Abstract:To the best of our knowledge, there was no prior report providing valuable preliminary data through a demonstration of the quantitative phytochemical and antioxidant activity of Gymnosporia senegalensis. The total contents of phenols, flavonoid, flavanol, tannin, and saponin were evaluated from different fractions extracted from the leaf, stem, and bark of G. senegalensis by using standards such as gallic acid, quercetin, rutin, tannic acid, and saponin quillaja. The antioxidant potential was measured by 2,2-diphenyl-1-picrylhydrazyl (DPPH), hydrogen peroxide scavenging (H2O2), superoxide anion radical scavenging, metal chelating ferrous ion, ferric reducing antioxidant power (FRAP), and total antioxidant capacity (TAC). Data were subjected to half-inhibitory concentration (IC50) and one-way analysis of variance (ANOVA) at p < 0.05 as a significant value. The total phenol content was found to be highest in the chloroform extract of stem at 97.7 ± 0.02 mg GAE/g. The total flavonoid and flavonol contents in the aqueous extract were 97.1 ± 0.03 mg QE/g and 96.7 ± 0.07 mg RE/g, respectively. The total tannin content in the ethyl acetate extract of leaf was 97.5 ± 0.01 mg TAE/g, and the total saponin content in the methanol extract of stem was 79.1 ± 0.06 mg SQE/g. The antioxidant analysis indicated that IC50 and percentage (%) inhibition were dose-dependent and showed the highest antioxidant activity (40.9 ± 0.9 μg/mL) in methanol extract of leaf for DPPH, (88.8 ± 1.12 μg/mL) in the chloroform extract of stem for H2O2, (43.9 ± 0.15 μg/mL) in the aqueous extract of bark for superoxide anion radical scavenging activity, (26.9 ± 0.11 μg/mL) in the chloroform extract of leaf for the metal chelating ferrous ion activity, (7.55 ± 0.10 mg/mL) in the benzene extract of leaf for FRAP, and (2.97 ± 0.01 mg/mL) in the methanol extract of bark for TAC. These results show that G. senegalensis has great potential in antioxidant activities. The isolation and characterization of specific bioactive compounds and the in vivo applicability of such activity await further extensive studies for drug discovery and development.

Abstract:Background: Irvingia gabonensis extracts are documented to possess antidiabetic, hypocholesterolemic, and antioxidant activities. The phytochemical components of medicinal plants are responsible for their therapeutic properties, particularly those with antioxidant properties. Objectives: The quantitative phytochemicals and in vitro antioxidant properties of the aqueous and ethanol extracts of I. gabonensis stem bark were assessed in this study. Materials and Methods: Standard techniques were used to measure the concentrations of phenols, flavonoids, tannins, alkaloids, steroids, and saponins in water and alcohol extracts of this plant. Total Antioxidant Power (TAP) and the extract's capacity to scavenge free radicals including superoxide, nitric oxide, 2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2-azino-bis-(3-ethylbenzoth iazoline-6-sulfonic acid) (ABTS+), β-carotene, and hydrogen peroxide were also evaluated. The extracts' Ferric-Reducing Potential (FRAP) was also assessed. Results: The alcohol extract had significantly (P < 0.05) higher concentrations of all the phytochemicals measured as well as near significantly (P <0.066) higher TAP in comparison to the aqueous extracts. In general, the in vitro antioxidant capacity of the ethanol extract surpassed that of the water extract. The ethanol extract had IC 50 values that were comparable to or less than the reference standards for its ability to neutralize nitric oxide, H 2 O 2 , superoxide, and DPPH radicals. Conclusion: The outcomes of the quantitative phytochemical analysis and in vitro antioxidant effects of aqueous and ethanol extracts of I. gabonensis stem bark clearly show their potential as an excellent reservoir of bioactive compounds and scavengers of deleterious oxidants; properties that could be explored therapeutically.

Abstract:Background Ziziphus mauritiana Lam. is a plant used in traditional medicine in Burkina Faso in the treatment of several diseases, of which diabetes is characterized by oxidative stress. The aim of this study was to evaluate the in vitro antioxidant potential of the extracts of leaves of this plant. Methods The crude hydroethanolic extract (HEE) of the leaves of Z. mauritiana and their partitionates in n-hexane, dichloromethane, and ethyl acetate, and in the residual aqueous solution (the F1, F2, F3, and F4 fractions, respectively) were first prepared. The content of polyphenols was determined and the antioxidant effects of the extracts were evaluated by their 1,1-diphenyl-2-picrylhydrazyl (DPPH) scavenging activity, inhibition of lipid peroxidation (TBARS), and the ferric reducing antioxidant power (FRAP). Results The HEE as well as the F3 and F4 fractions were rich in polyphenols with contents between 58 and 84 mg equivalent gallic acid per 100 mg. The flavonoid content was 4 mg quercetin equivalents in the HEE and the F4 fraction. Except for the F1 fraction, the HEE and the other fractions showed significant DPPH scavenging activity (IC50 between 8 and 12 μg/mL). The IC50 of TBARS by different extracts was in the range 1-5 μg/mL, and the FRAP activity was 7-85 mg ascorbic acid equivalent per 100 mg. Total polyphenol content was highly correlated with the antioxidant activities. Conclusions The HEE, F3, and F4 fractions were found to be the richest in polyphenols and had the best antioxidant activity. The antioxidant activity of the extracts of the leaves of Z. mauritiana is due to these polyphenolic compounds.

Abstract:The purpose of this study is to determine the phytochemical content and biological activities of the Astragalus gombo (endemic specie). While identification and quantification of individual secondary metabolites were performed by analysis HPLC. Antioxidant (DPPH and FRAP), anti-inflammatory, anti-hemolytic and antibacterial activities were evaluated. For analysis HPLC, we obtained 65 peaks and identified 6 major bioactive compounds. The total concentration in polyphenols, flavonoids and condensed tannins varied respectively from 66.306±0.88 mg GA eq/g, 34.312±1.4 mg Q eq/g and 5.343±2.74 mg Ca eq/g. In terms of antioxidant activity, we found that this extract had high inhibitory ratios equivalent to IC50=62.813±0.006 µg/mL for DPPH and IC50=19.375±0.041 µg/mL for FRAP. A high anti-inflammatory activity was estimated at 1615.81µg/mL, and a weak anti-hemolytic activity, the other hand the antibacterial activity is somewhat moderate against the five strains studied. This study demonstrated that the aqueous extract of A. gombo from El Oued region has tremendous antioxidant, anti-inflammatory and antibacterial abilities.
African Under-Utilized Medicinal Leafy Vegetables Studied by Microtiter Plate Assays and High-Performance Thin-Layer Chromatography–Planar Assays

Abstract:Biological activities of six under-utilized medicinal leafy vegetable plants indigenous to Africa, i.e., Basella alba, Crassocephalum rubens, Gnetum africanum, Launaea taraxacifolia, Solanecio biafrae, and Solanum macrocarpon, were investigated via two independent techniques. The total phenolic content (TPC) was determined, and six microtiter plate assays were applied after extraction and fractionation. Three were antioxidant in vitro assays, i.e., ferric reducing antioxidant power (FRAP), cupric reduction antioxidant capacity (CUPRAC), and 2,2-diphenyl-1-picrylhydrazyl (DPPH) scavenging, and the others were enzyme (acetylcholinesterase, butyrylcholinesterase, and tyrosinase) inhibition assays. The highest TPC and antioxidant activity from all the methods were obtained from polar and medium polar fractions of C. rubens, S. biafrae, and S. macrocarpon. The highest acetyl- and butyrylcholinesterase inhibition was exhibited by polar fractions of S. biafrae, C. rubens, and L. taraxacifolia, the latter comparable to galantamine. The highest tyrosinase inhibition was observed in the n-butanol fraction of C. rubens and ethyl acetate fraction of S. biafrae. In vitro assay results of the different extracts and fractions were mostly in agreement with the bioactivity profiling via high-performance thin-layer chromatography–multi-imaging–effect-directed analysis, exploiting nine different planar assays. Several separated compounds of the plant extracts showed antioxidant, α-glucosidase, α-amylase, acetyl- and butyrylcholinesterase-inhibiting, Gram-positive/-negative antimicrobial, cytotoxic, and genotoxic activities. A prominent apolar bioactive compound zone was tentatively assigned to fatty acids, in particular linolenic acid, via electrospray ionization high-resolution mass spectrometry. The detected antioxidant, antimicrobial, antidiabetic, anticholinesterase, cytotoxic, and genotoxic potentials of these vegetable plants, in particular C. rubens, S. biafrae, and S. macrocarpon, may validate some of their ethnomedicinal uses.

Abstract:Aframomum melegueta, commonly known as grains of paradise, is a medicinal plant celebrated for its rich phytochemical content and therapeutic properties. This study evaluated the antioxidant and cytotoxic potentials of its ethanolic and methanolic extracts, both in vitro and in vivo, while also analyzing their chemical profiles. HPLC analysis identified key compounds, including gallic acid, caffeic acid, caffeine, coumarin, rutin, catechin, ferulic acid, and quercetin. Chronic toxicity assessments confirmed the safety of the extracts, with no adverse effects on animal health, particularly in liver histopathology. Cytotoxicity results indicated reduced splenocyte viability at the highest concentrations. The extracts exhibited significant antioxidant activity in DPPH•, FRP, and phosphomolybdate assays, demonstrating their effectiveness as antiradical agents. In vivo antioxidant results showed a reduction in lipid peroxidation levels in serum and liver, highlighting the extracts' ability to mitigate oxidative stress. Additionally, the extracts provided protection against H2O2‐induced erythrocyte hemolysis and modulated NO production in peritoneal macrophages. These findings underscore the therapeutic potential of A. melegueta extracts, suggesting their promise in developing preventive strategies for oxidative stress‐related chronic diseases.

Abstract:The use of medicinal plants as a core component of the traditional African health care system is perhaps the oldest and most diverse of all therapeutic approaches. Traditional herbal healers are, in many parts of rural Africa, the most readily accessible and affordable health resource available to the local population and, at times, the only surviving treatment. Finding a suitable solvent is important to obtain a high yield of antioxidants in the phytochemicals present in C. africana. Among the solvents, methanol displayed the highest capacity in extracting glycosides (223.12Â±1.20 mg LE/g dry weight) phenolics (89.09Â±0.24 (mg GAE/g dry weight) and flavonoids (80.88Â±0.24 mg QE/g dry weight) from C. africana stem bark extract. Closely related trend was observed in glycosides, phenolics and flavonoids extracted with n-hexane. These findings suggest a high number of polar glycosides and phenolics in the stem extract of C. africana. These findings validate the pharmacological activities of the plant in Africa.
Global Chemical Composition and Antioxidant and Anti-Tuberculosis Activities of Various Extracts of Globularia alypum L. (Globulariaceae) Leaves

Abstract:In this work, an evaluation of the biological activities of Globularia alypum L. extracts and their global chemical composition was realized. Extracts from G. alypum were obtained by two extraction methods. The composition of polyphenols (8.5-139.95 g gallic acid equivalent/Kg of dry mass), tannins (1.39-18.65 g catechin equivalent/Kg of dry mass), anthocyanins (8.17-70.69 mg cyanidin equivalent/Kg of dry mass) and flavonoids (0.31-19.28 g quercetin equivalent/Kg of dry mass) was evaluated. The samples were subjected to a screening for their antioxidant activities using the DPPH* and ABTS*+ assays. For the first time, the anti-tuberculosis activity (H(37)Rv) for G. alypum was tested against Mycobacterium tuberculosis. The strongest antioxidant activity was obtained for the methanol extract (IC(50 ) = 15.58 ± 0.168 mg/L) and the best anti-tuberculosis activity was obtained for the petroleum ether extract (IC(50 )= 77 mg/L). We have found a positive correlation between the total phenolics content and the antioxidant activity R2 = 0.88 (DPPH*) and R2 = 0.97 (ABTS*+). We have found also a positive correlation between the flavonoid content and the antioxidant activity R2 = 0.91 (DPPH*) and R2 = 0.91 (ABTS*+).

Abstract:Anacardium occidentale and Vitellaria paradoxa are two medicinal plants traditionally used by Ivorian, particularly in the treatment of diabetes, arterial hypertension and diarrhea. The objective of this work was to evaluate the phytochemical constituents and the antioxidant activity of the stem barks of A. occidentale L. and V. paradoxa. The stem bark of both plants was used for the aqueous extraction. Major chemical groups were identified using the tube staining method followed by confirmation by thin layer chromatography (TLC). The antioxidant activity of the extracts was evaluated by DPPH method. The results of phytochemical screening of plant extracts revealed the presence of polyphenols, anthraquinones, tannins, sterols, terpenes and saponins. In addition, the evaluation of the antioxidant activity by spectrophotometry using the DPPH method showed that all extracts had significant and different antioxydant activity compared to the reference molecule (Vitamin C, IC50 = 0.80 ± 0.70 μg/mL) with respective IC50 of 0.42 ± 0.32 μg/mL for V. paradoxa and 0.87 ± 0.77 μg/mL for A. occidentale. V. paradoxa therefore has the best antioxydant activity. These results could explain the therapeutic virtues of A. occidentale and V. paradoxa in the treatment of various pathologies.

Abstract:Gynura nepalensis D.C. (family: Asteraceae) has abundant uses in the alternative medicinal practice, and this species is commonly used in the treatment of diabetes, rheumatism, cuts or wounds, asthma, kidney stones, cough, urinary tract bleeding, gall bladder stones, hepatitis, diarrhea, hemorrhoids, constipation, vomiting, fertility problems, blood poisoning, septicemia, skin allergy, indigestion, high cholesterol levels, and so on. This study aims to investigate the hepatoprotective and antioxidant potential of the methanol extract of the Gynura nepalensis D.C. (GNME) along with chemical profiling with phytochemical screening. Moreover, prospective phytocompounds have been screened virtually to present the binding affinity of the bioactive components to the hepatic and oxidative receptors. In the hepatoprotective study, alanine transaminase (ALT), aspartate aminotransferase (AST), alkaline phosphatase (ALP), total protein (TP), and lipid peroxidation (LP) and total bilirubin (TB) have been assessed, and in the antioxidant study, the DPPH free radical scavenging, total antioxidant flavonoid, and phenolic contents were determined. Moreover, the molecular binding affinity of the bioactive component of the plant has been analyzed using PyRx AutoDock Vina, Chimera, and Discovery Studio software. The plant extract showed dose-dependent hepatoprotective potential (p < 0.05, 0.01, 0.001) as well as strong antioxidant properties. Moreover, hepatoprotective and antioxidant molecular docking studies revealed a result varying from −2.90 kcal/mol to −10.1 kcal/mol. 4,5-dicaffeoylquinic acid and chlorogenic acid revealed the highest binding affinity among the selected molecules. However, the plant showed portent antioxidant and hepatoprotective properties in the in vitro, in vivo, and in silico models, and it is presumed that the hepatoprotective properties of the plant extract have occurred due to the presence of the vast bioactive chemical compounds as well as their antioxidant properties. Therefore, advanced studies are recommended to elucidate the pharmacological properties of the plant extracts.

Abstract:Magnetic circular dichroism~MCD! has been measured for ferromagnetic CoS 2 in the S 2p, Co 2p and 3p core absorption regions. MCD in the S 2 p region is discussed in terms of the exchange splitting of the unoccupied band. The Co 2 p MCD has revealed that the orbital angular momentum of the Co 3 d state remains finite in spite of its low-spin configuration. This situation is confirmed by a simple cluster model calculation.

Abstract:ETHNOPHARMACOLOGICAL RELEVANCE: Glinus oppositifolius (L.) Aug. DC. belongs to the family Molluginaceae, an annual prostrate herb traditionally used to treat inflammations, arthritis, malarial, wounds, fevers, diarrhoea, cancer, stomach discomfort, jaundice, and intestinal parasites. However, the anti-arthritic activity of the aerial part has still not been reported.AIM OF THE STUDY: To investigate the antioxidant and anti-arthritic activity of G. oppositifolius in Complete Freund's Adjuvant (CFA) induced rats.MATERIALS AND METHODS: The dried aerial parts of this plant material were defatted with n-hexane and extracted by methanol using a soxhlet apparatus. The in vitro anti-arthritic activity of methanolic extract of G. oppositifolius (MEGO) was evaluated in protein denaturation, membrane stabilization, and inhibition of proteinase assay at 25, 50, 100, 200, and 400 μg/ml concentrations. Female Wistar rats were immunized sub-dermally into the right hind paw with 0.1 ml of CFA. Rats were administered with MEGO at doses of 200 and 400 mg/kg once daily for fourteen days after arthritis induction. Assessment of arthritis was performed by measuring paw diameter, arthritic index, arthritic score, body weight, organ weight, and hematological and biochemical parameters, followed by the analysis of pro-inflammatory cytokines such as tumor necrosis factor-alpha (TNF-α), interleukin 6 (IL-6), interleukin-1-beta (IL-1β), cyclooxygenase-2 (COX-2), interleukin 13 (IL-13) and interleukin 10 (IL-10) and histopathological study. In vivo antioxidant effect was investigated in enzymatic assays. The presence of phytoconstituents was analyzed by Gas Chromatography-Mass Spectrometry (GC-MS) and Liquid Chromatography-Mass Spectrometry (LC-MS), respectively. In silico molecular docking study of the compounds was carried out against COX-2, IL-1β, IL-6, and TNF-α using AutoDock 4.2 and BIOVIA-Discovery Studio Visualizer software.RESULTS: MEGO's in vitro anti-arthritic activity showed dose-dependent inhibition of protein denaturation, membrane stabilization, and proteinase inhibition, followed by significant in vivo anti-arthritic activity. The rats treated with MEGO showed tremendous potential in managing arthritis-like symptoms by restoring hematological, biochemical, and histological changes in CFA-induced rats. MEGO (200 and 400 mg/kg) showed a significant alleviation in the levels of hyper expressed inflammatory mediators (TNF-α, IL-1β, and IL-6) and oxidative stress (SOD, CAT, GSH, and LPO) in CFA-induced rats. Spergulagenin-A as identified by LC-MS analysis, exhibited the highest binding affinity against COX-2 (-8.6), IL-1β (7.2 kcal/mol), IL-6 (-7.4 kcal/mol), and TNF-α (-6.5 kcal/mol).CONCLUSIONS: Provided with the comprehensive investigation, methanolic extract of G. oppositifolius against arthritic-like condition is a proof of concept that revalidates its ethnic claim. The presence of Spergulagenin-A might be responsible for the anti-arthritic activity.

Bioassay-Guided Isolation of Guggulsterol I and Evaluation of Its Antioxidant Properties in Ethanol Extracts From Commiphora caudata Leaves

Abstract:The secondary metabolites of various parts of Commiphora caudata have shown a range of biological activities under both in vivo and in vitro conditions, particularly anti-inflammatory and antioxidant properties. While E-guggulsterone from this plant has been proven to have anti-inflammatory effects, the antioxidant potential of phytochemicals present in the leaves of C. caudata is less explored. This investigation aimed to isolate an antioxidant phytoconstituent from the ethanolic extract of the dried leaves of C. caudata using a bioassay-guided approach. The dried leaves were successively extracted with organic solvents, including ethanol. The presence of phytochemicals was tested using high-performance thin-layer chromatography (HPTLC), and phytoconstituent from ethanol extract was purified by column chromatography. The antioxidant activity of both the crude extract and the purified compound was evaluated and then compared. The radical scavenging activity was measured using the 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay. The ethanolic extract of C. caudata showed 87.6% DPPH radical scavenging activity at a concentration of 250 µg/mL, while standard ascorbic acid showed 94.9% inhibition at the same concentration. The concentrated ethanolic extract of C. caudata was subjected to silica-based column chromatography with an ascending polarity of mobile phase solvents, ethyl acetate, and ethanol (25:75), yielding a single compound. The isolated compound was confirmed as guggulsterol I by ultraviolet (UV), infrared (IR), mass, and nuclear magnetic resonance (NMR) spectroscopy. The radical scavenging activity of the crude ethanolic extract of C. caudata leaves and the isolated compound guggulsterol I was concentration-dependent. The crude ethanolic extract of C. caudata showed significant antioxidant activity in comparison with the standard. However, the isolated guggulsterol I showed less antioxidant activity than the crude ethanolic extract. This study strongly suggests that the crude ethanolic extract of C. caudata leaves had better antioxidant activity due to the synergistic or additive effect of guggulsterol I and other phytoconstituents.

Abstract:Portulacaria afra, is indigenous to South Africa and has been identified to have several medicinal properties according to traditional knowledge and few studies. The drive around this research is to evaluate the medicinal properties of the leaves, stems and for the first time the roots extracts of Portulacaria afra, using four solvents with different polarities. The aqueous (60°C), methanol, n-hexane and ethyl acetate whole plant extracts of P. afra were investigated for their phytochemical properties, antimicrobial and antioxidant activity. The phytochemical screening revealed that the methanolic and aqueous extracts of the leaves displayed high presence of secondary metabolites compared to n-hexane and ethyl acetate extracts. The methanolic leaves extracts showed strong presence of quinones, phenols, steroid and coumarins while the aqueous leaves extracts contained a moderate presence of saponins, terpenoids, quinones and coumarins. Ethyl acetate leaves extracts revealed a strong presence of tannins, moderate presence of phytosteroids and a low presence of volatile oil. Meanwhile, the leaves extracts with n-hexane showed a considerable amount of saponins with a moderate presence, and a low presence of tannins, volatile oils and terpenoids. The methanolic stems extracts displayed the most significant presence of secondary metabolites, showing a high presence of terpenoids, steroids, phenols and coumarins. The aqueous stems extracts showed a strong presence of glycosides with a moderate presence of saponins. However, ethyl acetate and n-hexane stems extracts displayed a few secondary metabolites with their concentration ranging from medium to low. The ethyl acetate roots extracts displayed a significant elevated amount of quinones with a strong presence. n-hexane roots extracts showed a moderate presence of volatile oil and a low presence of tannins and steroids. Methanolic roots extracts showed a moderate presence of coumarins and glycosides while aqueous roots extracts showed a low presence of glycosides. The overall highest total phenolics contents (TPCs) and total flavonoids contents (TFCs) in all the plant parts, were found to be in the methanol stems extracts and aqueous roots extracts respectively. Next to the methanol leaves and aqueous leaves extracts respectively. However, in the root’s extracts, the aqueous extracts showed the highest total phenolics content while the water extracts had the highest total flavonoids contents. The antimicrobial activities of P. afra whole plant extracts with the various four solvents were tested against three microorganisms Escherichia coli, Staphylococcus aureus and Streptomyces griseus using agar-well diffusion method. The Antimicrobial activity of the n-hexane extracts of the leaves, stems and roots of P. afra presented a wide range of inhibition against all the test microorganisms, ethyl acetate leaves extract showed a considerable effect against Staphylococcus aureus while the methanolic extracts were not active. Aqueous roots extracts demonstrated a strong antimicrobial activity against Staphylococcus aureus while the other extracts were not active. The zones of inhibition ranged from 13 to 24 mm for the plant extracts. The antioxidant activity potential of the aqueous, methanol, n-hexane and ethyl acetate extracts of P. afra leaves, stems and roots extracts were observed through a 2, 2 diphenylpicryhydrazyl (DPPH) free radical assay, hydrogen peroxide scavenging (H2O2) and metal chelating activity assay. Ethyl acetate roots extracts exhibited the strongest hydrogen peroxide scavenging activity compared to the other extracts. Meanwhile, aqueous stems extracts showed the highest antioxidant activity against DPPH radical. Aqueous and n-hexane roots extracts displayed the strongest metal chelating ability. These findings reveal the efficacy of the use of several solvents with different polarities for effective and more accurate extraction of various compounds and indicate that the antimicrobial and antioxidant activity of P. afra parts are dependent on the solvent extracts.

Abstract:Adults of Haemaphysalis intermedia Warburton and Nuttall (= H. parva Neumann, 1908, preoccupied) are redescribed and illustrated, and immature stages of this species are de- scribed and illustrated. For this study, type material of H. parva Neumann, 1908, from Ceylon has been studied (and illustrated) and compared with a large number of specimens collected in nature in southern India and reared through F1 generations in the laboratory. The confusing and complex history of this taxon is reviewed in detail. H. intermedia is a member of the H. bispinosa group of ticks and exceptionally distinct characters for recog- nizing this previously inadequately delineated species are demonstrated. Haemaphysalis intermedia Warburton and Nuttall, 1909, has long been a confusing taxon. Through the kindness and helpfulness of a number of colleagues, older collections and type series of species associated in one way or another with this name have been made available for comparison with a large number of samples collected in nature and reared in the laboratory by personnel of the Virus Research Centre, Poona, collateral to epidemiological investigations on Kyasanur Forest disease. Abstract:BackgroundMany plant polysaccharides have shown high antioxidant and immunostimulating properties and can be explored as novel molecules with biological properties that can potentially improve immune function. The objective of this work was to characterize soluble and cell wall polysaccharides isolated from the stem bark of Allanblackia floribunda and Chromolaena odorata leaves and to evaluate their antioxidant and immunomodulatory properties.MethodsThree polysaccharide fractions: soluble polysaccharides (PoS), pectins (Pec) and hemicelluloses (Hem) were extracted from A. floribunda stem bark and C. odorata leaves. These samples were analysed for their proteins, phenolic compounds and total sugar contents. The monosaccharide composition was determined by gas chromatography and arabinogalactan proteins content in PoS was evaluated by rocket electrophoresis. The in vitro antioxidant activities were evaluated by 1, 1-diphenyl-2-picryl hydrazyl (DPPH) and 2,2′-azino-bis-3-éthylbenzylthiazoline-6-sulphonic acid (ABTS) radical scavenging assays and ferrous ions chelating activity. Immunomodulatory activities were performed on the peripheral blood mononuclear cells (PBMCs) using proliferation and enzyme linked immunospot (ELISPOT) method to determine the production of an interferon-gamma.ResultsThe characterization of the various fractions showed varied metabolites in each plant. In PoS fractions, Ara and Gal were the major monosaccharides found, indicating that arabinogalactans are the primary macromolecules. Hem fractions contained predominantly Xyl and GalA for A. floribunda and Xyl (upto 80 %) for and C. odorata. A. floribunda Hem fraction and C. odorata PoS fraction showed significant DPPH and ABTS radical scavenging activities and immunostimulatory activity via stimulation of PBMC and production of IFN-γ in a dose-dependent manner.ConclusionThe results obtained from this study support the ethnomedicinal use of the stem bark of A. floribunda and leaves of C. odorata. Further research is necessary to have supporting evidence that the antioxidative and immunomodulative activities of these fractions are really connected to the polysaccharides and not polyphenols.

Abstract:Most of biochemical reactions in the body generates Reactive Oxygen Species (ROS), which are involved in the pathogenesis of oxidative stress-related disorders like diabetes, nephrotoxicity, cancer, cardiovascular disorders, inflammation and neurological disorders when they attack biochemical molecules like proteins, lipids and nucleic acid. Antioxidants are used to protect the cells or tissues against potential attack by ROS. Most medicinal plants possess a rich source of antioxidants such as flavonoids, phenols, tannins, alkaloids among others. These phytochemicals are currently pursued as an alternative and complimentary drug. In this study, phytochemical components, antioxidant and acute toxicity study of the methanol extract of stem bark and root of F. sycomorus were carried out using standard methods. Findings from this study revealed the presence of some diagnostic microscopical features such as calcium oxalate, starch, gum/mucilage, lignin, Aleurone grain, suberized/Cuticular cell wall and inulin but calcium carbonate was absent in stem bark but present in the powdered root. Quantitative physical constants include moisture contents (6.40% and 7.82%), ash value (7.20% and 9.30 %) in stem bark and root respectively. Carbohydrates, alkaloid, flavonoids, saponins, tannins, glycoside, steroid, triterpenes and phenols were present in all the extracts. They were found to exhibit potent 1,1,-diphenyl 2-picryl hydrazyl (DPPH) free scavenging activity. The DPPH radical scavenging ability of the extracts showed the following trend Ascorbic acid < stem bark extract˃ root extract. Abstract:Natural remedies are being widely utilized in some disease's conditions including cancer, diabetes, neurodegenerative diseases, atherosclerosis hypertension and other cardiovascular diseases. The therapeutic intervention of medicinal plants and spices cannot be overemphasized in the management and control of diseases but the exact modes of action of these herbs and plants have not been fully elucidated. This research work was designed to study the expression of selected elements, zinc (Zn), iron (Fe), copper (Cu), selenium (Se), lead (Pb) and cadmium (Cd); the phytochemical and the in vitro antioxidant properties of ethanolic extracts of Moringa oleifera leaves, Allium sativum (Garlic) and Momordica charantia (ejinrin) leaves for their potential involvement in the prevention and management of cardiovascular diseases and cancer through different chemical methods. In vitro antioxidant properties were assayed by investigating 2,2-diphenyl-2-picryl-hydrazyl (DPPH) free radicals scavenging potentials and Ferric Reducing Antioxidant Potential (FRAP); total phenolic content was determined by using Folin-Ciocalteu assay and the elemental contents of these extracts was investigated through the use of Inductively Coupled Plasma Optical Emission Spectroscopy (ICP-OES). All tests were run in triplicates and analysed. Our results revealed that the ethanolic extracts of M. oleifera leaves, A. sativum (Garlic) and M. charantia (ejinrin) leaves possess significant antioxidant activities and express important and beneficial elements (in marginal and trace amounts) and phytochemicals. These results suggest that the medicinal attributes of these plants may be linked to the radical scavenging abilities, beneficial elements and phytochemicals expression of their ethanolic extracts which may likely be a good direction in the area of drug discovery and development.
