Abstract:A unique spectrofluorimetric protocol has been conceived for octreotide (a synthetic peptide drug) quantitation in both its authentic form and its application to dosage form. The protocol has been established simply upon condensation of octreotide by ninhydrin / phenyl acetaldehyde reagent in buffered media (pH 6.2). An intense fluorescence product has been formed and quantified at 463 nm (390 nm for excitation). After optimization for various experimental conditions, a wide linear interval (0.2-4.0 µg/ml) has been used to construct the calibration curve with a determination coefficient (r<sup>2</sup>) of 0.9994, a slope ± SD of 81.147 ± 0.7985, and a highly sensitive detection and quantitation limits nearly equal to 0.066 and 0.2 µg/ml, respectively. A proposed protocol has been checked in accordance with ICH validation guidelines, which indicate good accuracy and high precision of the proposed method. Furthermore, this protocol could be perfectly applied for the quantitative estimation of octreotide in its ampoules with a high degree of accuracy and precision. As a result, a developed protocol is ideally appropriate for fast and simple octreotide quantitative estimation in quality control laboratories.

Abstract:In the present study, two drugs for treating acromegaly are investigated which are either synthetic growth hormone inhibiting hormone (GHIH) or dopamine receptor agonist. Octreotide (OCT) and bromocriptine (BCT) were quantified with a quick, simple, and sensitive spectrophotometric approach in their authentic forms and commercial dosage forms. This approach was based upon formation of ion pair complex between erythrosine B and investigated drugs in an aqueous buffered solution. For OCT determination the higher sensitivity was obtained using ΔA at 525 nm. On the other hand, BCT was estimated using the absorbance at 556 nm. Under optimal conditions for the reaction, construction of calibration curves were performed within concentration range of 0.4-4 µg ml-1 for OCT and 1-9 µg ml-1 for BCT with with low detection limits (0.094 and 0.235 µg ml-1) and low quantitation limits (0.29 and 0.71 µg ml-1) for OCT and BCT respectively. The developed approach was assessed for linearity, accuracy, precision, limits of detection, and limits of quantitation in compliance with ICH validation recommendations. The suggested approaches demonstrated good linearity, as evidenced by determination coefficients (r2) of 0.999 with a slope 0.14 for OCT and 0.9989 with a slope 0.06 for BCT. Additionally, the environmental friendliness of the investigated method was assessed with some of the recent green analytical chemistry metrics.

Abstract:The present study is focused on the sensitive determination of newly FDA-approved glucagon-like-peptide agonists semaglutide (SEM) and tirzepatide (TIR). Direct, selective and label-free spectrofluorometric method was proposed and validated (according to ICH guidelines) for determination SEM and TIR in their pure form, newly approved pharmaceuticals and spiked human plasma. The developed method was based on measuring the native fluorescence of SEM and TIR in ethanol at 294.8 and 303 nm after being excited at 216 and 225 nm for SEM and TIR in order. The method sensibility allowed the quantification of both drugs in nano-scale up to 10 ng/mL. Several experimental variables including solvent type, surfactant, and pH were optimized after several attempts to get the best sensitivity for both drugs. The mean recovery percentage of SEM was compared and found in agreement with the reported method using student's t -test and the variance ratio F-test. Additionally, the greenness and whiteness profiles for this approach were evaluated using the GAPI, AGREE, and RGB algorithm; the positive results supported its use as great candidates for successful implementation in quality control labs and the pharmaceutical analysis companies.

Abstract:Octreotide as a synthetic cyclic octapeptide is a somatostatin analog with longer half-life and more selectivity for inhibition of the growth hormone. The acetate salt of octreotide is currently used for medical treatment of somatostatin-related disorders such as endocrine and carcinoid tumors, acromegaly, and gigantism. Octreotide contains both cysteine and tryptophan residues which make the hydrolysis part of its amino acid analysis procedure very challenging. The current paper introduces a fast and additive-free method which preserves tryptophan and cysteine residues during the hydrolysis. Using only 6 M HCl, this hydrolysis process is completed in 30 min at 150 °C. This fast hydrolysis method followed by pre-column derivatization of the released amino acids with 4-N,N-dimethylaminoazobenzene-4ʹ-sulfonyl chloride (DABS-Cl) which takes only 20 min, makes it possible to do the complete amino acid analysis of an octreotide sample in a few hours. The highly stable-colored DABS-Cl derivatives can be detected in 436 nm in a reversed phase chromatographic system, which eliminates spectral interferences to a great extent. The amino acid analysis of octreotide acetate including hydrolysis, derivatization, and reversed phase HPLC determination was validated according to International Conference of Harmonization (ICH) guidelines.

The joint use of synchronous spectrofluorimetry with second derivative mathematical treatment is for the first time to be developed and optimized in aid of using fluorescence data manager software generating second derivative peak amplitudes at 556.5 nm for ANT and 516.7 nm for TET. Linear responses have been represented over a wide range of concentration (0.5–12.0 μg/mL for ANT and 0.5–10.0 μg/mL for TET). Additionally, statistical comparison of the developed method with the official ones has been carried out where no significant difference was found. Additionally, greenness profile assessment was accomplished by means of four metric tools. Indeed, the method developed is found to be precise, sensitive, and discriminating to assess the cited drugs for regular analysis.

Abstract:A newly green method for the sensitive quantification of cloperastine, a cough suppressant, in spiked human plasma and its pharmaceutical formulation was designed for the first time. The established method depends on the enhancement of the weak fluorescence of cloperastine using 50 mM sulfuric acid to impair the photoinduced electron transfer produced from the nitrogen atom of piperidine moiety in cloperastine. This full protonation in an acid medium leads to an enhancement in the fluorescence of cloperastine, permitting its linear determination from 0.2 to 5.0 µg/mL with LOD and LOQ of 0.04 and 0.13 µg/mL, respectively. Moreover, the studied drug was estimated in its pharmaceutical market formulations as well as spiked human plasma. Furthermore, the greenness of the described method was evaluated.

Abstract:Green spectrophotometric and HPLC methods have been developed for the quantification of metoclopramide. In the spectrophotometric method, it was determined by direct absorbance measurement at 273 nm wavelength using ultrapure water as solvent. The Extend C18 column was used for the HPLC method. The mobile phase system consisted of a combination of ethanol and formic acid solution (pH 2.0; 30:70 v/v). Isocratic elution was applied and the flow rate was set at 1.0 mL min −1 . Metoclopramide was detected at 273 nm. The methods performed were economical, rapid, environmentally friendly, and simple, providing metoclopramide analysis within 5 min. The methods have been successfully applied in pharmaceutical products without matrix interference. The results of the application of the developed methods to pharmaceutical products were statistically compared and no significant difference was observed between the methods. In addition, the greenness assessment of the developed methods was performed using AGREE software. Our developed methods, based on the use of solvents such as ethanol and water, are proposed as a more environmentally and analyst-friendly option for the quantification of metoclopramide in pharmaceutical products than other methods currently in use.

Abstract:A green, simple, rapid, selective, and highly sensitive fluorimetric protocol has been established for quantitative analysis of the antibacterial drug moxifloxacin in its pure form and pharmaceutical dosage form. Owing to photoinduced electron transfer, moxifloxacin exhibits low native fluorescence in neutral media. Based on that, the developed fluorimetric protocol depends on inhibiting the photoinduced electron transfer effect of the nitrogen atom presented on the pyrrolidine ring in moxifloxacin by suitable adjusting of pH of the medium surrounding it, leading to its protonation. It is simply achieved by using 0.5 M acetic acid. This protonation enhances the native fluorescence of moxifloxacin, turning it into a stronger one. This fluorescence was measured at 498 nm after excitation at 295 nm with a linearity range from 10 to 60 ng mL-1 and a high correlation coefficient value of 0.9998. The fluorimetric approach could be applied for moxifloxacin detection with a limit of 0.89 ng mL-1 and a quantification limit of 2.69 ng mL-1. The developed method has been validated according to ICH guidelines, indicating high accuracy and excellent precision. Furthermore, the developed fluorimetric protocol was applied successfully for moxifloxacin analysis in pharmaceutical eye drops. As a result, the proposed protocol could be easily applied for quality control of moxifloxacin in different laboratories all over the world.

Abstract:Currently the development of technologies for labeling somatostatin with technetium-99m for diagnosing radionuclide neuroendocrine tumors is under way. Somatostatin analogues are binded with technetium99m only by the preliminary addition of a chelating agent. Therefore, it is important to develop a method for preparation of an octreotide derivative by modifying octreotide with precursors: ligands with high chelating ability for its tight binding with technetium-99m. ω-Bis(pyridin-2-ylmethyl)amino)aliphatic acids can be used successfully as such precursors. The purpose of the study was to develop a method for obtaining a new octreotide derivative for diagnosing neuroendocrine tumors. Materials and methods . The somatostatin octreotide analogue was used as the object of the study; succinimid-1-yl 6-(bis(pyridin-2-ylmethyl)amino)hexanoate was used as a chelating agent. Methods of high performance liquid chromatography and mass spectrometry were used to separate and analyze the synthesized compounds. Results . A method to produce an original octreotide derivative using a succinimid-1-yl 6-(bis(pyridin2-ylmethyl)amino)hexanoate as a chelating agent was proposed. The conditions of analytical and semipreparative HPLC for the analysis and purification of the active octreotide derivative (a monosubstituted derivative of the amino acid residue of D-phenylalanine) were suggested. Conclusion . The synthesized derivative of octreotide has a chelating center for strong binding to technetium-99m in its structure, which can be useful for diagnosing neuroendocrine tumors.

Abstract:The current study introduces the first analytical approach for the quantitative analysis of adipoRon, a synthetic adiponectin receptor agonist, using a facile, rapid, sensitive, and green spectrofluorimetric method. Nitrogen-doped carbon quantum dots (N-CQDs) were synthesized by adopting a fast microwave-assisted procedure using accessible and natural substrates (orange juice and urea). The suggested synthetic method produced CQDs with narrow particle size distribution (2–10 nm), high fluorescence stability, and good quantum yield (29.3%). The produced fluorescent QDs were used as fluorescent sensors and were quantitatively quenched by increasing adipoRon concentrations. A two-level full factorial design (2 2 FFD) was used to identify the most critical experimental factors affecting the relative fluorescence intensity of N-CQDs. The fluorescence quenching of N-CQDs by the drug was measured at 417 nm after excitation at 325 nm. Under the optimized conditions, the quenching of N-CQDs' fluorescent intensity displayed acceptable linearity with the adipoRon concentration range of 5.0–20.0 μg/mL with limits of quantitation and detection of 0.843 μg/mL and 0.278 μg/mL, respectively. Owing to the high sensitivity, the suggested method was efficiently applied for adipoRon estimation in different biological matrices, including spiked human plasma, real rat plasma, and liver homogenate samples with high % recoveries (87.41–103.17%) and low %RSD values (<2.87%). The method's greenness was evaluated using two metric tools, the complementary green analytical procedure index (ComplexGAPI) and the Analytical GREENNESS metric approach (AGREE), and the results showed that the method was excellent green. The proposed methodology was validated according to the ICHQ2 (R1) recommendations.

The parallel of our results for type I diabetes with those for ankylosing spondylitis is striking. In both there are strong associations with particular HLA markers and a significant increase in the proportion of non-secretors. This is further indirect evidence to support the suggestion that an infectious agent is implicated in the initiation of type I diabetes in some genetically predisposed paiients. Determining the secretor state of patients who have various viral infections would be a useful first step in investigating this hypothesis. We suggest that the initial susceptibility to some viral infections might be greater in the non-immune, non-secretor host. The specific immune response to the putative infectious agent, controlled by the class II HLA DR3/4 genes, might contribute to pathogenic sequelae leading to the diabetic state.

Abstract:Pemetrexed is a chemotherapeutic medicine, under the trade name Alimta. Malignant pleural mesothelioma patients. Its application in lung cancer has been studied. Here in, a second spectrofluorimetric method was advanced for quantifying of Pemetrexed in pharmaceutical formulation and spiked human plasma. That method depends on fluorescence derivatization of Pemetrexed with 4-chloro-7-nitrobenzo-2-oxa-1,3-diazole (NBD-Cl) at 75 °C in a (pH 9) of borate buffer to produce a fluorescent derivative which can be detected at 520 nm afterwards excitation at 460 nm. The method has been validated using ICH criteria, and it demonstrated linearity in a range of 2–120 ngmL-1. The proposed method was applied precisely and accurately for quantifying Pemetrexed within pharmaceutical formulation and spiking human plasma without any interferences. Moreover, the method's sustainability was evaluated and compared to the published method using two greenness assessment tools termed analytical eco-scale and Analytical GREENness (AGREE). That findings suggest that the method is more sustainable than the published method.

Abstract:In this study, an environmentally sustainable fluorimetric method for determination of Vonoprazan fumarate (VON) in dosage forms using nanoprobes consisting of nitrogen and sulfur co-doped carbon quantum dots (N, S-CQDs). The N, S-CQDs were prepared through a microwave-assisted method in 30 s. The resulting N, S-CQDs were characterized using transmission electron microscopy (TEM), high-resolution transmission electron microscopy (HRTEM), Fourier transform infrared spectroscopy (FTIR), and X-ray photoelectron spectroscopy (XPS). They exhibit fluorescence emission at 460 nm after excitation at 385 nm with a high quantum yield (60%). The analytical approach for VON determination relies on the quenching effect exerted by VON on the native fluorescence intensity of CQDs. The quenching mechanism was investigated using Stern-Volmer plots. The proposed method demonstrates linearity across a concentration range 10-80 μM (4.6-36.8 μg/mL) with corresponding limits of detection and quantitation calculated as 2.17 μM (0.99 μg/mL) and 6.58 μM (3.02 μg/mL), respectively. The method has been effectively utilized for the determination of VON in the pharmaceutical samples. Statistical comparison with reported RP-HPLC has been performed to verify the accuracy and precision of the developed method. The environmental sustainability of the developed method has been thoroughly examined through various greenness metrics.

Abstract:An eco‐friendly first derivative synchronous fluorescence approach was developed and validated for the determination of Donepezil HCl and trazodone HCl in their pure state, pharmaceutical dosage form and spiked human plasma. The study's objective is to establish an eco‐friendly, sensitive and economical quantitative methodology for the concurrent analysis of donepezil HCl (DPZ) and trazodone HCl (TRZ) in raw materials, tablets and human plasma. The first derivative synchronous fluorescence spectroscopic (FDSFS) technique was applied at constant wavelength difference (∆λ = 120) for assessment of DPZ and TRZ at each other's zero‐crossing point at 279 nm and 297 nm, respectively. The submitted technique was validated in accordance with ICH Q2 R1 guidelines and the linearity of the standard calibration curve was observed over the concentration range of 10–500 ng/ml for DPZ and 20–1,000 ng/ml for TRZ. The detection limits (LOD) were found to be 2.65 and 5.4 ng/ml, and the limits of quantitation (LOQ) were 8.05 and 16.3 ng/ml for DPZ and TRZ, respectively. This technique was used further to quantify the studied medications in their laboratory‐prepared mixtures, commercial tablets and spiked plasma samples. The results obtained were not significantly different from those acquired from the comparison methods, indicating the high accuracy and precision of the proposed method. Furthermore, the ecological friendliness of the suggested method was evaluated and proven to be excellent using Green Analytical Procedure Index (GAPI) and Analytical GREEnness (AGREE) evaluation tools.

Abstract:In this study, the simultaneous determination of bilastine and montelukast, two recently approved co-formulated antihistaminic medications, was accomplished using a quick, sensitive, environmentally friendly, and reasonably priced synchronous fluorescence spectroscopic approach for the first time. Enhancement of the method's sensitivity down to nanogram levels was achieved by the addition of sodium dodecyl sulfate (1.0% w/v) as a micellar system. According to the results, bilastine and montelukast's fluorescence was measured at 255.3 and 355.3 nm, respectively, using Δλ of 40.0 nm and distilled water as a green diluting solvent. With respect to the concentration ranges of bilastine (5.0-300.0 ng/ml) and montelukast (50.0-1000.0 ng/ml), the method showed excellent linearity (r ≥ 0.9998). The results showed that the suggested method is highly sensitive, with detection limits of 1.42 and 13.74 ng/ml for bilastine and montelukast, respectively. Within-run precisions (intra- and interday) per cent relative standard deviations (RSD) for both analytes were <0.59%. With high percentage recoveries and low percentage RSD values, the designed approach was successfully applied for the simultaneous estimation of the cited medications in their dosage form and human plasma samples. To evaluate the green profile of the suggested method, an analytical GREENNESS metric approach (AGREE) and green analytical procedure index (GAPI) metric tools were used. These two methods for evaluating greenness confirmed that the developed method met the highest number of green requirements, recommending its use as a green substitute for the routine analysis of the studied drugs. The proposed approach was validated according to ICHQ2 (R1) guidelines.

Abstract:This work introduces the first spectrofluorimetric approach for the estimation of lactoferrin, depending on the measurement of its native fluorescence, without the need for complicated instrumentation or large volumes of organic solvents. The proposed method was successfully applied for the determination of lactoferrin in different dosage forms and milk powder with high % recoveries. The excellent greenness profile of the developed method was confirmed using two greenness assessment tools: GAPI and AGREE. The current study presents the first spectrofluorimetric approach for the estimation of lactoferrin, depending on the measurement of its native fluorescence at 337 nm after excitation at 230 nm, without the need for any hazardous chemicals or reagents. It was found that the fluorescence intensity versus concentration calibration plot was linear over the concentration range of 0.1–10.0 μg/mL with quantitation and detection limits of 0.082 and 0.027 μg/mL, respectively. The method was accordingly validated according to the ICH recommendations. The developed method was applied for the estimation of lactoferrin in different dosage forms, including capsules and sachets with high percent recoveries (97.84–102.53) and low %RSD values (<1.95). Lactoferrin is one of the key nutrients in milk powder and a significant nutritional fortifier. In order to assess the quality of milk powder, it is essential to rapidly and accurately quantify the lactoferrin content of the product. Therefore, the presented study was successfully applied for the selective estimation of lactoferrin in milk powder with acceptable percent recoveries (96.45–104.92) and %RSD values (≤3.607). Finally, the green profile of the method was estimated using two assessment tools: Green Analytical Procedure Index (GAPI) and Analytical GREEnness (AGREE), which demonstrated its excellent greenness.

Abstract:The goal of the current research was to establish a quick and practical fluorometric technique for trifluridine analysis. The approach relied on the drug's complex formation with the zinc ion to produce a high‐fluorescence product. The fluorescence was further enhanced by adding sodium dodecyl sulfate, and it was observed at 450 nm following excitation at 400 nm. With a determination coefficient of 0.9994, the association between emission intensity and trifluridine concentration was linear between 1 and 125 ng mL−1. The quantitation limit was 0.987 ng mL−1 while 0.333 ng mL−1 was the detection limit. The buffer type, pH and concentration, type of surfactant and concentration, and finally the diluting solvent were among the reaction conditions that were closely examined. With great precision and reliability, the drug in question was quantified using this method in dosage formulations. The proposed method's level of greenness was assessed using two methodologies: the analytical greenness metric (AGREE) and the Green Analytical Procedure Index (GAPI).
