Abstract:The coupling of disposable solid-phase microextraction (SPME) with performance reference compounds (PRCs) has been recently introduced to measure time-averaged freely dissolved concentrations (Cfree) of hydrophobic organic contaminants in sediments under laboratory conditions. To explore the use of PRC-SPME for in situ sampling in seawater, disposable PDMS fibers (35-μm and 100-μm coating) preloaded with stable isotope labeled analogues as PRCs were deployed at six stations (each with three depths) in the open ocean of the Palos Verdes Shelf (CA, USA) Superfund site for 33d to measure Cfree of DDT and its degradates. The observed values of fractional equilibration (feq) of PRCs were mostly <0.85, suggesting nonequilibrium conditions at the end of deployment. The observed feqs for the samplers varied with compound, sampling station and depth, validating the need for calibration to derive accurate Cfree. The Cfree values of DDE and DDD determined with PRC-SPME were in good agreement with those previously measured by in situ large-volume water sampling or polyethylene devices. The highest Cfree in seawater 5m off the ocean floor was 750pgL−1 for o,p′-DDE, 2170pgL−1 for p,p′-DDE, 24pgL−1 for o,p′-DDD, and 75pgL−1 for p,p′-DDD. Results of this study demonstrated the feasibility and advantages of using disposable PDMS fiber coupled with PRCs for in situ sampling.
Tire-derived contaminants 6PPD and 6PPD-Q: Analysis, sample handling, and reconnaissance of United States stream exposures

Abstract:The environmental ubiquity of tire and road wear particles (TRWP) underscores the need to understand the occurrence, persistence, and environmental effects of tire-related chemicals in aquatic ecosystems. One such chemical is 6PPD-quinone (6PPD-Q), a transformation product of the tire antioxidant 6PPD. In urban stormwater runoff 6PPD-Q can exceed acute toxicity thresholds for several salmonid species and is being implicated in significant coho salmon losses in the Pacific Northwest. There is a critical need to understand the prevalence of 6PPD-Q across watersheds to identify habitats heavily affected by TRWPs. We conducted a reconnaissance of 6PPD and 6PPD-Q in surface waters across the United States from sites (N = 94) with varying land use (urban, agricultural, and forested) and streamflow to better understand stream exposures. A rapid, low-volume direct-inject, liquid chromatography mass spectrometry method was developed for the quantitation of 6PPD-Q and screening for 6PPD. Laboratory holding times, bottle material, headspace, and filter materials were investigated to inform best practices for 6PPD-Q sampling and analysis. Glass bottles with PTFE-lined caps minimized sorption and borosilicate glass fiber filters provided the highest recovery. 6PPD-Q was stable for at least 5 months in pure laboratory solutions and for 75 days at 5 °C with minimal headspace in the investigated surface water and stormwaters. Results also indicated samples can be frozen to extend holding times. 6PPD was not detected in any of the 526 analyzed samples and there were no detections of 6PPD-Q at agricultural or forested sites. 6PPD-Q was frequently detected in stormwater (57%, N = 90) and from urban impacted sites (45%, N = 276) with concentrations ranging from 0.002 to 0.29 μg/L. The highest concentrations, above the lethal level for coho salmon, occurred during stormwater runoff events. This highlights the importance of capturing episodic runoff events in urban areas near ecologically relevant habitat or nursery grounds for sensitive species.

Abstract:N-(1,3-dimethylbutyl)-N '-phenyl-p-phenylenediamine (6PPD) as a rubber antioxidant has attracted global concern, since its ozone-oxidation product 6PPD-quinone (6PPDQ) was found to be the primary toxicant responsible for urban runoff mortality syndrome in coho salmon. However, the biotransformation fate and associated toxicological mechanism of 6PPD have not received much study yet. In this work, the in vitro assays showed 6PPD can be transformed into 6PPDQ by cytochromes P450 (CYP450) in human liver microsomes (HLMs) with 0.98 % production rate, and the adducts of 6PPDQ with calf thymus DNA and the N-N coupling product between 6PPD and 6PPDQ were further identified after 6PPD incubation in HLMs. Further evidence for the 6PPDQ formation can be obtained from the in vivo assays that the 6PPDQ-DNA adducts and 6PPD-N-N6PPDQ dimer were detected in mice by oral gavage with 6PPD, and the latter dimer species was detected as well in 6PPD exposure to zebrafish larvae. Especially, the bioaccumulation property and high reactivity of 6PPDQ result in the continuous formation of the significant DNA adducts and 6PPD-N-N-6PPDQ dimer even in case of low production rate of biotransformation of 6PPD to 6PPDQ, which may provide potentially effective biomarkers for such process. DFT computations revealed the formation mechanism of 6PPDQ is the (N)Habstraction of 6PPD by CYP450, followed by amino radical rebound at the nearby ortho-carbon, yielding a quinol intermediate due to spin delocalization, that might readily undergo further oxidation by CYP450 into 6PPDQ.

Abstract:The chemical 6PPD-quinone is highly toxic to some fish species of the Oncorhynchus and Salvelinus genera and is the oxidation product of the common car tire additive 6PPD. We present a new sample preparation method that involves liquid-liquid extraction of water samples followed by silica-based solid phase extraction prior to LC–MS/MS analysis. The new sample preparation method showed good analyte recovery from spiked water samples (78%–91%) and a low ion suppression effect, surpassing previously published methods. This new method was successfully validated, achieving a limit of quantification of 5 ng/L and estimated expanded measurement uncertainty of 18.6%. In a proof-of-concept study, the method was applied to several water samples from various sources in Southern Norway. These were runoff samples from tunnel washing, from a tunnel runoff treatment plant and downstream of the plant drain. In addition, two water samples from puddles were included: one was run-off from an artificial soccer turf field and one from a puddle on a country road. The results of the analyses revealed that the concentration of 6PPD-quinone was above the LC50 reported for coho salmon ( Oncorhynchus kisutch ) in all samples except the samples from and downstream of the treatment plant. The highest measured concentration was 258 ng/L, which is the 2.7-fold of the reported LC50 in coho salmon (95 ng/L). Our initial data emphasize the need for more comprehensive environmental monitoring of 6PPD-quinone as well as toxicological studies in aquatic organisms.

Abstract:p-Phenylenediamines (PPDs) and PPD-derived quinones (PPD-Qs) have been considered emerging pollutants recently. Their available data on sediment and sewage sludge are limited, especially the ecological risks. Here, typical PPDs and PPD-Qs were measured in the sludge of wastewater treatment plants and surface sediment of a developed river basin (including reservoirs, estuaries, and rivers) and deep-sea troughs. The total concentrations of PPDs (∑PPD) were highest in sludge (range: 9.06-248 ng g-1), followed by surface sediment of the Dongjiang River basin, China (3.33-85.3 ng g-1), and lowest in sediment of the Okinawa Trough (0.01-7.46 ng g-1). The median value of ∑PPD in surface sediment of rivers (9.54 ng g-1) was higher than those in reservoirs (4.28 ng g-1) and estuaries (5.26 ng g-1). N-(1,3-Dimethylbutyl)-N'-phenyl-p-phenylenediamine (6PPD) was the major congener in all samples, accounting for over 60 % of ∑PPD. For quinones, 6PPD-Q and IPPD-Q were frequently detected in sludge, only trace 6PPD-Q was detected in the sediment of estuaries (nd-0.62 ng g-1) and rivers (nd-5.24 ng g-1), and both of them were absent from the sediment of the Okinawa Trough. The occurrence of PPDs in the trough may be the in-situ release of microplastics, and due to the low-light and weak alkaline conditions of deep-sea water, quinones may hardly photodegrade from PPDs. The PPD concentrations in sludge were positively correlated with local GDP, and the annual PPD emission from sludge will exceed 1370 kg in China. The results of ecological risk assessments indicated low risks for PPDs in sludge-amended soil, median risks for several PPDs in river sediment, but median to high risks for 6PPD-Q contamination sludge-amended soil. For the first time, we reported the occurrence of PPDs and related quinones in the sludge of WWTPs and found a potential environmental risk from 6PPD-Q in sludge used as a soil conditioner.

Abstract:The widespread occurrence of N-(1,3-Dimethylbutyl)-N′-phenyl-p-phenylenediamine-quinone (6PPD-Q) in aquatic environments and its hazards to aquatic species underscore the necessity of comprehending its environmental fate. Here, we investigated the transformation from 6PPD to 6PPD-Q and the attenuation of 6PPD-Q in surface water under natural conditions. Contrary to prior findings, this work revealed that 6PPD-Q and its precursor 6PPD-OH/6PPD-(OH) 2 , were not detected through target and suspect screening during 6PPD transformation in the surface water under the natural condition. 6PPD-Q predominantly accumulated in TWPs in ambient atmosphere with 1.28 % mass yield from the 6PPD dissipation. Subsequently, 6PPD-Q was eluted from TWPs and released to the water environment. The investigation on the natural attenuation of 6PPD-Q in the surface water demonstrated that direct and indirect photolysis facilitated the rapid dissipation of 6PPD-Q with a half-life of 2.57 h. Utilizing the liquid chromatography high resolution mass spectrometry (LC-HRMS), including both time of flight (TOF) MS and Orbitrap MS, twelve novel transformation products (TPs) of 6PPD-Q were identified by using a comprehensive non-targeted screening strategy. The results from two dimensions gas chromatography (GC×GC) TOF-MS revealed additional two TPs. Based on the molecular structure of TPs, four major pathways of 6PPD-Q attenuation were proposed, including bond cleavage, hydroxylation, quinone cleavage and rearrangement. All TPs were predicted to exhibit lower toxicity, indicating the natural attenuation of 6PPD-Q reduced its toxicity and potential environmental risks. This study provides crucial insights into the environmental fate of 6PPD-Q, highlighting the significance of understanding both its formation from 6PPD and its subsequent attenuation processes under natural conditions.

Abstract:Stormwater exposure can cause acute mortality of coho salmon (Oncorhynchus kisutch), and 6PPD-quinone (6PPD-Q) was identified as the primary causal toxicant. Commercial standards of 6PPD-Q recently became available; their analysis highlighted a systematic high bias in prior reporting concerning 6PPD-Q. A 6PPD-Q commercial standard was used to re-confirm toxicity estimates in juvenile coho salmon and develop a liquid chromatography-tandem mass spectrometry analytical method for quantification. Peak area responses of the commercial standard were ∼15 times higher than those of in-house standards, and the updated LC50 value (95 ng/L) was ∼8.3-fold lower than that previously reported. These data support prior relative comparisons of the occurrence and toxicity while confirming the substantial lethality of 6PPD-Q. While environmental concentrations are expected to be lower, 6PPD-Q also was more toxic than previously calculated and should be categorized as a “very highly toxic” pollutant for aquatic organisms. Isotope dilution-tandem mass spectrometry methods enabled accurate quantification (limits of quantification of <10 ng/L) within environmental samples.

Abstract:Existing data on source parameters of large crustal earthquakes (subduction events are not considered here) over a wide range of repeat times indicate that, for a given magnitude (M_s or M_w), earthquakes with long repeat times have shorter fault lengths than those with short repeat times. A shorter fault length for a given magnitude indicates a larger average stress drop which reflects the average strength of the fault zone. Our result therefore suggests that faults with longer repeat times are stronger than those with shorter repeat times. In terms of an asperity model in which the average strength of a fault zone is determined by the ratio, r_a, of the total area of the asperities (strong spots on a fault plane) to the total area of the fault zone, the above result suggests that r_a is proportional to the repeat time. Our result provides a method to estimate seismic source spectra from the fault length and the repeat time of a potential causative fault.
Occurrence and Fate of Substituted p-Phenylenediamine-Derived Quinones in Hong Kong Wastewater Treatment Plants

Abstract:para-Phenylenediamine quinones (PPD-Qs) are a newly discovered class of transformation products derived from para-phenylenediamine (PPD) antioxidants. These compounds are prevalent in runoff, roadside soil, and particulate matter. One compound among these, N-1,3-dimethylbutyl-n'-phenyl-p-phenylenediamine quinone (6PPD-Q), was found to induce acute mortality of coho salmon, rainbow trout, and brook trout, with the median lethal concentrations even lower than its appearance in the surface and receiving water system. However, there was limited knowledge about the occurrence and fate of these emerging environmental contaminants in wastewater treatment plants (WWTPs), which is crucial for effective pollutant removal via municipal wastewater networks. In the current study, we performed a comprehensive investigation of a suite of PPD-Qs along with their parent compounds across the influent, effluent, and biosolids during each processing unit in four typical WWTPs in Hong Kong. The total concentrations of PPDs and PPD-Qs in the influent were determined to be 2.7-90 and 14-830 ng/L. In the effluent, their concentrations decreased to 0.59-40 and 2.8-140 ng/L, respectively. The median removal efficiency for PPD-Qs varied between 53.0 and 91.0% across the WWTPs, indicating that a considerable proportion of these contaminants may not be fully eliminated through the current processing technology. Mass flow analyses revealed that relatively higher levels of PPD-Qs were retained in the sewage sludge (20.0%) rather than in the wastewater (16.9%). In comparison to PPDs, PPD-Qs with higher half-lives exhibited higher release levels via effluent wastewater, which raises particular concerns about their environmental consequences to aquatic ecosystems.

Abstract:The extensive use of synthetic antioxidants, notably N-(1,3-dimethylbutyl)-N'-phenyl-p-phenylenediamine (6-PPD), in rubber-related products, particularly in tire manufacturing, has induced concerns regarding their environmental impact and potential health hazards. Despite the identification of 6-PPD and its derivative, 6-PPD quinone (6-PPDQ), in various water samples and their lethal effects on certain aquatic species (e.g., coho salmon, rainbow trout and brook trout), the levels of airborne 6-PPD/6-PPDQ and their respiratory toxicity remain relatively unexplored. In this study, we aimed to evaluate the respiratory toxicity potential of 6-PPD and its derivatives, with a specific focus on detecting these compounds in airborne particulates and assessing their toxic effects on lung cells. Characterization of four airborne fine particulate (FP) samples revealed spherical morphologies with diameters ranging from 17.7 to 225.7 nm, displaying slight agglomeration and negative surface charge. methanol/acetonitrile extraction followed by liquid chromatography coupled with tandem mass spectrometry (LC-MS/MS) analysis confirmed the presence of both 6-PPD and 6-PPDQ on the surfaces of FPs, with significant variations (0.26-1.05 μg g-1) in loading capacity observed among the samples. Subsequent exposure of lung cells (THP-1, BEAS-2B, and A549) to 6-PPD and 6-PPDQ revealed dose-dependent declines in mitochondrial metabolic activity induced by 6-PPD, along with severe membrane damage, ATP depletion, and pro-inflammatory cytokine release. Conversely, 6-PPDQ exhibited negligible toxicity in all tested parameters. These findings underscore the potential health risks associated with airborne 6-PPD exposure and emphasize the importance of further research into the respiratory toxicity of 6-PPD derivatives.

Abstract:N , N ′-substituted p -phenylenediamines (PPDs) are widely used antioxidants in rubber tires, which could be released and accumulated in road dusts with rubber tires wear. As ozonation product of N -(1,3-dimethylbutyl)- N′ -phenyl- p -phenylenediamine (6PPD), 6PPD-quinone (6PPD-Q) exhibited higher toxicity to coho salmon. However, studies on their environmental behaviors are still limited. Road dust is the major medium PPDs exist, which significantly affects the levels of PPDs in other mediums, especially surface water and particulate matter. In this study, road dust samples were collected in 55 major cities of China to explore the distribution characteristics of PPDs and 6PPD-Q. The concentrations of total PPDs (ΣPPDs) and 6PPD-Q in urban trunk road dust samples were in the ranges of 7.90–727 and 3.00–349 ng/g, with median concentrations of 68 and 49 ng/g, respectively. 6PPD and 6PPD-Q are the dominant components in most road dusts. The functional region-dependent pollution characteristics of PPDs and 6PPD-Q give the first finding that urban tunnel road was the highly polluted region, followed by urban trunk roads. Suburban road dusts had a lower pollution level. Moreover, the estimated daily intake (EDI) of PPDs and 6PPD-Q for children was much higher than adults.

Abstract:To better understand the environmental impact of ubiquitous perfluoroalkyl substances (PFASs) in waters, reliable and robust measurement techniques are needed. As one of the most widely used passive sampling approaches, diffusive gradients in thin-films (DGT) is not only easy to handle but also provides time-weighted analyte concentrations. Based on DGT with XAD18 as a binding agent, we developed a new methodology to measure two frequently detected PFASs in surface waters and wastewaters, i.e. perfluorooctanoic acid (PFOA) and perfluorooctane sulfonate (PFOS). Their diffusion coefficients in the diffusive gel, measured using an independent diffusion cell, were 4.37 × 10-6 and 5.08 × 10-6 cm2 s-1 at 25 °C, respectively. DGT had a high capacity for PFOA and PFOS at 196 and 246 μg per gel disk, suggesting the DGT sampler was suitable for deployment of several weeks. Time-integrated concentrations of PFOA and PFOS in a natural lake and river, and a municipal wastewater treatment plant effluent using DGT samplers deployed in situ for 12-33 d were comparable to those measured by a solid-phase extraction method coupled with high-frequency grab sampling. This study demonstrates that DGT is an effective tool for in situ monitoring of PFASs in natural waters and wastewaters.

Abstract:Wearing of vehicle parts could release many chemical additives into the environment, such as benzotriazoles (BTRs), benzothiazoles (BTHs), and p-phenylenediamines (PPDs), which are potentially toxic to wildlife and humans. This study investigated the occurrence, source, and risks of BTRs, BTHs, and PPDs in a source catchment providing water to Guangzhou, a megacity in South China, covering groundwater, surface water, and stormwater. The results showed that BTRs and BTHs were predominant in surface water and groundwater. Unexpectedly, the BTR and BTH concentrations were lower in surface water than groundwater in a third of the paired samples. For the first time, 6PPD-quinone, a toxic ozonation product of N-(1,3-dimethylbutyl)-N'-phenyl-1,4-phenylenediamine (6PPD), was extensively detected in source waters. Stormwater decreased the BTR concentrations but increased the 6PPD-quinone concentrations in surface water owing to their affiliation to suspended particles. From natural to urban segments of Liuxi river, a downstream increasing trend in BTR and BTH concentrations was observed, confirming that they are indicative of urban anthropogenic activities. Strong correlations between industrial activities and BTR or BTH concentrations in surface water indicated that industrial activities were their main sources. Six compounds were prioritized as potentially persistent, mobile, and toxic (PMT) chemicals, combing our monitoring results and REACH criterion. This study improves our understanding of the environmental fates and risks of water-soluble tire-wear chemicals, which provides important information for chemical management, and indicates attention should be paid to the risk posed by 6PPD-quinone in the source water.

Abstract:Tire wear compounds N-(1,3-dimethylbutyl)-N′-phenyl-p-phenylenediamine (6PPD) and its derivative 6PPD-quinone have been considered as emerging pollutants and attracted much attention recently. As an antioxidant and antiozonant widely used, 6PPD would be released during the production or use of rubber-related products. Because of the mass production and wide use of rubber-related products, 6PPD and 6PPD-quinone have been identified to be ubiquitous in the environment. In this study, we firstly reviewed the current available literature on the analytical procedures, concentrations and distribution of 6PPD and 6PPD-quinone, and then investigated the potential toxic effects of these two compounds on aquatic organisms. Current studies have been mainly focused on the occurrence of 6PPD and 6PPD-quinone in dust and water, while available information on atmosphere, soil, sediments and organisms is limited. The fate and distribution of 6PPD and 6PPD-quinone would be influenced by environmental factors such as temperature, illumination, and storm events, etc. Although 6PPD and 6PPD-quinone have potential adverse effects on aquatic organisms, and 6PPD-quinone has species-specific toxicity, toxicological mechanisms of these compounds are still unclear. Based on the review and analysis of current studies, some suggestions for future research of 6PPD and 6PPD-quinone are given.

Abstract:The tire antioxidant 6-PPD has been widely used to enhance tire performance and extend tire lifespan. 6-PPD quinone (6-PPDQ), a quinone derivative derived from 6-PPD in the presence of ozone, has been recognized an emerging environmental contaminant. In addition to causing acute lethality to coho salmon, 6-PPDQ exhibits toxic effects on other aquatic species and mammals. Based on the existing evidence, we provide a critical overview on the human internal exposure, potential adverse effects on health, and prediction of human health risk of 6-PPDQ. 6-PPDQ could be detected in human samples, including human urine, blood, and cerebrospinal fluid. Human exposure to 6-PPDQ in the environment is inevitable and may lead to adverse health effects, including hepatotoxicity, enterotoxicity, pulmonary toxicity, neurotoxicity, reproductive toxicity, and cardiotoxicity. Additionally, potential human health risk to 6-PPDQ through exposure routes and human samples were predicted. This review is helpful to identify the existing knowledge gaps and future research directions regarding the human health effects of 6-PPDQ.
Nontarget analysis and characterization of p-phenylenediamine-quinones and -phenols in tire rubbers by LC-HRMS and chemical species-specific algorithm

Abstract:Background: N,N'-disubstituted p-phenylenediamine-quinones (PPDQs) are oxidization derivatives of p-phenylenediamines (PPDs) and have raised extensive concerns recently, due to their toxicities and prevalence in the environment, particularly in water environment. PPDQs are derived from tire rubbers, in which other PPD oxidization products besides reported PPDQs may also exist, e.g., unknown PPDQs and PPD-phenols (PPDPs). Results: This study implemented nontarget analysis and profiling for PPDQ/Ps in aged tire rubbers using liquid chromatography-high-resolution mass spectrometry and a species-specific algorithm. The algorithm took into account the ionization behaviors of PPDQ/Ps in both positive and negative electrospray ionization, and their specific carbon isotopologue distributions. A total of 47 formulas of PPDQ/Ps were found and elucidated with tentative or accurate structures, including 25 PPDQs, 18 PPDPs and 4 PPD-hydroxy-quinones (PPDHQs). The semiquantified total concentrations of PPDQ/Ps were 14.08-30.62 μg/g, and the concentrations followed the order as: PPDPs (6.48-17.39) > PPDQs (5.86-12.14) > PPDHQs (0.16-1.35 μg/g). Significance: The high concentrations and potential toxicities indicate that these PPDQ/Ps could seriously threaten the eco-environment, as they may finally enter the environment, accordingly requiring further investigation. The analysis strategy and data-processing algorithm can be extended to nontarget analysis for other zwitterionic pollutants, and the analysis results provide new understandings on the environmental occurrence of PPDQ/Ps from source and overall perspectives.

Abstract:Addressing the challenge of accurately monitoring polycyclic aromatic hydrocarbons (PAHs) in aquatic systems, this study employed diffusive gradients in thin-films (DGT) technique to achieve methods detection limits as low as 0.02 ng L −1 to 0.05 ng L −1 through in situ preconcentration and determination of time-integrated concentrations. The efficacy of the developed DGT samplers was validated under diverse environmental conditions, demonstrating independence from factors such as pH (5.03–9.01), dissolved organic matter (0–20 mg L −1 ), and ionic strength (0.0001–0.6 M). Notably, the introduction of a novel theoretical approach to calculate diffusion coefficients based on solvent-accessible volume tailored for PAHs significantly enhanced the method's applicability, particularly for organic pollutants with low solubility. Field deployments in coastal zones validated the DGT method against traditional grab sampling, with findings advocating a 4 to 7-day optimal deployment duration for balancing sensitivity and mitigating lag time effects. These results provide a sophisticated, efficient solution to the persistent challenge of monitoring hydrophobic organic pollutants in aquatic environments, broadening the scope and applicability of DGT in environmental science and providing a robust tool for researchers.
