Abstract:Virtually all temperate insects survive the winter by entering a physiological state of reduced metabolic activity termed diapause. However, there is increasing evidence that climate change is disrupting the diapause response resulting in non-diapause life stages encountering periods of winter cold. This is a significant problem for adult life stages in particular, as they must remain mobile, periodically feed, and potentially initiate reproductive development at a time when resources should be diverted to enhance stress tolerance. Here we present the first evidence of protein/meat feeding restricting rapid cold hardening (RCH) ability and increasing low temperature activity thresholds. No RCH response was noted in adult female blow flies (Calliphora vicina Robineau-Desvoidy) fed a sugar, water and liver (SWL) diet, while a strong RCH response was seen in females fed a diet of sugar and water (SW) only. The RCH response in SW flies was induced at temperatures as high as 10°C, but was strongest following 3h at 0°C. The CTmin (loss of coordinated movement) and chill coma (final appendage twitch) temperature of SWL females (-0.3 ± 0.5°C and -4.9 ± 0.5°C, respectively) was significantly higher than for SW females (-3.2 ± 0.8°C and -8.5 ± 0.6°C). We confirmed this was not directly the result of altered extracellular K+, as activity thresholds of alanine-fed adults were not significantly different from SW flies. Instead we suggest the loss of cold tolerance is more likely the result of diverting resource allocation to egg development. Between 2009 and 2013 winter air temperatures in Birmingham, UK, fell below the CTmin of SW and SWL flies on 63 and 195 days, respectively, suggesting differential exposure to chill injury depending on whether adults had access to meat or not. We conclude that disruption of diapause could significantly impact on winter survival through loss of synchrony in the timing of active feeding and reproductive development with favourable temperature conditions.
Phenotypic Plasticity Promotes Overwintering Survival in A Globally Invasive Crop Pest, Drosophila suzukii

Abstract:Spotted wing drosophila, Drosophila suzukii Matsumura, is a major pest of small fruit worldwide in temperate and subtropical growing regions. In Northern climates, D. suzukii likely overwinters locally under leaf litter and snow pack, but our understanding of the factors affecting thermal susceptibility is limited. While previous investigations of thermal susceptibility in this species have employed conventional static acclimation protocols, we aimed to determine whether gradual cooling, or dynamic acclimation, may extend the limits of known thermal tolerance by more closely approximating naturally occurring shifts in temperature. First, we assessed survival among adult and pupal D. suzukii using static acclimation. Then, we re-assessed survival using a novel dynamic acclimation method. We found that while static acclimation was sufficient to induce cold tolerance, dynamic acclimation significantly improved survival at temperatures as low as -7.5 °C. Following static acclimation, the lower lethal limit of adult D. suzukii was -1.1 °C in winter morphotype (WM) adults compared to 1.7 °C in non-acclimated summer morphotype (SM) adults. Dynamic acclimation reduced the lower limit to -5 °C in SM flies. At the end of our study 50% of WM flies survived 72 h at -7.5 °C. Below 0 °C pupal survival declined significantly regardless of acclimation procedure. However, pupal acclimation improved survival outcomes significantly compared to non-acclimated pupae, suggesting that while juvenile diapause is unlikely, cold hardening likely benefits those flies which may develop into the overwintering WM population. These data suggest that the degree of cold hardening is proportional to the thermal environment, a finding previously unrecognized in this species. Given the economic impact of this pest, these data may have important implications for offseason population monitoring and management. We discuss how phenotypic plasticity may drive geographical range expansion, and the impact of climate change on the spread of this species.
Effect of diapause on cold-resistance in different life-stages of an aphid parasitoid wasp

Abstract:To overwinter, insects from mild temperate areas can either enter diapause or remain active. Both strategies involve costs and benefits depending on the environment. In the first case, the emerging individuals will resist winter but have a reduced fitness because diapause entails physiological and ecological costs. In the second case, individuals need to be cold-resistant enough to withstand winter temperatures during their immature and adult stages, but could avoid diapause-associated costs. In mild temperate areas, the cost-benefit balance between the diapause and the non-diapause strategy would likely change in response to climate warming. A trade-off between these two strategies should lead to reduction of diapause expression in some populations. We explored the importance of such trade-off through the comparison of cold resistance capacities among different life stages in diapause and non-diapause individuals in a population of the aphid parasitoid Aphidius ervi (Hymenoptera: Braconidae) originating from western France where a decrease of diapause incidence was recently observed. As a proxy measure of insect physiological cold resistance, the Super Cooling Point (SCP) was determined for non-diapausing and diapausing prepupae and adults that went through prepupal diapause or not. Diapausing and non-diapausing prepupae were equally cold-resistant (-24.20 ±0.30°C vs. -24.74 ±0.36°C, respectively), and overall more resistant than adults. Adults that went through diapause as prepupae were less cold resistant (-17.85 ±1.10°C) than adults that have not undergone through diapause (-21.10 ±0.54°C). We also found that diapausing prepupae and adults that have undergone diapause were lighter than their non-diapausing counterparts, at a comparable size, suggesting higher energetic expenses during diapause. These results suggest a trade-off between diapause expression in prepupae and cold resistance and life-history-traits in adults. We conclude that selection could favor insects that do not enter diapause and thus avoid its associated costs while taking advantage at exploiting the mild winter environment.

Abstract:To adapt to changes in temperature, animals tend to invest more energy in thermal tolerance to enhance survival, which can have simultaneous costs on plastic traits. Would a decrease in genetic variability, due to global warming, affect the ability of populations with existing metabolic regulatory mechanisms to cope with extreme temperatures? To address this question, we conducted a series of experiments based on the A1B scenario of global warming, assessing within-population genetic variance in (a) morphological traits, (b) metabolic rate allometries, and (c) survival of a winter-diapausing predator ectotherm. Our study focused on the lacewing species Chrysoperla pallida , using both exogamic and endogamic artificial genetic lines. We discovered that both lines use their winter-diapausing phenotype to adapt to summer extreme temperatures caused by extreme heating conditions, but the exogamic line is prone to express phenotypic plasticity in metabolic scaling, with a trade-off between body size and mandible size, i.e. larger individuals tended to develop smaller mandibles to better survive. These findings highlight the significance of substantial phenotypic plasticity and pre-existing metabolic regulatory mechanisms in enabling ectotherms to cope with potential extreme heating occurring in global warming.

Abstract:The accumulation of trehalose has been suggested as a mechanism underlying insect cross-tolerance to cold/freezing and drought. Here we show that exposing diapausing larvae of the drosophilid fly, Chymomyza costata to dry conditions significantly stimulates their freeze tolerance. It does not, however, improve their tolerance to desiccation, nor does it significantly affect trehalose concentrations. Next, we use metabolomics to compare the complex alterations to intermediary metabolism pathways in response to three environmental factors with different ecological meanings: environmental drought (an environmental stressor causing mortality), decreasing ambient temperatures (an acclimation stimulus for improvement of cold hardiness), and short days (an environmental signal inducing diapause). We show that all three factors trigger qualitatively similar metabolic rearrangement and a similar phenotypic outcome—improved larval freeze tolerance. The similarities in metabolic response include (but are not restricted to) the accumulation of typical compatible solutes and the accumulation of energy-rich molecules (phosphagens). Based on these results, we suggest that transition to metabolic suppression (a state in which chemical energy demand is relatively low but need for stabilization of macromolecules is high) represents a common axis of metabolic pathway reorganization towards accumulation of non-toxic cytoprotective compounds, which in turn stimulates larval freeze tolerance.

Abstract:SUMMARY Insects are highly successful animals inhabiting marine, freshwater and terrestrial habitats from the equator to the poles. As a group, insects have limited ability to regulate their body temperature and have thus required a range of strategies to support life in thermally stressful environments, including behavioural avoidance through migration and seasonal changes in cold tolerance. With respect to overwintering strategies, insects have traditionally been divided into two main groups: freeze tolerant and freeze avoiding, although this simple classification is underpinned by a complex of interacting processes, i.e. synthesis of ice nucleating agents, cryoprotectants, antifreeze proteins and changes in membrane lipid composition. Also, in temperate and colder climates, the overwintering ability of many species is closely linked to the diapause state, which often increases cold tolerance ahead of temperature-induced seasonal acclimatisation. Importantly, even though most species can invoke one or both of these responses, the majority of insects die from the effects of cold rather than freezing. Most studies on the effects of a changing climate on insects have focused on processes that occur predominantly in summer (development, reproduction) and on changes in distributions rather than winter survival per se. For species that routinely experience cold stress, a general hypothesis would be that predicted temperature increases of 1°C to 5°C over the next 50-100 years would increase winter survival in some climatic zones. However, this is unlikely to be a universal effect. Negative impacts may occur if climate warming leads to a reduction or loss of winter snow cover in polar and sub-polar areas, resulting in exposure to more severe air temperatures, increasing frequency of freeze—thaw cycles and risks of ice encasement. Likewise, whilst the dominant diapause-inducing cue (photoperiod) will be unaffected by global climate change, higher temperatures may modify normal rates of development, leading to a decoupling of synchrony between diapause-sensitive life-cycle stages and critical photoperiods for diapause induction. In terms of climate warming and potential heat stress, the most recent predictions of summer temperatures in Europe of 40°C or higher in 50-75 years, are close to the current upper lethal limit of some insects. Long-term data sets on insect distributions and the timing of annual migrations provide strong evidence for ‘positive’ responses to higher winter temperatures over timescales of the past 20-50 years in North America, Europe and Asia.

Understanding how these trade‐offs vary among populations and across environments is therefore important to more accurately predict species' responses to future climate change. However, the extent to which responses vary across populations remains unknown because few studies investigate intraspecific differences. We performed a full‐factorial split‐brood common garden experiment to test how variation in rearing temperature affects developmental timing and other traits important for survival during diapause in the Glanville fritillary butterfly (Melitaea cinxia). Pre‐diapause larvae originating from four regions across a latitudinal cline across Europe were reared at four temperatures (25, 28, 31 and 34°C), and we used a reaction norm approach to test for evidence of genetic differentiation and variation in developmental plasticity across regions. We found clear signs of genetic differentiation in multiple developmental traits, as well as differences in developmental plasticity. Northern larvae entered diapause in the fourth instar when the temperatures were low, whereas southern larvae did so in the fifth instar. As a result, development time is canalized with regards to temperature in northern larvae: due to entering diapause one stage earlier, they develop fast even in the cold, whereas southern larvae always develop slower, especially at low temperatures. As a trade‐off, northern larvae have a lower body mass when reared at cooler temperatures compared to southern larvae, and they show increased plasticity in diapause mass. No clear clinal patterns were found in relative fat content. Our results show that trade‐offs between body size, development time and growth rate can vary within species living across environmental clines, possibly as a consequence of natural selection to local environmental conditions or other genetic constraints. This variation highlights the importance of recognizing the context dependency of relationships between important life‐history traits and their interactions with local environments in predicting species' responses to climate change. Abstract:Physiological time is important for understanding the development and seasonal timing of ectothermic animals, but has largely been applied to developmental processes that occur during spring and summer such as morphogenesis. There is a substantial knowledge gap in the relationship between temperature and development during winter, a season that is increasingly impacted by climate change. Most temperate insects overwinter in diapause, a developmental process with little obvious morphological change. We used principles from the physiological time literature to measure and model the thermal sensitivity of diapause development rate in the apple maggot fly , a univoltine fly whose diapause duration varies substantially within and among populations. We show that diapause duration can be predicted by modeling a relationship between temperature and development rate that is shifted towards lower temperatures compared to typical models of morphogenic, non-diapause development. However, incorporating interindividual variation and ontogenetic variation in the temperature-to-development-rate relationship was critical for accurately predicting fly emergence, as diapause development proceeded more quickly at high temperatures later in diapause. We conclude that the conceptual framework may be flexibly applied to other insects and discuss possible mechanisms of diapause timers and implications for phenology with warming winters.

Abstract:Introduction: Temperature fluctuations are important for the distribution and survival of insects. Rapid hardening, a type of phenotypic plasticity, is an adaptation that can help individuals better tolerate lethal temperatures because of earlier exposure to a sublethal but stressful temperature. Nutrition and sex are also known to influence a species ability to tolerate thermal stress. This study determined the effects of larval diet, adult diet, sex and hardening on the thermal tolerance of Ceratitis cosyra (Walker) (Diptera: Tephritidae) at lower and upper lethal temperatures. Methods: Larvae were raised on either an 8% torula yeast (high) or a 1% torula yeast (low) larval diet and then introduced to one of three dietary regimes as adults for thermal tolerance and hardening assays: no adult diet, sugar only, or sugar and hydrolysed yeast diet. Flies of known weight were then either heat- or cold-hardened for 2 hours before being exposed to a potentially lethal high or low temperature, respectively. Results: Both nutrition and hardening as well as their interaction affected C. cosyra tolerance of stressful temperatures. However, this interaction was dependent on the type of stress, with nutrient restriction and possible adult dietary compensation resulting in improved cold temperature resistance only. Discussion: The ability of the insect to both compensate for a low protein larval diet and undergo rapid cold hardening after a brief exposure to sublethal cold temperatures even when both the larva and the subsequent adult fed on low protein diets indicates that C. cosyra have a better chance of survival in environments with extreme temperature variability, particularly at low temperatures. However, there appears to be limitations to the ability of C. cosyra to cold harden and the species may be more at risk from long term chronic effects than from any exposure to acute thermal stress.

Abstract:We have used recombinant human CD4 presented on beads as an affinity matrix to screen a 2'-F-pyrimidine-containing RNA library with a complexity of approximately 10(14) molecules. Affinity-selected aptamers bind recombinant CD4 with low nanomolar equilibrium dissociation constants. These high-affinity aptamers conjugated to different fluorophores such as fluorescein and phycoerythrin were used to stain cells, expressing human CD4 on cell surface, for analysis by flow cytometry. Aptamers, conjugated to fluorophores, stained mouse T cells that express human CD4 on the surface, but not the control mouse T cells lacking human CD4. The control cells, however, do express mouse CD4 whose extracellular domain has 55% sequence identity to the human form. These human CD4-specific aptamers selectively stained CD4(+) T cells in a preparation of human peripheral blood mononuclear cells. These results and others suggest that aptamers are emerging as a versatile class of molecules that can be used for various diagnostic applications performed under different formats or platforms.
Temperature-mediated dynamics: Unravelling the impact of temperature on cuticular hydrocarbon profiles, mating behaviour, and life history traits in three Drosophila species

Abstract:In a world grappling with climate change, understanding the enduring impact of changes in temperatures on insect adult traits is crucial. It is proposed that cold- and warm-adapted species exhibit specialized behavioural and physiological responses to their respective temperature ranges. In contrast, generalist species maintain more stable metabolic and developmental rates across a broader range of temperatures, reflecting their ability to exploit diverse thermal niches. Here, we explored this intricate response to temperature exposure in three Drosophila species: Drosophila ezoana originating in Arctic regions, D. novamexicana in arid, hot environments, and in the cosmopolitan species D. virilis. Rearing these flies at 15, 20, 25, and 30 °C revealed striking variations in their cuticular hydrocarbon (CHC) profiles, known to mediate mate recognition and prevent water loss in insects. The cold-adapted D. ezoana consistently exhibited reduced CHC levels with increasing temperatures, while the warm-adapted D. novamexicana and the cosmopolitan D. virilis displayed more nuanced responses. Additionally, we observed a significant influence of rearing temperature on the mating behaviour of these flies, where those reared at the extreme temperatures, 15 and 30 °C, exhibiting reduced mating success. Consequently, this led to a decrease in the production of adult offspring. Also, these adult offspring underwent notable alterations in life history traits, reaching adulthood more rapidly at 25 and 30 °C but with lower weight and reduced longevity. Furthermore, among these offspring, those produced by the cold-adapted D. ezoana were more vulnerable to desiccation and starvation than those from the warm-adapted D. novamexicana and the cosmopolitan D. virilis. In summary, our research demonstrates that Drosophila species from diverse ecological regions exhibit distinct responses to temperature changes, as evidenced by variations in CHC profiles, mating behaviours, fertility, and life history traits. This provides valuable insights into how environmental conditions shape the biology and ecology of insects.

Abstract:Abstract Factors affecting the winter survival of temperate-zone insects are reviewed. Certain species suffer high winter mortality especially in cold years, or in years with below normal amounts of insulating snow. Survival depends on the choice of winter microhabitat and on cold-hardiness, but also on biological features that have not previously been emphasized. These include behaviour that results in placement of overwintering eggs in less severe sites and manufacture of cocoons or other structures that retard inoculation by ice, or desiccation. The possible role of habitat heterogeneity in facilitating population survival is stressed. Microhabitats are characterized mainly with respect to temperature. They differ markedly among geographic areas and not only in midwinter conditions (including the extent of variation), but also in the time at which entry to the microhabitat in fall is necessary to avoid the risk of frost. The cues that govern entry into these microhabitats include negative phototaxis in many species but have not been subjected to quantitative analysis. Deeper layers are warmer in winter but warm up more slowly in spring and therefore some species, especially in arctic habitats where rapid vernal development is advantageous because the season is short, overwinter in superficial or insolated sites. Supposed mechanisms of cold- and freezing-injury and its avoidance are reviewed. There are several theories of freezing-injury: many modem theories focus on damage to the cell membrane, apparently caused by changes in cell volume induced during freezing. Injury is avoided by suppression of haemolymph nucleators (enhancing supercooling); or by a variety of methods, especially those involving solutes, minimising damage to frozen tissues. Among these solutes, the prevalence of glycerol in overwintering insects is not unexpected since glycerol is a normal metabolite in animals and can play several possible roles in cryoprotection. Cold-hardiness is metabolically costly because of these solute adjustments. The cold-hardiness of a taxon depends partly on climatic history during its evolution. Faunal composition at the highest latitudes reflects this dependence. The winter survival of many high-latitude insects varies rather widely from year to year; but the many detailed studies on cold-hardiness that have been made in the laboratory have seldom been related to natural mortality. The overwintering stage depends partly on the taxon. It is often fixed within a genus, but less closely within higher taxa. Proportionately more species overwinter as larvae at higher latitudes, especially when the life cycle lasts more than one year. Habitat also influences the overwintering stage: aquatic species generally overwinter as larvae, although species of temporary pools frequently overwinter as eggs. Overwintering in more than one stage is not common. This review suggests that winter survival of a group depends on latitude (or its climatic equivalent), habitat, and evolutionary history. It might therefore be especially useful to compare cold-hardiness, developmental rates, control of the life cycle and other features among species at the same latitude from each extreme of a series, that contrasts cold-hardy groups in stable habitats (such as Chironomidae) with aerial or vegetation dwelling groups that are not cold-hardy (such as Orthoptera). Future study of winter survival also demands more ecological data (overwintering stage, microhabitat, and conditions experienced there) for a larger sample of the fauna, and the simultaneous measurement of cold-hardiness.

Abstract:Herbivorous insects having variable numbers of generations annually depending on climate and day length conditions are increasingly breeding additional generations driven by elevated temperature under the scenario of global warming, which will increase insect abundance and result in more frequent damage events. Theoretically, this relies on two premises, i.e., either an evolutionary shift to facultative diapause for an insect behaving an obligatory diapause or developmental plasticity to alter voltinism productively for an insect with facultative diapause before shortening photoperiods inducing diapause. Inter-population evidence supporting the premise (theory) comes primarily from a model system with voltinism linked to thermal gradients across latitude. We examined the intra-population evidence in the field (47°24′ N, 123°68′ E) with Ostrinia furnacalis, one of the most destructive pests, on corn in Asia and Pacific islands. The species was univoltine in high latitudinal areas (≤46° N). Divergence of the diapause feature (obligatory and facultative) was observed within the field populations from 2016 to 2021. Warmer climates would provoke more facultative diapause individuals to initiate a second generation, which will significantly drive the population to evolve toward facultative diapause (multi-voltinism). Both divergent diapause and temperature must be considered for accurate prediction of phenology and population dynamics in ACB.

However, it is unclear which morphological traits are the best estimators of abiotic conditions, how those traits are correlated with functional outcomes in cold tolerance, and whether there are regional differences in trait expression.We used a combination of controlled laboratory assays, and collaborative field collections of invasive Drosophila suzukii in different areas of the United States, to study the factors affecting phenotype variability of this temperate fruit pest now found globally.Laboratory studies demonstrated that winter morph (WM) trait expression is continuous within the developmental temperature niche of this species (10-25°C) and that wing length and abdominal melanization are the best predictors of the larval abiotic environment.However, the duration and timing of cold exposure also produced significant variation in development time, morphology, and survival at cold temperatures. During a stress test assay conducted at -5°C, although cold tolerance was greater among WM flies, long-term exposure to cold temperatures as adults significantly improved summer morph (SM) survival, indicating that these traits are not controlled by a single mechanism.Among wild D. suzukii populations, we found that regional variation in abiotic conditions differentially affects the expression of morphological traits, although further research is needed to determine whether these differences are genetic or environmental in origin and whether thermal susceptibility thresholds differ among populations within its invaded range.

Abstract:During diapause, a state of temporarily arrested development, insects require low winter temperatures to suppress their metabolism, conserve energy stores and acquire cold hardiness. A warmer winter could, thus, reduce diapause incidence and duration in many species, prematurely deplete their energy reserves and compromise post-diapause fitness. In this study, we investigated the combined effects of thermal stress and the diapause program on the expression of selected genes involved in antioxidant defense and heat shock response in the European corn borer Ostrinia nubilalis. By using qRT-PCR, it has been shown that response to chronic heat stress is characterized by raised mRNA levels of grx and trx, two important genes of the antioxidant defense system, as well as of hsp70 and, somewhat, of hsp90, two major heat shock response proteins. On the other hand, the expression of hsc70, hsp20.4 and hsp20.1 was discontinuous in the latter part of diapause, or was strongly controlled by the diapause program and refractory to heat stress, as was the case for mtn and fer, genes encoding two metal storage proteins crucial for metal ion homeostasis. This is the first time that the effects of high winter temperatures have been assessed on cold-hardy diapausing larvae and pupae of this important corn pest.

A quantitative model of temperature-dependent diapause progression

Abstract:Winter diapause in insects is commonly terminated through cold exposure, which, like vernalization in plants, prevents development before spring arrives. Currently, quantitative understanding of the temperature dependence of diapause termination is limited, likely because diapause phenotypes are generally cryptic to human eyes. We introduce a methodology to tackle this challenge. By consecutively moving butterfly pupae of the species Pieris napi from several different cold conditions to 20 °C, we show that diapause termination proceeds as a temperature-dependent rate process, with maximal rates at relatively cold temperatures and low rates at warm and extremely cold temperatures. Further, we show that the resulting thermal reaction norm can predict P. napi diapause termination timing under variable temperatures. Last, we show that once diapause is terminated in P. napi, subsequent development follows a typical thermal performance curve, with a maximal development rate at around 31 °C and a minimum at around 2 °C. The sequence of these thermally distinct processes (diapause termination and postdiapause development) facilitates synchronous spring eclosion in nature; cold microclimates where diapause progresses quickly do not promote fast postdiapause development, allowing individuals in warmer winter microclimates to catch up, and vice versa. The unveiling of diapause termination as one temperature-dependent rate process among others promotes a parsimonious, quantitative, and predictive model, wherein winter diapause functions both as an adaptation against premature development during fall and winter and for synchrony in spring.

Abstract:Aedes albopictus (Skuse 1894) mosquitoes can transmit deadly arboviruses and are globally invasive due to their ability to survive in both tropical and temperate climates. Although adults cannot survive harsh winters, females are capable of anticipating seasonal change and producing overwintering diapause (DP) eggs that remain in a state of arrested development over the winter and hatch when favorable conditions return in the spring. While low temperatures can facilitate DP entry under short photoperiods, temperature signals alone are not sufficient to induce DP. To identify maternal phenotypes predictive of DP egg production in laboratory conditions, we characterized aspects of maternal physiology and behavior to identify those that correlate with DP egg production and changes in photoperiod, versus changes in temperature. Neither changes in temperature nor photoperiod impacted protein preference, blood meal consumption, or total number of eggs produced per female. Egg retention and oviposition timing were influenced by temperature, independent of DP egg production. However, females housed under short photoperiod conditions showed increased starvation resistance, despite showing similar levels of locomotor activity and internal stores of triacylglycerols, glucose, glycogen, and trehalose compared to females housed in long photoperiods. These results suggest that temperature and photoperiod differentially affect maternal phenotypes and identify starvation resistance as a maternal phenotype that is influenced by photoperiod and correlates with DP egg status. Aedes albopictus mosquitoes can survive in temperate climates because females are able to detect changes in temperature and day length in the autumn to produce "diapause" eggs that are provisioned with extra nutrients to survive harsh winters by remaining in a state of arrest and hatching in the spring. Although temperature and day length normally change in concert with each other throughout the year, studies have shown that day length is the most important cue used by female mosquitoes to initiate diapause egg production. In this study, the authors examined aspects of feeding and reproduction to determine which of these correlated with day length. Many aspects of reproduction were influenced by temperature; however, housing female mosquitoes under autumn-like short-day conditions led to increased starvation resistance. Surprisingly these females survive longer even though they do not appear to expend less energy compared to females housed in spring-like long-day conditions at the same temperature. This suggests that the mosquito mothers undergo changes in their metabolism that correlate with the production of DP eggs that can survive the winter. This new knowledge may lead to novel targets to disrupt seasonal reproduction in mosquitoes and limit the parts of the world where they can survive.

Abstract:Animals are facing increasing thermal and nutritional stress under climate change. Genetic adaptation is necessary to buffer long-term stress. However, it's unclear whether adaptation to combined stress can occur without compromising viability and thermal plasticity. We tested thermotolerance and thermal plasticity in larval Drosophila melanogaster selected under different temperatures (18°C, 25°C, 28°C) and diets (standard, diluted, low protein:carbohydrate). Basal cold tolerance was affected by both low protein concentration and temperature; larvae evolved higher basal cold tolerance on diluted and low protein:carbohydrate diets at 18°C and 28°C. Hardening increased cold tolerance for most lines, except those selected at 18°C and 28°C on low protein:carbohydrate diets and 25°C on standard diets. Basal heat tolerance was affected by selection temperature, with higher tolerance in 25°C selection lines. An interaction between selection temperature, selection diet and hardening treatment affected plasticity in heat tolerance, with most selection lines showing reduced heat tolerance upon hardening, except for lines selected at 25°C on low protein diets and at 28°C on standard diets. Our results indicate that adaptation to combined stress allows basal cold tolerance and plasticity in cold tolerance to evolve independently, but not heat tolerance, highlighting the increased vulnerability of ectotherms to long-term climate change.
