Abstract:We sought to identify factors associated with being a reservoir district for wild poliovirus in Pakistan. Differences between reservoir and non-reservoir districts were identified using acute flaccid paralysis surveillance data, population census statistics and data from a survey of district health officials (DHOs). Of the 11 poliovirus reservoir districts identified, population density was significantly higher (median 550 persons/km2) than the non-reservoirs (median 175 persons/km2). DHOs from reservoir districts more often reported that planning was affected by refugees and they had more frequent DHO transfers compared with non-reservoir districts. Multivariate analysis confirmed that reservoirs more often had high population density and frequent DHO transfers. Assessment of district-level and management characteristics can supplement surveillance methods to further improve health programmes.

Abstract:SUMMARY Feedback data loss can severely degrade overall system performance. In addition, it can a ﬀ ect the control and computation of the Cyber-physical Systems (CPS). CPS hold enormous potential for a wide range of emerging applications that include di ﬀ erent data tra ﬃ c patterns. These data tra ﬃ c patterns have wide varieties of diversities. To recover various tra ﬃ c patterns we need to know the nature of their underlying property. In this paper, we propose a data recovery framework for di ﬀ erent tra ﬃ c patterns of CPS, which comprises data pre-processing step. In the proposed framework, we designed a Data Pattern Analyzer to classify the di ﬀ erent patterns and built a model based on the pattern as a data pre-processing step. Inside the framework, we propose a data recovery scheme, called Ef-ﬁcient Temporal and Spatial Data Recovery (ETSDR) algorithm to recover the incomplete feedback for CPS to maintain real time control. In this algorithm, we utilize the temporal model based on the tra ﬃ c pattern and consider the spatial correlation of the nearest neighbor sensors. Numerical results reveal that the proposed ETSDR outperforms both the weighted prediction (WP) and the exponentially weighted moving average (EWMA) algorithms regardless of the increment percentage of missing data in terms of the root mean square error, the mean absolute error, and the integral of absolute error.

Abstract:The retinal ganglion cells giving rise to retinohypothalamic projections in the rat were identified using retrograde transport of horseradish peroxidase (HRP) or FluoroGold injected into the suprachiasmatic nucleus (SCN), and using transneuronal transport of the Bartha strain of the swine herpesvirus (PRV-Bartha). When PRV-Bartha is injected into one eye, it is taken up by retinal ganglion cells, replicated, transported to axon terminals in the SCN, and released. There the virus may take one, or both, of two paths to retinal ganglion cells in the contralateral eye: 1) uptake by SCN neurons, replication, and release from the neurons with uptake and retrograde transport in retinal afferents originating in the contralateral retina; 2) transneuronal passage through axo-axonic appositions between retinal afferents in the SCN with subsequent retrograde transport of virus to the contralateral retina. The ganglion cells thus labeled are a homogeneous population of small neurons (mean diameter, 12.8 +/- 2.2 microns and mean area, 81.8 +/- 21.8 microns 2) with sparsely branching dendrites that are widely distributed over the retina. This population is best identified when virus labeling of retinal projections in areas beyond the hypothalamus is eliminated by lateral geniculate lesions that transect the optic tract at its entry into the geniculate complex. The same population is labeled with retrograde tracers but, with both HRP and FluoroGold, other ganglion cells are labeled, presumably from uptake by fibers of passage, indicating that the virus is a more reliable marker for ganglion cells giving rise to retinohypothalamic projections. The ganglion cells identified correspond to a subset of type III, or W, cells.

Abstract:Wheat germ agglutinin-horseradish peroxidase was injected into the entire (0.8 μL) or partial (rostral or caudal, 0.1-0.3 μL) superior cervical ganglion (SCG) of the rat (male Sprague-Dawley, N = 35) to examine the distribution of neurons in the middle (MCG) and inferior (ICG) cervical ganglion that send axons bypass the SCG. Whole-mounts of the SCG, cervical sympathetic trunk (CST), MCG, ICG, and sections of the brainstem and spinal cord were prepared. With entire SCG tracer injection, neurons were labeled evenly in the MCG (left: 258, right: 121), ICG (left: 848, right: 681), and CST (up to 770). Some neurons grouped in a single bulge just rostral to the MCG, which we termed as the "premiddle cervical ganglion" (pMCG). The left pMCG (120) is larger and has more neurons than the right pMCG (82). Centrally, neurons were labeled in lamina IX of cervical segments (C1: 18%, C2: 46%, C3: 33%, C4: 3%), intermediate zone of thoracic segments (T1: 31%, T2: 35%, T3: 27%, T4: 7%), and intermediate reticular nuclei (96%) and perifacial zone (4%) of brainstem. The rostral and caudal SCG injection selectively labeled neurons mainly in brainstem, C1-C2 and in T1-T2, respectively. Before projecting to their peripheral targets, many neurons in pMCG, MCG and ICG run rostrally within the CST rather than segmentally through the closest rami, from the level of SCG or above. Neurons in pMCG and MCG may have similar or complementary function and those in brainstem may be involved in the vestibulo-autonomic interaction. Abstract:After injection of horseradish peroxidase (HRP) into the pelvic nerve of the rat, a small number of HRP-labeled axon terminals were found in the gracile nucleus. Double labeling experiments were also performed: Fluoro-Gold (FG) was injected into the pelvic nerve, while cholera toxin B subunit (CTb) was injected into the gracile nucleus or dorsal faciculus at the fifth and sixth cervical cord segments ipsilateral to the FG injection. About 5% of FG-labeled neurons were labeled with CTb in the L6-, S1- and S2-dorsal root ganglia ipsilateral to the tracer injection. The results suggest that some primary afferent information from pelvic visceral organs may be directly conveyed to gracile nucleus by the primary afferent neurons.
Hypoglossal premotor neurons with rhythmical inspiratory-related activity in the cat: localization and projection to the phrenic nucleus

Abstract:Localization and projection to the phrenic (PH) nucleus were studied in a sample of premotor neurons that directly projected to hypoglossal motoneurons (XII Mns) and showed respiratory-related patterns of activity. The experiments were carried out in cats, under pentobarbital anesthesia. In the first part of the study, the retrograde double-labeling technique was used to reveal the existence of neurons projecting to both the XII and the PH nuclei. Injection of a fluorescent dye (fast blue, FB) into the XII nucleus and another (nuclear yellow, NY) into the PH nucleus retrogradely labeled, with either FB or NY, medullary reticular neurons mainly in the regions ventrolateral to the nucleus of the tractus solitarius (vl-NTS), ventrolateral to the hypoglossal nucleus (vl-XII), and dorsomedial to the nucleus ambiguus (dm-AMB) bilaterally. In addition, some neurons in these regions were labeled with both FB and NY. In the second part of the study, unitary activity was recorded extracellularly from medullary respiratory neurons. In the regions vl-NTS, vl-XII, and dm-AMB, inspiratory neurons were found which antidromically responded to stimulation of the XII nucleus. Some of them also responded antidromically to stimulation of the PH nucleus. Averaging of rectified and integrated XII and PH nerve discharges by spontaneous spikes of single inspiratory neurons in the vl-NTS and dm-AMB regions revealed a facilitation in either XII nerve discharge or both XII and PH nerve discharges after a short latency of monosynaptic range. It is concluded that in the vl-NTS and dm-AMB regions there are inspiratory neurons that are excitatory premotor neurons projecting to XII Mns showing the respiratory-related activity. Some of them have excitatory synaptic connections to XII and PH Mns via bifurcating axons.
Development of the rat phrenic nucleus and its connections with brainstem respiratory nuclei

Abstract:The development of phrenic motoneurons and descending bulbospinal projections to the cervical spinal cord have been examined in prenatal and early postnatal rats with the aid of the carbocyanine dyes DiI and DiA. Phrenic motoneurons could be identified by retrograde labelling as early as E13, while aggregation of phrenic motoneurons into a column and the formation of dendritic bundles became apparent from E16. The initial phrenic motoneuron dendritic bundles were oriented in the dorsolateral and ventromedial directions, while ventrolaterally directed bundles entering the marginal zone appeared by E16, and rostrocaudal bundles were clearly visible by E21. The column of phrenic motoneurons extended rostrocaudally from C2 to C6 at E13 and E14, but this became confined to the C3-5 segments by E21. Two-way tracing of connections between putative brainstem respiratory centres and cervical spinal cord with the carbocyanine dyes, DiI and DiA, indicated that brainstem bulbospinal neurons in the position of the adult ventral respiratory group (VRG) and medial parabrachial (MPB) nuclei appeared to project to the cervical cord white matter as early as E15 and may contribute axons to the grey matter of the cervical cord as early as E17 These findings are consistent with electrophysiological studies of respiratory function development in the fetal rat, which found relatively regular rhythmic phrenic discharge by E20 to 21. In summary, our findings indicate that the structural differentiation of phrenic motoneurons is well-advanced prior to birth and that the descending pathways involved in the control of respiratory function are in place several days before birth.

Abstract:There were HRP labeled cells in middle cervical ganglia, inferior cervical ganglia and C_(5)-C_(7) DRG on experimental sides. Most of the labeled cells were small or middle size. The sum of mean area and the mean optical density of HRP lebeled cells were larger in the middle cervical ganglia and C_(6) DRG than that in inferior cervical ganglia and C_(5) or C_(7) DRG separately (P<0.01). There wasn^t any HRP labeled cell in C_(1)-T_(1) DRG of control side and in trigeminal ganglia.
[Discharge Patterns of Respiratory Related Neurones in the Medial Areas of Nucleus Retrofacialis of Medulla].

Abstract:Experiments were performed on rabbits and SD rats, both anesthetized with urethane (1.0 g/kg, i.v. or i.p.). Discharges of phrenic nerve and the diaphragmatic muscle electric activities were monitored. Discharge patterns of the respiration-related neurons (RRNs) in the medial area of nucleus retrofacialis (mNRF) were recorded and analyzed. Among the 249 RRNs recorded in the mNRF of the rabbits, 118 were inspiratory (I, 47.4%), 91 expiratory (E, 36.5%), and 40 phase-spanning (PS, 16.1%). Both I and E neurons could be further subdivided into 4 subtypes, the PS neurons into E-I and I-E type. In the 153 RRNs recorded in rats, 68 were I (44.5%), 55 E (35.9%), 30 PS (19.6%). It was found that a group of E-I PS neurons that fire preceding phrenic nerve respiratory activity are located in the mNRF, which was believed to be important in the generation of the basic respiratory rhythm.

Abstract:1. Extracellular and intracellular recordings were made from dorsal horn neurones sending their axons through the dorsal columns in cats anaesthetized with chloralose and paralysed with gallamine triethiodide. 2. Seventeen neurones were injected with horseradish peroxidase through the intracellular micro-electrode, recovered from the histological material and shown to send their axons into the dorsal columns. 3. The cells had axonal conduction velocities of 30--47 ms-1; excitatory receptive fields that usually showed multireceptive characteristics, often including input from sensitive mechanoreceptors in glabrous skin; a third of the sample had a marked subliminal fringe to the excitatory field; inhibitory fields were usually situated proximal to the excitatory field and contiguous with it. 4. The cells were located in laminae III, IV and medial V. Dorsal cells had restricted dendritic trees that ascended in an essentially cylindrical volume of tissue through lamina II and often into I; cells intermediate in depth had more primary dendrites than the others, usually dorsally directed into lamina II, and a more extensive rostro-caudal development; deep, medial cells had dendritic trees that radiated extensively from the cell body but were restricted to the transverse plane. Two cells had axons that ascended the dorsolateral funiculus for a few mm before re-entering the dorsal horn, crossing it and reaching the dorsal columns. Collaterals were given off the axons in the grey matter, in the dorsolateral funiculus and the dorsal columns. 5. The form and function of the neurones are discussed.

Abstract:Objective To establish a set of explicit method of isolating phrenic and hypoglossal nerve and recording their discharges,which may lay a foundation for further investigating the function of respiratory motor nerve in physiological and pathological conditions.Methods Ten healthy SD rats (350-450 g) were anesthetized intraperitoneally and subject to cervical midline skin incision.The phrenic nerve,the hypoglossal nerve of the same side were isolated with the bilateral vagotomy under the surgical microscope.Two-polar electrodes were connected to the nerves and multi-channel physiological recorder was used to record the electrical activities of nerves.Results Brachial plexus and posterior belly of digastrics muscles are location markers of phrenic and hypoglossal lnerves respectively.For 60 seconds,the area of phrenic discharging was (36.3 ± 3.2) μV,the amplitude was (42.6 ± 8.30) μtV,maximum peak was 33.5 μV and the minimal peak was-19.5 μV.As to the charges of hypoglossal nerve,the discharging area was (18.7±4.2) μV,the amplitude was (33.6±9.30) μV,maximum peak was 27.9μVand the minimal peak was-22.0 μV.Conclusions There are certain difficulties of isolating and recording the phrenic and hypoglossal nerves,which may hinder the study of the function of respiratory motor nerves.The present study established a convenient way to isolat and record charges of respiratory nerve,which provide anatomic basis for further investigation.
Anatomic Research on Respiratory Function Reconstruction by Transposition of Accessory Nerve to Phrenic Nerve in the Rats with Upper Cervical Cord Injuries

Abstract:Objective:To provide microanatomical data for respiratory function reconstruction by transposition of accessory nerve to phrenic nerve in rats with upper cervical cord injuries. Methods:The bilateral accessory nerves,phrenic nerves,and their branches of 30 male rats were dissected under operating microscopy. The diameter, length of the accessory nerve and its branches, the distance between the accessory nerve and the main trunk of the phrenic nerve were measured adopting the slide gauge with precision of 0.001 millimeter. Results: The proximal part of the main trunk of phrenic nerve was about 0.207±0.051 mm in diameter. At the point of 4.191±0.135mm from jugular foramen, the accessory nerve gave off muscular branches for sternocleidomastoid muscle, which were 5.657±0.283 mm in length and 0.221±0.039 mm in diameter. The distance between the former and the main trunk of the phrenic nerve was about 10.041±3.181 millimeters. The supraclavicular section of the accessory nerve, between its two muscular branches for trapezius, was about 0.274±0.032 mm in diameter and about 5.154±0.782 mm parting from the proximal part of the main trunk of the phrenic nerve. The length of infraclavicular section of the accessory nerve was about (0.216±0.027)mm before giving off medial and lateral branches horizontally. Its distance from the proximal part of main trunk of the phrenic nerve was about -1.416±0.216 mm. Conclusions: The diameter of infraclavicular section of the accessory nerve before giving off medial and lateral branches is similar to that of the proximal part of main trunk of phrenic nerve. It is possible to suture them directly.

Abstract:We implement a binary collision approximation to study pulse propagation in a chain of o-rings. In particular, we arrive at analytic results from which the pulse velocity is obtained by simple quadrature. The predicted pulse velocity is compared to the velocity obtained from the far more resource-intensive numerical integration of the equations of motion. We study chains without precompression, chains precompressed by a constant force at the chain ends (constant precompression), and chains precompressed by gravity (variable precompression). The application of the binary collision approximation to precompressed chains provides an important generalization of a successful theory that had up to this point only been implemented to chains without precompression, that is, to chains in a sonic vacuum.

Abstract:This study describes the substructure of the dorsal lateral geniculate nucleus of the thalamus of the pigmented rat (Rattus norvegicus) based on the eye-of-origin of its retinal ganglion cell inputs. We made monocular intra-ocular injections of the B-subunit of cholera toxin (CTB), a sensitive anterograde tracer, in three adult male Long-Evans rats. In four additional subjects, we injected fluorophor-conjugated CTB in both eyes, using a different fluorophor in each eye. Brains of these subjects were fixed and sectioned, and the labeled retinal ganglion cell termini were imaged with wide-field sub-micron resolution slide scanners. Retinal termination zones were traced to reconstruct a three dimensional model of the ipsilateral and contralateral retinal termination zones in the dLGN on both sides of the brain. The dLGN volume was 1.58 \pm0.094 mm^{3}, comprising 70 \pm 3% the volume of the entire retinorecipient LGN. We find the retinal terminals to be well-segregated by eye of origin. We consistently found three or four spatially separated ipsilateral-recipient zones within each dLGN, rather than the single compact zone expected. It remains to be determined whether these subdomains represent distinct functional sublaminae.
Nitric Oxide‐synthesising Neurons in the Central Subnucleus of the Nucleus Tractus Solitarius Provide a Major Innervation of the Rostral Nucleus Ambiguus in the Rabbit

Abstract:We describe an intramedullary nitric oxide synthase (NOS) neural pathway that projects from the nucleus tractus solitarius (NTS) to the rostral nucleus ambiguus (NA) in the rabbit. With the use of NADPH diaphorase histochemistry and NOS immunohistochemistry, a compact group of NOS-positive perikarya was identified in the central subnucleus of the NTS dorsomedial to the tractus solitarius and rostral to the obex. A dense network of NOS terminals was seen in the rostral NA. We investigated whether NOS terminals in the NA derive from NOS perikarya in the central NTS and whether the central NOS pathway links esophageal afferents and efferents. In some rabbits, the central NTS was unilaterally lesioned. In others, Phaseolus vulgaris-leucoagglutinin (PHA-L) was injected into the central NTS, or cholera toxin-gold was injected into the NA, or cholera toxin-horseradish peroxidase (HRP) was injected into the wall of the esophagus. The medulla was subsequently processed to demonstrate PHA-L, cholera toxin-gold, HRP, and NOS reactivity. Seven days after the NTS lesion, we observed a marked decrease in the density of NOS terminals in the ipsilateral NA. After injection of PHA-L into the central NTS, a dense group of PHA-L fibres was seen in the rostral NA, principally ipsilaterally. Afferent fibres from the esophagus were found around the NOS cell bodies in the central NTS, and many of these NOS neurons were double labeled with cholera toxin-gold after injection of this tracer into the NA. NOS terminals were found around NA neurons that were retrogradely labelled from the esophagus. We conclude that the NOS neurons in the central NTS act as interneurons in a central pathway connecting esophageal afferents and efferents.
A comparative study of pre-sympathetic and Bötzinger neurons in the rostral ventrolateral medulla (RVLM) of the rat

Abstract:The aim of this study was to investigate the degree of functional and anatomical overlap between two major neuronal subpopulations in the rostral ventrolateral medulla: pre-sympathetic (sympathoexcitatory) neurons, and expiratory neurons of the Bötzinger complex. Extracellular recordings were made with dye-filled microelectrodes in pentobarbital anesthetized, paralyzed and artificially ventilated adult Wistar rats. Tests applied included stimulation of baroreceptor afferents, activation of peripheral chemoreceptors and lung stretch receptors, changes in central respiratory drive with hyper- or hypoventilation, nociceptive stimulation, and antidromic stimulation from the T2 segment of the spinal cord or medulla oblongata at obex level. The two groups of neurons showed different patterns of spontaneous activity and generally different responses to these stimuli. The recording positions showed some overlap, but the majority of Bötzinger neurons were dorsolateral to pre-sympathetic neurons. There was a large overlap between the location of pre-sympathetic neurons and the lateral part of the C1 adrenergic group, but only a small overlap between these adrenergic neurons and Bötzinger neurons. These results indicate that the anatomically adjacent pre-sympathetic and Bötzinger expiratory neurons form two functionally distinct neuronal subpopulations.
Organization of medullary adrenergic and noradrenergic projections to the periaqueductal gray matter in the rat

Abstract:The periaqueductal or midbrain central gray matter (CG) in the rat contains a dense network of adrenergic and noradrenergic fibers. We examined the origin of this innervation by using retrograde and anterograde axonal tracers combined with immunohistochemistry for the catecholamine biosynthetic enzymes tyrosine hydroxylase (TH), dopamine beta-hydroxylase (DBH), and phenylethanolamine N-methyltransferase (PNMT). Following injections of the fluorescent tracers Fast Blue or Fluorogold into the CG, double-labeled neurons in the medulla were identified mainly in the noradrenergic A1 group in the caudal ventrolateral medulla (VLM) and A2 group in the medial part of the nucleus of the solitary tract (NTS); and in the adrenergic C1 group in the rostral ventrolateral medulla and C3 group in the rostral dorsomedial medulla. Injections of Phaseolus vulgaris-leucoagglutinin (PHA-L) into these cell groups resulted in a distinct pattern of axonal labeling in various subdivisions of the CG. Anterogradely labeled fibers originating in the medial NTS were predominantly found in the lateral portion of the dorsal raphe nucleus and in the adjacent part of the lateroventral CG (CGlv). Following PHA-L injections into the C3 region the anterogradely labeled fibers were diffusely distributed in the CGlv and the dorsal raphe nucleus at caudal levels, but rostrally tended to be located laterally in the CGlv. In contrast, ascending fibers from the caudal and rostral VLM terminated in the rostral dorsal part of the CGlv and in the dorsal nucleus of the CG, whereas ventral parts of the CG, including the dorsal raphe nucleus, contained few afferent fibers. Double-label studies with antisera against DBH and PNMT confirmed that noradrenergic neurons in the A1 and A2 groups and adrenergic neurons in the C1 and C3 groups contributed to these innervation patterns in the CGlv. Noradrenergic and adrenergic projections from the medulla to the CG may play an important role in a variety of autonomic, sensory and behavioral processes.
Extrinsic Inhibitory Innervation to Rubral Neurons in Rat Brain-Stem Slices

Abstract:Synaptic connections between the neurons in the red nucleus (RN) and its extrinsic neurons were studied using rat brain-stem slices. Intracellular records were obtained from the RN neurons. Ipsilateral stimuli to areas in the dorsolateral mesencephalic reticular formation (DLMRF) or substantia nigra (SN) elicited monosynaptic hyperpolarizing postsynaptic potentials (PSPs) in about 95% of RN neurons recorded. The hyperpolarizing PSPs could be reversibly blocked by bicuculline, indicating that they were GABAAreceptor-mediated–Cl−-inhibitory PSPs. The sites of most inhibitory synapses arising from DLMRF and SN are possibly located on the proximal half of the soma–dendritic membrane of RN neurons, according to the analysis of the IPSPs with Rall’s model. In addition, tracing dyes were employed to examine the morphological pathways. After rhodamine B, a retrograde tracer, was applied to the RN in brain slices, the cell bodies of a number of neurons in DLMRF and SN were labeled. These labeled neurons were also immunopositive for glutamic acid decarboxylase (GAD) as revealed from double labeling with an anti-GAD antiserum. The anterograde tracer, tetramethylrhodamine dextran, was applied to the DLMRF or SN and taken up by many neurons in the areas. A portion of these cells extended their processes toward and terminated within the RN. Moreover, electron microscopic examination confirmed that the tetramethylrhodamine dextran-decorated synaptic terminals were present in the RN. The results indicate that the rubral neurons receive direct GABAAreceptor-mediated inhibitory inputs from neurons in the DLMRF and SN, which may participate in modulation of rubral outputs.
