Abstract:Objective:To study the electrophysiological role in locating the segment of lumbar nerve dorsal roots in rat.Method:L3-L5 never rooots of rat were stimulated by means of electrophysiological method,the maximum amplitudes of the sensory nerve action potentia(SNAP) from two subdermal needle electrode at different muscle of bilater lower limb were recorded.Result:The recorded waveform mostly was typically polyphasic wave.When stimulating L3,the maximum amplitudesrecoreded in quadriceps femoris(480.8±255.9μV) was higher than those in other muscles significantly(P<0.01);When stimulating L4,the maximum amplitudes recorded in quadriceps femoris(305.2±131.5μV) and tibialis anterior(340.2±310.4μV) were higher than those in biceps femoris(142.4±144.7μV) and gastrocnemius(138.2±127.6μV) significantly(P<0.01);When stimulating L5,the maximum amplitudes recorded in biceps femoris(377.5±264.4μV) and gastrocnemius(168.4±126.7μV)biceps femoris(121.5±120.6μV) were higher than those in quadriceps femoris(54.1±98.26μV) significantly(P<0.01).Conclusion:Stimulating different lumbar never dorsal rooots in rat will lead to different maximum amplitudes of SNAP(the earliest maximum amplitudes) recorded in different lower limb muscles of rat,which can been refered to locate the segment of the stimulated lumbar nerve dorsal roots.
Inter‐Strain Differences in the Lumbar Spinal Cord Anatomy and Neuromorphology: Wistar Versus Dark Agouti Rats

Abstract:Comparison of anatomy and morphology of spinal cord of Wistar (W) and Dark Agouti (DA) rats. Top: an algorithm of the analysis of the neurochemical labeling on the spinal cord slices using NeuN‐staining and a ration between the number of motor neurons and non‐motor neurons in DA and W rats. Bottom: a ratio between an area of the gray matter and white matter in DA and W rats; and a ratio between the number of neurons expressing parvalbumin (PV) and calbindin (CB) in regions of interest (ROI) in DA and W rats. Rat strains differ in physiology, behavior, and recovery after central nervous system injury. To assess these differences, we compared the gross and local anatomy and neuromorphology of the lumbar spinal cord of the Wistar and Dark Agouti (DA) strains. The key findings include (i) distinct spatial relationships between vertebrae and spinal segments in the two strains; (ii) Wistar rats have larger volumes of spinal cord gray and white matter; (iii) DA rats have smaller total neuronal populations, thus indicating an expectation of smaller local neuronal populations; (iv) this expectation was confirmed for interneurons expressing calbindin 28 kDa. But contrary to expectations, (v) DA rats had more numerous populations of the interneurons expressing parvalbumin and a population of α‐motoneurons. Consequently, these strains displayed divergent ratios in specific spinal neuronal populations. Researchers should consider these inter‐strain differences when comparing data across different strains.

Abstract:Objective: To investigate afferent pathways of rat sacroiliac joint with neural retro-tracing technique.Methods:Thirty male Sprague-Dawley rats were randomly divided into sympathectomy group(group A,of which left paravertebral sympathetic trunk were cut down)and unsympathectomy group(group B).20 μl 30% horseradish peroxidase (HRP) was injected into left sacroiliac joints in two groups.After 72 hours ,bilateral L_1-S_1 dorsal root ganglia (DRG) were harvested and processed by TMB method,in which HRP-positive neurons were observed and counted under light microscope.Results:There were significant differences in the number of HRP-positive neurons in left L_1,L_2 DRG between group A and group B (P<0.05).HRP-positive neurons in group B obviously decrease than group A.However,there was no significant differences in left L_3-S_1 DRG were observed between group A and group B (P>0.05).Conclusion:Afferent neurons of govern same side sacroiliac joint were included by L_1-S_1 DRG.The same side paravetebral sympathetic trunk might be an important neural pathway between sacroiliac joint and L_1,L_2 DRG.However,it might be not a key bypass from sacroiliac joint to L_3-S_1 DRG.
BOTH PEPTIDERGIC AND NONPEPTIDERGIC SMALL NEURONS IN RAT DRG EXPRESS GABABR --EVIDENCE FOR PAIN REGULATION BY PRESYNAPTIC MECHANISM IN SPINAL LEVEL

Abstract:It has been reported that the small type of neurons in the dorsal root ganglion (DRG) play an important role in pain regulation by a presynaptic mechanism via the metabotropic type-B γ-aminobutyric acid receptors ( GABABR ). In order to understand whether the 2populations of the small type of the neurons, peptidergic and nonpeptidergic, in DRG share the same role, immunoflourescent histochemical methods and confocal laser scanning microscope were employed to investigate the expression of the GABABR in the peptidergic and nonpeptidergic small DRG neurons. The results revealed that 92% of the peptidergic and 90% of nonpeptidergic small DRG neurons express GABABR in their perikarya and central processes, which distribute in the various laminae of the spinal dorsal horn. These results suggest both the peptidergic and nonpeptidergic populations of the small neurons in the DRG share similar role in pain modulation via presynaptic mechanisms but in given laminae of the spinal dorsal horn.

Abstract:Background/Purpose: The purpose of this experiment was to identify the neurons in the lumbosacral spinal cord involved in colon-rectal function and to compare normal and anorectal malformation of fetal rats.Methods: The authors quantified the sacral parasympathetic nucleus (SPN) innervation of the rectum by Fluorogold (FG) (Fluorochrome, Englewood, CO) retrograde tracing experiment in fetal rats with normal and anorectal malformation. Anorectal malformation was induced in rat fetuses by ethylenethiourea (ETU). The number of FG-labeled SPNs was scored and compared between male fetuses with or without-fed group and control groups. malformation in the ETU-fed group and control groups.Results: The number of FG-labeled SPNs in the fetuses without a defect, with ETU injected but without any defects of the anorecturn or neural tube, with low-type deformity, and with high-type deformity were (mean +/- SEM) 47.3 +/- 2.9, 45.6 +/- 3.2, 24.2 +/- 3.8, and 8.5 +/- 2.5, respectively. Fluorogold-labeled SPNs in the fetuses with high-type deformity were significantly fewer than those in fetuses without defects (P < .05) and in controls (P < .05).Conclusions: These findings suggest that defective SPN innervation to the rectum is a primary anomaly that coexists with the alimentary tract anomaly in anorectal malformation during fetal development. The intrinsic neural deficiency is an important factor likely to contribute to poor postoperative anorectal function despite surgical correction of the malformation. (c) 2007 Elsevier Inc. A 3D atlas of sexually dimorphic lumbosacral motor neurons that control and integrate pelvic visceral and somatic functions in rats

Abstract:The lumbosacral spinal motor system comprises somatic motor neurons (SMNs) that target striated muscle and visceral motor neurons (VMNs; autonomic preganglionic neurons) that target peripheral ganglia. The brain coordinates these pathways to mediate functions such as continence, voiding and sexual function by ensuring appropriate changes of organ state with striated muscle activity (e.g., sphincter opening, relevant postural changes). These spinal circuits are important therapeutic targets for bioelectronic medicine yet are poorly defined in comparison with limb SMNs. We aimed to define the structural features and relationships between SMNs and VMNs relevant to pelvic function in male and female rats, by building 3D maps of lumbosacral spinal cord. We achieved this by applying large-volume immunostaining (choline acetyltransferase), with tissue clearing and advanced whole mount microscopy (light sheet and ribbon scanning confocal microscopy). We identified VMNs specific to pelvic organ function by microinjecting cholera toxin (beta-subunit) into the major pelvic ganglia (MPG). These VMNS are primarily located in segments L1-L2 (dorsal commissural nucleus) and L6-S1 (intermediolateral nucleus). Unexpectedly, small groups of SMNs in the urethral rhabdosphincter, cremaster and levator ani motor pools also projected through the MPG. Motor neuron counts and analysis of dendritic fields detected sexual dimorphism in both SMNs and VMNs. Their colocation of dendritic bundles suggested a mechanism of coordinating activity. This study has provided the first multiscale 3D atlas of rat lumbosacral cord. This will be shared as a resource on an open science platform (sparc.science) to allow further exploration and modeling of network features and cellular morphology.

Abstract:The spinal cord is the most phylogenetically ancient part of the central nervous system. Due to a faster growth of the spinal column relative to the spinal cord in ontogenesis, some spinal segments in adult mammals are shifted rostrally relative to the eponymous vertebrae, which is called the ascent of the spinal cord. Based on the literature data, the skeletotopy of the lumbar spinal cord was compared in 17 mammalian species, while in 4 of them, it was additionally compared in newborn and adult animals. The length ratio of the L2 segment and VL2 vertebra, as well as the number of the vertebra that encloses the 29th spinal segment, were determined as measures characterizing the degree of spinal cord ascent. Based on the literature data, digital dexterity and the encephalization quotient were also determined as measures frequently used in comparative studies. It was shown that different species differ to a greater extent in the relative length of the rostral lumbar segments, whereas within the same species, newborns differ from adults in the relative length of the caudal lumbar segments. In most species, the degree of spinal cord ascent positively correlates with digital dexterity and the encephalization quotient. The above-mentioned macroanatomical characteristics of the spinal cord may be used to analyze the relationships between adaptive mechanisms in various mammalian species.

Peptidergic neurohemal system of an insect: mass spectrometric morphology

Abstract:Neuropeptides are by far the most diverse group of messenger molecules in insects. To understand cell signaling and function, it is essential to reveal the complete neuropeptide profile of a single neuron/nerve/neurohemal organ first. In this study, matrix-assisted laser desorption ionization time of flight (MALDI-TOF) mass spectrometry was used to analyze the peptidergic system of an insect, focussing on the neurohemal structures. Major neurohemal organs were investigated, including the retrocerebral complex, perisympathetic organs, and all nerves supplying these organs with neurosecretions. Additionally, peripheral neurohemal release sites such as the dilator muscle of the antennal circulatory organ and lateral heart nerves were studied, as well as parts of the stomatogastric nervous system. The following neuropeptide families were analyzed: kinins, allatostatins, leucomyosuppressin, corazonin, adipokinetic hormones, myoinhibitory peptide, sulfakinins, periviscerokinins, YLSamide, VEAacid, SKNacid, proctolin, the head peptide, and pyrokinins. Beyond a contribution to a map of the distribution of neuropeptides in a neurohemal system, the following conclusions can be drawn from these experiments. (1) Nearly all abundant peaks in the different mass spectra represent peptides that have already been identified. (2) Although only adult males were used in this study, variations in the peptide abundances were observed that are possibly correlated with different physiological/developmental conditions. (3) Peptides have a body-region-specific distribution in the neurohemal system. (4) A clear compartmentalisation of the retrocerebral complex could be observed.

Abstract:Kornhuber and Deecke (1965) first described a slowly increasing surface negative potential which starts about 800 ms before movement onset and was termed “Bereitschaftspotential” (BP). The BP had a widespread bilateral distribution over precental and parietal regions with a maximum at the vertex. In a series of studies it was shown that the BP amplitude was influenced by psychological and physical parameters of the movement. The psychological parameters of the movement included motivation (McAdam and Seals, 1969), attention, and intentional involvement of the subject (Kornhuber and Deecke, 1965). The physical parameters of the movement included speed (Becker et al.. 1976; Hazemann et al., 1978). force (Kutas and Donchin, 1974; Wilke and Lansing, 1973; Becker and Kristeva, 1980) and bilaterality of the movement (Shibasaki and Kato, 1975; Kristeva et al., 1979). All of these studies used a self-paced simple movement. There have been only two studies on the BP prior to a skilled motor task (Taylor, 1978; Papakostopoulos, 1980). While the components of the skilled motor tasks used by these authors were familiar, the sequence required was unfamiliar to the subjects and was acquired during the experiment. To our knowledge, there is no published account for the BP prior to a well-learned complex sequence of skilled motor movements. A good example of such a task is the finger movement of a piano player. Most probably these movements do not rely on sensory feedback since “there is no time to correct errors’’ (Cooper, 1980). These movements are therefore preprogrammed. The BP prior to such movements requiring a complex motor program would be expected to have a large amplitude and would start early. We studied movements of a pianist playing the piano. Two situations-playing a single note and playing a melody-were compared.

Abstract:Electrophysiological properties, reaction to substance P (SP) receptor agonist, and co-existence of glutamate (Glu) and SP in neurons of the spinal dorsal root ganglion (DRG) of the rat were investigated in vitro. The main results were: (1) According to the fiber conduction velocity, totally 135 intracellularly recorded DRG neurons were divided into A alpha/beta type (> 12 m/s) and C type (< 1.3 m/s). There were remarkable differences between the fast after hyperpolarization (fAHP) of action potential between the two types: fAHP of C type neurons had smaller amplititude and longer duration, whereas the fAHP of A alpha/beta type neurons had larger amplititude and shorter duration. (2) Among 22 DRG neurons of which overshoots of action potential appeared, specific SP receptor agonist Sar-SP induced depolorization in 42% (5/12) of the A alpha/beta type neurons and 80% (8/10) of the C type neurons. (3) Biocytin was intracellularly injected into the 22 Sar-SP administrated DRG neurons. After fixation and section, employing the immunofluorescent histochemical multi-staining technique for phosphate-activated glutaminase (PAG, a specific marker for glutamatergic neurons), SP and biocytin, 25% (3/12) of the A alpha/beta type neurons and 80% (8/10) of the C type neurons showed both PAG- and SP-like immunoreactivities. There was a great difference between the percentages of the PAG/SP co-existing A alpha/beta type neurons and C type neurons (P < 0.05). The present results indicated that there were some differences in chemicoanatomical and electrophysiological properties between A alpha/beta type and C type neurons, and SP might facilitate the discharge of DRG neurons through activating SP autoreceptors on their membrane.

Fiber arrangements of cutaneous and muscular rami in cervical nerves of the rat--regarding conception of the spinal nerve stratum-structure

Abstract:Fluorescence tracers DiO and DiI were applied to compare fiber arrangements of cutaneous and muscular rami in the proximal region of anterior and posterior branches of cervical nerves. Results show that a ventrodorsal relationship of cervical nerves is observed in gross level; however, fiber arrangements of cutaneous and muscular rami differ in anterior branches according to distance from the anterior root and dorsal root ganglia. That is, the fibers have a scattering course (no ventrodorsal relationship) in distal regions. Moreover, just before entering anterior roots, the motor fibers of muscular ramus run through the ventral part and sensory fibers of muscles and fibers of cutaneous ramus run through the dorsal part and intersect with motor fibers of the posterior muscular ramus. In contrast with the anterior one, in posterior branches of cervical nerves, cutaneous and muscular rami fibers are very regularly arranged in the distal and proximal region. Based on these results, the ventrodorsal relationship of fiber arrangements could exist in posterior branches and proximal region of anterior branches. Present results differ with those of the spinal nerve stratum-structure; therefore, they are insufficient to elucidate the relationship of peripheral nerves by conception of spinal nerve stratum-structure.

Abstract:Analysis of responses on electrical stimulation is one of the experimental paradigms to study the excitability of the nervous system. In particular, the technique of recording muscle responses evoked by electrical epidural stimulation (ES) of the spinal cord (SC) in humans and animals is widely used. In rats decerebrated at the precollicular level, responses of mm. tibialis anterior (TA) and gastrocnemius medialis (GM) on ES of the L2, L4, L6 spinal segments and transvertebral stimulation (TS) of the VL2, VL4, VL6 vertebrae with single and double pulses were analyzed. The currents at which the amplitude of the sensory component of the response for a single pulse and one of the pulses of the pair was maximum were determined. At the minimum of these currents, the ratio of the amplitudes of the sensory component of the response to the first and second pulses to the amplitude of the sensory component of the response to a single pulse was analyzed. For both muscles, a weakening of the response to both pulses of the pair was obtained with TS VL2 and VL4, while when stimulating VL2, the TA response to the second pulse was lower than to the first. On the contrary, with ES of all segments of interest, a facilitation of the response to the second pulse was obtained for both muscles. A similar facilitation was qualitatively observed for two other muscles, mm. iliacus and vastus lateralis . Thus, the use of double pulses during stimulation made it possible to identify the dependence of the response of SC neural networks on the method of their activation (TS or ES). The facilitation of the response to the second pulse during ES is presumably explained by a decrease in presynaptic inhibition due to decerebration.

Abstract:There were HRP labeled cells in middle cervical ganglia, inferior cervical ganglia and C_(5)-C_(7) DRG on experimental sides. Most of the labeled cells were small or middle size. The sum of mean area and the mean optical density of HRP lebeled cells were larger in the middle cervical ganglia and C_(6) DRG than that in inferior cervical ganglia and C_(5) or C_(7) DRG separately (P<0.01). There wasn^t any HRP labeled cell in C_(1)-T_(1) DRG of control side and in trigeminal ganglia.
Identification of CNS Neural Circuitry Involved in the Innervation of C7 Spinal Nerve: a Viral Transsynaptic Tracing Study.

Abstract:OBJECTIVE:Contralateral C7 spinal nerve transfer is a useful operation for the treatment of brachial plexus root avulsion. The recovery of the independent function at the ipsilateral side, however, depends on neural circuitry reorganization in the central nervous system (CNS). This study tried to locate the CNS neuronal elements involved in the innervation of C7 spinal nerve.METHOD:Pseudorabies virus (PRV, TK/gG(-), 2 microlitre), which expressed green fluorescent protein (GFP), was injected into the left C7 spinal nerve in 20 adult Sprague Dawley rats. After rats survived for 6 h, 12 h, 24 h and 36 h, the C1-C7 segments of the spinal cord and brain were processed using a polyclonal immunohistochemical antibody against PRV.RESULTS:PRV-labeled neurons were found mainly in gray matter of the C1-C7 segments of the spinal cord and at the following structures of the brain: lateral vestibular nucleus, lateral paragigantocellular nucleus, A5 cells, red nucleus, primary and secondary motor cortexes, primary and secondary somatosensory cortexes. Although located bilaterally, the PRV-labeled neurons existed predominantly in the ipsilateral side of the spinal cord and the contralateral side of the brain at 6-12 h after injection (p.i.). The number of PRV-labeled neurons in the CNS was increasing with rat's survival time and the distribution of these neurons turned bilateral with no obvious dominance to either side at 24 h and 36 h (p.i.).CONCLUSION:By use of transsynaptic tracing technique with PRV, the anatomically connected set of neurons, which modulates the activity of C7 spinal nerve, is located successfully in the CNS.

Abstract:The distribution of immunoreactivity to the neurokinin3 receptor (NK3R) was examined in segments C7, T11-12, L1-2, and L4-6 of the rat spinal cord. NK3R immunoreactivity was visualized by using two antisera generated against sequences of amino acids contained in the C-terminal region of the NK3R. NK3R-immunoreactive cells were numerous in the substantia gelatinosa of all spinal segments examined as well as the dorsal commissural nucleus of spinal segments L1-2. Isolated, immunoreactive cells were scattered throughout other regions of the spinal cord. The relationship of NK3R-immunoreactivity with neurons was demonstrated by colocalization with microtubule associated protein 2-immunoreactivity in individual cells. Within neurons, NK3R-immunoreactivity was associated predominately with the plasma membrane of cell bodies and dendrites. Within the substantia gelatinosa, 86% of nitric oxide synthase (NOS)-immunoreactive neurons were also NK3R-immunoreactive. Although NOS-immunoreactive neurons were found throughout all other regions of the spinal cord in the segments examined, these were not NK3R-immunoreactive. When preganglionic sympathetic neurons in spinal segments T11-12 and L1-2 were visualized by intraperitoneal injection of Fluorogold, less than 1% of the Fluorogold-labeled neurons were also immunoreactive for NK3R. The large number of NK3R-immunoreactive neurons in the substantia gelatinosa suggests that some effects of tachykinins on somatosensation may be mediated by NK3R.

Abstract:Neuropeptides are involved in numerous brain activities and are responsible for a wide spectrum of higher mental functions. The main purpose of this outline structural qualitative study was to identify the possible immunoreactivity of classical neuropeptides, as well as novel ones such as nesfatin-1, phoenixin (PNX), spexin (SPX), neuromedin U (NMU) and respective receptors within the rat claustrum for the first time. The study shows the novel identification of peptidergic neurotransmission in the rat claustrum which potentially implicates a contribution of this neuropeptide to numerous central neurosecretory mechanisms.

Axons of Passage and Inputs to Superior Cervical Ganglion in Rat

Abstract:Wheat germ agglutinin-horseradish peroxidase was injected into the entire (0.8 μL) or partial (rostral or caudal, 0.1-0.3 μL) superior cervical ganglion (SCG) of the rat (male Sprague-Dawley, N = 35) to examine the distribution of neurons in the middle (MCG) and inferior (ICG) cervical ganglion that send axons bypass the SCG. Whole-mounts of the SCG, cervical sympathetic trunk (CST), MCG, ICG, and sections of the brainstem and spinal cord were prepared. With entire SCG tracer injection, neurons were labeled evenly in the MCG (left: 258, right: 121), ICG (left: 848, right: 681), and CST (up to 770). Some neurons grouped in a single bulge just rostral to the MCG, which we termed as the "premiddle cervical ganglion" (pMCG). The left pMCG (120) is larger and has more neurons than the right pMCG (82). Centrally, neurons were labeled in lamina IX of cervical segments (C1: 18%, C2: 46%, C3: 33%, C4: 3%), intermediate zone of thoracic segments (T1: 31%, T2: 35%, T3: 27%, T4: 7%), and intermediate reticular nuclei (96%) and perifacial zone (4%) of brainstem. The rostral and caudal SCG injection selectively labeled neurons mainly in brainstem, C1-C2 and in T1-T2, respectively. Before projecting to their peripheral targets, many neurons in pMCG, MCG and ICG run rostrally within the CST rather than segmentally through the closest rami, from the level of SCG or above. Neurons in pMCG and MCG may have similar or complementary function and those in brainstem may be involved in the vestibulo-autonomic interaction. Heterogeneity of Rat Corticospinal Neurons.

Abstract:In order to examine the degree of diversity within a population of cortical projection neurons, rat corticospinal cells were retrogradely labeled in vivo by injecting rhodamine-tagged microspheres into the cervical spinal cord, and subsequently studied electrophysiologically and anatomically in neocortical slices maintained in vitro, by use of standard current clamp techniques and a double-labeling protocol (Tseng et al., J. Neurosci. Meth. 37.121-131, 1991). Three different subgroups were distinguished on the basis of their spiking behavior: (1) Adapting cells had a marked fast (50 ms) and slow phase (200 ms) of spike frequency adaptation; (2) regular spiking (RS) cells had only a period of fast adaptation; (3) some regular spiking neurons had prominent depolarizing afterpotentials (DAPs) and could generate bursts of spikes, often in repetitive fashion (RS(DAP) cells). Subgroups of RS(DAP) cells had different patterns of burst responses to depolarizing current pulses, suggesting differences in the types and/or sites of underlying ionic conductances.Adapting cells had a slightly higher membrane input resistance and more prominent slow hyperpolarizing afterpotentials than RS and RS(DAP) neurons; however, the activation of presumed anomalous rectifier current by intracellular hyperpolarizations was less prominent in adapting neurons. Orthodromic stimulation in. layer I evoked presumed excitatory and inhibitory postsynaptic potentials (EPSPs and IPSPs) in all three types of cells, but prominent short-latency IPSPs were found in a higher percentage of adapting neurons.The morphology of electrophysiologically characterized corticospinal neurons was studied following intracellular injection of biocytin. All three spiking types were typical layer V pyramids with apical dendrites reaching layer 1, basal dendrites in infragranular layers, and deep-directed axons that had a moderate density of local collaterals in lower cortical layers. The profuseness of dendrites, examined by Sholl's analysis of two-dimensional, camera lucida-reconstructed neurons was comparable in the three neuronal subgroups, although a smaller somatic area and more slender apical dendritic trunk were found in adapting neurons.Our results suggest that corticospinal cells in rats are a heterogeneous population of projection neurons with respect to their spiking behavior, membrane properties, synaptic connections, and, to a lesser extent, their morphology. This diversity revealed in vitro adds new complexity to the classification of corticospinal neurons. (C) 1993 Wiley-Liss, Inc.
