Abstract:Total oxidisable precursor assay (TOP assay) can degrade and convert “unknown” per- and polyfluoroalkyl substances (PFAS) to detectable PFAS. However, the detailed degradation pathway is still not known, particularly when the TOP assay is applied to analyse complex samples such as aqueous film-forming foam (AFFF). To gain insights into the pathway and the effectiveness of the TOP assay, several “known” compounds are first tested as controls, including sodium dodecyl benzene sulphate (SDBS), perfluorooctanoic acid (PFOA), perfluorooctanesulfonic acid (PFOS). Secondly, the test is expanded to several PFAS precursors such as 6:2 fluorotelomer sulfonate (6:2 FTS), 8:2 fluorotelomer sulfonate (8:2 FTS), and a cationic surfactant N-ethyl-N-(2-hydroxyethyl) perfluorooctyl sulfonamide (EtFOSE). Thirdly, the TOP assay is used to test “unknown” PFAS samples that have been previously used as AFFF in Australia. The degradation products are monitored, to compare the mass balance and propose the degradation pathway. While HPLC-MS/MS is typically employed to detect the individual TOP assay products, most of which are perfluoroalkyl carboxylic acids (PFCA), an app-based smartphone sensor can also provide semi-quantitative results as a sum. Overall, the results indicate the effectiveness of the TOP assay to assess the presence of PFAS precursors in the AFFF samples, with some variations in the end products. Recommendations for enhancement of the TOP assay are also provided.

Abstract:Total oxidisable precursor (TOP) assay can oxidise some per- and polyfluoroalkyl substances (PFASs) and their precursors, most of which cannot be quantitatively detected so far, and convert them to detectable PFASs, such as perfluoroalkyl acids (PFAAs). In this study, the TOP assay is modified to increase the oxidation and conversion efficiency by changing the initial concentration of target sample, increasing oxidising doses, time, temperature, etc. The modified TOP assay is applied to test several aqueous film-forming foams (AFFF) and a PFAS-contaminated soil extract. The sum concentrations of the detectable PFASs are increased by up to ∼534× in the AFFF samples and ∼7× in the PFAS-contaminated soil extract. The detectable fluorotelomer sulfonate (FTS, such as 6:2/8:2 FTS) is accounted as an oxidation indicator to monitor the oxidation and conversion progress of the oxidisable PFASs precursors to the detectable PFASs. Overall, the modified TOP assay could be an appropriate method for identifying missing PFASs mass in complex matrices by detecting the PFASs precursors effectively.

Abstract:Per- and polyfluoroalkyl substances (PFAS), that are widely used in different industrial and commercial activities, have become emerging contaminants of concern. Thus, the evaluation of PFAS presence in diverse environmental matrices is of paramount importance, leading to the necessity in development of different types of analytical methods to analyse the trace amounts of PFAS with varying selectivity and sensitivity. Analytical methods that have been advanced and employed for PFAS identification are critically described with their strengths, limitations and future potentials in this review. This review includes, (i) instrument analytical methodologies, mainly focusing on sample preparation, extraction and recovery analysis, (ii) nanoparticle-based sensors, and an app-based smartphone portable sensor, for the monitoring and sensing of PFAS in different environmental matrices. Furthermore, (iii) total oxidisable precursor assay (TOPA), content of total organic fluorine (TOF) as well as total fluorine (TF) are also reviewed, which provide fundamental tools towards achieving precursor quantification and closing mass balance analysis. Abstract:Per- and polyfluoroalkyl substances (PFASs) are a class of synthetic, organic chemicals that contaminate drinking water and natural ecosystems. PFAS source apportionment is challenging because there are many sources, and standard analytical methods quantify fewer than 100 of the thousands of PFASs in commerce. The total oxidizable precursor (TOP) assay augments the number of PFASs that can be quantified and is increasingly incorporated into routine site investigation. Here we examine the ability of the TOP assay to identify PFAS sources, including aqueous film-forming foam (AFFF) impacted sites, municipal wastewater treatment plants (WWTPs), and municipal solid waste landfills in 145 samples from 46 locations and three countries. The bootstrapped mean composition of PFASs from each source was dominated by precursors, particularly in samples from WWTPs where precursors that form short-chain perfluoroalkyl carboxylates during the TOP assay were most common. Compared to when TOP assay data were excluded, inclusion of TOP assay data in dimension-reducing algorithms, such as principal component analysis (PCA), improved separation among sources. We converted the PCA tool into a web application that allows users to initiate PFAS source apportionment efforts on data from sites where contaminant sources are unknown.
Differences in the internal PFAS patterns of herbivores, omnivores and carnivores - lessons learned from target screening and the total oxidizable precursor assay

Abstract:Per- and polyfluorinated alkyl substances (PFAS) are a group of anthropogenic chemicals, which are not (fully) biodegradable and accumulate in different environmental compartments worldwide. A comprehensive, quantitative analysis - consisting of target analysis (66 different analytes, including e. g. ultrashort-chain perfluorinated carboxylic acids (PFCAs), precursor compounds and novel substitutes) and the Total Oxidisable Precursor (TOP) assay (including trifluoroacetic acid (TFA)) - were conducted to analyse the PFAS concentrations and patterns in 12 mammalian and two bird species from different areas of Germany and Denmark. The PFAS contamination was investigated in dependance of the trophic class (herbivores, omnivores, carnivores), ecological habitat (terrestrial, (semi-) aquatic) and body tissue (liver, musculature). PFAS concentrations were highest in carnivores, followed by omnivores and herbivores, with ∑PFAS concentration ranging from 1274 μg/kg (Eurasian otter liver) to 22 μg/kg (roe deer liver). TFA dominated in the herbivorous species, whereas perfluorooctanesulfonic acid (PFOS) and the long-chain PFCAs covered the majority of the PFAS contamination in carnivorous species. Besides trophic class, ecological habitat also affected the PFAS levels in the different species, with terrestrial herbivores and omnivores showing higher PFAS concentration than their aquatic counterparts, whereas for carnivores this relationship was reversed. The TOP assay analysis indicated similar trends, with the PFCA formation pattern differing significantly between the trophic classes. TFA was formed predominantly in herbivorous and omnivorous species, whereas in carnivorous species a broad spectrum of PFCAs (chain-length C2-C14) was formed. Musculature tissue of six species exhibited significantly lower PFAS concentrations than the respective liver tissue, but with similar PFAS patterns. The comprehensive approach applied in the present study showed, that primarily the trophic class is decisive for the PFAS concentration, as herbivores, omnivores and carnivores clearly differed in their PFAS concentrations and patterns. Additionally, the TOP assay gave novel insights in the PFCA formation potential in biota samples.

Abstract:<p>Here we synthesize current understanding of the magnitudes and methods for assessing human and wildlife exposures to poly‐ and perfluoroalkyl substances (PFAS). Most human exposure assessments have focused on two to five legacy PFAS and wildlife assessments are typically limited to targeted PFAS (up to ~30 substances). However, shifts in chemical production are occurring rapidly and targeted methods for detecting PFAS have not kept pace with these changes. Total fluorine (TF) measurements complemented by suspect screening using high resolution mass spectrometry are thus emerging as essential tools for PFAS exposure assessment. Such methods enable researchers to better understand contributions from precursor compounds that degrade into terminal perfluoroalkyl acids (PFAA). Available data suggest that diet is the major human exposure pathway for some PFAS but there is large variability across populations and PFAS compounds. Additional data on TF in exposure media and the fraction of unidentified organofluorine are needed. Drinking water has been established as the major exposure source in contaminated communities. As water supplies are remediated, and for the general population, exposures from dust, personal care products, indoor environments and other sources may be more important. A major challenge for exposure assessments is the lack of statistically representative population surveys. For wildlife, bioaccumulation processes differ substantially between PFAS and neutral lipophilic organic compounds, prompting a revaluation of traditional bioaccumulation metrics. There is evidence that both phospholipids and proteins are important for the tissue partitioning and accumulation of PFAS. New mechanistic models for PFAS bioaccumulation are being developed that will assist in wildlife risk evaluations.</p><p>This article is protected by copyright. All rights reserved.</p>

Abstract:According to traditional neurophysiological theory, consciousness requires neocortical functioning, and children born without cerebral hemispheres necessarily remain indefinitely in a developmental vegetative state. Four children between 5 and 17 years old are reported with congenital brain malformations involving total or near‐total absence of cerebral cortex but who, nevertheless, possessed discriminative awareness: for example, distinguishing familiar from unfamiliar people and environments, social interaction, functional vision, orienting, musical preferences, appropriate affective responses, and associative learning. These abilities may reflect‘vertical’plasticity of brainstem and diencephalic structures. The relative rarity of manifest consciousness in congenitally decorticate children could be due largely to an inherent tendency of the label‘developmental vegetative state’to become a self‐fulfilling prophecy.

Abstract:Oil refinery activity can be an emission source of perfluoroalkyl and polyfluoroalkyl substances (PFAS) to the environment, while the contamination profiles in soils remain unknown. This study investigated 44 target PFAS in soil samples collected from an oil refinery in Southeastern China, identified novel PFAS, and characterized their behaviors by assessing their changes before and after employing advanced oxidation using a combination of nontarget analysis and a total oxidizable precursor (TOP) assay. Thirty-four target PFAS were detected in soil samples. Trifluoroacetic acid (TFA) and hexafluoropropylene oxide dimer acid (HFPO-DA) were the dominant PFAS. Twenty-three novel PFAS of 14 classes were identified, including 8 precursors, 11 products, and 4 stable PFAS characterized by the TOP assay. Particularly, three per-/polyfluorinated alcohols were identified for the first time, and hexafluoroisopropanol (HFIP) quantified up to 657 ng/g dw is a novel precursor for TFA. Bistriflimide (NTf2) potentially associated with an oil refinery was also reported for the first time in the soil samples. This study highlighted the advantage of embedding the TOP assay in nontarget analysis to reveal not only the presence of unknown PFAS but also their roles in environmental processes. Overall, this approach provides an efficient way to uncover contamination profiles of PFAS especially in source-impacted areas.

Abstract:We have developed a quantitative particle size analytical method at the single atomic level employing electron microscopy and image processing for the investigation of supported metal catalysts. In the present study, a supported gold (Au) catalyst containing sub-nano clusters and individual atoms was globally observed by high-resolution high-angle annular dark-field scanning transmission electron microscopy (HAADF-STEM) using spherical aberration (Cs)-corrected TEM. To fully extract structural information of the Au clusters and individual atoms from the HAADF-STEM images, a morphological image-processing operation was applied. The resulting mean particle size was in good agreement with particle sizes estimated from average information provided by X-ray absorption fine structure analysis. It is demonstrated that the present HAADF-STEM image analysis gives a quantitative particle size distribution measurement of supported Au clusters and individual atoms.
Comprehensive target analysis and TOP assay of per- and polyfluoroalkyl substances (PFAS) in wild boar livers indicate contamination hot-spots in the environment

Abstract:The suitability of wild boar liver as a bioindicator of per- and polyfluoroalkyl substances (PFAS) in the terrestrial environment was investigated. Samples from 50 animals in three different areas associated with (1) contaminated paper sludges distributed on arable land (PS), (2) industrial emissions of PFAS (IE) and (3) background contamination (BC) were analyzed for 66 PFAS, including legacy PFAS, novel substitutes and precursors of perfluoroalkyl acids (PFAAs). Additionally, the Total Oxidizable Precursor (TOP) assay was performed to determine the formation potential of PFAAs from precursors. In total, 31 PFAS were detected with site-specific contamination profiles. PFAS concentrations in livers from area PS and IE (567 and 944 μg kg-1 wet weight, respectively) were multiple times higher than from area BC (120 μg kg-1). The dominating PFAS were the legacy compounds perfluorooctane sulfonic acid (PFOS) in areas PS and BC (426 and 82 μg kg-1, respectively) and perfluorooctanoic acid (PFOA) in area IE (650 μg kg-1). In area IE, the compounds 4,8-dioxa-3H-perfluorononanoic acid (DONA) and hexafluoropropylene oxide dimer acid (HFPO-DA) - which are used as substitutes for PFOA - were determined at 15 and 0.29 μg kg-1, respectively. The formation potential of PFAAs was highest in area PS, but generally lower than the contamination with PFAAs. The pattern of perfluoroalkyl carboxylic acids (PFCAs) in wild boar liver reflects the contamination of the local soil at the two hot-spot areas IE and PS. This first comparison of PFAS contamination between wild boars and soil suggests that wild boar livers are suitable bioindicators for PFAS contamination in the terrestrial environment. Moreover, in terrestrial samples from area IE, legacy PFAS were found to be retained for a longer period as compared to riverine samples (suspended particulate matter and chub filet).

Abstract:Per- and polyfluoroalkyl substances (PFAS) raise significant concerns due to their persistence, bioaccumulation potential, and toxicity to both ecosystems and human health. However, the long-term trends of PFAS in aquatic environments remain inadequately explored. In this study, we systematically assessed the spatiotemporal distribution, periodic fluctuations, source apportionment, and risk evaluation of 12 PFAS in the Rhine River based on the long-term measuring data collected from 2007 to 2019. The study revealed that the mean concentration and mass flux of total PFAS during this period were 32.83 ng L−1 and 6.36 × 104 μg s−1, declining at an annual rate of 3.70% and 3.82%, respectively. Wavelet analysis demonstrated that the most prominent periodic oscillation of PFAS was 40-60 months. Regarding the sources of PFAS, we employed the self-organizing map (SOM) and the positive matrix factorization (PMF) model for source apportionment. The results indicated that the primary sources of PFAS were agrochemical, pharmaceutical and textile industries, accounting for 38.1% of the total concentration. The contribution from household contamination, tannery industry, and coating materials has increased annually. In contrast, the share of electrochemical fluorination and chemical recycling has shown a continuous decline. The risk quotient (RQ) and hazard quotient (HQ) calculations for three age groups indicated that PFAS exposure did not pose a significant risk to ecological or human health. Implementing source-oriented mitigation strategies is crucial to effectively reduce the ecological and human health risks of PFAS in receiving waters.

Abstract:The age and order of ossification of the triquetral, among the bones of the hand and wrist, were determined from serial radiographs of 108 males and 103 females of the Oxford Child Health Survey. Although the median ages of ossification agreed reasonably well with the mean ages reported by other authors, the distributions suggested that the former is the more appropriate statistic. The distributions of order of ossification were distinctly bimodal for both sexes because of the tendency of the triquetral to appear along with the epiphyseal centers. The triquetral appeared before the lunate in 184 children, after it in eight, and could not be sequentially distinguished in 19. Excluding these 19, triquetral sequence variability in the remaining was achieved, in 190, through alteration of its appearance relative to the epiphyseal centers and, in the other two through an alteration of the lunate appearance. Measures of median or average order of appearance are of very little value for the triquetral.

Abstract:Abstract High per- and polyfluorinated alkyl substance (PFAS) concentrations have been detected in agricultural soils in Southwest Germany. Discharges of PFAS-contaminated paper sludge and compost are suspected to be the cause of the contamination. Perfluorinated carboxylic acids (PFCAs) have been detected also in groundwater, drinking water, and plants in this area. Recently, previously unknown compounds have been identified by high-resolution mass spectrometry (HRMS). Major contaminants were polyfluorinated dialkylated phosphate esters (diPAPs) and N-ethyl perfluorooctane sulfonamide ethanol–based phosphate diester (diSAmPAP). In this study, HRMS screening for PFAS was applied to 14 soil samples from the contaminated area and 14 impregnated paper samples which were from a similar period than the contamination. The paper samples were characterized by diPAPs (from 4:2/6:2 to 12:2/12:2), fluorotelomer mercapto alkyl phosphates (FTMAPs; 6:2/6:2 to 10:2/10:2), and diSAmPAP. In soil samples, diPAPs and their transformation products (TPs) were the major contaminants, but also FTMAPs, diSAmPAP, and their TPs occurred. The distribution patterns of the carbon chain lengths of the precursor PFAS in soil samples were shown to resemble those in paper samples. This supports the hypothesis that paper sludge is a major source of contamination. The presence of major degradation products like PFCAs, FTSAs, or PFOS and their distribution of carbon chain lengths indicate the activity of biotic or abiotic degradation processes and selective leaching processes from the upper soil horizons. Developing potency factors for thyroid hormone disruption by PFASs using TTR-TRβ CALUX® bioassay and assessment of PFASs mixtures in technical products

Abstract:Background: Over the last decade, per- and polyfluoroalkyl substances (PFASs) have become one of the most heavily investigated persistent organohalogen compound class of environmental concern. However, knowledge about their toxicology is still scarce, although PFASs as individual compounds and their industrial mixtures were shown to exert effects on the thyroid hormone system. Methods: In vitro toxicity potency factors were established for thyroid hormone transport disruption potential using the novel TTR-TRβ CALUX® bioassay for major PFASs. We assessed technical PFASs mixtures, including aqueous film-forming foam (AFFF) surfactants and chromium mist suppressants (CMS) applications with and without total oxidizable precursor (TOP) by TTR-TRβ CALUX® assay for their thyroid hormone transport disrupting potential. Results: All PFASs listed in the German guideline for drinking water (German Environment Agency, 2017) affected the T4 binding to TTR, an important plasma thyroid hormone transport protein. For all tested PFASs, potency factors based on PC80 values relative to PFOA could be obtained and ranged between PFBA (0.0018) and PFOS (2.0). Applying in vitro potency factors obtained from the present in vitro TTR-TRβ CALUX® assay study and recently reported in vivo potency factors (Zeilmaker et al., 2018; Bil et al., 2021) on the above-mentioned German guideline for PFAS in drinking water, showed that the cumulative effect-based trigger values (in vivo and in vitro) are comparable (3.0 vs. 2.9 to 4.6 μg PFOA-EQ/l). Additionally, AFFF surfactants and CMS with and without TOP assay were tested. Highest activities were found in the older AFFF surfactants (2013/2014) due to higher PFOS/PFOA levels, which were already substituted with 6:2 FTS in 2019, resulting in much lower PFOA-EQ levels. As expected also the PFOA-EQ levels increased in the samples with TOP treatment compared to the original AFFF surfactants and CMS as confirmed here by biological and chemical PFOA-equivalents (PFOA-EQ) analysis. Additionally, CMS (which have been used in the electroplating chromium industry since the 1950s) as well as PFOS-free, but not PFAS-free fume suppressants (such as Fumetrol® 21) have been tested in the TTR-TRβ CALUX® assay and showed much lower activity levels then the AFFFs, confirmed by the similar potency determination based on chemical PFASs analysis followed by transformation to PFOA-EQ for comparison. The potency factor of 6:2 FTS, which is the main substitute for PFOS in CMS, indicates that it is approximately 100-times less potent as a thyroid hormone disruptor as compared to PFOS. Conclusion: Potency factors based on PC80 values from TTR-TRβ CALUX® relative to PFOA have been developed for major PFASs. In AFFF surfactants and CMS a trend of higher activities with higher amounts of PFOS and PFOA have been found. PFOA and PFOS showed high responses in the TTR-TRβ CALUX® assay and had the largest contributions to the PFOA-EQs in the AFFF surfactants and CMS applications. Using potency factors as determined in the TTR-TRβ CALUX® to convert PFASs assessed by chemical analysis to PFOA-EQ led to comparable results as compared to the results from PFASs measured directly by the TTR-TRβ CALUX® assay. This study supports the claim that semiquantitative effect- and group-based in vitro CALUX bioanalysis tools can be applied effectively to assess industrial products containing complex mixtures with PFAS compounds for which no instrumental analysis are established, and for many compounds where in vitro toxicity data are not yet available.
