Abstract:Scanning probe microscopy was used to simultaneously determine the molecular chain structure and intrinsic mechanical properties, including anisotropic elastic modulus and friction, for lamellae of highly oriented high-density polyethylene (HDPE) obtained by the melt-drawn method. The molecular-scale image of the highly oriented lamellae by friction force microscopy (FFM) clearly shows that the molecular chains are aligned parallel to the drawing direction, and the periodicities along and perpendicular to the drawing direction are 0.26 and 0.50 nm, respectively. The results indicate that the exposed planes of the lamellae resulting from the melt-drawn method are (200), which is consistent with results of transmission electron microscopy and electron diffraction. Because of the high degree of anisotropy in the sample, coming from alignment of the molecular chains along the drawing direction, the measured friction force, F, determined by FFM is strongly dependent on the angle, theta, between the scanning direction and the chain axis. The force increases as theta is increased from 0 degrees (i.e., parallel to the chain axis) to 90 degrees (i.e., perpendicular to the chain axis). The structural anisotropy was also found to strongly influence the measurements of the transverse chain modulus of the polymer by the nanoindentation technique. The measured value of 13.8 GPa with transverse modulus was larger than the value 4.3 GPa determined by wide-angle X-ray diffraction, which we attributed to anisotropic deformation of the lamellae during nanoindentation measurements that was not accounted for by the elastic treatment we adopted from Oliver and Pharr. The present approach using scanning probe microscopy has the advantage that direct correlations between the nanostructure, nanotribology, and nanomechanical properties of oriented samples can be determined simultaneously and simply.

Abstract:Two kinds of polyethylene chain aggregation with chain axis perpendicular and parallel to the supported substrate were designed and successfully obtained from melt under an electric field and by melt-drawn method, respectively. Their nanostructures were investigated by using transmission electron microscopy and atomic force microscopy. The chain modulus of the aggregations along and perpendicular to the chain axis were quantitatively measured to be 267 GPa and 13.8 GPa, respectively, by performing atomic force microscopy combining nanoindentation technique. This work provides new method to study polymer properties on the nanometer or molecular scale and insight into structure-property relationship on the nanometer scale for polymers, which was usually established based on micro-scale structure and macro-scale property.

Abstract:Nanoindentation technique and scanning force microscopy have been used to measure directly the polyethylene modulus along the chain axis. Single crystals of polyethylene were employed in order to obtain well-aligned chain segments. To minimize effects of scanner creep, a Z scan rate of 3Hz was employed. The “X Rotate” value of 25° was selected to eliminate effects of lateral tip motion. The results were analyzed by the Oliver–Pharr method for which direct observation and measurement of the contact area are not required. Considering the influence of tip roundness on the projected contact area, the nanoindentation results were analyzed by the Sawa method. The chain modulus obtained from the thinner polyethylene single crystal sample was 204±21GPa by the Oliver–Pharr method and 168±17GPa by the Sawa method. The lower values than expected were due to substrate effects and anisotropy of chain deformation during nanoindentation. An extrapolation of the chain modulus obtained by various strains to zero nanoindentation eliminated the effect of substrate and anisotropy of chain deformation. The corresponding chain modulus obtained from the thicker sample was 278GPa by the Oliver–Pharr method and 267GPa by the Sawa method, respectively, in better agreement with the value of 340GPa determined theoretically.

Abstract:The rigidity of peptide fibers is essential for their chemical and biological functions, despite that it remains largely unexplored. Here, we present the first direct measurement of the mechanics of individual fibers in peptide hydrogels by AFM imaging and statistical analysis and find that the intermolecular interactions play a considerable role.
Direct Measurement of Plowing Friction and Wear of a Polymer Thin Film Using the Atomic Force Microscope

Abstract:Nanometer-scale plowing friction and wear of a polycarbonate thin film were directly measured using an atomic force microscope (AFM) with nanoscratching capabilities. During the nanoscratch tests, lateral forces caused discrepancies between the maximum forces for the initial loading prior to the scratch and the unloading after the scratch. In the case of a nanoscratch test performed parallel to the cantilever probe axis, the plowing friction added another component to the moment acting at the cantilevered end compared to the case of nanoindentation, resulting in an increased deflection of the cantilever. Using free-body diagrams for the cases of nanoindentation and nanoscratch testing, the AFM force curves were analyzed to determine the plowing friction during nanoscratch testing. From the results of this analysis, the plowing friction was found to be proportional to the applied contact force, and the coefficient of plowing friction was measured to be 0.56 ± 0.02. Also, by the combination of nanoscratch and nanoindentation testing, the energetic wear rate of the polycarbonate thin film was measured to be 0.94 ± 0.05 mm3/(N m).

Abstract:We introduce nanoscale dynamic mechanical analysis (DMA) based on atomic force microscopy (AFM), a new mode for quantitative viscoelastic analysis of heterogeneous polymer materials at the nanoscale (AFM-nDMA). AFM-nDMA takes advantage of the exquisite force sensitivity, small contact radius, and nanoscale indentation depth of the AFM to provide dynamic mechanical analysis with 10 nm spatial resolution at rheologically relevant frequencies and variable temperature. This novel, non-resonant measurement is embedded in a force curve and typically occurs at a series of frequencies to provide spectra of storage modulus, loss modulus, and loss tangent. By using tailored probes and mitigating the effect of contact radius changes throughout the measurement, quantitative results are obtained that tie directly to bulk DMA and allow for time–temperature superposition analysis. The combination of quantitative results and 10 nm spatial resolution holds promise for the investigation of previously inaccessible microscopic domains, confinement effects, and interphases.

Abstract:Genetic analysis of natural variation in ecotypes of Arabidopsis thaliana can facilitate the discovery of new genes or of allelic variants of previously identified genes controlling physiological processes in plants. We mapped quantitative trait loci (QTL) for light response in recombinant inbred lines (RILs) derived from the Columbia and Kashmir accessions via two methods: composite interval mapping and eXtreme array mapping (XAM). After measuring seedling hypocotyl lengths in blue, red, far-red, and white light, and in darkness, eight QTL were identified by composite interval mapping and five localized near photoreceptor loci. Two QTL in blue light were associated with CRY1 and CRY2, two in red light were near PHYB and PHYC, and one in far-red light localized near PHYA. The RED2 and RED5 QTL were verified in segregating lines. XAM was tested for the identification of QTL in red light with pools of RILs selected for extreme phenotypes. Thousands of single feature polymorphisms detected by differential DNA hybridized to high-density oligo-nucleotide arrays were used to estimate allele frequency differences between the pools. The RED2 QTL was identified clearly; differences exceeded a threshold of significance determined by simulations. The sensitivities of XAM to population type and size and genetic models were also determined by simulation analysis.

Abstract:We quantitatively investigated the effect of iodine and pH on the stability of the helical structure formed by polyphenylacetylene derivatives using atomic force microscopy (AFM) based single-molecule force spectroscopy (SMFS) and circular dichroism (CD). The AFM measurement results showed that the addition of iodine into the system caused the decrease of the plateau heights in the stretching curves recorded during the SMFS experiments. In addition, the pH value increase can weaken the intra. polymer hydrogen bond and interrupt the integrity of the helical structure. However, the CD result did not show any detectable changes upon the addition of iodine and the change of pH value, which indicate that SMFS is a very sensitive and powerful method for subtle structure study.

Abstract:Here we demonstrate a simple, yet powerful method, atomic force microscopy (AFM) nanomechanical mapping, to directly visualize the interdiffusion and microstructure at the interface between two polymers. Nanomechanical measurements on the interface between poly(vinyl chloride) (PVC) and poly(caprolactone) (PCL) allow quantification of diffusion kinetics, observation of microstructure, and evaluation of mechanical properties of the interdiffusion regions. These results suggest that nanomechanical mapping of interdiffusion enables the quantification of diffusion with high resolution over large distances without the need of labeling and the assessment of mechanical property changes resulting from the interdiffusion.
Probing friction and adhesion properties of poly(vinyl methylether) homopolymer and blend films under nano-confinement using atomic-force microscopy

Abstract:The surface properties of poly(vinyl methylether) (PVME) homopolymer and blend films are characterized by atomic force microscopy (AFM) with the film thickness ranging from 5 nm to 400 nm. When the film thickness is less than 200 nm, lateral force of the PVME films decreases with the decreasing film thickness and has less dependence on the scanning rate. The local adhesion measurements indicate the adhesion force per unit area increases with the decreasing film thickness. These results are consistent to each other showing the increased polymer stiffness and decreased polymer chain mobility due to the confinement in the ultra-thin films. The effects of blending PVME with semicrystalline isotactic polypropylene (i-PP) on the lateral force of the film have also been investigated. Upon blending, both the scale of the lateral forces and their scanning rate dependences exhibit great changes. The lateral force of the blend film has a maximum when the fraction of PVME is around 0.8, similar to that when a polymer goes through the glass-transition. To cite this article: D. Wang, H. Abstract:Poly(N-isopropylacrylamide) (PNIPAM) is a highly versatile thermo-responsive hydrogel with immense potential for applications in biomedicine and tissue engineering. While PNIPAM has been extensively researched, there remains a significant gap in understanding its mechanical behavior at the atomistic scale. Addressing this challenge, our study employs molecular dynamics (MD) simulations to delve into the mechanical response of cross-linked PNIPAM hydrogels. The uniqueness of our research lies in our emphasis on providing atomic-level insights into the influence of chemical cross-linking and physical entanglements, as well as quantifying their effects during the application of strain. This represents a novel contribution to the study of hydrogels. Our MD simulations enable us to scrutinize the behavior of individual polymer chains and their intricate interactions in unprecedented detail, shedding light on microscale phenomena that are often inaccessible through experiments alone. To tackle the complexities associated with atomistic simulations of cross-linked hydrogels, we introduce a dynamic cross-linking algorithm. This innovative approach incorporates a nonreactive force field to construct realistic three-dimensional hydrogel microstructures, employing a stepwise bond formation strategy. Our findings underscore the remarkable impact of increasing the Degree of Cross-linking (DoC) and/or the Degree of Polymerization (DoP) on enhancing the robustness and elastic modulus of PNIPAM hydrogels. Additionally, we uncover the stochastic nature of chemical cross-linking, leading to the formation of anisotropic super-fragments when DoC exceeds a critical threshold. Furthermore, our research highlights the pivotal role of low strain rates on the order of 10 6 s −1 in accurately modeling the mechanical properties of hydrogels using MD simulations. This approach enables us to obtain meaningful quantitative mechanical properties that align with experimental results reported in the existing literature. In summary, our study offers an unprecedented level of insight into the intricate interplay of chemical cross-linking and physical entanglements, providing a foundation for future research in this domain.

Abstract:Polymeric materials are widely used in industries ranging from automotive to biomedical. Their mechanical properties play a crucial role in their application and function and arise from the nanoscale structures and interactions of their constitutive polymer molecules. Polymeric materials behave viscoelastically, i.e. their mechanical responses depend on the time scale of the measurements; quantifying these time-dependent rheological properties at the nanoscale is relevant to develop, for example, accurate models and simulations of those materials, which are needed for advanced industrial applications. In this paper, an atomic force microscopy (AFM) method based on the photothermal actuation of an AFM cantilever is developed to quantify the nanoscale loss tangent, storage modulus, and loss modulus of polymeric materials. The method is then validated on a styrene-butadiene rubber (SBR), demonstrating the method's ability to quantify nanoscale viscoelasticity over a continuous frequency range up to five orders of magnitude (0.2 Hz to 20,200 Hz). Furthermore, this method is combined with AFM viscoelastic mapping obtained with amplitude-modulation frequency-modulation (AM-FM) AFM, enabling the extension of viscoelastic quantification over an even broader frequency range, and demonstrating that the novel technique synergizes with preexisting AFM techniques for quantitative measurement of viscoelastic properties. The method presented here introduces a way to characterize the viscoelasticity of polymeric materials, and soft matter in general at the nanoscale, for any application.

Contactless Rheology of Soft Gels over a Broad Frequency Range

Abstract:We report contactless measurements of the viscoelastic rheological properties of soft gels. The experiments are performed using a colloidal-probe Atomic Force Microscope (AFM) in a liquid environment and in dynamic mode. The mechanical response is measured as a function of the liquid gap thickness for different oscillation frequencies. Our measurements reveal an elastohydrodynamic (EHD) coupling between the flow induced by the probe oscillation and the viscoelastic deformation of the gels. The data are quantitatively described by a viscoelastic lubrication model. The frequency-dependent storage and loss moduli of the polydimethylsiloxane (PDMS) gels are extracted from fits of the data to the model and are in good agreement with the Chasset--Thirion law. Our results demonstrate that contactless colloidal-probe methods are powerful tools that can be used for probing soft interfaces finely over a wide range of frequencies.

Nanoscale physicochemical properties of chain- and step-growth polymerized PEG hydrogels affect cell-material interactions

Abstract:Poly(ethylene glycol) (PEG) hydrogels provide a versatile platform to develop cell instructive materials through incorporation of a variety of cell adhesive ligands and degradable chemistries. Synthesis of PEG gels can be accomplished via two mechanisms: chain and step growth polymerizations. The mechanism dramatically impacts hydrogel nanostructure, whereby chain polymerized hydrogels are highly heterogeneous and step growth networks exhibit more uniform structures. Underpinning these alterations in nanostructure of chain polymerized hydrogels are densely-packed hydrophobic poly(methyl methacrylate) or poly(acrylate) kinetic chains between hydrophilic PEG crosslinkers. As cell-material interactions, such as those mediated by integrins, occur at the nanoscale and affect cell behavior, it is important to understand how different modes of polymerization translate into nanoscale mechanical and hydrophobic heterogeneities of hydrogels. Therefore, chain- and step-growth polymerized PEG hydrogels with macroscopically similar macromers and compliance (for example, methacrylate-functionalized PEG (PEGDM), MW = 10 kDa and norbornene-functionalized 4-arm PEG (PEGnorb), MW = 10 kDa) were used to examine potential nanoscale differences in hydrogel mechanics and hydrophobicity using atomic force microscopy (AFM). It was found that chain-growth polymerized network yielded greater heterogeneities in both stiffness and hydrophobicity as compared to step-growth polymerized networks. These nanoscale heterogeneities impact cell-material interactions, particularly human mesenchymal stem cell (hMSC) adhesion and spreading, which has implications in use of these hydrogels for tissue engineering applications. © 2017 Wiley Periodicals, Inc. Comparison of two case-based learning conditions with real patients in teaching occupational medicine

Abstract:Abstract Objective: To investigate the impact of three different training formats in occupational medicine (OM) on perceptions and performance of undergraduate students. Methods: A comparative study which included all fourth-year medical students was conducted over a three-year period. The year group in 2010 (211 students) received paper case studies followed by one small group session. The format used in 2011 actively engaged 188 students in the learning process by adding collaborative work and group discussions to the written information. In 2012, the approach comprised no longer constructed text cases but 212 students encountered real patients. Students’ perceptions were obtained by questionnaire. Their learning performance was assessed through review of written reports and score on oral presentations. Statistical differences in ratings were analyzed using Fisher’s exact and Kruskal–Wallis tests. Results: All three formats were found to equally achieve the stated learning objectives. The year groups with incorporation of active learning strategies and patient contacts had significant better test performance compared to those receiving only written case studies. Real patient students gave statistically significant higher rates for relevance, authenticity and appropriate difficulty level of the training than did students who discussed written case studies. Conclusion: Both approaches with augmented interaction in 2011 and 2012, improved performance and satisfaction among students. However, students valued the use of real patients higher than paper-form cases.

Abstract:The temperature dependence of nanomechanical properties of adsorbed poly-NIPAm microgel particles prepared by a semibatch polymerization process was investigated in an aqueous environment via indentation-based atomic force microscopy (AFM) methods. Poly-NIPAm microgel particles prepared by the classical batch process were also characterized for comparison. The local mechanical properties were measured between 26 and 35 °C, i.e., in the temperature range of the volume transition. Two different AFM tips with different shapes and end radii were utilized. The nanomechanical properties measured by the two kinds of tips showed a similar temperature dependence of the nanomechanical properties, but the actual values were found to depend on the size of the tip. The results suggest that the semibatch synthesis process results in the formation of more homogeneous microgel particles than the classical batch method. The methodological approach reported in this work is generally applicable to soft surface characterization in situ.This article has not yet been cited by other publications.

AFM Indentation and Material Property Identification of Soft Hydrogels

Abstract:There is growing evidence of the importance of mechanical deformations on various facets of cell functioning. This asks for a proper understanding of the cell’s characteristics as a mechanical system in different physiological and mechanical loading conditions. Many researchers use atomic force microscopy (AFM) indentation and the Hertz contact model for elastic material property identification under shallow indentation. For larger indentations, many of the Hertz assumptions are not inherently satisfied and the Hertz model is not directly useful for characterizing nonlinear elastic or inelastic material properties. We have used exponential hyperelastic material in FE simulations of the AFM indentation tests. A parameter identification approach is developed for hyperelastic material property determination from the simulated data. We collected AFM indentation data on agarose gel and developed a simple algorithm for contact point detection. The contact point correction improves the prediction of elastic modulus over the case of visual contact point identification. The modulus of 1% agarose gel was found to be about 15 kPa using the proposed correction, with mild but non-trival hardening with deeper indentation. The experimental data is compared with the results from the FE simulations and shows that over the hardening portion of the indentation response, our proposed parameter identification approach successfully captures the experimental data.
