Abstract:RGD has been demonstrated to improve implant osseointegration. However, few studies are known about an effect of RGD coating on a bone-bonding ability of screw-shaped porous implant. The aim of this study was to investigate the effect of RGD coating using the layer-by-layer self-assembly technique on the bone-bonding ability of porous implant. 60 implants of 10 mm in length (30 control and 30 RGD-coated) were inserted into femurs of 30 rabbits and 30 implants of 8 mm in length (15 control and 15 RGD-coated) were inserted into tibias of 15 rabbits. At 4, 8, and 12 weeks post-implantation, femurs and tibias were retrieved and prepared for removal torque tests (RTQ) and histomorphometric evaluation, respectively. No differences were found in the RTQ values between two implants at 4 weeks (p = 0.932). There were statistical significances in the RTQ values at 8 and 12 weeks (p = 0.002, 0.001, respectively). New bone was formed on both implant surfaces. The bone-implant contact pattern appeared to produce a broad-based direct contact in both implants. The RGD-coated implants showed a significantly greater BIC in the threads inside the cortical bone compared with the control implants at 4, 8, and 12 weeks (p = 0.024, 0.041, 0.022, respectively). No differences were found in the bone area within the same threads between two implants at 4 weeks (p = 0.806) whereas differences were found at 8 and 12 weeks (p = 0.009, 0.031, respectively). It was concluded that RGD coating using the layer-by-layer self-assembly technique has a positive effect on the bone-bonding ability of porous implant.
Surface Modification And Its Effect On Attachment, Spreading, And Proliferation Of Human Gingival Fibroblasts

Abstract:Purpose: The purpose of this study was to exploit potential methods of surface modification for improving the seal between the neck portion of a dental implant and the surrounding soft tissue. Materials and Methods: Titanium surfaces were modified by machining (SM-Ti group); machining and acid etching (AE-Ti group); or machining, acid etching, and depositing 4.5 collagen/hyaluronic acid (col/HA) polyelectrolyte bilayers (CHC-Ti group). These were analyzed using scanning electron microscopy, scanning force microscopy, x-ray photoelectron spectroscopy, contact angle measurement, and quartz crystal microbalance measurement. The degradation behavior of the col/HA multilayer coating was measured. Next, human gingival fibroblasts (HGFs) were cultured on the different surfaces, and cell morphology and spreading were observed using fluorescence microscopy and a shape factor measurement. Cell proliferation was examined by fluorometric quantification of the amount of cellular DNA. Matrix formation of HGFs was determined via enzyme-linked immunosorbent assay. Gene expression was analyzed via reverse transcriptase polymerase chain reaction. Results: Similar surface topology for these three groups was observable on a microscopic scale, and morphologic differences were apparent on the nanoscale. Both acid etching and col/HA deposition improved the hydrophilicity of the titanium surface, in contrast to machining alone. Each col/HA bilayer was about 5 nm thick. The col/HA coating degraded in about a week. Attachment and spreading of HGFs was better on the CHC-Ti surface than on the SM-Ti or AE-Ti surfaces. Moreover, the proliferation and differentiation of HGFs were greatly stimulated when cultured on CHC-Ti. Conclusion: In contrast to two control surfaces (one machined, one machined and acid-etched), col/HA treatment of Ti improved the attachment, spreading, proliferation, and differentiation of HGFs. Abstract:For dental/orthopedic implants to achieve better bone apposition and bone-implant bonding, various approaches to improve titanium surfaces have been developed. Recently, a fluoridated hydroxyapatite (FHA) coating on titanium (Ti) implants was made by sol–gel method and shown to be a possible applicative bone implant. The purpose of the current study was to evaluate biological responses and biomechanical bonding strength of FHA coated Ti implants as compared with that of the conventional Ti alloys and hydroxyapatite (HA) coated Ti implants. In vitro assays were made using human osteoblast-like cell (MG63) culture on different implants with cell attachment, morphology and differentiation evaluations. The implant plates were also implanted into the proximal metaphysis of New Zealand White rabbit tibiae. After 8 and 16 weeks implantation, mechanical and histological assessments were performed to evaluate biomechanical and biological behavior in vivo. The results showed that the cell adhesion and cell growth rate on the FHA and HA surface was higher than that on cp Ti surface (p<0.01), and insignificant difference was observed between two coated groups. Mechanical test demonstrated that the FHA implants had a higher interface shear strength than the both controls at 8 and 16 wks, with no significant difference with HA-Ti. Histologically, the coated implants revealed a significantly greater percentage of bone-implant contact when compared with the uncoated implants. Results demonstrated that the new FHA surface improved cell adhesion and proliferation. The coating exhibited a bioactive mechanical and histological behavior at bone-implant interface, suggesting that a useful approach by combined coating processes could optimize implant surfaces for bone deposition and early implant fixation.

Abstract:Objective. The objective of this study was to investigate bone responses to titanium implants surface-roughened by sandblasted and double-etched treatments in a rabbit model.Study design. Sixty implants of 10 mm in length (30 machined and 30 roughened) were inserted into femurs of 30 rabbits and 30 implants of 8 mm in length (15 machined and 15 roughened) were inserted into tibias of 15 rabbits. At 2, 4, and 8 weeks postimplantation, femurs and tibias were retrieved and prepared for removal torque tests (RTQ) and histomorphometric evaluation, respectively.Results. The roughened implants showed 66.21%, 89.06%, and 115.00% greater RTQ values than did the machined implants at 2, 4, and 8 weeks. Histomorphometric evaluation demonstrated the roughened implants significantly increased bone-implant contact and peri-implant bone formation during all observation periods.Conclusion. These results suggest this surface-roughened approach provides the implant surface with a considerable osteoconductive potential promoting a high level of bone integration with bone. Abstract:The aim of this study was to evaluate the influence of a roughened H2O2/HCl heat-treated titanium surface on peri-implant bone formation at an early stage in vivo. 24 Ti6Al4V alloy implants were used; half were treated by sandblasted and dual acid-etched treatments (control group), while the others were treated by sandblasted, dual acid-etched and H2O2/HCl heat treatments (test group). The morphology and roughness were analyzed by field emission SEM and atomic force microscopy. The implants were inserted into the femora of 12 adult white rabbits. After 2 and 4 weeks, femora block specimens were prepared for histological and histomorphometric analysis. SEM micrographs showed that multilevel and different sized pits were formed on both surfaces. New bone formation was observed on both implant surfaces. Test implants demonstrated a greater mean percentage of bone-implant contact as compared with controls at 2 (46.84 vs. 41.81, p=0.000) and 4 weeks (49.43 vs. 44.87, p=0.006) of healing. It is concluded that the H2O2/HCl heat-treated rough titanium surface promoted enhanced bone apposition during the early stages of new bone formation around the implant.

Abstract:OBJECTIVE The purpose of this study was to compare and evaluate the effects of various laser systems on surface roughness and shear bond strengths of zirconia ceramics. MATERIAL AND METHODS Seventy-two ceramic discs (10 mm×2 mm) obtained from two sintered zirconia ceramics (Zirkonzahn, Zirkonzahn Prettau(®)) were divided into two groups (n=36) according to the computer-aided design (CAD)-computer-aided manufacturing (CAM) technique and then further divided into three groups (n=12). Each group was treated with one of the following: (1) femtosecond (FS), (2) Nd:YAG (NY), and (3) Er:YAG (EY) lasers. After laser irradiation, a scanning electron micrograph (SEM) was taken at 500× magnification for qualitative examination. Following surface roughness measurement by profilometry, resin cement (Rely X U200) was bonded to the ceramic specimens using Teflon tubes (3 mm height, 4 mm diameter). Specimens were stored in distilled water at 37°C for 24 h and then thermal cycled for 5000 cycles. A shear bond strength (MPa) test was performed using a universal testing machine at a crosshead speed of 1 mm/min. Data were analyzed by two way analysis of variance (ANOVA) and Tukey honest significant difference (HSD) tests for pairwise comparisons among groups (p=0.05). RESULTS MPa and roughness were significantly affected by laser type (p<0.001), and there was no significant interaction between the two zircon ceramics. The group irradiated with FS laser had significantly higher (p<0.05) roughness and MPa mean values than those of the other groups. No significant difference was found between the groups irradiated with NY and EY laser (p>0.05). In the SEM, the surfaces of the FS group were rougher than those of the NY and EY groups. NY and EY surfaces were nearly smooth. CONCLUSIONS FS laser is an effective surface treatment for roughening surfaces of zirconia ceramics. Furthermore, it reveals the highest MPa.
Novel hydrophilic nanostructured microtexture on direct metal laser sintered Ti-6Al-4V surfaces enhances osteoblast response in vitro and osseointegration in a rabbit model

Abstract:The purpose of this study was to compare the biological effects in vivo of hierarchical surface roughness on laser sintered titanium-aluminum-vanadium (Ti-6Al-4V) implants to those of conventionally machined implants on osteoblast response in vitro and osseointegration. Laser sintered disks were fabricated to have micro-/nano-roughness and wettability. Control disks were computer numerical control (CNC) milled and then polished to be smooth (CNC-M). Laser sintered disks were polished smooth (LST-M), grit blasted (LST-B), or blasted and acid etched (LST-BE). LST-BE implants or implants manufactured by CNC milling and grit blasted (CNC-B) were implanted in the femurs of male New Zealand white rabbits. Most osteoblast differentiation markers and local factors were enhanced on rough LST-B and LST-BE surfaces in comparison to smooth CNC-M or LST-M surfaces for MG63 and normal human osteoblast cells. To determine if LST-BE implants were osteogenic in vivo, we compared them to implant surfaces used clinically. LST-BE implants had a unique surface with combined micro-/nano-roughness and higher wettability than conventional CNC-B implants. Histomorphometric analysis demonstrated a significant improvement in cortical bone-implant contact of LST-BE implants compared to CNC-B implants after 3 and 6 weeks. However, mechanical testing revealed no differences between implant pullout forces at those time points. LST surfaces enhanced osteoblast differentiation and production of local factors in vitro and improved the osseointegration process in vivo. © 2016 Wiley Periodicals, Inc. Impact of Laser Structuring on Medical-Grade Titanium: Surface Characterization and In Vitro Evaluation of Osteoblast Attachment

Abstract:Improved implant osteointegration offers meaningful potential for orthopedic, spinal, and dental implants. In this study, a laser treatment was used for the structuring of a titanium alloy (Ti6Al4V) surface combined with a titanium dioxide coating, whereby a porous surface was created. The objective was to characterize the pore structure shape, treatment-related metallographic changes, cytocompatibility, and attachment of osteoblast-like cells (MG-63). The treatment generated specific bottleneck pore shapes, offering the potential for the interlocking of osteoblasts within undercuts in the implant surface. The pore dimensions were a bottleneck diameter of 27 μm (SD: 4 μm), an inner pore width of 78 μm (SD: 6 μm), and a pore depth of 129 μm (SD: 8 μm). The introduced energy of the laser changed the metallic structure of the alloy within the heat-affected region (approximately 66 μm) without any indication of a micro cracking formation. The phase of the alloy (microcrystalline alpha + beta) was changed to a martensite alpha phase in the surface region and an alpha + beta phase in the transition region between the pores. The MG-63 cells adhered to the structured titanium surface within 30 min and grew with numerous filopodia over and into the pores over the following days. Cell viability was improved on the structured surface compared to pure titanium, indicating good cytocompatibility. In particular, the demonstrated affinity of MG-63 cells to grow into the pores offers the potential to provide significantly improved implant fixation in further in vivo studies.

For the best bone-implant contact, machined titanium is subjected to various surface treatments. In the present study, proliferation of human gingivial fibroblast (HGF-1) cells on Grade 5 titanium disks covered with Grade 23 titanium by selective laser melting technology was evaluated. Aim: The main aim was to provide a novel surface procedure providing more biocompatible external structure with a biomechanically intact inner structure and increasing cell proliferation. Materials and Methods: Forty-eight titanium Grade 5 machined disks with 5 mm of diameter was divided into four groups. Group 1, also known as the control group, was not subjected to any surface treatment. Group 2 was treated with sand-blasted, large-grid, acid-etching (SLA) technique and Group 3 was treated with selective laser melting (SLM) method. Group 4 was treated with both SLM and SLA. The surface topography was analyzed using scanning electron microscope and the roughness of the samples was evaluated via optic profilometer. Additionally, optical tensiometer was used to measure the surface wettability. To obtain further insights on biocompability of the samples, HGF-1 cell viability at 48 h was assessed with MTT assay. These results were also confirmed by fluorescent staining. Results: Although Ra value and wettability of Group 4 were the highest amongst the samples handled, based on 48 h MTT results and fluorescent staining, highest cell proliferation was observed in Group 3. Conclusions: It was verifed that the surface topography, roughness, and wettability are all crucial factors on healthy cell populations. Therefore, it was concluded that disks treated with SLM were shown to express the most suitable condition for biocompability.

Abstract:Purpose Attempts have been made to decontaminate the implant surface by using diode lasers. However, the parameters that provide efficient decontamination without altering the topography or surface characteristics of titanium implants are still unclear. The aim of the present study was to evaluate the effect of altering the power of diode laser (940 nm) application on the hydrophilicity, surface topography, and chemical composition of sandblasted, large grit, acid‐etched (SLA) treated titanium alloy implant surface. Materials and Methods Thirty‐six SLA‐treated titanium discs (Dentis Co., Ltd.) were used in this study. The hydrophilicity of all discs was measured by using a contact angle goniometer (190 CA; Rame‐hart Co, Ltd). Discs were randomly divided into four groups (n = 9 each) based on the power of the diode laser used. Group I (control, no lasing group), group II: treated with 1 W power, group III: 2 W power, and group IV: 3 W power. The chemical composition of the SLA discs was evaluated by using energy dispersive x‐ray spectroscopy (EDX) before laser application. Hydrophilicity was reevaluated after the application of laser irradiation. The surface topography of all discs was examined. Changes in the chemical composition of the titanium discs were investigated following the lasing procedure. Morphometric analysis of the surface area (μm2) of the indentations created following laser application was also evaluated. Data were collected and the Shapiro‐Wilk test of normality was used. Abstract:The successful implantation depends on the favorable biocompatibility of the implant. In this paper,microgrooves on the surface of Ti-6Al-4V titanium alloy implants were made by laser processing technology,scanning electron microscopy and surface profilometer were used to observe the laser textured titanium samples,firstly. Then, the liquid-solid contact angle-measuring instrument was used to characterize the wettability of titanium samples. Finally,using a microplate reader and laser confocal microscope, the cytological behavior of cell proliferation, adhesion, differentiation, and cell morphology on the surface of different samples were studied.It showed that laser microgrooved surfaces had reduced liquid-solid contact angles,increased surface roughness,and better proliferation and differentiation of osteoblasts, while most cells grew along the microgrooves. These results indicate that the increased wettability provides a better growth environment for cell growth. Microgrooves are crucial in cell contact guidance, while their parameters control cell behavior and deserve an in-depth study.

Nucleolar organizer regions (NORs) in normal and pathological liver: a quantitative analysis.

Abstract:By means of a silver staining method, Nucleolar Organizer Region-associated proteins (NORs) have been evaluated on paraffin sections of a series of 58 ultrasound-guided liver biopsy specimens. These included 12 normal livers, 12 cirrhotic livers, 12 cases of chronic hepatitis and 22 cases of hepatocellular carcinoma. A significant difference (P less than 0.001) was found between the mean AgNOR scores of the normal and pathological biopsies, and between the non-neoplastic and the carcinomatous lesions. The authors suggest that AgNOR counts, in combination with conventional histocytological criteria, may be a useful method in the diagnosis of liver diseases.

Abstract:The clinical success of dental titanium implants is profoundly linked to implant stability and osseointegration, which comprises pre-osteoblast proliferation, osteogenic differentiation, and extracellular mineralization. Because of the bio-inert nature of titanium, surface processing using subtractive or additive methods enhances osseointegration ability but limits the benefit due to accompanying surface contamination. By contrast, laser processing methods increase the roughness of the implant surface without contamination. However, the effects of laser-mediated distinct surface structures on the osteointegration level of osteoblasts are controversial. The role of a titanium surface with a laser-mediated microchannel structure in pre-osteoblast maturation remains unclear. This study aimed to elucidate the effect of laser-produced microchannels on pre-osteoblast maturation. Pre-osteoblast human embryonic palatal mesenchymal cells were seeded on a titanium plate treated with grinding (G), sandblasting with large grit and acid etching (SLA), or laser irradiation (L) for 3–18 days. The proliferation and morphology of pre-osteoblasts were evaluated using a Trypan Blue dye exclusion test and fluorescence microscopy. The mRNA expression, protein expression, and protein secretion of osteogenic differentiation markers in pre-osteoblasts were evaluated using reverse transcriptase quantitative polymerase chain reaction, a Western blot assay, and a multiplex assay, respectively. The extracellular calcium precipitation of pre-osteoblast was measured using Alizarin red S staining. Compared to G- and SLA-treated titanium surfaces, the laser-produced microchannel surfaces enhanced pre-osteoblast proliferation, the expression/secretion of osteogenic differentiation markers, and extracellular calcium precipitation. Laser-treated titanium implants may enhance the pre-osteoblast maturation process and provide extra benefits in clinical application.

Effect of surface finish on the osseointegration of laser-treated titanium alloy implants

Abstract:It was the purpose of this study to examine the osseointegration of laser-textured titanium alloy (Ti6Al4V) implants with pore sizes of 100, 200, and 300 microm, specifically comparing 200-microm implants with polished and corundum-blasted surfaces in a rabbit transcortical model. Using a distal and proximal implantation site in the distal femoral cortex, each animal received all four different implants in both femora. The bone-implant interface and the newly formed bone tissue within the pores and in peri-implant bone tissue were examined 3, 6, and 12 weeks post-implantation by static and dynamic histomorphometry. Here we show that additional surface blasting of laser-textured Ti6Al4V implants with 200-microm pores resulted in a profound improvement in osseointegration, 12 weeks postimplantation. Although lamellar bone formation was found in pores of all sizes, the amount of lamellar bone within pores was linearly related to pore size. In 100-microm pores, bone remodeling occurred with a pronounced time lag relative to larger pores. Implants with 300-microm pores showed a delayed osseointegration compared with 200-microm pores. We conclude that 200 microm may be the optimal pore size for laser-textured Ti6Al4V implants, and that laser treating in combination with surface blasting may be a very interesting technology for the structuring of implant surfaces.

Influence of Laser Treatment of Ti6Al4V on the Behavior of Biological Cells

Abstract:This article explores the enhancement of material surface properties of Ti6Al4V, potentially applicable to dental implants, through ultra-short pulse laser systems. This study investigates potential connections between surface wettability and biocompatibility, addressing the challenge of improving variability in material properties with specific laser treatment. Several designed microstructures were manufactured using a picosecond laser system. After that, the wettability of these structures was measured using the sessile drop method. The basic behavior and growth activity of biological cells (MG-63 cell line) on treated surfaces were also analyzed. While the conducted tests did not conclusively establish correlations between wettability and biocompatibility, the results indicated that laser treatment of Ti6Al4V could effectively enlarge the active surface to better biological cell colonization and adhesion and provide a focused moving orientation. This outcome suggests the potential application of laser treatment in producing special dental implants to mitigate the issues during and following implantation.

[The role of surface roughness in promoting osteointegration]

Abstract:Scientific evidence that has been gathered in the past 20 years established that certain endosseous dental implants--primarily screw-type implants made of commercially pure titanium can be successfully utilized as anchorage for dental prostheses. In recent years, an effort has been made to simplify the surgical procedure, in order to modify clinical treatment modalities. One of the trends is to increasingly utilize microrough titanium implants. Roughened implant surfaces have a long history in implant dentistry, and the most prominent surface is titanium plasma-sprayed (TPS). In recent years new implant surfaces have emerged, so-called microrough titanium surfaces produced with reducing techniques such as grit-blasting with Al2O3 or TiO2 particles, sandblasting and acid-etching, or acid-etching alone. These different titanium surfaces have been tested in numerous in-vivo studies utilizing different animal models. Summarizing the results of these studies, it can be concluded that there is currently sufficient evidence that titanium implants with a microrough surfaces achieve a faster bone integration, a higher percentage of Bone implant Contact (BIC), and a higher resistance to shear documented with higher Removal Torque Values (RTV) when compared with titanium implants with a polished or machined surface. In order to understand the mechanism through which surface roughness modulates its effects mentioned above, recent studies used in-vitro experimental methods to study cell response to implant surface topography. These studies have shown that osteoblasts are sensitive to surface roughness, exhibiting decreased proliferation and a more differentiated phenotype on rougher surfaces. PGE2 production is enhanced on rough surfaces, as is the production of TGF beta 1, suggesting that surface roughness can mediate autocrine and paracrine regulation of osteogenesis. Moreover, surface roughness was found to modulate the effect of systemic hormones like 1,25-(OH)2D3 on osteoblasts. The clinical advantages of implants with rough surface were observed in recently conducted clinical trials. It was found, in humans, that roughened titanium implants need shorter healing period before loading, 6-8 (SLA and Osseotite respectively) weeks instead of 12 weeks. The clinical advantages of shorter healing periods are obvious. Moreover, it was found that certain roughened implants can be used in shorter sizes (6-8 mm) then accepted today. The utilization of shorter implants offers the avoidance of extensive surgical procedures such as nerve lateralization in the mandible or sinus grafting in the maxilla. However, sufficient long term documentation is still lacking, and the predictability of such modalities has yet to be examined in long term prospective clinical trials.
Development of Microstructure on Titanium Implant Surface Using CO2 Laser Processing

Abstract:Background: Dental biomaterials made of titanium are commonly used. It depends on the implant surface texture to improve fixation and prevent the unwanted adhesion of bone cells. This study aimed to investigate whether continuous laser beam carbon dioxide (CNC - CO2) lasers produce specific textures on titanium surfaces with micrometer-sized indentations that influence cell behavior. Materials and method: (CNC - CO2) red laser device; with a fundamental wavelength of λ=10600 nm and power pulses of 34 W were applied, and textures on the surface of titanium discs were achieved. Results: Excellent degrees of uniformity and repeatability were achieved for the desired portions of the surface by creating different surface textures. The surface topography and chemical composition of the specimens were investigated by scanning electron microscopy, electron dispersive spectroscopy, X-ray diffraction, and surface roughness measurements. Also, a laser power of 34 watts raised the surface roughness, Ra (1.71 nm), and Rz (1.99 nm). Conclusion: Titanium surface textures with unique qualities can be formed in response to an increased heat input. When excessive laser power was used, the measured roughness increased because of instantaneous re-melting. The use of a right continuous-wave (CNC - CO2) laser on titanium used in dental implants can form specific surface textures.
Effect of RGD/biomimetic Calcium Phosphate Coatings on Adhesion, Proliferation and Differentiation of MC3T3-E1 Cells on Titanium

Abstract:Objective:To evaluate the effect of biomimetic calcium phosphate coatings containing RGD on adhesion,proliferation and differentiation of MC3T3-E1 cells.Methods:A 2-step approach was adopted to prepare biomimetic calcium phosphate coatings on the surface of titanium.The characterization of the coatings was analyzed by scanning electron microscopy (SEM),Fourier-transform infrared spectroscopy,and X ray diffractionometry.The RGD was incorporated into coatings through co-precipitation.MC3T3-E1 cells were seeded on Ti,Ti/biomimetic calcium phosphate coatings,and Ti/RGD/biomimetic calcium phosphate coatings,respectively.The morphology of MC3T3-E1 cells cultured on samples was observed by SEM.The attachment of MC3T3-E1 cells on various samples was determined by crystal violet staining assay.The proliferation of MC3T3-E1 cells was determined by MTT after cultured for 1,3 and 7 days.The ALP activity of the MC3T3-E1 cells was measured after cells cultured on various samples for 3,7 and 14 days.Results:The results of SEM and crystal violet assay showed that the adhesion of cells growing on RGD/biomimetic calcium phosphate coatings was greater than that of control groups.Moreover,MTT test showed that the proliferation of cells growing on RGD/biomimetic calcium phosphate coatings was greater compared with that in control groups.In addition,the ALP activity of MC3T3-E1 cells cultured on RGD/biomimetic calcium phosphate coatings was higher than that in control groups.Conclusion:RGD/biomimetic calcium phosphate coatings can promote adhesion,proliferation,and differentiation of MC3T3-E1 cells.

Abstract:BACKGROUND: Selective laser melting (SLM) titanium is an ideal option to manufacture customized implants with suitable surface modification to improve its bioactivity. The peri-implant soft tissues form a protective tissue barrier for the underlying osseointegration. Therefore, original microrough SLM surfaces should be treated for favorable attachment of surrounding soft tissues.MATERIAL AND METHODS: In this study, anodic oxidation (AO) was applied on the microrough SLM titanium substrate to form TiO2 nanotube arrays. After that, calcium phosphate (CaP) nanoparticles were embedded into the nanotubes or the interval of nanotubes by electrochemical deposition (AOC). These two samples were compared to untreated (SLM) samples and accepted mechanically polished (MP) SLM titanium samples. Scanning electron microscopy, energy dispersive spectrometry, X-ray diffraction, surface roughness, and water contact angle measurements were used for surface characterization. The primary human gingival epithelial cells (HGECs) and human gingival fibroblasts (HGFs) were cultured for cell assays to determine adhesion, proliferation, and adhesion-related gene expressions.RESULTS: For HGECs, AOC samples showed significantly higher adhesion, proliferation, and adhesion-related gene expressions than AO and SLM samples (P<0.05) and similar exceptional ability in above aspects to MP samples. At the same time, AOC samples showed the highest adhesion, proliferation, and adhesion-related gene expressions for HGFs (P<0.05).CONCLUSION: By comparison between each sample, we could confirm that both anodic oxidation and CaP nanoparticles had improved bioactivity, and their combined utilization may likely be superior to mechanical polishing, which is most commonly used and widely accepted. Our results indicated that creating appropriate micro-/nano-topographies can be an effective method to affect cell behavior and increase the stability of the peri-implant mucosal barrier on SLM titanium surfaces, which contributes to its application in dental and other biomedical implants.

Effects of the micro-nano surface topography of titanium alloy on the biological responses of osteoblast

Abstract:In clinical applications, osseointegration is essential for the long-term stability of dental implants. Inspired by the hierarchical structure of natural bone, we applied the electrochemical etching (EC) technique to form a micro-nano structure on a titanium alloy (Ti6Al4V) substrate, called EC surface. Sand blasting and acid etching (SLA) and machined (M) methods were employed to generate micro and smooth textures, respectively, as the control groups. The surface topographies of the three substrates were characterized using scanning electron microscopy (SEM). Then, human osteoblast-like cells (MG63) were cultured on substrates, and adhesion, proliferation, morphology, alkaline phosphatase activity (ALP), and gene expression levels of Runt-related transcription factor 2 (RUNX2), osteocalcin (OCN), osteopontin (OPN), and type I collagen (COLIA 1) were analyzed. MG63 cells cultured on the EC Ti alloy substrates displayed better cell adhesion, significant proliferation, and a higher production level of ALP, gene expressions of RUNX2, OCN, OPN and COLIA 1 (p < 0.01 or p < 0.05) compared with those of SLA and M substrates. These results indicate that the micro-nano structure fabricated by electrochemical etching method is beneficial for the biological functions of MG63 cells and may be a promising candidate in dental implants. © 2016 Wiley Periodicals, Inc.
