Abstract:Observing microplastics in manipulative experiments is of paramount importance for understanding the fate of microplastics in the environment, organisms, and food webs. Labeling microplastics with fluorescent dyes is a useful tool in laboratory experiments for tracking microplastics. However, literature using fluorescence-based detection is heavily biased toward the use of polystyrene and polyethylene microspheres, potentially due to their commercial availability. Consequently, much less is understood about the fate of nonspherical morphologies and other types of plastics common in the environment. Presented here is a heat-mediated microplastic dyeing protocol that facilitates the stable incorporation of inexpensive commercially available fluorescent disperse dyes directly into the polymer structure for use in laboratory-based studies. A Rapid Method for Detecting Microplastics Based on Fluorescence Lifetime Imaging Technology (FLIM)

Abstract:With the increasing use and release of plastic products, microplastics have rapidly accumulated in ecological environments. When microplastics enter the food chain, they cause serious harm to organisms and humans. Microplastics pollution has become a growing concern worldwide; however, there is still no standardized method for rapidly and accurately detecting microplastics. In this work, we used fluorescence lifetime imaging technology to detect four kinds of Nile red-stained and unstained microplastics, and the unique phasor fingerprints of different microplastics were obtained by phasor analysis. Tracing the corresponding pixels of the “fingerprint” in the fluorescence lifetime image allowed for the quick and intuitive identification of different microplastics and their location distributions in a mixed sample. In our work, compared with staining the four microplastics with a fluorescent dye, using the phasor “fingerprint library” formed by the autofluorescence lifetimes of the microplastics was more easily distinguished than microplastics in the mixed samples. The feasibility of this method was further tested by adding three single substances—SiO2, chitin and decabromodiphenyl ethane (DBDPE), and surface sediments to simulate interferent in the environment, and the results providing potential applications for the identification and analysis of microplastics in complex environments.

Abstract:The fast, affordable, and standardized detection of microplastics (MP) remains one of the biggest challenges in MP research. Comparable data are essential for appropriate risk assessments and the implementation of laws and limit values. The fluorescent staining of MP in environmental samples is a possible solution to this problem. This study investigates the optimization of a sample preparation process (hydrogen peroxide digestion) and the staining process (temperature, concentration, time, surfactants as staining aids) for using a specifically developed fluorescent dye for MP detection. The optimization is performed by comparing the sample preparation process and staining of MP from different polymers and natural particles. Further, the suitability of the optimized process for the detection of fluoropolymers and tire abrasion was tested. The results show that the optimized method (increased temperature and optimized stain concentration) can detect microplastics reliably with a total sample preparation and measurement time of 2.5–3 h per sample, reaching recovery rates of 93.3% (polypropylene) to 101.7% (polyester). Moreover, two of the three tested fluoropolymers could be detected reliably. Tire abrasion could not be detected with the here presented method, as the black color leads to strong quenching. A long-term study measuring the MP pollution in the effluent of a municipal wastewater treatment plant compared the optimized and original processes and confirmed the stability of the improved method for routine measurements and contamination control.

Abstract:1. Plastic pollution represents a global environmental issue with relevant associated risks for ecosystem and human health. Fluorescence-based detection techniques allowed rapid tracking and identification of micro/nanoplastics (MNPs) in numerous studies by combining pre-treatment procedures, specific staining protocols, and advanced analytical instruments. However, despite an increasing scientific attention, the detection of plastic debris in the submicron and nano size range is still challenging when targeting environmental complex samples. In particular, the suitable approaches are not consistently harmonized and generally case/system-specific, with detrimental consequences on data reproducibility, accuracy, and reliability. This review is intended to provide state-of-the-art information on the existing and promising fluorescence-based approaches to assess the occurrence of MNPs in environmental samples of different origin. More specifically, we identified and discussed the most used fluorescent dyes, staining protocols, and analytical instruments for the fluorescence-based detection of MNPs. Advantages and disadvantages of the applied methodological approaches were critically presented, by highlighting their potentialities and challenges in view of future needs to improve the current scientific knowledge on spread and fate of MNPs in the environment. Overall, fluorescence-based techniques are offering promising results and unique analytical opportunities for the detection and quantification of MNPs in the environment.

Abstract:Microplastics (MPs) have recently emerged as a new major and ubiquitous environmental pollutant with still undefined, yet potentially high risks for human health and ecosystems. This has prompted growing public concern along with an increasing number of scientific studies. In particular, recent research has highlighted the need for a standardized methodology to monitor microplastics in different media, particularly in water. This study reports on the use of a new particle counter for the quantitative detection of MPs in water samples based on real-time analysis of fluorescence emissions. The instrument was calibrated using two types of plastic particles, i.e., polyvinyl chloride and high-density polyethylene, selected as examples of high- and low-density plastics, respectively. Specific solvents were used to match the different plastic densities. Measurements were also carried out on particles obtained from wastewater samples collected at the inlet and outlet of specific units of a municipal wastewater treatment plant after sieving, filtering, digestion with hydrogen peroxide to remove degradable organic matter and resuspension in the solvents employed during the calibration step. A Fourier transform infrared spectroscopy analysis performed on the same wastewater samples confirmed the presence of MPs, and, in particular, of polyethylene, in some of the samples in which the highest concentrations were measured applying the proposed method. Therefore, the novel particle counter described in this paper could represent a promising method to quantitatively measure MP concentrations in water samples.

Abstract:Recently, the increasing amount of plastic waste has raised concerns about microplastics in aquatic environments. In this study, microplastics between 0.1 and 5 mm in samples from different points of three Drinking Water Treatment Plants (DWTP) were separated, quantified, and identified. Staining methods were used in combination with microscopic and spectroscopic techniques. On the one hand, the Rose Bengal dye was used to discriminate between natural and synthetic particles. On the other hand, Nile Red and iDye Pink reagents have been evaluated for staining microplastics, providing them with fluorescence. The nature of the particles is determined by comparison with a series of patterns by means of an epifluorescence microscope. In addition, "Micro Fourier Transform Spectrophotometer " and "Raman Spectroscopy" were used to identify the nature of the particles more accurately. Concerning the results, polyester was the most common material by fluorescence identification, and it was confirmed with Raman spectroscopy. Since most of the particles observed were microfibres, Raman proved to be a better identification technique than μ-FTIR, which could only identify large fragments. In addition, the global elimination of microplastics (MPs) resulted notable in the three DWTPs, being 81.47% for A, 88.98% for B, and 82.27% for C, thus guaranteeing a higher quality of drinking water.

Abstract:Microplastics (MP) are micrometric plastic particles present in drinking water, food and the environment that constitute an emerging pollutant and pose a menace to human health. Novel methods for the fast detection of these new contaminants are needed. Fluorescence-based detection exploits the use of specific probes to label the MP particles. This method can be environmentally friendly, low-cost, easily scalable but also very sensitive and specific. Here, we present the synthesis and application of a new probe based on perylene-diimide (PDI), which can be prepared in a few minutes by a one-pot reaction using a conventional microwave oven and can be used for the direct detection of MP in water without any further treatment of the sample. The green fluorescence is strongly quenched in water at neutral pH because of the formation dimers. The ability of the probe to label MP was tested for polyvinyl chloride (PVC), polyethylene (PE), polyethylene terephthalate (PET), polypropylene (PP), polystyrene (PS), poly methyl methacrylate (PMMA) and polytetrafluoroethylene (PTFE). The probe showed considerable selectivity to PVC MP, which presented an intense red emission after staining. Interestingly, the fluorescence of the MP after labeling could be detected, under excitation with a blue diode, with a conventional CMOS color camera. Good selectivity was achieved analyzing the red to green fluorescence intensity ratio. UV–Vis absorption, steady-state and time-resolved fluorescence spectroscopy, fluorescence anisotropy, fluorescence wide-field and confocal laser scanning microscopy allowed elucidating the mechanism of the staining in detail.

Abstract:Microplastics are globally recognized as contaminants in freshwater and marine aquatic systems. To date there is no universally accepted protocol for isolation and quantification of microplastics from aqueous media. Various methodologies exist, many of which are time consuming and have the potential to introduce contaminants into samples, thereby obscuring characterization of the environmental microplastic load. Here, we present first steps in the detection of microplastics in liquid samples, based on their fluorescent staining followed by high throughput analysis and quantification using Flow Cytometry. Using controlled laboratory settings nine polymer types [polystyrene (PS); polyethylene (PE); polyethylene terephthalate (PET/PETE); high density polyethylene (HDPE); low density polyethylene (LDPE); polyvinyl chloride (PVC); polypropylene (PP); nylon (PA); polycarbonate (PC)] were tested for identification and quantification in freshwater. All nine plastic types were stained with 10 μg/mL Nile Red in 10% dimethyl sulfoxide with a 10 min incubation time. The lowest spatial detectable limit for plastic particles was 200 nm. Out of the nine polymer types chosen for the study PS, PE, PET, and PC were well-identified; however, results for other plastic types (PVC, PP, PA, LDPE, and HDPE) were masked to certain extent by Nile Red aggregation and precipitation. The methodology presented here permits identification of a range of particle sizes and types. It represents a significant step in the quantification of microplastics by replacing visual data interpretation with a sensitive and automated method.

Abstract:As an emerging marine pollutant, microplastics represent a focal point in global monitoring and management efforts. With seawater accounts for 97 % of the total global water resources, scientific assessments of microplastics in seawater are crucial for pollution control and management of marine environments. This study focuses on investigating microplastics in near-shore seawater and proposes a rapid and accurate detection method using a constructed confocal Raman spectroscopy detection system. By optimizing the pretreatment process of seawater microplastic samples, the efficient removal of organic matter interference in microplastic detection is achieved. Employing fluorescent labeling addresses the issues of prolonged detection time and high false positive rates associated with traditional methods, enabling rapid differentiation between microplastics and other substances and significantly enhancing detection efficiency and accuracy. Additionally, the use of differential Raman spectroscopy effectively mitigates fluorescence signal interference, thus improving the signal-to-noise ratio of the spectra. By employing dual-wavelength laser excitation at 784 nm/785 nm, microplastics such as polyethylene (PE), polypropylene (PP), polystyrene (PS) ranging in size from 60 to 500 μm are successfully detected in seawater. The results demonstrate that the proposed pretreatment method for seawater microplastics and novel detection techniques enable rapid screening and comprehensive non-destructive detection of microplastics in seawater, thereby facilitating the characterization of marine microplastics and providing scientific support for enhancing the management of microplastic pollution and ecological risk control.

Abstract:Although plastics have benefited our lives in terms of cost and convenience, the disposal of end-of-life plastics poses environmental problems, such as microplastics (MPs). Although the separation (e.g., filtration) and staining of MPs with fluorescent dye/solvent are generally accepted steps to observe MPs in an environmental matrix, in this study, an in situ selective fluorescent illumination of the MPs in water was attempted with the aid of surfactant. Nonpolar fluorescent dye in combination with surfactant affords nanometer-sized dye particles in water, which adsorb on MPs and penetrate the polymer matrix for effective staining and stable fluorescent behaviors. The effects of different staining parameters, including different dyes, surfactants, staining temperatures, staining times, dye/surfactant ratios, dye/MP ratios, and MP concentrations in aqueous solutions were investigated to better understand staining conditions. More interestingly, non-adsorbed free dye molecules in the staining solution were almost completely fluorescence-quenched by introducing the quenching agent, aniline, while the fluorescence intensity of the stained MP was maintained. By staining MPs with a dye/surfactant combination and subsequently quenching with aniline, in situ selective fluorescent illumination of the MPs in water was successfully achieved, which may eliminate the tedious separation/filtration procedure of MPs to accomplish the quick detection or monitoring of MPs.

Abstract:Microplastics are of rising health concerns because they have been detected even in remote and pristine environments, from the Artic snow to the Marianne Trench. The occurrence and impact of nanoplastics in ecosystems is almost unknown, in particular due to analytical limitations such as very small sizes that fall below detection limits of current techniques. Here we take advantage of a common interference in analytical flow cytometry to develop a method for the quantification of the number of plastic particles in the 0.6–15 μm size range. Plastic particles are stained with the lipophilic dye Nile-Red then detected by flow cytometry, a method regularly used in biology for rapid quantification of fluorescent cells. We found that sample analysis lasts 90 s, which is hundreds of times faster than the analysis of filter portions by micro-Raman and other spectroscopic techniques. Our method is highly efficient in detecting polyethylene, with staining efficiency higher than 70% and signal linearity with concentration. Staining efficiency up to 96% was observed for polyvinylchloride and for polystyrene.

Abstract:Detection of microplastics in environmental samples requires fast, sensitive and selective analytical techniques, both in terms of the size of the microparticles and their concentration. Single particle inductively coupled plasma mass spectrometry (SP-ICP-MS) allows the detection of plastic particles down to ca. 1 μm and down to concentrations of 100 particles per mL. In SP-ICP-MS, detection of carbon-containing particles is hampered by the presence of other forms of carbon (carbonates, organic matter, microorganisms...). An acidic pre-treatment of river water samples with 10% (v/v) nitric acid for 24 h allowed the reduction of the presence of dissolved carbon to ultrapure water levels and the digestion of potential microorganisms in the samples, recovering polystyrene microparticles up to 80%. Carbon-containing particles were detected in most of the samples analysed from Spanish and French Pyrenean rivers. The presence of microplastics in these samples was confirmed by Raman microscopy and their morphology was defined by electron microscopy combined with energy-dispersive X-ray spectroscopy. The developed SP-ICP-MS method is suitable for the rapid screening of river waters for the presence of microplastics, which can then be analysed by inherently slower but more selective techniques (e.g., Raman microscopy).

Abstract:In this study, we developed a method for the on-site selective detection and quantification of microplastics in various water matrices using fluorescence-tagged peptides combined with electrochemical impedance spectroscopy (EIS). Among the types of plastics found in seawater, polystyrene (PS) microplastics were selected. Fluorometry, scanning electron microscopy (SEM), and Raman spectroscopy were used to verify the specific interaction of these peptides with PS spherical particles of different sizes (ranging from 0.1 to 250 µm). Principal component analysis (PCA) was employed to determine the effects of temperature (25-65 °C), incubation time (5 and 10 min), and particle size on plastic-peptide bonding efficiency, based on fluorescence intensity. For each water type (pure, tap, NaCl (0.5 M), and seawater), EIS plots (Nyquist and Bode) were generated. Significant factors affecting the EIS response, including particle size, shape, and material, were analyzed by measuring electrical parameters for different microplastic concentrations (50 ppb to 20 ppm). The EIS parameters changed with increasing plastic concentration, determining a limit of detection (LOD) of 50 ppb (ng/mL) for pure and tap water and 400 ppb for saline water, as the lowest concentration producing a significant change in EIS parameters compared to the baseline. The sensor proved highly effective for detecting microplastics in low ionic strength environments such as pure and tap water. However, in high ionic strength environments like saline and seawater, the detection capability diminished, likely due to the masking effect of ions on the EIS response.
Fluorescent Tagging of Polymer Particles with PBN for the Detection of Microplastics in Personal Care Goods

Abstract:Objectives : Plastic pollution has been aggravated due to the massive use of disposable products and single-use face masks since the coronavirus disease (COVID-19) pandemic. There is growing interest regarding detection and identification of plastic pollution in daily life. The aim of this study is (1) to investigate suitability of a fluorescent labelling method for the detection of polymers, (2) to detect microplastics in personal care goods, and (3) to identify the detected plastic particles.Methods : In order to detect plastic particles in real samples, a 1-pyrenebutyric acid N-hydroxysuccinimidyl ester (PBN) fluorophore was used as a dye for staining plastic particles. First, the capability of PBN was evaluated by staining a model polymer of polyethylene (PE) particles, and its desorption and photobleaching was also investigated. Using the optimum staining condition, microplastics were detected in a laboratory-controlled sample and commercially available products, specifically in cleansers, scrubs, and toothpastes. Finally, the detected microplastics were identified using an attenuated total reflection Fourier transform infrared spectroscopy (ATR FT-IR) analysis.Results and Discussion : The PE particles fluoresced blue light after being stained with PBN at a concentration of 100 μg/mL for 3-5 min without post treatments (e.g., fixation and drying), but the particles should be detected within 24 h due to the photobleaching of PBN. The desorption of PBN fluorophore occurred when the stained PE particles were washed with 25% acetone rather than rinse-off with deionized (DI) water. All of the tested personal care goods contained plastic particles and various sizes and shapes (e.g., spherical, rounded, and irregular shapes) of particles were found in different types and brands of products. The detected particles were identified as PE, polypropylene (PP), polycarbonate (PC), ethylene vinyl acetate (EVA), polystyrene (PS), and polyvinyl chloride (PVC) polymers.Conclusions : The PBN was shown to effectively stain polymer particles and it allowed visual identification of microplastics in personal care goods. Various types and sizes of polymers were included into the commercially available products as ingredients, indicating a primary source of plastic pollution in aquatic environment.

Abstract:This paper presents the results of a research aimed at establishing a novel method for the detection of primary and secondary micro- and nanoplastics (MNPs), by using the fluorescence properties of the dye Nile Red-n-heptane (NR-H). The method has been applied to the detection of laboratory degraded polymers (Polystyrene, PS and Polyethylene Terephthalate, PET) as well as traceable latex microspheres in aqueous environments, showing a remarkable detection capacity and avoiding the prior extraction or processing of MNPs in natural samples, with significant time savings compared to conventional methods. The study has been carried out on various types of water, including samples from wastewater treatment plants, boreholes, seawater and synthesized seawater. The effectiveness of the staining process was evaluated by scanning electron microscopy (SEM), dynamic light scattering (DLS) and optical microscopy. As a result, a novel standardizable protocol for the rapid detection of MNPs has been established, with the potential to improve environmental protection through fast in-situ detection and identification of plastic contaminants. The limitations of the protocol in the quantification of MNPs have also been identified and further studies are proposed to overcome these limitations.

Abstract:Staining microplastics (MPs) for fluorescence detection has been widely applied in MP analyses. However, there is a lack of standardized staining procedures and conditions, with different researchers using different dye concentrations, solvents, incubation times, and staining temperatures. Moreover, with the limited types and morphologies of commercially available MPs, a simple and optimized approach to making fluorescent MPs is needed. In this study, 4 different textile dyes, along with Nile red dye for comparison, are used to stain 17 different polymers under various conditions to optimize the staining procedure. The MPs included both virgin and naturally weathered polymers with different sizes and shapes (e.g., fragments, fibers, foams, pellets, beads). We show that the strongest fluorescence intensity occurred with aqueous staining at 70 °C for 3 h with a dye concentration of 5 mg/mL, 55 mg/mL, and 2 μg/mL for iDye dyes, Rit dyes, and Nile red, respectively. Red fluorescent signals are stronger and thus preferred over green ones. The staining procedure did not significantly alter the surface, mass, and chemical characteristics of the particles, based on FTIR and stereomicroscopy. Stained MPs were spiked into freshwater, saltwater, a sediment slurry, and wastewater-activated sludge; even after several days, the recovered particles are still strongly fluoresced. The approach described herein for producing customized fluorescent MPs and quantifying MPs in laboratory-controlled experiments is both straightforward and simple.

Development of an Inexpensive and Comparable Microplastic Detection Method Using Fluorescent Staining with Novel Nile Red Derivatives

Abstract:Fluorescent staining of microplastics as a detection method is consistently gaining importance in microplastics research, as it is fast, easy to use, and requires low technical effort. In this study, a complete procedure was developed, from sample collection to sample processing and detection, to measure microplastics with low cost and time requirements. The developed procedure was tested by measuring the microplastics in the effluent of a German wastewater treatment plant over a period of one year. The results show that the process is especially well suited to investigate temporal variations of microplastic contamination, which requires a large number of samples to be processed. Further, the precision and selectivity of the detection process could be improved by applying newly developed Nile red derivatives for fluorescent staining. A low budget modification of a microscope for fluorescent imaging is compared to a modification with precise optical bandpass filters. A script enabling automated microplastic detection and counting was developed, improving the accuracy and comparability of the process.

Abstract:Plastic pollution has become a global challenge, affecting water quality and health. Plastics including polystyrene (PS), polyvinyl chloride (PVC), polypropylene (PP), polyethylene terephthalate (PET), and high-density polyethylene (HDPE), are significant contributors to environmental pollution. With the growing need for investigation and detection of plastics found in natural waters, we propose the use of a portable laser diode (LD)-based fluorescence lidar system for in-situ detection of plastic litters in surface waters based on excitation-emission fluorescence spectroscopic data. The experiments were carried out in a controlled environment using a fluorescence lidar system with 405 nm excitation wavelength to determine the fluorescence signals of several plastics at 470 nm emission wavelength. Simultaneous detection of PET plastic and Chlorella vulgaris were also observed to determine the fluorescence influence of chlorophyll in surface waters. Attenuated total reflection Fourier transform infrared (ATR-FTIR) spectroscopy was employed to study the chemical composition of the plastics used before and after being submerged in the water. Scanning electron microscopy (SEM) and high-resolution camera microscopy were used to analyze the morphology of the submerged PET samples. This study provides a basis for a new in-situ technique using a fluorescence lidar system for submerged or transparent plastics in surface waters.

Abstract:During the last years, microplastics in the environment came to the fore in environmental science research. For an appropriate risk assessment, it is essential to know the levels of microplastic contamination in the environment. In the field of microplastic detection, extensive research has been carried out in recent years. While common methods such as Raman spectroscopy and pyrolysis GC-MS are time-consuming and require trained staff and expensive equipment, there is the need for a cheap and easily applicable method. Staining microplastics with the fluorescent dye Nile red (NR) has a high potential to fulfill these criteria. In our work, we tested Nile red and newly developed derivatives, with the aim of achieving greater selectivity for plastic particles and more intense fluorescence. In addition, the influence of using different solvents and water at different pH values in the dyeing process was investigated by analyzing solid sample fluorescence spectra of dyed microplastics and natural particles. Finally, the method developed from the acquired knowledge was tested for sea salt.
