Abstract:Letrozole is an anticancer medication prescribed for the management of estrogen receptor-positive breast cancer in postmenopausal women. Chronic pain is prevalent in patients receiving chemotherapy, leading to the use of adjuvant analgesics such as tramadol. This work introduces the first analytical approach for the concurrent quantification of letrozole and tramadol, two co-administered drugs, employing a rapid, highly sensitive, eco-friendly, and cost-effective first derivative synchronous spectrofluorimetric technique. The fluorescence of tramadol and letrozole was measured at wavelengths of 235.9 nm and 241.9 nm, respectively using a wavelength difference (Δλ) of 60.0 nm. The developed approach demonstrated exceptional linearity (r ˃ 0.999) within the specified concentration ranges for tramadol (10.0–1200.0 ng/mL) and letrozole (1.0–140.0 ng/mL). The results demonstrated that the proposed technique exhibits a high level of sensitivity, with detection limits of 0.569 and 0.143 ng/mL for tramadol and letrozole, respectively, indicating the good bioanalytical applicability. The within-run precisions, both intra-day and inter-day, for both analytes, were less than 0.71 % RSD. The developed approach was effectively applied to simultaneously estimate the mentioned drugs in their tablets and human plasma samples, achieving high percentage recoveries and low % RSD values. In order to assess the environmental sustainability of the developed approach, Analytical GREEnnessNNESS (AGREE) and the Green Analytical Procedure Index (GAPI) metric tools were employed. Both tools revealed that the developed approach is excellent green, suggesting its usage as an environmentally-friendly alternative for the routine assay of the investigated pharmaceuticals. The developed approach was validated according to the ICHQ2 (R1) requirements.

Abstract:Duvelisib (DUV) is a potent anticancer drug whereas Moxifloxacin (MOX) is an antimicrobial drug with anti-proliferative potency against cancerous cells, which is empirically administered in cancer treatment. DUV and MOX combination is commonly prescribed to combat infections in patients while they are under chemotherapy treatment. This study describes, for the first time, the development of a simple and green synchronous spectrofluorimetric (SSF) method for the simultaneous estimation of DUV and MOX in plasma. DUV and MOX were quantified at 273 and 362 nm, respectively without interference between each other at Δλof 120 nm. The experimental variables influencing fluorescence intensities were thoroughly investigated and the optimum conditions were established. At pH 3.5, the optimum synchronous fluorescence intensity (SFI) was achieved in water solvent by using sodium acetate buffer solution. Calibration curves for DUV and MOX, correlating the SFI with the corresponding drug concentration, were linear in the range of 50-1000 ng mL-1for both drugs, with good correlation coefficients. The method was extremely sensitive, with limits of detection of 24 and 22 ng mL-1, and limits of quantitation of 40 and 45 ngmL-1for DUV and MOX, respectively. The SSF method was validated according to the Food and Drug Administration (FDA) guidelines for validation of analytical procedures, and the validation parameters were acceptable. The proposed SSF method was applied to the pharmacokinetic and bioavailability studies in rats' plasma after single concurrent oral administration of both drugs. The results of the study revealed that caution should be taken with DUV dose when concurrently administered with MOX. The greenness of SSF method was assessed by three different metric tools namely Analytical Eco-scale, Green Analytical Procedure Index, and Analytical Greenness Calculator. The results confirmed that SSF method is an eco-friendly and green analytical approach. In conclusion, the proposed SSF method is a valuable tool for pharmacokinetic/bioavailability studies and therapeutic drug monitoring of simultaneously administered DUV and MOX.

Abstract:The combination of Curcumin (CRN), resveratrol (RSV), and quercetin (QRN) has significant antioxidant effects and is found to be more effective than a single polyphenol. Spectrophotometric methods are considered one of the most common analytical techniques for the determination of the drugs due to their sensitivity, rapidness, low cost, and reproducibility. Therefore, the presence of new, and simple methods for the determination of such compounds will be highly valuable, specially in the presence of spectral overlap. In this research, five different facile spectrophotometric methods were investigated for the simultaneous determination of that ternary mixture for the first time, including zero order (I), first derivative (II), ratio difference double divisor (III), first derivative ratio spectra (IV), and mean centering (V) methods. The designed approaches were linear over the concentration ranges of (1.0–10.0), (0.5–8.0), and (1.0–14.0) μg/mL, respectively for curcumin, resveratrol, and quercetin. The different methods were then validated as stated by the International Council of Harmonization. The accuracy and precision have been evaluated by statistical analysis including student t -test, variance ratio F-test, and ANOVA. Moreover, the greenness and whiteness of the proposed methods were assessed to ensure the adherence to the greenness characters.

Abstract:A specific, sensitive and rapid liquid chromatography tandem mass spectrometry (LC-MS/MS) method has been developed and validated for simultaneous quantification of doxorubicin and curcumin in rat plasma after intravenous administration. The analytes of doxorubicin and curcumin were extracted with methanol precipitation using glibenclamide as internal standard (IS). The chromatographic separation was performed on a C18 column with acetonitrile and 0.1% formic acid water as mobile phase and with gradient elution at a flow rate of 0.2 mL/min. Calibration curves were linear over the ranges of 2-8000 ng/mL for doxorubicin and 5-2000 ng/mL for curcumin (r > 0.99). The lower limit of quantification (LLOQ) was 2 ng/mL for DOX and 5 ng/mL for Cur. Finally, this developed method was successfully applied in the pharmacokinetic study of doxorubicin and curcumin in rats and evaluated the effects of curcumin on the absorption of doxorubicin after intravenous administration.
Simultaneous determination of diosmin and hesperidin in combined pharmaceutical preparation by synchronous fluorescence spectrofluorimetric method

Abstract:A sensitive, rapid and green synchronous spectrofluorimetric method was developed to simultaneously analyze a binary mixture of diosmin (DSM) and hesperidin (HSP). The RSFI of both medications was measured in methanol at Δλ of 100 nm. The results indicated that specific experimental factors had an impact on these intensities. The optimization and thorough examination of these parameters were conducted. The plots of synchronous fluorescence intensity-concentration for DSM and HSP were found to be linear within the concentration ranges of 0.5-5.0 µg ml-1 and 0.2-3.0 µg ml-1, respectively. The detection limits for DSM and HSP were 0.107 µg ml-1 and 0.048 µg ml-1, respectively. The limits of quantification were 0.323 µg ml-1 and 0.144 µg ml-1 for DSM and HSP, respectively. The method outlined in this study was successfully used to determine the quantities of both drugs present in commercially available mixed tablets. The results obtained using this method were subsequently compared to those of a comparison method. Greenness assessment of the suggested procedure was accomplished by applying the GAPI method. Consequently, the recommended method can be used in the routine quality control analysis of the two cited drugs with minimum harmful effect on the environment as well as the individuals.

Abstract:Recently, a brand-new combination of mirabegron and solifenacin succinate received approval for managing the overactive bladder syndrome's bothersome symptoms. Herein, we introduce six validated UV-spectrophotometric approaches relying on simple mathematical calculations for rapid and concurrent analysis of MBR and SOS in their synthetic mixtures, commercial tablets, and human urine without preliminary separation. The proposed approaches were dual-wavelength, induced dual-wavelength, Fourier self-deconvolution, area under the curve, ratio difference, and first derivative ratio methods. These approaches were applied successfully for the concurrent determination of MBR and SOS in their mixtures (suffered from severe spectral overlap) with excellent recoveries without interference from pharmaceutical excipients or biological matrices. The concurrent analysis of MBR and SOS in spiked biological fluids was accomplished for the first time in this study using the Fourier approach. The proposed approaches were linear in ranges of (50–450) μg/mL and (70–1200) μg/mL for MBR and SOS, in order. The proposed approaches' greenness was evaluated using two tools namely; penalty points scoring system and green analytical procedure index (GAPI), revealing their superiority in greenness over the reported LC method. The proposed approaches were compared statistically in terms of t- and F-values with the reported LC method, revealing no discernible differences concerning accuracy or precision at p = 0.05. Accordingly, the proposed approaches are valid for regular analysis of MBR and SOS as affordable substitutes in QC labs lacking expensive chromatographic instruments.

Abstract:In clinical treatment, the analytical quality assessment of the delivery of chemotherapeutic preparations is required to guarantee the patient’s safety regarding the dose and most importantly the appropriate anticancer drug. On its own, the development of rapid analytical methods allowing both qualitative and quantitative control of the formulation of prepared solutions could significantly enhance the hospital’s workflow, reducing costs, and potentially providing optimal patient care. UV-visible spectroscopy is a nondestructive, fast, and economical technique for molecular characterization of samples. A discrimination and quantification study of three chemotherapeutic drugs doxorubicin, daunorubicin, and epirubicin was conducted, using clinically relevant concentration ranges prepared in 0.9% NaCl solutions. The application of the partial least square discriminant analysis PLS-DA method on the UV-visible spectral data shows a perfect discrimination of the three drugs with a sensitivity and specificity of 100%. The use of partial least square regression PLS shows high quantification performance of these molecules in solution represented by the low value of root mean square error of calibration (RMSEC) and root mean square error of cross validation (RMSCECV) on the one hand and the high value of R -square on the other hand. This study demonstrated the viability of UV-visible fingerprinting (routine approach) coupled with chemometric tools for the classification and quantification of chemotherapeutic drugs during clinical preparation.

Abstract:The cutting-edge combination of fluvoxamine (FVM) and ivermectin (IVM) has been presented as a proposed dosage form for the treatment of COVID-19 infections in early diagnosed patients. The main objective of this work is to develop simple, sensitive, and efficient methods for the synchronous quantification of FVM and IVM without any prior separation. Four green UV-methods were employed for the synchronous quantification, namely: Fourier functions convolution of absorption spectra, FFAS, Fourier functions convolution of derivative spectra of absorption curves, FFDS, Fourier function convolution of ratio spectra of absorption curves, FFRS and the dual-wavelength method, DWM. FFRS and DWM approaches can be able to reconcile the two components' significantly interfering spectrum presented in this commixture. Good linearity was checked in the range of 5–40, and 2.5–25 μg/mL for the FVM, and IVM, respectively. All approaches developed have been recommended in compliance with ICH principles. Furthermore, the approaches' greenness was predestined by "National Environmental Method Index" (NEMI), "Analytical GREEnness metric (AGREE)", the "Analytical Eco-Scale", and the "Green Analytical Procedure Index" (GAPI). In addition, spider diagram was utilized for the assessment of the greenness index of the solvent used. Beside greenness, the sustainability of our methods was investigated using the HEXAGON tool. Continuing the constant pursuit of greenness, drug-drug interactions (DDIs) between FVM & IVM were predicted by insilico tools to ensure the safety of the suggested mixture as a preliminary step before invitro and in vivo studies. Because they were deemed sustainable, affordable, and successful, the suggested UV-methods may be used for routine quality control investigations of the indicated formulations FVM & IVM.

Abstract:First derivative synchronous spectrofluorimetric analysis of two binary mixtures containing duloxetine with either avanafil or tadalafil. Application to pharmaceutical dosage forms and human plasma is achieved with excellent recoveries. Eco‐friendly approaches using ethanol as a green solvent and an assessment of the greenness of the methods are carried out. Antidepressants can cause sexual dysfunction side effects, necessitating the co‐administration of phosphodiesterase type 5 inhibitors. The simultaneous determination of these drugs in biological fluids is critical for therapeutic drug monitoring. For the first time, two binary mixtures containing duloxetine with either avanafil or tadalafil were estimated utilizing simple green spectrofluorimetric methods without the need for a previous separation step. The study was based on first derivative synchronous spectrofluorimetry in ethanol using a change in wavelength difference (∆λ) of 20 and 25 nm for the first and second combinations, respectively. Duloxetine and avanafil were estimated at 297.7 and 331 nm in their binary mixture, while duloxetine and tadalafil were determined at 290.3 and 297.7 nm, respectively. The linearity was achieved over the ranges of 0.1–1.5 μg mL−1 for both duloxetine and avanafil and 0.01–0.40 μg mL−1 for tadalafil, with limits of detection of 0.013, 0.022, and 0.004 μg mL−1 for duloxetine, avanafil, and tadalafil, respectively. Successful application of the developed approaches was accomplished for the estimation of the two mixtures in dosage forms as well as human plasma with excellent percentage recoveries (96–103.75% in plasma), which supports their suitability for use in quality control laboratories and pharmacokinetic studies. Moreover, the adopted approaches' greenness was evidenced by applying three tools.

Abstract:A specific and sensitive LC-MS/MS with protein precipitation- ultrasonic breaking method has been developed and validated for simultaneous determination of doxorubicin (DOX) and curcumin (Cur) in DOX and Cur co-loaded hyaluronic acid-vitamin E succinatemicelles [(DOX+Cur)-polymeric micelles (PMs)] in subcellular compartments of resistant MCF-7/Adr cells. Sequential extraction of four subcellular protein fractions (cytosolic, membrane/organelle, nucleic and cytoskeleton) was performed directly from MCF-7/Adr cells after incubation with (DOX+Cur)-PMs. An ultrasonic breaking-methanol precipitation method was used for extraction of the fractions, and the micelle breaking efficiency with methanol was 98.1 and 97.6% for DOX and Cur, respectively. The analytes were analyzed using positive electrospray ionization coupled with multiple reaction monitoring. The calibration curves were linear over a concentration range of 0.5-400ng/mL for DOX and 2-2000ng/mL for Cur, and the recovery for the two analytes were >85% with negligible matrix effect. The intra-day and inter-day precision was <10.80% and relative error was within +/- 7.70%. The developed method was successfully applied for subcellular determination of DOX and Cur in MCF-7/Adr cells. Moreover, Cur and (DOX+Cur)-PMs had a marked promoting effect on the distribution of DOX in the nucleic protein fraction.

Abstract:Pemetrexed is a chemotherapeutic medicine, under the trade name Alimta. Malignant pleural mesothelioma patients. Its application in lung cancer has been studied. Here in, a second spectrofluorimetric method was advanced for quantifying of Pemetrexed in pharmaceutical formulation and spiked human plasma. That method depends on fluorescence derivatization of Pemetrexed with 4-chloro-7-nitrobenzo-2-oxa-1,3-diazole (NBD-Cl) at 75 °C in a (pH 9) of borate buffer to produce a fluorescent derivative which can be detected at 520 nm afterwards excitation at 460 nm. The method has been validated using ICH criteria, and it demonstrated linearity in a range of 2–120 ngmL-1. The proposed method was applied precisely and accurately for quantifying Pemetrexed within pharmaceutical formulation and spiking human plasma without any interferences. Moreover, the method's sustainability was evaluated and compared to the published method using two greenness assessment tools termed analytical eco-scale and Analytical GREENness (AGREE). That findings suggest that the method is more sustainable than the published method.

Development of a gradient high performance liquid chromatography assay for simultaneous analysis of hydrophilic gemcitabine and lipophilic curcumin using a central composite design and its application in liposome development

Abstract:A rapid and reliable gradient high performance liquid chromatography (HPLC) method was developed for the simultaneous analysis of a hydrophilic anticancer drug, gemcitabine, and a lipophilic phytochemical, curcumin. A central composite design (CCD) was employed to optimize the gradient elution programme to obtain an acceptable retention time and peak width for curcumin with the aid of the desirability function (DF). Dual wavelengths (gemcitabine, 270nm; curcumin, 420nm) were selected to give maximal specific detection using a diode-array detector. The optimized DF was obtained with a mobile phase (water:acetonitrile) of 95:5 for 4.5min, followed by 30:70 for 5min, which gave retention times within 11min with peak widths <0.10min, respectively. No interference from the formulation ingredients was detected. The method was linear over the range of 0.125-5μM for gemcitabine, and 1.25-50μM for curcumin. The intra-day and inter-day variability were less than 5%, with accuracies between 90 and 110% of the true values. The limit of detection (LOD) for gemcitabine and curcumin was 0.04μM and 0.03μM, respectively. Using this method, release study for a dual-drug loaded formulation revealed a Fickian diffusion process for both. The stability of curcumin was pH dependent in aqueous solutions with a half-life of 3h at 25°C and pH 7.4. This was significantly improved after incorporation into the liposomes with no degradation for at least 3 months. In conclusion, the combination of CCD and DF offers a new approach to optimize a gradient HPLC method with high efficiency. A liposomal carrier system may provide an option for tumour dual-drug therapy with the additional advantage of chemical stabilization of the encapsulated compound.

Abstract:Curcumin (Cur) is commonly employed as a food additive in the pharmaceutical and food industries. However, improper use of curcumin can lead to adverse effects on human health. In this research, we developed an electrochemical approach for the synthesis of green fluorescence nitrogen and sulfur co-doped carbon dots (G-CDs) specifically designed for curcumin detection. The prepared G-CDs displayed strong fluorescent emissions at 540 nm, along with notable attributes such as excellent fluorescence stability against photobleaching, and resilience to changes in ionic strength. The G-CDs demonstrated selective recognition of curcumin, resulting in fluorescence quenching in a linear range of 0–140 μmol/L, with an impressive detection limit of 46.4nmol/L. Five distinct food samples were effectively detected for curcumin using the developed G-CDs nanoprobe, yielding satisfactory outcomes. This newly proposed approach offers simplicity, outstanding sensitivity, and selectivity, showcasing its promising capabilities as an alternative method for Cur detection without the need for specialized instruments.

Abstract:Two fluorescence‐based techniques to determine Ambroxol (AMX) and Guaifenesin (GFN) in their isolated forms or combined in pharmaceutical formulations without interference from each other. Ambroxol hydrochloride (AMX) and guaifenesin (GFN) are approved drugs utilized to treat coughs through their potent mucolytic and expectorant properties. Due to their massive, combined administration in many illnesses, there is a persistent need for their concurrent estimation in different pharmaceutical formulations. Two sensitive, environmentally friendly spectrofluorimetric methods were developed. AMX was determined using the first method (I) without interference from GFN. This method depends on the quenching of Erythrosine B (EB) native fluorescence at 552 nm after excitation at 527 nm due to the formation of a non‐fluorescent AMX‐EB ion‐pair complex in Britton–Robinson buffer (BRB) solution pH (3.5). The concentration plot is linear over the 0.25–5.0 μg/mL range, with a mean percent found value of 99.74%. Method (II) depends on measuring the native fluorescence of aqueous GFN solution at two analytical wavelengths, either 300 or 600 nm, after excitation at 274 nm. Relative fluorescence intensity (RFI)–concentration plots are linear over the ranges of 0.02–0.5 and 0.1–2.0 μg/ml, with mean percent found at 99.96% and 99.91% at dual wavelengths, respectively. The proposed methods were successfully applied to assay both drugs in raw materials and different single and combined pharmaceutical formulations. These methods have been thoroughly validated following International Committee on Harmonisation (ICH) guidelines. National Environmental Methods Index, Analytical Eco‐Scale, and Green Analytical Procedure Index were used to prove greenness, thereby enhancing their applicability. The proposed techniques provide straightforward, precise, and cost‐effective solutions for routine formulation analysis in quality control laboratories.

Abstract:Curcumin (Cur) is a heavily used complementary derived drug from cancer patients. Spheroid samples derived from 82 patients were prepared and treated after 48 h with two Cur formulations (CurA, CurB) in mono- and combination therapy. After 72 h, cell viability and morphology were assessed. The Cur formulations had significant inhibitory effects of -8.47% (p < 0.001), CurA of -10.01% (-50.14-23.11%, p = 0.001) and CurB of -6.30% (-33.50-19.30%, p = 0.006), compared to their solvent controls Polyethylene-glycol, β-Cyclodextrin (CurA) and Kolliphor-ELP, Citrate (CurB). Cur formulations were more effective in prostate cancer (-19.54%) and less effective in gynecological non-breast cancers (0.30%). CurA showed better responses in samples of patients <40 (-13.81%) and >70 years of age (-17.74%). CurB had stronger effects in metastasized and heavily pretreated tumors. Combinations of Cur formulations and standard therapies were superior in 20/47 samples (42.55%) and inferior in 7/47 (14.89%). CurB stimulated chemo-doublets more strongly than monotherapies (-0.53% vs. -6.51%, p = 0.022) and more effectively than CurA (-6.51% vs. 3.33%, p = 0.005). Combinations of Cur formulations with Artesunate, Resveratrol and vitamin C were superior in 35/70 (50.00%) and inferior in 16/70 (22.86%) of samples. Cur formulations were significantly enhanced by combination with Artesunate (p = 0.020). Cur formulations showed a high variance in their anti-cancer effects, suggesting a need for individual testing before administration.

Abstract:This subject merits reappraisal since it now appears from data accumulated in the literature as well as our own observations that serious and usually fatal gastrointestinal ulceration in neurologic disorders of children occurs fairly often. Abstract:In this study, the simultaneous determination of bilastine and montelukast, two recently approved co-formulated antihistaminic medications, was accomplished using a quick, sensitive, environmentally friendly, and reasonably priced synchronous fluorescence spectroscopic approach for the first time. Enhancement of the method's sensitivity down to nanogram levels was achieved by the addition of sodium dodecyl sulfate (1.0% w/v) as a micellar system. According to the results, bilastine and montelukast's fluorescence was measured at 255.3 and 355.3 nm, respectively, using Δλ of 40.0 nm and distilled water as a green diluting solvent. With respect to the concentration ranges of bilastine (5.0-300.0 ng/ml) and montelukast (50.0-1000.0 ng/ml), the method showed excellent linearity (r ≥ 0.9998). The results showed that the suggested method is highly sensitive, with detection limits of 1.42 and 13.74 ng/ml for bilastine and montelukast, respectively. Within-run precisions (intra- and interday) per cent relative standard deviations (RSD) for both analytes were <0.59%. With high percentage recoveries and low percentage RSD values, the designed approach was successfully applied for the simultaneous estimation of the cited medications in their dosage form and human plasma samples. To evaluate the green profile of the suggested method, an analytical GREENNESS metric approach (AGREE) and green analytical procedure index (GAPI) metric tools were used. These two methods for evaluating greenness confirmed that the developed method met the highest number of green requirements, recommending its use as a green substitute for the routine analysis of the studied drugs. The proposed approach was validated according to ICHQ2 (R1) guidelines.
Fast concurrent determination of guaifenesin and pholcodine in formulations and spiked plasma using first derivative synchronous spectrofluorimetric approach

Abstract:Green first derivative synchronous spectrofluorometry was used for simultaneous analysis of guaifenesin and pholcodine at 283 nm for guaifenesin and at 275 nm for pholcodine using a wavelength difference (Δλ) of 40 nm; the procedure was successful in determining these drugs in vitro in spiked plasma samples. Guaifenesin and pholcodine are frequently co‐formulated in certain dosage forms. A new fast first derivative synchronous spectrofluorometric method has been used for their simultaneous analysis in mixtures. Here, first derivative synchronous spectrofluorometry enabled the successful simultaneous estimation of guaifenesin at 283 nm and pholcodine at 275 nm using a wavelength difference (Δλ) of 40 nm. The method was fully validated following International Council of Harmonization guidelines. For guaifenesin and pholcodine, linearity was determined within the corresponding ranges of 0.05–0.30 and 0.10–6.0 μg/ml. The two drugs were effectively analyzed using the developed approach in their respective formulations, and the results showed good agreement with those attained using reference methods. The method demonstrated excellent sensitivity, with detection limits down to 0.007 and 0.030 μg/ml and quantitation limits of 0.020 and 0.010 μg/ml for guaifenesin and pholcodine, respectively. Therefore, the procedure was successful in determining these drugs simultaneously in vitro in spiked plasma samples and syrup dosage form. The developed methodology also offered an environmentally friendly advantage by utilizing water as the optimal diluting solvent throughout the whole work. Different greenness approaches were investigated to ensure the method's ecofriendly properties.

Spectrophotometric resolution for quantitative analysis of aspirin and rivaroxaban combination therapy in biological fluids using simple and eco-friendly procedure

Abstract:Patients diagnosed with symptomatic peripheral artery disease (PAD) in the lower extremities have a higher likelihood of suffering from major vascular events. Recently, FDA has approved the combination therapy of aspirin (ASP) and rivaroxaban (ROX) to reduce acute limb ischemia and other comorbidities in (PAD) patients. Zero order and ratio absorption spectra were employed in three simple and accurate spectrophotometric techniques (dual wavelength (DW), ratio difference (RD) and derivative ratio ( 1 DD) for concurrent detection and quantification of ASP and ROX in their pure forms, lab synthetic mixtures and in biological fluid. Our approach involves careful parameter optimization, including solvent selection, sample volumes, and instrumental settings, to reduce the analysis environmental impact. The acquired recovery percentages of accuracy were within 98–102% for pure active pharmaceutical ingredients and 90–110% for pharmaceutical formulations and biological determinations. A comprehensive assessment was done to compare the three methods regarding their ease of use, linearity, sensitivity, conditions, and limitations. The specificity of the proposed methods was evaluated by analyzing the lab synthetic mixtures. The suggested spectrophotometric methods were validated in compliance with ICH guidelines to confirm the validity claims. Also, statistical analysis was done to compare the outcomes obtained from the suggested methods with those obtained from the official ones and they agreed with null hypothesis regarding accuracy and precision. Furthermore, a comprehensive assessment of the environmental sustainability of the developed method was carried out using the Analytical Greenness Calculator, AGREE algorithm. The selected drugs can be efficiently, safely and economically analyzed by the suggested methods in pharmaceutical and biological matrices with no pretreatment or preliminary separation steps and thereby increasing their greenness level.

Abstract:Background: Curcumin shows degradation in the solution of high pH. There is a need for development of a method which can estimate the drug release precisely and accurately. The UV spectroscopy due to its simplicity and specificity is the most popular for method development for the determination of drugs. Objective: The present study was aimed to develop the UV-Visible spectrophotometric method for the assessment of curcumin and tetrahydrocurcumin (THC) using a stable solvent system. Method: The solubility of curcumin and tetrahydrocurcumin was determined at various physiological pH levels. Non-ionic surfactant, tween 80 was used with selected media to avoid the degradation of drugs. The selected solvent system was further validated according to guidelines of the international conference on harmonization (ICH) and the analytical parameters like linearity, veracity, sensitivity and accuracy were studied. Results: The solvent systems Phosphate buffer of pH 6.8+2% tween 80 and Phosphate buffer of pH 6.8+1% tween 80 were found optimum for estimation of curcumin and THC respectively. All the validation parameters were found within the range for developed methods. Conclusion: The proposed method is very simple and can be used for routine quality control testing of curcumin and tetrahydrocurcumin.
