Abstract:This article describes emulsification in a microfluidic double droplet generator (DDR) comprising two consecutive flow-focusing devices with locally modified surface chemistry. We generated W/O/W, O/O/W and O/W/O double emulsions with precisely controlled sizes and morphology of droplets. Secondly, by combining two mechanisms of droplet formation (the flow-focusing mechanism and the break up of liquid threads at T-junction) we produced multiple populations of droplets with varying size and/or composition. These droplets were used as the structural units for the formation of complex dynamic lattices.

Abstract:That vitamin A has a marked antikeratinizing effect has been established experimentally in animals and in humans. 1 It has also been shown that this effect may be produced by topical application. 2,3 In view of reports suggesting a favorable action of vitamin A given by injection in vulvar leukoplakia 4 and senile keratoses, 5 a clinical trial of this compound in topical form seemed indicated. Ten patients with typical oral leukoplakia of from 3 months' to 10 years' duration, ranging in severity from Grades II to IV of Thoma, 6 were treated with oral troches containing 150,000 units of vitamin A each. * The patients were instructed to remove their dentures and allow one troche to dissolve slowly in the mouth twice or three times daily. Treatment periods varied from two to six months. All patients were examined every four to six weeks, and photographs were taken for the record.
Novel glass capillary microfluidic devices for the flexible and simple production of multi-cored double emulsions

Abstract:Hypothesis: Double emulsions with many monodispersed internal droplets are required for the fabrication of multicompartment microcapsules and tissue-like synthetic materials. These double emulsions can also help to optically resolve different coalescence mechanisms contributing to double emulsion destabilization. Up to date microfluidic double emulsions are limited to either core-shell droplets or droplets with eight or less inner droplets. By applying a two-step jet break-up within one setup, double emulsion droplets filled with up to several hundred monodispersed inner droplets can be achieved. Experiments: Modular interconnected CNC-milled Lego®-inspired blocks were used to create two separated droplet break-up points within coaxial glass capillaries. Inner droplets were formed by countercurrent flow focusing within a small inner capillary, while outer droplets were formed by co-flow in an outer capillary. The size of inner and outer droplets was independently controlled since the two droplet break-up processes were decoupled. Findings: With the developed setup W/O/W and O/W/O double emulsions were produced with different surfactants, oils, and viscosity modifiers to encapsulate 25-400 inner droplets in each outer drop with a volume percentage of inner phase between 7% and 50%. From these emulsions monodispersed multicompartment microcapsules were obtained. The report offers insights on the relationship between the coalescence of internal droplets and their release.
A versatile and robust microfluidic device for capillary-sized simple or multiple emulsions production

Abstract:Ultrasound-vaporizable microdroplets can be exploited for targeted drug delivery. However, it requires customized microfluidic techniques able to produce monodisperse, capillary-sized and biocompatible multiple emulsions. Recent development of microfluidic devices led to the optimization of microdroplet production with high yields, low polydispersity and well-defined diameters. So far, only few were shown to be efficient for simple droplets or multiple emulsions production below 5 microns in diameter, which is required to prevent microembolism after intravenous injection. Here, we present a versatile microchip for both simple and multiple emulsion production. This parallelized system based on microchannel emulsification was designed to produce perfluorocarbon in water or water within perfluorocarbon in water emulsions with capillary sizes (<5 $\mu$m) and polydispersity index down to 5 % for in vivo applications such as spatiotemporally-triggered drug delivery using Ultrasound. We show that droplet production at this scale is mainly controlled by interfacial tension forces, how capillary and viscosity ratios influence droplet characteristics and how different production regimes may take place. The better understanding of droplet formation and its relation to applied pressures is supported by observations with a high-speed camera. Compared to previous microchips, this device opens perspectives to produce injectable and biocompatible droplets with a reasonable yield in order to realize preclinical studies in mice.
Manipulating The Generation Of Uniform O/W And W/O Emulsions Using Microfluidic T-Junction

Abstract:The purpose of this study is to generate monodisperse oil-in-water (O/W) and water-in-oil (W/O) emulsions using microfluidic T-junction. The materials of poly methyl methacrylate (PMMA) (hydrophobic) and glass (hydrophilic) were employed for microfluidic chip fabrication. Our strategy is based on the shear focus effect and sheath effect (focusing) to form uniform self-assembling sphere structures, the so-called O/W and W/O emulsions, in the T-junction microchannel. Results show that the size from 31 mu m to 43 mu m in diameter of O/W emulsions was generated by using the glass-based microfluidic chip. In addition, results show that the size from 70 mu m to 220 mu m of W/O emulsions was available by utilizing the PMMA-based microfluidic chip. These W/O emulsions were further applied to Ca-alginate microparticles (Ca-alg MPs) synthesis. The proposed microfluidic chips are capable of generating relatively uniform micro-droplets and have the advantages of actively controlling the droplet diameter, and having a simple and low cost process, with a high throughput.

Abstract:Profile switching of two-phase flows is often required in microfluidic systems. Manipulation of flow profiles can be realized by control of local surface energy of micro channel through wettability-patterning of channel surface. This article presents a facile approach for wettability-patterning of the micro channels of glass chips. Commercially available octadecyltrichlorosilane (OTS) was used to hydrophobilize the channels via the formation of OTS self-assembly monolayer (SAM), and a UV-source that mainly emits deep UV-light of 254 and 185nm was employed to degrade the in-channel formed OTS-SAM. The architecture of soda-lime glass/quartz glass hybrid chip was designed to facilitate the deep UV-light effective degrading the OTS-SAM. The established approach, together with the side-by-side laminar-flow patterning technique, was applied to prepare various finely patterned channel networks for different tasks of flow profile switching. The micro device capable of conducting the profile switch from W/O droplets to two separated continuous phases was demonstrated to perform on-chip quick liquid–liquid extraction for the determination of partition coefficients of pharmaceuticals.
Microfluidic synthesis of atto-liter scale double emulsions toward ultrafine hollow silica spheres with hierarchical pore networks

Abstract:A facile PDMS-glass hybrid microfluidic device is developed for generating uniform submicrometer-scale double emulsion droplets with unprecedented simplicity and controllability. Compared with planar flow-focusing geometries, our three-dimensional flow-focusing geometry is advantageous for stably producing femto- to atto-liter droplets without the retraction problem of the dispersed phase fluid. In addition, this microfluidic platform can withstand the use of strong organic solvents (e.g. tetrahydrofuran (THF) and toluene) as a dispersed phase without deforming PDMS devices because the dispersed phase containing organic solvents does not directly contact the PDMS wall. In particular, monodisperse double emulsions are generated spontaneously via the internal phase separation of single emulsions driven by the diffusion of a co-solvent (tetrahydrofuran) in microfluidic devices. Finally, we demonstrated that the double emulsions can be used as morphological templates of ultrafine spherical silica capsules with controlled hierarchical pore networks via the evaporation-induced self-assembly (EISA) method. During EISA, triblock copolymers (Pluronic F127) act as a surfactant barrier separating the internal droplet from the continuous oil phase, resulting in the 'inverse' morphology (i.e. hydrophobic polymer-in-water-in-oil emulsions). Depending on the precursor composition and kinetic condition, various structural and morphological features, such as mesoporous hollow silica spheres with a single central core, multi-cores, or a combination of these with robust controllability can be seen. Electron microscopy (SEM, STEM, HR-TEM), small angle X-ray scattering (SAXS), and N(2) adsorption-desorption confirm the well-controlled hierarchical pore structure of the resulting particles.

Abstract:Conventional droplet-based microfluidic systems require expensive, bulky external apparatuses, such as electric power supplies and pressure-driven pumps for fluid transportation. This study demonstrates an alternative way to produce emulsion droplets by autonomous fluid-handling based on the gas permeability of poly(dimethylsiloxane) (PDMS). Furthermore, basic concepts of fluid-handling are expanded to control the direction of the microfluid in the microfluidic device. The alternative pumping energy resulting from the high gas permeability of PDMS is used to generate water-in-oil (W/O) emulsions, which require no additional structures apart from microchannels. We can produce emulsion droplets by simple loading of the oil and aqueous solutions into the inlet reservoirs. During the operation of the microfluidic device, changes in droplet size, volumetric flow rate, and droplet generation frequency were quantitatively analyzed. As a result, we found that changes in the wetting properties of the microchannel greatly influence the volumetric flow rate and droplet generation frequency. This alternative microfluidic approach for preparing emulsion droplets in a simple and efficient manner is designed to improve the availability of emulsion droplets for point of care bioanalytical applications, in situ synthesis of materials, and on-site sample preparation tools.

Abstract:Aqueous microdroplets are of significant interest in biological fields, while the employment of water-in-oil (w/o) emulsion and lack of spatial control precluded its widespread application. Herein a novel microfluidic approach is developed to generate water-in-water (w/w) microdroplets embedded in hydrogel microfibers. Aqueous two phase system (ATPS) is applied to generate w/w droplets, and alginate is introduced to continuous phase to form microfibers, which offers spatial restriction and manipulation possibility to droplets. The size and pattern of aqueous droplets can be precisely controlled, and immobilization within hydrogel fiber facilitates easy manipulation and observation. The microdroplets surrounded by hydrophilic environment can act as a cell-cell interaction model, and their potential for biological and environmental applications are demonstrated by long-term culture of encapsulated cells and water remediation of Bacillus subtilis.

Abstract:Here we introduce a one-step microfluidic device for the controllable generation of gas/water/oil (G/W/O) double emulsions. The structure of the double emulsions can be tuned easily by the three phase flow rates and the dimensional range of the encapsulated microbubbles can be expanded by integrating CO2 adsorption. The G/W/O double emulsions were successfully in situ photo-polymerized to fabricate hollow hydrogel microspheres with tunable structures.

Abstract:The efficient generation of monodispersed droplets holds great promise for micro-/nanoparticle synthesis and biochemical analysis. However, it remains a challenge to achieve high-throughput generation of monodispersed droplets in a portable and/or parallel manner in nonexpert biomedical laboratories. Herein, a versatile microchannel array (mu CA) device is reported that is portable, multifaceted, reliable, and mass-manufacturable, supporting the high-throughput generation of monodispersed droplets in different ways. This device consists of a silicon-based mu CA chip based on the step emulsification principle, as well as two matching plastic containers, both of which can be mass-manufactured by traditional microfabrication methods at low material costs. With the mu CA device, aqueous solution can be dispersed into emulsion droplets by various modes, such as mechanical pump-based large-scale, handheld syringe-based portable, and gas pump-based highly parallel droplet generation. Furthermore, it is demonstrated that our cost-effective device can be applied for digital polymerase chain reaction analysis, supporting the need for more accessible microfluidic systems. Thus, the present device is expected to have a significant impact on both benchside and bedside applications. A versatile microchannel array device that is portable, multifaceted, reliable, and mass-manufacturable, supports the high-throughput generation of monodispersed droplets in different ways, which provides a versatile droplet platform for a myriad of different applications.image (c) 2024 WILEY-VCH GmbH

Abstract:Giant liposomes, or giant unilamellar vesicles (GUVs), have been utilized as cell-size bioreactors to replicate the physical and chemical properties of biological cells. However, conventional methods for preparing GUVs typically lack precise control over their size. Several research groups have recently developed microfluidic techniques to create monodisperse GUVs by generating water-in-oil-in-water (W/O/W) droplets with a thin oil layer that subsequently transform into GUVs. However, the formation of a thin oil shell requires the intricate control of the flow rate, which can be both challenging and unstable. In this study, we investigated the design of a two-step flow-focusing microfluidic channel to produce stable W/O/W droplets. These droplets underwent substantial oil layer reduction through spontaneous removal by fluidic shear forces. Consequently, the majority of the oil layer in the W/O/W droplets was reduced, improving uniformity of GUVs.

Abstract:Double emulsions are useful geometries as templates for core-shell particles, hollow sphere capsules, and for the production of biomedical delivery vehicles. In microfluidics, two approaches are currently being pursued for the preparation of microfluidic double emulsion devices. The first approach utilizes soft lithography, where many identical double-flow-focusing channel geometries are produced in a hydrophobic silicone matrix. This technique requires selective surface modification of the respective channel sections to facilitate alternating wetting conditions of the channel walls to obtain monodisperse double emulsion droplets. The second technique relies on tapered glass capillaries, which are coaxially aligned, so that double emulsions are produced after flow focusing of two co-flowing streams. This technique does not require surface modification of the capillaries, as only the continuous phase is in contact with the emulsifying orifice; however, these devices cannot be fabricated in a reproducible manner, which results in polydisperse double emulsion droplets, if these capillary devices were to be parallelized. Here, we present 3D printing as a means to generate four identical and parallelized capillary device architectures, which produce monodisperse double emulsions with droplet diameters in the range of 500 μm. We demonstrate high throughput synthesis of W/O/W and O/W/O double emulsions, without the need for time-consuming surface treatment of the 3D printed microfluidic device architecture. Finally, we show that we can apply this device platform to generate hollow sphere microgels.

Abstract:Microfluidic devices can form monodisperse double emulsions, but the fabrication steps are complicated and require specialized equipment. Recently, a method to convert single emulsions into double emulsions using vortex mixers has been proposed. Using this method, we demonstrate the production of double emulsions using commercially available single-emulsion microfluidic chips. We characterize the effect of vortex speed, vortex duration and the number of vortex/flick cycles on the average diameter and coefficient of variation of the double emulsions. Using fluorescent nanoparticles as tracers, we show that droplet breakup occurs during the second emulsification (using the vortex), but did not observe any fusion between the cores of double emulsion droplets. We also found that some inverted double emulsion droplets containing the outer water phase in their core were produced during vortex emulsification. Finally, while commercial chips only exist with a finite range of channel size that sets the monodispersed emulsion droplet radius, we show that the double-emulsion droplet radius can be adjusted using osmotic pumping. Our method is simple, available and user-friendly for biomedical researchers.

Abstract:In this work, we generated gas-in-water-in-oil (G/W/O) multi-cores double emulsions with a one-step microfluidic device and found a new stable flow pattern, where multi-cores double emulsions were easily fabricated. The mechanism of the particular flow pattern was discussed. Moreover, we systematically studied the effects of the three flow rates on the size and structure of the double emulsions. We then developed mathematical models to predict the size and structure of the double emulsions. On this basis, porous polyacrylamide hydrogel microspheres were successfully fabricated by in situ UV polymerization method. Abstract:This article presents a simple and novel approach to prepare monodispersed gas-in-oil-in-water (G/O/W) and gas-in-water-in-oil (G/W/O) double-emulsions in the same dual-coaxial microfluidic device. The effects of three phase flow rates on the sizes of microbubbles and droplets and the number of the encapsulated microbubbles were systematically studied. We successfully synthesized two different types of gas/liquid/liquid (G/L/L) double emulsions with different inner structures in the same geometry by adjusting the flow rates sequentially. Mathematical models were developed to predict the size and structures of the double emulsions. This simple approach gives a new idea for preparing hollow and porous microspheres with microbubbles as the direct core/pores templates.
Genetic variation in proinflammatory and anti-inflammatory cytokine production in multiple organ dysfunction syndrome*

Abstract:ObjectivesThe objectives of this study were to examine the prevalence of genetic variation for in vitro cytokine production (tumor necrosis factor [TNF]-&agr;, interleukin-10, transforming growth factor-&bgr;1) in patients with multiple organ dysfunction syndrome, to measure circulating cytokine levels and relate these to genotype, and to identify the relationship between genetic variation and outcome. DesignProspective analysis. SettingIntensive care unit of a university teaching hospital. PatientsEighty-eight critically ill patients with multiple organ dysfunction syndrome. Measurements and Main ResultsThe frequency of the different interleukin-10 genotypes (corresponding to high, intermediate, and low interleukin-10 production in vitro) were significantly different between controls and multiple organ dysfunction syndrome patients. High interleukin-10 producers were under-represented in the multiple organ dysfunction syndrome group: This genotype occurred in 30% of controls but in only 6% of patients (p < .001). There was no relationship between interleukin-10 genotype and mortality. The frequency of TNF-&agr; genotypes was also significantly different between patients and controls. Intermediate TNF-&agr; producers were under-represented (5.7% vs. 23%) and high TNF-&agr; producers over-represented (35.2% vs. 16%) in the patient group (p < .001). TNF-&agr; genotype was not related to mortality. The distribution of TNF-&bgr; genotypes (homozygous B1, homozygous B2, and heterozygotes) was also different between controls and patients (p = .008). The B2/B2 genotype (associated with high TNF-&agr; production) tended to occur less frequently in the intensive care unit population (31% vs. 50%) and was associated with a higher mortality rate than either the B1/B1 or B1/B2 genotypes (48% vs. 11% and 33% respectively, p = .115). The combination of proinflammatory (TNF-&agr;/TNF-&bgr;) and anti-inflammatory (interleukin-10/transforming growth factor-&bgr;1) cytokine genotypes was associated with prolonged patient survival time. Patients predisposed to produce a balanced cytokine response (e.g., intermediate interleukin-10/TNF-&agr; producers) demonstrated the longest survival times, although overall mortality was no different. ConclusionA genetic predisposition to high interleukin-10 production or intermediate TNF-&agr; production in vitro may be protective of admission to the intensive care unit, although once admitted, any protection provided by these genotypes seems to be lost. TNF-&bgr; genotype conferred no advantage to patients with multiple organ dysfunction syndrome, the TNFB2 allele being associated with increased mortality. The combination of proinflammatory and anti-inflammatory cytokine genotypes supports the idea that a balanced cytokine response is favorable and was associated with prolonged patient survival time.
Concentric Capillary Microfluidic Devices Designed for Robust Production of Multiple-Emulsion Droplets

Abstract:Multiple emulsions are used as templates for producing functional microcapsules due to their unique core-shell geometry. Employing glass capillary devices with coaxial channels has proven effective in creating uniform multiple-emulsion droplets. However, the use of partially miscible fluids, crucial for microcapsule production, often results in clogging and disrupts the stability of these devices. Here, we introduce innovative capillary microfluidic devices with concentric capillary channels, specifically designed to optimize the production of multiple-emulsion droplets while mitigating issues of precipitation and clogging. The key aspect of these devices is their configuration of two or three concentrically aligned capillaries, which form separate, coaxial microchannels for fluid injection. This unique alignment, achieved through rotational adjustments that leverage the natural off-center positioning of tapered capillaries, facilitates the simultaneous coaxial injection of various fluids into a droplet-forming junction, significantly reducing fluid contact before emulsification. The devices, featuring double and triple concentric capillary channels, consistently produce highly uniform double-, triple-, and quadruple-emulsion droplets with precisely controlled diameters and layer thicknesses. The minimal contact between fluids prior to emulsification in these devices broadens the usable range of fluid combinations, heralding new possibilities in microcapsule development for pharmaceutical and cosmetic applications.
