Abstract:A compact ultrahigh-pressure nanoflow liquid chromatograph (LC) was developed with the purpose in mind of creating a portable system that could be easily moved to various testing locations or placed in close proximity to other instruments for optimal coupling, such as with mass spectrometry (MS). The system utilized innovative nanoflow pumps integrated with a very low volume stop-flow injector and mixing tee. The system weighed only 5.9 kg (13 lbs) or 4.5 kg (10 lbs) without a controller and could hold up to 1100 bar (16000 psi) of pressure. The total volume pump capacity was 60 μL. In this study, the sample injection volume was determined by either a 60 nL internal sample groove machined in a high-pressure valve rotor or by a 1 μL external sample loop, although other sample grooves or loops could be selected. The gradient dwell volume was approximately 640 nL, which allowed significant reduction in sample analysis time. Gradient performance was evaluated by determining the gradient step accuracy. A low RSD (0.6%, n = 4) was obtained for day-to-day experiments. Linear gradient reproducibility was evaluated by separating a three-component polycyclic aromatic hydrocarbon mixture on a commercial 150 μm inner diameter capillary column packed with 1.7 μm particles. Good retention-time reproducibility (RSD < 0.17%) demonstrated that the pumping system could successfully generate ultrahigh pressures for use in capillary LC. The system was successfully coupled to an LTQ Orbitrap MS in a simple and efficient way; LC-MS of a trypsin-digested bovine serum albumin (BSA) sample provided narrow peaks, short dwell time, and good peptide coverage.
A Pressure-Driven Capillary Electrophoretic System with Injection Valve Sampling.

Abstract:To improve repeatability and efficiency and to simplify the operation procedure of capillary electrophoresis (CE), a pressurized CE system (p-CE) with injection valve sampling was developed. It consisted of one high-pressure pump, a six-port injection valve, a PEEK cross, a separation and back pressure capillary, an ultraviolet-visible detector and a high voltage power supply. The pressure-driven flow ranging from 4.5 nL min(-1) to 0.81 μL min(-1) in the separation capillary was produced by splitting to the flow from the high-pressure pumps (0.005-0.4 mL min(-1)). Nano-volume sample injection (<10 nL) was conducted by a micro-volume rotary injector (0.5-5 μL) with flow splitting. In the p-CE system, the new commercial capillary could be directly used without any wash, and the capillary-flush process between runs was also eliminated. In this case, the analytes were driven toward the outlet of the separation capillary by the pressurized flow, the electric field force and minute electroosmotic flow, and they were separated owing to the electrophoretic mobility. The p-CE system allows for the independent variation of the pressurized flow rate and electrical field and electrophoretic separation of good repeatability (below 3%) under high electrical fields (500-1000 V cm(-1)) and flow rate gradient modes. The feasibility of the p-CE system in real analysis was demonstrated by iodate quantification in iodized table salts. The separation of iodide and iodate was realized within 0.3 min, proving its high analytical speed.
Valveless gated injection for microfluidic chip-based liquid chromatography system with polymer monolithic column.

Abstract:In this work, we developed a microfluidic chip-based liquid chromatography (LC) system with valveless gated sample injection method and monolithic columns. The valveless LC system consisted of a microchip with a separation column and a balance column, two syringe pumps and a laser induced fluorescence detector. Nanoliter-scale sample injection was achieved by simply switching the ON and OFF states of the syringe pumps under gated injection mode, without the need of any mechanical valve. The balance column facilitated the gated injection process by balancing the back pressure in the sample channel with that in the separation channel. Under the gated injection mode, the sample injection volumes could be varied in the range of 0.4-2.4 nL. We investigated the effects of the mobile-phase composition, flow rate, and the sample injection volume on the performance of the LC system. Under the optimized conditions, two fluorescently labeled amines were successfully separated in less than 220 s with a column length of 1cm. Theoretical plates ranging from 35,700 to 29,400 per meter, corresponding to plate heights ranging from 28 to 34 μm were obtained. The advantages of the present system include simple system structure, ease of operation, convenience for varying injection volume, and high sample loading capacity.

Abstract:A miniaturized capillary electrophoresis (CE) system with UV-Vis detection was coupled to a flow injection (FI) system for achieving high throughput continuous sample introduction. The cassette of a commercial CE instrument was modified to hold a 6.5cm long silica capillary and a flow-through waste reservoir. The cassette was inserted into the flow-cell chamber of a commercial UV detector, with the light beam focused on the capillary and collected by two ball lenses on the cassette. The capillary inlet, left outside the cassette and detector, was positioned on the top of a vertical 3.5mm diameter glass rod, in close contact with an electrode. Samples injected through the FI system dropped freely on top of the pillar, covering the capillary inlet and electrode. Continuous sample introduction was achieved for CE separations under non-interrupted separation voltage, which was isolated from the FI system through the discontinuity of droplets. The newly developed interface and UV detection system was used for fast separation of sulphamethoxazole (SMZ) and trimethoprim (TMP) in sulphatrim tablets, achieving a high throughput of over 48h−1, and a low carryover of 2%. Separation efficiencies of 8μm plate height and detection limits of 1.0mgl−1 for SMZ and 0.5mgl−1 (3σ) for TMP were obtained.

Abstract:For direct analyses of micro-samples and the hyphenation with nanoflow separation techniques, a low-cost demountable nanoflow nebulizer was fabricated for inductively coupled plasma mass spectrometry (ICPMS). A small-bore capillary (20 mm) with a tapered tip served as the sample capillary of the nebulizer. The gas orifice diameter, the internal diameter and the wall thickness at the tip of the sample capillary were 160, 20, and 5 mm, respectively, which ensured stable generation of aerosol by the nebulizer at nanoflow rates down to 50 nL min(-1) and an ultra-low aspiration rate (80 nL min(-1)). Narrower aerosol size distribution and higher analyte transport efficiency (nearly 100%) were obtained with the proposed nebulizer than with the previous demountable capillary microflow nebulizer. The sensitivity and detection limit of the proposed nebulizer at 500 nL min(-1) were about one third of those obtained by using the demountable capillary microflow nebulizer at 5 mL min(-1), whereas the precision of the former at 500 nL min(-1) was even to some extent superior over that obtained with the latter at 5 mL min(-1). Furthermore, the ICP-MS instrument could tolerate direct introduction of 100% organic solvents by using the developed nebulizer operating at nanoflow rates. It was utilized to sheathless interface nano-high performance liquid chromatography with ICP-MS for arsenic speciation analysis with good accuracy and precision, proving its strong robustness and good suitability.

Abstract:A wide variety of analytical techniques have been employed for monitoring chemical reactions, with online instrumentation providing additional benefits compared to offline analysis. A challenge in the past for online monitoring has been placement of the monitoring instrumentation as close as possible to the reaction vessel to maximize sampling temporal resolution and preserve sample composition integrity. Furthermore, the ability to sample very small volumes from bench-scale reactions allows the use of small reaction vessels and conservation of expensive reagents. In this study, a compact capillary LC instrument was used for online monitoring of as small as 1 mL total volume of a chemical reaction mixture, with automated sampling of nL-scale volumes directly from the reaction vessel used for analysis. Analyses to demonstrate short term (~2 h) and long term (~ 50 h) reactions were conducted using tandem on-capillary ultraviolet absorbance followed by in-line MS detection or ultraviolet absorbance detection alone, respectively. For both short term and long term reactions (10 and 250 injections, respectively), sampling approaches using syringe pumps minimized the overall sample loss to ~0.2% of the total reaction volume.

Abstract:The development of efficient sample introduction and pretreatment systems for microfluidic chip-based analytical systems is important for their application to real-life samples. In this work, world-to-chip interfacing was achieved by a novel flow-through sampling reservoir featuring a guided overflow design. The flow-through reservoir was fabricated on a 30 x 60 x 3 mm planar glass chip of crossed-channel design used for capillary electrophoresis separations. The 20-microL sample reservoir was produced from a section of plastic pipet tip and fixed at one end of the sampling channel. Sample change was performed by pumping 80-microL samples sandwiched between air segments at approximately 0.48 mL/min flow rate through the flow-through reservoir, introduced from an access hole on the bottom side of the chip. A filter paper collar wrapped tightly around the reservoir guided the overflowing sample solution into a plastic trough surrounding the reservoir and then to waste. The performance of the system was demonstrated in the separation and determination of FITC-labeled arginine, glycine, phenylalanine, and glutamic acid with LIF detection, by continuously introducing a train of different samples through the system without electrical interruption. Employing a separation channel of 4 cm (2-cm effective separation length) and 1.4-kV separation voltage, maximum throughputs of 80/h were achieved with <4.1% carryover and precisions ranging from 1.5% for arginine to 2.6% RSD (n = 11) for glycine. The sampling system was tested in the continuous monitoring of the derivatizing process of amino acids by FITC over a period of 4 h, involving 166 analytical cycles. An outstanding overall precision of 4.8% RSD (n = 166) was achieved for the fluorescein internal standard.

Abstract:A novel microfluidic picoliter-scale sample introduction approach was developed by combining the spontaneous injection approach with a capillary electrophoresis (CE) system based on a short capillary and slotted-vial array. A droplet splitting phenomenon at the capillary inlet end during the spontaneous sample introduction process was observed for the first time. On the basis of this phenomenon, a translational spontaneous injection approach was established to reduce sample injection volumes to the sub-100 pL range. A versatile high-speed capillary electrophoresis (HSCE) system was built on the basis of this sample injection approach with separation performance comparable to or even better than those reported in microfluidic chip-based CE systems. The HSCE system was composed of a short fused-silica capillary and an automated sample introduction system with slotted sample and buffer reservoirs and a computer-programmed translational stage. The translational spontaneous sample injection was performed by linearly moving the stage, allowing the capillary inlet first to enter the sample solution and then removing it. A droplet was left at the tip end and spontaneously drawn into the capillary by surface tension effect to achieve sample injection. The stage was continuously moved to allow the capillary inlet to be immersed into the buffer solution, and CE separation was performed by applying a high voltage between the buffer and waste reservoirs. With the use of the novel system, high-speed and efficient capillary zone electrophoresis (CZE) separation of a mixture of five fluorescein isothiocyanate (FITC) labeled amino acids was achieved within 5.4 s in a short capillary with a separation length of 15 mm, reaching separation efficiencies up to 0.40 microm plate height. Outstanding peak height precisions ranging from 1.2% to 3.7% RSD were achieved in 51 consecutive separations. By extension of the separation length to 50 mm, both high-speed and high-resolution CZE separation of eight FITC-labeled amino acids could be obtained in less than 21 s with theoretical plates ranging from 163,000 to 251,000 (corresponding to 0.31-0.20 microm plate heights). The present HSCE system also allowed fast chiral separations of FITC-labeled amino acids under micellar electrokinetic chromatography (MEKC) mode within 6.5 s.

Abstract:A newly developed portable capillary liquid chromatograph was investigated for the separation of various pharmaceutical and illicit drug compounds. The system consists of two high-pressure syringe pumps capable of delivering capillary-scale flow rates at pressures up to 10,000 psi. Capillary LC columns packed with sub-2 μm particles are housed in cartridges that can be inserted into the system and easily connected through high-pressure fluidic contact points by simply applying a specific, pre-determined torque rather than using standard fittings and less precise sealing protocols. Several over-the-counter analgesic drug separations are demonstrated, along with a simple on-line measurement of tablet dissolution. Twenty illicit drug compounds were also separated across six targeted drug panels. The results described in this study demonstrate the capability of this compact LC instrument to address several important drug-related applications while simplifying system operation, and greatly reducing solvent usage and waste generation essential for on-site analysis. Abstract:In recent years, miniaturized analytical instruments have been developing to meet the needs of portable and rapid analysis. The key of miniaturized analytical equipment is the miniaturization and integration of functional modules. This paper aims to develop a miniaturized photometric detector and separation microfluidic chip for a liquid chromatography (LC) system. The detector uses a light-emitting diode to emit ultraviolet light, which is collimated by an internal double lens. A Z-shaped flow cell with a long optical path is designed and fabricated in the separation microfluidic chip with a three-layer structure, which provides a tubing-free connection between the separation and detection unit. Detector performance is evaluated using hemoglobin (Hb) samples, with an upper limit of detection linearity (95 %) of 0.345 AU and stray light level as low as 0.08 %. Additionally, the microchip channel can be filled with cation exchange resin and C18 particles. Finally, an ion LC system and a reversed-phase LC system were constructed based on the miniaturized photometric detector and two microchips with different packed columns, respectively, and were successfully used in the separation and detection of two metabolic markers (glycated hemoglobin or bilirubin). The results of this study are expected to facilitate the development of a portable LC system and their application in community health services and family health management of chronic diseases.

Abstract:An automated microfluidic sequential injection analysis system that efficiently manipulates sample and reagent solutions in the nanoliter range in approximately 10 s per analytical cycle is described. The system consisted of a 6-cm-long, typically 75-microm i.d., fused-silica capillary (which functioned as a sampling probe and reactor as well as a flow-through detection cell), a horizontally oriented waste reservoir that provided liquid level differences for inducing gravity-driven flows, an autosampling device holding samples and reagents with horizontally fixed slotted microvials, and a laser-induced fluorescence detection system. Sample and reagent zones were sequentially introduced via gravity-driven flow by scanning the capillary tip (functioning as the sampling probe) through the vial slots, while vials containing sample, reagent, and carrier were sequentially rotated to the probe by programmed movement of the vial holders. Sequentially injected nanoliter zones were rapidly mixed by convection and diffusion within the carrier flow, demonstrating a behavior that conformed well to the Taylor dispersion model, and zone penetration effects were characterized and optimized under Taylor's dispersion theory guidelines. For the determination of fluorescein, a high throughput of 400 h(-1) was achieved, rapidly producing calibration curves (five points) within 45 s. Owing to its adaptability to the Taylor's dispersion model, the system was used also for measuring diffusion coefficients of fluorescent species. Potentials for using the system in enzyme inhibition assays were demonstrated by a reaction involving the conversion of fluorescein digalactoside to fluorescent hydrolysates via beta-galactosidase and the inhibition of beta-galactosidase by diethylenetriaminepentaacetic acid.

Abstract:The design of a miniaturized LED-based UV-absorption detector was significantly improved for on-column nanoflow LC. The detector measures approximately 27mm×24mm×10mm and weighs only 30g. Detection limits down to the nanomolar range and linearity across 3 orders of magnitude were obtained using sodium anthraquinone-2-sulfonate as a test analyte. Using two miniaturized detectors, a dual-detector system was assembled containing 255nm and 275nm LEDs with only 216nL volume between the detectors A 100μm slit was used for on-column detection with a 150μm i.d. packed capillary column. Chromatographic separation of a phenol mixture was demonstrated using the dual-detector system, with each detector producing a unique chromatogram. Less than 6% variation in the ratios of absorbances measured at the two wavelengths for specific analytes was obtained across 3 orders of magnitude concentration, which demonstrates the potential of using absorption ratio measurements for target analyte detection. The dual-detector system was used for simple, but accurate, mobile phase flow rate measurement at the exit of the column. With a flow rate range from 200 to 2000nL/min, less than 3% variation was observed.

Abstract:Abstract The speed of analysis in capillary gas chromatography can be substantially increased by reduction of the column inner diameter. However, special demands are then posed upon instrumental design. In particular, the sampling system is highly critical because it has to be capable of delivering extremely small injection band widths which must be compatible with the column inside diameter. This study focuses on the evaluation of two potentially suitable sample introduction systems with respect to input band width and detection limits and their compatibility with small bore (≦ 100 μm) columns in capillary gas chromatography. One of them allows liquid on‐column injection, based on liquid splitting, of only a few nl onto small bore (≦ 100 ◂m) fused silica columns. For gases, input band widths as low as 1 ms are obtained with this system. The other one is part of a miniaturized gas chromatograph with extremely low dead volume interfaces and detector volumes. It allows input band widths for gases of a few ms. Without any preconcentration ppm concentrations are measured in gaseous samples with a 80 ◂m thick film capillary column. It will be shown that a further reduction of the minimum detectable amount and analysis time is possible with this equipment.

Abstract:The aim of this study was to use a systematic schedule, including urodynamics, to describe the rate of coexisting overactive bladder (OB) in patients with bladder outlet obstruction (BOO) caused by benign prostatic hyperplasia (BPH). We also identified differences between the patients with pure BOO compared with those with BOO combined with OB (BOO + OB). One hundred and sixty‐two men referred to our clinic due to LUTS were included. Patients with a history that might affect their bladder function were excluded. After cystometry and pressure–flow studies, the patients were divided into pure BOO and BOO + OB. Of the 162 men, 55% had pure BOO. BOO + OB was found in 45%. Age, s‐PSA, voided volume, and obstruction grade differed significantly between the groups. The patients with BOO + OB were older, had a higher s‐PSA, voided smaller volumes, and were more obstructed. We found no differences in TRUS‐volume, Q‐max, IPS score, or PVR. There was a strong association between OB and BOO, the percentage of OB increasing with increased obstruction. TRUS‐volume, Q‐max, IPS score, and PVR did not predict whether the patients had a combined BOO + OB or not. Abstract:In this paper, we use the concept of the HPLC 3D calibration surface for evaluating reaction performance in micro-mole scale self-optimizing flow systems. This approach enables comparing the analyte and internal standard relative levels for a wide range of internal standard concentrations. This approach considers fluctuations in response factors and potential nonlinearities in the Beer-Lambert law, which arise from performing HPLC analyses at different concentration ranges. The concept is validated by constructing a calibration surface for a formal [3 + 3] cycloaddition. The robustness of the 3D calibration surface is assessed in an autonomous flow self-optimization in a non-steady state regime, where a standard 2D calibration curve produces inconsistent results due to variable concentration HPLC injections.
