Abstract:A benchtop miniature mass spectrometer system, Mini 12, with ambient ionization source and tandem mass spectrometry capabilities has been developed and characterized. This instrument was developed as a self-contained system to produce quantitative results for unprocessed samples of small volumes including nonvolatile analytes. The ion processing system, vacuum system, and control system are detailed. An integrated sample loading system facilitates automated operation. A user interface has been developed to acquire and to interpret analytical results for personnel who have limited mass spectrometry knowledge. Peak widths of Δm/z 0.6 Th (full width at half-maximum) and a mass range of up to m/z 900 are demonstrated with the rectilinear ion trap mass analyzer. Multistage experiments up to MS(5) are accomplished. Consumable cartridges have been designed for use in ambient paper spray ionization, and the recently developed extraction spray ionization method has been employed to improve the quantitative performance. Monitoring of trace-levels of chemicals in therapeutic drugs, as well as in food safety and environmental protection operations is demonstrated. Dual MS/MS scans are implemented to obtain the intensities of the fragment ions from the analyte and its internal standard, and the ratio is used in quantitative analysis of complex samples. Limits of quantitation (LOQ) of 7.5 ng/mL, with relative standard deviations below 10%, have been obtained for selected therapeutic drugs in whole blood throughout their individual therapeutic ranges.

Abstract:A recently developed hand-held, rectilinear ion trap mass spectrometer, capable of performing in situ analysis, has been evaluated for a variety of environmentally relevant analytes. Different sampling and ionization methods were implemented, demonstrating the considerable versatility of this instrument. A discontinuous (viz. pulsed) atmospheric pressure inlet (DAPI) was used to introduce externally-generated analyte ions. Nitro compounds were ionized by electrosonic spray ionization (ESSI) yielding the protonated and sodiated forms of the molecular ion, as well as fragment ions. The amines 2,2,6,6-tetramethylpiperidine, triethylamine and 2,6-diphenylpyridine showed low parts per billion (ppb) detection limits. Vapor phase external ionization was used to examine the chemical warfare simulant dimethyl methylphosphonate and the insect repellant N,N-diethyl- m-toluamide. Membrane introduction mass spectrometry (MIMS) was used as the introduction system for hydrophobic analytes using a selectively permeable (polydimethylsiloxane) membrane placed within the vacuum manifold with subsequent ionization of the thermally desorbed neutral compounds inside the ion trap. MIMS allowed the quantitation of trace levels (a few ppb) of fluorinated compounds in the vapor phase. MIMS was also applied to the quantitation of aqueous polycyclic aromatic hydrocarbons (PAH's) with limits of detection again in the low ppb range for naphthalene, acenaphthene, anthracene and phenanthrene.

Abstract:We report a micro-fractionation device for high performance liquid chromatography-mass spectrometry to archive chromatographic separations on an array of optimized surface energy traps (SETs). The method has the potential to significantly alter nanoflow LC-MS workflow, decoupling separation and analysis. The wetting characteristics of the SETs cause the HPLC eluent stream to spontaneously split into droplet microfractions. The droplet mirofractions are then dried down to enable facile storage and transport of the archived separation. Discontinuously dewetting array parameters were explored to maximize array volume and resolution using a combination of SET design, shape, size, and spacing. Mass spectrometry analysis is performed utilizing a liquid micro-junction surface sampling probe to extract dried analytes from the surface of the SETs followed by electrospray ionisation. A reverse phase separation of pharmaceutical compounds is "recorded" using the micro-fractionation device followed by "reading" the chromatographic trace with a mass spectrometer 24 hours after the separation was performed/archived, demonstrating a true decoupling of LC, and MS. Additionally, we demonstrate the ability to collect microfractions with sub-one-second integration time, approaching the scan time of a mass spectrometer or UV-Vis detector. With further improvements to the device, sub-1-second micro-fractionation may enable seamless reconstruction of archived chromatograms indistinguishable from online LC-MS data, while also providing the benefits of easy storage and transport of archived separations.

Abstract:For direct analyses of micro-samples and the hyphenation with nanoflow separation techniques, a low-cost demountable nanoflow nebulizer was fabricated for inductively coupled plasma mass spectrometry (ICPMS). A small-bore capillary (20 mm) with a tapered tip served as the sample capillary of the nebulizer. The gas orifice diameter, the internal diameter and the wall thickness at the tip of the sample capillary were 160, 20, and 5 mm, respectively, which ensured stable generation of aerosol by the nebulizer at nanoflow rates down to 50 nL min(-1) and an ultra-low aspiration rate (80 nL min(-1)). Narrower aerosol size distribution and higher analyte transport efficiency (nearly 100%) were obtained with the proposed nebulizer than with the previous demountable capillary microflow nebulizer. The sensitivity and detection limit of the proposed nebulizer at 500 nL min(-1) were about one third of those obtained by using the demountable capillary microflow nebulizer at 5 mL min(-1), whereas the precision of the former at 500 nL min(-1) was even to some extent superior over that obtained with the latter at 5 mL min(-1). Furthermore, the ICP-MS instrument could tolerate direct introduction of 100% organic solvents by using the developed nebulizer operating at nanoflow rates. It was utilized to sheathless interface nano-high performance liquid chromatography with ICP-MS for arsenic speciation analysis with good accuracy and precision, proving its strong robustness and good suitability.

Abstract:A major design objective of portable mass spectrometers is the ability to perform in situ chemical analysis on target samples in their native states in the undisturbed environment. The miniature instrument described here is fully contained in a wearable backpack (10 kg) with a geometry-independent low-temperature plasma (LTP) ion source integrated into a hand-held head unit (2 kg) to allow direct surface sampling and analysis. Detection of chemical warfare agent (CWA) simulants, illicit drugs, and explosives is demonstrated at nanogram levels directly from surfaces in near real time including those that have complex geometries, those that are heat-sensitive, and those bearing complex sample matrices. The instrument consumes an average of 65 W of power and can be operated autonomously under battery power for ca. 1.5 h, including the initial pump-down of the manifold. The maximum mass-to-charge ratio is 925 Th with mass resolution of 1-2 amu full width at half-maximun (fwhm) across the mass range. Multiple stages of tandem analysis can be performed to identify individual compounds in complex mixtures. Both positive and negative ion modes are available. A graphical user interface (GUI) is available for novice users to facilitate data acquisition and real-time spectral matching.

Abstract:A microfluidic system for introducing nanolitre sample in inductively coupled plasma mass spectrometry using electrokinetic flow combined with hydrodynamic flow was developed, which consisted of a microfluidic chip, a syringe pump, a high voltage power supply, a homemade microflow nebulizer and a heated single pass spray chamber. In the microfluidic chip, the sample in the reservoir S automatically flew across the intersection P-1 to the reservoir SR under hydrodynamic pressure. Two porous polymer plugs C-1 and C-2 were created on the microchip to isolate electrokinetic flow from the pressure-driven flow. Once a high voltage between S and buffer reservoir B was applied, the sample was driven through the porous polymer plug C-1 into the metering channel by electrokinetic force. Simply changing the sampling time can freely optimize the sampling volume down to 0.18 nL, which was injected into the nebulizer by the pressure-driven makeup solution. The nanolitre sample introducing system has the advantages of low sample consumption, satisfactory precision, low sampling dead volume (0.38 nL) and low sampling dead time (0.42 s). The proposed system was successfully applied to determine platinum content in a serum sample of a cancer patient on cisplatin chemotherapy. The sample consumption of 1.8 nL was about 10(5) times less than that with the conventional sampling system. The sample throughput up to 112 h(-1) could be achieved with the detection limit of 64 ng L-1 for Pt-195. The content of Pt in the sample was 8.4 +/- 0.3 mu g L-1, agreeing well with that (8.5 +/- 0.3 mu g L-1) determined by the conventional sampling system.

Abstract:A shoebox-sized, 10-kg, handheld mass spectrometer, Mini 10, based on a rectilinear ion trap mass analyzer has been designed, built, and characterized. This instrument has evolved from a decade-long experimental and simulation program in mass spectrometer miniaturization. The rectilinear ion trap has a simplified geometry and high trapping capacity, and when used with a miniature and ruggedized pumping system, it allows chemical analysis while the instrument is being carried. Compact electronics, including an air core RF drive coil, were developed to control the instrument and to record mass spectra. The instrument runs on battery power, consuming less than 70 W, similar to a laptop computer. Wired and wireless networking capabilities are implemented. The instrument gives unit resolution and a mass range of over m/z 500. Tandem mass spectrometry capabilities are implemented using collision-induced dissociation, and they are used to provide confirmation of chemical structure during in situ analysis. Continuous monitoring of air and solution samples is demonstrated, and a limit of detection of 50 ppb was obtained for toluene vapor in air and for an aqueous naphthalene solution using membrane sample introduction.

Abstract:Metal speciation analysis of microsamples in the clinical, biological and forensic fields is important for elucidating the metals' toxicity, mobility and metabolic behaviors of metals. Such analysis may be achieved by nanoliter high-performance liquid chromatography (nanoHPLC) hyphenated with inductively coupled plasma mass spectrometry (ICP-MS). In this work, an in-column high-pressure nebulizer (ICHPN) was developed to enhance the sensitivity and the separation efficiency of the sheathless nanoHPLC-ICP-MS coupling. The ICHPN consists of two concentric fused-silica capillaries with tapered tips connected via a PEEK tee, where C-18 silica particles with a size of 5 mu m were packed in the tapered inner capillary based on the keystone effect. Combining a heated single pass spray chamber with a makeup gas, the ICHPN was capable of independently optimizing the nebulization efficiency and the transport efficiency. The ICHPN offered high sensitivities, low detection limits and good precision at nanoflow rates. Compared with commercial nebulizers, the ICHPN fabrication was simple, rapid, reproducible and inexpensive. By implementing the ICHPN, we achieved rapid separation of four mercury species, i.e., Hg2+ and methyl-, ethyl- and phenylmercuric chloride, within 8.0 min with good resolution (2.0-13.9). Detection limits of 0.044-0.13 mu g L-1 were obtained with precisions of peak heights and areas ranging from 1.5 to 3.5% for a 50 mu g L-1 standard solution. Good agreement between the determined and certified values of mer- cury species in a certified reference material of caprine blood (SRM 955c) together with good recoveries (93-101%) validated the accuracy of the method. Abstract:The search for targets of FMRP (the product of FMR1, the mutated gene in Fragile X syndrome) has predominantly focused on identifying transcripts that are regulated by this RNA-binding protein. This study introduces the use of two-dimensional gel electrophoresis (2D PAGE) as a novel approach for demonstrating changes in protein synthesis secondary to FMRP deficit. By a standardized 2D PAGE protocol, we studied leukocyte homogenates from 30 males with different patterns of FMR1 mutation and different levels of FMRP. Samples from these subjects were compared to those of 12 normal control males and eight subjects with other mental retardation-associated conditions (i.e., Rett and Down syndromes). We found an abnormal pattern of a major leukocytic protein, identified by 2D PAGE datasets and immunoblotting as annexin-1 (Anx-1). Anx-1 appeared in subjects with Fragile X as multiple rather than 1-2 spots, at approximately 37 kd, in the pI 5-7 range. The presence and intensity of this Anx-1 pattern was relatively independent of Anx-1 levels and inversely related to total and high MW FMRP immunoreactivities. Based on the 2D PAGE pattern, without obvious MW change, and on dephosphorylation assays, we concluded that Anx-1's abnormality represents an aberrant posttranslational modification other than phosphorylation. Comparisons of our data with published cytoskeletal protein 2D profiles suggest that Anx-1 may be abnormally acetylated and, consequently, incapable of establishing appropriate N-terminal protein-protein interactions. In addition to its peripheral anti-inflammatory function, Anx-1 mediates glucocorticoid inhibition of the hypothalamo-pituitary-adrenal axis. As the latter seems to be disrupted in Fragile X syndrome, the reported Anx-1 abnormality could be responsible for some aspects of the Fragile X neurobehavioral phenotype. Our data also emphasize the feasibility of using 2D PAGE for disclosing molecular abnormalities in Fragile X and other genetic disorders.

Abstract:A compact ultrahigh-pressure nanoflow liquid chromatograph (LC) was developed with the purpose in mind of creating a portable system that could be easily moved to various testing locations or placed in close proximity to other instruments for optimal coupling, such as with mass spectrometry (MS). The system utilized innovative nanoflow pumps integrated with a very low volume stop-flow injector and mixing tee. The system weighed only 5.9 kg (13 lbs) or 4.5 kg (10 lbs) without a controller and could hold up to 1100 bar (16000 psi) of pressure. The total volume pump capacity was 60 μL. In this study, the sample injection volume was determined by either a 60 nL internal sample groove machined in a high-pressure valve rotor or by a 1 μL external sample loop, although other sample grooves or loops could be selected. The gradient dwell volume was approximately 640 nL, which allowed significant reduction in sample analysis time. Gradient performance was evaluated by determining the gradient step accuracy. A low RSD (0.6%, n = 4) was obtained for day-to-day experiments. Linear gradient reproducibility was evaluated by separating a three-component polycyclic aromatic hydrocarbon mixture on a commercial 150 μm inner diameter capillary column packed with 1.7 μm particles. Good retention-time reproducibility (RSD < 0.17%) demonstrated that the pumping system could successfully generate ultrahigh pressures for use in capillary LC. The system was successfully coupled to an LTQ Orbitrap MS in a simple and efficient way; LC-MS of a trypsin-digested bovine serum albumin (BSA) sample provided narrow peaks, short dwell time, and good peptide coverage.

Abstract:An electrospray/ultraviolet lamp dual-source ion trap mass spectrometer was developed for the rapid detection of gas and liquid samples. A discontinuous atmospheric pressure sampling system was used in the instrument, in which both the electrospray ions and gaseous analytes were sampled and transferred by a pinch valve device. The two ionization sources used here were generally suitable for different kinds of analytes and provide complementary applications. Electrospray was used for the ionization of polar compounds in solution, while the UV ionization source was mainly applied to the analysis of gaseous organic compounds. A variety of samples such as anisole, toluene, 2,4-dimethylaniline, arginine, reserpine and aspartame were employed to test the performance in different working mode of the instrument. The result showed that the two sources was suitable for ionization of different samples, and different types of molecular ions were generated when analyzing 2,4-dimethylaniline. Two ionization sources could be used alternately or simultaneously without interference with the other and the working mode could also be switched to fit the application requirements. Dual-source configuration was an effective way to extend the applications for miniature mass spectrometers. It did not significantly increase the size of the instrument, but provided more versatile analysis to meet the need for the measurement of different types of samples.

Abstract:A miniature mass spectrometry system with a sampling probe has been developed for real-time analysis of chemicals from sample surfaces. The sampling probe is 1.5 m in length and is comprised of one channel for introducing the spray and the other channel for transferring the charged species back to the Mini MS. This system provides a solution to the problem of real-time mass spectrometry analysis of a three-dimensional object in the field and is successful with compounds including those in inks, agrochemicals, explosives, and animal tissues. This system can be implemented in the form of a backpack MS with a sampling probe for forensic analysis or in the form of a compact MS with an intrasurgical probe for tissue analysis.

Abstract:Miniature mass spectrometry analytical systems of backpack configuration fitted with sampling probes could potentially be of significant interest for in-field, real-time chemical analysis. In this study, various configurations were explored in which a long narrow tube was used to connect the turbo and backing pumps used to create and maintain vacuum. Also, for the first time we introduced two new types of pumps for miniature mass spectrometers, the Creare 130 g drag pump and Creare 350 g scroll backing pump. These pumps, along with another Creare 550 turbo pump and the commercially available Pfeiffer HiPace 10 turbo and KnF diaphragm backing pumps, were tested with the backpack configurations. The system performance, especially the scan time, was characterized when used with a discontinuous atmospheric pressure interface (DAPI) for ion introduction. The pumping performance in the pressure region above 1 mtorr is critical for DAPI operation. The 550 g turbo pump was shown to have a relatively higher pumping speed above 1 mtorr and gave a scan time of 300 ms, almost half the value obtained with the larger, heavier HiPace 10 often used with miniature mass spectrometers. The 350 g scroll pump was also found to be an improvement over the diaphragm pumps generally used as backing pumps. With a coaxial low temperature plasma ion source, direct analysis of low volatility compounds glass slides was demonstrated, including 1 ng DNP (2,4-Dinitrophenol) and 10 ng TNT (2,4,6-trinitrotoluene) with Creare 550 g turbo pump as well as 10 ng cocaine and 20 ng DNP with Creare 130 g drag pump. Abstract:The first coupling of atmospheric pressure ionization methods, electrospray ionization (ESI) and desorption electrospray ionization (DESI), to a miniature hand-held mass spectrometer is reported. The instrument employs a rectilinear ion trap (RIT) mass analyzer and is battery-operated, hand-portable, and rugged (total system: 10 kg, 0.014 m(3), 75 W power consumption). The mass spectrometer was fitted with an atmospheric inlet, consisting of a 10 cm x 127 microm inner diameter stainless steel capillary tube which was used to introduce gas into the vacuum chamber at 13 mL/min. The operating pressure was 15 mTorr. Ions, generated by the atmospheric pressure ion source, were directed by the inlet along the axis of the ion trap, entering through an aperture in the dc-biased end plate, which was also operated as an ion gate. ESI and DESI sources were used to generate ions; ESI-MS analysis of an aqueous mixture of drugs yielded detection limits in the low parts-per-billion range. Signal response was linear over more than 3 orders of magnitude. Tandem mass spectrometry experiments were used to identify components of this mixture. ESI was also applied to the analysis of peptides and in this case multiply charged species were observed for compounds of molecular weight up to 1200 Da. Cocaine samples deposited or already present on different surfaces, including currency, were rapidly analyzed in situ by DESI. A geometry-independent version of the DESI ion source was also coupled to the miniature mass spectrometer. These results demonstrate that atmospheric pressure ionization can be implemented on simple portable mass spectrometry systems.
LC-Swan Probe: an Integrated In-Situ Sampling Interface for Liquid Chromatography Separation and Mass Spectrometry Analysis.

Abstract:In situ sampling mass spectrometry (MS) systems can achieve rapid analysis of samples, while most of them do not have the pretreatment capability of chromatographic separation. This Article describes the design, fabrication, and application of a swan-shaped in situ sampling MS probe with liquid chromatography (LC) separation capacity. The LC-Swan probe was fabricated based on a single capillary with a micrometer-sized hole at its U-shaped bottom for sampling, a monolithic column for separation, and a tapered tip for electrospray. Four functions including in situ sampling, sample injection, chromatographic separation, and MS electrospray were integrated in the LC-Swan probe. Direct sampling and contacting-dissolution-injection sampling modes were developed to perform in situ sampling and injection of liquid samples and dry spot samples, respectively, in the high flow-resistance LC system. A pressing-sealing method was also developed using a polydimethylsiloxane (PDMS) sealer to achieve the sealing of the probe sampling hole during the high-pressure chromatographic separation process. The LC-Swan probe-based system exhibited effective desalting capacity in the analysis of angiotensin H with similar relative standard deviations (RSDs) of retention time and peak area below 3% and 19% (n = 3) for both salt-containing and salt-free samples. The present system was applied for analyzing cytochrome C digest to test its separation capability for samples with complex compositions, and 19 peptides were detected in 13 min with an amino acid coverage of 85%. We also applied the system in metabolite analysis of mouse organ sections of brain, liver, and kidney to preliminarily demonstrate its application potential in MS imaging analysis.

Abstract:The present study reports on a streamlined analytical platform for rapid, straightforward, and on-site sample analysis using a miniature mass spectrometer with adequate tandem mass spectrometry (MS/MS) capability. An “all-in-one” workflow is developed combining accelerated air-assisted in-syringe extraction, sorbent-packed membrane clean-up, and needle spray ionization with an in-house built syringe assembly. A custom-made metal needle with a sharp, thin, and conical tip of micrometer-scale served as an integral component of the syringe assembly. This needle was capable of both aspirating a high-speed airflow for a highly efficient in-syringe extraction and performing stable electrospray for direct miniature mass spectrometry analysis. As a proof-of-concept demonstration, the developed protocol was implemented for the identification of 40 adulterated cosmetic ingredients and validated in terms of sensitivity, linearity, matrix effect, and recovery. A custom expandable mass spectral library was established in-house for smart suspect screening. Compound search and identification were conducted following a two-stage strategy using the full-scan MS and MS/MS data. The methodology demonstrates excellent potential for outside-the-laboratory chemical analysis by reducing the high time and labor costs associated with traditional sample preparation and chromatographic separation procedures, which would meet product quality and safety inspection requirements for on-site compliance check and regulatory enforcement at the testing sites such as supermarkets, department stores, wholesale markets, and roadside stalls.

Abstract:Ambient mass spectrometry (MS) has revolutionized the way of MS analysis and broadened its application in various fields. This paper describes the use of microfluidic techniques to simplify the setup and improve the functions of ambient MS by integrating the sampling probe, electrospray emitter probe, and online mixer on a single glass microchip. Two types of sampling probes, including a parallel-channel probe and a U-shaped channel probe, were designed for dry-spot and liquid-phase droplet samples, respectively. We demonstrated that the microfabrication techniques not only enhanced the capability of ambient MS methods in analysis of dry-spot samples on various surfaces, but also enabled new applications in the analysis of nanoliter-scale chemical reactions in an array of droplets. The versatility of the microchip-based ambient MS method was demonstrated in multiple different applications including evaluation of residual pesticide on fruit surfaces, sensitive analysis of low-ionizable analytes using postsampling derivatization, and high-throughput screening of Ugi-type multicomponent reactions.

Abstract:A newly developed portable capillary liquid chromatograph was investigated for the separation of various pharmaceutical and illicit drug compounds. The system consists of two high-pressure syringe pumps capable of delivering capillary-scale flow rates at pressures up to 10,000 psi. Capillary LC columns packed with sub-2 μm particles are housed in cartridges that can be inserted into the system and easily connected through high-pressure fluidic contact points by simply applying a specific, pre-determined torque rather than using standard fittings and less precise sealing protocols. Several over-the-counter analgesic drug separations are demonstrated, along with a simple on-line measurement of tablet dissolution. Twenty illicit drug compounds were also separated across six targeted drug panels. The results described in this study demonstrate the capability of this compact LC instrument to address several important drug-related applications while simplifying system operation, and greatly reducing solvent usage and waste generation essential for on-site analysis.
