Abstract:Phthalate esters (PAEs) are a class of the emerging pollutants that pose a potential environmental threat to marine ecosystems. In this study, a simple analytical method using ultrasound-assisted extraction combined with gas purges microsyringe extraction (GP-MSE) coupled with GC-MS was utilized for the reliable and rapid determination of PAEs in different types of marine sediment. The analytical results showed that the method exhibited excellent reproducibility, linear responses, and detection limits, which verified the suitability of the method for the determination of PAEs in marine sediment. This approach requires minimal reagents, solvents, and sample pretreatment procedures as well as a short analysis time; thus, procedural blanks can be kept to a minimum. This method was demonstrated to be a highly efficient and sensitive quantitative analytical method for the simple detection of PAEs in marine sediment.

Abstract:Aliphatic linear polyamines, from diamines to hexaamines, tertiary branched tetraamines, and quaternary branched pentaamines are widely distributed in eubacteria, archaebacteria, and cyanobacteria. Twenty-four linear types and four branched types are acid extractable from bacterial cells and can be chromatographically analyzed and identified. The varieties of polyamines are due to the combination of amino acid decarboxylase activities to form diamines, aminopropyl- and aminobutyl-transfer activities mediated by aminopropyltransferases or Schiff-base complex formation, and hydroxylation activity. The absence or presence of spermidine, norspermidine or homospermidine and the occurrence of 2-hydroxyputrescine and diaminopropane are related to grouping into the alpha, beta, gamma, and delta subclasses within Proteobacteria. Flavobacterium complex and green bacteria contain putrescine and homospermidine. Gram-negative thermophiles contain long linear and branched polyamines; however, their distribution profiles are species specific. Gram-positive eubacteria, which comprise Bacillus cluster, anaerobes, and actinomycetes, ubiquitously contain putrescine and spermidine, while the occurrence of spermine is limited to thermophiles. Archaebacteria are separated into polyamine-absent methanogens and halophiles, homospermidine-dominant methanogens, spermidine-dominant methanogens, and spermidine- and norspermidine-containing thermophiles. Cyanobacteria comprise two types; one group contains homospermidine and the other spermidine. The polyamine distribution pattern can serve as a chemotaxonomic marker in bacterial classification and is associated with bacterial systematics on the level of order, family, or genus.

Abstract:The present work provides the first data on the occurrence of different classes of pharmaceuticals and personal care products (PPCPs) in surface marine sediments from an Arctic fjord (Kongsfjorden, Svalbard Islands, Norway). The target compounds included: ciprofloxacin; enrofloxacin; amoxicillin; erythromycin; sulfamethoxazole; carbamazepine; diclofenac; ibuprofen; acetylsalicylic acid; paracetamol; caffeine; triclosan; N , N -diethyl-meta-toluamide; 17β-estradiol; 17α-ethinyl estradiol and estrone. Sampling was performed in the late summer, when high sedimentation rates occur, and over 5 years (2018–2022). Based on the environmental concentrations (MECs) found of emerging contaminants and the relative predicted no-effect concentrations (PNECs), an environmental risk assessment (ERA) for sediments was performed, including the estimation of the Risk Quotients (RQs) of selection and propagation of antimicrobial resistance (AMR) in this Arctic marine ecosystem. Sediments were extracted by Pressurized Liquid Extraction (PLE) and the extracts were purified by Solid Phase Extraction (SPE). Analytical determination was conducted with liquid chromatography-high-resolution mass spectrometry (HPLC-HRMS). PPCPs were detected in the sediments along the fjord in all the years investigated, with overall concentrations similar in most cases to those reported in urbanized areas of the planet and ranging from a minimum of 6.85 ng/g for triclosan to a maximum of 684.5 ng/g for ciprofloxacin. This latter was the only antibiotic detected but was the most abundant compound (32 %) followed by antipyretics (16 %), hormones (14 %), anti-inflammatories (13 %), insect repellents (11 %), stimulants (9 %), and disinfectants (5 %). Highest concentrations of all PPCPs detected were found close to the Ny-Ålesund research village, where human activities and the lack of appropriate wastewater treatment technologies were recognized as primary causes of local contamination. Finally, due to the presence in the sediments of the PPCPs investigated, the ERA highlights a medium (0.1 < RQ 1) for organisms living in this Arctic marine ecosystem, including high risk of the spread of AMR.

Novel polar AhR-active chemicals detected in sediments of an industrial area using effect-directed analysis based on in vitro bioassays with full-scan high resolution mass spectrometric screening

Abstract:Studies investigating aryl hydrocarbon receptor (AhR)-active compounds in the environment typically focus on non- and mid-polar substances, such as PAHs; while, information on polar AhR agonists remains limited. Here, we identified polar AhR agonists in sediments collected from the inland creeks of an industrialized area (Lake Sihwa, Korea) using effect-directed analysis combined with full-scan screening analysis (FSA; using LC-QTOFMS). Strong AhR-mediated potencies were observed for the polar and latter fractions of RP-HPLC (F3.5-F3.8) from sediment organic extracts in the H4IIE-luc in vitro bioassays. FSA was performed on the corresponding fractions. Twenty-eight tentative AhR agonists were chosen using a five-step process. Toxicological confirmation using bioassay revealed that canrenone, rutaecarpine, ciprofloxacin, mepanipyrim, genistein, protopine, hydrocortisone, and medroxyprogesterone were significantly active. The relative potencies of these AhR-active compounds compared to that of benzo[a]pyrene ranged from 0.00002 to 2.0. Potency balance analysis showed that polar AhR agonists explained, on average, ~6% of total AhR-mediated potencies in samples. Some novel polar AhR agonists also exhibited endocrine-disrupting potentials capable of binding to estrogen and glucocorticoid receptors, as identified by QSAR modeling. In conclusion, the focused studies on distributions, sources, fate, and ecotoxicological effects of novel polar AhR agonists in the environment are necessary.

Abstract:The Ross Sea, among the least human-impacted marine environments worldwide, recently became the first marine protected area in Antarctica. To assess the impact of the Italian research station Mario Zucchelli (MZS) on the surrounding waters, passive sampling - as well as spot sampling for comparison - took place in the effluent of the wastewater treatment plant (WWTP) and the receiving surface marine waters. Polar Organic Chemical Integrative Samplers (POCIS) were deployed for six consecutive 2-week periods from November to February in a reservoir collecting the wastewater effluent. Passive samplers were also deployed at shallow depth offshore from the wastewater effluent outlet from MZS for two separate 3-week periods (November 2021 and January 2022). Grab water samples were collected alongside each POCIS deployment, for comparison with passive sampling results. POCIS, used for the first time in Antarctica, demonstrated to be advantageous to estimate time-averaged concentrations in waters and the results were comparable to those obtained by repeated spot samplings. Among the 23 studied ECs - including drugs, UV-filters, perfluorinated substances, caffeine - 15 were detected in both grab and passive sampling in the WWTP effluent and followed similar concentration profiles in both types of sampling. High concentrations of caffeine, naproxen and ketoprofen in the dozens of μg L-1 were detected. Other compounds, including drugs and several UV filters, were detected down to sub- μg L-1 concentrations. In marine waters close to the effluent output, only traces of a drug (4.8 ng L-1) and two UV filters (up to 0.04 μg L-1) were quantified.

[Nontarget Screening and Ecological Risk Assessment of Polar Organic Pollutants in Surface Water on the West Bank of Taihu Lake].

Abstract:Emerging pollutants occur in the environment, which has become a pressing issue for environmental research. In order to comprehensively screen potential polar organic pollutants in surface water of Wujin and Yixing in the Taihu Lake Basin, nontarget screening was carried out by high performance liquid chromatography(HPLC) and time of flight mass spectrometry(TOF-MS). Screened by accurate mass, isotope distribution, and MS/MS information, 162 organic compounds were identified, including 46 pesticides, 34 drugs, 8 personal care products, and 27 additives; 17 organic synthetic intermediates and 30 metabolites, 45 of which have been verified by reference standards. Through the quantitative analysis of 42 pollutants and ecological risk assessment of 3 trophic model species, it was found that 25 pollutants posed medium risk while 12 pollutants presented high risk. Nontarget screening can be used to identify potential pollutants with no prior information or standards. It is not only fast, accurate, and has high analytical flux, but also provides an important basis for subsequent ecological risk assessment.

Abstract:Gamcheon Harbor in Busan, the largest port city in South Korea, is contaminated with persistent toxic substances, including polycyclic aromatic hydrocarbons (92 to 1700 ng g −1 dry mass (dm)) and styrene oligomers (17 to 520 ng g −1 dm). This study applied effect-directed analysis and nontarget screening (NTS) to identify aryl hydrocarbon receptor (AhR)-active substances in Gamcheon harbor sediments. Relatively great AhR-mediated potencies were found in RP-HPLC fractions, F2.7–F2.8 (mid-polar, log K OW 6–8) and F3.6–F3.7 (polar, log K OW 5–7). Target AhR agonists comprised up to 43% of total AhR-mediated potencies. NTS using GC-QTOFMS and LC-QTOFMS identified daphnoretin and isorhamnetin as significant AhR agonists, with relative potency values of 0.4 × 10 −3 and 6.5 × 10 −5 , respectively, compared to benzo[ a ]pyrene. The major AhR agonists in the coastal sediments of Korea appeared to be region-specific. This approach is useful for identifying and managing key toxic substances in coastal ecosystems.

Abstract:Despite being one of the most remote areas on the planet, the Antarctic continent is subject to anthropogenic influences. The presence of various groups of contaminants, including persistent organic pollutants (POPs) and polycyclic aromatic hydrocarbons (PAHs), has been documented in the region over the past decades. However, a significant knowledge gap remains regarding the detection of new pollutants, such as emerging contaminants (ECs), in Antarctic coastal environments. This study analyzed the occurrence and levels of selected POPs, PAHs, ECs in surface sediments from Admiralty Bay, Antarctica Peninsula. Non-target screening was employed to identify potential novel contaminants in the region. Samples ( n = 17) were extracted using an accelerated solvent extraction (ASE) system and instrumental analyses were performed using gas chromatography coupled to a triple-quadrupole mass spectrometer (GC/MS-MS). Regarding regulated contaminants, concentrations of Σ5PCBs ranged from <lod 0.5="" g −1, while Σ16PAHs ranged from 2.6 to 617.4 ng g −1 . These compounds were predominant in the Martel inlet, suggesting influences of local input sources, especially for PAHs. ECs were detected in some sites of the Ezcurra and Martel inlets, especially nearby the Brazilian and Polish research stations. The UV filters homosalate (0.3 to 251.3 ng g −1 ) and octocrylene (0.7 to 93.8 ng g −1 ) were the most abundant EC, followed by TPP (0.1 to 87.4 ng g −1 ) and galaxolide (0.2 to 55.0 ng g −1 ). Applying non-target analysis, phthalates, and the fragrance OTNE (octahydro-tetramethyl-naphthalenyl-ethanone) were identified for the first time in Antarctic sediments. The data highlights the contribution of human activities in the region, possibly through wastewater effluents. This study provides data on the current levels of PAHs and POPs in the region, which are steadily decreasing over the years.

Abstract:Microplastics (MPs) cause an increased threat to the freshwater environment by adsorbing pollutants on their large surface area. Considering their adsorption characteristics, non-polar pollutants with high distribution coefficients have been studied extensively. However, comprehensive research on the types of polar pollutants adsorbed by MPs is lacking. In this study, a nontarget screening strategy, including classification and identification, was performed to analyze the pollutants adsorbed by MPs in Tai Lake and the Yangtze River. Compared with the pollutants adsorbed or added to raw plastics, more types of polar pollutants were found on MPs from freshwater. The nontarget classification of 4723 features on MPs from freshwater and 680 features from raw plastics were annotated based on the mass spectrometry spectra. Further identification with multiple platforms identified hundreds of pollutants absorbed by MPs in Tai Lake and Yangtze River, including industrial intermediates, medicines, and surfactants, exceeding those adsorbed by raw plastics, showing an enrichment of the pollutants on MPs in freshwater by secondary adsorption. Our study is the first to use nontarget analysis to comprehensively demonstrate MP adsorption and release of pollutants in freshwater environment, providing a significant reference for the research of MPs and the management of the water environment.
Persistent organic pollutants in the Antarctic marine environment: The influence impacts of human activity, regulations, and climate change

Abstract:This study investigates the presence, distribution, and potential impacts of perfluoroalkyl substances (PFASs) and hexabromocyclododecanes (HBCDs) on the Antarctic marine environment. The analysis results from the King Sejong Station, the Jang Bogo Station, and Cape Evans revealed the highest concentrations of both PFASs and HBCDs at King Sejong Station, indicating the significant influence of human activity. Short-chain perfluorocarboxylic acids (PFCAs) dominated the seawater samples, with PFPeA at the highest concentration (0.076 ng/L) at King Sejong Station, whereas perfluorosulfonic acids (PFSAs) were prevalent in the sediments, with PFHxS reaching 0.985 ng/g. Total PFASs in benthos ranged from N.D. to 2.40 ng/g ww across all stations. This indicated the effects of long-range transport and glacial meltwater. α-HBCD was the most common diastereomer in benthos samples, detected in 58.3% of samples, suggesting its selective persistency. Although risk quotient analysis revealed low immediate risks to lower-trophic organisms, potential risks remain owing to their persistence and bioaccumulation potential. Contaminant patterns changed after regulations: perfluorooctanesulfonic acid (PFOS) and perfluorooctanoic acid (PFOA) levels decreased, unregulated PFASs increased, HBCD stereoisomer ratios shifted towards α-HBCD dominance, and overall HBCD concentrations declined. Widespread persistence of regulated substances was observed in Antarctic environments, highlighting the need for comprehensive and long-term monitoring strategies. This study provides essential baseline data on contaminant distributions across the Southern Ocean, contributing to our understanding of emerging pollutants in Antarctic regions and informing future environmental protection efforts.
Exploiting Molecular Ions for Screening Hydrophobic Contaminants in Sediments using Gas Chromatography-Atmospheric Pressure Chemical Ionization-Ion Mobility-Mass Spectrometry

Abstract:Hydrophobic organic compounds (HOCs) are conventionally screened by matching experimental- to reference electron ionization (EI) mass spectra acquired using gas chromatography-mass spectrometry (GC-MS). However, for identification of novel substances that are absent from EI databases, de novo structure elucidation using EI is hampered by extensive in-source fragmentation. To address this problem, a new method based on GC-atmospheric pressure chemical ionization (APCI) coupled to ion mobility-high resolution mass spectrometry (IM-HRMS) was developed for simultaneous target, suspect, and non-target screening of HOCs. Of 102 target chemicals, 85.3% produced (quasi-)molecular ions as base peaks, with 71.6% displaying method detection limits lower than those of GC-EI-low resolution MS. The optimized method was applied to standard reference sediment and sediments from the Baltic Sea, an Arctic shelf, and a Norwegian lake. In total, we quantified 56 target chemicals with concentrations ranging from 4.86 pg g-1 dwt to 124 ng g-1 dwt. Further, using a combination of exact mass, collision cross section (CCS), retention time, and MS2 spectra, a total of 54 suspects were identified at Confidence Level (CL) 2. Among the remaining features, 169 were prioritized using a halogen-selective CCS cutoff (100 Å2 + 20% mass), leading to annotation 54 substances (CL ≤3). Notably, a suite of fluorotelomer thiols, disulfides, and alkyl sulfones were identified in sediment (CL 1-2) for the first time. Overall, this work demonstrates the potential of GC-APCI-IM-HRMS as a next-generation technique for resolving complex HOC mixtures in environmental samples through exploitation of molecular ions.

A targeted and non-targeted discovery screening approach for poly-and per-fluoroalkyl substances in model environmental biota samples

Abstract:A comprehensive analytical approach for targeted and non-targeted discovery screening of per- and polyfluoroalkyl substances (PFAS) was developed and applied to model complex environmental biotic samples. Samples were extracted by formic acid-acetonitrile solution and cleaned up and fractionated by SPE (WAX). Target PFAS quantification was performed by ultra-high performance liquid chromatography interfaced with a triple quadrupole mass spectrometer (UPLC-QqQ-MS/MS). Non-targeted analysis (NTA) PFAS screening was performed with UPLC coupled with a quadrupole-Exactive orbitrap high resolution mass spectrometer (UPLC-Q-Exactive-HRMS). An iterative exclusion (IE) approach was applied to data acquisition for NTA suspect screening to increase the potential for unknown PFAS discovery with MS/MS. A complex workflow in Compound Discoverer was set up to automate data processing of the PFAS suspects search. New mass lists and MS/MS databases, which included a large number of PFAS, were set up and introduced into the search for high-throughput structure identification using HRMS techniques. The integrated targeted-NTA method successfully analyzed for legacy and alternative PFAS in model environmental biota samples, namely polar bear liver and bird egg samples. Targeted analysis provided unequivocal identification of well known/established PFAS (mainly perfluoroalkyl acids) with quantification at very low levels. The NTA suspect screening was able to determine a broader range of PFAS. The data analysis method offered high-confidence annotations for PFAS despite lacking available authentic standards. Overall, the analytical coverage of PFAS was greater and elucidated other PFAS present in these model apex predators.
Screening for Polar Chemicals in Water by Trifunctional Mixed-Mode Liquid Chromatography-High Resolution Mass Spectrometry

Abstract:The presence of persistent and mobile organic contaminants (PMOC) in aquatic environments is a matter of high concern due to their capability of crossing through natural and anthropogenic barriers, even reaching drinking water. Most analytical methods rely on reversed-phase liquid chromatography (RPLC), which is quite limited for the detection of very polar chemicals. Thus, many of these PMOCs may have not been recognized as water pollutants yet, due to the lack of analytical methods capable to detect them. Mixed-mode LC (MMLC), providing the combination of RP and ion-exchange functionalities is explored in this work with a trifunctional column, combining RPLC, anion and cation exchange, which allows the simultaneous determination of analytes with extremely different properties. A nondiscriminant sample concentration step followed by a MMLC-high resolution mass spectrometry method was developed for a group of 37 very polar model chemicals with different acid/base functionalities. The overall method performance was satisfactory with a mean limit of detection of 50 ng/L, relative standard deviation lower than 20% and overall recoveries (including matrix effects) higher than 60% for 54% of model compounds. Then, the method was applied to 15 real water samples, by a suspect screening approach. For those detected PMOC with standard available, a preliminary estimation of concentrations was also performed. Thus, 22 compounds were unequivocally identified in a range of expected concentrations from 6 ng/L to 540 μg/L. Some of them are well-known PMOC, such as acesulfame, perfluorobutanoic acid or metformin, but other novel pollutants were also identified, as for example di-o-tolylguanidine or trifluoromethanesulfonic acid, which had not or were scarcely studied in water so far.
Single injection LC-MS/MS analytical method for the quantification of diverse families of micropollutants, including PFAS and organotins, in Gammarus fossarum

Abstract:Since the last decades, light has been shed on the pollution of aquatic ecosystems. Considering apolar compounds in water, analysis of the dissolved phase is not sufficient due to their possible bioaccumulation. Additional analysis of sediments, biota, or sentinel species is necessary. Among sentinel species, Gammarus fossarum is a small shrimp of 30 mg that lives naturally in the river. Its ability to bioaccumulate makes it a good bioindicator of river pollution. Nevertheless, micropollutants are difficult to extract from gammarids due to their small size and their high level of lipids. Extracted interferences can lead to analytical difficulties. Targeted micropollutants in this work were organotins, personal and pharmaceutical care products (PPCPs), pesticides, flame retardants, and perfluorinated compounds. A sample preparation based on QuEChERS followed by liquid chromatography coupled to tandem mass spectrometry (LC-MS/MS) was developed: different salts (acetate and citrate) and purification (addition of hexane, dispersive SPE) were tested and validated. Targeted molecules used to be analyzed by LC-MS/MS, except organotins which are principally analyzed in gas chromatography. One of the main challenges was to quantify them also in LC-MS/MS to implement an original multi-residue method. The analysis of 40 compounds was finally validated according to ICH guidelines, with LOQs ranging from 0.04 ng.g-1 to 313.5 ng.g-1. Regarding Perfluorooctanesulfonic acid (PFOS) and the sum of hexabromocyclododecane, LOQs reached the environmental quality standards in biota which are respectively of 9.1 and 167 ng.g-1. Finally, the method was applied to 15 real samples. Many compounds were quantified: perfluorinated compounds, drugs such as ketoprofen, and even cosmetics products such as octocrylene. This is the first study to quantify as many emerging compounds, especially organotins, in a sentinel species such as G. fossarum.
Vertical distribution and risk assessment of pharmaceuticals and other micropollutants in southern North Sea coastal waters

Abstract:Pharmaceutical compounds are micropollutants of emerging concern, as well as other classes of chemicals such as UV filters and artificial sweeteners. They enter marine environments via wastewater treatment plants, aquaculture runoff, hospital effluents, and shipping activities. While many studies have investigated the presence and distribution of these pollutants in numerous coastal areas, our study is the first to focus on their occurrence, spatial distribution, and vertical distribution in the sea surface microlayer (SML) and the near-surface layer of marine environments. We analyzed 62 pharmaceutical compounds, one UV filter, and six artificial sweeteners from the SML to the corresponding underlying water (0 cm, 20 cm, 50 cm, 100 cm, and 150 cm) at four stations in the southern North Sea. One station is the enclosed Jade Bay, one is the Weser estuary at Bremerhaven, and the other two stations (NS_7 and NS_8) are in the open German Bight. Jade Bay receives pollutants from surrounding wastewater treatment plants, while the Weser estuary receives pollutants from cities like Bremerhaven, which has dense populations and industrial activities. Concentrations of pharmaceutical compounds were higher in the upper water layers (from the SML to 20 cm). Eleven pharmaceutical compounds (caffeine, carbamazepine, gemfibrozil, ibuprofen, metoprolol, salicylic acid, clarithromycin, novobiocin, clindamycin, trimethoprim, and tylosin) were detected in >95 % of our samples. One UV filter (benzophenone-4) was found in 83 % and three artificial sweeteners (acesulfame, saccharin, and sucralose) in 100 % of all our samples. All artificial sweeteners posed high risks to the freshwater invertebrate Daphnia magna . Understanding the spatial and vertical distribution of pharmaceuticals and other micropollutants in marine environments may be essential in assessing their dispersal and detection in other aquatic environments.

Abstract:While the coastal pollution of persistent toxic substances (PTSs) has been widely documented, information on offshore environments remains limited. Here, we investigated the spatial distribution and sources of PTSs in the offshore sediments (n = 34) of South Korea. Sediment samples collected from the Yellow Sea (n = 18), the South Sea (n = 10), and the East Sea (n = 6), in 2017-18 were analyzed for a total of 71 PTSs. Target compounds include 31 PCBs, 15 PAHs, 9 emerging PAHs (e-PAHs), 10 styrene oligomers (SOs), and 6 alkylphenols (APs). Sedimentary PCBs showed relatively low concentrations with no significant difference across the three seas (0.16-6.9 ng g-1 normalized organic carbon, OC). Low-chlorinated PCBs (tri- and tetra Cl-CBs) were predominant (mean: 77%), primarily indicating atmospheric inputs. PAHs widely accumulated in the three seas with low to moderate level (22-250 ng g-1 OC), and dominated by high molecular weight PAHs (4-6 rings). PMF analysis revealed coast-specific PAHs sources; i.e., originated from mainly coke production (77%) in the Yellow Sea, vehicle emissions (68%) in the South Sea, and fossil fuel combustion (49%) in the East Sea. SOs showed significant contamination than other PTSs, with elevated concentrations in the Yellow Sea (mean: 350 ng g-1 OC). APs showed a similar regional distribution to SOs, but concentrations were much lower (mean: 17 ng g-1 OC). SOs and APs seemed to be introduced from rivers and estuaries on the west coast of Korea, where industrial and municipal activities are concentrated, then might be transported to offshore through tide or currents. Overall, the novel data presented for various PTSs in offshore Korean sediments warrant the necessity of a long-term monitoring effort and urgent management practice to protect marine ecosystem.

Abstract:A highly sensitive peptide mapping method using derivatization and fluorescence detection is described. Bovine cytochrome c was digested using a buffer compatible with the derivatization that followed. The derivatization was performed with 6-aminoquinolyl-N-hydroxysuccinimidyl carbamate. The peptide mapping of the tagged digest was conducted with both HPLC and capillary LC (CLC) systems. A capillary LC-electrospray ionization mass spectrometer (MS) was set up for measuring the molecular weights of the tagged peptides. Optimization was made of the conditions used for digestion, derivatization, and mapping. MS measurements of the tagged peptides suggested that there was only one derivatization product produced from all peptides (except one) and that all the identified peptides were fully tagged. Peptide mapping of the tagged digest reviews a larger number of peptides, covering almost the entire sequence. Peptide mapping of a 20 fmol amount of tagged digest was readily performed with the CLC system. By using derivatization and fluorescence detection, the sensitivity of peptide mapping could be improved 2000 times compared to that observed with uv detection of untagged peptides.

Abstract:The 'heat separator' is a very simple device for attaining a temperature drop of the order of 30 centigrade degrees by means of a stream of air. An air jet enters a long cylindrical chamber near one end, and is directed so as to maintain a high velocity vortex. Air drawn off from the centre of the vortex is cooled, while the remainder, flowing down the length of the chamber, is warmed. The present paper describes results of a quantitative study of the performance of the device. Although the maximum efficiency is only about 15%, as compared with an ideal adiabatic expansion of the total flow, it provides a practical means of cooling for many purposes.

Abstract:Freshwater systems are facing a number of pressures due to the inputs of polar organic contaminants from a range of sources including agriculture, domestic and industry. The River Itchen and River Test are two sensitive chalk streams in Southern England that are experiencing a decline in invertebrate communities. We used Chemcatcher passive samplers to measure time-weighted average concentrations (14 days) of polar pollutants at nine sites on the River Itchen and eight sites on the River Test over a 12-month period. Sampler extracts were analysed using a targeted LC/MS method. In total, 121 plant protection products and pharmaceutical and personal care products were quantified (range of log Kow from - 1.5 to 7). Concentrations (sub ng L-1 to >500 ng L-1) in both rivers showed spatial and temporal variations. A greater number of compounds and higher concentrations were found in the River Test. The chemical profile was dominated by inputs from wastewater treatment plants and legacy plant protection products. On the River Itchen, high concentrations (∼100 ng L-1) of caffeine were observed directly downstream of a fish farm. Using the NORMAN database, the predicted no effect concentration (PNEC) freshwater values were exceeded by only five contaminants (2-hydroxy-terbuthylazine, alprazolam, azithromycin, diclofenac and imidacloprid). In addition, venlafaxine was detected above its EU Watch List concentration. These exceedances were mainly downstream of direct inputs from treatment plants. These compounds are known to have ecotoxicological effects on a range of aquatic biota including macroinvertebrates. Of concern is the ubiquitous presence of the ectoparasiticide imidacloprid, highlighting the need to control its use. The impact of the cocktail of pollutants found in this study on the long-term effects on chalk stream ecosystems remains unknown and needs further investigation.
