting process. First, a nanoknife with sharp angle 5° is fabricated from a commercial atomic force microscope cantilever by focused ion beam etching and a vibrating system is constructed from a piezo actuator. Then, they are integrated with a self-developed nanorobotic manipulation system inside an environment scanning electron microscope. After that, we choose yeast cells as the sample to implement the vibrating cutting and investigate the effect of vibrating parameters (frequency and amplitude) on cell cutting quality. The results clearly indicate that the vibrating nanoknife is able to reduce the cutting force and improve the cutting quality. It is also suggested that the repeated load-unload (impact) cycle is the main reason for the better performance of vibrating cutting. The effect of vibrating parameters at small scale benefits our fundamental understanding on cell mechanics, and this research paves a way for the low-destructive non-embedded cell cutting and promotes the practical cell cutting techniques.
