Abstract:Metal-Organic Framework (MOF) based nanozymes with clear structure are beneficial for exploration of structural-performance and exhibit broad prospects in improving activity. In this study, the prepared bimetallic Fe3Ni-MOF nanozyme was superior to single metal MOF in the peroxidase-like activity. Subsequently, a derivative nanozyme (Fe3Ni-MOF-Ar) was prepared by pyrolysis using Fe3Ni-MOF as the precursor in argon atomoshere with controlled temperature. The investigated of Fe3Ni-MOF-Ar revealed that the irregular macroporous state and the presence of heterovalent FeIII/FeII sites of Fe3Ni-MOF-Ar enable the retention, exposure, and electronic structure regulation of active sites, promoting the dual mechanism (the generation of •OH and electron transfer mechanism) and significantly increasing the peroxidase-like activity. Fe3Ni-MOF-Ar exhibited a strong affinity for substrate H2O2, which is higher than horseradish peroxidase. Ascorbic acid and cysteine are typical substances of antioxidants. Fe3Ni-MOF-Ar was used for sensitive colorimetric detection of ascorbic acid and cysteine, and the detection limit was as low as 150 and 60 nM. In addition, the smartphone devices was used to detection of antioxidant equivalent ascorbic acid, with a detection range of 0.5-120 μM. Fe3Ni-MOF-Ar nanozyme is feasible for sensitive detection of saliva total antioxidant capacity.

Abstract:A novel Fe-MoOx nanozyme, engineered with enhanced peroxidase (POD)-like activity through strategic doping and the creation of oxygen vacancies, is introduced to catalyze the oxidation of TMB with high efficiency. Furthermore, Fe-MoOx is responsive to single electron transfer (SET) and hydrogen atom transfer (HAT) mechanisms related to antioxidants and can serve as a desirable nanozyme for total antioxidant capacity (TAC) determination. The TAC colorimetric platform can reach a low LOD of 0.512 μM in solution and 24.316 μM in the smartphone-mediated RGB hydrogel (AA as the standard). As proof of concept, the practical application in real samples was explored. The work paves a promising avenue to design diverse nanozymes for visual on-site inspection of food quality.

Abstract:Accurate on-site analysis of food quality, environmental pollutants, and disease biomarkers is of great significance for safeguarding public health. In this work, based on the novel nanozymes, MnCo oxides@carbon yolk-shell nanocages (MnCo@C NCs), a portable colorimetric sensor with smartphone platform has been developed for rapid, on-site and quantitative analysis of total antioxidant capacity (TAC) and phenolic compounds. The MnCo@C NCs are synthesized via one-step calcination of polydopamine-coated MnCo Prussian blue analogs (MnCo-PBA@PDA). The PDA-derived carbon shell is found to be able to protect the nanocages from collapsing, thus increasing their specific surface areas and porosity. Benefiting from the unique structure and multivalent MnCo bimetallic oxides, the MnCo@C NCs perform outstanding catalytic performance and multiple enzyme-mimicking activities including oxidase, laccase and catalase. Hence, a multifunctional application platform integrated smartphone has been constructed for rapid and sensitive colorimetric detection of three model analytes (i.e., ascorbic acid (AA), 2,4-dichlorophenol (2,4-DP), and epinephrine) with extremely low detection limits of 0.29 μM, 0.76 μM, and 0.70 μM, respectively. This sensor device is successfully applied in TAC analysis in vegetables, fruits, and beverages, as well as epinephrine determination in human serum samples. This work provides new insights into designing multifunctional nanozymes to advance the instant detection technology in the field of food supervision, environment monitoring, and human health.

Abstract:The facile detection of total antioxidant capacity (TAC) is limited by in-situ analysis, because it usually requires complex laboratory equipments. Here, a colorimetric assay for TAC detection is developed based on the peroxidase-like activity of iron-doped carbon nitride (Fe/NC) and the smartphone platform. The peroxidase-like activity of carbon nitride is greatly improved by the introduction of Fe atoms, and the active sites turn to Fe-N x coordination groups in the Fe/NC. The inhibition mechanism of the chromogenic reaction for different kinds of antioxidants is also studied. The colorimetric assay is fabricated by the relationship of absorbance-color-antioxidant content and applied successfully to the TAC detection of several fruit juicesand commercial beverages. This work not only provides a promising approach for convenient in-situ TAC assay without the use of large instruments, but also expands the application of nanozymes in nutritional value assessment of foods.

Abstract:The level alterations of antioxidants are highly associated with various oxidative stress-related diseases, food quality and drug safety problems, which enables the reliable and accurate determination of total antioxidant capacity (TAC) become progressively significant. Herein, for the first time, we explored the catalytic effect of MnSiO 3 nanozyme with oxidase-like activity towards different fluorescent substrates, and introduced ratiometric fluorescence to MnSiO 3 -based biosensing. By harnessing MnSiO 3 -propelled catalytic oxidation of two fluorescent substrates with contrary responses (Scopoletin, SC and o-phenylenediamine, OPD), we constructed a facile, cost-effective ratiometric fluorescent (RF) platform for TAC measurement. In the absence of antioxidants, SC and OPD can be oxidized, and the synergistic effect of fluorescence resonance energy transfer (FRET) and inner-filter-effect (IFE) between SC and oxidized OPD (Ox-OPD) will result in the weak fluorescence of Ox-SC at 465 nm and the strong one of Ox-OPD at 562 nm, respectively. However, the addition of antioxidants will dissociate MnSiO 3 , generating the recovered high/low fluorescence values of both substrates. Through monitoring the FI 465 /FI 562 changes, the RF analysis of different antioxidants (Cys, AA, GSH) with satisfactory sensitivity is performed and the TAC measurements in diverse real samples (human serums, beverages, and drugs) are successfully accomplished. Moreover, the system demonstrates high selectivity and could compete with the commercial TAC test kits.

Abstract:A portable sensor is needed for the quick screening of additives in food samples, to ensure food safety. The consumption of roxarsone (ROX) contaminated meat increases arsenic toxicity, risk of cancer and neurodegenerative diseases. In this research work, we report the synthesis of thulium (III)-benzene 1,3,5-tricarboxylic acid-based metal-organic framework (Tm-BTC MOF) and its use as an excellent electrocatalyst for the determination of ROX at point-of-care. A hydrothermal procedure was developed for the synthesis of Tm-BTC MOF using a single polarity solvent. A Tm-BTC MOF modified electrode was fabricated and its electrocatalytic sensing ability towards ROX was explored. Three kinds of special chemical interactions form a special affinity between ROX and Tm-BTC MOF. In association with differential pulse voltammetry, the Tm-BTC MOF/GCE showed a low detection limit of 100 pM. The Tm-BTC MOF modified electrode was integrated into a portable sensing device via coupling it with a miniaturized potentiostat (KAUSTat) operated wirelessly by a smartphone. The developed smartphone-based sensor was successful in measuring ROX not only in lab samples but also in real (spiked) food samples. The method can be suitable for on-site testing of food quality and safety in resource-limited settings.

Abstract:Total Antioxidant Capacity (TAC) Assay plays an important role in evaluating the quality of antioxidant food and monitoring the oxidative stress level of human body. It is mainly achieved by measuring the contents of antioxidants such as AA, L-Cys and GSH, while TAC can be detected by using peroxidase-like activity of artificial nanoenzyme materials. In this work, the N-Doped, defect-rich N-MoS2NFs nano-materials were used to build the nano enzyme, which has strong stability and high peroxidase-like activity. H2O2 was detected because it can be catalyzed to generate the intermediate ·OH and make TMB appears blue. However, when H2O2, AA, L-Cys and GSH coexist in solution, due to the oxidation resistance of AA, L-Cys and GSH, they can competitively react with ·OH in solution or reduce TMB in oxidation state (oxTMB), which reduces the characteristic absorption of oxTMB, indirectly achieves the purpose of detecting AA, L-Cys and GSH, and finally realizes the determination of TAC, even in actual serum and saliva samples. At the same time, the N-MoS2 NFs/NH2-MIL-53(Al)+OPD system is further constructed. Based on the fluorescence resonance energy transfer (FRET) between NH2-MIL-53(Al) and oxidized OPD (oxOPD), the purpose of detecting TAC by fluorescence method was realized.

Abstract:Natural enzymes have excellent catalytic activity. However, due to their unstable nature and high cost, current research has turned to the synthesis and development of enzyme-like nanomaterials and single-atomic nanozymes. In this study, a single-atomic palladium-loaded nitrogen-doped porous carbon catalyst (SA-Pd/NPC) was prepared and used as a mimetic peroxidase to catalyze the substrates oxidation. The catalytic capability of the SA-Pd/NPC was tested by the TMB-H 2 O 2 system, and it expressed a superior catalytic capability owing to the plentiful catalytic centers of the single-atom Pd, its high porosity, the large specific surface area, and the strong electron transfer capability of the NPC. For the color reaction of TMB, thiol antioxidants (e.g., glutathione, GSH) and non-thiol antioxidants (e.g., ascorbic acid, AA) are suitable for different inhibition mechanisms. GSH and AA are typical substances of these two main antioxidant types, respectively. Here, we demonstrate that this prepared catalyst could be used to simultaneously determine a variety of major known physiologically relevant thiol-containing and thiol-free antioxidants, accompanied by a blue color gradient change with UV–Vis spectra at 652 nm through the SA-Pd/NPC-catalyzed TMB-H 2 O 2 system. Linear responses to GSH and AA could be obtained in the concentration ranges of 0.01–0.10 mM and 1–13 μM (both R 2 values were greater than 0.970), respectively, while the limits of detection were 3 μM and 0.3 μM, respectively. The ability of the nanozyme to detect overall antioxidant levels (TAL) was also confirmed in subsequent tests on artificial saliva and biological samples.

Abstract:In recent years, there has been a continuously growing interest in antioxidants by both customers and food industry. The beneficial health effects of antioxidants led to their widespread use in fortified functional foods, as dietary supplements and as preservatives. A variety of analytical methods are available to evaluate the total antioxidant capacity (TAC) of food extracts and beverages. However, most of them are expensive, time-consuming, and require laboratory instrumentation. Therefore, simple, cheap, and fast portable sensors for point-of-need measurement of antioxidants in food samples are needed. Here, we describe a smartphone-based chemosensor for on-site assessment of TAC of aqueous matrices, relying on the antioxidant-induced formation of gold nanoparticles. The reaction takes place in ready-to-use analytical cartridges containing an hydrogel reaction medium preloaded with Au(III) and is monitored by using the smartphone's CMOS camera. An analytical device including an LED-based lighting system was developed to ensure uniform and reproducible illumination of the analytical cartridge. The chemosensor permitted rapid TAC measurements of aqueous samples, including teas, herbal infusions, beverages, and extra virgin olive oil extracts, providing results that correlated with those of the reference methods for TAC assessment, e.g., oxygen radical absorbance capacity (ORAC).

Abstract:Analysis techniques have been done to provide the physical evidence that the electrical signature observed from a single event gate rupture (SEGR) event on a deep trench oxide capacitor from 90nm bulk complementary metal oxide semiconductor (CMOS) technology, used for the reduction of single event upsets (SEU), does identify that dielectric breakdown has occurred. SEGR damaged trench oxides were identified via a voltage contrast technique using a focused ion beam (FIB). The FIB was used to delayered and expose the deep trenches. A wet chemical etch was used to identify the location of SEGR leakage path. The oxide rupture location was observed at the top of the deep trench capacitor.

Abstract:Redox imbalance and oxidative stress are increasingly recognized as significant factors in health disorders such as neurodegenerative disorders, premature aging and cancer. However, detecting antioxidant levels that is crucial for managing oxidative stress, can be challenging due to existing assays' limitations, such as insensitivity to thiol-containing antioxidants. This study presents a simple fluorescence-based assay for antioxidant detection employing the enhanced photocatalytic oxidase-like activity of dithiothreitol (DTT)-assisted bovine serum albumin (BSA)-stabilized gold nanoclusters (DTT@BSA-AuNCs). The reported nanozyme exhibits remarkable stability, versatility, and catalytic activity. Under LED irradiation, DTT@BSA-AuNCs generate singlet oxygen, which converts non-fluorescent thiamine to fluorescent thiochrome, utilizing dissolved oxygen for catalysis. Antioxidants inhibit thiochrome formation, leading to fluorescence quenching. This method enables sensitive detection of antioxidants such as ascorbic acid and glutathione with limits of detection of 0.08 μM and 0.32 μM, respectively, under neutral pH, outperforming previous studies. The assay successfully detects antioxidants in human saliva and cancer cell models. The DTT@BSA-AuNCs-based assay offers a cost-effective, sensitive, and straightforward approach for detecting antioxidants in biological samples, facilitating improved monitoring of oxidative stress in various diseases.

Abstract:Total antioxidant capacity (TAC) is currently considered as a vital indicator of food quality in antioxidant ability and attracts much attention for human healthcare. It is thus of great significance to realize the accurate and rapid detection of TAC in foods. Herein, we have constructed a preferable hybrid nanozyme based on the mesoporous silica-stabilized CuO composited Fe3O4 nanoparticles (Fe3O4@MSNs@CuO, FMC NPs), which possess the enhanced peroxidase (POD)-like activity via cascade response for specific and sensitive determination of TAC in fruit foods. The results showed the hybrid nanozyme displayed a remarkable POD-like activity, excellent selectivity and sensitivity, and the limit of detection (LOD) of the colorimetric sensor was 6.13 mM with the concentration range from 10 to 45 mM. Therefore, the fabricated hybrid nanozyme can be regarded as an effective biosensor for the evaluation of antioxidant quality in fruit foods in future.Key points• The stabilized bimetallic nanozyme was constructed for TAC analysis in fruits.• The hybrid nanozyme possessed the enhanced POD-like activity by cascading effects.• The nanozyme was an effective biosensor for antioxidant quality evaluation in fruits.

Abstract:This study presents a potent paper-based analytical device (PAD) for quantifying various sugars using an innovative bi-nanozyme made from a 2-dimensional Fe/Ce metal-organic framework (FeCe-BTC). The MOF showed excellent bifunctional peroxidase-oxidase activities, efficiently catalyzing luminol's chemiluminescence (CL) reaction. As a peroxidase-like nanozyme, FeCe-BTC could facilitate the dissociation of hydrogen peroxide (H 2 O 2 ) into hydroxyl radicals, which then oxidize luminol. Additionally, it was also discovered that when reacting with H 2 O 2 , the MOF turns into a mixed-valence MOF, and acts as an oxidase nanozyme. This activity is caused by the generated Ce 4+ ions in the structure of MOF that can directly oxidize luminol. The MOF was directly synthesized on the PAD and cascaded with specific natural enzymes to establish simple, rapid, and selective CL sensors for the measurement of different sugars. A cell phone was also used to record light intensities, which were then correlated to the analyte concentration. The designed PAD showed a wide linear range of 0.1–10 mM for glucose, fructose, and sucrose, with detection limits of 0.03, 0.04, and 0.04 mM, respectively. It showed satisfactory results in food and biological samples with recovery values ranging from 95.8 to 102.4 %, which makes it a promising candidate for point-of-care (POC) testing for food control and medicinal purposes.

Abstract:It is well known that an excess of hydroxyl radicals (˙OH) in the human body is responsible for oxidative stress-related diseases. An understanding of the relationship between the concentration of ˙OH and those diseases could contribute to better diagnosis and prevention. Here we present a supersensitive nanosensor integrated with an electrochemical method to measure the concentration of ˙OH in vitro. The electrochemical sensor consists of a composite comprised of ultrasmall cerium oxide nanoclusters (<2 nm) grafted to a highly conductive carbon deposited on a screen-printed carbon electrode (SPCE). Cyclic voltammetry (CV) and electrochemical impedance spectroscopy (EIS) were used to analyze the interaction between cerium oxide nanoclusters and ˙OH. The CV results demonstrated that this electrochemical sensor had the capacity of detecting ˙OH with a high degree of accuracy and selectivity, achieving a consistent performance. Additionally, EIS results confirmed that our electrochemical sensor was able to differentiate ˙OH from hydrogen peroxide (H2O2), which is another common reactive oxygen species (ROS) found in the human body. The limit of detection (LOD) observed with this electrochemical sensor was of 0.6 μM. Furthermore, this nanosized cerium oxide-based electrochemical sensor successfully detected in vitro the presence of ˙OH in preosteoblast cells from newborn mouse bone tissue. The supersensitive electrochemical sensor is expected to be beneficially used in multiple applications, including medical diagnosis, fuel-cell technology, and food and cosmetic industries.

Abstract:ABSTRACT Eukaryotic initiation factor 4G (eIF4G) promotes mRNA recruitment to the ribosome by binding to the mRNA cap- and poly(A) tail-binding proteins eIF4E and Pap1p. eIF4G also binds eIF4A at a distinct HEAT domain composed of five stacks of antiparallel α-helices. The role of eIF4G in the later steps of initiation, such as scanning and AUG recognition, has not been defined. Here we show that the entire HEAT domain and flanking residues of Saccharomyces cerevisiae eIF4G2 are required for the optimal interaction with the AUG recognition factors eIF5 and eIF1. eIF1 binds simultaneously to eIF4G and eIF3c in vitro, as shown previously for the C-terminal domain of eIF5. In vivo, cooverexpression of eIF1 or eIF5 reverses the genetic suppression of an eIF4G HEAT domain Ts− mutation by eIF4A overexpression. In addition, excess eIF1 inhibits growth of a second eIF4G mutant defective in eIF4E binding, which was also reversed by cooverexpression of eIF4A. Interestingly, excess eIF1 carrying the sui1-1 mutation, known to relax the accuracy of start site selection, did not inhibit the growth of the eIF4G mutant, and sui1-1 reduced the interaction between eIF4G and eIF1 in vitro. Moreover, a HEAT domain mutation altering eIF4G moderately enhances translation from a non-AUG codon. These results strongly suggest that the binding of the eIF4G HEAT domain to eIF1 and eIF5 is important for maintaining the integrity of the scanning ribosomal preinitiation complex.

Abstract:Two issues relevant to competency-based teacher training were investigated-the specification of acceptable implementation levels for validated techniques and the necessity and feasibility of providing training on those techniques. First, a descriptive study was conducted to collect data on two direct-instruction teaching techniques-rate of presentation and signalling-that have been demonstrated to be functionally related to child performance in earlier studies. Data collected on 13 teachers, who received intensive preservice and inservice training, were then used as a standard for comparison in a multiple-baseline design across three untrained teachers to evaluate the effects of training on the two techniques. The experimental study served to determine whether training on the two techniques was necessary; and, if training was necessary, whether a training package, including supervised practice, unsupervised practice, and self-critique, would result in adequate implementation levels for the two techniques. Low implementation levels during baseline for three untrained teachers indicated that training was necessary. With training, all teachers increased their levels of appropriately signalling pupil responses and accelerated their rates of presentation well above the levels of the comparison standard. Observations made one week and again four weeks after training ended showed that performance levels achieved during training were maintained.

Abstract:Nanozymes, serving as synthetic alternatives to natural enzymes, offer several benefits including cost-effectiveness, enzyme-like catalytic abilities, enhanced stability, adjustable catalytic activity, easy recyclability, mild reaction conditions, and environmental friendliness. Nonetheless, the ongoing quest to develop nanozymes with enhanced activity and to delve into the catalytic mechanism remains a challenge. In our research, we effectively developed Au@CuO nanocomposites (Au@CuO Nc), replicating the functions of four enzymes found in nature: peroxidase (POD), catalase (CAT), glutathione peroxidase (GPx), and oxidase (OXD). The catalytic efficiency of Au@CuO Nc for TMB oxidation (oxTMB) was approximately 4.8 times greater than that of plain Cu2O cubes, attributed to the synergistic catalytic impact between the Au element and Cu2O within Au@CuO Nc. Mechanistic studies revealed that the novel Au@CuO Nc nanozyme greatly enhances the decomposition of H2O2 to reactive oxygen species (ROS) intermediates (˙OH, ˙O2- and 1O2), resulting in increased POD-like activity of the single-component Cu2O cubes. When an antioxidant like TA was added to the chromogenic system, it converted oxTMB into a colorless form of TMB, enabling further evaluation of TA. Hence, a colorimetric sensor was developed for the rapid and precise quantitative measurement of TA, demonstrating strong linearity between 0.3 and 2.4 μM and featuring a low detection threshold of 0.25 μM. Moreover, this sensor was effectively utilized for the assessment of TA in actual tea samples. This work innovatively proposes a simplified and reliable strategy for the advanced design of highly effective Cu-based nanozymes, enhancing enzyme-like reactions for simultaneous, on-site colorimetric probing of antioxidants.
Cerium dioxide-based nanostructures as signal nanolabels for current detection in the immunosensing determination of salivary myeloperoxidase

Abstract:This work reports the first electrochemical immunoplatform involving the use of synthesized cerium dioxide nanoparticles (CeO 2 NPs) / multi-walled carbon nanotubes (MWCNTs) composites as signal nanolabels for the determination of salivary myeloperoxidase (MPO), one of the proteins suggested as diagnostic biomarkers of periodontitis. The immunosensing device profits the benefits of CeO 2 NPs in terms of fast electron transfer and the catalytic activity and prevents the CeO 2 NPs tendency to agglomerate by decoration over MWCNTs, also providing high sensitivity. After a thorough characterization of the CeO 2 NPs/MWCNTs using various techniques and optimization of the variables involved in the preparation of the immunoplatform, a sandwich-type immunoassay was implemented with specific MPO capture and biotinylated detection antibodies. The use of horseradish peroxidase-streptavidin (HRP-Strept) or CeO 2 NPs/MWCNTs-(HRP-Strept) as labels for the amperometric detection at screen printed carbon electrodes (SPCEs) at −0.20 V vs. Ag pseudo-reference electrode using the H 2 O 2 /hydroquinone (HQ) system, was compared. The method developed involving CeO 2 NPs/MWCNTs-(HRP-Strept) provided a linear calibration plot for MPO over the 1.0 to 100.0 ng mL −1 (R 2 = 0.994) range with a limit of detection value of 0.40 ng mL −1 . The immunoplatform was applied to the analysis of saliva from volunteers showing that the method allowed discrimination between healthy individuals and patients diagnosed with periodontitis through the determination of MPO in 1000-times diluted saliva. The obtained results agreed with those provided by an ELISA kit, this latter requiring a much longer assay time (4 h vs 2 h).
