Influence of Dental Titanium Implants with Different Surface Treatments Using Femtosecond and Nanosecond Lasers on Biofilm Formation

Abstract:This study aimed to evaluate the impact of different surface treatments (machined; sandblasted, large grit, and acid-etched (SLA); hydrophilic; and hydrophobic) on dental titanium (Ti) implant surface morphology, roughness, and biofilm formation. Four groups of Ti disks were prepared using distinct surface treatments, including femtosecond and nanosecond lasers for hydrophilic and hydrophobic treatments. Surface morphology, wettability, and roughness were assessed. Biofilm formation was evaluated by counting the colonies of Aggregatibacter actinomycetemcomitans (Aa), Porphyromonas gingivalis (Pg), and Prevotella intermedia (Pi) at 48 and 72 h. Statistical analysis was conducted to compare the groups using the Kruskal-Wallis H test and the Wilcoxon signed-rank test (α = 0.05). The analysis revealed that the hydrophobic group had the highest surface contact angle and roughness (p < 0.05), whereas the machined group had significantly higher bacterial counts across all biofilms (p < 0.05). At 48 h, the lowest bacterial counts were observed in the SLA group for Aa and the SLA and hydrophobic groups for Pg and Pi. At 72 h, low bacterial counts were observed in the SLA, hydrophilic, and hydrophobic groups. The results indicate that various surface treatments affect implant surface properties, with the hydrophobic surface using femtosecond laser treatment exerting a particularly inhibitory effect on initial biofilm growth (Pg and Pi).

Abstract:This article explores the enhancement of material surface properties of Ti6Al4V, potentially applicable to dental implants, through ultra-short pulse laser systems. This study investigates potential connections between surface wettability and biocompatibility, addressing the challenge of improving variability in material properties with specific laser treatment. Several designed microstructures were manufactured using a picosecond laser system. After that, the wettability of these structures was measured using the sessile drop method. The basic behavior and growth activity of biological cells (MG-63 cell line) on treated surfaces were also analyzed. While the conducted tests did not conclusively establish correlations between wettability and biocompatibility, the results indicated that laser treatment of Ti6Al4V could effectively enlarge the active surface to better biological cell colonization and adhesion and provide a focused moving orientation. This outcome suggests the potential application of laser treatment in producing special dental implants to mitigate the issues during and following implantation.

Abstract:Pure Titanium (Titanium grade 2) and Ti6Al4V (Titanium grade 5) alloys are commonly used biocompatible materials for medical bone implants and dental protheses. As some implants have to be removed after a certain amount of time after the implantation, the osteo repellent surface is one of the most important property for the removable bone implants such as bone screws and plates. On contrary, for the dental protheses the property of the most importance is osseointegration. Femtosecond laser treatment causes ablation and produces LIPSS ("laser induced periodic surface structures") which can consist of cones on macro scale and ripples on nano scale which showed structures and roughness similar to that of a bone. Therefore, surface modifications by electrochemical anodization and femtosecond laser could be seen as promising methods for improving both osteo-repellence and osteointegration properties of the bone implants and dental protheses. The aim of this work is electrochemical and surface characterization of the anodized Titanium and Ti6Al4V alloy treated by the femtosecond laser. Firstly, Titanium and Ti6Al4V samples were anodized potentiostaticaly up to 100V in 25% phosphoric acid solution for 120s. Following the potentiostatic anodization, samples were structured with femtosecond laser to produce micro cones and nano ripples on top of the cones. As a final step, laser treated spots were potentiodynamicaly anodized using a SDCM (Scanning Droplet Cell Microscopy) setup. Influence of different potentials, scan rates and electrolytes were studied and optimized, consequently. Morphology and topography were examined by the means of SEM (Scanning Electron Microscopy) and AFM (Atomic Force Microscopy), while crystallography was determined by the XRD (X-Ray Diffraction). Furthermore, the composition of the oxide layer was determined by the XPS (X-ray photoelectron spectroscopy). Oxide forming factor (k ox ), a relative permittivity ε r , and ECSA (Electrochemical surface area) were obtained from the cyclic voltammograms (CV) and electrochemical impedance spectroscopy (EIS) experimental data. Electrochemical impedance spectroscopy data was modeled and compared between not preanodized, preanodized and anodized and not anodized after femtosecond laser structuring. Finally, bioassessment with osteoblast was done to examine the growth of the osteoblast cells on the modified surface structures. Value of the oxide forming factor (k ox ) for the pure Titanium and Titanium grade 5 were successfully calculated and compared with a Scanning Electron Microscopy. Calculated values of the relative permittivity ε r are in agreement with the literature values. Finally, obtained results indicate that anodized and subsequently femtosecond laser treated samples could be used as medical bone and dental implants.

Abstract:Ceramic dental materials, especially alumina (20 %vol)-yttrium stabilized tetragonal zirconia poly crystal (A-Y-TZP20), have been considered as alternatives to metals for endosseous dental implant application. For increasing the bone-to-implant contact as well as the speed of bone formation, a new surface modification can be effective. The aim of this study was to design microgroove patterns by femtosecond laser on A-Y-TZP20 nano-composite disks for endosseous dental implant application. The phase composition and the morphology of the A-Y-TZP20 nano-composite samples were characterized using X-ray diffraction and Scanning electron microscopy equipped with energy dispersive X-ray spectroscopy techniques. Statistical analysis was submitted to Kolmogorov-Smirnov test and Student's t test for independent variables, with a 5 % significance level. EDAX analysis revealed a significant decrease in the relative content of contaminants like carbon (p < 0.05) in laser surface-treated group as compared to non surface-treated group. X-ray diffraction did not show any change in the crystalline structure induced by laser processing. It was concluded that the femtosecond laser is a clean and safe method for surface modification of A-Y-TZP20.

Abstract:Purpose Attempts have been made to decontaminate the implant surface by using diode lasers. However, the parameters that provide efficient decontamination without altering the topography or surface characteristics of titanium implants are still unclear. The aim of the present study was to evaluate the effect of altering the power of diode laser (940 nm) application on the hydrophilicity, surface topography, and chemical composition of sandblasted, large grit, acid‐etched (SLA) treated titanium alloy implant surface. Materials and Methods Thirty‐six SLA‐treated titanium discs (Dentis Co., Ltd.) were used in this study. The hydrophilicity of all discs was measured by using a contact angle goniometer (190 CA; Rame‐hart Co, Ltd). Discs were randomly divided into four groups (n = 9 each) based on the power of the diode laser used. Group I (control, no lasing group), group II: treated with 1 W power, group III: 2 W power, and group IV: 3 W power. The chemical composition of the SLA discs was evaluated by using energy dispersive x‐ray spectroscopy (EDX) before laser application. Hydrophilicity was reevaluated after the application of laser irradiation. The surface topography of all discs was examined. Changes in the chemical composition of the titanium discs were investigated following the lasing procedure. Morphometric analysis of the surface area (μm2) of the indentations created following laser application was also evaluated. Data were collected and the Shapiro‐Wilk test of normality was used. Abstract:To elucidate the cytotoxic induction mechanisms of the hydrophilic HEMA, the comparative cytotoxic activities of HEMA and the hydrophobic monomers TEGDMA and MMA were studied, using erythrocytes, gingival fibroblasts and a salivary gland carcinoma cell line. Also, the gel-to-fluid phase transition properties (i.e. temperature, Tm; cooperativity, H/HHW; enthalpy, ΔH) of dipalmitoylphosphatidylcholine (DPPC) and DPPC/cholesterol (CS) liposomes (as a model for biological membranes) induced by methacrylates were investigated, using differential scanning calorimetry (DSC). In addition, the methacrylate-chemical-shifts in DPPC liposomes were assayed using NMR spectroscopy. Both the hemolytic and cytotoxic activity declined in the order: TEGDMA> HEMA>MMA. The changes in Tm increased in the order: HEMA >TEGDMA, while in contrast, that of HEMA was slightly increased without changes in the ΔH. The DSC changes in DPPC/CS liposomes with HEMA were the largest of those recorded. The cytotoxicity of HEMA may be induced by the hydrophobic interaction derived from the molecular association of OH groups of HEMA and, in addition, by the preferential interaction with CS.

Abstract:Surface design plays a critical role in determining the integration of dental implants with bone tissue. Femtosecond laser‐texturing has emerged as a breakthrough technology offering excellent uniformity and reproducibility in implant surface features. However, when compared to state‐of‐the‐art sandblasted and acid‐etched surfaces, laser‐textured surface designs typically underperform in terms of osseointegration. This study investigated the capacity of a bio‐inspired femtosecond laser‐textured surface design to enhance osseointegration compared to state‐of‐the‐art sandblasted & acid‐etched surfaces. Laser‐texturing facilitates the production of an organized trabeculae‐like microarchitecture with superimposed nano‐scale laser‐induced periodic surface structures on both 2D and 3D samples of titanium‐zirconium‐alloy. Following a boiling treatment to modify the surface chemistry, improving wettability to a contact angle of 10°, laser‐textured surfaces enhance fibrin network formation when in contact with human whole blood, comparable to state‐of‐the‐art surfaces. In vitro experiments demonstrate that laser‐textured surfaces significantly outperform state‐of‐the‐art surfaces with a 2.5‐fold higher level of mineralization by bone progenitor cells after 28 days of culture. Furthermore, in vivo evaluations reveal superior biomechanical integration of laser‐textured surfaces after 28 days of implantation. Notably, during abiological pull‐out tests, laser‐textured surfaces exhibit comparable performance, suggesting that the observed enhanced osseointegration is primarily driven by the biological response to the surface. Abstract:Aim: The purpose of the study was the evaluation of the esthetic and physical changes produced on colored titanium Grade 5 (Ti6Al4V) laser treated surfaces to be used in implant dentistry for esthetic success. Materials and methods: Colored titanium surfaces were obtained with laser treatment. The physical and topographic properties were evaluated by stereo, light, and electron microscopy and profilometric analyses. L*a*b* colorimetric coordinates were measured by spectrometry, and the superficial chemical characteristics were evaluated by energy dispersive X-ray analysis. Results: Within the complete palette of titanium colors, pinks (P1-P2), incarnadine (I), and white (W) obtained by laser were selected. The topography, texture, hues, saturation, roughness, and porosity of the samples were compared with those of machined (M) and sand-blasted and etched (SBAE) control surfaces. P1, P2, and I, similar in hue and roughness (Ra ≅ 0.5 μm), had a microgroove spacing of 56 μm and a decreasing porosity. The W sample with a “checkerboard” texture and a light color (L* 96.31) was similar to the M samples (Ra = 0.32 μm), but different from SBAE (Ra = 1.41 μm, L* 65.47). Discussion: The aspects of hard and soft tissue could result in an esthetic failure of the dental implant by showing the dark color of the fixture or abutment. The two different pinks and incarnadine surfaces showed favorable esthetic and physical features to promote dental implant success even in the maxillary anterior area with gingival recession, asymmetry, and deficiency. Conclusion: Titanium colored laser surfaces represent a valid alternative to those currently traditionally obtained and interesting and potential perspectives in the management of dental implants’ esthetic failure.

Abstract:Titanium is widely used in implants but can cause adverse reactions like inflammation and tissue rejection. Surface modifications like acid-etching and anodization improve implant properties and osseointegration. This study investigated how micro and nanoporous TiO 2 films affect the implant-host interaction, focusing on physical–chemical and biological aspects. Surface treated implants were obtained by acid-etched and anodization of commercially pure titanium (CPTi). Samples were characterized by FE- and FIB-SEM, XRD, EDX, WCA, profilometry, and electrochemical behavior was also investigated. Biocompatibility was accessed by protein adsorption (albumin and serum proteins), hemolysis, and in vitro fibroblast interaction. Anodic samples exhibited the highest corrosion resistance and apatite deposition. Protein adsorption was related to surface roughness and wettability. Samples followed the Langmuir model isotherm (Type I), except for galvanostatic samples (Type III isotherm). Kinetic adsorption was modeled by a pseudo-first-order model, wherein rate-limiting step is albumin diffusion into pores. All surfaces met the hemocompatibility requirements for its use as biomaterial (hemolysis index < 2 %). The upper surface roughness threshold was identified at approximately Sa ≈ 1 μm for better fibroblast viability, adhesion, and proliferation. Combining the results, nanoporous samples present optimized surface conditions that enhance body-fluid contact and improve cell adhesion, thereby contributing to long-term stability.

Abstract:3 mol% yttria-stabilized zirconia ceramics have been gaining attention as promising restorative materials that are extensively used in dental implant applications. However, implant failure due to bacterial infection and its bioinert surface slow osseointegration in vivo, which are significant issues in clinical applications. In this work, surface modification was achieved using a continuous wave carbon dioxide laser at a wavelength of 10.6 μm in an air atmosphere. Changes in the surface characteristics were evaluated using X-ray diffraction (XRD), field-emission scanning electron microscopy (FESEM), atomic force microscopy (AFM), and 2D roughness and hardness tests. The bioactivity of the laser-treated samples was studied by examining their behavior when immersed in the SBF solution. The formation of the hydroxyapatite phase on the laser-treated sample was much more uniform than that of its untreated counterparts. The antibacterial properties of surface-treated zirconia ceramics against Streptococcus mutans and Escherichia coli bacteria were rigorously examined. These results indicate that the laser-induced nanoscale grooves significantly improved antibacterial activity by creating hydrophobic surfaces. The cellular response was evaluated for 7 days on microtextures on the zirconia surfaces and an untreated sample with MC3T3-E1 pre-osteoblast cell line cultured under basal conditions. Surface topography was revealed to improve the cellular response with increased metabolic activity compared to the untreated sample and showed modulation of cell morphology for the entire time. These results suggest that laser modification can be an appropriate non-contact method for designing nanoscale microtextures to improve the biological response and antibacterial behavior of zirconia ceramics in restorative dentistry.

Abstract:The use of dental implants of titanium and its alloys has proved to be effective, through well established and documented parameters, both in the dimensions and in the manufacturing processes and also in the surgical techniques. There are clinical situations where there is a need to reduce the diameter of the implants, below 3.75 mm in diameter. In the current state of art of the implant technology it is desirable that these also have surfaces capable of decreasing the period of osseointegration. In the present work, to improve the mechanical strength of the material, an alloy of 80% of Ti and 20% of Zr % in mass was proposed and elaborated, aiming its use as biomaterial. Physical, chemical, microstructural and mechanical characterization was carried out. The surfaces of the treated samples were observed using: scanning electron microscopy (SEM); semi quantitatively chemically analyzed using dispersive energy spectroscopy (EDS: wettability of the samples was determined and, finally, the roughness was measured using optical profilometry. For the conditions used in the present work, it was concluded, that the best surface treatment for the TiZr 80/20 alloy was acid etching with 1% vol. hydrofluoric acid for 5 minutes, as this treatment presented the most prominent results of wettability and roughness simultaneously.
Effect of different laser texture configurations on improving surface wettability and wear characteristics of Ti6Al4V implant material

Abstract:In this paper, the effect of different laser-textured Ti6Al4V alloy surface topologies on improving wettability and tribology characteristics is reported. Three texture patterns, namely dimple, moat and hybrid configurations, using an Nd-YAG laser source with varying scanning speed were prepared on the implant material surface. Surface properties, such as micro-hardness, wettability, friction, wear and elemental analysis, were studied. The morphology and features of the textured surfaces were investigated using a scanning electron microscope. The surface oxidation was evidenced through EDS analysis showing the presence of oxygen (22.9%) and carbon (11.7%) on the laser-textured surface. The wettability analysis has shown the formation of hydrophilicity on the textured surfaces. The moat pattern surface has the highest textured density of 58% and the least contact angle of 51.5°. The wear test results show an improved tribological property for textured surfaces than the non-textured surface. This study shows that the moat type-textured configuration is more effective for creating functional surfaces of Ti6Al4V bio-implant material followed by hybrid and dimple configurations.

Abstract:Abstract The initial stability after implantology is paramount to the survival of the dental implant, and the surface roughness of the implant plays a vital role in this regard. The characterisation of surface topography is a complicated branch of metrology, with a huge range of parameters available. Each parameter contributes significantly towards the survival and mechanical properties of three-dimensional printed specimens. The purpose of this paper is to experimentally investigate the effect of surface roughness of three-dimensional printed dental implants and three-dimensional printed dogbone tensile samples under areal height parameters, amplitude parameters (average of ordinates), skewness parameters and mechanical properties. During the experiment, roughness values were analysed, and the results showed that the skewness parameter demonstrated a minimum value of 0.59%. The three-dimensional printed dental implant recorded the arithmetic mean deviation of the assessed profile with a 3.4-mm diameter at 43.23% and the three-dimensional printed dental implant with a 4.3-mm diameter at 26.18%. Samples with a complex geometry exhibited a higher roughness surface, which was the greatest difficulty of additive manufacturing when evaluating surface finish. The results show that when the ultimate tensile stress decreases from 968.35 to 955.25 MPa, the arithmetic mean deviation increases by 1.4%, and when ultimate tensile stress increases to 961.18 MPa, the arithmetic mean deviation increases by 0.6%. When the cycle decreases from 262,142 to 137,433, the arithmetic mean deviation shows that less than a 90.74% increase in the cycle is obtained. For the three-dimensional printed dental implants, the higher the surface roughness, the lower the mechanical properties, ultimately leading to decreased implant life and poor performance.

Abstract:Background: Titanium dental implants are today widely used with osseointegration mainly dependently on the implant surface properties. Different processing routes lead to different surface characteristics resulting, of course, in different in situ behaviors of the implants. Materials: The effect of different treatments, whether mechanical or chemical, on the surface morphology of titanium implants were investigated. To this aim, various experimental methods, including roughness analysis as well scanning electron microscope (SEM) observations, were applied. Results: The results showed that, in contrast to the mechanical treatments, the chemical ones gave rise to a more irregular surface. SEM observations suggested that where commercial pure titanium was used, the chemical treatments provided implant surfaces without contaminations. In contrast, sandblasted implants could cause potential risks of surface contamination because of the presence of blasting particles remnants. Conclusions: The examined implant surfaces showed different roughness levels in relation to the superficial treatment applied. The acid-etched surfaces were characterized by the presence of deeper valleys and higher peaks than the sandblasted surfaces. For this reason, acid-etched surfaces can be more easily damaged by the stress produced by the peri-implant bone during surgical implant placement.

Abstract:This review covers several basic methodologies of surface treatment and their effects on titanium (Ti) implants. The importance of each treatment and its effects will be discussed in detail in order to compare their effectiveness in promoting osseointegration. Published literature for the last 18 years was selected with the use of keywords like titanium dental implant, surface roughness, coating, and osseointegration. Significant surface roughness played an important role in providing effective surface for bone implant contact, cell proliferation, and removal torque, despite having good mechanical properties. Overall, published studies indicated that an acid etched surface-modified and a coating application on commercial pure titanium implant was most preferable in producing the good surface roughness. Thus, a combination of a good surface roughness and mechanical properties of titanium could lead to successful dental implants.

Abstract:Titanium (Ti) materials are highly valued in the medical field for their outstanding biocompatibility and corrosion resistance. However, challenges such as suboptimal wettability and wear resistance can impact the tribological properties of titanium implants, potentially leading to implant failure. This study explores the application of ultrafast pulsed laser processing to create two distinct structures, circular pits and grooves, on the surface of titanium materials. The samples underwent low-surface-energy treatment, after which the wettability and wear resistance of the textured surfaces were evaluated. The findings indicate that the textured surfaces exhibit improved hydrophobic properties and reduced surface wear. Specifically, the textured surfaces demonstrated a remarkable 73.68% reduction in wear compared to the untextured surfaces. These results underscore the potential of etching textured structures onto titanium surfaces to enhance their wear resistance, thereby offering promising implications for the improvement of titanium implant performance.
