Abstract:To date, poly- and perfluoroalkyl substances (PFASs) represent a real threat for both their environmental persistence, wide physicochemical variability, and their potential toxicity. Thus far a large portion of these chemicals remain structurally unknown. These chemicals, therefore, require the implementation of complex non-targeted analysis workflows using high resolution mass spectrometry coupled with liquid chromatography (LC-HRMS) for their comprehensive detection and monitoring. This approach, even though comprehensive, does not always provide the much needed analytical resolution for the analysis of complex PFAS mixtures such as fire-fighting aqueous-film forming foams (AFFFs). This study consolidates the advantages of LC×LC technique hyphenated with high-resolution tandem mass spectrometry (HRMS/MS) for the identification of PFASs in AFFF mixtures. A total of 57 PFAS homologue series (HSs) were identified in 3M and Orchidee AFFF mixtures thanks to the (i) high chromatographic peak capacity (n’2D,c~300) and the (i) increased mass domain resolution provided by the “remainder of Kendrick Mass” (RKM) analysis on the HRMS data. Then, we attempted to annotate PFASs of each HSs exploiting the available reference standards and the FluoroMatch workflow in combination with the RKM defect by different fluorine repeating units, such as CF2, CF2O, and C2F4O. This approach resulted in 12 identified PFAS HSs, including compounds belonging to the HSs of PFACAs, PFASAs, SF5-PFASAs, N-SPAmP-FASA, and N-CMAmP-FASA. The annotated categories of perfluoroalkyl aldehydes, and chlorinated PFASAs represent a first record of PFAS HSs in the investigated AFFF samples.

What problem does this paper attempt to address?
The problem this paper attempts to address is: how to efficiently and comprehensively detect and identify polyfluoroalkyl and perfluoroalkyl substances (PFASs) in complex fluorinated surfactant (AFFF) mixtures. Specifically, the paper improves the detection and annotation capabilities of PFASs by integrating two-dimensional liquid chromatography with high-resolution tandem mass spectrometry (LC×LC-HRMS/MS) technology. ### Main Issues 1. **Complexity and Unknown Nature of PFASs**: - PFASs have become a major environmental issue due to their environmental persistence and wide range of physicochemical properties. - The structures of most PFASs are still unclear, requiring complex non-targeted analytical methods for detection and monitoring. 2. **Limitations of Existing Technologies**: - Although single-dimensional liquid chromatography with high-resolution mass spectrometry (LC-HRMS) can provide comprehensive detection, its resolution is insufficient when analyzing highly complex PFAS mixtures, such as firefighting foams (AFFF). - The high complexity of the PFAS chemical space can lead to co-elution of homologous series (HSs), affecting the quality of data processing and feature detection, and subsequently impacting the quality of tandem mass spectrometry (MS/MS) experiments. ### Solutions 1. **Integration of LC×LC-HRMS/MS Technology**: - Utilize orthogonal stationary phases in two-dimensional liquid chromatography (LC×LC) to increase chromatographic peak capacity and improve resolution. - Combine high-resolution tandem mass spectrometry (HRMS/MS) to generate high-quality mass spectrometry data, enhancing the detection and annotation accuracy of PFASs. 2. **Data Processing and Analysis Methods**: - Use "Kendrick Mass Defect" (KMD) and "Residual Kendrick Mass" (RKM) analysis methods to improve the detection and grouping capabilities of PFAS homologous series. - Employ the FluoroMatch platform in combination with MS/MS data for PFAS annotation, including the annotation of known standards and unknown compounds. ### Experimental Results - Using LC×LC-HRMS/MS technology, 57 PFAS homologous series (HSs) were successfully identified in 3M and Orchidee AFFF mixtures. - Among them, 12 PFAS homologous series were annotated, including categories such as PFACAs, PFASAs, SF5-PFASAs, N-SPAmP-FASA, and N-CMAmP-FASA. - Some PFAS homologous series were detected for the first time in AFFF samples, such as perfluoroalkyl aldehydes and chlorinated PFASAs. ### Summary By integrating LC×LC-HRMS/MS technology and advanced data processing methods, this paper significantly improves the detection and annotation capabilities of complex PFAS mixtures, providing strong support for environmental monitoring and risk assessment.
