Abstract:Aedes albopictus (Skuse 1894) mosquitoes can transmit deadly arboviruses and are globally invasive due to their ability to survive in both tropical and temperate climates. Although adults cannot survive harsh winters, females are capable of anticipating seasonal change and producing overwintering diapause (DP) eggs that remain in a state of arrested development over the winter and hatch when favorable conditions return in the spring. While low temperatures can facilitate DP entry under short photoperiods, temperature signals alone are not sufficient to induce DP. To identify maternal phenotypes predictive of DP egg production in laboratory conditions, we characterized aspects of maternal physiology and behavior to identify those that correlate with DP egg production and changes in photoperiod, versus changes in temperature. Neither changes in temperature nor photoperiod impacted protein preference, blood meal consumption, or total number of eggs produced per female. Egg retention and oviposition timing were influenced by temperature, independent of DP egg production. However, females housed under short photoperiod conditions showed increased starvation resistance, despite showing similar levels of locomotor activity and internal stores of triacylglycerols, glucose, glycogen, and trehalose compared to females housed in long photoperiods. These results suggest that temperature and photoperiod differentially affect maternal phenotypes and identify starvation resistance as a maternal phenotype that is influenced by photoperiod and correlates with DP egg status. Aedes albopictus mosquitoes can survive in temperate climates because females are able to detect changes in temperature and day length in the autumn to produce "diapause" eggs that are provisioned with extra nutrients to survive harsh winters by remaining in a state of arrest and hatching in the spring. Although temperature and day length normally change in concert with each other throughout the year, studies have shown that day length is the most important cue used by female mosquitoes to initiate diapause egg production. In this study, the authors examined aspects of feeding and reproduction to determine which of these correlated with day length. Many aspects of reproduction were influenced by temperature; however, housing female mosquitoes under autumn-like short-day conditions led to increased starvation resistance. Surprisingly these females survive longer even though they do not appear to expend less energy compared to females housed in spring-like long-day conditions at the same temperature. This suggests that the mosquito mothers undergo changes in their metabolism that correlate with the production of DP eggs that can survive the winter. This new knowledge may lead to novel targets to disrupt seasonal reproduction in mosquitoes and limit the parts of the world where they can survive.

Thus, diapausing insects must also be able to survive these prewinter conditions which warm temperatures could make quite energetically taxing despite relative inactivity.We tested for both immediate and delayed fitness effects of prewinter conditions in diapausing Pieris napi butterfly pupae, experimentally exposing them to different prewinter treatments in a factorial design. We placed diapausing pupae at one of three temperatures (15, 20 and 25°C) for 1 to 16 weeks, followed by the same standardized winter for all individuals.We monitored survival of pupae at multiple points during the experiment, including after winter, as well as their change in mass. For a subset of individuals, we also made repeated metabolic measurements.We found substantial weight loss during prewinter warm periods, greater during longer prewinter treatments at higher temperatures. This weight loss was associated with elevated metabolic rates at higher temperatures which increased over the duration of the prewinter treatment.Although we found little prewinter mortality associated with these conditions, mortality was much greater post‐winter for individuals in long, warm prewinter treatments and the dry mass of adults that did survive these conditions was lower, highlighting the need to understand chronic or delayed effects of stress on fitness.Ultimately, we found substantial fitness consequences of prewinter conditions for a diapausing insect. Abstract:We show that developmental temperature impacts Aedes albopictus biology, including its fitness and its physiology. We further show that development at 32°C is perceived as a stress, while development at 18°C constrains Wolbachia proliferation. Overall, our data support local phenotypic thermal adaptation in Ae. albopictus. The increase of environmental temperature due to current global warming is not only favouring the expansion of the distribution range of many insect species, but it is also changing their phenology. Insect phenology is tightly linked to developmental timing, which is regulated by environmental temperatures. However, the degree to which the effects of developmental temperatures extend across developmental stages and their inter‐stage relationships have not been thoroughly quantified in mosquitoes. Here, we used the mosquito Aedes albopictus, which is an aggressive invasive species and an arboviral vector, to study how developmental temperature influences fitness across developmental stages, thermal traits, energy reserves, transcriptome and Wolbachia prevalence in laboratory‐reared populations originally collected from either temperate or tropical regions. We show that hatchability, larval and pupal viability and developmental speed are strongly influenced by temperature, and these effects extend to wing length, body mass, longevity and content of water, protein and lipids in adults in a population‐specific manner. On the contrary, neither adult thermal preference nor heat resistance significantly change with temperature. Wolbachia density was generally lower in adult mosquitoes reared at 18°C than at other tested temperatures, and transcriptome analysis showed enrichment for functions linked to stress responses (i.e. cuticle proteins and chitin, cytochrome p450 and heat shock proteins) in mosquitoes reared at both 18 and 32°C. Our data showed an overall reduced vector fitness performance when mosquitoes were reared at 32°C, and the absence of isomorphy in the relationship between developmental stages and temperature in the laboratory population deriving from larvae collected in northern Italy. Altogether, these results have important implications for reliable model projections of the invasion potentials of Ae. albopictus and its epidemiological impact.

Abstract:Abstract In temperate environments most species of insects enter an arrested state of development, known as diapause, that enables them to survive the adverse environmental conditions associated with winter. Although diapause is restricted to a single life stage within species of insects, there are examples of insects that overwinter in the egg, larval, pupal and adult stages. Here we offer a targeted, non-systematic literature review examining how overwintering impacts subsequent reproduction in female insects. Several factors, including the lifestage at which insects overwinter, the type of energy investment strategy females use for breeding, elements of the winter environment, and contributions from male insects can influence trade-offs that female insects face between overwintering survival and post-diapause reproduction. Additionally, climate change and elements of the urban environment, including light pollution and higher temperatures in cities, can exacerbate or ameliorate trade-offs faced by reproducing female insects. Better understanding trade-offs between overwintering survival and reproduction in insects not only enhances our understanding of the underlying physiological mechanisms and ecological processes governing diapause and reproduction, but also provide opportunities to better manage insect pests and/or support beneficial insects.

Effect of diapause on cold-resistance in different life-stages of an aphid parasitoid wasp

Abstract:To overwinter, insects from mild temperate areas can either enter diapause or remain active. Both strategies involve costs and benefits depending on the environment. In the first case, the emerging individuals will resist winter but have a reduced fitness because diapause entails physiological and ecological costs. In the second case, individuals need to be cold-resistant enough to withstand winter temperatures during their immature and adult stages, but could avoid diapause-associated costs. In mild temperate areas, the cost-benefit balance between the diapause and the non-diapause strategy would likely change in response to climate warming. A trade-off between these two strategies should lead to reduction of diapause expression in some populations. We explored the importance of such trade-off through the comparison of cold resistance capacities among different life stages in diapause and non-diapause individuals in a population of the aphid parasitoid Aphidius ervi (Hymenoptera: Braconidae) originating from western France where a decrease of diapause incidence was recently observed. As a proxy measure of insect physiological cold resistance, the Super Cooling Point (SCP) was determined for non-diapausing and diapausing prepupae and adults that went through prepupal diapause or not. Diapausing and non-diapausing prepupae were equally cold-resistant (-24.20 ±0.30°C vs. -24.74 ±0.36°C, respectively), and overall more resistant than adults. Adults that went through diapause as prepupae were less cold resistant (-17.85 ±1.10°C) than adults that have not undergone through diapause (-21.10 ±0.54°C). We also found that diapausing prepupae and adults that have undergone diapause were lighter than their non-diapausing counterparts, at a comparable size, suggesting higher energetic expenses during diapause. These results suggest a trade-off between diapause expression in prepupae and cold resistance and life-history-traits in adults. We conclude that selection could favor insects that do not enter diapause and thus avoid its associated costs while taking advantage at exploiting the mild winter environment.

Abstract:SUMMARY Insects are highly successful animals inhabiting marine, freshwater and terrestrial habitats from the equator to the poles. As a group, insects have limited ability to regulate their body temperature and have thus required a range of strategies to support life in thermally stressful environments, including behavioural avoidance through migration and seasonal changes in cold tolerance. With respect to overwintering strategies, insects have traditionally been divided into two main groups: freeze tolerant and freeze avoiding, although this simple classification is underpinned by a complex of interacting processes, i.e. synthesis of ice nucleating agents, cryoprotectants, antifreeze proteins and changes in membrane lipid composition. Also, in temperate and colder climates, the overwintering ability of many species is closely linked to the diapause state, which often increases cold tolerance ahead of temperature-induced seasonal acclimatisation. Importantly, even though most species can invoke one or both of these responses, the majority of insects die from the effects of cold rather than freezing. Most studies on the effects of a changing climate on insects have focused on processes that occur predominantly in summer (development, reproduction) and on changes in distributions rather than winter survival per se. For species that routinely experience cold stress, a general hypothesis would be that predicted temperature increases of 1°C to 5°C over the next 50-100 years would increase winter survival in some climatic zones. However, this is unlikely to be a universal effect. Negative impacts may occur if climate warming leads to a reduction or loss of winter snow cover in polar and sub-polar areas, resulting in exposure to more severe air temperatures, increasing frequency of freeze—thaw cycles and risks of ice encasement. Likewise, whilst the dominant diapause-inducing cue (photoperiod) will be unaffected by global climate change, higher temperatures may modify normal rates of development, leading to a decoupling of synchrony between diapause-sensitive life-cycle stages and critical photoperiods for diapause induction. In terms of climate warming and potential heat stress, the most recent predictions of summer temperatures in Europe of 40°C or higher in 50-75 years, are close to the current upper lethal limit of some insects. Long-term data sets on insect distributions and the timing of annual migrations provide strong evidence for ‘positive’ responses to higher winter temperatures over timescales of the past 20-50 years in North America, Europe and Asia.

Abstract:Spotted wing drosophila, Drosophila suzukii Matsumura, is a major pest of small fruit worldwide in temperate and subtropical growing regions. In Northern climates, D. suzukii likely overwinters locally under leaf litter and snow pack, but our understanding of the factors affecting thermal susceptibility is limited. While previous investigations of thermal susceptibility in this species have employed conventional static acclimation protocols, we aimed to determine whether gradual cooling, or dynamic acclimation, may extend the limits of known thermal tolerance by more closely approximating naturally occurring shifts in temperature. First, we assessed survival among adult and pupal D. suzukii using static acclimation. Then, we re-assessed survival using a novel dynamic acclimation method. We found that while static acclimation was sufficient to induce cold tolerance, dynamic acclimation significantly improved survival at temperatures as low as -7.5 °C. Following static acclimation, the lower lethal limit of adult D. suzukii was -1.1 °C in winter morphotype (WM) adults compared to 1.7 °C in non-acclimated summer morphotype (SM) adults. Dynamic acclimation reduced the lower limit to -5 °C in SM flies. At the end of our study 50% of WM flies survived 72 h at -7.5 °C. Below 0 °C pupal survival declined significantly regardless of acclimation procedure. However, pupal acclimation improved survival outcomes significantly compared to non-acclimated pupae, suggesting that while juvenile diapause is unlikely, cold hardening likely benefits those flies which may develop into the overwintering WM population. These data suggest that the degree of cold hardening is proportional to the thermal environment, a finding previously unrecognized in this species. Given the economic impact of this pest, these data may have important implications for offseason population monitoring and management. We discuss how phenotypic plasticity may drive geographical range expansion, and the impact of climate change on the spread of this species.
The long summer: pre-wintering temperatures affect metabolic expenditure and winter survival in a solitary bee

Abstract:The impact of climate change on insect populations depends on specific life cycle traits and physiological adaptations. The solitary bee Osmia lignaria winters as a pre-emergent adult, and requires a period of cold temperature for winter diapause completion. It is a univoltine species, and diapause induction does not depend on photoperiod. To understand the potential effects of longer summers on O. lignaria populations, we exposed individuals to three treatments simulating early, mid and late winter arrivals, and measured respiration rates, metabolic expenditure, weight loss, fat body depletion, lipid levels and winter mortality. The early-winter treatment disrupted diapause development, but had no apparent negative effects on fitness. In contrast, late-winter bees had a greater energetic expenditure (1.5-fold), weight (1.4-fold) and lipid (2-fold) loss, greater fat body depletion, and a 19% increase in mortality compared to mid-winter bees. We also monitored adult eclosion and arrival of winter temperatures under natural conditions in four years. We found a positive correlation between mean degree-day accumulation during pre-wintering (a measure of asynchrony between adult eclosion and winter arrival) and yearly winter mortality. Individually, bees experiencing greater degree-day accumulations exhibited reduced post-winter longevity. Timing of adult eclosion in O. lignaria is dependent on the duration of the prepupal period, which occurs in mid-summer, is also diapause-mediated, and is longer in populations from southerly latitudes. In a global warming scenario, we expect long summer diapause phenotypes to replace short summer diapause phenotypes, effectively maintaining short pre-wintering periods in spite of delayed winter arrivals.
Light Pollution Disrupts Seasonal Differences in the Daily Activity and Metabolic Profiles of the Northern House Mosquito, Culex pipiens

Abstract:The Northern House mosquito, Culex pipiens, is an important disease vector, and females are capable of surviving the winter in a state of overwintering diapause. This species' diapause response has been extensively studied, and recent evidence suggests that the circadian clock is involved in measuring seasonal changes in daylength to initiate the diapause response. However, differences in the circadian activity of diapausing and non-diapausing Cx. pipiens have not been thoroughly investigated. Additionally, recent findings indicate that artificial light at night (ALAN) can disrupt mosquito diapause, potentially prolonging the mosquito biting season. We compared the circadian locomotor activity of mosquitoes reared in diapause-averting, long-day conditions and diapause-inducing, short-day conditions with and without ALAN to elucidate the interplay between circadian activity, diapause, and light pollution. We also uncovered metabolic differences between mosquitoes reared under diapausing and non-diapausing photoperiods with and without ALAN by measuring the concentration of protein, fructose, glycogen, water-soluble carbohydrates, and lipids. We found that ALAN exposure altered several diapause-associated phenotypes including slightly, but not significantly, increasing activity levels in short day-reared mosquitoes; and preventing some short day-reared mosquitoes from accumulating lipids. ALAN also significantly reduced glycogen and water-soluble carbohydrate levels in long day-reared mosquitoes. Based on our findings, light pollution may decrease insect fitness by perturbing metabolism, and may also impact several phenotypes associated with insect diapause, potentially extending the mosquito biting season and preventing insects in urban environments from overwintering successfully.

Abstract:We have used recombinant human CD4 presented on beads as an affinity matrix to screen a 2'-F-pyrimidine-containing RNA library with a complexity of approximately 10(14) molecules. Affinity-selected aptamers bind recombinant CD4 with low nanomolar equilibrium dissociation constants. These high-affinity aptamers conjugated to different fluorophores such as fluorescein and phycoerythrin were used to stain cells, expressing human CD4 on cell surface, for analysis by flow cytometry. Aptamers, conjugated to fluorophores, stained mouse T cells that express human CD4 on the surface, but not the control mouse T cells lacking human CD4. The control cells, however, do express mouse CD4 whose extracellular domain has 55% sequence identity to the human form. These human CD4-specific aptamers selectively stained CD4(+) T cells in a preparation of human peripheral blood mononuclear cells. These results and others suggest that aptamers are emerging as a versatile class of molecules that can be used for various diagnostic applications performed under different formats or platforms.

Abstract:Background: The mosquito Aedes albopitus is a competent vector for the transmission of many blood-borne pathogens. An important factor that affects the mosquitoes' development and spreading is climate, such as temperature, precipitation and photoperiod. Existing climate-driven mechanistic models overlook the seasonal pattern of diapause, referred to as the survival strategy of mosquito eggs being dormant and unable to hatch under extreme weather. With respect to diapause, several issues remain unaddressed, including identifying the time when diapause eggs are laid and hatched under different climatic conditions, demarcating the thresholds of diapause and non-diapause periods, and considering the mortality rate of diapause eggs. Methods: Here we propose a generic climate-driven mechanistic population model of Ae. albopitus applicable to most Ae. albopictus-colonized areas. The new model is an improvement over the previous work by incorporating the diapause behaviors with many modifications to the stage-specific mechanism of the mosquitoes' life-cycle. monthly Container Index (CI) of Ae. albopitus collected in two Chinese cities, Guangzhou and Shanghai is used for model validation. Results: The simulation results by the proposed model is validated with entomological field data by the Pearson correlation coefficient r (2) in Guangzhou (r (2) = 0.84) and in Shanghai (r (2) = 0.90). In addition, by consolidating the effect of diapause-related adjustments and temperature-related parameters in the model, the improvement is significant over the basic model. Conclusions: The model highlights the importance of considering diapause in simulating Ae. albopitus population. It also corroborates that temperature and photoperiod are significant in affecting the population dynamics of the mosquito. By refining the relationship between Ae. albopitus population and climatic factors, the model serves to establish a mechanistic relation to the growth and decline of the species. Understanding this relationship in a better way will benefit studying the transmission and the spatiotemporal distribution of mosquito-borne epidemics and eventually facilitating the early warning and control of the diseases.
Phenotypical aspects of Culex pipiens biotype pipiens during diapause: Lipid utilization, body size, insemination, and parity

Abstract:In temperate regions, female Culex pipiens biotype pipiens mosquitoes undergo diapause in winter. Diapausing biotype pipiens mosquitoes are potentially important winter reservoirs for mosquito-borne viruses, such as West Nile virus (WNV), Usutu virus (USUV), and Sindbis virus (SINV). Mosquitoes in diapause have not taken a bloodmeal prior to winter. Therefore, they do not become infected by taking an infectious bloodmeal and as a consequence, vertical transmission is considered the primary mechanism of mosquito-borne virus overwintering. Prior to winter, biotype pipiens mosquitoes build up fat reserves, which they utilize throughout winter. Furthermore, earlier studies have indicated that larger body size is correlated with increased survival during winter. However, not much is known about lipid utilization and body size of wild biotype pipiens mosquitoes in diapause. Therefore, we performed monthly collections of diapausing biotype pipiens mosquitoes in two consecutive winters (2020/2021 and 2021/2022) in bunkers of the New Hollandic Waterline in the Netherlands. Every month, we checked the proportion of inseminated and parous females via microscopy. In addition, we measured wing length as proxy for body size, and assessed total lipid content. Furthermore, we monitored indoor temperature in the overwintering locations. We show that the overwintering sites in our study provide relatively stable environments, in which temperatures rarely drop below 0 °C. The vast majority of biotype pipiens females were inseminated (84.1 %) and nulliparous (97.5 %). We detected differences in body size between but not within the two years of sampling. Additionally, we detected a difference in lipid content between the sampling years. We confirm that the vast majority of diapausing biotype pipiens females are inseminated and nulliparous. This indicates that they did not blood feed prior to winter, which underscores the likeliness of vertical transmission being the primary mechanism behind virus overwintering. The detected difference in body size between years can most likely be attributed to differences in summer conditions the mosquitoes were exposed to as larvae, although this needs confirmation. The difference in lipid depletion could not be explained by differences in climatic conditions. To shed more light on the links between climatic conditions, body size, lipid depletion and the consequences for mosquito population dynamics and arbovirus transmission, future experimental work, for example by arbovirus exposure followed by artificially induced diapause, is desired.
Effects of overwintering on the transcriptome and fitness traits in a damselfly with variable voltinism across two latitudes

Abstract:Winter diapause consists of cessation of development that allows individuals to survive unfavourable conditions. Winter diapause may bear various costs and questions have been raised about the evolutionary mechanisms maintaining facultative diapause. Here, we explored to what extent a facultative winter diapause affects life-history traits and the transcriptome in the damselfly Ischnura elegans, and whether these effects were latitude-specific. We collected adult females at central and high latitudes and raised their larvae in growth chambers. Larvae were split into a non-diapausing and post-winter (diapausing) cohort, were phenotyped and collected for a gene expression analysis. At the phenotypic level, we found no difference in survival between the two cohorts, and the post-winter cohort was larger and heavier than the non-winter cohort. These effects were mostly independent of the latitude of origin. At the transcriptomic level, wintering affected gene expression with a small fraction of genes significantly overlapping across latitudes, especially those related to morphogenesis. In conclusion, we found clear effects of diapause on the phenotype but little evidence for latitudinal-specific effects of diapause. Our results showed a shared transcriptomic basis underpinning diapause demonstrated, here, at the intraspecific level and supported the idea of evolutionary convergence of the response to diapause across organisms.

Prolonged diapause has sex-specific fertility and fitness costs

Abstract:Abstract Diapause in seasonal environments allows insects to survive adverse seasons. However, individuals can sometimes enter a prolonged diapause for more than a year, and also skip favourable seasons, which can bring additional costs through e.g. loss of metabolic resources. At the same time, prolonged diapause can be beneficial if it allows individuals to have a risk-spreading strategy to skip potentially suboptimal breeding seasons. We studied if prolonged diapause (2-year diapause) negatively affects the fertility and fitness of female and male Colorado potato beetles ( Leptinotarsa decemlineata ) compared to control (1-year diapause) beetles. We also tested the parental effects on the subsequent chemical stress tolerance of their offspring. We found that prolonged diapause carried fertility costs only for females who were less fertile than the control females. However, no differences in fertility were observed in males. Furthermore, prolonged diapause in females resulted in offspring with lower larvae-to-adult survival even though these offspring had accelerated development times. In contrast, paternal diapause duration had no effects on their offspring larvae-to adult survival, but prolonged diapause males sired offspring with slower development times than control males. Perhaps to compensate the costs related to prolonged diapause both older parents produced or sired offspring with higher body mass than control parents. Despite the differences in emergence mass, parental diapause duration did not affect offspring insecticide stress tolerance. The difference between females and males most likely results from the observed differences in prolonged diapause females’ capacity to fight against cellular oxidative damage which was poorer compared to the control females. Even though prolonged diapause allows individuals to have a risk-spreading strategy it carries sex-specific fertility and fitness costs indicating that selection could favour this in males but not in females.

Phenotypic plasticity of a winter-diapause mechanism copes with the effects of summer global warming in an ectothermic predator

Abstract:To adapt to changes in temperature, animals tend to invest more energy in thermal tolerance to enhance survival, which can have simultaneous costs on plastic traits. Would a decrease in genetic variability, due to global warming, affect the ability of populations with existing metabolic regulatory mechanisms to cope with extreme temperatures? To address this question, we conducted a series of experiments based on the A1B scenario of global warming, assessing within-population genetic variance in (a) morphological traits, (b) metabolic rate allometries, and (c) survival of a winter-diapausing predator ectotherm. Our study focused on the lacewing species Chrysoperla pallida , using both exogamic and endogamic artificial genetic lines. We discovered that both lines use their winter-diapausing phenotype to adapt to summer extreme temperatures caused by extreme heating conditions, but the exogamic line is prone to express phenotypic plasticity in metabolic scaling, with a trade-off between body size and mandible size, i.e. larger individuals tended to develop smaller mandibles to better survive. These findings highlight the significance of substantial phenotypic plasticity and pre-existing metabolic regulatory mechanisms in enabling ectotherms to cope with potential extreme heating occurring in global warming.

Abstract:Anthropogenic challenges, particularly climate change-associated factors, are strongly impacting the behavior, distribution, and survival of insects. Yet how these changes affect pests such as Drosophila suzukii, a cosmopolitan pest of soft-skinned small fruits, remains poorly understood. This polyphagous pest is chill-susceptible, with cold temperatures causing multiple stresses, including desiccation and starvation, also challenging the immune system. Since the invasion of Europe and the United States of America in 2009, it has been rapidly spreading to several European and American countries (both North and South American) and North African and Asian countries. However, globalization and global warming are allowing an altitudinal and latitudinal expansion of the species, and thus the colonization of colder regions. This review explores how D. suzukii adapts to survive during cold seasons. We focus on overwintering strategies of behavioral adaptations such as migration or sheltering, seasonal polyphenism, reproductive adaptations, as well as metabolic and transcriptomic changes in response to cold. Finally, we discuss how the continuation of climate change may promote the ability of this species to survive and spread, and what mitigation measures could be employed to overcome cold-adapted D. suzukii.

Abstract:Our findings challenge the positive association between body weight and diapause (dormancy) phenotypes in the model flesh fly, Sarcophaga crassipalpis. Nutrient limitation alters the relationship between body weight and diapause phenotypes, suggesting that the relationships between body‐weight and phenotypes need to be considered within ecological contexts. During the winter, animals face limited food availability. Many animals enter dormancy to reduce their winter energy expenditure. Most insects spend the winter in diapause, a state of programmed dormancy. It is often assumed that diapausing insects need nutrient stores to fuel their many months of basal metabolism and must grow heavier than their non‐diapause‐programmed counterparts. However, the extent to which food limitation affects body weight during overwintering preparation as well as the likelihood and duration of diapause remains unclear. We limited the duration of the feeding period and thus the total quantity of food available to diapause‐destined larvae of the pupal‐diapausing flesh fly, Sarcophaga crassipalpis, to test how food limitation affects body weight in the context of diapause programming. We also tested the extent to which food deprivation and body weight affect the likelihood and duration of diapause. We hypothesized that diapause‐destined larvae grow more quickly and pupariate at a heavier body weight than non‐diapause larvae. We also hypothesized that body weight is more dramatically reduced by food limitations when a larva is programmed for diapause. Finally, we hypothesized that larvae with lighter body weight (i.e., food limited) are less likely to enter pupal diapause and also stay in diapause for a shorter duration than heavier, well‐fed, individuals. Contrary to our hypotheses that diapausing insects are heavier than their non‐diapausing counterparts, we found diapausing pupae weighed less than non‐diapausing pupae, especially when larvae received limited food. We found light pupae did not abort their diapause program. In both diapausing and non‐diapausing pupae, body weight was positively correlated with simulated winter survival. However, above a weight threshold, body weight no longer affected simulated winter survival in diapausing pupae. Contrary to our predictions and the general consensus in much of the diapause literature, we also found that lighter pupae stayed in diapause longer than heavier pupae. Overall, our results challenge the precept that body weight and diapause are positively associated. The relationship between body weight and diapause is complex and may be affected by the availability of food before and after winter, the availability of high‐quality overwintering sites, and the life history of a particular insect.

Abstract:1 The aim of this study was to investigate the effect of various antagonists, selective for the tachykinin NK1 or NK2 receptor, on the atropine‐resistant ascending excitatory reflex (AER) to the circular muscle of the guinea‐pig ileum elicited by radial stretch (balloon distension) or electrical field stimulation. 2 Submaximal and maximal atropine‐ (1 μm) resistant AER elicited by balloon distension averaged about 40–50% and 70–90% of maximal circular spasm to 80 mm KCl, respectively. The NK1 receptor antagonist, (±)‐CP 96,345 (1 μm) inhibited both maximal and submaximal AER. FK 888 (1–3 μm) inhibited submaximal AER only. RP 67,580 (1 μm) was ineffective. The NK2 receptor antagonist, GR 94,800, inhibited both maximal and submaximal AER at all concentrations tested (0.1–3.0 μm), while SR 48,968 was effective only at 1.0 μm. The NK2 receptor antagonists, MEN 10,376 and MEN 10,573 inhibited both submaximal and maximal AER at 10 and 1.0 μm, respectively. 3 In other experiments, an NK1 receptor antagonist, (±)‐CP 96,345 or FK 888 (1.0 μm in each case) was administered first and the effect of GR 94,800 (1.0 μm) on the residual AER response was determined; or GR 94,800 was administered first and the effect of (±)‐CP 96,345 or FK 888 was determined. The results of these experiments indicated an additive effect produced by the combined treatment with NK1 and NK2 receptor antagonists. 4 Electrical field stimulation (10 Hz for 0.5 s, 10–20 V, 0.15–0.3 ms pulse width) with electrodes placed at 1.4–1.8 cm anal to the recording site, produced ascending contractions which were almost abolished by 10 μm hexamethonium (electrically‐evoked AER). In the presence of apamin (0.1 μm) and NG‐nitro‐l‐arginine (30 μm) these contractions were reproducible over 10 consecutive stimulation cycles. GR 94,800 (1 μm) and FK 888 (1 μm) both produced a partial inhibition of the electrically‐evoked AER and their combined administration produced an inhibitory effect which was larger than that induced by each antagonist alone. 5 FK 888 (1–3 μm), GR 94,800 (1–3 μm), MEN 10,573 (1 μm) and MEN 10,376 (10 μm) did not significantly affect the atropine‐sensitive twitch contractions produced by electrical field stimulation of the guinea‐pig ileum longitudinal muscle‐myenteric plexus preparation, which were abolished by 10–30 μm procaine, 1 μm tetrodotoxin or 1 μm atropine. (±)‐CP 96,345 (1 μm) and SR 48,968 (1 μm) produced 12% and 27% inhibition of cholinergic twitches in the longitudinal muscle of the ileum, respectively. 6 We conclude that both NK1 and NK2 receptors mediate the atropine‐resistant AER to the circular muscle of the ileum. NK2 receptor activation plays a more important role than NK1 receptor activation in the AER evoked by radial stretch. Since a consistent fraction of the distension‐ and electrically‐evoked atropine‐resistant AER persists in the presence of combined NK1 and NK2 receptor blockade, the existence of a third excitatory transmitter to the circular muscle of the ileum, in addition to acetylcholine and tachykinins, is suggested.
Postdiapause reproduction of spotted-wing drosophila (Diptera: Drosophilidae) in realistically simulated cold climatic springtime conditions of Québec, Canada

Abstract:Abstract Knowing the biology and ecology of Drosophila suzukii (Diptera: Drosophilidae) is important to predict its potential for permanent establishment in cold climatic regions. We studied the reproductive potential of postdiapause male and female flies collected in lowbush blueberry fields in late season over two years. After being submitted to a six-month cold-winter regime, survivors were exposed to a simulated springtime warming regime to monitor reproductive potential and expected population growth. Overwintered fly survivors of both sexes that were aged up to nine months after field capture reproduced successfully in spring and early summer when mated to nondiapause flies experimentally made available for mating as potential migrant colonisers from lower latitudes. Relative to control nondiapause flies, the lifetime fertility of postdiapause flies was reduced by 84% in females and 70% in males, revealing large fitness costs of reproductive diapause under cold stress. Modelling potential reproduction of overwintered flies in warming conditions using models parametrised for age-dependent fertility rate and diapause status showed that nondiapause migrant flies would be strongly advantaged over locally overwintered postdiapause flies due to precocious reproductive maturity, broader pattern of age-specific fertility, and higher lifetime fertility.
