Abstract:Per- and polyfluoroalkyl substances (PFAS), that are widely used in different industrial and commercial activities, have become emerging contaminants of concern. Thus, the evaluation of PFAS presence in diverse environmental matrices is of paramount importance, leading to the necessity in development of different types of analytical methods to analyse the trace amounts of PFAS with varying selectivity and sensitivity. Analytical methods that have been advanced and employed for PFAS identification are critically described with their strengths, limitations and future potentials in this review. This review includes, (i) instrument analytical methodologies, mainly focusing on sample preparation, extraction and recovery analysis, (ii) nanoparticle-based sensors, and an app-based smartphone portable sensor, for the monitoring and sensing of PFAS in different environmental matrices. Furthermore, (iii) total oxidisable precursor assay (TOPA), content of total organic fluorine (TOF) as well as total fluorine (TF) are also reviewed, which provide fundamental tools towards achieving precursor quantification and closing mass balance analysis. Abstract:Per- and polyfluoroalkyl substances (PFAS) are a group of over 4700 heterogeneous compounds with amphipathic properties and exceptional stability to chemical and thermal degradation. The unique properties of PFAS compounds has been exploited for almost 60 years and has largely contributed to their wide applicability over a vast range of industrial, professional and non-professional uses. However, increasing evidence indicate that these compounds represent also a serious concern for both wildlife and human health as a result of their ubiquitous distribution, their extreme persistence and their bioaccumulative potential. In light of the adverse effects that have been already documented in biota and human populations or that might occur in absence of prompt interventions, the competent authorities in matter of health and environment protection, the industries as well as scientists are cooperating to identify the most appropriate regulatory measures, substitution plans and remediation technologies to mitigate PFAS impacts. In this review, starting from PFAS chemistry, uses and environmental fate, we summarize the current knowledge on PFAS occurrence in different environmental media and their effects on living organisms, with a particular emphasis on humans. Also, we describe present and provisional legislative measures in the European Union framework strategy to regulate PFAS manufacture, import and use as well as some of the most promising treatment technologies designed to remediate PFAS contamination in different environmental compartments.

Abstract:Per- and polyfluoroalkyl substances (PFAS) is an emerging class of organic pollutants of concern and is now prevalent in environmental matrices including water, soil, air, and biological. So far, several standard analytical methods have been developed to systematically analyze PFAS in different environmental matrices. However, the complexity of environmental matrices makes the effective extraction of PFAS difficult, and the legacy PFAS is gradually changing into a new PFAS with short chain and unknown structure in production, which makes the analysis of PFAS challenging. In this review, the following aspects are summarized: (1) the advances in standard analytical methods for PFAS in different environmental matrices, and further generalizes the updating novel extraction and detection methods; (2) the analysis of unknown PFAS, the suspect and non-targeted screening analysis method of PFAS based on high-resolution mass spectrometry (HRMS) is systematically described.

Abstract:Here we synthesize current understanding of the magnitudes and methods for assessing human and wildlife exposures to poly‐ and perfluoroalkyl substances (PFAS). Most human exposure assessments have focused on two to five legacy PFAS and wildlife assessments are typically limited to targeted PFAS (up to ~30 substances). However, shifts in chemical production are occurring rapidly and targeted methods for detecting PFAS have not kept pace with these changes. Total fluorine (TF) measurements complemented by suspect screening using high resolution mass spectrometry are thus emerging as essential tools for PFAS exposure assessment. Such methods enable researchers to better understand contributions from precursor compounds that degrade into terminal perfluoroalkyl acids (PFAA). Available data suggest that diet is the major human exposure pathway for some PFAS but there is large variability across populations and PFAS compounds. Additional data on TF in exposure media and the fraction of unidentified organofluorine are needed. Drinking water has been established as the major exposure source in contaminated communities. As water supplies are remediated, and for the general population, exposures from dust, personal care products, indoor environments and other sources may be more important. A major challenge for exposure assessments is the lack of statistically representative population surveys. For wildlife, bioaccumulation processes differ substantially between PFAS and neutral lipophilic organic compounds, prompting a revaluation of traditional bioaccumulation metrics. There is evidence that both phospholipids and proteins are important for the tissue partitioning and accumulation of PFAS. New mechanistic models for PFAS bioaccumulation are being developed that will assist in wildlife risk evaluations.This article is protected by copyright. All rights reserved.

Global performance and trends of research on per- and polyfluoroalkyl substances (PFASs) between 2001 and 2018 using bibliometric analysis

Abstract:Per- and polyfluoroalkyl substances (PFASs) are widely used in food packaging, non-stick pots, and surfactants. However, their persistence in the environment, hazardous nature, and potential for bioaccumulation and longrange transport have alarmed an increasing number of scholars and research institutions. Although several literature reviews on PFASs research exist, only a few of them have considered bibliometric indices. In this study, 3,373 PFASs-related articles published between 2001 and 2018 were analyzed using a bibliometric analysis method based on the Science Citation Index (SCI) Expanded. The software tools for mapping knowledge domain (MKD) (VOSviewer and Science of Science (Sci2)) were used to analyze the performance of contributors and PFASs research topics, hotspots, and trends. Our results reveal that the number of PFASs-related articles published annually has increased significantly, with most originating from the United States (followed by those from China). The Chinese Academy of Sciences has published the most articles. A comprehensive analysis of title, keywords, and keywords plus showed that PFASs research hotspots include humans, precursors, and detection methods, with the main focuses being environmental science, toxicology, and environmental engineering. The four main research topics of PFASs were identified, and a literature review was carried out for each one. Overall, this study can supply researchers with a deeper understanding of the development of PFASs studies and provide a comprehensive data reference for researchers to further grasp the research direction in this field.

Abstract:The increasingly stringent restrictions on legacy per- and polyfluoroalkyl substances (PFASs) have led to the compensatory use of new fluorinated replacements. These new compounds, hereafter referred to as emerging PFASs, continue to be discovered and are now showing ubiquity in abiotic and biotic environments. Thus, there is an urgent need for robust, yet sensitive analytical methods to determine their occurrence and understand their behavior, fate, impact, and toxicity. Here, we review the up-to-date sample preparation and analytical methodologies for emerging PFASs based on peer-reviewed literature published in the past three years (2015–2018). The determination of emerging PFASs is similar to that of legacy PFASs, with satisfactory performances for most emerging PFASs achieved using conventional extraction and analytical approaches. However, the determination of certain specific analytes remains challenging due to the unavailability of standards and reference materials, low recoveries and matrix effects, background contamination, and poor sensitivities due to in-source fragmentation. Despite recent progress in identifying ionic semi-volatile PFASs with liquid chromatograph-high-resolution mass spectrometry (LC-HRMS), our knowledge on new types of neutral volatile PFASs remains poor due to limited non-target analysis using gas chromatograph (GC)-HRMS.
Method development and evaluation for the determination of perfluoroalkyl and polyfluoroalkyl substances in multiple food matrices

Abstract:A method for the determination of 21 perfluorinated and 10 polyfluorinated alkyl substances (PFAS) was developed for application in different food matrices. Acetonitrile was used as the extraction solvent with solid phase extraction weak anion-exchange (SPE-WAX) clean up, with LC-MS/MS analysis using both surrogate and performance standards to correct for losses during sample preparation and matrix effects. The method has been evaluated in four different matrices (fish, pizza, chicken nuggets and spinach). Originally, the focus was to develop a method for foods commonly thought to be a source of PFASs (e.g. fish). It was expanded to include foods where PFAS exposure would be possible through their presence in grease-proof food packaging (e.g. pizza, chicken nuggets). Vegetables (lettuce) and fruit (tomato) have recently been considered as part of proficiency testing programmes, so the inclusion of some testing in a vegetable matrix (i.e. spinach) was also added to the testing. Limits of quantification ranged from 0.018 ng g-1 (L-PFDS) to 5.28 ng g-1 (FHEA), although method quantification limits for PFBA (12.4 ng g-1), 6:2 PAP (8.96 ng g-1) and 8:2 PAP (3.49 ng g-1) were elevated above instrumental limits owing to their consistent detection in reagent blank samples. PFAS analyses were strongly impacted by matrix, therefore the use of isotopically labelled internal standards was critical to the development of accurate results. The accuracy of the method using numerous proficiency testing schemes or interlaboratory comparison studies has shown the developed method to be successful with z-scores for all concerned analytes in all test matrices remaining within ±2.0, with the exception of PFBA in wheat flour which was -2.4.

Abstract:A population of stem-like cells in tumors, the so-called cancer stem cells (CSCs), are being held responsible for therapy resistance and tumor recurrence. In analogy with normal stem cells, CSCs possess the capacity of long term self-renewal and multilineage differentiation. CSCs are believed to be more resistant to various therapies compared to their differentiated offspring and therefore the cause of tumor relapse. Markers for CSCs have been identified using xenograft transplantation assays and lineage tracing in mouse models, however the specificity and validity of many of these markers is under debate. Recently, low proteasome activity has been postulated as a novel CSC marker. In several solid malignancies a small subset of low proteasomal activity cells with CSC characteristics were identified, suggesting that proteasomal activity might be a functional marker for CSCs. In this perspective, we will discuss a recent study by Munakata et al., describing a population of colorectal cancer cells with CSC properties, characterized by low proteasome activity and treatment resistance. We will put this finding in a broader view by discussing the challenges and issues inherent with CSC identification, as well as some emerging insights in the CSC concept.

Abstract:Emerging persistent organic pollutants (ePOPs) include polybrominated diphenyl ethers (PBDEs) and perfluorooctane sulfonyl fluoride/perfluorooctane sulfonate (POSF/PFOS), which are newly listed in the Stockholm Convention. Other ePOPs, which have not been regulated, include organophosphate flame retardants (PFRs), novel brominated flame retardants (NBFRs) and other perfluoroalkyl substances (PFASs). Often ePOPs data related to occurrence, toxicity, impact or environmental behavior are insufficient or inadequate because of the lack of proper analytical methods to obtain them. Thus, a critical review of the analytical procedures proposed in the last six years (2011–2017) for determining ePOPs by chromatographic methods in the different compartments of the aquatic ecosystems is presented. The overall analytical procedure, from sampling to final determination, is emphasized presenting recent developments in the extraction, pre-concentration, and instrumental detection needed for the accurate quantification of ePOPs in environmental samples. Finally, this review examines the basic challenges we face in order to anticipate future directions and urgent needs of this field.

Perfluoroalkyl substances: a risk for the aquatifc environment? A 1-year case study in river waters of central Italy

Abstract:Perfluoroalkyl substances (PFASs) are a large class of persistent emerging pollutants, ubiquitous in different environmental compartments. In this study, twenty-one PFASs were determined in seventy-eight water samples collected from six different rivers in the Umbria region (central Italy) during a 13-month monitoring campaign. The sum of the twenty-one target analytes (Σ 21 PFASs) ranged from 2.0 to 68.5 ng L −1 , with a mean value of 22.0 ng L −1 . The highest concentrations of Σ 21 PFASs were recorded in the warmest months (from June to September) due to reduced river streamflow caused by low rainfall and high temperatures. PFASs with a number of carbon atoms between four and nine prevail over C10–C18 congeners due to their higher water solubility and to their increased use in industry. PFBA, followed by PFPeA, PFHxA, and PFOA, was the most abundant congeners detected in the analyzed river water samples. Finally, the calculation of risk quotients (∑RQs) has allowed to assess the risk for three aquatic organisms (fish, algae, and daphnid) deriving from the exposure to PFASs. The survey showed that the risk for the three aquatic organisms during the four seasons and throughout the year was always negligible. The only exception was a low risk for fish and daphnid in GEN river considering the annual exposure.

Abstract:The ubiquity of perfluoroalkyl substances has raised concerns about the unintended consequences of PFAS exposure on human health. In the present study, an eco-friendly ultra-performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) method was developed for the simultaneous determination of 17 PFAS in human serum and semen samples. QuEChERS salts MgSO 4 :NaCl 4:1 (w/w) were used for the extraction. The separation of analytes was performed on an ACQUITY BEH C 18 column (100 × 2.1 mm, 1.7 μm), using water:methanol 95:5 and methanol as mobile phases A and B, respectively, both containing 2 mM ammonium acetate. Multiple reaction monitoring (MRM) in negative ion mode was used, selecting two transitions for each analyte, except for perfluorobutanoic acid (PFBA) and perfluoropentanoic acid (PFPeA). The analytical method was validated according to the Organization of Scientific Area Committees (OSAC) for Forensic Sciences guidelines and AGREE approach software was used to evaluate the greenness of the method. The developed procedure was applied to the analysis of 10 paired human serum and semen samples, proving the suitability in high throughput laboratories due to the easy preparation and the reduced volume of toxic solvents. Moreover, it allows to perform further investigation on the correlation between serum and semen PFAS concentration, focusing on male reproductive system correlated pathologies, such as male infertility.

Abstract:To date, poly- and perfluoroalkyl substances (PFAS) represent a real threat for their environmental persistence, wide physicochemical variability, and their potential toxicity. Thus far a large portion of these chemicals remain structurally unknown. These chemicals, therefore, require the implementation of complex non-targeted analysis workflows using liquid chromatography coupled with high-resolution mass spectrometry (LC-HRMS) for their comprehensive detection and monitoring. This approach, even though comprehensive, does not always provide the much-needed analytical resolution for the analysis of complex PFAS mixtures such as fire-fighting aqueous film-forming foams (AFFFs). This study consolidates the advantages of the LC×LC technique hyphenated with high-resolution tandem mass spectrometry (HRMS/MS) for the identification of PFAS in AFFF mixtures. A total of 57 PFAS homolog series (HS) were identified in 3M and Orchidee AFFF mixtures thanks to the (i) high chromatographic peak capacity ( n' 2D,c ~ 300) and the (i) increased mass domain resolution provided by the "remainder of Kendrick Mass" (RKM) analysis on the HRMS data. Then, we attempted to annotate the PFAS of each HS by exploiting the available reference standards and the FluoroMatch workflow in combination with the RKM defect by different fluorine repeating units, such as CF 2 , CF 2 O, and C 2 F 4 O. This approach resulted in 12 identified PFAS HS, including compounds belonging to the HS of perfluoroalkyl carboxylic acids (PFACAs), perfluoroalkyl sulfonic acids (PFASAs), (N-pentafluoro(5)sulfide)-perfluoroalkane sulfonates (SF5-PFASAs), N -sulfopropyldimethylammoniopropyl perfluoroalkane sulfonamides (N-SPAmP-FASA), and N -carboxymethyldimethylammoniopropyl perfluoroalkane sulfonamide (N-CMAmP-FASA). The annotated categories of perfluoroalkyl aldehydes and chlorinated PFASAs represent the first record of PFAS HS in the investigated AFFF samples.

Non-targeted metabolomics and associations with per- and polyfluoroalkyl substances (PFAS) exposure in humans: A scoping review

Abstract:OBJECTIVE: To summarize the application of non-targeted metabolomics in epidemiological studies that assessed metabolite and metabolic pathway alterations associated with per- and polyfluoroalkyl substances (PFAS) exposure.RECENT FINDINGS: Eleven human studies published before April 1st, 2021 were identified through database searches (PubMed, Dimensions, Web of Science Core Collection, Embase, Scopus), and citation chaining (Citationchaser). The sample sizes of these studies ranged from 40 to 965, involving children and adolescents (n = 3), non-pregnant adults (n = 5), or pregnant women (n = 3). High-resolution liquid chromatography-mass spectrometry was the primary analytical platform to measure both PFAS and metabolome. PFAS were measured in either plasma (n = 6) or serum (n = 5), while metabolomic profiles were assessed using plasma (n = 6), serum (n = 4), or urine (n = 1). Four types of PFAS (perfluorooctane sulfonate(n = 11), perfluorooctanoic acid (n = 10), perfluorohexane sulfonate (n = 9), perfluorononanoic acid (n = 5)) and PFAS mixtures (n = 7) were the most studied. We found that alterations to tryptophan metabolism and the urea cycle were most reported PFAS-associated metabolomic signatures. Numerous lipid metabolites were also suggested to be associated with PFAS exposure, especially key metabolites in glycerophospholipid metabolism which is critical for biological membrane functions, and fatty acids and carnitines which are relevant to the energy supply pathway of fatty acid oxidation. Other important metabolome changes reported included the tricarboxylic acid (TCA) cycle regarding energy generation, and purine and pyrimidine metabolism in cellular energy systems.CONCLUSIONS: There is growing interest in using non-targeted metabolomics to study the human physiological changes associated with PFAS exposure. Multiple PFAS were reported to be associated with alterations in amino acid and lipid metabolism, but these results are driven by one predominant type of pathway analysis thus require further confirmation. Standardizing research methods and reporting are recommended to facilitate result comparison. Future studies should consider potential differences in study methodology, use of prospective design, and influence from confounding bias and measurement errors.

Ultrashort pfas: a review of the topic

Abstract:Background: Ultrashort perfluoroalkyl substances (UsPFAS), characterized by carbon chain lengths of 2 to 4 atoms, represent a distinct subgroup within the broader PFAS class of chemicals. Despite their lower molecular weight, ultrashort PFAS have gained significant attention due to their widespread presence. This article provides a comprehensive review of the topic. Methods: We analyzed 33 articles published between 2017 and 2024 found on Pubmed using "ultrashort PFAS" as keyword. We compared: 1- environmental matrices in which ultrashort PFAS were found, 2- different distribution patterns, 3- degradation pathways, to better understand whether they may resemble longer-chain PFAS, which are known for their bioaccumulation and persistence as "forever chemicals". Conclusions: The occurrence of ultrashort PFAS has been documented in different environmental matrices (including surface waters, waste waters, precipitations, sediments, soils, and also human serum and urine) indicating widespread environmental exposure. Advanced analytical techniques, such as liquid chromatography-tandem mass spectrometry (LC-MS/MS) and gas chromatography-mass spectrometry (GC-MS), have enabled the sensitive detection and quantification of these compounds at trace levels. Studies keep finding new ultrashort PFAS and have focused on elucidating their sources and fate pathways to better understand their behavior in natural systems. The toxicity of ultrashort PFAS remains an area of active in-vestigation. While less is known about their health effects, studies suggest potential adverse impacts on organisms and ecosystems. This underscores the importance of continued research to assess the eco-logical consequences of ultrashort PFAS exposure and bring up risk management strategies.

Abstract:Perfluorinated and polyfluorinated alkyl substances (PFAS), more than 4700 in number, are a group of widely used man-made chemicals that accumulate in living things and the environment over time. They are known as "forever chemicals" because they are extremely persistent in our environment and body. Because PFAS have been widely used for many decades, their presence is evident globally, and their persistence and potential toxicity create concern for animals, humans and environmental health. They can have multiple adverse health effects, such as liver damage, thyroid disease, obesity, fertility problems, and cancer. The most significant source of living exposure to PFAS is dietary intake (food and water), but given massive industrial and domestic use, these substances are now punctually present not only domestically but also in the outdoor environment. For example, livestock and wildlife can be exposed to PFAS through contaminated water, soil, substrate, air, or food. In this review, we have analyzed and exposed the characteristics of PFAS and their various uses and reported data on their presence in the environment, from industrialized to less populated areas. In several areas of the planet, even in areas far from large population centers, the presence of PFAS was confirmed, both in marine and terrestrial animals (organisms). Among the most common PFAS identified are undoubtedly perfluorooctanesulfonate (PFOS) and perfluorooctanoic acid (PFOA), two of the most widely used and, to date, among the most studied in terms of toxicokinetics and toxicodynamics. The objective of this review is to provide insights into the toxic potential of PFAS, their exposure, and related mechanisms.

Abstract:Per- and polyfluoroalkyl substances (PFASs) have emerged as a prominent environmental pollutant in recent years, primarily due to their tendency to accumulate and magnify in both the environment and living organisms. The entry of PFASs into the environment can have detrimental effects on human health. Hence, it is crucial to actively monitor and detect the presence of PFASs. The current standard detection method of PFAS is the combination of chromatography and mass spectrometry. However, this requires expensive instruments, extra sample pretreatment steps, complicated operation and long analysis time. As a result, new methods that do not rely on chromatography and mass spectrometry have been developed and applied. These alternative methods mainly include optical and electrochemical sensor methods, which offer great potential in terms of real-time field detection, instrument miniaturization, shorter analysis time, and reduced detection cost. This review provides a summary of recent advancements in PFAS detection sensors. We categorize and explain the principles and mechanisms of these sensors, and compare their limits of detection and sensitivity. Finally, we discuss the future challenges and improvements needed for PFAS sensors, such as field application, commercialization, and other related issues.

Abstract:PFASs (perfluoroalkyl and polyfluoroalkyl substances) are highly fluorinated, aliphatic, synthetic compounds with high thermal and chemical stability as well as unique amphiphilic properties which make them ingredients in a range of industrial processes. PFASs have attracted consideration due to their persistence, toxicity and bioaccumulation tendency in the environment. Recently, attention has begun to be addressed to shorter-chain PFASs, such as perfluorohexane sulfonate [PFHxS], apparently less toxic to and more easily eliminated from lab animals. However, short-chain PFASs represent end-products from the transformation of fluorotelomers whose biotic breakdown reactions have not been identified to date. This means that such emergent pollutants will tend to accumulate and persist in ecosystems. Since we are just learning about the interaction between short-chain PFASs and microorganisms, this study reports on the response to PFHxS of two Pseudomonas sp. strains isolated from environmental matrices contaminated by PFASs. The PFHxS bioaccumulation potential of these strains was unveiled by exploiting different physiological conditions as either axenic or mixed cultures under alkanothrofic settings. Moreover, electron microscopy revealed nonorthodox features of the bacterial cells, as a consequence of the stress caused by both organic solvents and PFHxS in the culturing substrate.

Abstract:To date, poly- and perfluoroalkyl substances (PFASs) represent a real threat for both their environmental persistence, wide physicochemical variability, and their potential toxicity. Thus far a large portion of these chemicals remain structurally unknown. These chemicals, therefore, require the implementation of complex non-targeted analysis workflows using high resolution mass spectrometry coupled with liquid chromatography (LC-HRMS) for their comprehensive detection and monitoring. This approach, even though comprehensive, does not always provide the much needed analytical resolution for the analysis of complex PFAS mixtures such as fire-fighting aqueous-film forming foams (AFFFs). This study consolidates the advantages of LC×LC technique hyphenated with high-resolution tandem mass spectrometry (HRMS/MS) for the identification of PFASs in AFFF mixtures. A total of 57 PFAS homologue series (HSs) were identified in 3M and Orchidee AFFF mixtures thanks to the (i) high chromatographic peak capacity (n’2D,c~300) and the (i) increased mass domain resolution provided by the “remainder of Kendrick Mass” (RKM) analysis on the HRMS data. Then, we attempted to annotate PFASs of each HSs exploiting the available reference standards and the FluoroMatch workflow in combination with the RKM defect by different fluorine repeating units, such as CF2, CF2O, and C2F4O. This approach resulted in 12 identified PFAS HSs, including compounds belonging to the HSs of PFACAs, PFASAs, SF5-PFASAs, N-SPAmP-FASA, and N-CMAmP-FASA. The annotated categories of perfluoroalkyl aldehydes, and chlorinated PFASAs represent a first record of PFAS HSs in the investigated AFFF samples.

Abstract:Per- and polyfluoroalkyl substances (PFAS) have been manufactured for over seven decades. Owing to the unique characteristics of C-F bond, it has been widely used in the industrial and commercial products, which leading to its wide presence in the environmental and biological matrix. In this critical review, we systematically summarized the existing literature on the behaviors and mechanisms of bioaccumulation, transportation, and elimination of PFAS in humans and pointed out the deficiencies and gaps of the existing research. Additionally, we calculated the binding affinities between a total of 56 PFAS and four important renal PFAS transporters (organic anion transporter 1, 3, and 4, together with urate transporter 4) to elucidate the renal elimination behaviors of PFAS in human body by molecular docking method for the first time. Furthermore, we put forwarded perspectives for future research to call on researchers to fill in the gaps of the existing studies and increase the investigations on the research topic. The understanding of the behaviors and mechanisms of PFAS in human beings are significant for the health risks assessment of human exposed to PFAS. Nevertheless, the research on the topic is still very limited. This review provides a comprehensive view of PFAS in human body, explores the potential human renal elimination mechanisms of PFAS, and presents insightful perspectives for further research on the relationship between PFAS exposure and its potential health effects on human beings. Abstract:Perfluorooctane sulfonate (PFOS) found extensive use for over 60 years up until its restriction in the early 2000s, culminating in its listing under the Stockholm Convention on Persistent Organic Pollutants (POPs) in 2009. Efforts to minimise human body burdens are hindered by uncertainty over their precise origins. While diet appears the principal source for the majority of western populations (with other pathways like dust ingestion, drinking water and inhalation also important contributors); the role played by exposure to PFOS-precursor compounds followed by in vivo metabolism to PFOS as the ultimate highly stable end-product is unclear. Such PFOS-precursor compounds include perfluorooctane sulfonamide derivates, e.g., perfluorooctane sulfonamides (FOSAs) and sulfonamidoethanols (FOSEs). Understanding the indirect contribution of such precursors to human body burdens of PFOS is important as a significant contribution from this pathway would render the margin of safety between the current exposure limits and estimates of external exposure to PFOS alone, narrower than hitherto appreciated. Estimates derived from mathematical modelling studies, put the contribution of so-called "precursor exposure" at between 10% and 40% of total PFOS body burdens. However, there are substantial uncertainties associated with such approaches. This paper reviews current understanding of human exposure to PFOS, with particular reference to recent research highlighting the potential of environmental forensics approaches based on the relative abundance and chiral signatures of branched chain PFOS isomers to provide definitive insights into the role played by "precursor exposure".
