Abstract:The facile detection of total antioxidant capacity (TAC) is limited by in-situ analysis, because it usually requires complex laboratory equipments. Here, a colorimetric assay for TAC detection is developed based on the peroxidase-like activity of iron-doped carbon nitride (Fe/NC) and the smartphone platform. The peroxidase-like activity of carbon nitride is greatly improved by the introduction of Fe atoms, and the active sites turn to Fe-N x coordination groups in the Fe/NC. The inhibition mechanism of the chromogenic reaction for different kinds of antioxidants is also studied. The colorimetric assay is fabricated by the relationship of absorbance-color-antioxidant content and applied successfully to the TAC detection of several fruit juicesand commercial beverages. This work not only provides a promising approach for convenient in-situ TAC assay without the use of large instruments, but also expands the application of nanozymes in nutritional value assessment of foods.

Abstract:A novel Fe-MoOx nanozyme, engineered with enhanced peroxidase (POD)-like activity through strategic doping and the creation of oxygen vacancies, is introduced to catalyze the oxidation of TMB with high efficiency. Furthermore, Fe-MoOx is responsive to single electron transfer (SET) and hydrogen atom transfer (HAT) mechanisms related to antioxidants and can serve as a desirable nanozyme for total antioxidant capacity (TAC) determination. The TAC colorimetric platform can reach a low LOD of 0.512 μM in solution and 24.316 μM in the smartphone-mediated RGB hydrogel (AA as the standard). As proof of concept, the practical application in real samples was explored. The work paves a promising avenue to design diverse nanozymes for visual on-site inspection of food quality.

Abstract:A total antioxidant capacity (TAC) assay plays an essential role in evaluating antioxidant behaviors. It mainly involves examining the dosages of antioxidants such as ascorbic acid (AA), while TAC can be measured by nanozyme with peroxidase-like activity. Herein, nitrogen-doped carbon nanoflowers decorated with uniform PtNi nanoparticles (PtNi/NCFs) were fabricated by a simple pyrolysis strategy. The obtained PtNi/NCFs showed high peroxidase-mimicking activity to catalyze 3,3′,5,5′-tetramethylbenzidine (TMB) oxidation by generating rich hydroxyl radicals (·OH) in the H2O2 system. By virtue of the PtNi/NCFs nanozyme, a visual colorimetric sensor was developed for assays of H2O2 and AA. The limits of detection were as low as 45.3 and 0.94 μM for determination of H2O2 and AA with linear ranges of 100–1000 and 1.0–20.0 μM, respectively. Moreover, such sensor was also successfully employed to detect AA and H2O2 in real tablets and juices. The PtNi/NCFs nanozyme holds great potential for practical applications.

Abstract:Total Antioxidant Capacity (TAC) Assay plays an important role in evaluating the quality of antioxidant food and monitoring the oxidative stress level of human body. It is mainly achieved by measuring the contents of antioxidants such as AA, L-Cys and GSH, while TAC can be detected by using peroxidase-like activity of artificial nanoenzyme materials. In this work, the N-Doped, defect-rich N-MoS2NFs nano-materials were used to build the nano enzyme, which has strong stability and high peroxidase-like activity. H2O2 was detected because it can be catalyzed to generate the intermediate ·OH and make TMB appears blue. However, when H2O2, AA, L-Cys and GSH coexist in solution, due to the oxidation resistance of AA, L-Cys and GSH, they can competitively react with ·OH in solution or reduce TMB in oxidation state (oxTMB), which reduces the characteristic absorption of oxTMB, indirectly achieves the purpose of detecting AA, L-Cys and GSH, and finally realizes the determination of TAC, even in actual serum and saliva samples. At the same time, the N-MoS2 NFs/NH2-MIL-53(Al)+OPD system is further constructed. Based on the fluorescence resonance energy transfer (FRET) between NH2-MIL-53(Al) and oxidized OPD (oxOPD), the purpose of detecting TAC by fluorescence method was realized.

Abstract:N-doping strategy has been explored to enhance the activity of carbon nanozymes because the reconstructed electronic structures in N-doped carbon nanozymes are advantageous for the catalytic process. However, carbon nanozymes with high N content are still difficult to obtain due to the instability of N element under high calcination temperatures. To address this challenge, here we proposed a new N-doping strategy to fabricate highly active and specific peroxidase-like carbon nanozymes by using a high N-containing polymer (i.e., polyethylenimine (PEI)) as the N source and a natural clay mineral (i.e., montmorillonite (MMT)) as a template, respectively. We showed that the assembly of MMT with PEI protected N loss under high calcination temperatures and thus retained more catalytically active N sites. The mechanism study showed that the hydroxyl radical could be the key intermediate involved in the peroxidase-like catalysis. We then used the optimized carbon nanozyme with high and specific peroxidase-like activity (i.e., CP600-6) to detect H2O2, glucose, and ascorbic acid. Moreover, we successfully determined the total antioxidant capacity (TAC) in real samples including four commercial beverages, fresh orange juice, and three kinds of vitamin C tablets. The current study not only provides a new strategy for fabricating peroxidase-like nanozymes but also develops a facile TAC assay for future use in evaluation of antioxidant food quality and oxidative stress in healthcare.

Abstract:Iron-anchored nitrogen/doped carbon single-atom nanozymes (Fe-N/C), which possess homogeneous active sites and adjustable catalytic environment, represent an exemplary model for investigating the structure-function relationship and catalytic activity. However, the development of pyrolysis-free synthesis technique for Fe-N/C with adjustable enzyme-mimicking activity still presents a significant challenge. Herein, Fe-N/C anchored three carrier morphologies were created via a pyrolysis-free approach by covalent organic polymers. The peroxidase-like activity of these Fe-N/C nanozymes was regulated via the pores of the anchored carrier, resulting in varying electron transfer efficiency due to disparities in contact efficacy between substrates and catalytic sites within diverse microenvironments. Additionally, a colorimetric sensor array for identifying antioxidants was developed: (1) the Fe-N/C catalytically oxidized two substrates TMB and ABTS, respectively; (2) the development of a colorimetric sensor array utilizing oxTMB and oxABTS as sensing channels enabled accurate discrimination of antioxidants such as ascorbic acid (AsA), glutathione (GSH), cysteine (Cys), gallic acid (GA), and caffeic acid (CA). Subsequently, the sensor array underwent rigorous testing to validate its performance, including assessment of antioxidant mixtures and individual antioxidants at varying concentrations, as well as target antioxidants and interfering substances. In general, the present study offered valuable insights into the active origin and rational design of nanozyme materials, and highlighting their potential applications in food analysis.

Abstract:Natural enzymes have excellent catalytic activity. However, due to their unstable nature and high cost, current research has turned to the synthesis and development of enzyme-like nanomaterials and single-atomic nanozymes. In this study, a single-atomic palladium-loaded nitrogen-doped porous carbon catalyst (SA-Pd/NPC) was prepared and used as a mimetic peroxidase to catalyze the substrates oxidation. The catalytic capability of the SA-Pd/NPC was tested by the TMB-H 2 O 2 system, and it expressed a superior catalytic capability owing to the plentiful catalytic centers of the single-atom Pd, its high porosity, the large specific surface area, and the strong electron transfer capability of the NPC. For the color reaction of TMB, thiol antioxidants (e.g., glutathione, GSH) and non-thiol antioxidants (e.g., ascorbic acid, AA) are suitable for different inhibition mechanisms. GSH and AA are typical substances of these two main antioxidant types, respectively. Here, we demonstrate that this prepared catalyst could be used to simultaneously determine a variety of major known physiologically relevant thiol-containing and thiol-free antioxidants, accompanied by a blue color gradient change with UV–Vis spectra at 652 nm through the SA-Pd/NPC-catalyzed TMB-H 2 O 2 system. Linear responses to GSH and AA could be obtained in the concentration ranges of 0.01–0.10 mM and 1–13 μM (both R 2 values were greater than 0.970), respectively, while the limits of detection were 3 μM and 0.3 μM, respectively. The ability of the nanozyme to detect overall antioxidant levels (TAL) was also confirmed in subsequent tests on artificial saliva and biological samples.

A noble metal-enhanced Au@CuO heterostructure with multienzyme-mimicking activities for colorimetric detection of tannic acid

Abstract:Nanozymes, serving as synthetic alternatives to natural enzymes, offer several benefits including cost-effectiveness, enzyme-like catalytic abilities, enhanced stability, adjustable catalytic activity, easy recyclability, mild reaction conditions, and environmental friendliness. Nonetheless, the ongoing quest to develop nanozymes with enhanced activity and to delve into the catalytic mechanism remains a challenge. In our research, we effectively developed Au@CuO nanocomposites (Au@CuO Nc), replicating the functions of four enzymes found in nature: peroxidase (POD), catalase (CAT), glutathione peroxidase (GPx), and oxidase (OXD). The catalytic efficiency of Au@CuO Nc for TMB oxidation (oxTMB) was approximately 4.8 times greater than that of plain Cu2O cubes, attributed to the synergistic catalytic impact between the Au element and Cu2O within Au@CuO Nc. Mechanistic studies revealed that the novel Au@CuO Nc nanozyme greatly enhances the decomposition of H2O2 to reactive oxygen species (ROS) intermediates (˙OH, ˙O2- and 1O2), resulting in increased POD-like activity of the single-component Cu2O cubes. When an antioxidant like TA was added to the chromogenic system, it converted oxTMB into a colorless form of TMB, enabling further evaluation of TA. Hence, a colorimetric sensor was developed for the rapid and precise quantitative measurement of TA, demonstrating strong linearity between 0.3 and 2.4 μM and featuring a low detection threshold of 0.25 μM. Moreover, this sensor was effectively utilized for the assessment of TA in actual tea samples. This work innovatively proposes a simplified and reliable strategy for the advanced design of highly effective Cu-based nanozymes, enhancing enzyme-like reactions for simultaneous, on-site colorimetric probing of antioxidants.
Multifunctional MnCo@C yolk-shell nanozymes with smartphone platform for rapid colorimetric analysis of total antioxidant capacity and phenolic compounds

Abstract:Accurate on-site analysis of food quality, environmental pollutants, and disease biomarkers is of great significance for safeguarding public health. In this work, based on the novel nanozymes, MnCo oxides@carbon yolk-shell nanocages (MnCo@C NCs), a portable colorimetric sensor with smartphone platform has been developed for rapid, on-site and quantitative analysis of total antioxidant capacity (TAC) and phenolic compounds. The MnCo@C NCs are synthesized via one-step calcination of polydopamine-coated MnCo Prussian blue analogs (MnCo-PBA@PDA). The PDA-derived carbon shell is found to be able to protect the nanocages from collapsing, thus increasing their specific surface areas and porosity. Benefiting from the unique structure and multivalent MnCo bimetallic oxides, the MnCo@C NCs perform outstanding catalytic performance and multiple enzyme-mimicking activities including oxidase, laccase and catalase. Hence, a multifunctional application platform integrated smartphone has been constructed for rapid and sensitive colorimetric detection of three model analytes (i.e., ascorbic acid (AA), 2,4-dichlorophenol (2,4-DP), and epinephrine) with extremely low detection limits of 0.29 μM, 0.76 μM, and 0.70 μM, respectively. This sensor device is successfully applied in TAC analysis in vegetables, fruits, and beverages, as well as epinephrine determination in human serum samples. This work provides new insights into designing multifunctional nanozymes to advance the instant detection technology in the field of food supervision, environment monitoring, and human health.

Abstract:A free radical scavenging system based on FeO@Au/MOF-ABTS˙ has formed the basis of a novel method for the highly sensitive and specific spectrophotometric determination of ascorbic acid (AA). The FeO@Au/MOF nanozyme with magnetic separation properties was effectively prepared and evaluated using an environmentally friendly technique. Nanomaterials have the advantages of superparamagnetism, biocompatibility, chemical stability, and enhanced synergistic peroxidase-like activity, which can be utilized in catalysis to oxidise the peroxidase substrate 2,2'-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid) (ABTS) into a green-colored product in the presence of HO. AA as an antioxidant has scavenging effects on ABTS radicals and can reduce green ABTS˙ to uncolored ABTS, contributing to a substantial reduction in green color. Based on such a premise, a highly selective and sensitive chromogenic sensing method depending on the peroxidase-like activity of the nanocomposites was developed in order to achieve the efficient detection of AA in real samples. Under optimum conditions, the proposed technique had a detection range of 0.001-0.1 mmol L, a limit of detection of 0.098 μmol L, and a detection time of only 30 seconds. The newly proposed colorimetric analysis method devoid of enzymes has broad application potential in the areas of quality control and quality and safety detection.
Integrated gold nanorods-based dual-signal platform for accurate total antioxidant capacity assessment in food samples

Abstract:Accurate assessment of Total Antioxidant Capacity (TAC) in food is crucial for evaluating nutritional quality and potential health benefits. This study aims to enhance the sensitivity and reliability of TAC detection through a dual-signal method, combining colorimetric and photothermal signals. Gold nanorods (AuNRs) were utilized to establish a dual-signal method duo to the colorimetric and photothermal properties. Fenton reaction can etch the AuNRs from the tips, as a result, a blue shift in the longitudinal LSPR absorption peak was obtained, leading to significant changes in color and photothermal effects, facilitating discrimination through both visual observation and thermometer measurements. In the presence of antioxidants, the Fenton reaction was suppressed or inhibited, protecting the AuNRs from etching. The colorimetric and photothermal signals were therefore positively correlated with TAC levels, enabling dual-signal detection of TAC. The linear range of AA was 4-100 μM in both colorimetry and photothermal modes, with detection limits of 1.60 μM and 1.38 μM, respectively. This dual-signal approach achieves low detection limits, enhancing precision and sensitivity. The method thus has the potential to act as a promising candidate for TAC detection in food samples, contributing to improved food quality and safety assessment.
Direct and Sensitive Electrochemical Determination of Total Antioxidant Capacity in Foods Using Nanochannel-Based Enrichment of Redox Probes

Abstract:Simple and sensitive determination of total antioxidant capacity (TAC) in food samples is highly desirable. In this work, an electrochemical platform was established based on a silica nanochannel film (SNF)-modified electrode, facilitating fast and highly sensitive analysis of TAC in colored food samples. SNF was grown on low-cost and readily available tin indium oxide (ITO) electrode. Fe3+-phenanthroline complex-Fe(III)(phen)3 was applied as the probe, and underwent chemical reduction to form Fe2+-phenanthroline complex-Fe(II)(phen)3 in the presence of antioxidants. Utilizing an oxidative voltage of +1 V, chronoamperometry was employed to measure the current generated by the electrochemical oxidation of Fe(II)(phen)3, allowing for the assessment of antioxidants. As the negatively charged SNF displayed remarkable enrichment towards positively charged Fe(II)(phen)3, the sensitivity of detection can be significantly improved. When Trolox was employed as the standard antioxidant, the electrochemical sensor demonstrated a linear detection range from 0.01 μM to 1 μM and from 1 μM to 1000 μM, with a limit of detection (LOD) of 3.9 nM. The detection performance is better that that of the conventional colorimetric method with a linear de range from 1 μM to 40 μM. Owing to the anti-interfering ability of nanochannels, direct determination of TAC in colored samples including coffee, tea, and edible oils was realized.

Abstract:Total antioxidant capacity (TAC) is currently considered as a vital indicator of food quality in antioxidant ability and attracts much attention for human healthcare. It is thus of great significance to realize the accurate and rapid detection of TAC in foods. Herein, we have constructed a preferable hybrid nanozyme based on the mesoporous silica-stabilized CuO composited Fe3O4 nanoparticles (Fe3O4@MSNs@CuO, FMC NPs), which possess the enhanced peroxidase (POD)-like activity via cascade response for specific and sensitive determination of TAC in fruit foods. The results showed the hybrid nanozyme displayed a remarkable POD-like activity, excellent selectivity and sensitivity, and the limit of detection (LOD) of the colorimetric sensor was 6.13 mM with the concentration range from 10 to 45 mM. Therefore, the fabricated hybrid nanozyme can be regarded as an effective biosensor for the evaluation of antioxidant quality in fruit foods in future.Key points• The stabilized bimetallic nanozyme was constructed for TAC analysis in fruits.• The hybrid nanozyme possessed the enhanced POD-like activity by cascading effects.• The nanozyme was an effective biosensor for antioxidant quality evaluation in fruits.

Abstract:A fluorescent assay for total antioxidant capacity (TAC) detection based on the light-responsive oxidase-like activity of bovine serum albumin-stabilized gold nanoclusters (BSA-AuNCs) has been developed. Thiamine (TH) as the peroxidase substrate usually works at alkaline conditions and thus limits its practical applications. Here, by utilization the light-responsive oxidase-like activity of BSA-AuNCs, TH is oxidized to fluorescent thiochrome under neutral condition in two minutes due to the single oxygen generated by BSA-AuNCs upon light irradiation. After the introduction of antioxidants into the BSA-AuNCs-TH system, the formation of thiochrome is inhibited resulting in the fluorescence decrease. On the basis of the above facts, BSA-AuNCs-TH-based assay has been fabricated and applied successfully to detect antioxidants and the TAC of vitamin C tablets as well as some commercial fruit juice with satisfied results. This work may provide novel insights into developing light-responsive nanozymes-based fluorescent assays.

Abstract:Utilizing the peroxidase (POD) activity of nanozymes to construct the colorimetric sensors is a popular strategy for assessing the antioxidant capacity of biological molecules, but the detection sensitivity is often compromised by their low enzymatic activity. Pt-based nanozymes, favored by their regulated nanostructures, well provide numerous binding sites for H 2 O 2 , and exhibit enhanced POD performance. Here, we utilized a simple hydrothermal method to synthesize PtTeNi nanomaterials featured with the ultrathin sheets for detecting biological antioxidant (ascorbic acid (AA) as the model). With the TMB-H 2 O 2 system as the colorimetric substrate, PtTeNi nanomaterial exhibited superior POD-like activity with a higher Vmax (5.02 × 10 −4 mM s −1 ) and lower Km (3.14 mM)) in contrast to natural HRP. AA significantly inhibited the colorimetric reaction of PtTeNi-TMB-H 2 O 2 system via competitively binding OH generated by PtTeNi-mediated H 2 O 2 catalysis and further blocking oxTMB formation. Inspired by this fact, the PtTeNi-TMB-H 2 O 2 colorimetric sensor successfully realized highly sensitive and selective detection of AA with a good linear range from 5 μM to 500 μM and the detection limit of 2.94 μM. For analyzing the total antioxidant capability and AA contents in actual samples (commercial drinks and vitamin C tablets), the PtTeNi-based colormetric method displayed comparable results with the commercial total antioxidant capacity assay kit, with the advantages of easy operation, convenient storage and high stability. This work fully affirms the excellent enzyme-like performance of Pt-based nanomaterial and confirms the feasibility of nanozymes-based colorimetric sensing strategy in detecting antioxidant capability of biological molecules.

Abstract:The level alterations of antioxidants are highly associated with various oxidative stress-related diseases, food quality and drug safety problems, which enables the reliable and accurate determination of total antioxidant capacity (TAC) become progressively significant. Herein, for the first time, we explored the catalytic effect of MnSiO 3 nanozyme with oxidase-like activity towards different fluorescent substrates, and introduced ratiometric fluorescence to MnSiO 3 -based biosensing. By harnessing MnSiO 3 -propelled catalytic oxidation of two fluorescent substrates with contrary responses (Scopoletin, SC and o-phenylenediamine, OPD), we constructed a facile, cost-effective ratiometric fluorescent (RF) platform for TAC measurement. In the absence of antioxidants, SC and OPD can be oxidized, and the synergistic effect of fluorescence resonance energy transfer (FRET) and inner-filter-effect (IFE) between SC and oxidized OPD (Ox-OPD) will result in the weak fluorescence of Ox-SC at 465 nm and the strong one of Ox-OPD at 562 nm, respectively. However, the addition of antioxidants will dissociate MnSiO 3 , generating the recovered high/low fluorescence values of both substrates. Through monitoring the FI 465 /FI 562 changes, the RF analysis of different antioxidants (Cys, AA, GSH) with satisfactory sensitivity is performed and the TAC measurements in diverse real samples (human serums, beverages, and drugs) are successfully accomplished. Moreover, the system demonstrates high selectivity and could compete with the commercial TAC test kits.

Abstract:Herein, spore@Cu-trimesic acid (TMA) biocomposites were prepared by self-assembling Cu-based metal-organic framework on the surface of Bacillus velezensis spores. The laccase-like activity of spore@Cu-TMA biocomposites was enhanced by 14.9 times compared with that of pure spores due to the reaction of Cu 2+ ions with laccase on the spore surface and the microporous structure of Cu-TMA shell promoting material transport and increasing substrate accessibility. Spore@Cu-TMA rapidly oxidized and transformed 2,2′-azino-bis (3-ethylbenzthiazoline-6-sulfonic acid) (ABTS) into ABTS ●+ without using H 2 O 2 . Under optimum conditions, the ABTS ●+ could be stored for 21 days at 4 °C and 7 days at 37 °C without the addition of any stabilizers, allowing for the large-scale preparation and long-term storage of ABTS ●+ . The ultrarobust stable ABTS ●+ obtained with the use of Cu-TMA could effectively reduce the "back reaction" by preventing the leaching of the metabolites released by the spores. On the basis of these findings, a rapid, low-cost, and eco-friendly colorimetric platform was successfully developed for the detection of antioxidant capacity. Determination of antioxidant capacity for several antioxidants such as caffeic acid, glutathione, and Trolox revealed their corresponding limits of detection at 4.83, 8.89, and 7.39 nM, respectively, with linear ranges of 0.01–130, 0.01–140, and 0.01–180 μM, respectively. This study provides a facile way to prepare ultrarobust stable ABTS ●+ and presents a potential application of spore@Cu-TMA biocomposites in food detection and bioanalysis.

Abstract:The escalating oxidative stress has heightened the daily human demand for diverse antioxidants. Therefore, development of the novel approaches to assess the total antioxidant capacity (TAC) of various nutrients is essential. In this study, drawing inspiration from the active site of native peroxidase enzymes, a novel peroxidase (POD)-like nanozyme was developed based on the cobalt ferrite (CoFe2O4) nanoparticles functionalized with different catalytic amino acids. Based on the TMB/H2O2 colorimetric system, the most substantial enhancement in POD-like activity was obtained by the glutamic acid coating among different charged amino acids studied, with more than 74% increase in specific activity compared to the bare CoFe2O4. A signal-off colorimetric sensing platform based on the obtained nanobiocatalyst was developed for the accurate quantification of the antioxidant capacity of phenolic compounds and vitamin C. The sensitive and selective quantification of ascorbic acid, tannic acid, gallic acid, cyanidin-3-glucoside, and quercetin was obtained by this colorimetric method.

Abstract:In general, the total antioxidant capacity (TAC) in food is vital for evaluating the quality of antioxidant food and health monitoring. Most studies about the access of the TAC in food are mainly focused on typical colorimetric strategies. Herein, a simple and sensitive strategy for TAC in beverages and fruit has been proposed based on amino/hydroxyl-functionalized carbon dots (N2H4-CDs) and Fe3+. The fluorescence (FL) of N2H4-CDs at 650 nm can be effectively and selectively quenched via the formation of the nonfluorescence complex (Fe3+/N2H4-CDs). Moreover, the FL of N2H4-CDs is concentrated in the red region, which can eliminate the interference in complex biological environments. Interestingly, based on the theoretical calculations of density functional theory (DFT), we have confirmed that the surface amino and hydroxyl play an essential role in selective association with Fe3+, resulting in effective quenching. Moreover, because Fe3+ can be reduced by antioxidants to Fe2+, the quenched FL of N2H4-CDs can be restored in the presence of ascorbic acid (AA) and glutathione (GSH). Therefore, the typical antioxidants (AA) and especially certain thiol antioxidants (GSH) can also be analyzed by the proposed strategy. More importantly, we have proved that this strategy is successfully utilized for analyzing the TAC in beverages and fruit.
