Abstract:The integration of microfluidics with electric field control, commonly referred to as electrofluidics, has led to new opportunities for biomedical analysis. The requirement for closed microcapillary channels in microfluidics, typically formed via complex microlithographic fabrication approaches, limits the direct accessibility to the separation processes during conventional electrofluidic devices. Textile structures provide an alternative and low‐cost approach to overcome these limitations via providing open and surface‐accessible capillary channels. Herein, we investigate the potential of different 3D textile structures for electrofluidics. In this study, 12 polyester yarns were braided around nylon monofilament cores of different diameters to produce functional 3D core–shell textile structures. Capillary electrophoresis performances of these 3D core–shell textile structures both before and after removing the nylon core were evaluated in terms of mobility and bandwidth of a fluorescence marker compound. It was shown that the fibre arrangement and density govern the inherent capillary formation within these textile structures which also impacts upon the solute analyte mobility and separation bandwidth during electrophoretic studies. Core–shell textile structures with a 0.47 mm nylon core exhibited the highest fluorescein mobility and presented a narrower separation bandwidth. This optimal textile structure was readily converted to different geometries via a simple heat‐setting of the central nylon core. This approach can be used to fabricate an array of miniaturized devices that possess many of the basic functionalities required in electrofluidics while maintaining open surface access that is otherwise impractical in classical approaches.

Abstract:Electric field–driven microfluidics, known as electrofluidics, is a novel attractive analytical tool when it is integrated with low‐cost textile substrate. Textile‐based electrofluidics, primarily explored on yarn substrates, is in its early stages, with few studies on 3D structures. Further, textile structures have rarely been used in cellular analysis as a low‐cost alternative. Herein, we investigated novel 3D textile structures and develop optimal electrophoretic designs and conditions that are favourable for direct 3D cell culture integration, developing an integrated cell culture textile‐based electrofluidic platform that was optimised to balance electrokinetic performance and cell viability requirements. Significantly, there were contrasting electrolyte compositional conditions that were required to satisfy cell viability and electrophoretic mobility requiring the development of and electrolyte that satisfied the minimum requirements of both these components within the one platform. Human dermal fibroblast cell cultures were successfully integrated with gelatine methacryloyl (GelMA) hydrogel‐coated electrofluidic platform and studied under different electric fields using 5 mM TRIS/HEPES/300 mM glucose. Higher analyte mobility was observed on 2.5% GelMA‐coated textile which also facilitated excellent cell attachment, viability and proliferation. Cell viability also increased by decreasing the magnitude and time duration of applied electric field with good cell viability at field of up to 20 V cm−1.

Abstract:Paper and thread are widely used as the substrates for fabricating low-cost, disposable, and portable microfluidic analytical devices used in clinical, environmental, and food safety monitoring. Concerning separation methods including chromatography and electrophoresis, these substrates provide unique platforms for developing portable devices. This review focuses on summarizing recent research on the miniaturization of the separation techniques using paper and thread. Preconcentration, purification, desalination, and separation of various analytes are achievable using electrophoresis and chromatography methods integrated with modified or unmodified paper/thread wicking channels. A variety of 2D and 3D designs of paper/thread platforms for zone electrophoresis, capillary electrophoresis, and modified/unmodified chromatography are discussed with emphasis on their limitation and improvements. The current progress in the signal amplification strategies such as isoelectric focusing, isotachophoresis, ion concentration polarization, isoelectric focusing, and stacking methods in paper-based devices are reviewed. Different strategies for chromatographic separations based on paper/thread will be explained. The separation of target species from complex samples and their determination by integration with other analytical methods like spectroscopy and electrochemistry are well-listed. Furthermore, the innovations for plasma and cell separation from blood as an important human biofluid are presented, and the related paper/thread modification methods are explored.
EB66 cell line, a duck embryonic stem cell-derived substrate for the industrial production of therapeutic monoclonal antibodies with enhanced ADCC activity

Abstract:Monoclonal antibodies (mAbs) represent the fastest growing class of therapeutic proteins. The increasing demand for mAb manufacturing and the associated high production costs call for the pharmaceutical industry to improve its current production processes or develop more efficient alternative production platforms. The experimental control of IgG fucosylation to enhance antibody dependent cell cytotoxicity (ADCC) activity constitutes one of the promising strategies to improve the efficacy of monoclonal antibodies and to potentially reduce the therapeutic cost. We report here that the EB66 cell line derived from duck embryonic stem cells can be efficiently genetically engineered to produce mAbs at yields beyond a 1 g/L, as suspension cells grown in serum-free culture media. EB66 cells display additional attractive grown characteristics such as a very short population doubling time of 12 to 14 hours, a capacity to reach very high cell density (> 30 million cells/mL) and a unique metabolic profile resulting in low ammonium and lactate accumulation and low glutamine consumption, even at high cell densities. Furthermore, mAbs produced on EB66 cells display a naturally reduced fucose content resulting in strongly enhanced ADCC activity. The EB66 cells have therefore the potential to evolve as a novel cellular platform for the production of high potency therapeutic antibodies.
Microfluidic Co-Flow of Newtonian and Viscoelastic Fluids for High-Resolution Separation of Microparticles.

Abstract:The microfluidic passive control of microparticles largely relies on the hydrodynamic effects of the carrier media such as Newtonian fluids and viscoelastic fluids. Yet the viscoelastic/Newtonian interfacial effect has been scarcely investigated, especially for high-resolution particle separation. Here we report a microfluidic co-flow of Newtonian (water or PBS) and viscoelastic fluids (PEO) for the size-dependent separation of microparticles. The co-flow condition generates a stable viscoelastic/Newtonian interface, giving rise to the wall-directed elastic lift forces that compete with the center-directed lift forces, and efficiently hinders the migration of microparticles from the Newtonian to the viscoelastic fluid in a size-dependent manner. An almost complete separation of a binary mixture of 1 μm and 2 μm polystyrene particles is achieved by the co-flow of water and a very dilute PEO solution (100 ppm), whereas the sole use of water or PEO could not lead to an efficient separation. This co-flow microfluidic system is also applied for the separation of Staphylococcus aureus (1 μm) from platelets (2-3 μm) with >90% efficiencies and purities.
Fabrication of A Su-8-Based Polymer-Enclosed Channel with A Penetrating Uv/Ozone-Modified Interior Surface for Electrokinetic Separation of Proteins

Abstract:This paper introduces electrokinetic separation inside fully cross-linked epoxy-based polymer channels that were batch modified on the inner surfaces using a penetrating UV/ozone treatment from the outside. The treatment can employ either a 254 nm UV source in an ozone-rich environment or a stand-alone 172 nm UV source to directly generate C=O hydrophilic functional groups on the embedded polymer channel wall surfaces. Short-wavelength UV radiation was employed to break polymer surface bonds inside the channel. Ozone generated directly from air or supplied externally oxidized the reaction site on the activated polymer surface to generate the desired functional groups. An epoxy-based photoresist compound, SU-8 (MicroChem, MA), which is widely used in microfluidic systems, was employed to demonstrate the surface modification. Fourier transform infrared spectroscopy (FTIR) and high resolution x-ray photoelectron spectroscopy (HRXPS) were employed to characterize the functional groups that formed after the UV/ozone surface modification and to confirm the formation of O-H functional groups from the phenol group covalently bonded to the SU-8 surface, attributed mostly to the surface hydrophilicity modification. Water contact angles on the modified surface ranged from 72 degrees to 12 degrees depending on the processing time, UV power and ozone concentration. These angles were retained for at least 4 weeks after the process. Finally, the inner wall surfaces of the SU-8-enclosed channels were successfully modified using this technology, and rapid water transportation and EOF pumping were visualized inside the channel after surface modification. Successful electrokinetic separation of 10 mM BSA and 10 mM anti-rabbit IgG labeled with FITC inside the channel was also carried out. The polymer channel revealed a surface charge density of 75% of the zeta potential on a microslide glass surface, indicating the potential for molecule separation using polymer channels instead of glass channels. This simple process provides a novel way to integrate surface modification into microfluidic structures after fabrication for fully polymer-based lab-on-a-chip systems.
A numerical study on microfluidic devices to maintain the concentration and purity of dielectrophoresis-induced separated fractions of analyte

Abstract:Determining the physical and chemical properties of biologically important particles such as cells, organelles, viruses, exosomes, complexes, nucleotides, and proteins is needed to understand their function. These properties are determined with common analytical tools (mass spectrometry, cryo-EM, NMR, various spectroscopies, nucleotide sequencing, etc.) whose function can be improved when samples are pure and concentrated. Separations science plays a central role in conditioning samples, ranging from low-resolution benchtop operations like precipitations or extractions to higher-resolution chromatography and electrophoresis. In the last two decades, gradient insulator-based dielectrophoresis (g-iDEP) has emerged as a high-resolution separation technique capable of highly selective enrichment of cells, viruses, exosomes, and proteins. Specific evidence has been shown that pure homogeneous and concentrated fractions of cells and exosomes can be generated from complex mixtures. However, recovering those fractions for analysis has not been developed, limiting the technique to an analytical rather than a preparative one. Here, a finite element analysis was undertaken to identify geometries and operational parameters to efficiently remove the enriched fraction while retaining maximum concentration and providing total mass transfer. Geometric factors (e.g., side channel width and distance from the gradient-inducing gap) were studied, along with the addition of a second inlet side channel. Two flow-generating mechanisms-electroosmosis and hydrostatic pressure-were evaluated for semi-optimized device designs, including a comparison of the one- and two-inlet designs. Simulations indicate effectively one hundred percent mass transfer and a concentration increase by an order of magnitude for several device configurations and operational parameters.

Abstract:A method based on the in situ polymerization of methyl methacrylate (MMA) has :been developed for the rapid fabrication of a novel separation platform, fiber electrophoresis microchip. To demonstrate the concept, prepolymerized MMA molding solution containing a UV initiator was sandwiched between a poly(methyl methacrylate) (PMMA) cover plate and a PMMA base plate bearing glycerol-permeated fiberglass bundles and was exposed to UV light. During the UV-initiated polymerization, the fiberglass bundles were embedded in the PMMA substrate to form fiberglass-packed microchannels. When the glycerol in the fiberglass bundles was flushed away with water, the obtained porous fiberglass-packed channels could be employed to perform electrophoresis separation. Scanning electron micrographs (SEMs) and microscopic images offered insights into the fiber electrophoresis microchip. The analytical performance of the novel microchip has been demonstrated by separating and detecting dopamine and catechol in connection with end-column amperometric detection. The fiber-based microchips can be fabricated by the new approach without the need for complicated and expensive lithography-based microfabrication techniques, indicating great promise for the low-cost production of microchips, and should find a wide range of applications.
Joule Heating-Induced Dispersion in Open Microfluidic Electrophoretic Cytometry

Abstract:While protein electrophoresis conducted in capillaries and microchannels offers high-resolution separations, such formats can be cumbersome to parallelize for single-cell analysis. One approach for realizing large numbers of concurrent separations is open microfluidics (i.e., no microchannels). In an open microfluidic device adapted for single-cell electrophoresis, we perform 100s to 1000s of simultaneous separations of endogenous proteins. The microscope slide-sized device contains cells isolated in microwells located in a ∼40 μm polyacrylamide gel. The gel supports protein electrophoresis after concurrent in situ chemical lysis of each isolated cell. During electrophoresis, Joule (or resistive) heating degrades separation performance. Joule heating effects are expected to be acute in open microfluidic devices, where a single, high-conductivity buffer expedites the transition from cell lysis to protein electrophoresis. Here, we test three key assertions. First, Joule heating substantially impacts analytical sensitivity due to diffusive losses of protein out of the open microfluidic electrophoretic (EP) cytometry device. Second, increased analyte diffusivity due to autothermal runaway Joule heating is a dominant mechanism that reduces separation resolution in EP cytometry. Finally, buffer exchange reduces diffusive losses and band broadening, even when handling single-cell lysate protein concentrations in an open device. We develop numerical simulations of Joule heating-enhanced diffusion during electrophoresis and observe ∼50% protein loss out of the gel, which is reduced using the buffer exchange. Informed by analytical model predictions of separation resolution (with Joule heating), we empirically demonstrate nearly fully resolved separations of proteins with molecular mass differences of just 4 kDa or 12% (GAPDH, 36 kDa; PS6, 32 kDa) in each of 129 single cells. The attained separation performance with buffer exchange is relevant to detection of currently unmeasurable protein isoforms responsible for cancer progression.

Abstract:This study addresses the challenge of separating bacteria with similar structures such as Escherichia coli and Aeromonas hydrophila . This approach employs pulsed field dielectrophoresis assisted by laminar flow fractionation in a lab-on-a-chip system with integrated optical detection. Bacterial cells passed through 30-μm microelectrodes subjected at 1 MHz and 14 V peak-to-peak in pulsed mode, while fluid flow carried bacteria towards the chamber's end. The on-and-off electric field at specific pulse intervals expose bacterial cells to diverse forces, including kinetics, dielectrophoresis, gravity, drag, and diffusion, resulting in a net force facilitating their movement. Variations of pulsing time, flow rates, and voltage were investigated to identify the optimal combination for efficient separation. Next, the bacteria were detected using an optical fibre based on their absorbance. Results demonstrated a 30% separation efficiency in 90 min at 9.6 μL min −1 flow rates, 4 s pulsing time, and 40 μS cm −1 medium conductivity. A. hydrophila aggregates experienced greater DEP force and retained at microelectrodes during electric field application compared to E. coli , which moved faster towards optical detection. The separation mechanism with and without electric field was different, and precise control of cell movement during field-off periods is important to minimize uncontrolled diffusion. While the optical detection part has been successful, longer time and separation length are recommended for better separation. A carefully tuned combination of pulsing time, flow rates, voltage, and microelectrode design is crucial for this integrated lab-on-chip system to be efficient for separating and detecting closely related microorganisms.

Abstract:Sorting and separation of microparticles is a challenging problem of interdisciplinary nature. Existing technologies can differentiate microparticles by their bulk properties, such as size, density, electric polarizability, etc. The next level of challenge is to separate particles that show identical bulk properties and differ only in subtle surface features, such as functionalization with ligands. In this work, we propose a technique to sort and separate particles and fluid droplets that differ in surface properties. As a dispersive medium, we use a nematic liquid crystal (LC) rather than an isotropic fluid, which allows us to amplify the difference in surface properties through distinct perturbations of LC order around the dispersed particles. The particles are placed in a LC cell with spatially distorted molecular orientation subject to an alternating current electric field. The gradients of the molecular orientation perform two functions. First, elastic interactions between these pre-imposed gradients and distortions around the particles separate the particles with different surface properties in space. Second, these pre-imposed patterns create electro-osmotic flows powered by the electric field that transport the sorted particles to different locations thus separating them. The demonstrated unique sorting and separation capability opens opportunities in lab-on-a-chip, cell sorting and bio-sensing applications.

Abstract:Electrophoretic separation of a fluorescent dye mixture, containing rhodamine B (RB) and fluorescein, in liquid foams stabilized by anionic, cationic, or non-ionic surfactants in water-glycerol mixtures was studied in a custom-designed foam separation device. The effects of the external electric field applied across the foam and the initial pH of the solution on the effectiveness of separation were also studied. The fluid motion due to electroosmosis and the resulting back pressure within the foam and local pH changes were found to be complex and affected the separation. Fluorescein dye molecules, which have a positive or negative charge depending on the solution pH, aggregated in the vicinity of an electrode, leaving a pure band of neutral dye RB. The effectiveness of the separation was quantified by the percentage width of the pure RB band, which was found to be between 29 and 42%. This study demonstrates the potential of liquid foam as a platform for electrophoretic separation.

Abstract:Micro free-flow electrophoresis (μFFE) provides a rapid and straightforward route for the high-performance online separation and purification of targeted liquid samples in a mild manner. However, the facile fabrication of a μFFE device with high throughput and high stability remains a challenge due to the technical barriers of electrode integration and structural design for the removal of bubbles for conventional methods. To address this, the design and fabrication of a high-throughput μFFE chip are proposed using laser-assisted chemical etching of glass followed by electrode integration and subsequent low-temperature bonding. The careful design of the height ratio of the separation chamber and electrode channels combined with a high flow rate of buffer solution allows the efficient removal of electrolysis-generated bubbles along the deep electrode channels during continuous-flow separation. The introduction of microchannel arrays further enhances the stability of on-chip high-throughput separation. As a proof-of-concept, high-performance purification of fluorescein sodium solution with a separation purity of ~97.9% at a voltage of 250 V from the mixture sample solution of fluorescein sodium and rhodamine 6G solution is demonstrated.

Tunable membrane-less dielectrophoretic microfiltration by crossing interdigitated electrodes

Abstract:Filtration is a crucial step in the analysis of living microparticles. In particular, the selective microfiltration of phytoplankton by size and shape remains an open problem, even though these criteria are essential for their gender and/or species identification. However, microfiltration devices necessitate physical membranes which complicate their fabrication, reduce the sample flow rate and can cause unwanted particle clogging. Recent advances in microfabrication such as electrode High Precision Capillary Printing allow to rapidly build electrode patterns over wide areas. In this study, we introduce a new concept of membrane-less dielectrophoretic (DEP) microfiltration suitable for large scale microfabrication processes. The proposed design involves two pairs of interdigitated electrodes at the top and the bottom of a microfluidic channel. We use finite-element calculations to analyse how the DEP force field throughout the channel, as well as the resulting trajectories of particles depend on the geometry of the system, on the physical properties of the particles and suspending medium and on the imposed voltage and flow rates. We numerically show that in the negative DEP regime, particles are focused in the channel mid-planes and that virtual pillars array leads either to their trapping at specific stagnation points, or to their focusing along specific lines, depending on their dielectrophoretic mobility. Simulations allow to understand how particles can be captured and to quantify the particle filtration conditions by introducing a critical dielectrophoretic mobility. We further illustrate the principle of membrane-less dielectrophoretic microfiltration using the proposed setup, by considering the separation of a binary mixture of polystyrene particles with different diameters, and validate it experimentally.

Preparative continuous flow electrophoretic instrumentation for purification of biological samples

Abstract:We constructed preparative instrumentation and developed the method that are based on separation of the samples by bidirectional isotachophoresis/moving boundary electrophoresis in continuous divergent flow. The described instrumentation can be used for variety of the samples, however, it can be easily optimized and tailored for the specific sample. The trapezoid separation bed from non-woven textile exhibited minimum adsorption effect for sample and it can be used repeatedly. By addition of different spacers via separation space inlets, the sections of pH gradient can be modified to enhance the separation. The liquid flow from two inlets positioned on each side of the sample inlet prevented the contact of the sample with anolyte and catholyte at the analysis beginning. One pair of thin electrodes (graphite and stainless-steel) was placed at the separation space output. The electrode products were washed out into drains without disturbing the focusing process. The influence of EOF was managed by tilting the separation bed in direction from cathodic to anodic side. The components of spirulina supernatant and color pI markers were separated in the pH gradient from 3.9 to 10.1. pH gradient was stable for at least 4.5 hours and spirulina supernatant from about 0.12 g of dry powder was processed. Compared to other preparative methods used for spirulina separation, the presented method/instrumentation working with continuous divergent flow had essential advantages. The efficient separation was fast, and no intermediate steps were necessary to obtain liquid fractions with separated components compatible with further biological experiments.This article is protected by copyright. Abstract:The concept of generating an oscillatory electroosmotic flux inside the porous particle to enhance the intra-particle mass transport was presented and a new kind of electrochromatography carried out in a five-compartment electrolyzer were developed. The adsorbent was packed in the central compartment, while the neighboring compartments were used as the elution compartments and the electrode compartments, respectively. Chromatographic separations of human serum albumin on Blue Sepharose Fast Flow, bovine serum albumin (BSA) on DEAE-Sepharose Fast Flow, and BSA on hydroxyapatite were carried out, respectively. The adsorption isotherms were shown to be independent of electric field, while the increase in the electric field strength resulted in a linear increase in the magnitude of electroosmotic flux and the improvement of the breakthrough behavior in all cases. The experiment results have demonstrated the effectiveness of the oscillatory electroosmosis in enhancing intra- and inter-particle mass transport and its high potential to large-scale chromatography.
Development of a gel monolithic column polydimethylsiloxane microfluidic device for rapid electrophoresis separation.

Abstract:A β-cyclodextrin (β-CD)-bonded gel monolithic column polydimethylsiloxane (PDMS) microfluidic device was developed in a simple and feasible way. Before preparation of gel monolithic column in PDMS microchannel, PDMS surface was activated by UV light to create silanol groups, which is an active molecule to covalently bond 3-(trimethoxysilyl)-propyl methacrylate (Bind-Silane) and seal microfluidic device. By the way, Bind-Silane is a bifunctional molecule to link polyacrylamide (PAA) gel and inner wall of PDMS microchannel covalently. Allyl-β-CD was used not only as a multifunctional crosslinker in PAA gel to control the size of the pores, but also as a chiral selector for the enantioseparation. The stability, transferring heat and optical characteristic of the microfluidic device were examined. The separation capability of the gel monolithic column was confirmed by the successful separation of fluorescein isothiocyanate (FITC)-labeled arginine (Arg), glutamine acid (Glu), tryptophan (Try), cysteine (Cysteine) and phenylalanine (Phe) in the PDMS microfluidic device less than 100s at 36mm effective separation length. A maximum of 2.06×105 theoretical plates was obtained by the potential strength of 490V/cm. A pair of FITC-labeled dansyl-d,l-threonine (Dns-Thr) was separated absolutely.

Abstract:Joule heating, arising from the electric current passing through the capillary, causes many undesired effects in CE that ultimately result in band broadening. The use of narrow-bore capillaries helps to solve this problem as smaller cross-sectional area results in decreased Joule heating and the rate of heat dissipation is increased by the larger surface-to-volume ratio. Issues arising from such small capillaries, such as poor detection sensitivity, low loading capacity and high flow-induced backpressure (complicating capillary loading) can be avoided by using a bundle of small capillaries operating simultaneously that share buffer reservoirs. Microstructured fibres, originally designed as waveguides in the telecommunication industry, are essentially a bundle of parallel ∼5 μm id channels that extend the length of a fibre having otherwise similar dimensions to conventional CE capillaries. This work presents the use of microstructured fibres for CZE, taking advantage of their relatively high surface-to-volume ratio and the small individual size of each channel to effect highly efficient separations, particularly for dye-labelled peptides.

Abstract:Conventional microfluidic devices are typically complex and expensive. The devices require the use of pneumatic control systems or highly precise pumps to control the flow in the devices. This work investigates an alternative method using paper based microfluidic devices to replace conventional microfluidic devices. Size based separation and extraction experiments conducted were able to separate free dye from a mixed protein and dye solution. Experimental results showed that pure fluorescein isothiocyanate could be separated from a solution of mixed fluorescein isothiocyanate and fluorescein isothiocyanate labeled bovine serum albumin. The analysis readings obtained from a spectrophotometer clearly show that the extracted tartrazine sample did not contain any amount of Blue-BSA, because its absorbance value was 0.000 measured at a wavelength of 590nm, which correlated to Blue-BSA. These demonstrate that paper based microfluidic devices, which are inexpensive and easy to implement, can potentially replace their conventional counterparts by the use of simple geometry designs and the capillary action. These findings will potentially help in future developments of paper based microfluidic devices.
