Abstract:The randomization design employed to gather the data is the basis for the exact distributions of the permutation tests. One of the designs that is frequently used in clinical trials to force balance and remove experimental bias is the truncated binomial design. The exact distribution of the weighted log-rank class of tests for censored cluster medical data under the truncated binomial design is examined in this paper. For p-values in this class, a double saddlepoint approximation is developed using the truncated binomial design. With the right censored cluster data, the saddlepoint approximation's speed and accuracy over the normal asymptotic make it easier to invert the weighted log-rank tests and find nominal 95% confidence intervals for the treatment effect.

Abstract:Clustered data frequently occur in biomedical research fields and clinical trials. The log-rank tests are widely used for two-independent samples of clustered data tests. The randomized block design and truncated binomial design are used for forcing balance in clinical trials and reducing selection bias. In this paper, survival clustered data are randomized by generalized randomized block, and subsequently clustered data in each block are randomized by truncated binomial design. Consequently, the p-values of the null permutation distribution of log-rank tests for clustered data are approximated via the double saddlepoint approximation method. Comprehensive numerical studies are carried out to assess the accuracy of the saddlepoint approximation. This approximation has a great accuracy over the asymptotic normal approximation.

Abstract:Clinical studies sometimes provide clustered data with censored failure times. A crucial factor of the randomized design that lessens selection bias is the random allocation rule. Given this, the weighted rank tests' p-values for stratified survival clustered sampling based on the random allocation rule are approximated using the double saddle-point approximation technique. For tests of significance and confidence intervals for the treatment effect, this approximation can be utilized. Through simulation experiments, the accuracy of the saddle-point approximation is examined by comparing saddle-point and normal approximations to the exact underlying permutation distribution.

Abstract:Motivated by Prentice's (1978) generalization of linear rank statistics for censored data, we introduce a similar kernel for deriving linear rank statistics for truncated data. They take very similar forms to those of Prentice's statistics. Moreover, we also show that they can be expressed equivalently in weighted log rank-type forms, thus providing natural variance estimators. Explicit forms corresponding to Wilcoxon and log rank tests are obtained. Extensions to data subject to both left truncation and right censoring are provided.

Abstract:The weighted log-rank class is the common and widely used class of two-sample tests for clustered data. Clustered data with censored failure times often arise in tumorigenicity investigations and clinical trials. The randomized block design is a significant design that reduces both unintentional bias and selection bias. Accordingly, the p-values of the null permutation distribution of weighted log-rank class for clustered data are approximated using the double saddlepoint approximation technique. Comprehensive simulation studies are carried out to appraise the accuracy of the saddlepoint approximation. This approximation exhibits a significant improvement in precision over the asymptotic approximation. This precision motivates us to determine the approximated confidence intervals for the treatment impact.

Abstract:Quantile regression for right‐ or left‐censored outcomes has attracted attention due to its ability to accommodate heterogeneity in regression analysis of survival times. Rank‐based inferential methods have desirable properties for quantile regression analysis, but censored data poses challenges to the general concept of ranking. In this article, we propose a notion of censored quantile regression rank scores, which enables us to construct rank‐based tests for quantile regression coefficients at a single quantile or over a quantile region. A model‐based bootstrap algorithm is proposed to implement the tests. We also illustrate the advantage of focusing on a quantile region instead of a single quantile level when testing the effect of certain covariates in a quantile regression framework. Résumé En analyse de survie pour données censurées à gauche ou à droite, la régression quantile a suscité beaucoup d'attention en raison de sa capacité à accommoder l'hétérogénéité dans le cadre d'analyses de régression. Les méthodes inférentielles fondées sur les rangs ont de nombreuses propriétés attrayantes pour des analyse de régression quantile, mais elles sont confrontées à des défis posés par le concept d'ordre en présence de censure. Pour contourner cette difficulté, les auteurs de ce travail proposent un concept de régression quantile à scores de rangs censurés. Cette approche permet de construire des tests de rangs pour les coefficients du modèle de régression quantile en question, que ce soit pour un seul quantile ou tout un ensemble de quantiles. L'implantation de tels tests repose sur un algorithme bootstrap fondé sur un modèle. Enfin, les auteurs illustrent l'avantage de cibler une région quantile plutôt qu'un quantile unique lorsqu'il est question de tester l'effet de certaines covariables dans un cadre de régression quantile.

Abstract:Recently linear rank statistics with censored data have been used as the estimating functions for the regression parameters in the linear model with an unspecified error distribution. The resulting rank estimators are consistent and asymptotically normal. However, the asymptotic variances of these estimators are complicated and are difficult to estimate well with censored data. In this paper, we propose some simple methods for making inference about a subset of the regression coefficients while regarding others as nuisance parameters. A lack-of-fit test for the linear model is also presented. The proposed procedures are illustrated with an example.

Abstract:A class of rank estimators is introduced for regression analysis in the presence of both left-truncation and right-censoring on the response variable. By making use of martingale theory and a tightness lemma for stochastic integrals of multiparameter empirical processes, the asymptotic normality of the estimators is established under certain assumptions. Adaptive choice of the score functions to give asymptotically efficient rank estimators is also discussed.

Abstract:The main idea of this paper is to approximate the exact p-value of a class of non-parametric, two-sample location-scale tests. In this paper, the most famous non-parametric two-sample location-scale tests are formulated in a class of linear rank tests. The permutation distribution of this class is derived from a random allocation design. This allows us to approximate the exact p-value of the non-parametric two-sample location-scale tests of the considered class using the saddlepoint approximation method. The proposed method shows high accuracy in approximating the exact p-value compared to the normal approximation method. Moreover, the proposed method only requires a few calculations and time, as in the case of the simulated method. The procedures of the proposed method are clarified through four sets of real data that represent applications for a number of different fields. In addition, a simulation study compares the proposed method with the traditional methods to approximate the exact p-value of the specified class of the non-parametric two-sample location-scale tests.

Abstract:This paper deals with a class of nonparametric two-sample location-scale tests. The purpose of this paper is to approximate the exact p-value of the considered class under a randomized block design. The exact p-value of the considered class is approximated by the saddlepoint approximation method, also by the traditional method which is the normal approximation method. The saddlepoint approximation method is more accurate than the normal approximation method in approximating the exact p-value, and does not take a lot of time like the simulation method. This accuracy is proved by applying the mentioned methods to two real data sets and a simulation study.

Abstract:A multivariate data analysis (MVDA) is a powerful statistical approach to simultaneously analyze datasets with multiple variables. Unlike univariate or bivariate analyses, which simultaneously focus on one or two variables, respectively, MVDA considers the interactions and relationships among multiple variables within a dataset. Several nonparametric tests can be used in the context of one-sample multivariate location problems. The exact distributions of such tests cannot be analytically computed and are usually approximated using an asymptotic approximation. This article proposes the saddlepoint approximation method to approximate the tail probability for multivariate sign and signed-rank tests. It is suggested as a more accurate alternative to the traditional asymptotic approximation method and an alternative to the simulation method. It requires a lot of time as it depends on all possible permutations. Real data examples were provided to illustrate the calculation of p-values, and a simulation study was conducted to compare the accuracy of the saddlepoint approximation method with the simulation method (permutation-based, so time-consuming) and an asymptotic normal approximation method. The study results show that the saddlepoint approximation provides highly accurate approximations to the p-values of the considered statistics, and it often outperforms the normal approximation. Additionally, the results show that the proposed method's computation time is much less than that of the time-consuming simulation method.

Abstract:Bivariate data are frequently encountered in many applied fields, including econometrics, engineering, physiology, biology, and medicine. For bivariate analysis, a wide range of non-parametric and parametric techniques can be applied. There are fewer requirements needed for non-parametric procedures than for parametric ones. In this paper, the saddlepoint approximation method is used to approximate the exact p-values of some non-parametric bivariate tests. The saddlepoint approximation is an approximation method used to approximate the mass or density function and the cumulative distribution function of a random variable based on its moment generating function. The saddlepoint approximation method is proposed in this article as an alternative to the asymptotic normal approximation. A comparison between the proposed method and the normal asymptotic approximation method is performed by conducting Monte Carlo simulation study and analyzing three numerical examples representing bivariate real data sets. In general, the results of the simulation study show the superiority of the proposed method over the asymptotic normal approximation method.

Abstract:Many statisticians resort to using the asymptotic normal approximation method to carry out statistical inference for many statistical tests, especially nonparametric ones. In this article, the saddlepoint approximation method is proposed as an alternative to the asymptotic normal approximation method to carry out statistical inference for a number of nonparametric tests for an important type of data that appears frequently in many clinical studies such as cancer and tumorigenicity studies. In clinical trials, there are many strategies through which treatments are assigned to patients. Equal allocation of both treatments is a largely prevalent approach in clinical trials to eliminate experimental bias and increase power. Accordingly, the statistical analysis is carried out based on the truncated binomial design, which is one of the designs that achieve a perfect balance between the two treatments. To clarify the accuracy of the proposed approximation method, two sets of real data are analyzed, and for the same purpose, a comprehensive simulation study is carried out.

Abstract:To compare the survival functions based on right-truncated data, Lagakos et al. proposed a weighted logrank test based on a reverse time scale. This is in contrast to Bilker and Wang, who suggested a semi-parametric version of the Mann-Whitney test by assuming that the distribution of truncation times is known or can be estimated parametrically. The approach of Lagakos et al. is simple and elegant, but the weight function in their method depends on the underlying cumulative hazard functions even under proportional hazards models. On the other hand, a semi-parametric test may have better efficiency, but it may be sensitive to misspecification of the distribution of truncation times. Therefore, this paper proposes a non-parametric test statistic based on the integrated weighted difference between two estimated survival functions in forward time. The comparative results from a simulation study are presented and the implementation of these methods to a real data set is demonstrated.

A Large Sample Study of Rank Estimation for Censored Regression Data

Abstract:Large sample approximations are developed to establish asymptotic linearity of the commonly used linear rank estimating functions, defined as stochastic integrals of counting processes over the whole line, for censored regression data. These approximations lead to asymptotic normality of the resulting rank estimators defined as solutions of the linear rank estimating equations. A second kind of approximations is also developed to show that the estimating functions can be uniformly approximated by certain more manageable nonrandom functions, resulting in a simple condition that guarantees consistency of the rank estimators. This condition is verified for the two-sample problem, thereby extending earlier results by Louis and Wei and Gail, as well as in the case when the underlying error distribution has increasing failure rate, which includes most parametric regression models in survival analysis. Techniques to handle the delicate tail fluctuations are provided and discussed in detail.
Exact and efficient multivariate two-sample tests through generalized linear rank statistics

Abstract:So-called linear rank statistics provide a means for distribution-free (even in finite samples), yet highly flexible, two-sample testing in the setting of univariate random variables. Their flexibility derives from a choice of weights that can be adapted to any given (simple) alternative hypothesis to achieve efficiency in case of correct specification of said alternative, while their non-parametric nature guarantees well-calibrated $p$-values even under misspecification. By drawing connections to (generalized) maximum likelihood estimation, and expanding on recent work on ranks in multiple dimensions, we extend linear rank statistics both to multivariate random variables and composite alternatives. Doing so yields non-parametric, multivariate two-sample tests that mirror efficiency properties of likelihood ratio tests, while remaining robust against model misspecification and computationally tractable. We prove non-parametric versions of the classical Wald and score tests facilitating hypothesis testing in the asymptotic regime, and relate these generalized linear rank statistics to linear spacing statistics enabling exact $p$-value computations in the small to moderate sample setting. Moreover, viewing rank statistics through the lens of likelihood ratios affords applications beyond fully efficient two-sample testing, of which we demonstrate three: testing in the presence of nuisance alternatives, simultaneous detection of location and scale shifts, and $K$-sample testing.

Abstract:The problem of sample size determination based on three commonly used non-parametric rank based tests, namely, one-sample Wilcoxon's rank sum test, two-sample's Wilcoxon's rank sum test, and the rank-based test for independence is studied. Explicit formulas for variabilities of the test statistics under the alternative hypotheses are derived. Consequently, close forms of power functions of these test statistics are obtained for sample size determination utilizing the concept of higher order polynominal equations. Simulation studies were performed to evaluate the finite samples performance of the derived sample size formulas. The results indicates that the derived methods work well with moderate sample size.
On two-sample testing for data with arbitrarily missing values

Abstract:We develop a new rank-based approach for univariate two-sample testing in the presence of missing data which makes no assumptions about the missingness mechanism. This approach is a theoretical extension of the Wilcoxon-Mann-Whitney test that controls the Type I error by providing exact bounds for the test statistic after accounting for the number of missing values. Greater statistical power is shown when the method is extended to account for a bounded domain. Furthermore, exact bounds are provided on the proportions of data that can be missing in the two samples while yielding a significant result. Simulations demonstrate that our method has good power, typically for cases of $10\%$ to $20\%$ missing data, while standard imputation approaches fail to control the Type I error. We illustrate our method on complex clinical trial data in which patients' withdrawal from the trial lead to missing values.

Abstract:A Kolmogorov–Smirnov-type test for the two-sample problem with left-truncated data is proposed. The asymptotic null distribution of the test statistic and its omnibus consistency are established. A bootstrap resampling plan to approximate the null distribution of the test statistic is introduced. The finite sample performance of the proposed test is investigated through simulations. Comparison to the well-known log-rank test and a real data illustration are included.
