Abstract:Natural remedies are being widely utilized in some disease's conditions including cancer, diabetes, neurodegenerative diseases, atherosclerosis hypertension and other cardiovascular diseases. The therapeutic intervention of medicinal plants and spices cannot be overemphasized in the management and control of diseases but the exact modes of action of these herbs and plants have not been fully elucidated. This research work was designed to study the expression of selected elements, zinc (Zn), iron (Fe), copper (Cu), selenium (Se), lead (Pb) and cadmium (Cd); the phytochemical and the in vitro antioxidant properties of ethanolic extracts of Moringa oleifera leaves, Allium sativum (Garlic) and Momordica charantia (ejinrin) leaves for their potential involvement in the prevention and management of cardiovascular diseases and cancer through different chemical methods. In vitro antioxidant properties were assayed by investigating 2,2-diphenyl-2-picryl-hydrazyl (DPPH) free radicals scavenging potentials and Ferric Reducing Antioxidant Potential (FRAP); total phenolic content was determined by using Folin-Ciocalteu assay and the elemental contents of these extracts was investigated through the use of Inductively Coupled Plasma Optical Emission Spectroscopy (ICP-OES). All tests were run in triplicates and analysed. Our results revealed that the ethanolic extracts of M. oleifera leaves, A. sativum (Garlic) and M. charantia (ejinrin) leaves possess significant antioxidant activities and express important and beneficial elements (in marginal and trace amounts) and phytochemicals. These results suggest that the medicinal attributes of these plants may be linked to the radical scavenging abilities, beneficial elements and phytochemicals expression of their ethanolic extracts which may likely be a good direction in the area of drug discovery and development.

Abstract:Occult hydrocephalus has received increasing attention during the last few years due to the reversibility of the serious, associated, symptoms, which include dementia and apraxia of gait. A vast number of these patients may be rescued by the insertion of an atrioventricular shunt or similar procedure. Abstract:Plants are universally recognized as a vital part of the world’s natural heritage and up to 80% of the population rely on plants for their primary healthcare. Varieties of medicinal plants are recognized as a source of natural antioxidants that can protect from oxidative stress, thus playing an important role in chemoprevention of diseases. In the present investigation, 22 extracts from different parts of eight medicinal plants (Punica granatum Linn. (Punicaceae), Caesalpinia bonducella Flem. (Fabaceae), Hibiscus subdariffa Linn. [(Malvaceae), Moringa oleifera Lam. (Moringaceae), Garcinia indica Linn. (Clusiaceae), Emblica officinalis Gaertn. (Euphorbiaceae), Momordica charantia Linn. (Cucurbitaceae), and Matricaria chamomilla Linn. (Asteraceae)] were screened for their protection against oxidative stress in erythrocytes induced by hydrogen peroxide (2 mM) and tert-butyl hydroperoxide (0.01 mM). The effect was also compared with known antioxidants and flavonoids. Subjecting erythrocytes to oxidative stress by incubation with hydrogen peroxide and tert-butyl hydroperoxide caused a significant alteration in reduced glutathione (GSH) and malondialdehyde (MDA) concentration compared to the control. Our results show that medicinal plant extracts protect erythrocytes from hydrogen peroxide and tert-butyl hydroperoxide induced oxidative stress; known antioxidant (vitamin C, E, and β-carotene) and flavonoid (quercetin) also showed a similar protective effect. Our observations may, in part, suggest the use of the spent/waste parts of medicinal plants. This could be an economically viable source of natural and potent antioxidants effective against complications arising from oxidative stress. The results may also improve the ethanopharmacological knowledge of medicinal plants.

Abstract:Magnetic circular dichroism~MCD! has been measured for ferromagnetic CoS 2 in the S 2p, Co 2p and 3p core absorption regions. MCD in the S 2 p region is discussed in terms of the exchange splitting of the unoccupied band. The Co 2 p MCD has revealed that the orbital angular momentum of the Co 3 d state remains finite in spite of its low-spin configuration. This situation is confirmed by a simple cluster model calculation.

Abstract:The quest for novel antioxidant and anti-inflammatory medications from medicinal plants is crucial since the plants contain bioactive compounds with a better efficacy and safety profile than orthodox therapy. This study harnesses the capabilities of mechatronics-driven Agilent Gas Chromatography, deploying in vitro, in vivo, and in silico models to unravel the antioxidant and anti-inflammatory attributes within Combretum paniculatum ethanol extract (CPEE). Employing gas chromatography-mass spectroscopy (GC-MS), our analysis efficiently segregates and evaluates volatile compound mixtures, a technique renowned for identifying organic compounds, as exemplified by its success in detecting fatty acids in food and resin acids in water. Using gas chromatography-mass spectrometry (GC-MS) and GC-FID analyses, this paper ascertains the comprehensive phytochemical composition of CPEE. Also, Molecular interactions of identified compounds with cyclooxygenase (COX-2) implicated in inflammatory urpsurge is verified. GC-MS and GC-FID analyses unveil 41 phytoconstituents within CPEE. Based on the in vitro research, CPEE demonstrated potential in inhibiting thiobarbituric acid-reactive substances, nitric oxide, and phospholipase lipase A2 with inhibition rates of 2.284, 6.547, and 66.8 μg/mL respectively. In vivo experiments confirm CPEE's efficacy in inhibiting granuloma tissue formation, lipid peroxidation, and neutrophil counts compared to untreated rats. Moreover, CPEE elicited a significant (P < 0.05) increase in the activities of SOD, CAT, and GSH concentrations while decreasing C-reactive protein, signifying promising therapeutic potential. Highlighting interactions between top-scoring phytoligands (epicatechin, catechin, and kaempferol) and COX-2, the findings underscore their drug-like characteristics, favorable pharmacokinetics, and enhanced safety toxicity profiles. Results from in vitro, in vivo, and in silico studies, highlights CPEE remarkable antioxidant and anti-inflammatory potentials.

Abstract:The in vitro study of the antioxidant properties of the hydroalcoholic extracts of various Indian medicinal plants can logically help to develop a better and safer way of amelioration from oxidative stress. As aimed, the present study has been done to estimate and thereby conclude regarding the antioxidant activities of a few Indian medicinal plants, viz., Terminalia chebula, Terminalia belerica, Emblica officinalis, Caesalpinia crista, Cajanus cajan, and Tinospora cordifolia. The extracts of the plants have been subjected to the evaluation of antioxidant properties through scavenging assays for reactive oxygen species like superoxide, nitric oxide, peroxynitrite, hypochlorous acid, singlet oxygen, etc. and measurement of TEAC values and other phytochemical parameters. The phenolic and flavonoid contents of each plant have been found to be correlated to their individual antioxidant activity. The results showed the hydroalcoholic extracts of the plants were efficient indicators of their antioxidant capacity thus concreting their basis to be used as natural antioxidant.

Abstract:Caralluma edulis is a leafless succulent herb native to India and Pakistan, where it is used as emergency food and as potential herbal medicine. It was investigated for its bioactive chemicals and further medicinal properties. The objective of the current study was to accomplish the chemical and biological characterization of C. edulis . Chemical profiling was done through estimation of total bioactive contents and UHPLMS analysis. Biological screening was achieved through six different antioxidant and enzyme inhibition assays. Estimation of its phenolic and flavonoid contents revealed that ethyl acetate fraction (Ce-E) contains 14.15 mg GAE/g extract and 26.13 mg RE/g extract of phenolics and flavonoids, respectively, followed by the methanolic (Ce-M) and the water (Ce-W) soluble fractions. In free radical inhibitory assays, Ce-E fraction also exhibited highest activity (DPPH: 19.72 mg TE/g extract, ABTS: 48.66 mg TE/g extract) followed by Ce-M, whereas, in CUPRAC and FRAP assays, all the extracts, except water fraction, exhibited nearly equal potential in the range of 53.85–57.96 and 25.42–32.12 mg TE/g extract, respectively. In metal chelating antioxidant assay, only Ce-M and Ce-W fractions displayed considerable activities (25.88 and 23.55 mg EDTAE/g extract respectively), whereas, Ce-H exhibited the highest activity in phosphomolybdenum assay (1.03 mmolTE/g extract), against BChE (6.43 mg GALAE/g extract), α-glucosidase and α-amylase (6.98 and 0.47 mmol ACAE/g, respectively) and tyrosinase (64.46 mg KAE/g extract) enzymes. Ce-M and Ce-E also exhibited significant activity against tyrosinase (59.85 and 58.40 mg KAE/g extract, respectively). Crude methanolic extract (Ce-M) was analyzed to unveil its secondary metabolic picture; the UHPLCMS analysis discloses 105 compounds as phenolic acids, flavonoids, steroids and their glycosides, which makes the C. edulis extract a strong candidate for nutraceutical and functional food ingredient. Some of the metabolites were docked against all the tested enzymes to predict mode of actions and to substantiate the pharmaceutical nature of C. edulis .

Abstract:The medicinal plants have been one of the major sources of medicines since the beginning of civilization. There is high demand for plant based medicines, nutraceuticals and cosmeceuticals all across the globe. Herbal medicines are becoming more and more popular in recent years with their over increasing acceptability in both developing and developed countries. All parts of the plant leaf, stem, bark, root, flowers, fruits, seeds, peels, are therapeutically useful. But flowers, seeds and peels are less utilized. Seeds are generally thrown into environment and these waste disposal has become a critical global problem. But they have a rich source of many valuable bioactive compounds especially phenols and flavonoids because of which they can be used therapeutically. In the present work, 6 plant seeds viz. Annona squamosa L., Carica papaya Linn. (Un-ripe), Carica papaya Linn. (Ripe), Ceiba pentandra L. Gaertn., Trachyspermum ammi L. and Trigonella foenum - graecum L. were screened for their phytochemical profile and antioxidant efficacy using different extraction techniques. The seeds were extracted by different extraction methods viz. Traditional methods -decoction, maceration and Modern methods - microwave assisted extraction and ultrasonic assisted extraction. The phytochemicals or bioactive components evaluated were total phenols, phenolic acids, flavonoids, flavonols and proanthocyanidin content. In vitro antioxidant activity in terms of 2, 2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging activity, Superoxide anion free radical (O2-) scavenging activity, 2, 2’-Azino-bis-(3-ethyl) benzothiazoline-6-sulfonic acid (ABTS) radical cation scavenging activity and Ferric reducing antioxidant power (FRAP) was evaluated. All the extracts showed varied levels of phytochemical and antioxidant activity. But the best activity was shown by modern methods especially UAE. Thus, seeds can be profitably, beneficially employed as a rich source of bioactive compounds which can be effectively utilized as a source of natural antioxidant and reduce environmental pollution.

Abstract:Introduction: Natural antioxidants are vital to promote health and treat critical disease conditions in the modern healthcare system. This work adds to the index of natural medicines by exploring the antioxidant potential of Dodonaea viscosa Jacq. (Plant-DV). Material and Methods: The aqueous extract of leaves and flower-containing seeds from plant-DV in freshly prepared phosphate buffer is evaluated for antioxidant potential. In vitro antioxidant potential of the nascent and oxidatively stressed extracts was analyzed through glutathione (GSH) assay, hydrogen peroxide (H2O2) scavenging effect, glutathione-S-transferase (GST) assay, and catalase (CAT) activity. In vivo therapeutic assessment is performed in Wistar Albino rats using vitamin C as a positive control. The livers and kidneys of individual animals are probed for glutathione, glutathione-S-transferase, and catalase activities. Results: flower-containing seeds have GSH contents (59.61 µM) and leaves (32.87 µM) in the fresh aqueous extracts. The hydrogen peroxide scavenging effect of leaves is superior to flower-containing seeds with 17.25% and 14.18% respectively after 30 min incubation. However, oxidatively stressed extracts with Ag(I) and Hg(II) show declining GSH and GST levels. The plant extracts are non-toxic in rats at 5000 mg/Kg body weight. Liver and kidneys homogenate reveal an increase in GSH, GST, and CAT levels after treatment with 150 ± 2 mg/kg and 300 ± 2 mg/kg body weight plant extract compared with normal saline-treated negative and vitamin C treated positive control. Discussion: The crude aqueous extracts of leaves and flower-containing seeds of plant-DV show promising antioxidant potential both in in vitro and in vivo evaluation.

Abstract:Introduction and Aim: Free radicals have been established detrimental effects on human health, mostly because of oxidative stress. Antioxidants are essential in reducing these effects and protecting cells from oxidative damage. As a result, there is rising interest in finding natural antioxidants. In this work, Polystichum acrostichoides and Adiantum lunulatum aerial parts, both widely utilized in medical systems for their therapeutic effects and as sources of free radicals, were examined to see how solvent extraction affected their antioxidant activity. Materials and Methods: In both plants' methanolic and hexane extracts, the amounts of phenolic and flavonoid compounds were measured. Utilizing the ABTS, DPPH, and hydroxyl radical scavenging tests, the antioxidant activity of the extracts was evaluated. Results: In both plant species, the methanolic extracts had higher phenolic and flavonoid contents than the hexane extracts. Notably, the ABTS, DPPH, and hydroxyl radical scavenging assays revealed that the methanol extract had the best antioxidant potency. On the other hand, there was no apparent antioxidant action in the hexane extracts. Conclusion: The results suggest that the aerial parts of Polystichum acrostichoides and Adiantum lunulatum could be effective sources of antioxidants. In the ABTS, DPPH, and hydroxyl radical scavenging experiments, the methanolic extracts had reduced IC50 values, which indicated that they had considerable antioxidant activity. These results demonstrate the potential of these ferns in the creation of innovative treatments for a range of oxidative stress-related diseases.
Screening of Phytochemical Profile and Biological Activities in the Leaves, Stems and Roots of South African Portulacaria Afra using Four Extraction Solvents

Abstract:Portulacaria afra, is indigenous to South Africa and has been identified to have several medicinal properties according to traditional knowledge and few studies. The drive around this research is to evaluate the medicinal properties of the leaves, stems and for the first time the roots extracts of Portulacaria afra, using four solvents with different polarities. The aqueous (60°C), methanol, n-hexane and ethyl acetate whole plant extracts of P. afra were investigated for their phytochemical properties, antimicrobial and antioxidant activity. The phytochemical screening revealed that the methanolic and aqueous extracts of the leaves displayed high presence of secondary metabolites compared to n-hexane and ethyl acetate extracts. The methanolic leaves extracts showed strong presence of quinones, phenols, steroid and coumarins while the aqueous leaves extracts contained a moderate presence of saponins, terpenoids, quinones and coumarins. Ethyl acetate leaves extracts revealed a strong presence of tannins, moderate presence of phytosteroids and a low presence of volatile oil. Meanwhile, the leaves extracts with n-hexane showed a considerable amount of saponins with a moderate presence, and a low presence of tannins, volatile oils and terpenoids. The methanolic stems extracts displayed the most significant presence of secondary metabolites, showing a high presence of terpenoids, steroids, phenols and coumarins. The aqueous stems extracts showed a strong presence of glycosides with a moderate presence of saponins. However, ethyl acetate and n-hexane stems extracts displayed a few secondary metabolites with their concentration ranging from medium to low. The ethyl acetate roots extracts displayed a significant elevated amount of quinones with a strong presence. n-hexane roots extracts showed a moderate presence of volatile oil and a low presence of tannins and steroids. Methanolic roots extracts showed a moderate presence of coumarins and glycosides while aqueous roots extracts showed a low presence of glycosides. The overall highest total phenolics contents (TPCs) and total flavonoids contents (TFCs) in all the plant parts, were found to be in the methanol stems extracts and aqueous roots extracts respectively. Next to the methanol leaves and aqueous leaves extracts respectively. However, in the root’s extracts, the aqueous extracts showed the highest total phenolics content while the water extracts had the highest total flavonoids contents. The antimicrobial activities of P. afra whole plant extracts with the various four solvents were tested against three microorganisms Escherichia coli, Staphylococcus aureus and Streptomyces griseus using agar-well diffusion method. The Antimicrobial activity of the n-hexane extracts of the leaves, stems and roots of P. afra presented a wide range of inhibition against all the test microorganisms, ethyl acetate leaves extract showed a considerable effect against Staphylococcus aureus while the methanolic extracts were not active. Aqueous roots extracts demonstrated a strong antimicrobial activity against Staphylococcus aureus while the other extracts were not active. The zones of inhibition ranged from 13 to 24 mm for the plant extracts. The antioxidant activity potential of the aqueous, methanol, n-hexane and ethyl acetate extracts of P. afra leaves, stems and roots extracts were observed through a 2, 2 diphenylpicryhydrazyl (DPPH) free radical assay, hydrogen peroxide scavenging (H2O2) and metal chelating activity assay. Ethyl acetate roots extracts exhibited the strongest hydrogen peroxide scavenging activity compared to the other extracts. Meanwhile, aqueous stems extracts showed the highest antioxidant activity against DPPH radical. Aqueous and n-hexane roots extracts displayed the strongest metal chelating ability. These findings reveal the efficacy of the use of several solvents with different polarities for effective and more accurate extraction of various compounds and indicate that the antimicrobial and antioxidant activity of P. afra parts are dependent on the solvent extracts.

Abstract:Background: Irvingia gabonensis extracts are documented to possess antidiabetic, hypocholesterolemic, and antioxidant activities. The phytochemical components of medicinal plants are responsible for their therapeutic properties, particularly those with antioxidant properties. Objectives: The quantitative phytochemicals and in vitro antioxidant properties of the aqueous and ethanol extracts of I. gabonensis stem bark were assessed in this study. Materials and Methods: Standard techniques were used to measure the concentrations of phenols, flavonoids, tannins, alkaloids, steroids, and saponins in water and alcohol extracts of this plant. Total Antioxidant Power (TAP) and the extract's capacity to scavenge free radicals including superoxide, nitric oxide, 2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2-azino-bis-(3-ethylbenzoth iazoline-6-sulfonic acid) (ABTS+), β-carotene, and hydrogen peroxide were also evaluated. The extracts' Ferric-Reducing Potential (FRAP) was also assessed. Results: The alcohol extract had significantly (P < 0.05) higher concentrations of all the phytochemicals measured as well as near significantly (P <0.066) higher TAP in comparison to the aqueous extracts. In general, the in vitro antioxidant capacity of the ethanol extract surpassed that of the water extract. The ethanol extract had IC 50 values that were comparable to or less than the reference standards for its ability to neutralize nitric oxide, H 2 O 2 , superoxide, and DPPH radicals. Conclusion: The outcomes of the quantitative phytochemical analysis and in vitro antioxidant effects of aqueous and ethanol extracts of I. gabonensis stem bark clearly show their potential as an excellent reservoir of bioactive compounds and scavengers of deleterious oxidants; properties that could be explored therapeutically.

Abstract:Abstract Background Rural populations in Uganda rely heavily on medicinal plants for the treatment of bacterial skin infections. However, the efficacy of these medicinal plants for their pharmacological action is not known. The study aimed at evaluating the antibacterial, antioxidant, and sun protection potential of Spermacoce princeae , Psorospermum febrifugum , Plectranthus caespitosus , and Erlangea tomentosa extracts. Methods The plant samples were extracted by maceration sequentially using hexane, dichloromethane, ethyl acetate, methanol, and distilled water. Antibacterial activity of each extract was carried out using an agar well diffusion assay against Pseudomonas aeruginosa , Staphylococcus aureus , Escherichia coli , Klebsiella pneumonie , Streptococcus pyogenes , and Salmonella typhi . Acute dermal toxicity of the aqueous extract of S. princeae and P. febrifugum , and E. tomentosa was assessed in young adult healthy Wistar albino rats at a dose of 8000 and 10,000 mg/kg body weight. The antioxidant activity of each extract was carried out using a 1,1-diphenyl-2-picryl-hydrazyl (DPPH) radical scavenging assay. The sun protection factor was determined using Shimadzu UltraViolet-Visible double beam spectrophotometer between 290 and 320 nm. Results The plant extracts showed good antibacterial activity against the tested bacterial strains with minimum inhibitory concentration (MIC) ranging between 3.12 and 12.5 mg/ml. There was no significant change in the levels of creatinine, alanine aminotransferase, and aspartate aminotransferase in the rats even at a higher dose of 10,000 mg/kg, which was related to the results of biochemical analysis of the blood samples from the treated and control groups. The aqueous and methanol extracts of S. princeae showed potential antioxidant properties, with half maximal inhibitory concentration (IC 50 ) values of 59.82 and 61.20 μg/ml respectively. The organic and aqueous extracts of P. caespitosus showed high levels of protection against Ultraviolet light with sun protection potential values ranging between 30.67 and 37.84. Conclusions The study demonstrated that the selected medicinal plants possessed good antibacterial, antioxidant, and sun protection properties. Therefore, the plants are alternative sources of antibacterial, antioxidant, and sun protection agents in managing bacterial skin infections.
African Under-Utilized Medicinal Leafy Vegetables Studied by Microtiter Plate Assays and High-Performance Thin-Layer Chromatography–Planar Assays

Abstract:Biological activities of six under-utilized medicinal leafy vegetable plants indigenous to Africa, i.e., Basella alba, Crassocephalum rubens, Gnetum africanum, Launaea taraxacifolia, Solanecio biafrae, and Solanum macrocarpon, were investigated via two independent techniques. The total phenolic content (TPC) was determined, and six microtiter plate assays were applied after extraction and fractionation. Three were antioxidant in vitro assays, i.e., ferric reducing antioxidant power (FRAP), cupric reduction antioxidant capacity (CUPRAC), and 2,2-diphenyl-1-picrylhydrazyl (DPPH) scavenging, and the others were enzyme (acetylcholinesterase, butyrylcholinesterase, and tyrosinase) inhibition assays. The highest TPC and antioxidant activity from all the methods were obtained from polar and medium polar fractions of C. rubens, S. biafrae, and S. macrocarpon. The highest acetyl- and butyrylcholinesterase inhibition was exhibited by polar fractions of S. biafrae, C. rubens, and L. taraxacifolia, the latter comparable to galantamine. The highest tyrosinase inhibition was observed in the n-butanol fraction of C. rubens and ethyl acetate fraction of S. biafrae. In vitro assay results of the different extracts and fractions were mostly in agreement with the bioactivity profiling via high-performance thin-layer chromatography–multi-imaging–effect-directed analysis, exploiting nine different planar assays. Several separated compounds of the plant extracts showed antioxidant, α-glucosidase, α-amylase, acetyl- and butyrylcholinesterase-inhibiting, Gram-positive/-negative antimicrobial, cytotoxic, and genotoxic activities. A prominent apolar bioactive compound zone was tentatively assigned to fatty acids, in particular linolenic acid, via electrospray ionization high-resolution mass spectrometry. The detected antioxidant, antimicrobial, antidiabetic, anticholinesterase, cytotoxic, and genotoxic potentials of these vegetable plants, in particular C. rubens, S. biafrae, and S. macrocarpon, may validate some of their ethnomedicinal uses.

Abstract:Carica papaya is widely used in folk medicine as herbal remedy to prevent, protect against, and cure several diseases. These curative properties are based on the presence in different parts of the plant of phytochemical nutrients with antioxidant effect. Seeds are the less exploited part; thus this study is aimed at assessing the antioxidant activities of the C. papaya seeds water extract against hydrogen peroxide (H 2 O 2 ) oxidative stress in human skin Detroit 550 fibroblasts. C. papaya seeds water extract is not toxic and acts as a potent free radical scavenger, providing protection to Detroit 550 fibroblasts that underwent H 2 O 2 oxidative stress. Data show that (i) the maximum protective effect is achieved by the simultaneous administration of the extract with 1 mM H 2 O 2 ; (ii) the extract in presence of an oxidative stress does not increase catalase activity and prevents the release of cytochrome C and the inner mitochondrial transmembrane potential ( Δ ψ m ) loss; (iii) the extract is more efficient than vitamin C to hamper the oxidative damage; (iv) the purified subfractions of the seeds water extract exert the same antioxidant effect of whole extract. In conclusion, C. papaya seeds water extract is potentially useful for protection against oxidative stress.

Abstract:Oxidative stress is a common characteristic of psychiatric, neurological, and neurodegenerative disorders. Therefore, compounds that are neuroprotective and reduce oxidative stress may be of interest as novel therapeutics. Phenolic, flavonoid and anthocyanin content, ORAC and DPPH free radical scavenging, and Cu2+ and Fe2+ chelating capacities were examined in variations (fresh/capsule) of Queen Garnet plum (QGP, Prunus salicina), black pepper (Piper nigrum) clove (Syzygium aromaticum), elderberry (Sambucus nigra), lemon balm (Melissa officinalis) and sage (Salvia officinalis), plus two blends (Astralagus membranaceus-lemon balm-rich, WC and R8). The ability of samples to prevent and treat H2O2-induced oxidative stress in SH-SY5Y cells was investigated. Pre-treatment with WC, elderberry, QGP, and clove prevented the oxidative stress-induced reduction in cell viability, demonstrating a neuroprotective effect. Elderberry increased cell viability following oxidative stress induction, demonstrating treatment effects. Clove had the highest phenolic and flavonoid content, DPPH, and Cu2+ chelating capacities, whereas QGP and elderberry were highest in anthocyanins. Black pepper had the highest ORAC and Fe2+ chelating capacity. These findings demonstrate that plant extracts can prevent and treat oxidative stress-induced apoptosis of neuron-like cells in vitro. Further research into phytochemicals as novel therapeutics for oxidative stress in the brain is needed.

Abstract:This paper summarizes our research for herbal extracts with potent antioxidant activity obtained from a large scale screening based on superoxide radical (O(2) (•-)) scavenging activity followed by characterization of antioxidant properties. Firstly, scavenging activity against O(2) (•-) was extensively screened from ethanol extracts of approximately 1000 kinds of herbs by applying an electron spin resonance (ESR)-spin trapping method, and we chose four edible herbal extracts with prominently potent ability to scavenge O(2) (•-). They are the extracts from Punica granatum (Peel), Syzygium aromaticum (Bud), Mangifera indica (Kernel), and Phyllanthus emblica (Fruit). These extracts were further examined to determine if they also scavenge hydroxyl radical ((•)OH), by applying the ESR spin-trapping method, and if they have heat resistance as a desirable characteristic feature. Experiments with the Fenton reaction and photolysis of H(2)O(2) induced by UV irradiation demonstrated that all four extracts have potent ability to directly scavenge (•)OH. Furthermore, the scavenging activities against O(2) (•-) and (•)OH of the extracts of P. granatum (peel), M. indica (kernel) and P. emblica (fruit) proved to be heat-resistant.The results of the review might give useful information when choosing a potent antioxidant as a foodstuff. For instance, the four herbal extracts chosen from extensive screening possess desirable antioxidant properties. In particular, the extracts of the aforementioned three herbs are expected to be suitable for food processing in which thermal devices are used, because of their heat resistance.

Abstract:Introduction: Phenolic phytochemicals are known for antioxidant-mediated pharmacological effects in various diseases (diabetes, cancer, CVDs, obesity, inflammatory and neurodegenerative disorders). However, individual compounds may not exert the same biological potency as in combination with other phytochemicals. Cyamopsis tetragonoloba (Guar), an underutilized semi-arid legume which has been used as a traditional food in Rajasthan (India), is also a source of the important industrial product guar gum. However, studies on its biological activity, like antioxidant, are limited. Methods: We tested the effect of C. tetragonoloba seed extract to enhance the antioxidant activity of well-known dietary flavonoids (quercetin, kaempferol, luteolin, myricetin, and catechin) and non-flavonoid phenolics (caffeic acid, ellagic acid, taxifolin, epigallocatechin gallate (EGCG), and chlorogenic acid) using DPPH radical scavenging assay. The most synergistic combination was further validated for its cytoprotective and anti-lipid peroxidative effects in in vitro cell culture system, at different concentrations of the extract. LC-MS analysis of purified guar extract was also performed. Results and discussion: In most cases, we observed synergy at lower concentrations of the seed extract (0.5-1 mg/ml). The extract concentration of 0.5 mg/ml enhanced the antioxidant activity of Epigallocatechin gallate (20 µg/ml) by 2.07-folds, implicating its potential to act as an antioxidant activity enhancer. This synergistic seed extract-EGCG combination diminished the oxidative stress nearly by double-fold when compared with individual phytochemical treatments in in vitro cell culture. LC-MS analysis of the purified guar extract revealed some previously unreported metabolites, including catechin hydrate, myricetin-3-galactoside, gossypetin-8-glucoside, and puerarin (daidzein-8-C-glucoside) which possibly explains its antioxidant enhancer effect. The outcomes of this study could be used for development of effective nutraceutical/dietary supplements.
