Abstract:Results of three consecutive completed UK trials (1980-1997) for childhood lymphoblastic leukaemia are presented. National accrual has progressively increased so that over 90% of all the country's ALL cases were treated on the latest trial reported, UKALLXI. From 1980 to 1990, event-free and overall survival progressively improved, following adoption of an American therapy template and use of two post-remission intensification modules. Since 1990 despite demonstration of the benefit of a third intensification module overall event-free survival (EFS) has not improved further. Survival remains high due to a good retrieval rate especially for those relapsing off treatment after receipt of two intensification pulses. Possible reasons for the plateau in event-free survival (including type and dose of induction steroid, dropping of induction anthracycline, type and dose of asparaginase, gaps early in therapy following intensification, and overall lack of compliance in maintenance) are being explored in the latest protocol ALL '97. Cranial irradiation had been successfully replaced by a long course of intrathecal methotrexate injections for the majority of patients. Age (<1 year >10 years) sex (male) and white count >50 x 10(9)/l plus slow initial bone marrow clearance were consistently the most important independent prognostic indicators during this time period. Rome/NCI criteria accurately predict standard and high-risk groups for B cell lineage, but not consistently for T cell disease. This international collaborative venture might help us to define those truly at highest risk, and how we can optimise therapy for specific subgroups including T-ALL and those with unfavourable cytogenetics.

Abstract:Pesticides and biocides (PaB) are ubiquitously present in aquatic ecosystems due to their widespread application and have been detected in rivers at concentrations that may cause distress to aquatic life. Many of these compounds accumulate in sediments acting as long-term source for aquatic ecosystems. However, data on sediment contamination with current-use PaB in Europe are scarce. Thus, in this study, we elucidated PaB patterns and associated risks in sediments of seven major European rivers focusing on their last stretch as an integrative sink of particles transported by these rivers. Sediments were extracted with pressurized liquid extraction (PLE) using a broad-spectrum method recovering many compound classes with a wide range of physicochemical properties. Altogether 126 compounds were analyzed and 81 of them were detected with LC-HRMS and GC-NCI-MS/MS at least in one of the sediments. The highest number of compounds was detected (59) in River Elbe sediments close to Cuxhaven with outstanding concentrations ranging from 0.8 to 1691 mg/g organic carbon. Multivariate analysis identified a cluster with 3 ubiquitous compounds (cyhalothrin, carbendazim, fenpropimorph) and three clusters of chemicals with higher variability within and between rivers. Risk assessment indicates an acute toxic risk to benthic crustaceans at all investigated sites with the pyrethroids tefluthrin and cyfluthrin together with the fungicide carbendazim as the main drivers. Risks to algae were driven at most sites almost exclusively by photosynthesis inhibitors with estuary-specific herbicide mixtures, while in the rivers Po and Gironde cell division inhibitors played an important role at some sites. Mixtures of specific concern have been defined and suggested for integration in future monitoring programs.

Abstract:The investigation of pollutant inputs via stormwater runoff and subsequent effects in receiving waters is becoming increasingly urgent in view of climate change with accompanying extreme weather situations such as heavy rainfall events. In this study, two sampling areas, one urban and one rural but dominated by a highway, were investigated using effect-directed analysis to identify endocrine and neurotoxic effects and potentially responsible substances in stormwater structures and receiving waters. For this purpose, a transgenic yeast cell assay for the simultaneous detection of estrogenic, androgenic, and progestogenic effects (YMEES) was performed directly on high-performance thin-layer chromatography (HPTLC) plates. Concomitantly, estrogens were analyzed by GC-MS/MS and other micropollutants typical for wastewater and stormwater by LC-MS/MS. Discharges from the combined sewer overflow (CSO) contribute a large portion of the endocrine load to the studied water body, even surpassing the load from a nearby wastewater treatment plant (WWTP). An effect pattern similar to the CSO sample was shown in the receiving water after the CSO with lower intensities, consisting of an estrogenic, androgenic, and progestogenic effect. In contrast, after the WWTP, only one estrogenic effect with a lower intensity was detected. Concentrations of E1, 17α-E2, 17β-E2, EE2, and E3 in the CSO sample were 2000, 410, 1100, 560, and 2700 pg/L, respectively. HPTLC-YMEES and GC-MS/MS complement each other very well and help to elucidate endocrine stresses. An Acetylcholinesterase (AChE) inhibitory effect could not be assigned to a causative compound by suspect and non-target analysis using LC-HRMS. However, the workflow showed how information from HPTLC separation, effect-based methods, and other meta-information on the sampling area and substance properties can contribute to an identification of effect-responsible substances. Overall, the study demonstrated that effect-based methods in combination with HPTLC and instrumental analysis can be implemented to investigate pollution by stormwater run-off particularly regarding heavy rain events due to climate change.

Abstract:Wearing of vehicle parts could release many chemical additives into the environment, such as benzotriazoles (BTRs), benzothiazoles (BTHs), and p-phenylenediamines (PPDs), which are potentially toxic to wildlife and humans. This study investigated the occurrence, source, and risks of BTRs, BTHs, and PPDs in a source catchment providing water to Guangzhou, a megacity in South China, covering groundwater, surface water, and stormwater. The results showed that BTRs and BTHs were predominant in surface water and groundwater. Unexpectedly, the BTR and BTH concentrations were lower in surface water than groundwater in a third of the paired samples. For the first time, 6PPD-quinone, a toxic ozonation product of N-(1,3-dimethylbutyl)-N'-phenyl-1,4-phenylenediamine (6PPD), was extensively detected in source waters. Stormwater decreased the BTR concentrations but increased the 6PPD-quinone concentrations in surface water owing to their affiliation to suspended particles. From natural to urban segments of Liuxi river, a downstream increasing trend in BTR and BTH concentrations was observed, confirming that they are indicative of urban anthropogenic activities. Strong correlations between industrial activities and BTR or BTH concentrations in surface water indicated that industrial activities were their main sources. Six compounds were prioritized as potentially persistent, mobile, and toxic (PMT) chemicals, combing our monitoring results and REACH criterion. This study improves our understanding of the environmental fates and risks of water-soluble tire-wear chemicals, which provides important information for chemical management, and indicates attention should be paid to the risk posed by 6PPD-quinone in the source water.

Assessment of stormwater discharge contamination and toxicity for a cold-climate urban landscape

Abstract:Background Stormwater is water resulting from precipitation events and snowmelt running off the urban landscape, collecting in storm sewers, and typically being released into receiving water bodies through outfalls with minimal to no treatment. Despite a growing body of evidence observing its deleterious pollution impacts, stormwater management and treatment in cold climates remains limited, partly due to a lack of quality and loading data and modeling parameters. This study examines the quality of stormwater discharging during the summer season in a cold-climate, semi-arid Canadian city (Saskatoon, Saskatchewan). Results Seven stormwater outfalls with mixed-land-use urban catchments > 100 km 2 were sampled for four summer (June–August 2019) storm events and analyzed for a suite of quality parameters, including total suspended solids (TSS), chemical oxygen demand (COD), dissolved organic carbon (DOC), metals, and targeted polyaromatic hydrocarbons (PAHs). In addition, assessment of stormwater toxicity was done using the two toxicity assays Raphidocelis subcapitata (algae) and Vibrio fischeri (bacteria). Notable single-event, single-outfall contaminant pulses included of arsenic (420 μg/L), cadmium (16.4 μg/L), zinc (924 μg/L), fluorene (4.95 μg/L), benzo[a]pyrene (0.949 μg/L), pyrene (0.934 μg/L), phenanthrene (1.39 μg/L), and anthracene (1.40 μg/L). The IC 50 in both R. subcapitata and V. fischeri was observed, if at all, above expected toxicity thresholds for individual contaminant species. Principal component analysis (PCA) showed no clear trends for individual sampling sites or sampling dates. In contrast, parameters were correlated with each other in groups including DOC, COD, TSS, and reduced algal toxicity; and total dissolved solids (TDS), sum of metals, and pH. Conclusions In general, stormwater characteristics were similar to those of previous studies, with a bulk of contamination carried by the first volume of runoff, influenced by a combination of rainfall depth, antecedent dry period, land use, and activity within the catchment. Roads, highways, and industrial areas contribute the bulk of estimated contaminant loadings. More intensive sampling strategies are necessary to contextualize stormwater data in the context of contaminant and runoff volume peaks.

Abstract:Aliphatic linear polyamines, from diamines to hexaamines, tertiary branched tetraamines, and quaternary branched pentaamines are widely distributed in eubacteria, archaebacteria, and cyanobacteria. Twenty-four linear types and four branched types are acid extractable from bacterial cells and can be chromatographically analyzed and identified. The varieties of polyamines are due to the combination of amino acid decarboxylase activities to form diamines, aminopropyl- and aminobutyl-transfer activities mediated by aminopropyltransferases or Schiff-base complex formation, and hydroxylation activity. The absence or presence of spermidine, norspermidine or homospermidine and the occurrence of 2-hydroxyputrescine and diaminopropane are related to grouping into the alpha, beta, gamma, and delta subclasses within Proteobacteria. Flavobacterium complex and green bacteria contain putrescine and homospermidine. Gram-negative thermophiles contain long linear and branched polyamines; however, their distribution profiles are species specific. Gram-positive eubacteria, which comprise Bacillus cluster, anaerobes, and actinomycetes, ubiquitously contain putrescine and spermidine, while the occurrence of spermine is limited to thermophiles. Archaebacteria are separated into polyamine-absent methanogens and halophiles, homospermidine-dominant methanogens, spermidine-dominant methanogens, and spermidine- and norspermidine-containing thermophiles. Cyanobacteria comprise two types; one group contains homospermidine and the other spermidine. The polyamine distribution pattern can serve as a chemotaxonomic marker in bacterial classification and is associated with bacterial systematics on the level of order, family, or genus.
Analysis of emerging organic contaminants in water, fish and suspended particulate matter (SPM) in the Joint Danube Survey using solid-phase extraction followed by UHPLC-MS-MS and GC-MS analysis

Abstract:In the third Joint Danube Survey (JDS3), emerging organic contaminants were analysed in the dissolved water phase of samples from the Danube River and its major tributaries. Analyses were performed using solid-phase extraction (SPE) followed by ultra-high-pressure liquid chromatography triple-quadrupole mass spectrometry (UHPLC-MS-MS) and gas chromatography-mass spectrometry (GC-MS). The polar organic compounds analysed by UHPLC-MS-MS were 1H-benzotriazole, methylbenzotriazoles, carbamazepine, 10,11-dihydro-10,11-dihydroxy-carbamazepine, diclofenac, sulfamethox-azole, 2,4-D (2,4-dichlorophenoxyacetic acid), MCPA (2-methyl-4-chlorophenoxyacetic acid), metolachlor, cybutryne (irgarol), terbutryn, DEET (N,N-diethyl-m-toluamide), and several perfluoroalkyl acids (C6-C9; C8=perfluorooctanoic acid (PFOA)) and perfluorooctansulfonic acid (PFOS). In addition, several organophosphorus flame retardants were analysed by GC-MS. The most relevant compounds identified in the 71 water samples, in terms of highest median and maximum concentrations, were 1H-benzotriazole, tris(1-chloro-2-propyl)phosphate (TCPP), methylbenzotriazoles, carbama-zepine and its metabolite, DEET, sulfamethoxazole, tris(isobutyl)phosphate (TiBP), tris(2-chloroethyl)phosphate (TCEP), PFOA, PFOS and diclofenac. The concentrations of these compounds in the samples were generally below the environmental quality standard (EQS) threshold values, with the exception of PFOS, the concentration of which exceeded the annual average water EQS limit of 0.65ng/L along the whole river, and also exceeded the fish biota EQS of 9.1μg/kg. In addition, the proposed EQS for diclofenac, of 0.1μg/L, was exceeded in the Arges River in Romania (255ng/L).
Non-targeted screening and photolysis transformation of tire-related compounds in roadway runoff

Abstract:Roadway runoff serves as a crucial pathway for transporting contaminants of emerging concern (CECs) from urban environments to receiving water bodies. Tire-related compounds originating from tire wear particles (TWPs) have been frequently detected, posing a potential ecological threat. Yet, the photolysis of tire-related compounds within roadway runoff remains inadequately acknowledged. Addressing this deficit, our study utilized high-resolution mass spectrometry (HRMS) to characterize the chemical profile of roadway runoff across eight strategically selected sites in Guangzhou, China. 219 chemicals were identified or detected within different confidence levels. Among them, 29 tire-related contaminants were validated with reference standards, including hexa(methoxymethyl)melamine (HMMM), 1,3-diphenylguanidine (DPG), dicyclohexylurea (DCU), and N-cyclohexyl-2-benzothiazol-amine (DCMA). HMMM exhibited with the abundance ranging from 2.30 × 104-3.10 × 106, followed by DPG, 1.69 × 104-8.34 × 106. Runoff sample were exposed to irradiation of 500 W mercury lamp for photodegradation experiment. Photolysis results indicated that tire-related compounds with a low photolysis rate, notably DCU, DCMA, and DPG, are more likely to persist within the runoff. The photolytic rates were significantly correlated with the spatial distribution patterns of these contaminants. Our findings underscore TWPs as a significant source of pollution in water bodies, emphasizing the need for enhanced environmental monitoring and assessment strategies.
Identification of Microplastic Pathways within a typical European Urban Wastewater System

Abstract:In recent years, ThermoExtraction/Desorption‐Gas Chromatography/Mass Spectrometry (TED‐GC/MS) has been developed as a rapid detection method for the determination of microplastics (MP) mass contents in numerous environmentally relevant matrices and, in particular, for the measurement of polymers in water samples without time‐consuming sample preparation. The TED‐GC/MS method was applied to investigate a typical European municipal wastewater system for possible MP masses. Such investigations are important in view of the recent revision of the Urban Wastewater Treatment Directive. Four different representative sampling sites were selected: greywater (domestic wastewater without toilet), combined sewer, and influent and effluent of a wastewater treatment plant (WWTP). All samples were collected by fractional filtration. Filtration was carried out over mesh sizes of 500, 100, 50, and in some cases 5 μm. Polyethylene (PE), polypropylene (PP) and polystyrene (PS) were detected in all samples, with the PE fraction dominating in all cases. Styrene‐butadiene rubber which serves as an indication of tire abrasion was only found in the influent of the WWTP. The highest MP mass contents were found in the combined sewer, so MP can become a source of pollution during heavy rain events when the capacity limits of the effluent are reached, and the polluted effluent is released uncontrolled into the environment. Based on the studies, MP retention from the WWTP could be estimated to be approximately 96 %. Little trends in polymer type or mass contents were detected within the different fractions of the samples or when comparing samples to each other. This article is protected by copyright. Abstract:The presence of persistent and mobile organic contaminants (PMOC) in aquatic environments is a matter of high concern due to their capability of crossing through natural and anthropogenic barriers, even reaching drinking water. Most analytical methods rely on reversed-phase liquid chromatography (RPLC), which is quite limited for the detection of very polar chemicals. Thus, many of these PMOCs may have not been recognized as water pollutants yet, due to the lack of analytical methods capable to detect them. Mixed-mode LC (MMLC), providing the combination of RP and ion-exchange functionalities is explored in this work with a trifunctional column, combining RPLC, anion and cation exchange, which allows the simultaneous determination of analytes with extremely different properties. A nondiscriminant sample concentration step followed by a MMLC-high resolution mass spectrometry method was developed for a group of 37 very polar model chemicals with different acid/base functionalities. The overall method performance was satisfactory with a mean limit of detection of 50 ng/L, relative standard deviation lower than 20% and overall recoveries (including matrix effects) higher than 60% for 54% of model compounds. Then, the method was applied to 15 real water samples, by a suspect screening approach. For those detected PMOC with standard available, a preliminary estimation of concentrations was also performed. Thus, 22 compounds were unequivocally identified in a range of expected concentrations from 6 ng/L to 540 μg/L. Some of them are well-known PMOC, such as acesulfame, perfluorobutanoic acid or metformin, but other novel pollutants were also identified, as for example di-o-tolylguanidine or trifluoromethanesulfonic acid, which had not or were scarcely studied in water so far.

Abstract:Benzotriazoles (BTR) and bisphenols (BP) are artificial contaminants of emerging concern (CECs) commonly found in everyday products. This study focuses on urban runoff to investigate the occurrence of BTRs and BPs in Iceland, regarded as one of the least polluted places in the world, which made it reasonable to confirm or deny the presence of these micropollutants in its environment. Samples collected in February 2023 (SC1) and August 2024 (SC2) from seven locations along Iceland's Ring Road were evaluated to determine the occurrence of seven BTRs (1H-BTR, 4Me-BTR, 5Me-BTR, 5Cl-BTR, UV-P, UV-326, UV-329) and six BPs (BPF, BPE, BPA, BPZ, BPAP, BPM) in the runoff, using the ultrasound-assisted emulsification-microextraction for analytes isolation and gas chromatography-mass spectrometry for detection (USAEME-GC/MS). All locations showed detectable and varying levels of BTRs and BPs, with 5Cl-BTR (11.6 μg/L) and BPF (56.3 μg/L), both in SC1, demonstrating the highest concentrations, providing valuable insights into their prevalence and distribution. A correlational analysis investigated the connection between these pollutants and various characteristics associated with the locations along the Ring Road. This study contributes to the essential comprehension of these CECs, serving as input for future strategies for monitoring and mitigating their impact.
Quantification and risk assessment of polar organic contaminants in two chalk streams in Hampshire, UK using the Chemcatcher passive sampler

Abstract:Freshwater systems are facing a number of pressures due to the inputs of polar organic contaminants from a range of sources including agriculture, domestic and industry. The River Itchen and River Test are two sensitive chalk streams in Southern England that are experiencing a decline in invertebrate communities. We used Chemcatcher passive samplers to measure time-weighted average concentrations (14 days) of polar pollutants at nine sites on the River Itchen and eight sites on the River Test over a 12-month period. Sampler extracts were analysed using a targeted LC/MS method. In total, 121 plant protection products and pharmaceutical and personal care products were quantified (range of log Kow from - 1.5 to 7). Concentrations (sub ng L-1 to >500 ng L-1) in both rivers showed spatial and temporal variations. A greater number of compounds and higher concentrations were found in the River Test. The chemical profile was dominated by inputs from wastewater treatment plants and legacy plant protection products. On the River Itchen, high concentrations (∼100 ng L-1) of caffeine were observed directly downstream of a fish farm. Using the NORMAN database, the predicted no effect concentration (PNEC) freshwater values were exceeded by only five contaminants (2-hydroxy-terbuthylazine, alprazolam, azithromycin, diclofenac and imidacloprid). In addition, venlafaxine was detected above its EU Watch List concentration. These exceedances were mainly downstream of direct inputs from treatment plants. These compounds are known to have ecotoxicological effects on a range of aquatic biota including macroinvertebrates. Of concern is the ubiquitous presence of the ectoparasiticide imidacloprid, highlighting the need to control its use. The impact of the cocktail of pollutants found in this study on the long-term effects on chalk stream ecosystems remains unknown and needs further investigation.

Heterocyclic organic compounds BTRs are an emerging threat, under-recognized and under-analyzed in most environmental and water legislation. They are hazardous, potentially mutagenic, and carcinogenic micropollutants, not susceptible to effective biodegradation, and they move easily through the trophic chain, contaminating the environment and water resources. Traffic activities are a common source of BTR emissions in the urban environment, directly polluting human habitats through the different routes and numerous vehicles circulating in the cities. Using twelve heterogeneous locations scattered over a metropolitan area in Poland as a case study, this research analyzed the presence of BTRs in water samples from runoff produced from rainwater and snowmelt. 1H-BTR, 4Me-BTR, 5Me-BTR and 5Cl-BTR were detected in the tested runoff water. 5Cl-BTR was present in all samples and in the highest concentrations reaching 47,000 ng/L. Risk quotients calculated on the basis of the determined concentrations indicate that the highest environmental risk is associated with the presence of 5Cl-BTR and the sum of 4Me-BTR and 5Me-BTR, and the most sensitive organisms are bacteria and invertebrates. The results indicate that it is possible to associate the occurrence of these contaminants with the type of cover, traffic intensity, and vehicle type.

Abstract:The Haihe River System (HRS) drains the Chinese megacities Beijing and Tianjin, forming a large-scale irrigation system severely impacted by wastewater-borne pollution. The origin, temporal magnitudes, and annual mass fluxes of a wide range of pharmaceuticals, household chemicals, and pesticides were investigated in the HRS, which drains 70% of the wastewater discharged by 20 million people living in Beijing. Based on Chinese consumption statistics and our initial screening for 268 micropollutants using high-resolution mass spectrometry, 62 compounds were examined in space and time (2009-2010). The median concentrations ranged from 3 ng/L for metolachlor to 1100 ng/L for benzotriazole and sucralose. Concentrations of carbendazim, clarithromycin, diclofenac, and diuron exceed levels of ecotoxicological concern. Mass-flux analyses revealed that pharmaceuticals (5930 kg/year) and most household chemicals (5660 kg/year) originated from urban wastewaters, while the corrosion inhibitor benzotriazole entered the rivers through other pathways. Total pesticide residues amounted to 1550 kg/year. Per capita loads of pharmaceuticals in wastewater were lower than those in Europe, but are expected to increase in the near future. As 95% of the river water is diverted to irrigate agricultural soil, the loads of polar organic micropollutants transported with the water might pose a serious threat to food safety and groundwater quality.

Abstract:The presence of tyre and road wear particles (TRWP) in the environment is an underestimated threat due to their potential impact on ecosystems and human health. However, their mode of action and potential impacts on aquatic ecosystems remain largely unknown. In the present study, we adopted a sediment exposure scenario to investigate the influence of sediment coming from an urban runoff sedimentation basin on the life cycle of Chironomus riparius. Targeted broad-spectrum chemical analysis helped to characterise the urban sediments and confirmed the significant contribution of contaminants from traffic (e.g. tyre wear contribution, Polycyclic Aromatic Hydrocarbons [PAHs], metals, tyre rubber additives). First-stage chironomid larvae were subjected to increasing concentrations of urban whole sediment. The results showed that exposure to this urban sediment influenced all measured endpoints. In vivo quantification of ROS showed that larvae exposed to the lowest concentration of contaminated sediment exhibited increased fluorescence. The contaminated sediment conditions increased mortality by almost 30 %, but this effect was surprisingly not concentration-dependent. Fertility decreased significantly and concentration-dependently. The results of the Mean Emergence Time (EmT50) and larval size showed an optimality curve. Furthermore, as a consequence of the effects on fitness, the Population Growth Rate (PGR) exhibited a significant decrease, which was concentration-dependent. Therefore, after a single generation, PGR calculation can be adopted as a sensitive tool to monitor pollution caused by complex matrices, i.e. composed of several contaminants. Our research highlights the importance of effective management of road runoff and underlines the need for further investigation to better understand the toxicity of TRWPs.
