Abstract:Trace amounts of Hg2+ are detected by using a microcantilever coated with gold. The microcantilever undergoes bending due to accumulation of Hg2+ on the gold surface. It is found that a concentration of 10(-11) M Hg2+ can be detected using this technology. Other cations, such as K+, Na+ Pb2+, Zn2+, Ni2+, Cd2+, Cu2+, and Ca2+ have little or no effect on the deflection of the cantilever. The selectivity of the Hg2+ sensor could be improved by coating the gold surface of microcantilever with a self-assembled monolayer of a long-chain thiol compound.

Abstract:Invisible mercury ion is an extremely poisonous environmental pollutant, therefore, a fast and highly sensitive detection method is of significant importance. In this study, a liquid-gated graphene field-effect transistor (GFET) array biosensor (6 × 6 GFETs on the chip) was fabricated and applied for Hg2+ quantitate detection based on single-stranded DNA (ssDNA) aptamer. The biosensor showed outstanding selectivity to Hg2+ in mixed solutions containing various metal ions. Moreover, the sensing capability of the biosensor was demonstrated by real-time responses and showed a fairly low detection limit of 40 pM, a wide detection ranged from 100 pM to 100 nM and rapid response time below one second. These results suggest that the GFET array biosensor based on ssDNA aptamer offers a simple fabrication procedure and quite fast method for mercury ion contaminant detection and are promising for various analytical applications.

Abstract:Contamination of aquatic environment by heavy metals is serious because of the toxicity of such metals on the whole ecological system. This paper describes a double-side nanoband electrode array, which consisted of gold and platinum electrodes on the opposite surface and was used for heavy metals detection. The fabrication quality of the array was validated by scanning electron microscope (SEM) and energy dispersive X-ray (EDX). Cyclic voltammetry was utilized to characterize the electrochemical behavior of the array. Well correlation was found between experimental data and theoretical models. The analytical performance of the array was investigated using anodic stripping voltammetry, and detection limits below mu g.L(-1) level were obtained for lead and copper ions. The Au nanoband electrode array without plating mercury was applied to the detection of heavy metals in sea water, where agreement was found with the standard analytical method (Atomic Absorption Spectrometry, AAS). This paper proposed a mercury-free double-side nanoband electrode array, offing the possibility for in situ, on line heavy metals detection.

Abstract:This paper presents a novel hybrid sensor array for heavy metal determination with self-calibration process by multivariate nonlinear regression (MNLR). Microelectrode array (MEA) and light addressable potentiometric sensor (LAPS) were synchronously fabricated on the same wafer by microfabrication technology. The electrochemical behavior of gold MEAs in different sizes was characterized by cyclic voltammetry. Meanwhile, the detection and quantification of zinc, lead and copper were conducted by differential pulse stripping voltammetry. The sensitivity of zinc, lead and copper are 36.3 nA/ppb, 11.2 nA/ppb and 4.6 nA/ppb, respectively. The electrochemical performance of pH in LAPS was studied with sensitivity of 52.1 mV/pH. The reproducibility and long-term stability were investigated in which relative standard deviation of merely 1.45% was observed and less than 4 mV deviation was presented in a continuous test of 2 h. The acidity of solutions influences the stripping process to great extent due to hydrogen evolution, hydrolysis of heavy metal and hydroxylation of the working electrode. Therefore, a self-calibration process through MNLR was carried out to calibrate the influence of pH and to enhance the detection performance based on the outcomes of heavy metal in MEA and pH in LAPS. The results validate good capability of the hybrid sensor array for trace heavy metal analysis. (C) 2013 Elsevier B.V. All rights reserved.

Abstract:Based on stripping voltammetry, a novel gold microelectrode array (MEA) sensor chip with mercury film plated was used for simultaneously detection of trace heavy metal ions Zn2+, Cd2+, Pb2+ and Cu2+ in water pollution. The electrochemical behavior and the actual surface area of the MEA were investigated by cyclic voltammetry in K3[Fe(CN)6] solution. Electrochemical impedance spectrum (EIS) was utilized to examine the mercury deposition extent on the surface of the MEA. Mercury film plated MEA was then applied to simultaneously detect four heavy metal elements Zn2+, Cd2+, Pb2+ and Cu2+ in sample solutions using differential pulse anodic stripping voltammetry, where good linearities of the standard working curves were obtained for Cd2+, Pb2+ and Cu2+ with the detection limits of 0.1, 0.5 and 0.3 μg/L, separately. But the linearity of the standard working curve was poor for Zn2+, which was mainly caused by the excursion of the testing baseline and the inaccurate addition.

Abstract:A multiple signal amplification strategy was designed for an ultrasensitive competitive immunoassay for Hg2+. This strategy was achieved using graphene conjugated with a large number of CdSe quantum dots to enhance the basal signal and enormous horseradish peroxidase (HRP) labeled with gold nanoparticles (AuNPs) to consume the coreactant H2O2 generated in situ. The immunosensor was constructed by immobilization of coating antigen on poly(diallyldimethylammonium chloride) graphene CdSe composites (PDDA-GN-CdSe), and a strong electrochemiluminescence (ECL) signal was obtained. When the immunosensor was immersed in antibody AuNPs HRP composites, the ECL signal greatly decreased, which was ascribed to the bound enzyme on the electrode surface. The self-produced coreactant H2O2 was consumed by o-phenylenediamine in the presence of enzyme, effectively decreasing the ECL intensity from the quantum dots. The Hg2+ in solution and the corresponding coating antigen competed for the limited antibody, and thus, the ECL intensity was linearly dependent on the logarithm of the mercury(II) concentration from 0.2 to 1000 ng mL(-1) with a detection limit of 0.06 ng mL(-1). The immunoassay exhibited good stability and accuracy and acceptable reproducibility, indicating that it provides a promising approach for the detection of trace mercury and other small molecular compounds in environmental samples.

Abstract:Monitoring the levels of potentially toxic metal ions such as Hg(2+) in aquatic ecosystems is important because this ion can have severe effects on human health and the environment. Thus, a novel chemiluminescence (CL) sensor is developed for the highly selective and ultrasensitive detection of Hg(2+) ions in aqueous solution, based on thymine-Hg(2+)-thymine (T-Hg(2+)-T) coordination chemistry and subsequent formation of gold nanoparticles (Au NPs) in a HAuCl(4)/NH(2)OH reaction. The thus-formed Au NPs trigger the reaction between luminol and AgNO(3), producing CL emission. This novel CL technique has several advantages including high sensitivity (0.01 ppb) and selectivity over a spectrum of interfering metal ions. In view of these advantages, as well as the cost-effective, minimized working steps and portable features of the CL techniques, we expect that this CL sensor will be a promising candidate for the field detection of toxic Hg(2+) ions in environment, water, and food samples.

Abstract:An electrochemical sensor was developed for attomolar Hg(2+) detection. Three single-stranded DNA probes were rationally designed for selective and sensitive detection of the target, which combined T-Hg(2+)-T coordination chemistry and the characteristic of convenient modification of electrochemical signal indicator. Graphene and nanoAu were successively electrodeposited on a glass carbon electrode surface to improve the electrode conductivity and functionalize with the 10-mer thymine-rich DNA probe (P1). NanoAu carriers functionalized with 29-mer guanine-rich DNA probe (P3) labeled methyl blue (MB-nanoAu-P 3s) were used to further strengthen signal response. In the presence of Hg(2+), a T-T mismatched dsDNA would occur between P1 and a 22-mer thymine-rich DNA probe (P2) on the electrode surface due to T-Hg(2+)-T coordination chemistry. Followed by adding the MB-nanoAu-P 3s for hybridization with P2, square wave voltammetry was executed. Under optimal conditions, Hg(2+) could be detected in the range from 1.0 aM to 100 nM with a detection limit of 0.001 aM. Selectivity measurements reveal that the sensor is specific for Hg(2+) even with interference by high concentrations of other metal ions. Three different environmental samples were analyzed by the sensor and the results were compared with that from an atomic fluorescence spectrometry. The developed sensor was demonstrated to achieve excellent detectability. It may be applied to development of ultrasensitive detection strategies.
Ultrasensitive Electrochemical Sensing of Hg 2+ Based on Thymine-Hg 2+ -Thymine Interaction and Signal Amplification of Alkaline Phosphatase Catalyzed Silver Deposition

Abstract:We report an ultrasensitive electrochemical sensor for Hg2+ detection, on the basis of two Hg2+-specific oligonucleotide probes (a thiolated capture probe and a biotinated signal probe), "terminal" signal amplification of alkaline phosphatase catalyzed deposition of silver and in situ microliter-droplet anodic stripping voltammetry (ASV). The thiolated capture probe is immobilized on an Au-plated glassy carbon electrode, and the biotinated signal probe is then attached to the electrode surface through the thymine-Hg2+-thymine interaction in the presence of Hg2+. A streptavidin-alkaline phosphatase (ALP) composite is then immobilized on the electrode surface via biotin-streptavidin interaction. The immobilized ALP can catalyze the hydrolyzation of ascorbic acid 2-phosphate trisodium salt to generate ascorbic acid, which can reduce AgNO3 to form silver deposit on the modified electrode. Quantitative analysis of Hg2+ is conducted through microliter-droplet ASV of silver after its simultaneous chemical dissolution and cathodic preconcentration on the modified electrode. The sensor can linearly respond to the common logarithm of Hg2+ concentration from 0.1nM to 250μM with a limit of detection of 0.01nM (2 ppt, S/N = 3) and a sensitivity as high as 227μAdec-1.

Abstract:Herein, we describe a competitive-type electrochemiluminescence (ECL) strategy for Hg2+ determination based on the peroxydisulfate/oxygen (S2O82-/O-2) system that uses Pt/Pd nanodendrites (Pt/Pd NDs)-thiosemicarbazide/norfloxacin (TN)-covered gold nanoparticles (Pt/Pd-TNG(50)) as a signal enhancer. The Pt/Pd NDs, a dense array of Pt branches on a Pd core, possessed excellent catalytic properties to enhance ECL intensity by accelerating electron transfer. In addition, the binary intramolecular synergy of TN, which had cooperative interactions of powerful - stacking with a larger conjugated surface, could extremely enhance the ECL signal of the S2O82-/O-2 system. Furthermore, we designed a competitive immunoassay method using a structured sensor where a monoclonal antibody (mAb) against Hg2+ exhibited high specificity and recognition of Hg2+, which greatly improved the specificity and sensitivity of the immunosensor. As a result, the proposed immunosensor gave Hg2+ detection with a low detection limit (16 pg mL(-1)) and displayed high sensitivity and stability. Importantly, this work not only, for the first time to our knowledge, utilized Pt/Pd NDs as promising ECL emitters for bioprobe construction but also opened an efficient way for the detection of Hg2+ in environmental monitoring.

Abstract:In this work, an unmodified homogeneous electrochemical sensor based on electrochemical bonding and catalytic hairpin assembly (CHA) was first constructed for the high sensitivity detection of Hg2+. Herein, tetraferrocene, a synthesized compound, was used as a signal marker that modified both ends of the hairpin probe to amplify the electrochemical signal. The interaction of T-Hg2+-T could induce the catalytic self-assembly of hairpins by means of auxiliary DNA. The rigid DNA triangle that was formed easily reaches the electrode and induced Au-S self-assembly assisted by potential, allowing tetraferrocene to reach the electrode surface and generate a sensitive electrochemical signal. CHA and tetraferrocene signal markers accomplished dual signal amplification, and the limit of detection was 0.12 pM. Differential pulse voltammetry experiments in the presence of tetraferrocene redox indicator show that the linear response range of electrochemical biosensors to mercury ions is 0.2-2000 pM, This technology offers good selectivity and high recognition efficiency for the detection of mercury ions and has broad application prospects in actual sample detection.

Abstract:Facile and ultrasensitive detection of Hg2+ in water environment remains challenging. Exonuclease III (Exo-III)-assisted target recycling is one of the most popular amplification strategies. Although the magnesium (II) ions are widely acting as cofactors of Exo-III, we recognized that Mg2+ cofactors would strongly disturb the charge distribution on citrate-stablized gold nanoparticles (in the general sense, unmodified AuNPs) surface, thus generate false positive colorimetric signals. To address this issue, we first put forward the view that the cobalt (II) ions can function as the Exo-III cofactor and successfully construct a novel label-free colorimetric aptasensor for facile and ultrasensitive detection of Hg2+ using Hg2+ -triggered Exo-III-assisted signal amplification and unmodified AuNPs as indicators. A hairpin-looped DNA probe was rationally designed with thymine-rich recognition termini and specifically recognized trace Hg2+ by a stable T-Hg2+-T structure. A blue-to-red color change of AuNPs with the addition of Hg2+ provided the quantitative detection of Hg2+ with a limit of detection of 0.2 nM and a linear working range from 0.5 nM to 5.0 nM. The whole testing time for one assay was approximately 40 min. Real water samples, even containing Hg2+ at 1 nM, could be determined by the aptasensor with recovery rates from 97% to 103%.

Abstract:In this paper, an electrochemical aptamer sensor with high sensitivity and selectivity was proposed for the detection of mercury ion (Hg 2+ ). The orderly assembled gold nanorods-modified screen-printed electrode (AuNRs/SPE) was used to improve the sensor with higher sensitivity and data reproducibility. The sensor was designed based on the specific recognition of Hg 2+ with aptamer to form T-Hg 2+ -T binding structure. Two aptamer chains were applied in the designed sensor, aptamer S1 with terminal thiolate group was self-assembled on the AuNRs/SPE via Au-S bond, and complementary S2 labeled with ferrocene (Fc) would generate a strong redox electrical signal. The addition of Hg 2+ would mediate the folding of the T-base of S1 to form a hairpin structure, resulting in the dissociation of the Fc-labelled S2 from the sensing system and a significant reduction in the redox current. The electrochemical properties of the sensor were further investigated using cyclic voltammetry (CV), differential pulse voltammetry (DPV) and electrochemical impedance spectroscopy (EIS). The effects of aptamer reaction time, concentration, and target incubation time on the sensor response were also investigated. The results showed that the sensor had a sensitive response to Hg 2+ at concentrations ranging from 1 pM to 1 μM, with a detection limit of 0.3 pM. Furthermore, the proposed sensor was applied in the determination of Hg 2+ in real samples, demonstrating its potential practical value.

Abstract:Mercuric ion (Hg2+) in aqueous media is extremely toxic to the environment and organisms. Therefore, the ultra-trace electrochemical determination of Hg2+ in the environment is of critical importance. In this work, a new electrochemical Hg2+ sensing platform based on porous activated carbon (BC/Cu2O) modified with cuprous oxide was developed using a simple impregnation pyrolysis method. Differential pulse anodic stripping voltammetry (DPASV) was used to investigate the sensing capability of the BC/Cu2O electrode towards Hg2+. Due to the excellent conductivity and large specific surface area of BC, and the excellent catalytic activity of Cu2O nanoparticles, the prepared BC/Cu2O electrode exhibited excellent electrochemical activity. The high sensitivity of the proposed system resulted in a low detection limit of 0.3 ng·L-1 and a wide linear response in the ranges from 1.0 ng·L-1 to 1.0 mg·L-1. In addition, this sensor was found to have good accuracy, acceptable precision, and reproducibility. All of these results show that the BC/Cu2O composite is a promising material for Hg2+ electrochemical detection.

Abstract:A triple-channel optical signal probe has been designed for the detection of Hg2+ ions. In the absence of Hg2+, the thymine-rich ssDNA wrapped AuNPs were well dispersed. Thus, the colour of the solution is pink-red; the resonance light scattering (RLS) signal is low; the fluorescence of acridine orange (AO) is powerfully quenched. In the presence of Hg2+, the ssDNA formed a T-Hg2+-T configuration, which produces a colour change, RLS signal enhancement, and fluorescence of AO restoration. Under the optimum conditions, the system exhibits a dynamic response range for Hg2+ from 50 nM to 5 mu M with a detection limit of 30 nM.

Abstract:In this paper, we developed an electrochemical aptasensor with high sensitivity based on the stable T-Hg2+-T structure for Hg2+ determination. The three-dimensional reduced graphene oxide-modified gold nanoparticles (3D-rGO/AuNPs) was served as support material, which offered more immobilization sites for thiolated probe (P1). In the presence of Hg2+, P1 could interact with another probe (P2) by forming T-Hg2+-T structure. Further, the partial sequence of P2 that did not interact with P1 was employed as an initiator sequence, which resulted in the formation of the extended double-stranded DNA through the hybridization chain reaction (HCR) between two harpin DNA strands (HP1 and HP2). Toluidine blue (TB) was used to interact with the double-stranded DNA and bring in effective electrochemical responses. Under the optimal experimental conditions, the aptasensor exhibited pronounced electrochemical signals towards Hg2+ ranging from 10 fM to 50 nM, with a low detection limit of 3.3 fM. Besides, the aptasensor showed excellent performance in water samples for Hg2+ determination.

A Simple Colorimetric Sensor Based on Anti-Aggregation of Gold Nanoparticles for Hg2+ Detection

Abstract:A new method has been proposed to realize the visual detection of Hg2+ via anti-aggregation of gold nanoparticles (Au NPs). The positively charged Au NPs were prepared using poly(diallyldimethylammonium) chloride (PDDA) as reducer and stabilizer. In the presence of cysteine, the color of Au NPs solution turned from ruby red to royal purple, indicating the aggregation of Au NPs. Owing to the high stability constant of cysteine with Hg2+, the pre-incubation of Hg2+ with cysteine would form Hg2+–cysteine complexes and significantly reduce the concentration of free cysteine molecules, thus the aggregation of Au NPs was interrupted since there was not enough inducer. With the increase of Hg2+ concentration, the color of the Au NPs solution would progress from purple to red, allowing the visual detection of Hg2+ ranging from 5.0×10−8 to 1.0×10−5M, with a detection limit of 2.5×10−8M. The proposed method is convenient, low-cost and free of complex equipment, making it possible to analyze Hg2+ in drinking water, rain water or water extracted air samples.

Abstract:Heavy metal ions (Hg( II )) were detected in fresh chicken liver and snail shell extract samples using novel synthesised SDA/MWCNT-modified electrodes. The synthesized N , N ′-bis(salicylaldehyde)-1,2-diaminobenzene (SDA) ligand was characterized via FT-IR, 1 H-NMR, and 13 C-NMR spectroscopy. The hydroxyl and imine functional groups present in SDA act as active sites and bind to the MWCNT surface. The surface morphology of the modified SDA/MWCNT electrode exhibited a star-like crystal structure and the preconcentration of Hg( II )-SDA/MWCNTs lead to a crystal cloud structure, as characterized by SEM with EDX. The enhancement of current and conductance of the SDA/MWCNT- and MWCNT-modified electrode was investigated by cyclic voltammetry (CV) and electrochemical impedance spectroscopy (EIS). The conductance ( σ ) values for the MWCNT- and SDA/MWCNT-modified electrodes are 234.1 × 10 −5 S cm −1 and 358.4 × 10 −5 S cm −1 , respectively, as determined by electrochemical impedance spectroscopy. Consequently, an electrochemical sensor with outstanding performance in terms of reproducibility, stability and anti-interference ability was fabricated. The stripping analysis of Hg( II ) was performed using square wave anodic stripping voltammetry (SWASV) and cyclic voltammetry (CV). Using SWASV, a linear range of Hg( II ) response was found to be 1.3 to 158 μg L −1 , and the limit of detection (LOD) was 0.24 μg L −1 . Finally, the results of the recovered value of Hg( II ) in freshly prepared chicken liver and snail shell extract samples by SWASV were compared with the atomic absorption spectroscopy (AAS) results.

Highly Selective and Sensitive Detection of Hydrogen Sulfide by the Diffraction Peak of Periodic Au Nanoparticle Array with Silver Coating

Abstract:The two-dimensional (2D) periodic Au nanosphere array with silver coating was prepared by using a colloidal monolayer template to obtain a Au nanosphere array and subsequently depositing silver thin coating on it, which could be used as an optical sensor to effectively detect H2S. Such periodic Au nanosphere array with silver coating displayed a surface plasmonic resonance (SPR) peak and an optical diffraction peak. Compared with the SPR peak, the diffraction peak, originated from the periodic arrangements of the obtained array, demonstrated a more sensitive optical change to detect H2S with a significant redshift as the H2S concentration increased. It was attributed to the increase of the refractive index of the environment around the Au nanosphere arrays with silver coating due to the partial formation of Ag2S after detecting H2S. Furthermore, the H2S sensor based on the change of the optical diffraction peak, showed an excellent selectivity and it was very sensitive to detect H2S from 2 to 30 μM. This method was investigated by the analysis in H2S-spiked blood samples, which indicates that the method has the potential to detect H2S in blood samples. Abstract:Mercury is a toxic environmental contaminant that can cause serious health problems. This work describes a new type of eco-friendly three-electrode plastic sensor chip for the determination of trace Hg(II) by means of anodic stripping voltammetry (ASV). The sensor chip is entirely fabricated by injection moulding, which is a sustainable manufacturing method, and consists of three conductive carbon-based electrodes embedded in a plastic holder while the reference electrode is coated with Ag using e-beam evaporation. The sample is spiked with Au(III) which deposits on the working electrode in the form of gold nanoparicles during the analysis; the target Hg(II) co-deposits on the gold nanoparticles forming a Au(Hg) amalgam in situ. The accumulated Hg is stripped off the electrode and quantified by an anodic square wave potential scan. The relevant conditions and the potential interferences are investigated. The limit of detection for Hg(II) is 0.4 μg L-1 and the repeatability at the 20 μg L-1 Hg(III) level (n = 10) is 5.3%. The sensor is applied to water, honey, fish oil and mussel samples with recoveries between 98 and 107%.
